Abstract:Urolithiasis commonly occurs in kidney and ureteral, and may cause local organ/tissue damage, even kidney failure. The incidence of this disease is increasing worldwide, in which calcium oxalate is the major composition forming the urinary calculus. Therefore, to monitor this disease for the prevention and treatment, measuring the oxalate in the urine is of great significance. Here, a rapid and sensitive colorimetric method was developed based on 3,3',5,5'-tetramethylbenzidine-manganese dioxide (TMB-MnO2) nanosheets for oxalate detection. MnO2 nanosheets acted as an efficient biomimetic oxidase to catalyze the reaction with TMB and oxalate. Pale yellow TMB can be oxidized to blue oxide TMB catalyzed by BSA-stabilized MnO2 nanosheets, and oxalate can selectively inhibit this reaction by consuming and reacting with MnO2 nanosheets, thus achieving the quantitative detection of oxalate. Moreover, a home-made bionic electronic-eye (E-eye) system was developed as a portable in-situ detection platform to efficiently measure the oxalate concentrations in 10 s by direct photographing. By optimizing experimental conditions, this method shows a wide linear range (7.8 μM to 250 μM) and a low detection limit (0.91 μM) for oxalate detection. Besides, this method exhibits high selectivity even with 80-fold interfering chemicals. Furthermore, the performance of the method was validated by testing the artificial urine samples, indicating its great potential for monitoring and diagnosis of urolithiasis in point-of-care applications.

Abstract:Improving the sensitivity of the fluorescence method is crucial in biosensor fields. In this study, an ultrasensitive and excellent selectivity method was developed for the detection of trace Cu2+ ions using 3-mercaptopropionic acid capped CdSSe/CdS/ZnS quantum dots (MPA-CdSSe/CdS/ZnS QDs) as a fluorescent probe. Through energy band engineering to optimize the interior inorganic structure and passivate the surface defects, the photoluminescence quantum yield of the MPA-CdSSe/CdS/ZnS QDs in aqueous solution had as high as 70 %. The X-ray diffraction (XRD) and Transmission Electron Microscope (TEM) analysis revealed the prepared CdSSe/CdS/ZnS QDs were cubic zinc blende structure with average diameter 7.2 nm. Further, the fluorescence of the MPA-CdSSe/CdS/ZnS QDs was significantly quenched by the Cu2+ with fast response time through an efficient photo-induced electron transfer (PET) progress. By optimizing the thickness of CdS shell, the MPA-CdSSe/CdS/ZnS QDs probe with appropriate CdS shell thickness exhibited excellent linear linearity and sensitivity for the detection of Cu2+ ions with limit of detection as low as 0.05 nM. Additionally, ions interference experiments demonstrated the good selectivity of the probe towards Cu2+. Furthermore, this sensing system could also be applied to the measurement of Cu2+ ions in real water samples with satisfactory recoveries. This work provided new insights to design and synthesize high-performance QDs-based fluorescence probes with controllable exciton emission behavior for their applications in biosensor and biological fields.
Xanthine oxidase immobilized cellulose membrane-based colorimetric biosensor for screening and detecting the bioactivity of xanthine oxidase inhibitors

Abstract:Xanthine oxidase (XO) is a typical target for hyperuricemia and gout, for which there are only three commercial xanthine oxidase inhibitors (XOIs): febuxostat, topiroxostat and allopurinol. However, these inhibitors have problems such as low bioactivity and several side effects. Therefore, the development of novel XOIs with high bioactivity for the treatment of hyperuricemia and gout is urgently needed. In this work we constructed a XO immobilized cellulose membrane colorimetric biosensor (XNCM) by the TEMPO oxidation, amide bond coupling and nitro blue tetrazolium chloride (NBT) loading method. As expected, the XNCM was able to detect xanthine, with high selectivity and sensitivity by colorimetric method with a distinctive color change from yellow to purple, which can be easily observed by the naked-eye in just 8 min without any complex instrumentation. In addition, the XNCM sensor performed screening of 21 different compounds and have been successfully pre-screened out XOIs with biological activity. Most importantly, the XNCM was able to quantitatively detect the IC50 values of two commercial inhibitors (febuxostat and allopurinol). All the results confirmed that the XNCM is a simple and effective tool which can be used for the accelerated screening of XOIs and has the potential to uncover additional XOIs.

Abstract:In this work, we developed a ''naked-eye'' colorimetric and ratiometric fluorescence probe for a very important biomarker of uric acid (UA). The method was based on the oxidation of UA by uricase to allantoin and hydrogen peroxide, and then o-Phenylenediamine (OPD) was oxidized to the yellow-colored 2,3-diaminophenazine (oxOPD) in the presence of horseradish peroxidase (HRP) and hydrogen peroxide. The fluorescence emission of glutathione functionalized Ti3C2 MQDs (GSH-Ti3C2 MQDs) centered at 430 nm overlaps with the UV absorption of oxOPD at 425 nm to a large extent, which facilitates fluorescence resonance energy transfer (FRET) between GSH-Ti3C2 MQDs and oxOPD. With the increase of the UA concentration, the emission at 430 nm of GSH-Ti3C2 MQDs is progressively quenched and the emission at 568 nm of oxOPD was gradually increased. Moreover, the probe we designed is easier to distinguish with color change by naked eye for the detection of UA. This is the first report about the determination of UA by a ''naked-eye'' colorimetric and ratiometric fluorescence method combining GSH-Ti3C2 MQDs and uricase/HRP enzymes. Enhanced peroxidase‐like activity of MnFe2O4 nanoparticles on halloysite nanotubes for uric acid detection

Abstract:Nanozymes have significantly advanced sensing assays by replicating native enzyme functions. However, designing nanozymes with high catalytic activity and easy recyclability remains challenging. The study presented here has resulted in the development of a highly efficient and sensitive colorimetric system for the detection of uric acid, utilizing MnFe2O4@HNTs—a novel composite material consisting of MnFe2O4 loaded onto halloysite nanotubes. These nanocomposites exhibited outstanding peroxidase‐like activity and attractive magnetic properties. The catalytic efficiency of the MnFe2O4@HNTs in the oxidation of 3,3′,5,5′‐tetramethylbenzidine, in the presence of H2O2, was remarkable, leading to a distinct color change from colorless to blue. A linear relationship was observed between absorbance and UA concentration in the range of 1–20 μM, with a detection limit as low as 52 nM. Mechanistic investigations revealed that reactive oxygen species (ROS), specifically singlet oxygen (1O2), were generated through the decomposition of H2O2, which is responsible for the peroxidase‐like activity demonstrated by the MnFe2O4@HNTs. The method showed minimal interference from serum substances and high selectivity. Magnetic MnFe2O4 allowed easy separation and maintained over 95% activity after seven reuse cycles. The developed assay was successfully applied to the detection of uric acid in human serum, achieving recoveries greater than 98.60%. This research significantly advances the design of recyclable high‐performance nanozymes and establishes an effective colorimetric sensing platform for UA detection in clinical samples, potentially improving diagnostic tools for healthcare applications.

Abstract:Serum levels of uric acid (UA) play an important role in the prevention of diseases. Developing a rapid and accurate way to detect UA is still a meaningful task. Hence, positively charged manganese dioxide nanosheets (MnO 2 NSs) with an average latter size of 100 nm and an ultra-thin thickness of below 1 nm have been prepared. They can be well dispersed in water and form stable yellow-brown solutions. The MnO 2 NSs can be decomposed by UA via redox reaction, leading to a decline of a characteristic absorption peak (374 nm) and a color fading of MnO 2 NSs solution. On this basis, an enzyme-free colorimetric sensing system for the detection of UA has been developed. The sensing system shows many advantages, including a wide linear range of 0.10–50.0 μmol/L, a limit of quantitation (LOQ) of 0.10 μmol/L, a low limit of detection (LOD) of 0.047 μmol/L (3σ/m), and rapid response without need of strict time control. Moreover, a simple and convenient visual sensor for UA detection has also been developed by adding an appropriate amount of phthalocyanine to provide a blue background color, which helps to increase visual discrimination. Finally, the strategy has been successfully applied to detect UA in human serum and urine samples.

Abstract:The fabrication of efficient fluorescent probes that possess an excellent sensitivity and selectivity for uric acid is highly desirable and challenging. In this study, composites of silver nanoparticles (AgNPs) wrapped with nitrogen-doped carbon quantum dots (N-CQDs) were synthesised utilising N-CQDs as the reducing and stabilising agents in a single reaction with AgNO3. The morphology and structure, absorption properties, functional groups, and fluorescence properties were characterised by transmission electron microscopy, X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy, ultraviolet spectroscopy, fluorescence spectroscopy, and X-ray diffraction spectroscopy. In addition, we developed a novel method based on AgNPs/N-CQDs for the detection of uric acid using the enzymatic reaction of uric acid oxidase. The fluorescence enhancement of the AgNPs/N-CQDs composite was linear (R2 = 0.9971) in the range of 2.0–60 μmol/L, and gave a detection limit of 0.53 μmol/L. Trace uric acid was successfully determined in real serum samples from the serum of 10 healthy candidates and 10 gout patients, and the results were consistent with those recorded by Qianxinan Prefecture People's Hospital. These results indicate that the developed AgNP/N-CQD system can provide a universal platform for detecting the multispecies ratio fluorescence of H2O2 generation in other biological systems.

Abstract:Hypoxanthine concentration is a potential indicator to evaluate the freshness in the early post-mortem of several aquatic products. Based on MnO2 in-situ coated upconversion nanoparticles (UCNPs) and xanthine oxidase (XOD), a novel sensor was conducted for the efficient, sensitive determination of hypoxanthine. In this strategy, upconversion fluorescence quenched by MnO2 would be restored by H2O2 and uric acid (UA), two products from the XOD-catalyzed reactions of hypoxanthine. Through pretreatment with short-time heating and alkylation by N-ethylmaleimide (NEM) to avoid potential interference from reducing substances in the food matrix, this method exhibited satisfactory selectivity. The fluorescence intensity of green emission Igreen was positively proportional to hypoxanthine concentration at a wide range of 0.5-50 mg/L with a detection limit of 0.14 mg/L. Moreover, this convenient method was employed to quantify the hypoxanthine in fish, shrimp, and shellfish samples, showing excellent potential for the application in quality control of aquatic products.

Abstract:A dual-emission ratiometric fluorescent sensing system based on boron-doped carbon quantum dots (B-CQDs) and gold nanoclusters (AuNCs) has been developed for the determination of xanthine. The blue fluorescence of B-CQDs at 445 nm is then reduced by the AuNCs through the inner filter effect (IFE) under a single excitation wavelength of 370 nm. By the catalysis of xanthine oxidase (XOD), xanthine is oxidized by oxygen dissolved in the solution to produce H2O2. The horseradish peroxidase (HRP) catalyzes H2O2 to generate hydroxyl radicals, which can quench the fluorescence of AuNCs, leading to the recovery of the fluorescence of B-CQDs. Based on the relationship between the fluorescence intensity ratio (F445/F665) and the concentration of xanthine, the designed method exhibits a good linearity range of 1.2–500.0 μmol L −1 and a limit of detection of 0.37 μmol L −1. The ratiometric fluorescent is applied to determine xanthine in human urine samples. Good recoveries of spiked samples in the range 99.2–105.0% are obtained by the proposed assay, with relative standard deviations (RSD) ranging from 0.9 to 2.6%.

Abstract:Twitter has been recognized as a useful channel for the sharing and dissemination of health information, owing in part to its “retweet” function. This study reports findings from a content analysis of frequently retweeted obesity-related tweets to identify the prevalent beliefs and attitudes about obesity on Twitter, as well as key message features that prompt retweeting behavior conducive to maximizing the reach of health messages on Twitter. The findings show that tweets that are emotionally evocative, humorous, and concern individual-level causes for obesity were more frequently retweeted than their counterparts. Specifically, tweets that evoke amusement were retweeted most frequently, followed by tweets evoking contentment, surprise, and anger. In regard to humor, derogatory jokes were more frequently retweeted than nonderogatory ones, and in terms of specific types of humor, weight-related puns, repartee, and parody were shared frequently. Consistent with extant literature about obesity, the findings demonstrated the predominance of the individual-level (e.g., problematic diet, lack of exercise) over social-level causes for obesity (e.g., availability of cheap and unhealthy food). Implications for designing social-media-based health campaign messages are discussed.
A Biosensor Based on the Biomimetic Oxidase Fe3O4@MnO2 for Colorimetric Determination of Uric Acid

Abstract:High plasma urate is closely related to gout, cardiovascular and other diseases. Therefore, monitoring the content of uric acid (UA) in plasma is of great significance for the treatment of gout and the prevention of other related diseases. Herein, a biosensor based on the biomimetic oxidase Fe3O4 nanoparticles (NPs) @MnO2 nanosheets (Fe3O4@MnO2 NS) was constructed for colorimetric determination of UA. MnO2 NS is an efficient biomimetic oxidase, and we found that the intrinsic oxidase activity of MnO2 NS doped with Fe3O4 NPs can be significantly enhanced. The chromogenic substrate TMB can be catalyzed by Fe3O4 @MnO2 NS to generate blue oxidized TMB, and UA can decompose the MnO2 NS to inhibit the color reaction of TMB selectively, thereby realizing the quantitative detection of UA. In addition, the UA biosensor can perform colorimetric analysis of UA level through three methods: naked eye, smartphone and ultraviolet-visible (UV-vis) spectrophotometer. The linear ranges of UV-vis spectrophotometry and colorimetry with smartphone were 1-70 mu M and 200-650 mu M, respectively, and the limits of detection (LOD) were 0.27 mu M and 21 mu M. The analysis results of human plasma samples showed that the method had good selectivity and practicability.

Abstract:Xanthine oxidase (XO) is a crucial enzyme in the development of hyperuricemia and gout. This study focuses on LWM and ALPM, two food-derived inhibitors of XO. We used molecular docking to obtain three systems and then conducted 200 ns molecular dynamics simulations for the Apo, LWM, and ALPM systems. The results reveal a stronger binding affinity of the LWM peptide to XO, potentially due to increased hydrogen bond formation. Notable changes were observed in the XO tunnel upon inhibitor binding, particularly with LWM, which showed a thinner, longer, and more twisted configuration compared to ALPM. The study highlights the importance of residue F914 in the allosteric pathway. Methodologically, we utilized the perturbed response scan (PRS) based on Python, enhancing tools for MD analysis. These findings deepen our understanding of food-derived anti-XO inhibitors and could inform the development of food-based therapeutics for reducing uric acid levels with minimal side effects.

Abstract:In this study, using diethylenetriaminepentaacetic acid (DPTA), diethylenetriamine (DETA) and Fe (NO3)3.9H2O, Fe, N co-doped carbon dots (Fe, N-CDs) were successfully synthesized by a simple one-pot hydrothermal method. Fe, N-CDs possessed fluorescence and peroxidase-like properties. When H2O2 was present, o-phenylenediamine (OPD) can be oxidized by Fe, N-CDs to oxidized phenylenediamine (oxOPD), and oxOPD further quenched the fluorescence of Fe, N-CDs because of inner filter effect. There were two peaks at 450 nm and 560 nm for the system of Fe, N-CDs and oxOPD. Results showed that the concentration of H2O2 can not only affect the fluorescence intensity but also the color of the oxOPD. Considering that the xanthine oxidase can oxidize xanthine to produce H2O2, thus, a ratiometric fluorescent and colorimetric dual-signal sensor for H2O2 and xanthine was developed. This new method was demonstrated to have high sensitivity and excellent selectivity for determination of H2O2 and xanthine. The detection limits (LOD) for H2O2 and xanthine were 0.047 μM and 0.02 μM with a ratiometric fluorescence method, and were 0.05 μM and 0.023 μM with a colorimetric method, respectively. This dual-signal sensor was used successfully for xanthine detection in human serum and urine.

Abstract:A simple dual-signal assay that combined colorimetric and fluorometric strategy for uric acid (UA) rapid detection was designed based on the versatility of facile synthesized MnO 2 nanosheet. The oxidization of 3,3',5,5'-tetramethylbenzidine (TMB) and the fluorescence quenching of quantum dots (QDs) could occur simultaneously by MnO 2 nanosheet. The present of UA was able to decompose MnO 2 nanosheet into Mn 2+ , resulting in the fluorescence recovery of QDs, along with the blue of ox TMB be faded. Based on the principles above, the detection of UA could be realized by the change of the dual signals (colorimetric and fluorometric). The linear range of the colorimetric mode was 5 ~ 60 μmol/L, and the limit of detection (LOD) was 2.65 μmol/L; the more wide linear range of the fluorescence mode was 5 ~ 120 μmol/L, and the LOD could be as low as 1.33 μmol/L. The method was successfully analyzing UA level in human serum samples, indicating that this new dual-signal method was able to apply in clinical diagnosis.

Abstract:In the human body, oxalate tends to form calcium oxalate with calcium ions, which can trigger the formation of stones in the urinary system. Therefore, oxalate in urine is usually utilized as a crucial biomarker in clinical urolithiasis diagnoses. In this work, a turn-on fluorescent nanoprobe was developed based on nitrogen-doped carbon dots (N-CDs) and MnO2 nanosheets (NSs) nanocomposites for oxalate sensing in urolithiasis patients. MnO2 NSs is a good sensing platform with high extinction coefficients and rich redox chemistry. The fluorescent N-CDs can be quenched efficiently by MnO2 NSs through the inner filter effect (IFE) to form N-CDs-MnO2 nanocomposites. The reductive oxalate could operate the decomposition of MnO2 NSs to Mn2+ resulting in the dissociation of the N-CDs-MnO2 nanocomposites and fluorescence recovery of N-CDs. Under optimal conditions, the developed sensor revealed good specificity toward oxalate with a limit of detection (LOD) of 0.69 μM. The developed sensor was successfully applied to quantify oxalate content in 47 urine samples (41 urolithiasis patients and 6 healthy persons). The results showed great consistency with clinical diagnostic reports and computed tomography images. This novel method retains several unique advantages, including affordable, rapid, and validating potential clinical application.

Abstract:A novel uricase‐free colorimetric method was developed for the sensitive and selective detection of uric acid based on the light‐induced oxidase‐mimicking activity of a new photosensitized covalent organic framework. As large numbers of people are suffering from gout, an accurate, rapid, and sensitive method for the detection of gout biomarker, uric acid, is important for its effective control, diagnosis, and therapy. Although colorimetric detection methods based on uricase have been considered, they still have limitations as they produce toxic H2O2 and are expensive and not stable. Here, a novel uricase‐free colorimetric method was developed for the sensitive and selective detection of uric acid based on the light‐induced oxidase‐mimicking activity of a new photosensitized covalent organic framework (COF) (2,4,6‐trimethylpyridine‐3,5‐dicarbonitrile–4‐[2‐(4‐formylphenyl)ethynyl]benzaldehyde COF [DCTP–EDA COF]). DCTP–EDA COF has a strong ability to harvest visible light, and it could catalyze the oxidation of 1,4‐dioxane, 3,3′,5,5′‐tetramethylbenzidine under visible light irradiation to produce obvious color changes. With the addition of uric acid, however, the significant inhibition of the oxidase‐mimicking activity of DCTP–EDA COF remarkably faded the color, and thus uric acid could be colorimetrically detected in the range of 2.0–150 μM with a limit of detection of 0.62 μM (3σ/K). Moreover, the present colorimetric method exhibited high selectivity; uric acid level in serum samples was successfully determined, and the recoveries ranged from 96.5% to 105.64%, suggesting the high accuracy of the present colorimetric method, which demonstrates great promise in clinical analysis.

"Where's the Interface?" Enhanced Use Models for Mobile Interaction

Abstract:This paper is an overview of my dissertation work, which addresses challenges involved in designing use models, interfaces and interaction styles for mobile technology that go beyond notions and metaphors found in desktop computing. A model of attention and a model of use are presented to explain distinctive requirements of mobile technology. Prototypes are being developed in collaboration with ABB, Scandinavian Airlines System and The Interactive Institute to demonstrate three different use models, where the embodied use model—which blends the gap between the virtual and the physical—is of particular interest.

Abstract:Small-molecule drugs play an important role in the treatment of various diseases. The screening of enzyme inhibitors is one of the most important means in developing therapeutic drugs. Herein, we demonstrate a liquid crystal (LC)-based screening assay assisted with enzyme catalysis-induced aptamer release for screening xanthine oxidase (XOD) inhibitors. The oxidation of xanthine by XOD prevents the specific binding of xanthine and its aptamer, which induces a bright image of LCs. However, when XOD is inhibited, xanthine specifically binds to the aptamer. Correspondingly, LCs display a dark image. Three compounds are identified as potent XOD inhibitors by screening a small library of triazole derivatives using this method. Molecular docking verifies the occupation of the active site by the inhibitor, which also exhibits excellent biocompatibility to HEK293 cells and HeLa cells. This strategy takes advantages of the unique aptamer-target binding, specific enzymatic reaction, and simple LC-based screening assay, which allows high-throughput and label-free screening of inhibitors with high sensitivity and remarkable accuracy. Overall, this study provides a competent and promising approach to facilitate the screening of enzyme inhibitors using the LC-based assay assisted with the enzyme catalysis-induced aptamer release.

Abstract:Given the metabolization of xanthine into uric acid, and the relevance with diseases and food safety, it is significantly desirable to propose a rapid, sensitive, and reliable strategy to monitor the expression level of xanthine in biological liquids. Herein, a H2O2-responsive colorimetric sensing system was triumphantly developed for xanthine assay by combining peroxidase-mimetic magnetic Fe nanoparticles (Fe NPs) and xanthine oxidase (XOD). XOD catalyzed xanthine oxidizing into H2O2, which cooperated with Fe NPs to catalyze the oxidation of chromogenic substrate 3,3′,5,5′-tetramethylbenzidine (TMB) into oxidized (oxTMB) along with the absorbance at 652 nm appearing and increasing. Therefore, a sensitive, rapid, and convenient colorimetric platform for H2O2 and xanthine sensing was successfully developed with a limit of detection (LOD) of 0.029 μM for H2O2 and 0.034 μM for xanthine. In general, the Fe NPs nanozyme with high peroxidase-like activity features the low-cost, high-performance, and ease of preparation, then providing an efficient and sensitive sensing platform for blood xanthine monitoring as an alternative to traditional strategies.
