Simultaneous Motion and Respiration Measurement in Adult Zebrafish Based on the Instantaneous Spatial Distribution of Separated Ventilatory Signals

Abstract:In this study, a novel system was developed for the simultaneous respiration and motion measurement of zebrafish for behavioral research. We propose a model to capture the instantaneous spatial distribution of ventilatory signals for estimating parameters originating from the fish's head and tail. By using these estimated parameters, the proposed method accurately determines the swimming position, body orientation, and respiratory frequency of fish. The measurement uncertainty analysis shows a distance error of approximately mm, mean velocity error of 0.01 mm/s with limits of agreement (LOA) in the range of [−14.26, 13.32] mm/s, and mean body-orientation angle error of 0.03 rad with LOA in the range of [−1.29, 1.35] rad, all compared with standard video camera analysis. Furthermore, this study investigated the effects of caffeine on zebrafish and confirmed a significant increase in respiratory frequency. Therefore, the proposed method can be used to study behavioral patterns in adult zebrafish and advance research in this field.
