Abstract:Carbon monoxide (CO) is an important gas transporter and is closely associated with a series of physiological and pathological processes. So far, it is clear that plenty of fluorescent probes have been prepared, but most of them still have some disadvantages and is unsuitable for imaging in vivo. Herein, a novel fluorecent probe named HPQ-BI-CO is synthesized and used to detect CO. In this probe, the benindole group is introduced to extend the wavelength. This probe possess a variety of advantages such as long emission wavelength, high sensitivity and selectivity, and rapid response capability. Moreover, the probe possesses a low cytotoxicity, which make it apply in the imaging of exogenous and endogenous CO in living 4T1 cells. Furthermore, this probe can be utilized as a promising tool for fluorescent bioimaging of CO in vivo like zebrafish.

Abstract:Carbon monoxide (CO) is a highly emitted atmospheric pollutant that can pose a serious health risk when its concentration is too high. However, endogenous CO plays an important role in maintaining human health. The development of tools capable of detecting CO in environmental samples and biological samples is therefore essential. Although CO fluorescent probes have been reported, their further applications are limited by factors such as low sensitivity and emission wavelength. Here, we have constructed a CO probe with simple structure, high sensitivity, good selectivity and near-infrared (NIR) emission characteristics. The probe uses the dicyanoisophorone derivative as fluorophore and connects allyl moiety as the reaction site. After the response, the intramolecular charge transfer (ICT) process turns on and releases strong near-infrared fluorescence at 675 nm. The linear range of fluorescence intensity of the probe for CO concentration is 1-10 mu M and the detection limit is as low as 58 nM. The high sensitivity and NIR emission give the probe excellent imaging of cellular exogenous and endogenous CO. Moreover, the probe can also be used to image CO in zebrafish. There is reason to believe that the probe can be a powerful tool for detecting CO concentrations in environmental samples and for analyzing the biological role of CO in organisms.
The construction of a near-infrared fluorescent probe with dual advantages for imaging carbon monoxide in cells and <i>in vivo</i>

Abstract:As a kind of toxic gas, carbon monoxide (CO) can hinder uptake of oxygen in humans. However, more and more studies have shown that CO is an important gaseous messenger in the body and playing an indispensable role in intracellular signaling pathways. So, it is necessary to develop an analytical method to study CO in living organisms. Although there are many CO-responsive probes, most of them have the disadvantages of a small Stokes shift or short emission wavelength. In order to address the above issue, a novel probe (FDX-CO) with a large Stokes shift (190 nm) and long emission wavelength (770 nm) was firstly synthesized to detect CO. The probe shows high sensitivity and superior selectivity toward CO. Moreover, the probe was successfully used for visualizing exogenous and endogenous CO in cells by fluorescence imaging, 3D quantification analysis and flow cytometric analysis. More importantly, FDX-CO could excellently detect CO in mice, which suggests that this probe has the potential ability to image CO in vivo. This probe can be viewed as a useful tool in the research of CO.

Abstract:Carbon monoxide (CO), as one of significant gas transmitter, is closely associated with a variety of physiological and pathological processes. Although plenty of fluorescent probes have been prepared for detecting CO, most of them suffer from water-soluble fluorophores and short emission wavelength, which tends to diffuse and is limited to apply in vivo. Herein, a novel water-soluble fluorescent probe (HPQ-MQ-CO) is prepared to detect CO by releasing a precipitating fluorochrome (HPQ-MQ-OH), which is developed by introducing the 1-ethyl-2-methylquinoline group into HPQ to obtain long emission wavelength and good diffusion resistant ability. Allyl formate, as the identification unit of CO, has good water solubility and quenches the fluorescence of HPQ-MQ-CO. When the probe reacts with CO and Pd2+, an long-emission and solid-state fluorescence signal at 650 nm can be observed, which is based on excited-state intramolecular proton transfer (ESIPT) mechanism. When the concentration of CO is raised to 100.0 μM, the fluorescence is increased 29 times, indicating the sensitivity of the probe. Moreover, this probe shows prominent selectivity for CO compared with other interfering species. Given these advantages, HPQ-MQ-CO can be used for CO detection in HepG2 cells and zebrafish by in-situ and long-term fluorescence imaging. In addition, this probe can monitor the up-regulation of CO in HepG2 cells and zebrafish during drug-induced liver injury (DILI).

Abstract:Carbon monoxide (CO) can be endogenously produced in mammalian cells and regarded as an essential cell signaling molecule in the body. And it is closely related with a range of physiological and pathological processes. Although some fluorescence probes for tracking CO are reported, near-infrared (NIR) fluorescence probe for CO with a large Stokes shift remains scarce. Herein, a NIR fluorescence probe named DCX-CO is designed and synthesized. In the probe, the allyl formate group is chosen as recognizing moiety and benzopyranonitrile-xanthene-based dye is selected as fluorophore. Upon the addition of CO, the probe shows NIR emission at 762 nm with a large Stokes shift (164 nm). Moreover, the probe displays high sensitivity for CO with 16-fold fluorescence enhancement and excellent selectivity to CO over other potential interference in biological sample. Based on the low cytotoxicity, the probe can be used for the detection of exogenous and endogenous CO in HepG2 cells and HCT116 cells. In addition, taking advantage of NIR emission with a large Stokes shift, DCX-CO is applied for monitoring CO in vivo.

Abstract:Carbon monoxide (CO) is an important gas signaling molecule and has been widely involved in regulating important life processes. Effective monitoring of CO in living systems is critical. Combined with the accuracy of ratio detection and the advantages of two-photon imaging, a simple ratiometric two-photon fluorescent probe RTFP was rationally designed and synthesized using 7-(diethylamino)-4-hydroxycoumarin as a two-photon fluorophore and allyl carbonate as the reactive unit. Probe RTFP exhibited excellent selectivity and sensitivity towards CO, and was successfully applied to image endogenous CO in living cells and zebrafish.

Abstract:In this work, by taking full advantage of the transformation of nitro group into amino moiety under the direct reduction of CO, a new metal-free colorimetric and near-infrared fluorescent probe NIR-CO for rapid and specific tracing of intracellular CO has been constructed, and it also become the first reaction-based near-infrared fluorescent probe for CO detection without additional reagents. Probe NIR-CO could quantitatively detect CO at the nanomolar level and the detection limit was calculated as low as 6.1 nM. Additionally, probe NIR-CO with preeminent sensing properties was successfully applied to monitor the fluctuations of endogenous CO levels in living systems. In particular, the up-regulation of heme oxygenase-1 (HO-1) caused by transient glucose deprivation (TGD) in living cells and the down-regulation of HO-1 inhibited by high glucose (HG) in zebrafish were confirmed for the first time via mapping the glucose-induced fluctuations of CO levels. Thus, we anticipate that probe NIR-CO not only can serve as a powerful tool to investigate the detailed biological functions of CO in the pathophysiological processes, but also possesses potential applications of in situ monitoring HO-1 activity in the complex biological systems.

Abstract:As an important cellular signaling molecule, carbon monoxide (CO) is of great significance in maintaining the physiological activities of biological systems. Exploring techniques for detecting of CO at the subcellular fraction is crucial for understanding its various cellular functions. In this work, a simple and effective fluorescence probe (MNP-CO) with mitochondria-targeting was reported for detecting CO in mitochondria. The nitro group was chosen as the recognizing moiety, triphenylphosphine as the mitochondrial target unit and naphthalimide as a fluorescent group. This probe displays high selectivity, high sensitivity and excellent photo-stability. Moreover, biological imaging experiments have shown that MNP-CO has excellent fluorescence imaging ability for CO in cells and zebrafish, and has mitochondrial targeting ability.

Abstract:Hydrogen peroxide (H2O2), as one of important reactive oxygen species (ROS), can be endogenously produced in the liver, and used as a biomarker for predicting drug-induced liver injury (DILI). So, it is important to construct an efficient method for monitoring H2O2 in the liver to assess DILI. Herein, a hepatocyte-targeted fluorescent probe (GCP) for ratiometric detection of H2O2 is developed. GCP consists of two main parts: coumarin-pyran-based fluorophore and galactose-based hepatocyte-targetable group. The probe itself shows near-infrared (NIR) emission owing to its extended p-conjugation. Meanwhile, a large emission shift from 706 to 482 nm (224 nm) is observed after the reaction with H2O2. Taking advantage of the spectroscopic property, GCP exhibits high sensitivity toward H2O2 with 19-fold fluorescence ratio enhancement and is highly selective over other ROS. Moreover, the probe possesses a good hepatocyte-targeting ability and is applied for ratiometric monitoring endogenous H2O2 in HepG2 cells. Furthermore, the probe can trace H2O2 level during DILI in HepG2 cells. Most importantly, GCP can locate in the liver of zebrafish and detect H2O2 level in the liver during DILI, which will be a potentially useful tool to assess DILI.

Abstract:Carbon monoxide (CO) not only causes damage to life and health as an environmental pollutant, but also undertakes many physiological functions in organisms. In particular, developing means that can be used for the determination of CO in organelles will provide insight into the vital role it plays. Studies have shown that mitochondrial respiration is closely related to CO concentrations, so it is critical to develop tools for CO detection in mitochondria. Here, we use a rhodamine derivative that can target mitochondria as fluorophores to construct a mitochondrial-labeled CO fluorescence probe (Rh-CO) with high sensitivity (detection limit: 9.4 nM), excellent water-solubility, and long emission (λem = 630 nm). Prominently, the probe has outstanding mitochondria-targeting capabilities. Moreover, we used transient glucose deprivation (TGD) and heme to stimulate endogenous CO production in living cells and zebrafish, respectively, and the probe exhibited excellent imaging capabilities. All in all, we expect this probe to contribute to a deeper understanding of the role played by CO in mitochondria.

Abstract:Developing a reliable tool for monitoring peroxynitrite (ONOO-) in the complex organisms is critical to uncover its detailed cellular functions in numerous physiological and pathological processes. Thus, a novel ratiometric fluorescent probe was reasonably designed for ONOO-, in which the rhodol-HBT conjugate dye was chosen as a fluorophore, and hydrazine moiety was adopted as recognition receptor of ONOO-. The probe exhibited outstanding sensitivity, specificity, and a fast response towards ONOO-. More importantly, the probe with a large emission shift (Delta lambda(em)= 127 nm) has been proven to be capable of tracking ONOO- with high resolution in live cells and zebrafish. This work provides a simple and reliable tool for detecting and imaging ONOO- in the living systems, and thus we anticipate that the designed probe could be used as a powerful imaging tool in unraveling biological functions of ONOO-.

Abstract:As an important gaseous signal molecule, carbon monoxide (CO) plays a major role in multiple physiological and pathological processes. Therefore, it is significantly meaningful to develop fluorescent probes with features such as in situ imaging to detect CO in biological samples. However, the reported CO fluorescent probes still suffer from low sensitivity, poor selectivity, and low water solubility. In particular, due to the very low concentration of CO in organisms, these probes must have high sensitivity to achieve the detection of CO. To address this problem, we constructed a CO fluorescence probe ( HS-CO ) with a reasonable design, which can greatly increase the rigid plane of the probe through spontaneous intramolecular cyclization after the reaction with CO. The substantial increase in the fluorescence quantum yield after the reaction enabled it to achieve a strong change in the fluorescence signal and exhibit high sensitivity for detecting CO (LOD = 4.5 nM). Importantly, probe HS-CO was successfully used to detect endogenous and exogenous CO in HeLa cells. In addition, probe HS-CO successfully detected CO in zebrafish. These results together identified the great potential of the probe as a tool for functional analysis of CO in organisms.

Abstract:The sensitive and accurate detection of intracellular gasotransmitter carbon monoxide (CO) is of great significance for unraveling its diverse cellular functions. Herein, we report a simple highly specific and sensitive 4-hydroxynaphthalimide-based ratiometric fluorescent probe Ratio-CO for monitoring CO levels in living cells. Probe Ratio-CO could quantitatively detect CO in the range of 0-50 M by the ratiometric fluorescence spectroscopy method and the detection limit was measured to be about 17.9 nM. Additionally, probe Ratio-CO has been proven to possess preeminent selectivity towards CO over other bioactive species. Most importantly, the excellent response properties of probe Ratio-CO enable its applications in the monitoring of the fluctuations of CO levels in living cells by the ratiometric fluorescence imaging technique, and we thus anticipate that this probe would be a novel tool for further elucidating the biological functions of CO in living systems.
Golgi-targetable fluorescent probe for ratiometric imaging of CO in cells and zebrafish

Abstract:Carbon monoxide (CO) plays a vital role in living organisms. The detection of CO at the subcellular level is of great significance to clarify its biological functions. Increasing evidence shows that CO is important in Golgi apparatus, but so far, no probe has been reported to detect CO in Golgi. In this paper, an effective fluorescence probe Gol-CO was reported for CO imaging in Golgi apparatus. Gol-CO uses a Golgi-targetable aminoquinoline derivative as the fluorophore and importantly, it shows significant ratiometric fluorescence signal changes at 520/425 nm in aqueous solutions with high selectivity and sensitivity (about 41 nM of detection limit) for CO. Imaging experiments showed that Gol-CO has excellent targeting ability to Golgi apparatus and excellent ratiometric fluorescence imaging ability for CO in cells and zebrafish. Gol-CO also succeeded in visualizing the level increase of CO in living cells under oxidative stress.

Abstract:Peroxynitrite (ONOO-) has been proven to participate in various physiological and pathological processes, and may also be a contributing factor in many diseases. In view of this, there is a need to develop detection tools for unambiguously tracking a small amount of endogenous ONOO- to reveal its exact mechanisms. In this paper, a colorimetric and red-emitting fluorescent probe Red-PN, based on a rhodamine-type fluorophore and hydrazide reactive site is described. The probe Red-PN possesses the advantages of rapid response (within 5 s), visual color change (from colorless to pink), preeminent sensitivity (detection limit = 4.3 nM) and selectivity. Because of these outstanding performances, it was possible to accurately detect endogenous ONOO-. It was encouraging that the probe Red-PN could be used effectively for tracking the relatively low levels of endogenous and exogenous ONOO-in living cells and zebrafish. Thus, it is envisioned that the probe Red-PN would have promising prospects in applications for imaging ONOO- in a variety of biological settings.

Abstract:Hydrogen peroxide(H2O2), as a reliable signaling biomolecule for oxidative stress, its accurate detection during agent-stimulated oxidative stress plays a vital role in pathological and physiological mechanism exploration for disease theranostics. It's necessary to develop an efficient method for their detection. In view of the advantages of fluorescent probes, we rationally constructed a novel fluorescent probe Compound 2 based on 4-(Bromomethyl)benzeneboronic acid pinacol ester_Herein, a small molecule fluorescent probe was fabricated using isoflore nitrile as fluorescent group, phenylboronic acid pinacol ester as the response group, to detect H2O2. The probe Compound 2 has a strong fluorescence intensity at 575 nm, indicating that the structure of the probe molecule is reasonably designed, and the Stokes shift is up to 172 nm. While the detection time is as low as 30 s and the LOD of the probe for H2O2 is as low as 3.7 μmol/L,the quantum yield is Φ = 40.31 %. It has been successfully used for imaging detection of H2O2 in HepG2 cells and zebrafish for its low toxicity. It can be found that this small molecule fluorescent probe can identify H2O2 in tumor cells significantly and efficiently, which would realize the early diagnosis of tumor.

Abstract:As one of the major reactive oxygen species (ROS), superoxide anion (O2•-) is engaged in maintaining redox homeostasis in the cell microenvironment. To identify the pathological roles in related disorders caused by abnormal expression of O2•-, it is of great significance to monitor and track the fluctuation of O2•- concentration in vivo. However, the low concentration of O2•- and the interference caused by tissue autofluorescence make the development of an ideal detection methodology full of challenges. Herein, a "Turn-On" chemical response near-infrared (NIR) fluorescence probe Dcm-Cu-OTf for O2•- detection in inflamed models, was constructed by conjugating the NIR fluorophore (dicyanisophorone derivative) with an O2•- sensing moiety (trifluoromethanesulfonate). Dcm-Cu-OTf exerted about 140-fold fluorescence enhancement after reacting 200 μM O2•- with an excellent limited of detection (LOD) as low as 149 nM. Additionally, Dcm-Cu-OTf exhibited a super large Stokes shift (260 nm) and high selectivity over other bio-analytes in stimulated conditions. Importantly, Dcm-Cu-OTf showed low toxicity and enabled imaging of the generation of O2•- in the Lipopolysaccharide (LPS)-stimulated HeLa cells, zebrafish, and LPS-induced inflamed mice. The present study provided a potential and reliable detection tool to inspect the physiological and pathological progress of O2•- in living biosystems.

Abstract:CO-releasing molecule-2 (CORM-2), as a safe donor for releasing CO, has been widely adopted to disclose the physiopathological roles of CO and used as therapeutic drug replacing gaseous CO. The accurate determination of CORM-2 levels in biological systems is central for understanding its safe use as therapeutic agent and the cellular functions of CO. In this paper, a novel fluorescent probe CPD-NB was synthesized, which provided a safe and convenient way for detecting CORM-2 without the requirement of Pd2+ as an additional substrate. It featured a robust fluorescence response to CORM-2 with desirable sensitivity (the detection limit was as low as 4 nM), and showed high selectivity toward CORM-2 over other biologically relevant species. Moreover, with the low cytotoxicity of probe CPD-NB, the visualizations of CORM-2 changes in live cells and zebrafish were also achieved. Thus, we expected that probe CPD-NB could be as an ideal platform for safe and powerful imaging of CORM-2 in biological systems, and may thus promote the progress of CO-related research.
