Abstract:Background. Electromagnetic fields (EMEs) used in stem-cell tissue engineering can help elucidate their biological principles. Objective. The aim of this study was to investigate the effects of low-intensity EMFs on cell proliferation, differentiation, and cycle in mouse bone marrow stromal cells (BMSCs) and the in vivo effects of EMFs on BMSC. Methods. Harvested BMSCs were cultured for 3 generations and divided into 4 groups. The methylthiotetrazole (MTT) assay was used to evaluate cell proliferation, and alkaline phosphatase activity was measured via a colorimetric assay on the 3rd, 7th, and 10th days. Changes in cell cycle also were analyzed on the 7th day, and bone nodule formation was analyzed on the 12th day. Additionally, the expression of the collagen I gene was examined by reverse transcription-polymerase chain reaction (RT-PCR) on the 10th day. The BMSCs of the irradiated group and the control group were transplanted into cortical bone of different mice femurs separately, with poly(lactic-co-glycolic acid) (PLGA) serving as a scaffold. After 4 and 8 weeks, bone the bone specimens of mice were sliced and stained by hematoxylin and eosin separately. Results. The results showed that EMFs (0.5 mT, 50 Hz) accelerated cellular proliferation, enhanced cellular differentiation, and increased the percentage of cells in the G(2)/M+S (postsynthetic gap 2 period/mitotic phase + S phase) of the stimulation. The EMF-exposed groups had significantly higher collagen I messenger RNA levels than the control group. The EMF + osteogenic medium-treated group readily formed bone nodules. Hematoxylin and eosin staining showed a clear flaking of bone tissue in the irradiated group. Conclusion. Irradiation of BMSCs with low-intensity EMEs (0.5 mT, 50 Hz) increased cell proliferation and induced cell differentiation. The results of this study did not establish a stricter animal model for studying osteogenesis, and only short-term results were investigated. Further study of the mechanism of EMF is needed.
Thyroxine Differentially Modulates the Peripheral Clock: Lessons from the Human Hair Follicle

Abstract:The human hair follicle (HF) exhibits peripheral clock activity, with knock-down of clock genes (BMAL1 and PER1) prolonging active hair growth (anagen) and increasing pigmentation. Similarly, thyroid hormones prolong anagen and stimulate pigmentation in cultured human HFs. In addition they are recognized as key regulators of the central clock that controls circadian rhythmicity. Therefore, we asked whether thyroxine (T4) also influences peripheral clock activity in the human HF. Over 24 hours we found a significant reduction in protein levels of BMAL1 and PER1, with their transcript levels also decreasing significantly. Furthermore, while all clock genes maintained their rhythmicity in both the control and T4 treated HFs, there was a significant reduction in the amplitude of BMAL1 and PER1 in T4 (100 nM) treated HFs. Accompanying this, cell-cycle progression marker Cyclin D1 was also assessed appearing to show an induced circadian rhythmicity by T4 however, this was not significant. Contrary to short term cultures, after 6 days, transcript and/or protein levels of all core clock genes (BMAL1, PER1, clock, CRY1, CRY2) were up-regulated in T4 treated HFs. BMAL1 and PER1 mRNA was also up-regulated in the HF bulge, the location of HF epithelial stem cells. Together this provides the first direct evidence that T4 modulates the expression of the peripheral molecular clock. Thus, patients with thyroid dysfunction may also show a disordered peripheral clock, which raises the possibility that short term, pulsatile treatment with T4 might permit one to modulate circadian activity in peripheral tissues as a target to treat clock-related disease.

Abstract:OBJECTIVETo study the effects of GSM 1800 MHz radiofrequency electromagnetic fields (RF EMF) on DNA damage in Chinese hamster lung (CHL) cells.METHODSThe cells were intermittently exposed or sham-exposed to GSM 1800 MHz RF EMF (5 minutes on/10 minutes off) at a special absorption rate (SAR) of 3.0 W/kg for 1 hour or 24 hours. Meanwhile, cells exposed to 2-acetylaminofluorene, a DNA damage agent, at a final concentration of 20 mg/L for 2 hours were used as positive control. After exposure, cells were fixed by using 4% paraformaldehyde and processed for phosphorylated form of H2AX (gammaH2AX) immunofluorescence measurement. The primary antibody used for immunofluorescence was mouse monoclonal antibody against gammaH2AX and the secondary antibody was fluorescein isothiocyanate (FITC)-conjugated goat anti-mouse IgG. Nuclei were counterstained with 4, 6-diamidino-2-phenylindole (DAPI). The gammaH2AX foci and nuclei were visualized with an Olympus AX70 fluorescent microscope. Image Pro-Plus software was used to count the gammaH2AX foci in each cell. For each exposure condition, at least 50 cells were selected to detect gammaH2AX foci. Cells were classified as positive when more than five foci were detected. The percentage of gammaH2AX foci positive cells was adopted as the index of DNA damage.RESULTSThe percentage of gammaH2AX foci positive cell of 1800 MHz RF EMF exposure for 24 hours (37.9 +/- 8.6)% or 2-acetylaminofluorene exposure (50.9 +/- 9.4)% was significantly higher compared with the sham-exposure (28.0 +/- 8.4)%. However, there was no significant difference between the sham-exposure and RF EMF exposure for 1 hour (31.8 +/- 8.7)%.CONCLUSION1800 MHz RF EMF (SAR, 3.0 W/kg) for 24 hours might induce DNA damage in CHL cells.

Abstract:Magnetic resonance image (MRI) systems with a much higher magnetic flux density were developed and applied for potential use in medical diagnostic. Recently, much attention has been paid to the biological effects of static, strong magnetic fields (SMF). With the 13 T SMF facility in the Institute of Plasma Physics, Chinese Academy of Sciences, the present study focused on the cellular effects of the SMF with 13 T on the cell viability and the cell cycle distribution in immortalized hamster cells, such as human-hamster hybrid (AL) cells, Chinese hamster ovary (CHO) cells, DNA double-strand break repair deficient mutant (XRS-5) cells, and human primary skin fibroblasts (AG1522) cells. It was found that the exposure of 13 T SMF had less effect on the colony formation in either nonsynchronized or synchronized AL cells. Moreover, as compared to non-exposed groups, there were slight differences in the cell cycle distribution no matter in either synchronized or nonsynchronized immortalized hamster cells after exposure to 13 T SMF. However, it should be noted that the percentage of exposed AG1522 cells at G0/G1 phase was decreased by 10% as compared to the controls. Our data indicated that although 13 T SMF had minimal effects in immortalized hamster cells, the cell cycle distribution was slightly modified by SMF in human primary fibroblasts.

Abstract:Objectives: To evaluate chromosomal damage and cytotoxicity in exfoliated buccal mucosa cells, cells of the tongue and epithelial gingival cells from adults following CBCT scan and to compare the sensitivity of the different exfoliated cells to a same dosage of ionizing radiation. Methods: The study included 46 healthy participants (median age 27 years; age range 23-42 years) who had a CBCT scan. Exfoliated mucosa cells were collected immediately before the CBCT scan and 10 days after. Cells were centrifuged, fixed in the fluid of methanol : glacial acetic acid (3 : 1) and stained using the method of Schiff's reagent and fast green. One observer analyzed all the slides. For interobserver variances, a second observer scored 16 slides chosen from all the subjects. The same set of 16 slides were analyzed once again a month later for intraobserver variances. Results: There is no significant differences for micronucleated cells before and after a CBCT scan in exfoliated buccal mucosa cells (p = 0.476), cells of the tongue (p = 0.884) and epithelial gingival cells (p = 0.362). The frequencies of pyknosis cell and karyolysis cell had significantly increased after CBCT scan in the three groups. No significant difference was found among the three kinds of mucosa cells (p = 0.557). The interobserver (p = 0.624) and intraobserver (p = 0.193) variances were not significant. Conclusions: A CBCT scan may induce cytotoxicity but not chromosomal damage in the oral mucosa cells, including buccal mucosa cells, cells of the tongue or epithelial gingival cells. The sensitivity of the different exfoliated cells to the same dosage of radiation had no statistically significant difference.
Decreasing of the Mitotic Checkpoint Function of Tramsformed CHL Cells

Abstract:Objective To observe the chromosome instability (ClN), mitotic checkpoint function and the resisance to the colchicine cytotoxicity of transformed Chinese hamster lung cells (CHL). Methods Karyotype analysis was used to evaluate the chromosome stability ofthe transformed CHL cells induced by MNNG and the control CHL cells, The ratio of cells arresting at metaphase induced by colchicimes was determimed to evaluate the mitotic checkpoint function;MTT method was used to test the cytotoxicity of colchicimes. Results At the 3rd week after treating CHL cells with 2 μg/ml MNNG for 24 hours, many transformation foci appeared, and the cloning effciency of transformed cells (54.2%)was higher than that of the control cells (1%)in soft agar.These results showed that the treated cell had been transformed malignantly, Karyotype analysis showed that the number of chromosome in the transformed CHL cells varied from 14 to 48, the cells with 25chromosomes only accoumt for 7%, while 81%in the control CHL cells. These indicated that the transformed CHL cells appeared ClN, After the cells were treated with 0.5 μg/ml colchicines for different time, the ratios of the transformed cells arresting at metaphase were lower than those of the comtrol cells, in addition, after the cells were treated with different doses of colchicine for 24 hours, the transformed cells appeared to be resistant to the cytotoxicity of colchicine. The results suggested that the function of mitotic checkpoint decreased. Conclusion The decreasing of mitotic checkpoint function could play an important role during cell transformation.

Abstract:The chiasmatic association of the heteromorphic sex chromosomes in the spermatocytes of the Chinese hamster was observed in squash and/or air-dried preparations. The pairing arm of the Y was invariably its short arm. Although the X in diakinesis did not show distinct long and short arm as in mitotic metaphase, the DNA replication patterns of the sex chromosomes in spermatogonia suggested that the distal segment of the long arm of the X is homologous to the short arm of the Y.

Abstract:To explore whether synchrotron radiation hard X-ray has any effects on biological tissue structure and re- lated antigen expression of fixed bone, cartilage, liver and lung, diffraction-enhanced X-ray imaging (DEI) and in-line holography with synchrotron radiation hard X-ray from Beijing Synchrotron Radiation Facility (BSRF) were applied to observe the morphology of the bone defect of rabbit, the lung cancer of mouse, the hepatic fibrosis and hepatocir- rhosis of SD rat as well as their respective control. The histologic changes were observed by microscope. The semi- quantitative analysis of antigen-III, S-100, FN and ubiquitin were also performed by immunohistochemistry. Imaged tissue was found to be intact, and there was no difference in these antigen expressions between the imaged and control groups. It is concluded that synchrotron radiation hard X-ray has no effect on biological tissue structure and related antigen expression of fixed bone, cartilage, liver and lung. The present results have provided experimental evidences for the development of synchrotron hard X-ray-phase technique of in vitro biological and medical studies.
Evaluation of the Cytotoxic and Genotoxic Effects by Melamine and Cyanuric Acid Co-Exposure in Human Embryonic Kidney 293 Cells

Abstract:The melamine and cyanuric acid (CA) complex has been suggested to cause the toxic effects observed in melamine-contaminated food or milk. However, the cytotoxic and genotoxic effects of co-exposure to melamine and CA are not fully clear. Therefore, the cytotoxic effects of melamine and CA were first examined by co‐exposure in human kidney 293 cells using the MTT assay. During a 24-h period for the three concentrations tested (0.5, 1, and 5 mg/mL), neither melamine nor CA alone showed significant toxic effects on 293 cells at 0.5 mg/mL, while higher concentrations led to decreased in cell viability. However, co-exposure to several combinations of melamine and CA [100:1, 10:1, 1:10, and 1:100 (v:v), at a final concentration of 0.5 mg/mL] did cause cytotoxicity with higher levels of CA leading to higher cytotoxicity. By contrast, while neither melamine nor CA alone induced phosphorylated-H2AX (γH2AX) foci formation, melamine and CA at a 100:1 ratio induced γH2AX foci 24 h post-treatment. The alkaline comet assay also revealed the presence of DNA damage following melamine and CA co-exposure. In vivo assay also revealed the presence of melamine-CA complex in the kidney. These data indicated that the cytotoxic and genotoxic effects of melamine and CA co-exposure differ from those of melamine or CA alone.

Abstract:This study aims to investigate the cellular effects of radiofrequency exposure, 1950 MHz, long-term evolution (LTE) signal, administered alone and in combination with mitomycin-C (MMC), a well-known cytotoxic agent. Chinese hamster lung fibroblast (V79) cells were exposed/sham exposed in a waveguide-based system under strictly controlled conditions of both electromagnetic and environmental parameters, at specific absorption rate (SAR) of 0.3 and 1.25 W/kg. Chromosomal damage (micronuclei formation), oxidative stress (reactive oxygen species [ROS] formation), and cell cycle progression were analyzed after exposure and coexposure. No differences between exposed samples and sham-controls were detected following radiofrequency exposure alone, for all the experimental conditions tested and biological endpoints investigated. When radiofrequency exposure was followed by MMC treatment, 3 h pre-exposure did not modify MMC-induced micronuclei. Pre-exposure of 20 h at 0.3 W/kg did not modify the number of micronuclei induced by MMC, while 1.25 W/kg resulted in a significant reduction of MMC-induced damage. Absence of effects was also detected when CW was used, at both SAR levels. MMC-induced ROS formation resulted significantly decreased at both SAR levels investigated, while cell proliferation and cell cycle progression were not affected by coexposures. The results here reported provide no evidence of direct effects of 1950 MHz, LTE signal. Moreover, they further support our previous findings on the capability of radiofrequency pre-exposure to induce protection from a subsequent toxic treatment, and the key role of the modulated signals and the experimental conditions adopted in eliciting the effect.

Abstract:Objective:To investigate the effect of Zinc Gluconate and Manganous cholride which were used solely and together on DNA damage induced by silica dioxide in Chinese hamster lung cells.Methods:Fibroblast cells were cultured with silica,at the same time Zinc Gluconate and Manganous Chloride which were used solely and together were added respectively.DNA damage were measured through comet assay.Results:Zinc Gluconate,Manganous Chloride and Zinc Gluconate together with Manganous chloride have the effects to lighten DNA damage induced by silicon dioxide in Chinese hamster lung cells.The most suitable combination of the two compounds should be 1.0 μg/ml of Zinc Gluconate with 0.5 μg/ml of Manganous Chloride.Conclusion:Zinc Gluconate and Manganous Chloride which were used solely and together could reduce DNA damage induced by silicon dioxide in Chinese hamster lung cells.

Abstract:Abstract This study aimed to investigate the impact of chronic low-level exposure to chemical carcinogens with different modes of action on the cellular response to ionising radiation. Human lymphoblastoid GM1899A cells were cultured in the presence of 4-nitroquinoline N-oxide (4NQO), N -nitroso- N -methylurea (MNU) and hydrogen peroxide (H 2 O 2 ) for up to 6 months at the highest non-(geno)toxic concentration identified in pilot experiments. Acute challenge doses of 1 Gy X-rays were given and chromosome damage (dicentrics, acentric fragments, micronuclei, chromatid gaps/breaks) was scored. Chronic exposure to 20 ng/ml 4NQO, 0.25 μg/ml MNU or 10 μM H 2 O 2 hardly induced dicentrics and did not significantly alter the yield of X-ray-induced dicentrics. Significant levels of acentric fragments were induced by all chemicals, which did not change during long-term exposure. Fragment data in combined treatment samples compared to single treatments were consistent with an additive effect of chemical and radiation exposure. Low level exposure to 4NQO induced micronuclei, the yields of which did not change throughout the 6 month exposure period. As for fragments, micronuclei yields for combined treatments were consistent with an additive effect of chemical and radiation. These results suggest that cellular radiation responses are not affected by long-term low-level chemical exposure.

Abstract:The purpose of this study was to determine the influence of iodinated contrast agents on the formation of DNA double-strand breaks in vitro in lymphocytes and to verify these results in patients undergoing diagnostic computed tomography examinations. Blood samples were irradiated in vitro in the presence of iodinated X-ray contrast agent. Controls were irradiated without contrast agent. Fourteen patients were investigated using contrast-enhanced computed tomography (CT), and 14 other patients with unenhanced CT. Blood samples were taken prior to and 5 min and 1, 2 and 24 h after the CT examination. In these blood samples the average number of γH2Ax-foci per lymphocyte was enumerated by fluorescence microscopy. Statistical differences between foci numbers developed in the presence and absence of contrast agent were tested using an independent sample t-test. In vitro foci numbers after irradiation were significantly higher when contrast agent was present during irradiation. In vivo, γH2Ax-foci levels were 58% higher in patients undergoing contrast-enhanced CT compared with those undergoing unenhanced CT. In the presence of iodinated contrast agents DNA, damage is increased and the radiation dose is not the only factor affecting the amount of DNA damage. Individual patient characteristics and biological dosimetry applications, e.g. the analysis of γH2Ax-foci, have to be considered.

Abstract:ABSTRACT Objective: To explore if low dose ionizing radiation could induce crossadaptive response to cytogenetic damage in embryonic cells caused by subsequent cadmium chloride. Methods: The mice were wholebody irradiated with60Co γrays 1,2,4 and 8 cGy respectively on gestation d 9. 4 h after, the mice were injected CdCl2 solution according to Cd 4 mg/kg. Chromosome aberrations of embryonic cells of gestation d 10 were analysed. Results: 1,2,4 and 8 cGy60Co γrays induced resistance to cytogenetic damage of embryonic cells caused by subsequent Cd 4 mg/kg body weight. Conclusion: The results first demonstrated that low dose ionizing radiation can induce crossadaptive response to cytogenetic damage of embryonic cells caused by subsequent cadmium.

Abstract:PURPOSE:The purpose of this study is to link both numeric and structural chromosomal aberrations to the effectiveness of radiotherapy in chemotherapy refractory tumor cells. MATERIALS AND METHODS:Neuroblastoma (LAN-1) and 79HF6 glioblastoma cells derived from patients and their chemoresistant sublines were artificially cultured as neurospheres and irradiated by X-rays and heavy ions sources. All the cell lines were irradiated by Carbon-SIS with LET of 100 keV/μm. However, 79HF6 cells and LAN-1 cells were also irradiated by Carbon-UNILAC with LET of 168 keV/μm and Nickel ions with LET of 174 keV/μm, respectively. The effect of radiation on the survival and proliferation of cells was addressed by standard clonogenic assays. In order to analyze cell karyotype standard Giemsa staining, multicolor fluorescence in situ hybridization (mFISH) and multicolor banding (mBAND) techniques were applied. RESULTS:Relative biological effectiveness values of heavy ion beams relative to X-rays at the D10 values were found between 2.3 and 2.6 with Carbon-SIS and Nickel for LAN-1 and between 2.5 and 3.4 with Carbon-SIS and Carbon-UNILAC for 79HF6 cells. Chemorefractory LAN-1(RETO) cells were found more radioresistant than untreated LAN-1(WT) cells. 79HF6(RETO) glioblastoma cells were found more radiosensitive than cytostatic sensitive cells 79HF6(WT). Sphere formation assay showed that LAN-1(RETO) cells were able to form spheres in serum-free culture, whereas 79HF6 cells could not. Most of 79HF6(WT) cells revealed a number of 71-90 chromosomes, whereas 79HF6(RETO) revealed a number of 52-83 chromosomes. The majority of LAN-1(WT) cells revealed a number of 40-44 chromosomes. mFISH analysis showed some stable aberrations, especially on chromosome 10 as judged by the impossibility to label this region with specific probes. This was corroborated using mBAND analysis. CONCLUSION:Heavy ion irradiation was more effective than X-ray in both cytostatic naive cancer and chemoresistant cell lines. LAN-1(RETO) chemoresistant neuroblastoma cells were found to be more radioresistant than the cytostatic naive cells (LAN-1(WT)), whereas this effect was not found in 79HF6 cells.

Abstract:This study evaluates the magnitude and the effects of scatter of low kV X-ray in cone-beam micro-CT(micro-computed tomography) system for small animals.The magnitude of scatter is based on scatter-to-primary ratio(SPR) calculated using the beam stop method and estimated by polynormail-fitting extrapolation to 0 mm size stoppers.Lead foils with various slit widths were used to change the cone angle for the micro-CT imaging and its effect of the X-ray scattering.The standard phantom consisted of a uniform resin(40 mm diameter) representing a small mouse.The SPR at the center of the cone-beam system is 4.78% at the cone angle of 0.247° and is 27.58% at the cone angle of 5.67°.The effects of X-ray scattering on the reconstructed image uniformity and contrast ratio are also evaluated.

Abstract:Objective To discuss the effect of Crocus sativus L. cultured cell on peripheral blood index of irradiated mice. Methods 100 Kunming male mice were randomly divided into 5 groups,in addition to the blank control group,four groups were given 2. 5 Gy 60Coγ ray irradiation,including model control group and three treatment groups which delivered high,medium and low dose Crocus sativus L. cultured cell after irradiation. The number of white blood cells(WBC),red blood cells(RBC),lymphocyte(LYM),platelets (PLT),the content of hemoglobin(HGB) and red blood cells related indicators in peripheral blood were examined regularly to investi-gate the Crocus sativus L. cultured cell protective effect on irradiated mice. Results The number of WBC,LYM,RBC,HGB,PLT and the content of HMG in mice peripheral blood were decreased after 2. 5 Gy 60Coγ ray radiation. By oral delivery of Crocus sativus L. cultured cell,descent rate of WBC,RBC,HGB,LYM and PLT in mice peripheral blood were significantly delayed. Conclusion Crocus sativus L. cultured cell has protective effect on peripheral blood cells of irradiated mice.
