Abstract:A deeper understanding of the key processes that determine the particle morphologies generated during aerosol droplet drying is highly desirable for spray-drying of powdered pharmaceuticals and foods, predicting the properties of atmospheric particles, and monitoring disease transmission. Particle morphologies are affected by the drying kinetics of the evaporating droplets, which are in turn influenced by the composition of the initial droplet as well as the drying conditions. Herein, we use polymerization-induced self-assembly (PISA) to prepare three types of sterically stabilized diblock copolymer nanoparticles comprising the same steric stabilizer block and differing core blocks with z-average diameters ranging from 32 to 238 nm. These well-defined nanoparticles enable a systematic investigation of the effect of the nanoparticle size and composition on the drying kinetics of aqueous aerosol droplets (20-28 μm radius) and the final morphology of the resulting microparticles. A comparative kinetics electrodynamic balance was used to obtain evaporation profiles for 10 examples of nanoparticles at a relative humidity (RH) of 0, 45, or 65%. Nanoparticles comprising the same core block with mean diameters of 32, 79, and 214 nm were used to produce microparticles, which were dried under different RH conditions in a falling droplet column. Scanning electron microscopy was used to examine how the drying kinetics influenced the final microparticle morphology. For dilute droplets, the chemical composition of the nanoparticles had no effect on the evaporation rate. However, employing smaller nanoparticles led to the formation of dried microparticles with a greater degree of buckling.

Abstract:Spray drying is an approach employed in automotive, food, and pharmaceutical industries as a robust and cost efficient liquid atomization technique offering direct control over droplet dimensions. The majority of commercially available spray nozzles are designed for large throughput spray drying applications or uniform surface coating, but microfluidic nebulizers have recently been developed as small scale alternatives. Here, we explore the physical parameters that define the droplet size and formation under supersonic flow conditions commonly found in microfluidic spray drying systems. We examined the spray nozzle operation using high speed imaging and laser scattering measurements, which allowed us to describe the spray regimes and droplet size distributions. It was determined that by using this spray nozzle device, droplets with diameters of 4–8 <i>μ</i>m could be generated. Moreover, we show that the supersonic de Laval nozzle model can be used to predict the average droplet size. Our approach can be used as a platform for interfacing fluid microprocessing with gas phase detection and characterization.

Abstract:Spray freeze-drying has emerged as a valid alternative to traditional spray drying to produce therapeutic dry microparticles. In particular, the spherical shape and high porosity of spray freeze-dried microparticles make them suitable for pulmonary drug delivery through dry powder inhalers. However, an appropriate particle size and fine particle fraction are required to guarantee lung deposition. This study used ultrasonic spray freeze-drying to generate dry microparticles composed of mannitol either alone or added with the bronchodilator salbutamol sulphate. The influence of the solid concentration and the feed flow rate on the particle size, morphology, surface area, porosity, and crystallinity was investigated. Growing particle size was observed, increasing the concentration and feed flow rate. Similarly, the addition of the drug led to a larger particle size and surface area. The in vitro simulation of drug deposition highlighted the dependence of the aerodynamic properties on the solid concentration and feed flow rate. Due to the lower density and particle geometric size, the highest fine particle fraction (26%) and smallest mass median aerodynamic diameter (4.4 μm) were reached at the lowest solid concentration and feed flow rate.

Abstract:Spray drying is gaining traction in the pharmaceutical industry as one of the processing methods of choice for the manufacture of solid dosage forms intended for pulmonary, oral, and parenteral delivery. This process is particularly advantageous because of its ability to produce engineered particles with improved efficacy and stability by combining active pharmaceutical ingredients or biologics with appropriate excipients. Moreover, due to its high throughput, continuous operation, and ability to produce thermostable solid powders, spray drying can be a manufacturing method of choice in the production of drugs and other formulations, including vaccines, for global distribution. Formulation design based on a mechanistic understanding of the different phenomena that occur during the spray drying of powders is complicated and can therefore make the use of available particle formation models difficult for the practitioner. This review aims to provide step-by-step guidance accompanied by critical background information for the successful formulation design of spray-dried microparticles. These include discussion of the tools needed to estimate the surface concentration of each solute during droplet drying, their times and modes of solidification, and the amount of glass stabilizers and shell formers required to produce stable and dispersible powders.

Abstract:Spray drying and electrospraying are well-established drying processes that already have proven their value in the pharmaceutical field. However, there is currently still a lack of knowledge on the fundamentals of the particle formation process, thereby hampering fast and cost-effective particle engineering. To get a better understanding of how functional particles are formed with respect to process and formulation parameters, it is indispensable to offer a comprehensive overview of critical aspects of the droplet drying and particle formation process. This review therefore closely relates single droplet drying to pharmaceutical applications. Although excellent reviews exist of the different aspects, there is, to the best of our knowledge, no single review that describes all steps that one should consider when trying to engineer a certain type of particle morphology. The findings presented in this article have strengthened the predictive value of single droplet drying for pharmaceutical drying applications like spray drying and electrospraying. Continuous follow-up of the particle formation process in single droplet drying experiments hence allows optimization of manufacturing processes and particle engineering approaches and acceleration of process development.

Abstract:Introduction: The rising demand for pharmaceutical particles with tailored physicochemical properties has opened new markets for spray drying especially for solubility enhancement, improving inhalation medicines and stabilization of biopharmaceuticals. Despite this, the spray drying literature is scattered and often does not address the principles underpinning robust development of pharmaceuticals. It is therefore necessary to present clearer picture of the field and highlight the factors influencing particle design and scale-up. Areas covered: The review presents a systematic analysis of the trends in development of particle delivery systems using spray drying. This is followed by exploring the mechanisms governing particle formation in the process stages. Particle design factors including those of equipment configurations and feed/process attributes were highlighted. Finally, the review summarises the current industrial approaches for upscaling pharmaceutical spray drying. Expert opinion: Spray drying provides the ability to design particles of the desired functionality. This greatly benefits the pharmaceutical sector especially as product specifications are becoming more encompassing and exacting. One of the biggest barriers to product translation remains one of scale-up/scale-down. A shift from trial and error approaches to model-based particle design helps to enhance control over product properties. To this end, process innovations and advanced manufacturing technologies are particularly welcomed.
Advantages and challenges of the spray-drying technology for the production of pure drug particles and drug-loaded polymeric carriers

Abstract:Spray-drying is a rapid, continuous, cost-effective, reproducible and scalable process for the production of dry powders from a fluid material by atomization through an atomizer into a hot drying gas medium, usually air. Often spray-drying is considered only a dehydration process, though it also can be used for the encapsulation of hydrophilic and hydrophobic active compounds within different carriers without substantial thermal degradation, even of heat-sensitive substances due to fast drying (seconds or milliseconds) and relatively short exposure time to heat. The solid particles obtained present relatively narrow size distribution at the submicron-to-micron scale. Generally, the yield% of spray-drying at laboratory scale with conventional spray-dryers is not optimal (20-70%) due to the loss of product in the walls of the drying chamber and the low capacity of the cyclone to separate fine particles (<2 μm). Aiming to overcome this crucial drawback in early development stages, new devices that enable the production of submicron particles with high yield, even for small sample amounts, have been introduced into the market. This review describes the most outstanding advantages and challenges of the spray-drying method for the production of pure drug particles and drug-loaded polymeric particles and discusses the potential of this technique and the more advanced equipment to pave the way toward reproducible and scalable processes that are critical to the bench-to-bedside translation of innovative pharmaceutical products.

Abstract:Theimpact of particle hydrophobicity on the morphology and structureof hollow microspheres prepared by spray drying of silica sol wasinvestigated through modification with & gamma;-methylacryloxypropyltrimethoxysilane(& gamma;-MPS). Hydrophobic attraction among the hydrophobic silicaparticles in the aqueous phase drives particles to gather inward thedroplet at the early stages of dehydration, which delays the shellformation of the droplets. During the later stages of dehydration,hydrophobic particles form hydrophobic nanoconfined channels amongthemselves. Afterward, water undergoes rapid evaporation via the formationof molecular clusters, facilitating the formation of a thicker andstronger shell, which mitigates the formation of the tail vortex resultingfrom the adverse effects of airflow. After continued dehydration,regular hollow microspheres are formed. Cationic surfactant cetyltrimethylammoniumbromide (CTAB) was absorbed onto the negatively charged particle surfaceof silica sol, thereby inducing hydrophobicity via electrostatic attraction.Subsequently, the CTAB-adsorbed silica sol is spray dried in a mixedsolvent phase of ethanol and water, resulting in the preparation ofregular, morphologically uniform hollow microspheres. The robust synthesisand the underlying mechanistic study provide a scalable and sustainableroute to prepare hollow microspheres for potential application incosmetics.

Abstract:Inhalation is used for local therapy of the lungs and as an alternative route for systemic drug delivery. Modern powder inhalation systems try to target the required site of action/absorption in the respiratory tract. Large porous particles (LPPs) with a size >5 μm and a low mass density (usually measured as bulk or tapped) of <0.4 g/cm3 can avoid protective lung mechanisms. Their suitable aerodynamic properties make them perspective formulations for deep lung deposition. This experiment studied the effect of spray-drying process parameters on LPP properties. An experimental design of twelve experiments with a central point was realized using the Box–Behnken method. Three process parameters (drying temperature, pump speed, and air speed) were combined on three levels. Particles were formed from a D-mannitol solution, representing a perspective material for lung microparticles. The microparticles were characterized in terms of physical size (laser diffraction), aerodynamic diameter (aerodynamic particle sizer), morphology (SEM), and densities. The novelty and main goal of this research were to describe how the complex parameters of the spray-drying process affect the properties of mannitol LPPs. New findings can provide valuable data to other researchers, leading to the easy tuning of the properties of spray-dried particles by changing the process setup.

Abstract:Liquid atomization is applied in nanostructure dense coating technology to inject suspended nano-size powder materials into a suspension plasma spray (SPS) torch. This paper presents the effects of the atomization parameters on the nanoparticle processing. A numerical model was developed to simulate the dynamic behaviors of the suspension droplets, the solid nanoparticles or agglomerates, as well as the interactions between them and the plasma gas. The plasma gas was calculated as compressible, multi-component, turbulent jet flow in Eulerian scheme. The droplets and the solid particles were calculated as discrete Lagrangian entities, being tracked through the spray process. The motion and thermal histories of the particles were given in this paper and their release and melting status were observed. The key parameters of atomization, including droplet size, injection angle and velocity were also analyzed. The study revealed that the nanoparticle processing in SPS preferred small droplets with better atomization and less aggregation from suspension preparation. The injection angle and velocity influenced the nanoparticle release percentage. Small angle and low initial velocity might have more nanoparticles released. Besides, the melting percentage of nanoparticles and agglomerates were studied, and the critical droplet diameter to ensure solid melting was drawn. Results showed that most released nanoparticles were well melted, but the agglomerates might be totally melted, partially melted, or even not melted at all, mainly depending on the agglomerate size. For better coating quality, the suspension droplet size should be limited to a critical droplet diameter, which was inversely proportional to the cubic root of weight content, for given critical agglomerate diameter of being totally melted.

Abstract:Spray freeze drying is an attractive technology for fabricating pharmaceutical powder, but it is still challenging to tailor the microparticle physical and chemical structure. Herein, a series of uniform mannitol microparticles with controllable morphology and crystal properties were successfully fabricated via a self-developed micro-fluidic jet spray freeze tower. The effects of feed solution composition and freezing temperature on microparticle size, morphology, surface architecture and crystal property were investigated to propose the particle formation mechanism. Freezing temperature showed more apparent effect on the microparticle characteristics with higher solid content (10 and 15 w/w%) in aqueous solution; Whereas the addition of ethanol as a co-solvent had a notable impact on microparticle morphology, concurrently perturbing the crystallization tendencies of mannitol. These results provide new insights for producing spray freeze dried pharmaceutical microparticles.

Abstract:Spray freeze-drying is an emerging manufacturing technology that offers many advantages, including long-term stability, consistent particle size distribution, and enhanced bioavailability. However, its implementation on an industrial scale is still hampered by several technical problems relating to the design of the equipment and the selection of appropriate operating conditions. This study aims to clarify the relationship between the atomization conditions (atomizing power, feed flow rate, and viscosity) and the characteristics of the lyophilized powder (particle size distribution and morphology) for two model products (sucrose- and mannitol-based formulations). Independently of the formulation, the particle morphology only depended on its solid content, while the average particle size increased with the feed flow rate and viscosity. Lastly, the specific surface area of the lyophilized powder varied with the initial solid content and the type of excipient as well, with mannitol-based particles having the highest specific surface area.

Abstract:In this review, benefits and drawbacks of the process of spray drying and nano spray drying with regard to the manufacturing of polymeric particles for pharmaceutical applications are discussed. Spray drying has been used for many years in the food, chemical and pharmaceutical industries for converting liquids into solids, in order to form products of uniform appearance. The construction of spray dryer enables to atomize the liquid into small droplets, which ensures a large surface area for heat and mass transfer, and significantly shortens the processing. Each droplet dries to an individual solid microparticle of characteristic features that can be tailored by optimizing formulation variables and critical process parameters. Since spray drying technology is easy to scale up and can be used for drying almost any drug in a solution or suspension, there are numerous examples of products in clinical use, in which this process has been successfully applied to improve drug stability, enhance bioavailability or control its release rate. In recent years, nano spray drying technology has been proposed as a method for lab-scale manufacturing of nanoparticles. Such an approach is of particular interest at early stages of drug development, when a small amount of new chemical entities is available. Here, the nebulization technique is used for feed atomization, while laminar gas flow in the drying chamber ensures gentle drying conditions. Moreover, electrostatic collectors have gradually replaced cyclone separators, ensuring high effectiveness in producing solid nanoparticles, even if a small volume of the sample is processed.

Abstract:The research is devoted to the generation of fine particles and droplet flows with specified characteristics for a wide range of scientific problems. The aspects of aerodynamic fine atomization and the effects of atomizing gas density, gas velocity and mass flow rate, and liquid film thickness on the droplet formation are investigated. The hypothesis of highly efficient utilization of secondary droplets from a coarse polydisperse flow to produce fine particles is suggested and experimentally confirmed. A prototype device to implement the idea of separating the desired droplets fraction from a primary polydisperse flow was developed. In the case of fine liquid atomization, the developed spraying system enabled an increase in the gas-droplet flow concentration. The possibility of producing particles with different dispersiveness and morphology by employing the methods of spray separation and dilute solution atomization is demonstrated. The criteria of the system settings to generate the droplet flow with specified characteristics were determined. A Laval nozzle design is suggested for liquid atomization. A series of numerical experiments showed that a significant decrease in gas pressure (density) at the nozzle outlet provides an increase in the gas flow rate. The numerical estimation showed that the Laval nozzle proposed can improve the fine atomization efficiency by 30 percent.
Dynamic Modeling of Micro- and Nano-Sized Particles Impinging on the Substrate During Suspension Plasma Spraying

Abstract:Suspension plasma spraying (SPS) can be utilized to manufacture finely structured coatings. In this process, liquid suspended with micro- or nano-sized solid particles is injected into a plasma jet. It involves droplet injection, solvent evaporation, and discharge, acceleration, heating, and melting of the solid particles. The high-speed and high-temperature particles finally impact on the substrate wall, to form a thin layer coating. In this study, a comprehensive numerical model was developed to simulate the dynamic behaviors of the suspension droplets and the solid particles, as well as the interactions between them and the plasma gas. The plasma gas was treated as compressible, multi-component, turbulent jet flow, using Navier-Stokes equations solved by the Eulerian method. The droplets and solid particles were treated as discrete Lagrangian entities, being tracked through the spray process. The drag force, Saffman lift force, and Brownian force were taken into account for the aerodynamic drag force, aerodynamic lift force, and random fluctuation force imposed on the particles. Spatial distributions of the micro- and nano-sized particles are given in this paper and their motion histories were observed. The key parameters of spray distribution, including particle size and axial spray distance, were also analyzed. The critical size of particle that follows well with the plasma jet was deduced for the specified operating conditions. Results show that in the downstream, the substrate influences the flow field structure and the particle characteristics. The appropriate spray distances were obtained for different micro- and nano-sized particles.

Abstract:Early development drug formulation is exacerbated by increasingly poor bioavailability of potential candidates. Prevention of attrition due to formulation problems necessitates physicochemical analysis and formulation studies at a very early stage during development, where the availability of a new substance is limited to small quantities, thus impeding extensive experiments. Miniaturization of common formulation processes is a strategy to overcome those limitations. We present a versatile technique for fabricating drug nanoformulations using a microfluidic spray dryer. Nanoparticles are formed by evaporative precipitation of the drug-loaded spray in air at room temperature. Using danazol as a model drug, amorphous nanoparticles of 20-60 nm in diameter are prepared with a narrow size distribution. We design the device with a geometry that allows the injection of two separate solvent streams, thus enabling co-spray drying of two substances for the production of drug co-precipitates with tailor-made composition for optimization of therapeutic efficiency.

Abstract:This study was dealing with the spray drying of tailored mannitol particles as carriers in dry powder inhalation formulations. A self-constructed spray tower equipped with a laminar rotary atomizer was used to generate very narrow particle-size distributions. A design of experiments with rotation speed and drying temperature as factors was applied for the preparation of a broad set of different mannitol particles. Drying parameters affected particle properties like particle size, particle shape, surface roughness, and flowability significantly. Here, rotation speed was mainly affecting droplet and therefore particle size. Further, it was found that the inner particle temperature, and so the pressure inside the early particle shell, plays an important role during particle formation as emerging water vapor at higher temperatures served for indented particles and rough surface structures. Lower drying temperatures resulted in spherical and smooth particles as the inner temperature remained below the boiling temperature. The occurrence of these particle properties could further be related to mannitol carrier particle flow dynamics as large indented carriers were found to be less flowable than small spherical ones. In general, it was possible to control particle properties by the parameters of the spray drying process.

Abstract:Nanoparticle-based therapeutics have been used in pulmonary formulations to enhance delivery of poorly water-soluble drugs, protect drugs against degradation and achieve modified release and drug targeting. This review focuses on the use of spray drying as a solidification technique to produce microparticles containing nanoparticles (i.e., nanoparticle (NP) agglomerates) with suitable properties as dry powders for inhalation. The review covers the general aspects of pulmonary drug delivery with emphasis on nanoparticle-based dry powders for inhalation and the principles of spray drying as a method for the conversion of nanosuspensions to microparticles. The production and therapeutic applications of the following types of NP agglomerates are presented: nanoporous microparticles, nanocrystalline agglomerates, lipid-based and polymeric formulations. The use of alternative spray-drying techniques, namely nano spray drying, and supercritical CO2-assisted spray drying is also discussed as a way to produce inhalable NP agglomerates.

Abstract:Spray pyrolysis process has been used for the synthesis of non-oxide ceramic powders from liquid precursors in the Si/C/N system. Particles with a high thermal stability and with variable composition and size distribution have been obtained. In this process, the mechanisms involved in precursor decomposition and gas phase recombination of species are still unknown. The final aim of this work consists in improving the whole process comprehension by an experimental/modelling approach that helps to connect the synthesized particles characteristics to the precursor properties and process operating parameters. It includes the following steps: aerosol formation by a piezoelectric nebulizer, its transport and the chemical-physical phenomena involved in the reaction processes. This paper focuses on the aerosol characterization to understand the relationship between the liquid precursor properties and the liquid droplet diameter distribution. Liquids with properties close to the precursor of interest (hexamethyldisilazane) have been used. Experiments have been performed using a shadowgraphy technique to determine the drop size distribution of the aerosol. For all operating parameters of the nebulizer device and liquids used, bimodal droplet size distributions have been obtained. Correlations proposed in the literature for the droplet size prediction by ultrasonic nebulization were used and adapted to the specific nebulizer device used in this study, showing rather good agreement with experimental values.
