Abstract:In situ gelling thermosensitive hydrogel formulation has been reported to effectively sustain the release of macromolecules for a long time. However, the low-molecular-weight hydrophilic drugs, such as doxorubicin (DOX), are not suitable for intratumoral injection because the release will complete within one day. In this study, liposomal doxorubicin (LipDOX) was added into the hydrogel to form a novel thermosensitive formulation which prolonged the sustained release of DOX. DOX+C/GP (doxorubicin in chitosan/β-glycerophosphate) was prepared to compare with LipDOX+C/GP (liposomal doxorubicin in chitosan/β-glycerophosphate hydrogel). The particle size of DOX-loaded liposome was 94.2nm and the encapsulation efficiency of DOX was near 98%. In vitro release experiments, the release of DOX in both DOX+C/GP group and LipDOX+C/GP group increased along with the increasing pH of buffers. However, the LipDOX+C/GP group with lower initial burst release had a much longer releasing duration than DOX+C/GP group (21days vs. 24h). In vitro and in vivo antitumor experiments demonstrated that LipDOX+C/GP group had better antineoplastic effect and less toxicity than DOX+C/GP group. Pharmacokinetics study showed LipDOX+C/GP exhibited a higher AUC0-t and longer MRT than DOX+C/GP in blood and tumor, which indicated that LipDOX+C/GP obtained an enhanced antitumor activity compared with DOX+C/GP. In addition, the lower distribution index (the ratio of AUC of normal tissue/AUC of tumor tissue) of the LipDOX+C/GP implied it had lower toxicity to normal tissues than DOX+C/GP. Therefore, the novel thermosensitive hydrogel formulation was potential for clinical application in cancer treatment.

Abstract:Population conferences have evolved through three overlapping stages, each stage reflecting a different perspective on the relationship between national policy formulation and the international system. Initially, at Rome in 1954 and in Belgrade in 1965, participants were invited on the basis of their expertise and were not expected to represent their governments. By the time of the Bucharest Conference in 1974, the United Nations, in an attempt to give conferences a greater role in shaping population policy and to inspire member governments to show greater concern for their own population problems, decided that conferences would be intergovernmental gatherings and that national delegations would be selected by governments. The effect of this change was that governments gave less weight to scientific expertise and, conversely, greater weight to political and bureaucratic considerations. At the present time, although conferences remain intergovernmental gatherings, the door has been opened for nongovernmental organizations--civil society--to play a more active role in the conference process and in deliberations. As conferences have become more inclusive, their focus has veered from what has conventionally been regarded as population concerns. The changing composition of participants at UN conferences has had the effect of altering the policy agenda in the international population field and, as some have argued since Cairo, has redefined the meaning of population. Conferences in the twenty-first century likely will be compelled to confront diverse demographic problems in addition to social issues demanding the attention of the political system at every level.
Immunocytochemical localization of growth factors and their receptors in human pre-embryos and Fallopian tubes.

Abstract:We utilized indirect immunocytochemistry to demonstrate the presence of growth factors and their receptors in human pre-embryos and Fallopian tubes. In pre-embryos, only transforming growth factor-alpha (TGF-alpha) and the intracellular domain of epidermal growth factor receptor (EGFR) were found at the 4-cell stage. In 8- to 14-cell pre-embryos, TGF-alpha, the intracellular and extracellular domains of EGFR, and insulin-like growth factor-I and its receptor were found. Antibodies against TGF-alpha stained all Fallopian tube specimens, while the extracellular domains of EGFR was only found in specimens from patients with either blood type A or AB. These results suggest a cross-reactivity between the extracellular domain of the EGFR and blood group antigens. Our novel demonstration of growth factor receptor staining in human pre-embryos shows that growth factor receptor localization is dependent on the developmental stage of human pre-embryos. We have also established a potentially important link between the Fallopian tube which secretes growth factors and the localization of growth factor receptors in pre-embryos. These findings are compatible with the hypothesis that tubal secretions are embryotrophic for the early development of the pre-embryo.
In Vitro Evaluation of Lysozyme-loaded Microspheres in Thermosensitive Methylcellulose-based Hydrogel

Abstract:Long-term injectable microspheres have some inherent disadvantages such as migration of microspheres from the original site and the burst effect. In order to avoid these problems, microsphere-loaded thermosensitive hydrogel system was designed and expected to achieve a zero-order release of biomolecular drugs in relative high initial drug loadings. Lysozyme, an antibacterial protein usually used to reduce prosthetic valve endocarditis, was selected as the model drug. Poly (DL-lactide-co-glycolide) (PLGA) microspheres, prepared by solvent evaporation method, were employed to encapsulate lysozyme and dispersed into thermosensitive pre-gel solution containing methylcellulose (MC), polyethylene glycol (PEG), sodium citrate (SC), and sodium alginate (SA). The mixture could act as a drug reservoir by performing sol-gel transition rapidly if the temperature was raised from room temperature to 37 degrees C. The in vitro release results showed that the burst effect was avoided due to strengthening of diffusion resistance in the gel. The formulation was able to deliver lysozyme for over 30 days in a nearly zero-order release profile with a rate of 32.8 mu g.d(-1) which exhibits its remarkable potential for effective application in long-term drug delivery.
Liposomes interiorly thickened with thermosensitive nanogels as novel drug delivery systems.

Abstract:The fundamental structure of liposomes suffers from drawbacks of physical instability. To overcome this problem, the hypothesis of this study was to thicken the liposomal interior by incorporating thermosensitive in situ gel. The so called gelliposomes (GLs) were prepared by a thin-film method using poloxamer solutions as interior aqueous phase. Interior thermosensitive gelation was proved by observation of sustained dissolving of the poloxamer gel after destroying the lipid bilayers with Triton X-100; structural transformation as observed under optical microscopy in a heating–cooling circle also proved the fact of interior gelling. The sol–gel transition temperatures of GLs were in good correlation with those of the bulk poloxamer solution counterparts, which could be easily tailored by adjusting the concentration and ratio of poloxamer 407 (P407) to poloxamer 188 (P188). Membrane anisotropy measurement indicated increased membrane rigidity. In vitro release of the model drug cytosine arabinoside from GLs showed sustained release characteristics for at least one week with typical biphasic kinetics. Study on storage stability and protection against the destroying effect by membrane destroyers indicated improved physical stability in comparison with conventional liposomes. In situ evading of phagocytic uptake by macrophages was observed for GLs, which however should be attributed to the effect of exteriorly adsorbed poloxamers. In conclusion, GLs present distinct characteristics to be used as potential drug delivery systems.

Abstract:Abstract The article presents a hepatocellular carcinoma cell surface-specific ligand glycyrrhetinic acid (GA) and cell-penetrating peptide (TAT) with good cell membrane penetration to modify the anti-tumor drug pingyangmycin (PYM) liver delivery system, which achieve targeted delivery of drugs and improve anti-tumor efficiency. In this study, we synthesized the pingyangmycin liposome modified by glycyrrhetinic acid and cell penetrating peptide(GA-TAT-PYM-L) and evaluated the anti-tumor effect of GA-TAT-PYM-L in vitro . Using the 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenylte-trazolium bromidecell proliferation method, GA-TAT-PYM-L had a stronger inhibitory effect on HepG2 cells than the free drug PYM at the same concentration. Acridine orange-ethidium bromide staining assays showed that GA-TAT-PYM-L had stronger apoptosis promotion effects on HepG2 cells in comparison to PYM. Pharmacokinetic studies indicated that, compared with PYM, GA-TAT-PYM-L enhanced mean residence time (MRT 0–∞ ) and area under curve (AUC 0–∞ ) by about 2.79-fold and 2.45-fold. The T 1/2 was prolonged to 140.23 ± 14.13 min. Tissue distribution results showed that the PYM concentrations in livers from the GA-TAT-PYM-L group were always higher than other tissues at each monitoring period after 5 min, indicating that GA-TAT-PYM-L can achieve liver targeting.

Abstract:To enhance the antimicrobial activity of chloramphenicol (CAP) and extend ocular retention, chitosan nanoparticles carrying chloramphenicol (CAP-CNPs) were prepared by ionotropic gelation technique and further incorporated into thermosensitive in situ gel (CAP-CNPs-Gel). The formulation of the thermosensitive gel was optimized using a central composite design-response surface methodology (CCD-RSM). The optimized CAP-CNPs-Gel was selected to investigate its drug release kinetics, antimicrobial activity, and ocular irritancy. The average particle size and zeta potential of the optimized CAP-CNPs were 137.5 ± 3.6 nm and 29.9 ± 0.3 mV, respectively. The gel matrix was found to be most effective with a combination of 20.5% (w/v) poloxamer 407 and 5.0% (w/v) poloxamer 188. The CAP-CNPs-Gel formulation demonstrated sustained drug release properties, with drug release lasting up to 18 h. Furthermore, CAP-CNPs-Gel exhibited superior antibacterial activity when compared to both CAP eye drops and CAP gel. Through in vivo studies, CAP-CNPs-Gel was shown to be well tolerated by the eyes and capable of prolonging precorneal retention time for up to 40 min. These promising results suggest that CAP-CNPs-Gel has the potential to serve as a reliable treatment for eye infections. Moreover, the thermosensitive nanoparticulate gelling system utilized in this study provides a valuable carrier for effective CAP delivery.

Abstract:Non-invasive drug delivery offers a safe treatment while improving patient compliance. However, due to the particular physiological structure of the ocular, long-term retention and sustained drug release of the drug delivery system is crucial. Herein, this study aimed to design mPEG-CS-modified flexible liposomes-reinforced thermosensitive sol-gel reversible hydrogels (mPEG-CS-FL-TSG) for the delivery of astragaloside IV (AS-IV) and tetramethylpyrazine (TMP) to treat age-related macular degeneration. In vitro biological properties of mPEG-CS-FL and mPEG-CS-FL-TSG showed that they could be successfully taken up by ARPE-19 cells, and the uptake rate of mPEG-CS-FL-TSG was higher. Not only that, the release rate of mPEG-CS-FL-TSG was slower. More significantly, the results showed that the cytotoxicity of mPEG-CS-FL-TSG was lower than that of mPEG-CS-FL. In vivo result revealed that the drug delivery system could prominently enhance the ocular bioavailability of AS-IV and TMP, which is the enhanced synergism of well-permeable liposome and slow-releasing hydrogel. In summary, the mPEG-CS-FL-TSG can compensate for the short retention time and sudden release of liposome, as well as the low drug penetration of hydrogel, in order to show great promise in the non-invasive delivery of multiple drugs for the treatment of posterior ocular diseases.

Abstract:The aim of this study was to prepare and investigate the physical properties of a thermosensitive crosslinked chitosan pregel solution, and evaluate the in vitro release profiles of macromolecules from this sol-gel transition system. Chitosan and poly (vinyl alcohol) were used to form an interpenetrating polymeric network with glutaraldehyde as the crosslinker, and glycerophosphate (GP) was added to transform the pH-dependent solutions into thermosensitive pH-dependent solutions. Rheological study showed that the gelation was dependent on the crosslink degree and GP concentration of the solution. The crosslinked gel had excellent mechanic properties and no apparent "pores" and formed an integrated hydrogel texture according to scanning electronic micrograph. Gas chromatography test guaranteed the medication safety with no detection of glutaraldehyde remnants in the hydrogels. In vitro release study showed that the gelation does not significantly affect the macromolecules diffusion but the crosslinking degree does. These results indicated that the hydrogel formed an intensified three-dimensional hybrid network with interpenetrating molecules, which effectively buffered or delayed the macromolecules diffusion. The hydrogels sustained the drug release over 30 days and could be potentially used as in situ gelling implants. (c) 2006 Wiley Periodicals, Inc.

Abstract:Thermosensitive hydrogels promise to be the injectable implants for long-term controlled drug release; however, body response to the implanted hydrogels and its unpredictable impacts on drug release complicates their applications. In the present study, hydrophilic polymer poly(vinyl alcohol) (PVA) was blended into the thermosensitive hydrogel composed of chitosan and glycerophosphate to mitigate the body responses and promote the drug bioavailability. The effects of PVA on the surface properties of the hydrogel were evaluated by zeta-potential, water contact angle, and cell attachment. Body responses were explored by histological examination via subcutaneously implanting the hydrogels into Sprague-Dawley rats. Drug release in vivo and bioavailability were determined with cyclosporine A (CsA) employed as the model drug. The results showed that, on one hand, the presence of PVA improved the surface hydrophilicity of the hydrogel and inhibited the cell attachment on the hydrogel, which alleviated the further cell infiltration and tissue integration in body; and on the other hand, blending of PVA led to the more rapid gel formation and more compact network, which resisted the dehydration and survived the hydrogel from cell division. Abstract:For improving the effectiveness of cancer chemotherapy and avoiding rapid clearance of solid lipid nonoparticles (SLN) from the systemic circulation following systemic administration, 2-methoxyestradiol (2-ME) as model drug, PLGA-PEG-PLGA as hydrogel material, an injectable SLN loaded hydrogel was developed. Integrity of SLN within and released from the hydrogel was confirmed by direct visualization by a scanning electron microscope (SEM), particle size measurement by laser light scattering, and free drug concentration in the release medium by ultracentrifugation. Moreover, in vitro release, thermo-sensitive properties and rheological behavior were investigated. The results indicated that SLN were stable in the hydrogel. In the release medium, most 2-ME existed in the SLN and intact 2-ME SLN could be released from the hydrogel for a prolonged period over 46 days. Their concentration showed a significant effect on the release rate, in contrast to particle size and pH value of the release medium. In addition, the SLN loaded hydrogel could still exhibit reversible thermo-sensitive properties and better syringeability. These results suggested that the SLN loaded hydrogel could transport SLN to the target site and control prolonged release of SLN, which may increase the efficacy of cancer chemotherapy.

Abstract:Chitosan-grafted-mPEG(CS-mPEG) was obtained by modifying chitosan with methoxy poly(ethylene glycol)(mPEG) to improve the solubility and hydrophilicity of the material in neutral solutions.The CS-mPEG thermosensitive hydrogel exhibits better hydrophilicity with potential as the injectable polymer matrix for the delivery of biomolecules.Optimization of the modification method gave a CS-mPEG with a PEG-substitution degree of 21% to 25% with good water solubility and reversible thermosensibiliy.The sol-gel transformation finished within 10 min at 37 ℃.The in vitro release profiles show that release of the model biomolecular drug lysozyme(Lys) and the model chemical drug naltrexone(NTX) are well controlled from the CS-PEG hydrogel.Since the hydrophilic mPEG chain prevents inactivation of the biomolecules,the release rate of active lysozyme is markably increased in the chitosan-g-PEG hydrogel compared to the chitosan hydrogel.

Abstract:Chemotherapeutic agent gemcitabine is generally used in clinic by continuous intravenous infusion due to its short half life ,resulting in serious side effects .Gemcitabine-loaded thermosensitive hydrogel was prepared and evaluated with respect to both in vitro drug release and rheology and in vivo anti-tumor efficacy and systemic toxicity .The results show that gemcitabine-loaded thermosensitive hydrogel can release drug at a slow and steady rate ,improved in vivo tumor growth inhibition and alleviated systemic toxicity ,indicating good potential of thermosensitive hydrogel for sustained-release administration of gemcitabine for lo-cal chemotherapy .

Abstract:OBJECTIVE:To develop and study the properties of ion-exchange polyvinyl alcohol-acrylic acid microspheres (PVA-AA-Ms) for embolization.METHODS:The PVA-AA-Ms were produced by the method of inverse suspension polymerization. The morphology and particle size were determined by optical microscope; FT-IR was used to investigate the special functional groups of PVA-AA-Ms; the carboxyl content of PVA-AA-Ms was measured by chemical titration; the compression elasticity was examined by texture analyzer (TA-plus). Pingyangmycin (bleomycin A(5)) was used as model drug to prepare drug-loaded PVA-AA-Ms. Drug loading and entrapment efficiency were measured through UV-spectrophotometer; in vitro drug release characteristic was detected by constant temperature vibration dialysis assay.RESULTS:The PVA-AA-Ms were round and integrated. The average diameter of PVA-AA-Ms was 500 microm with a range of 150-1 000 microm. The carboxyl vibration was demonstrated by FT-IR and the content of carboxyl was 8.905 mmol/g. PVA-AA-Ms were mechanically stable with appropriate elasticity. Drug loading and entrapment efficiency were 30 g/L and 99.4%, respectively. The drug release rate was slow in phosphate buffer solution (PBS), 88.3% of the drug was released after 24 h and the t(50) was 2.19 h.CONCLUSION:PVA-AA-Ms prepared in this study were supposed to be suitable for clinical embolization according to the physicochemical properties. The high carboxyl content of PVA-AA-Ms which allowed them to load cationic drugs (e.g., drug with amino group) through ion-exchange mechanism brought broad prospects for combination of embolization and chemotherapy.

Abstract:A novel copolymer, poly(N-isopropylacrylamide)-chitosan (PNIPAAm-CS), was investigated for its thermosensitive in situ gel-forming properties and potential utilization for ocular drug delivery. The thermal sensitivity and low critical solution temperature (LCST) were determined by the cloud point method. PNIPAAm-CS had a LCST of 32 degrees C, which is close to the surface temperature of the eye. The in vivo ocular pharmacokinetics of timolol maleate in PNIPAAm-CS solution were evaluated and compared to that in conventional eye drop solution by using rabbits according to the microdialysis method. The C(max) of timolol maleate in aqueous fluid for the PNIPAAm-CS solution was 11.2 microg/ml, which is two-fold higher than that of the conventional eye drop, along with greater AUC. Furthermore, the PNIPAAm-CS gel-forming solution of timolol maleate had a stronger capacity to reduce the intra-ocular pressure (IOP) than that of the conventional eye drop of same concentration over a period of 12 h. In addition, the MTT assay showed that there is little cytotoxicity of PNIPAAm-CS at concentration range of 0.5-400 microg/ml. These results suggest that PNIPAAm-CS is a potential thermosensitive in situ gel-forming material for ocular drug delivery, and it may improve the bio-availability, efficacy, and compliance of some eye drugs.

Abstract:Journal of Biomaterials Applications, Ahead of Print. Due to the heterogeneity and the complexity of the tumor microenvironment, combination therapy, especially the combination of chemotherapy and photothermal therapy (PTT), had received increasing attention. However, the co-delivery of small molecule drugs for chemotherapy and photothermal agents was a key issue. Herein, we prepared a novel thermo-sensitive hydrogel loading with elemene (ELE)-loaded and nano graphene oxide (NGO)-based liposomes for enhanced combined therapy. ELE was applied as the model drug for chemotherapy because it was a natural sesquiterpene drug with broad-spectrum and efficient antitumor activity. NGO was applied as drug carrier and photothermal agent simultaneously due to its two-dimensional structure and high photo-thermal conversion efficacy. NGO was further modified with glycyrrhetinic acid (GA) to improve its water dispersion, biocompatibility and tumor-targeting ability. ELE was loaded by GA-modified NGO (GA/NGO) to prepare the liposomes designated as ELE-GA/NGO-Lip, which was further mixed with chitosan (CS) solution and β-glycerin sodium phosphate (β-GP) solution to prepare the thermo-sensitive hydrogel designated as ELE-GA/NGO-Lip-gel. The obtained ELE-GA/NGO-Lip-gel had the gelling temperature of 37°C, temperature and pH-response gel dissolution and high photo-thermal conversion effect. More importantly, ELE-GA/NGO-Lip-gel upon 808 nm laser irradiation had relative high anti-tumor efficiency against SMMC-7721 cells in vitro. This research might provide a potent platform for the application of thermos-sensitive injectable hydrogel in combined tumor therapy.

Abstract:The auto-gelling and drug release properties of the thermosensitive chitosan-β-glycerophosphate formulation were investigated. According to rheological study, gelation lag time of chitosan/β-glycerophosphate (GP)solutions varied from 2 to 60min with different deacetylation degree of chitosan, pH, gelation temperature, and the particles in the sol. The gelation properties were also found to influence the release profiles of a hydrophilic drug,5-fluorouracil (5-FU). Morphological examination by scanning electron microphotography demonstrated that large "pores" occurred during the gel-forming process, which created hydrophilic environment and led to the rapid initial release of the drug (85% in first 8h). Poly-3-hydroxybutyrate (PHB), a biodegradable material, was applied here as scaffold to capture 5-FU into microparticles with high encapsulation efficiency by solvent-nonsolvent method.Combination of these microparticles into the chitosan-β-GP formulation could drop the rapid initial release from 85% down to 29% in the optimized PHB content (75%, by mass). The release could sustain for about 10 months.This study provided an understanding of the potential of injectable implant using thermosensitive chitosan-β-GP formulation containing PHB based particles for the water soluble drugs that need the property of long-term delivery.

Abstract:Biodegradable in situ thermo-sensitive hydrogels have potential applications in ophthalmic pharmaceutical preparations. Herein, a thermo-gel of PNICL-PEG-PNICL loaded with the natural compound epigallocatechin gallate (EGCG) used as a drug sustained release carrier was prepared by block bending strategy. Compared to mono polymers that cannot form a gel, mixing two block copolymers through a block blending strategy allows for the preparation of thermo-sensitive hydrogels. The temperature sensitive behavior of the system can be easily regulated by varying the blending ratios of the two polymers. Furthermore, the internal structure of the gel was investigated that indicated the system formed a permeable network through micelle aggregation. The biosafety of the system and its ability to sustainably release EGCG at 37 o C for 240 h were preliminarily demonstrated in vitro . This study has revealed the internal structural changes of block blending thermo-gels and tentatively justified their potential as drug carriers.

Abstract:The polysaccharide-based pH-responsive compounds, namely, N,N,N-trimethyl chitosan (TMC), polyethylene glycolated hyaluronic acid (PEG-HA), and polysaccharide-based nano-conjugate of hyaluronic acid, chitosan oligosaccharide and alanine [HA-Ala-Chito(oligo)] were chemically synthesized using biopolymers chitosan and hyaluronic acid, and applied here to observe the changes in morphology, pH-stability, mechanical and drug-release behavior, and cytotoxicity of thermo-responsive polymer: Poloxamer 407 (PF127)-based drug delivery systems for traditional Chinese medicine Cortex Moutan (CM). The thermo-responsive hydrogel of PF127 loaded with CM (GelC) was used as control. The dual-responsive (pH/temperature) hydrogels: PF127/TMC/PEG-HA (Gel1) and PF127/HA-Ala-Chito(oligo) (Gel2) showed improved mechanical behavior as obtained by rheology and mechanical agitation study, and pH-stability under various external pH conditions, and those improvements occurred due to the addition of polysaccharide-based pH-responsive compounds in the systems. Both, Gel1 and Gel2 showed better morphology than GelC as obtained by SEM or TEM suggesting that interaction of polysaccharide-based pH-responsive compounds with PF127 in either gel or sol state gave better porous network structure in the hydrogels or more dispersed micellar arrangements in sol-state, respectively. Gel1 showed the highest cumulative drug release (86.5%) after 5 days under mild acidic condition (pH 6.4) suggesting that release behavior of a hydrogel drug carrier was dependent on morphology, mechanical behavior, and pH-stability. The transdermal release (ex-vivo) results indicated that gallic acid, the active marker of CM passed through porcine ear skin and all the formulations showed more or less similar transdermal release properties. The hydrogels loaded with CM showed no cytotoxicity (cell viability >90.0%) on human HaCaT keratinocytes within concentration range of 0.0-20.0 μg/ml as obtained by MTT assay, and cell viability was more than 100% at a concentration of 20.0 μg/ml for Gel2. The formulations without loaded drug namely, Gel1-CM and Gel2-CM exhibited strong anti-bacterial action against gram positive bacteria Staphylococcus aureus.
