Abstract:Although it has been known for half a century that the physical aging of glasses in experiments is described well by a linear thermal-history convolution integral over the so-called material time, the microscopic definition and interpretation of the material time remains a mystery. We propose that the material-time increase over a given time interval reflects the distance traveled by the system's particles. Different possible distance measures are discussed, starting from the standard mean-square displacement and its inherent-state version that excludes the vibrational contribution. The viewpoint adopted, which is inspired by and closely related to pioneering works of Cugliandolo and Kurchan of the 1990s, implies a "geometric reversibility" and a "unique-triangle property" characterizing the system's path in configuration space during aging. Both of these properties are inherited from equilibrium; they are confirmed by computer simulations of an aging binary Lennard-Jones system. Our simulations show that the slow particles control the material time. This motivates a "dynamic-rigidity-percolation" picture of physical aging. The numerical data show that the material time is dominated by the slowest particles' inherent mean-square displacement, which is conveniently quantified by the inherent harmonic mean-square displacement. This distance measure collapses data for potential-energy aging well in the sense that the normalized relaxation functions following different temperature jumps are almost the same function of the material time. Finally, the standard Tool-Narayanaswamy linear material-time convolution integral description of physical aging is derived from the assumption that when time is replaced by distance in the above sense, an aging system is described by the same expression as that of linear-response theory.

Abstract:Here we consider human aging and time perception by operating with time, as a physical quantity, linked to periodic motions of the Earth. Taking into account the periodicity of Earth's revolutions, the model of aging in a form like Fermat's spiral has been proposed. This model demonstrates the progressive extension of the paces of aging implying accelerated accumulation of adverse changes over life, which is, as a vector, directed to the center of the spiral. In this regard, an internal clock underpinning time perception that was evolved with life under the permanent influence of the Earth's motion events can accelerate with aging presumably due to progressive extension of the paces of aging.

Abstract:Having made substantial progress understanding molecules, cells, genes and pathways, aging biology research is now moving toward integration of these parts, attempting to understand how their joint dynamics may contribute to aging. Such a shift of perspective requires the adoption of a formal complex systems framework, a transition being facilitated by large-scale data collection and new analytical tools. Here, we provide a theoretical framework to orient researchers around key concepts for this transition, notably emergence, interaction networks and resilience. Drawing on evolutionary theory, network theory and principles of homeostasis, we propose that organismal function is accomplished by the integration of regulatory mechanisms at multiple hierarchical scales, and that the disruption of this ensemble causes the phenotypic and functional manifestations of aging. We present key examples at scales ranging from sub-organismal biology to clinical geriatrics, outlining how this approach can potentially enrich our understanding of aging.

Abstract:Author's early work on aging is developed to yield a relationship between life spans and the velocity of aging. The mathematical analysis shows that the mean extent of the advancement of aging throughout one's life is conserved, or equivalently, the product of the mean life span, and the mean rate of aging is constant. The result is in harmony with our experiences: It accounts for the unlimited replicability of tumor cells, and predicts the prolonged life spans of hibernating hamsters, in accordance with the Lyman and coworkers experiment. Comparing the present result and the empirical relationship between life spans of various mammals and basal metabolic rates, it is suggested that the mean rate of aging is intimately connected with the mean basal metabolic rate. With the help of this information, we inquire the reason of the difference in mean life spans between women and men, the result showing that the relative mean life span of women to men is 1 08, for various nations, which is close to the corresponding relative value of the basal metabolic rate. The present theory suggests, however, that this relationship between life spans and basal metabolic rates must be treated with caution.

Abstract:The gradual accumulation of damage and dysregulation during the aging of living organisms can be quantified. Even so, the aging process is complex and has multiple interacting physiological scales -- from the molecular to cellular to whole tissues. In the face of this complexity, we can significantly advance our understanding of aging with the use of computational models that simulate realistic individual trajectories of health as well as mortality. To do so, they must be systems-level models that incorporate interactions between measurable aspects of age-associated changes. To incorporate individual variability in the aging process, models must be stochastic. To be useful they should also be predictive, and so must be fit or parameterized by data from large populations of aging individuals. In this perspective, we outline where we have been, where we are, and where we hope to go with such computational models of aging. Our focus is on data-driven systems-level models, and on their great potential in aging research.

Abstract:Time perception is crucial for a coherent human experience. As life progresses, our perception of the passage of time becomes increasingly non-uniform, often feeling as though it accelerates with age. While various causes for this phenomenon have been theorized, a comprehensive mathematical and theoretical framework remains underexplored. This study aims to elucidate the mechanisms behind perceived time dilation by integrating classical and revised psychophysical theorems with a novel mathematical approach. Utilizing Weber-Fechner laws as foundational elements, we develop a model that transitions from exponential to logarithmic functions to represent changes in time perception across the human lifespan. Our results indicate that the perception of time shifts significantly around the age of mental maturity, aligning with a proposed inversion point where sensitivity to temporal stimuli decreases, eventually plateauing out at a constant rate. This model not only explains the underlying causes of time perception changes but also provides analytical values to quantify this acceleration. These findings offer valuable insights into the cognitive and neurological processes influencing how we experience time as we go through life.

Abstract:In humans, age-associated degrading changes, widely observed in molecular and cellular processes underly the time-dependent decline in spatial navigation, time perception, cognitive and psychological abilities, and memory. Cross-talk of biological, cognitive, and psychological clocks provides an integrative contribution to healthy and advanced aging. At the molecular level, genome, proteome, and lipidome instability are widely recognized as the primary causal factors in aging. We narrow attention to the roles of protein aging linked to prevalent amino acids chirality, enzymatic and spontaneous (non-enzymatic) post-translational modifications (PTMs SP), and non-equilibrium phase transitions. The homochirality of protein synthesis, resulting in the steady-state non-equilibrium condition of protein structure, makes them prone to multiple types of enzymatic and spontaneous PTMs, including racemization and isomerization. Spontaneous racemization leads to the loss of the balanced prevalent chirality. Advanced biological aging related to irreversible PTMs SP has been associated with the nontrivial interplay between somatic (molecular aging) and mental (psychological aging) health conditions. Through stress response systems (SRS), the environmental and psychological stressors contribute to the age-associated “collapse” of protein homochirality. The role of prevalent protein chirality and entropy of protein folding in biological aging is mainly overlooked. In a more generalized context, the time-dependent shift from enzymatic to the non-enzymatic transformation of biochirality might represent an important and yet underappreciated hallmark of aging. We provide the experimental arguments in support of the racemization theory of aging.

Abstract:The question, "Can aging be modified, delayed, or reversed?" has profound social and economic implications for rapidly aging societies today. Interventions, ideally, would intercept functional decline and extend healthspan by delaying late-life morbidity (known as "squaring the curve"). These have proven elusive, but examples of differential aging in the animal world abound, suggesting aging itself is a malleable process. We present a novel multi-scale theoretical framework for entropic aging, and apply it to recently published DNA methylation data from 348 evolutionarily distant mammalian species. Our analysis identified modules or correlated DNA methylation changes associated with reversible pathway activation in key biological processes. We discovered a single species-dependent scaling factor controlling the magnitude of fluctuations across biological pathways. It acts as the organism's "effective temperature", quantifying intrinsic biological noise within networks and is unrelated to physical body temperature. Furthermore, we find a distinct stochastic damage signature and an associated extreme value (Gumbel) distribution of activation barriers controlling site-specific damage rates of individual CpG sites. This implies that aging is driven by rare, high-energy transitions on rugged energy landscape, most likely simultaneous and hence practically irreversible failures in highly redundant systems. While the overall rate of damage accumulation and hence the maximum lifespan does not depend on the effective temperature driving the noise in leading pathways, effective temperature does influence both initial mortality rate and the mortality rate doubling time -- thereby shaping the survival curve. Lowering effective temperature must, therefore, be a promising Geroscience strategy, aimed directly at squaring the curve of aging. The example shows that targeting the thermodynamic forces driving mammalian aging may provide powerful strategies for the development of truly meaningful interventions to combat aging in humans.

Abstract:Living systems are subject to the arrow of time; from birth, they undergo complex transformations (self-organization) in a constant battle for survival, but inevitably ageing and disease trap them to death. Can ageing be understood and eventually reversed? What tools can be employed to further our understanding of ageing? The present article is an invitation for biologists and clinicians to consider key conceptual ideas and computational tools (known to mathematicians and physicists), which potentially may help dissect some of the underlying processes of ageing and disease. Specifically, we first discuss how to classify and analyze complex systems, as well as highlight critical theoretical difficulties that make complex systems hard to study. Subsequently, we introduce Topological Data Analysis - a novel Big Data tool - which may help in the study of complex systems since it extracts knowledge from data in a holistic approach via topological considerations. These conceptual ideas and tools are discussed in a rather informal way to pave future discussions and collaborations between mathematicians and biologists studying ageing.

Abstract:We describe a 3D percolation-type approach to modeling of the processes of aging and certain other properties of tissues analyzed as systems consisting of interacting cells. Lattice sites are designated as regular (healthy) cells, senescent cells, or vacancies left by dead (apoptotic) cells. The system is then studied dynamically with the ongoing processes including regular cell dividing to fill vacant sites, healthy cells becoming senescent or dying, and senescent cells dying. Statistical-mechanics description can provide patterns of time dependence and snapshots of morphological system properties. The developed theoretical modeling approach is found not only to corroborate recent experimental findings that inhibition of senescence can lead to extended lifespan, but also to confirm that, unlike 2D, in 3D senescent cells can contribute to tissue's connectivity/mechanical stability. The latter effect occurs by senescent cells forming the second infinite cluster in the regime when the regular (healthy) cell's infinite cluster still exits.

Abstract:Although the evolutionary theories of aging are quite well established, our knowledge about how we age is still very limited. The abundance and heterogeneity of available mechanistic theories of aging implicitly suggest that this phenomenon is overly complex and unlikely to be explained by a single pathway. Moreover, although aging remains a unique process, it is characterized by heterogeneous manifestations, not only determining inter-individual variations, but even intra-individual diversities. Such heterogeneity renders the inner nature of the aging process of difficult evaluation in older persons due to the potential biases introduced by multiple age-related social, biological, and clinical factors (and responsible for the evidence-based issue in geriatrics). Moving from the difficulties in translating anti-aging preclinical interventions into clinical trials, an alternative approach is illustrated. We encourage moving to a holistic evaluation of aging by adopting specific and consequent modifications in the design and conduction of clinical research. Such approach is today commonly applied in the clinical setting where the complexity of older patients often requires multidimensional interventions to adequately target the geriatric syndromes. Consistently, interventions targeting the aging process may result ineffective if too focused on a single underlying causal mechanism and/or failing to capture the complexity of the phenomenon. In this context, frailty (a geriatric syndrome characterized by age-related declines occurring across multiple physiologic systems) may indeed represent a clinically relevant threshold throughout the continuum of the aging process and a promising benchmark to test multidomain interventions against age-related conditions.

Abstract:We report a case of sudden onset of respiratory distress caused by pulmonary edema due to laryngospasm. The diagnosis was established by the clinical context and chest X-ray. A CT-scan was performed to narrow down the differential diagnosis and to confirm the diagnosis. Postextubation pulmonary edema due to laryngospasm is a rare entity with a typical clinical and radiographic presentation.

Abstract:Aging can be defined as a process of progressive decline in the physiological capacity of an organism, manifested by accumulated alteration and destabilization at the whole system level. Systems biology approaches offer a promising new perspective to examine the old problem of aging. We begin this review by introducing the concepts of systems biology, and then illustrate the application of systems biology approaches to aging research, from gene expression profiling to network analysis. We then introduce the network that can be constructed using known lifespan and aging regulators, and conclude with a look forward to the future of systems biology in aging research. In summary, systems biology is not only a young field that may help us understand aging at a higher level, but also an important platform that can link different levels of knowledge on aging, moving us closer to a more comprehensive control of systematic decline during aging.

Abstract:Aging is one of the greatest challenges for biomedical research in the developed world. The continuing increase in lifespan without accompanying increases in health span has extreme economic and societal implications for an aging society. The majority of adult‐onset diseases such as cancer, cardiovascular disease, and Alzheimer's are a direct consequence of aging, and with changing demographics, more and more people are afflicted. If we knew how to slow down the negative effects of aging, we could delay or prevent all of these diseases at once. We therefore need to understand the underlying molecular mechanisms of aging and longevity so as to develop applications that ultimately improve health span.

Abstract:Although species longevity is subject to a diverse range of selective forces, the mortality curves of a wide variety of organisms are rather similar. We argue that aging and its universal characteristics may have evolved by means of a gradual increase in the systemic interdependence between a large collection of biochemical or mechanical components. Modeling the organism as a dependency network which we create using a constructive evolutionary process, we age it by allowing nodes to be broken or repaired according to a probabilistic algorithm that accounts for random failures/repairs and dependencies. Our simulations show that the network slowly accumulates damage and then catastrophically collapses. We use our simulations to fit experimental data for the time dependent mortality rates of a variety of multicellular organisms and even complex machines such as automobiles. Our study suggests that aging is an emergent finite-size effect in networks with dynamical dependencies and that the qualitative and quantitative features of aging are not sensitively dependent on the details of system structure.

Abstract:Widespread interest in non-destructive biomarkers of aging has led to a curse of plenty: a multitude of biological ages that each proffers a 'true' health-adjusted age of an individual. While each measure provides salient information on the aging process, they are each univariate, in contrast to the "hallmark" and "pillar" theories of aging which are explicitly multidimensional, multicausal and multiscale. Fortunately, multiple biological ages can be systematically combined into a multidimensional network representation. The interaction network between these biological ages permits analysis of the multidimensional effects of aging, as well as quantification of causal influences during both natural aging and, potentially, after anti-aging intervention. The behaviour of the system as a whole can then be explored using dynamical network stability analysis which identifies new, efficient biomarkers that quantify long term resilience scores on the timescale between measurements (years). We demonstrate this approach using a set of 8 biological ages from the longitudinal Swedish Adoption/Twin Study of Aging (SATSA). After extracting an interaction network between these biological ages, we observed that physiological age, a proxy for cardiometabolic health, serves as a central node in the network, implicating it as a key vulnerability for slow, age-related decline. We furthermore show that while the system as a whole is stable, there is a weakly stable direction along which recovery is slow - on the timescale of a human lifespan. This slow direction provides an aging biomarker which correlates strongly with chronological age and predicts longitudinal decline in health - suggesting that it estimates an important driver of age-related changes.
