Abstract:Biological preservation methods with bacterial antagonists have emerged as alternatives to chemical sanitizers for extending shelf-life and reducing the population of pathogenic microorganisms. In addition, calcium plays an important role in maintaining the quality of fruit, and postharvest calcium treatments might determine the potential of fruit for processing. The objective of this work was to evaluate the effect of the postharvest application of calcium and biopreservation with the CPA-7 strain of Pseudomonas graminis on the quality parameters of fresh-cut pears. After harvest, whole pears were dipped in calcium chloride solution (1%, w/v) or water (control) for 10 min at 25 ℃ and stored for five months at temperatures ranging from 0 to -0.5 ℃. Both batches of fruit were minimally processed and dipped in a solution containing CPA-7 and an antioxidant solution or kept untreated, and both groups were stored at 4 ℃ for six days. The postharvest calcium treatment had no remarkable effect on the quality of the whole and fresh-cut pears. The enzymatic activities (PPO, PME and PG) related to browning and softening were constant in fresh-cut pears after storage, and the application of P. graminis CPA-7 had a positive effect on the activity of PPO. Finally, a combined effect of the biocontrol agent and calcium treatment was not demonstrated.

Abstract:Fresh-cut pear are popular for their nutritive, fresh and convenience. However, the oxidative stress caused by wounding aggravates quality loss. In the present study, the effects of 10 % CO2 on reactive oxygen species (ROS) metabolism and antioxidant capacity in fresh-cut pear were investigated. The results showed that 10 % CO2 effectively inhibited ROS accumulation during a 5-day storage, accompanied by reducing the content of hydrogen peroxide (H2O2) and superoxide anion radical (O2−). Furthermore, non-enzymatic antioxidant properties including free radical scavenging ability (DPPH), total antioxidant capacity (ABTS), and ferrous reducing ability (FRAP) in CO2-treated group were increased by 5.5 %, 23 % and 27 % respectively at the end of storage. Additionally, 10 % CO2 treatment enhanced the activities and genes expression levels of catalase (CAT), glutathione peroxidase (GPX), ascorbate peroxidase (APX), glutathione reductase (GR), superoxide dismutase (SOD) and peroxidase (POD), while NADPH oxidase (NOX) were inhibited. The accumulation of ascorbic acid (AsA) and reduced glutathione (GSH) were also accelerated by CO2. This study indicated that the generation of ROS was inhibited with inducing effects of genes and enzymes related to antioxidant capacity in response to 10 % CO2, which eventually alleviated oxidative damage and conserved storage quality of fresh-cut pear.

Abstract:Superficial scald, which appears as black or brown necrotic patches on the skin, is a serious postharvest physiological disorder of pear fruit. The occurrence of scald in 'Chili' pear (Pyrus bretschneideri) fruit during storage at 2 degrees C in response to preharvest polyethylene (PE) bagging and non-woven fabric bagging has been investigated. The non-woven fabric bagging treatment of fruit prevented scald development, while the PE bagged fruit had a higher incidence of scald than the untreated fruit. Fruit from the PE bagging treatment had lower Ca content, less distribution and lower flux rate of Ca2+ than no bagging and non-woven fabric bagging fruit. The treatment also was associated with greater expression of genes encoding calmodulin-like (CML) proteins, such as PbCML19, PbCML5, PbCML38, PbCML42-1, and PbCML42-2. In contrast, non-woven fabric bagging did not affect the expression of these genes in the fruit. In addition, CaCl2 treatment reduced scald development in PE bagged pear fruit during storage. Our results suggest that Ca2+ may play a role in regulating the occurrence of superficial scald in pear fruit.
Efficacy of Pseudomonas graminis CPA-7 against Salmonella spp. and Listeria monocytogenes on fresh-cut pear and setting up of the conditions for its commercial application

Abstract:Pseudomonas graminis CPA-7 has been reported to control foodborne pathogens on fresh-cut apple, peach and melon. The first aim of this study was to assess its antagonistic activity against Salmonella spp. and L. monocytogenes on fresh-cut pear. CPA-7 was able to control both pathogens on fresh-cut pear stored in air conditions at 5, 10 and 20 °C. However, when CPA-7 antagonistic effect was tested by simulating commercial application (with antioxidant solution and passive modified atmosphere packaging), its effect decreased and no reductions of foodborne pathogens were reported at 10 °C. Therefore, the second aim was to optimise the antioxidant solution and the packaging in order to retain its antagonistic capacity. The selected antioxidant solution was 2% ascorbic acid +2% sodium citrate +1% CaCl2 according to growth and effect of CPA-7. Film permeability, which affects gas composition inside fruit packages, influenced CPA-7 efficacy. If the biopreservative strain is used, film has to be sufficiently gas permeable to allow CPA-7 function and at the same time to maintain product quality.

Abstract:After harvesting fruits and vegetables and during storage and transportation, its sensorial, nutritional and sensorial quality decreases due to its high moisture content, microbial growth, environmental factors, maturity and senescence. Considering their very short shelf life, fruits and vegetables need immediate post-harvest care to increase it. The most common conservation methods are based on the control of the transpiration rate, respiration and microbial spoiling. However, traditional methods have proven to be vulnerable. Between 25 and 40% of fruits and vegetables are lost before consumption because of inadequate post-harvest treatments. In recent years, the development of alternative methods to assure microbial safety of fresh fruits and vegetables has been an important topic of investigation. Among these new methods, biopreservation using Lactic Acid Bacteria has been gaining interest, since they are classified as “generally recognized as safe” and have shown antimicrobial capacities; additionally, it is considered an environmentally friendly method. On the other hand, microencapsulation in a biodegradable matrix protects Lactic acid bacteria against unfavorable environmental conditions, which maintains their viability for a longer period. Therefore, the use of microencapsulated Lactic acid bacteria promises to be an effective technique to guarantee safety and to extend the useful life of fruits and vegetables during post-harvest. This review describes the main methods of preservation, as well as the emerging methods used to preserve fresh fruits and vegetables, with emphasis on bio-preservation as a proposal for future research.
Mucilage-Based and Calcium Ascorbate Edible Coatings Improve Postharvest Quality and Storability of Minimally Processed Cactus Pear Fruit Stored under Passive Atmosphere

Abstract:The minimally processed fruit and vegetable industry showed rapid growth worldwide, primarily due to the increasing consumer need for ready-to-eat fresh products characterized by high nutritional, sensory and healthy value. The postharvest life of peeled cactus pear fruits is relatively short, due to the processing operations that affect fruit integrity and cause metabolic disfunctions, as well as pulp browning, microbial growth, loss of firmness, off-flavor development, and nutraceutical value loss. In this study, we investigated the effects of mucilage-based (OFI) and calcium ascorbate edible coating on minimally processed cactus pear summer-ripening fruit, cold stored under passive atmosphere. The effect of the edible coating on the postharvest life, quality attributes, and nutraceutical value of fruit was evaluated by colors, total soluble solids content, carbohydrates; titratable acidity, ascorbic acid, betalains, DPPH, visual quality, and sensorial analysis. Our data showed a significant effect of mucilage-based and calcium ascorbate-based coating on preserving quality, nutritional value, sensorial parameters, and improving postharvest life of minimally processed cactus pear fruits; OFI had the most effective barrier effect. Furthermore, both coating treatments did not negatively affect the natural taste of minimally processed cactus pear fruits, which is an important aspect regarding the use of edible coatings when taste modification is undesirable.

Abstract:Table grapes are perishable crops with a high susceptibility to postharvest phytopathogens. Use of natural phytochemicals as elicitors for improving crop quality and enhancing the natural resistance against postharvest pathogens is a novel approach. Calcium ascorbate (CaAs) is an ecofriendly and inexpensive compound used as anti-browning agent in fresh cut fruits and minimally processed products. Thompson seedless table grapes ( Vitis vinifera L.) were treated with calcium ascorbate (CaAs) at 0, 5% and 10% 48 h before harvest and stored for 60 days at 1 ± 1 °C after harvest. CaAs of 5% significantly enhanced the berry total acids, total phenolics, flavonoids and carotenoids contents, and total antioxidant, ascorbate peroxidase, superoxide dismutase and catalase enzymes activities and decreased total soluble solids content, malondialdehyde, electrolyte leakage and pH. The treatment at both concentrations enhanced the berry firmness and decreased browning and color change indices at harvest time and during storage. After 2 months of storage, the highest ascorbate peroxidase activity, flavonoids and carotenoids contents was recorded in fruit treated with 10% CaAs, while the lowest decay extension rate, electrolyte leakage, weight loss, browning and color change was recorded in the berries treated with 5% CaAs. Furthermore, CaAs at 5% caused a significant decrease in weight loss and decreased berry abscission and decay extension rates during storage.

Suitability of combination of calcium propionate and chitosan for preserving minimally processed banana quality

Abstract:Background: The marketability of fresh-cut banana slices is limited by the rapid rate of fruit softening and browning. However, there is no scientific literature available about the role of postharvest calcium propionate and chitosan treatment on the quality attributes of fresh-cut banana. Therefore, the aim of the present study was to investigate these effects. Results: The application of calcium propionate plus chitosan (CaP+Chit) retained higher firmness, higher ascorbic acid content, higher total antioxidant activity and higher total phenolic compounds, along with lower browning, lower polyphenol oxidase, lower peroxidase, lower polygalacturonase and lower pectin methyl esterase activities and microbial growth, compared to control banana slices after 5 days of cold storage. Conclusion: The results of the present study show that CaP+Chit could be used to slow the loss of quality at the same time as maintaining quality and inhibiting microbial loads. © 2017 Society of Chemical Industry.

Abstract:Abstract Two types of plasminogen activator (PA), t-PA (tissue type) and u-PA (urokinase type), are released from endometrial tissue in organ culture, as judged by immunological identification and molecular weight. Addition of estradiol to the medium greatly enhanced the release of u-PA, whereas that of t-PA was not low. Addition of progesterone, on the other hand, after priming of the endometrial tissue with estradiol, resulted in a much lower release of both types of PA. This pattern of PA release in response to hormonal stimulation in vitro agrees with previous observations of the PA activity of endometrial secretion in vivo. Endometrial tissue also released a PA inhibitor with molecular weight of approximately 50,000, which complexed both t-PA and u-PA. In cultures stimulated with estradiol the amount of free u-PA increased gradually during incubation and minor amounts of free t-PA appeared after 4-6 days culture. The amount of complexes, and thus the amount of PA inhibitor also increased under influence of estradiol. In cultures stimulated with progesterone, on the other hand, only minor amounts of free u-PA and no free t-PA was detected. The inhibitor might be of either the endothelial or the placental type, or both.

Abstract:Putrescine and MAP treatments reduced the storage losses and decay ratio during cold storage in pear. Putrescine and MAP treatments maintained the fruit quality and bioactive compounds of pear during cold storage. Putrescine and MAP treatments have a significant effect on extending the post‐harvest life of pear. Pear (P. communis L.), which is a climacteric fruit species, has a very short storage and shelf life, and significant losses occur due to high metabolic activity and the fruit's respiration rate after harvest. Therefore, preventing or reducing post‐harvest quality losses in pear is one of the most basic problems awaiting solution. In this study, we planned for this purpose; the fruits of the Ankara pear cultivar treated with modified atmosphere packaging (MAP), putrescine (1 mM), and MAP + putrescine were stored for 120 days at 1°C and 90 ± 5% relative humidity. The quality analyses and measurements, such as weight loss, decay rate, fruit firmness, soluble solids content (SSC), titratable acidity (TA), total phenolic compounds, antioxidant capacity, organic acids, and specific phenolic compounds, were performed on the 30th, 60th, 90th, and 120th days. Weight loss and decay ratios were lower for putrescine and putrescine + MAP‐applied fruit. With these applications, the softening of the fruit was slowed down, and the increasing SSC in the fruit and the decreasing TA rates were lower, and thus the ripening of the fruit was delayed. Changes in individual phenolic content and organic acids were lower in MAP and putrescine‐applied fruit. The study revealed that MAP and putrescine applications in pear can be used effectively to maintain fruit quality after harvest.

Abstract:Fungal pathogens are the main causal agents of postharvest diseases of fruits and vegetables. To prevent this problem and avoid the use of harmful chemical fungicides, safer and greener alternatives have been sought. One of these alternatives is the use of plant-growth-promoting bacteria (PGPB). In this study, we evaluated in vitro four well-known PGPB strains (Pseudomonas fluorescens UM270, Bacillus toyonensis COPE52, Bacillus sp. E25, and Bacillus thuringiensis CR71) for their biocontrol potential against nineteen postharvest fungal pathogens. In vivo assays were also performed, and bacterial cells were inoculated on harvested strawberries and grapes with the pathogens Botrytis cinerea, Alternaria alternata, and Fusarium brachygibbosum to evaluate loss of firmness and disease incidence. Our results show that the four strains antagonized fungi in direct and indirect confrontation assays. Stronger antagonism was observed by the action of diffusible metabolites (DMs) compared to volatile organic compound (VOC) activity. All PGPB significantly improved the fruit firmness and reduced disease incidence caused by the fungal pathogens tested. However, strain UM270 showed excellent biocontrol activity, reducing the disease incidence of Fusarium brachygibbosum, Botrytis cinerea, and Alternaria alternata on strawberry fruits by 60%, 55%, and 65%, respectively. Diffusible antifungals and VOCs such as 2,4-diacetyl phloroglucinol, siderophores, auxins, fengycins, and N, N-dimethyl-hexadecyl amine, among others, might be responsible for the beneficial activities observed. These results suggest excellent biocontrol activities to inhibit postharvest pathogenic fungi and improve harvested fruit quality.

Abstract:Spoilage caused by microorganisms is the main problem of fresh-cut fruits during storage, leading to quality deterioration and short shelf life. Lactic acid bacteria (LAB) contribute to fruit preservation by producing secondary metabolites with antibacterial activity, like bactericins, organic acids, and extracellular polysaccharides, etc. In the study, the effects of Lactiplantibacillus plantarum BX62 and LP1-4 on the composition and dynamics of the microbiome on the surface of fresh-cut apples were investigated. L. plantarum survived and remained stable (>7 log CFU/mL). The cell suspensions (9 log cells/mL) of BX62, LP1-4, and BX62+LP1-4 showed varying degrees of antagonistic activity against microorganisms on the surface of fresh-cut apples during cold storage. Moreover, L. plantarum treatment inhibited the increase of bacterial diversity, and their cell suspensions also reduced the relative abundance of Actinobacteria, Acidobacteria, Basidiomycota, Rhodococcus, Pantoea, Ralstonia, Vishniacozyma, and Cystofilobasidium on the surface of fresh-cut apples. The results provide new insight into the characterization of microbial communities on the surface of fresh-cut apples after LAB treatment, which might assist in developing new biocontrol agents to control the microbial contamination of fruits.

Abstract:Rhodotorula glutinis was evaluated for its activity in reducing postharvest gray mold decay and blue mold decay of pear caused by Botrytis cinerea and Penicillium expansum respectively, and in reducing natural decay development of pear fruits, as well as its effects on postharvest quality of fruits. There was a significant negative correlation between concentrations of the yeast cells and infectivity of the pathogens. At concentrations of R. glutinis at 5x108 CFU/ml, the gray mold decay was completely inhibited after 7 days incubation at 20 degrees C, while the control fruit had 100% disease incidence and 2.15 cm lesion diameter respectively, at challenged with B. cinerea spores suspension of 1x105 spores/ml; No completely control was got to blue mold, when pear fruits stored at 20 degrees C for 7 d (challenged with P. expansum spores suspension of 5x104 spores/ml), but the decay was distinctly prevented with 20% and 0.60 cm of disease incidence and lesion diameter respectively, while the control fruits were 100% and 2.74 cm, respectively. Rapid colonization of the yeast in wounds was observed during the first 1 d at 20 degrees C, and then the populations stabilized for the remaining storage period. On pear wounds kept at 4 degrees C, rapid colonization of the yeast in wounds was observed during the first 3 d, and then the increase in population density of R. glutinis turned slow, which continued over 6 d after application of the antagonist until it reached a high level. Then, the populations stabilized for the remaining storage period. In the test on PDA plates, R. glutinis significantly inhibit the growth of B. cinerea and P. expansum. Spore germination of pathogens in PDB was greatly controlled in the present of living yeast cell suspensions. R. glutinis significantly reduced the natural development of decay of pear following storage at 20 degrees C for 7 days or at 4 degrees C for 30 days followed by 20 degrees C for 5 days, and did not impair quality parameters, including mass loss, firmness, TSS, ascorbic acid or titratable acidity. Thus, the application of R. glutinis can be an alternative to chemicals for control of postharvest diseases on pear fruits.

Abstract:The demand for healthy and ready-to-eat products, such asfreshly-harvested fruits, has been growing steadily over the years. However, these products are very susceptible to spoilage and have a short shelf-life. In this research, edible coatings based on sodium alginate and its blends with ascorbic acid (a natural antioxidant and anti-browning agent) were applied on fresh-cut pineapple samples, and the changes in their physicochemical properties were monitored during 10 d of storage at 4ºC. Initially, the surface of the coated fruits was brighter and statistically significant differences were not found between uncoated and coated samples (P<0.05); similar values were obtained in the parameters of soluble solids (~11 ºBrix), pH (~3.74) and titratable acidity (~0.64%). During storage, coated samples were more protected against changes in appearance compared to uncoated fresh-cut pineapple samples. The current results will be beneficial for further research that focuses on the preservation of minimally processed fruits such as pineapple.

Prevention of microbes-induced spoilage in sodium chloride-free cucumber fermentations employing preservatives

Abstract:This study evaluated preservatives to stabilize sodium chloride (NaCl)-free-cucumber fermentations. The brining of air-purged laboratory cucumber fermentations with 100.0 mM calcium chloride (CaCl2 ) and 25.0 mM acetic acid resulted in immediate rises in pH, the chemical reduction of the medium, and malodors. Supplementation with 3.0 mM sodium benzoate or 3.0 mM potassium sorbate enabled a decline in pH, a continuous oxidative state of the medium, and delayed rising pH spoilage. However, lactic and acetic acids eventually disappeared in fermentations supplemented with preservatives. The amount of preservatives needed to suppress growth of brined-cucumber-spoilage microbes was determined in Fermented Cucumber Juice Medium (FCJM). Supplementation of FCJM with 10.0 mM sodium benzoate was inhibitory for the spoilage yeasts, Issatchenkia occidentalis and Pichia manshurica, and the lactobacilli, Lentilactobacillus buchneri and Lentilactobacillus parafarraginis, but not of Zygosaccharomyces globiformis. Potassium sorbate inhibited the spoilage yeasts at 15.0 mM in FCJM but not the lactobacilli. Supplementation of FCJM with 20.0 mM fumaric acid had a bactericidal effect on the spoilage-associated lactobacilli. As expected, NaCl-free-commercial cucumber fermentations brined with 100 mM CaCl2 , no acetic acid, and 6 mM potassium sorbate resulted in complete fermentations, but supported rising pH, microbially induced spoilage during long-term storage. Post-fermentation supplementation with 12 mM sodium benzoate, 10 mM fumaric acid, a combination of the two, or 10 mM fumaric acid and 2 mM AITC prevented microbial activity during long-term bulk storage. PRACTICAL APPLICATION: Several preservative-based strategies for stabilizing NaCl-free cucumber fermentation in a commercial production setting were developed, enabling the implementation of a processing technology that reduces wastewater volumes and environmental impact.

Abstract:Eating fresh fruits and vegetables is, undoubtedly, a healthy habit that should be adopted by everyone (particularly due to the nutrients and functional properties of fruits and vegetables). However, at the same time, due to their production in the external environment, there is an increased risk of their being infected with various pathogenic microorganisms, some of which cause serious foodborne illnesses. In order to preserve and distribute safe, raw, and minimally processed fruits and vegetables, many strategies have been proposed, including bioprotection. The use of lactic acid bacteria in raw and minimally processed fruits and vegetables helps to better maintain their quality by extending their shelf life, causing a significant reduction and inhibition of the action of important foodborne pathogens. The antibacterial effect of lactic acid bacteria is attributed to its ability to produce antimicrobial compounds, including bacteriocins, with strong competitive action against many microorganisms. The use of bacteriocins, both separately and in combination with edible coatings, is considered a very promising approach for microbiological quality, and safety for postharvest storage of raw and minimally processed fruits and vegetables. Therefore, the purpose of the review is to discuss the biopreservation of fresh fruits and vegetables through the use of lactic acid bacteria as a green and safe technique.

Abstract:The effects of whole cells of three different O serotypes of Vibrio anguillarum on the murine immune response were studied. The addition of different doses (1–100/ig/ml) of V. anguillarum cells, as well as Salmonella typhimurium lipopolysaccharide, markedly increased the incorporation of [3H] thymidine into in vitro cultured spleen cells of C57BL/6 mice. All three serotype strains of V. anguillarum were able to induce the mitogenic effect at 10 μg/ml and 100 μg/ml, but serotype I strains were more potent than the others. Since pretreatment of spleen cells with rabbit anti‐mouse thymocyte antiserum did not affect the mitogenic activity of V. anguillarum, Vibrio cells may be a B‐lymphocyte mitogen.
