Abstract:Multiplexed quantification of low-abundance protein biomarkers in complex biofluids is important for biomedical research and clinical diagnostics. However, in situ sampling without perturbing biological systems remains challenging. In this work, we report a buoyant biosensor that enables in situ monitoring of protein analytes at attomolar concentrations with a 15 min temporal resolution. The buoyant biosensor implemented with fluorescent nanolabels enabled the ultrasensitive and multiplexed detection and quantification of cytokines. Implementing the biosensor in a digital manner (i.e., counting the individual nanolabels) further improves the low detection limit. We demonstrate that the biosensor enables the detection and quantification of the time-varying concentrations of cytokines (e.g., IL-6 and TNF-α) in macrophage culture media without perturbing the live cells. The easy-to-apply biosensor with attomolar sensitivity and multiplexing capability can enable an in situ analysis of protein biomarkers in various biofluids and tissues to aid in understanding biological processes and diagnosing and treating diverse diseases.

Abstract:This study introduces an advanced bioanalytical platform that combines digital microfluidics (DMF) with Raman spectroscopy, effectively addressing common issues in bioanalysis such as sample contamination, excessive consumption of samples and reagents, and manual handling. Our innovative system is engineered to handle diverse sample types and enables both sample preparation and in-situ analysis on a single device, utilizing less than 5μL of samples and reagents. It incorporates a Translucent Raman Enhancement Stack (TRES) sensor, which boosts the detection signal, and includes droplet-driving functionality for automated processing of complex samples in a compact setting. The hydrophilic surface of the TRES sensor draws analytes into the detection zone, expediting sample attachment and enhancing the surface-enhanced Raman scattering (SERS). Moreover, the TRES sensors are cost-effective, straightforward to manufacture, and scalable, making them suitable for widespread production and single-use bioanalytical applications. This integrated and automated approach streamlines the sample preparation and analysis processes, enhances detection efficiency, and achieves high sensitivity, excellent linearity, and effective detection of different biochemical analytes. We validate the system's accuracy through on-device enrichment and analysis of exosomes from serum, demonstrating its capability for real-time, on-site analysis of complex biological samples.

Abstract:Functional assays on intact tumor biopsies can complement genomics-based approaches for precision oncology, drug testing, and organs-on-chips cancer disease models by capturing key therapeutic response determinants, such as tissue architecture, tumor heterogeneity, and the tumor microenvironment. Most of these assays rely on fluorescent labeling, a semiquantitative method best suited for single-time-point assays or labor-intensive immunostaining analysis. Here, we report integrated aptamer electrochemical sensors for on-chip, real-time monitoring of cytochrome C, a cell death indicator, from intact microdissected tissues with high affinity and specificity. The platform features a multi-well sensor layout and a multiplexed electronic setup. The aptasensors measure increases in cytochrome C in the supernatant of mouse or human microdissected tumors after exposure to various drug treatments. Because of the sensor's high affinity, it primarily tracks rising concentrations of cytochrome C, capturing dynamic changes during apoptosis. This approach could help develop more advanced cancer disease models and apply to other complex in vitro disease models, such as organs-on-chips and organoids.
Multi-Channel Cell Co-Culture for Drug Development Based on Glass Microfluidic Chip-Mass Spectrometry Coupled Platform

Abstract:RATIONALE: Cell-based drug assay plays an essential role in drug development. By coupling a microfluidic chip with mass spectrometry (MS), we developed a multifunctional platform. Cell co-culture, cell apoptosis assay, fluorescence and MS detection of intracellular drug absorption could be simultaneously conducted on this platform.METHODS: Three micro-channels were fabricated through photolithography technology to conduct the cell co-culture. Cell apoptosis after drug treatment was assayed by fluorescent probes (Hoechst 33342). Intracellular Dox absorption was analyzed by confocal fluorescent microscopy. With a high voltage (similar to 4.5 kV) applied onto the microfluidic chip, the ionization spray was successfully generated by dropping isopropanol onto it. By coupling with a Shimadzu LCMS-2010 A mass spectrometer, intracellular CPA absorption was detected on the microfluidic chip.RESULTS: The microfluidic chip-MS coupled platform showed high biocompatibility. Distinction of cell apoptosis between co-cultured and mono-cultured cells was detected. The results of intracellular drug absorption well explained the different cell apoptosis rate.CONCLUSIONS: Cell-based drug assay was facilely and successfully conducted on the microfluidic chip-MS coupled platform. Abstract:Protein is a complex chemical substance essential for human survival. Traditional protein detection methods, such as colorimetry, electrochemical analysis, and enzyme-linked immunosorbent assays, have shown good specificity and accuracy for the protein detection. However, all these methods require specialized instruments, and the detection procedures are laborious and time-consuming. As a result, a rapid, sensitive, label-free protein detection method is urgently needed. Herein, we have developed an ultra-sensitive biosensor for the detection of low-concentration protein molecules, employing liquid crystal (LC)-amplified optofluidic resonator. Since the orientations of LCs highly depend on the surface biomolecular binding processes, LCs can be employed to realize the extremely sensitive detection of biomolecules. Immobilized protein molecules interfere with the orientation of LCs by reducing the vertical anchoring force from the alignment layer in which the spectral wavelength shift was monitored as a sensing parameter. A biosensing platform based on an LC-amplified optofluidic whispering gallery mode (WGM) resonator was designed and studied accordingly. Due to the simultaneous interaction of the WGM and the LCs in the optofluidic resonator, the changes caused by the injection of protein molecules will be amplified, resulting in a shift in the resonance wavelength. Total wavelength shifts scale proportionally to the molecular concentrations of the protein within a certain range. The detection limit for streptavidin (SA) can reach as low as the femtometer level, which is significantly higher than the detection limit in the classic polarized optical microscope (POM) method visible with the naked eye. In addition to SA, the LC-based optofluidic resonator can also be applied to detect a variety of protein molecules. Our study demonstrates that LC-amplified optofluidic resonator can provide a novel solution for ultrasensitive real-time characterization of biosensing and biomolecular interactions.

Abstract:Despite the promise of therapeutic antibodies in engaging the immune system to eliminate malignant cells, many aspects of the complex interplay between immune cells and cancer cells induced by antibody therapy remain incompletely understood. This study aimed to develop a biosensor system that can evaluate direct cell-cell physical contact and interactions between immune effector and target cells induced by therapeutic antibodies in physiologically relevant environments. The system uses two structural complementary luciferase units (SmBit and LgBit) expressed on the respective membranes of effector and target cells. Upon cell-cell contact, the two subunits form active NanoLuc, generating a luminescent signal, allowing for real-time monitoring of cell-cell interactions and quantitatively assessing the pharmacological effects of therapeutic antibodies. We optimized the system to ensure selectivity by adjusting the spacer lengths between two luciferase units to minimize interference from nonspecific intercellular contact. The system was applied to quantitatively monitor cell-cell interactions between NK and target cells induced by rituximab and between T and target cells induced by blinatumomab in a 3D cell culture system. The biosensor system has the potential to characterize antibody pharmacology through a deeper understanding of antibody-mediated cell-cell interactions.

Abstract:The slow light sensor techniques have been applied to bio-related detection in the past decades. However, similar testing-systems are too large to carry to a remote area for diagnosis or point-of-care testing. This study demonstrated a fully automatic portable biosensing system based on the microring resonator. An optical-fiber array mounted on a controller based micro-positioning system, which can be interfaced with MATLAB to locate a tentative position for light source and waveguide coupling alignment. Chip adapter and microfluidic channel could be packaged as a product such that it is cheap to be manufactured and can be disposed of after every test conducted. Thus, the platform can be more easily operated via an ordinary user without expertise in photonics. It is designed based on conventional optical communication wavelength range. The C-band superluminescent-light-emitting-diode light source couples in/out the microring sensor to obtain quasi-TE mode by grating coupler techniques. For keeping a stable chemical binding reaction, the cost-effective microfluidic pump was developed to offer a specific flow rate of 20 μL/min by using a servo-motor, an Arduino board, and a motor driver. The subwavelength grating metamaterial ring resonator shows highly sensitive sensing performance via surface index changes due to biomarker adhered on the sensor. The real-time peak-shift monitoring shows 10 μg/mL streptavidin detection of limit based on the biotin-streptavidin binding reaction. Through the different specific receptors immobilized on the sensor surface, the system can be utilized on the open applications such as heavy metal detection, gas sensing, virus examination, and cancer marker diagnosis.

Abstract:The binding fluorescence dye by supramolecular interactions to form optical chemosensors is a powerful sensing method allowing ultra-high selectivity for the target analyte and fast response of the detection. While notable success has been made to develop various supramolecular optical chemosensors, there is little effects contributed to the high throughput assays. Here we introduce a droplet-based microfluidic detection and screening system using supramolecular optical chemosensors to measure the enzyme activity and elucidate their prospects for high throughput screening. The supramolecular optical chemosensors derives from cucurbit[8]uril (CB[8])-mediated host-guest chemistry for macrocycle-dye complex formation. The encapsulation of CB[8]-dye in millions of picolitre microdroplets provides an efficient platform for handing a large number of parallel experiments, allowing for shorter processing time and reducing the needs for analyte volume. Further, the supramolecular optical chemosensors in microdroplets are used in measuring of the biocatalysis efficiency of enzyme immobilized hydrogel microparticles (HMPs), which also has been applied for high throughput screening. This label-free and robust approach of supramolecular optical chemosensors in microdroplets platform could be transferable to single-cell analysis and rapid selection of cells.

Abstract:A microfluidic-based surface-enhanced Raman scattering (SERS) platform for analyzing cytokines secreted by single cells is reported based on the elaborate bioconjugation of the immuno-sandwich complex on the probed cell surface. This platform integrates the dual functions of microfluidic droplet separation of single cells and SERS measurement. Two immune nanoprobes (capture probe and SERS probe) are introduced into a microfluidic droplet along with a single cell. They were anchored to the cell membrane protein surface by capturing secreted cytokines to form an immune sandwich structure, realizing the enrichment effect of cytokines above the cell membrane surface and the amplification effect of SERS detection probes. This single-cell analytical platform was applied to track specific cell-secreted vascular endothelial growth factor (VEGF) of different cell lines (MCF-7, SGC, and T24), and highly sensitive detection of VEGF was achieved. Chemometric methods (principal component analysis and <i>t</i>-distributed stochastic neighbor embedding) were adopted for the SERS data analysis, and the support vector machine (SVM) discriminant model was established to test the data. These chemometric methods successfully identify significant differences in the secreting ability of cytokines among three kinds of cancer cell lines, revealing cell heterogeneity. In addition, the behavior of single cells secreting VEGF was monitored time-dependently and was shown to increase with time. This work demonstrates the importance of tracking specific cells secreting cytokines based on the cell surface bioconjugation strategy. Our developed platform provides guidelines for using the single-cell exocytosis factors as biomarkers to assess the early diagnosis of cancer and provide physiological cues for learning single-cell secretions.

Abstract:Developing biochemical and cell biological assay for screening biomolecules, evaluating their characteristics in biological processes, and determining their pharmacological effects represents a key technology in biomedical research. A PDMS-based integrated microfluidic platform was fabricated and tested for facilitating the labeling of ligand on the nanogram scale and sequential cell binding analysis in a manner that saves both time and reagents. Within this microfluidic platform, ligand labeling, cell immobolization, and optical analysis are performed in a miniaturized, continuous and semi-automated manner. This microfluidic device for ligand labeling and cell analysis is composed of two functional modules: (i) a circular reaction loop for fluorophore-labeling of the ligand and (ii) four parallel-oriented incubation chambers for immobilization of cells, binding of ligand to different cell populations, and optical evaluation of interactions between the labeled ligand and its cell targets. Epidermal growth factor (EGF) as the ligand and different cell lines with various levels of EGF receptor expression have been utilized to test the feasiblity of this microfluidic platform. When compared to studies with traditional Petri dish handling of cells and tissues, or even microwell analyses, experiments with the microfluidic platform described here are much less time consuming, conserve reagents, and are programmable, which makes these platforms a very promising new tool for biological studies.

Abstract:Dysregulations in metabolic and physiological cellular processes are generally accompanied by changes in concentration profiles of multiple biomarkers. Measuring dynamic changes in concentrations of several affected biomarkers can enhance our comprehension of the underlying disease pathophysiology and/or mechanisms of cytotoxicity. Most analytical detection techniques require samples to be obtained at pre-defined times for offline analysis. However, the inadequate temporal resolution of these methods can lead to key biological events that occur at shorter timescales, being overlooked. Electrochemical biosensors are relatively simple, cost-effective detection techniques that have been used extensively for continuous analyte monitoring, and more recently, for sensing multiple analytes. We describe the design and functionality of a novel electrochemical detection platform, comprising a screen-printed Pt electrode array, a microfluidic chip, and an automated pump and valve system, capable of quantitatively detecting up to eight analytes. As a proof of concept, we show the simultaneous, sensitive, and selective amperometric detection of four essential biomarkers- glucose, lactate, glutamate, and acetylcholine, in real time. We also demonstrate the biological application of this platform by continuously monitoring exocytotic glutamate release from human induced pluripotent stem cell-derived neuronal cultures over time. Excessive accumulation of glutamate in the synaptic cleft leads to prolonged stimulation of glutamate receptors on neurons, triggering a cascade of neurotoxic events that ultimately results in loss of neuronal function and cell death. The results suggest that our multianalyte electrochemical sensor platform can be coupled with more complex biological systems such as in vitro organotypic models, for the accurate monitoring of biomarker changes during adverse events like acute neurotoxicity and microbial infection.

Abstract:A novel microfluidic platform integrated with a flexible PDMS-based electrochemical cytosensor was developed for real-time monitoring of the proliferation and apoptosis of HeLa cells. The PDMS-gold film, which had a conductive smooth surface and was semi-transparent, facilitated electrochemical measurements and optical microscope observations. We observed distinct increases and decreases in peak current intensity, corresponding to cell proliferation in culture medium and apoptosis in the presence of an anticancer drug, respectively. This electrochemical analysis method permitted real-time, label-free monitoring of cell behavior, and the electrochemical results were confirmed with optical microscopy. The flexible microfluidic electrochemical platform presented here is suitable for on-site monitoring of cell behavior in microenvironments.

Abstract:By enhancing the speed, accuracy, and sensitivity, it opens up a new possibility for direct analysis of biologically relevant entities such as nucleic acids and proteins in single cell level. Nanofluidic systems with critical dimensions which were comparable to the molecular scales can provide the aforementioned performance, offering a novel basis for ultrasensitive and high-resolution bio-detections and medical diagnostics. Inspired by this concept, a nanofluidic system, integrated with a novel multi-nanochannel filter, an electrokinetic nano-preconcentrator, and a nanofluidic DAEKF detector for the detection of single cell releasing, is developed. A diluted solution of culture medium from PC-12 cells excited with excess nicotine concentration was detected with near single cell level amounts (c.a. 3000-10000 molecules) in this system after all processing in 20 min. Results showed that this multi-depth micro/nanofluidic chip exhibits superior sensitivity, efficiency, reliability, and reproducibility than those of conventional microchip electrophoresis /electrochromatography systems.
A digital microfluidic device integrated with electrochemical sensor and 3D matrix for detecting soluble PD-L1

Abstract:PD1/PD-L1 checkpoint inhibitors are at the forefront of cancer immunotherapies. However, the overall response rate remains only 10-30%. Even among initial responders, drug resistance often occurs, which can lead to prolonged use of a futile therapy in the race with the fatal disease. It would be ideal to closely monitor key indicators of patients' immune responsiveness, such as circulating PD-L1 levels. Traditional PD-L1 detection methods, such as ELISA, are limited in sensitivity and rely on core lab facilities, preventing their use for the regular monitoring. Electrochemical sensors exist as an attractive candidate for point-of-care tool, yet, streamlining multiple processes in a single platform remains a challenge. To overcome this challenge, this work integrated electrochemical sensor arrays into a digital microfluidic device to combine their distinct merits, so that soluble PD-L1 (sPD-L1) molecules can be rapidly detected in a programmed and automated manner. This new platform featured microscale electrochemical sensor arrays modified with electrically conductive 3D matrix, and can detect as low as 1 pg/mL sPD-L1 with high specificity. The sensors also have desired repeatability and can obtain reproducible results on different days. To demonstrate the functionality of the device to process more complex biofluids, we used the device to detect sPD-L1 molecules secreted by human breast cancer cell line in culture media directly and observed 2X increase in signal compared with control experiment. This novel platform holds promise for the close monitoring of sPD-L1 level in human physiological fluids to evaluate the efficacy of PD-1/PD-L1 immunotherapy.
Liquid crystal-amplified optofluidic biosensor for ultra-highly sensitive and stable protein assay

Abstract:Abstract Protein assays show great importance in medical research and disease diagnoses. Liquid crystals (LCs), as a branch of sensitive materials, offer promising applicability in the field of biosensing. Herein, we developed an ultrasensitive biosensor for the detection of low-concentration protein molecules, employing LC-amplified optofluidic resonators. In this design, the orientation of LCs was disturbed by immobilized protein molecules through the reduction of the vertical anchoring force from the alignment layer. A biosensing platform based on the whispering-gallery mode (WGM) from the LC-amplified optofluidic resonator was developed and explored, in which the spectral wavelength shift was monitored as the sensing parameter. The microbubble structure provided a stable and reliable WGM resonator with a high Q factor for LCs. It is demonstrated that the wall thickness of the microbubble played a key role in enhancing the sensitivity of the LC-amplified WGM microcavity. It is also found that protein molecules coated on the internal surface of microbubble led to their interactions with laser beams and the orientation transition of LCs. Both effects amplified the target information and triggered a sensitive wavelength shift in WGM spectra. A detection limit of 1 fM for bovine serum albumin (BSA) was achieved to demonstrate the high-sensitivity of our sensing platform in protein assays. Compared to the detection using a conventional polarized optical microscope (POM), the sensitivity was improved by seven orders of magnitude. Furthermore, multiple types of proteins and specific biosensing were also investigated to verify the potential of LC-amplified optofluidic resonators in the biomolecular detection. Our studies indicate that LC-amplified optofluidic resonators offer a new solution for the ultrasensitive real-time biosensing and the characterization of biomolecular interactions.

Abstract:Micro and nanoscale particles are crucial in various fields, from biomedical imaging to environmental processes. While conventional spectroscopy and microscopy methods for characterizing these particles often involve bulky equipment and complex sample preparation, optical micro-sensors have emerged as a promising alternative. However, their broad applicability is limited by the need for surface binding and difficulty in differentiating between sensing targets. This study introduces an optofluidic, high-throughput optical microresonator sensor that captures subtle acoustic signals generated by particles absorbing pulsed light energy. This novel approach enables real-time, label-free detection and interrogation of particles and cells in their native environments across an extended sensing volume. By leveraging unique optical absorption properties, our technique selectively detects and classifies flowing particles without surface binding, even in complex matrices like whole blood samples. We demonstrate the measurement of gold nanoparticles with diverse geometries and different species of red blood cells amidst other cellular elements and proteins. These particles are identified and classified based on their photoacoustic fingerprint, which captures shape, composition, and morphology features. This work opens new avenues for rapid, reliable, and high-throughput particle and cell identification in clinical and industrial applications, offering a valuable tool for understanding complex biological and environmental systems.

Abstract:Phenotyping single cells based on molecules that they secrete or consume is a key bottleneck in many biotechnology applications. Here, we demonstrate a new approach for detecting secretions from individual single cells, namely, plasmonic colloidosome-based single cell detector (PCSD). This strategy uses colloidosomes to encapsulate single cells and aid molecular detection. Colloidosomes are constructed by emulsifying lactate probe and hydrophobic ligand dual-functionalized silver nanoparticles on an immiscible liquid-liquid interface. The established colloidosome-based platform exhibits negligible surface-enhanced Raman scattering (SERS) background interference, ultrasensitive SERS response, and excellent signal reproducibility in response to acidification of the medium inside the colloidosomes. Taking lactate as a model molecule, the acidification induced by single cells confined in the colloidosomes is detected. The approach shows promising applicability in single cell analysis based on extracellular metabolites production or consumption.
