Abstract:With the rapid developments of food industry and the improvement of living condi-tions,the phenomenon of economically motivated adulteration( EMA)is an extremely issue of concern due to people’s increasing requirements for food safety. Nowadays,the major EMA incidents include substitution of varieties,mislabeling geographical origin,interfusion of poor quality,incorporation of inferiority, etc. Therefore, an efficient authentication technology needs to be established. Metabolomics is defined as the quantitative and qualitative analyses of large quantity of metabolites as a whole in a given sample,which has huge advantages in non-targeted analysis. Metabolomics has been adopted extensively by both domestic and abroad researchers to study the food safety problems,which provided an alternative research tool to identify the EMA incidents. In this paper,the progress and applications of metabolomics in identifying food varieties,geographical origin,quality and counterfeits have been reviewed, which provide technical support to ensure food quality and safety and protect the interests of consumers.
Mislabeling Identification of Fresh Retail Beef Cuts Using Machine Learning – Guided REIMS Lipidomic Fingerprints

Abstract:Fraudulent activities such as mislabeling of beef occurred frequently. In this study, a fast and reliable methodology for specific discrimination of fresh retail beef cuts (ribeye, striploin, brisket, beef shank, and fore shank) was established based on electrosurgical knife - rapid evaporative ionization mass spectrometry (iKnife-REIMS) technique integrated with machine learning (ML). REIMS lipidomic fingerprints of various beef cuts were characterized by ML models to explore meaningful information. Authenticity identification models were built by main ML algorithms including discriminant analysis (DA), support vector machines (SVM), and k-nearest neighbor (KNN) to realize the discrimination of mislabeling in beef cuts. Mislabeling discrimination rates of DA, SVM, and KNN were 95.54%, 99.91%, and 100%, respectively. Model performances were evaluated by confusion matrix, receiver operating characteristic curve, and area under the curve. The validation of the proposed REIMS method for detecting mislabeled beef cuts with an accuracy of 96.80% was performed. Results demonstrated that this artificial intelligent method coupled to ML – guided REIMS analysis was efficient for authenticity detection of mislabeled beef cuts.
Fast discriminating of purity on minced mutton using electronic tongue

Abstract:Cheaper animal protein, such as Chicken as an example, has been fraudulently used as a substitute for more expensive animal proteins, like mutton and beef. The adulteration of mutton has attracted increasing attention. It requires reliable methods for the authentication of mutton adulteration. An electronic tongue with chemically modified field-effect-transistor sensors was employed to analysis the adulteration of chicken in minced mutton. The effects of sample weight on the sensor responses of electronic tongue were studied at three different extraction solutions. Analysis of variance found that the sample weight affected the responses of the sensor significantly. With the help of Principle component analysis (PCA), the optimum experimental parameters were acquired:15 g sample extracted by 100 mL KCl solution. The adulterated mutton was made by mixing mutton with chicken at levels of 0, 20%, 40%, 60%, 80%, and 100% by weight, respectively. With the optimum experimental parameters, 168 samples of adulterated mutton were detected, and the signals were analyzed by pattern recognition techniques to build models for classification of adulterated mutton with different content of chicken, and prediction of the content of chicken in minced mutton. With PCA, the adulterated mutton samples were grouped according to their content of chicken with good classification results, except that samples containing 80%and 100%chicken partially overlapped with each other. Better classification results were found when canonical discriminant analysis (CDA) was employed, as samples containing 80% and 100% chicken were clearly grouped and separated. Multiple linear regression (MLR) and Partial least square analysis (PLS) were employed to build the predictive model for the content of chicken adulterated into minced mutton. Both models could predict the adulteration with a high determination coefficient as high as 0.9925 and 0.9923, respectively. MLR was more effective for the prediction of chicken content. The E-nose proved to be a useful authentication method for meat adulteration detection for its efficiency and high accuracy.

Abstract:The immune system of any organism must maintain a fine balance between activation and inhibition. It must possess adequate reactivity to raise an effective immune response to target non-self molecules while not harming the organism itself. Essential to this process is the ability to control the timing and place of activation and to limit the extent of, and eventually terminate, activation. Failure to maintain this balance will result in either immunodeficiency or autoimmunity. Inhibitory receptors are involved in this regulation, a number having been shown to be critical in controlling the B cell immune response. There are two broad classes of inhibitory receptor most are of the immunoglobulin (Ig) superfamily while the remainder are lectin-like molecules. They share a number of structural and functional similarities. Each inhibitory receptor contains one or more immunoreceptor tyrosine-based inhibitory motifs (ITIMs) within its cytoplasmic domain essential for generation and transduction of inhibitory signals. The ITIM consists of a six amino acid consensus sequence (Ile/Val/Leu/Ser)-X-Tyr-X-X(Leu/Val). Ligation of the inhibitory receptor to an immunoreceptor tyrosine-based activatory motif (ITAM)-containing activatory molecule results in tyrosine kinase phosphorylation of the tyrosine residue within the ITIM by lyn. Tyrosine phosphorylation of the ITIM allows it to bind and activate phosphatases containing an src homology 2 (SH2) domain. Two classes of SH2-containing inhibitory phosphatases have been identified: the protein tyrosine phosphatases SHP-1 and SHP-2, and the phosphoinositol phosphatases SHIP and SHIP2. These classes have separate downstream signalling pathways through which they modulate cellular inhibition. In general, each class of phosphatase interacts with the ITIMs of different inhibitory receptors but each inhibitory receptor appears to act predominantly through only one class of phosphatase. A number of inhibitory receptors have been described on B cells, the details of which are summarized in Fig. 1 and Table 1. We will concentrate on three of these, FcgRII, CD22 and PD 1, and in addition will discuss lyn, SHP-1 and SHIP, which are crucial elements in the signalling pathways of the inhibitory receptors. We will describe their probable physiological roles in immune regulation, and then review the evidence from knockout mice, spontaneous mouse models of autoimmunity and human disease that defective regulation by B cell inhibitory receptors can lead to autoimmunity.

Abstract:In recent years, meat authenticity awareness has increased and, in the fight to combat meat fraud, various analytical methods have been proposed and subsequently evaluated. Although these methods have shown the potential to detect low levels of adulteration with high reliability, they are destructive, time-consuming, labour-intensive, and expensive. Therefore, rendering them inappropriate for rapid analysis and early detection, particularly under the fast-paced production and processing environment of the meat industry. However, modern analytical methods could improve this process as the food industry moves towards methods that are non-destructive, non-invasive, simple, and on-line. This review investigates the feasibility of different non-destructive techniques used for processed meat authentication which could provide the meat industry with reliable and accurate real-time monitoring, in the near future.

Abstract:The measurement of the ratio of 228Th vs 228Ra, two isotopes from the natural radioactivity, is an ancient, but not entirely reliable, method of dating biological materials. It has been applied for the dating of fishes with mitigated results in many cases. It has been also experimented with mixed success in forensics cases where the question of how long a person whose bones were found was dead. Here too, the precision of the results is not satisfactory. Other applications concern age dating oil and gas waste waters spills. Our goal, while based on the same measurement of the ratio of the two radiosiotopes, is different. We aim at assessing the age of an animal at the date of its death, in particular for the animals intended for human consumption. The measurement technique relies on gamma spectrometry of the bones of 228Th and 228Ra stemming from ingested food and from the environment. A strong incentive to our research is fraudulent sales of meat with falsified identification. For instance, mutton sold as lamb, which is more expensive. Depending on the country of origin, this is complicated by varying legal definitions of the animal category, which renders controls for imported meat difficult. Preliminary measurements with retail samples of approximate identification validate the procedure. Further and more precise inquiries in a collaborative work with the French Ministry's Anti-Fraud Agency (equivalent of the U.S. Food and Drug Administration) are on their way.

Abstract:<p>Increasing accounts of fraud and persistent labeling problems have brought the authenticity of leather products into question. In this study, we developed an extremely simplified workflow for real-time, <em>in situ</em>, and unambiguous authentication of leather samples using rapid evaporative ionization mass spectrometry (REIMS) coupled with an electric soldering iron. Initially, authentic leather samples from cattle, sheep, pig, deer, ostrich, crocodile, and snake were used to create a chemometric model based on principal component analysis and linear discriminant analysis algorithms. The validated multivariate statistical model was then used to analyze and generate live classifications of commercial leather samples. In addition to REIMS analysis, the microstructures of leathers were characterized by scanning electron microscopy to provide complementary information. The current study is expected to provide a high-throughput tool with superior efficiency and accuracy for authenticating the identity of leathers and other consumer products of biogenic origin.</p>

Rapid visual sensing and quantitative identification of duck meat in adulterated beef with a lateral flow strip platform

Abstract:A novel high-sensitivity authentication method has been demonstrated for the rapid visual detection of adulterated meat based on both the lateral flow strip (LFS) platform and on polymerase chain reaction (PCR). After the rapid extraction of genomic components from meat, the on-site amplification of the target DNA of adulterated duck meat is carried out with the rationally designed functional FITC- and biotin-modified primer set, thereby producing numerous double-stranded DNA (dsDNA) products dually labelled with FITC and biotin. The FITC-labelled terminal end of the products binds to the pre-immobilized FITC antibody on the test line of the strip, and the biotin-labelled terminal end binds to the streptavidin-conjugated gold nanoparticles, resulting in a visible test line on the LFS for the rapid identification of duck meat in adulterated beef. After optimization, an adulteration ratio as low as 0.05% can be easily measured, which is more sensitive than other common adulteration authentication methods and is even comparable to instrumental methods. Moreover, 22 commercial processed meat samples were tested with this new strategy, and 4 adulterated samples were successfully identified by both the classic method and our method. In essence, the present authentication method is simple in design, convenient in operation, and can be easily extended to the identification of other adulteration components just by replacing the modified primers.

Abstract:The "no controversy" status of the important role of nutrition in maintaining good health and disease management is becoming a gold standard in medical practice. Medical schools in developed countries and some in developing countries are beginning to renew interest in nutrition education for medical professionals. Despite difficulties envisaged in modifying the medical school curricula, it is inevitable that medical professionals need some basic nutrition knowledge appropriate for medical counselling in disease prevention and management. Cost effectiveness in providing health care services is now an important policy tool, which emphasizes preventive medical care. This is dependent on good nutrition education, which informs not only on food but healthcare and good environment. This article examines the need for nutrition education in medical schools, approaches that can be used to introduce nutrition education, using the available information from medical schools that have already integrated nutrition into their medical education curricula. It also identifies some of the barriers and strategies to overcome including specific actions for nutrition educators or whoever may be saddled with the responsibilities for initiating the development of nutrition curriculum for medical education.

Abstract:There is a growing enthusiasm among consumers for high value lamb, especially the lamb with certain geographical indications, which may result in possible fraudulent labeling due to the opportunity for large economic profits. On such a basis, the aim of this study was to improve the discrimination rate of geographical traceability of lamb. To achieve this objective, lamb samples obtained from four different regions from China and New Zealand were analyzed. The chemical profiles of lamb samples from different origins were characterized by stable isotope analysis, multi-element determination, and fatty-acid profile, and further subjected to a series of statistical analyses, including principal component analysis, hierarchical cluster analysis, and principal component analysis-linear discriminant analysis. Compared with the result obtained by using each method individually, data fusion by ratio modulation of multi-technique clearly improved the discrimination performance for origin traceability. A satisfactory discrimination rate of 100% was obtained after ratio modulation of these techniques. Comprehensively, this study offers proof of concept for a promising method that could be used for lamb provenance authentication and fraud detection.

Abstract:Muscle foods, valued for their significant nutrient content such as high-quality protein, vitamins, and minerals, are vulnerable to adulteration and fraud, stemming from dishonest vendor practices and insufficient market oversight. Traditional analytical methods, often limited to laboratory-scale., may not effectively detect adulteration and fraud in complex applications. Raman spectroscopy (RS), encompassing techniques like Surface-enhanced RS (SERS), Dispersive RS (DRS), Fourier transform RS (FTRS), Resonance Raman spectroscopy (RRS), and Spatially offset RS (SORS) combined with chemometrics, presents a potent approach for both qualitative and quantitative analysis of muscle food adulteration. This technology is characterized by its efficiency, rapidity, and noninvasive nature. This paper systematically summarizes and comparatively analyzes RS technology principles, emphasizing its practicality and efficacy in detecting muscle food adulteration and fraud when combined with chemometrics. The paper also discusses the existing challenges and future prospects in this field, providing essential insights for reviews and scientific research in related fields.

Abstract:Meat adulteration have become a global issue, which has increasingly raised concerns due to not only economic losses and religious issues, but also public safety and its negative effects on human health. Using optimal primers for seven target species, a multiplex PCR method was developed for the molecular authentication of camel, cattle, dog, pig, chicken, sheep and duck in one tube reaction. Species-specific amplification from the premixed total DNA of seven species was corroborated by DNA sequencing. The limit of detection (LOD) is as low as 0.025 ng DNA for the simultaneous identification of seven species in both raw and heat-processed meat or target meat: as little as 0.1% (w/w) of the total meat weight. This method is strongly reproducible even while exposed to intensively heat-processed meat and meat mixtures, which renders it able to trace meat origins in real-world foodstuffs based on the authenticity assessment of commercial meat samples. Therefore, this method is a powerful tool for the inspection of meat adulterants and has broad application prospects.

Abstract:In recent years, the problem of food adulteration has become increasingly rampant, seriously hindering the development of food production, consumption, and management. The common analytical methods used to determine food authenticity present challenges, such as complicated analysis processes and time-consuming procedures, necessitating the development of rapid, efficient analysis technology for food authentication. Spectroscopic techniques, ambient ionization mass spectrometry (AIMS), electronic sensors, and DNA-based technology have gradually been applied for food authentication due to advantages such as rapid analysis and simple operation. This paper summarizes the current research on rapid food authenticity analysis technology from three perspectives, including breeds or species determination, quality fraud detection, and geographical origin identification, and introduces chemometrics method adapted to rapid analysis techniques. It aims to promote the development of rapid analysis technology in the food authenticity field.

Abstract:Fraud occurs frequently in the fishery and aquatic product markets. Therefore, the authentication of fishery and aquatic products has become a crucial issue. Recently, elemental and stable isotope analyses have become popular for aquatic food authentication. In this review, relevant literature was collected to highlight the current progress in the authentication of fishery and aquatic products using multi‐element and stable isotope analyses combined with chemometrics. Both technologies provide valuable insight regarding geographic origin, culture method, and species identification of fishery and aquatic products. Their classification rate varied from 60% to 100% in most studies. The formation of both elemental and stable isotope profiles of fishery and aquatic products is complex and affected by many factors, including diet, biological differences, and culture environment. The classification rate of these methods may be increased by combining multiple technologies, selecting the correct elements and stable isotopes for specific purposes, selecting and verifying proper discrimination methods, and increasing sample sizes. This review provides valuable information to build reliable and stable tracing methods for fishery and aquatic products.

Abstract:Nowadays, stable isotope technology, DNA technology, spectroscopic technology, volatilomics technology, metabolomic analysis, mineral elements analysis and fatty acid analysis have been used to trace the origin of beef. Stable isotope technique is often combined with mineral elements analysis. Both of them have the advantages of high accuracy and low detection limit. Stable isotopes may be affected by geography and dietary history. The mineral elements analysis has high requirements for experimental operation. DNA technology is not affected by geography and dietary history, so it has unique advantages. It faces the problem of selecting molecular markers. Spectroscopic technology and volatilomics technology based on electronic nose do not require sample pretreatment. The limitation is that infrared and Raman are insensitive, and the data processing of hyperspectral image is difficult. Raman is much less sensitive than Infrared. The use of electronic nose has higher requirements for environment. Volatilomics technology based on mass spectrometry (MS), metabolomic analysis and fatty acid analysis have been used to analyze organic components. They can provide comprehensive information, but they have higher requirements for sample preservation. Metabolomic analysis equipment is expensive. The research on traceability and authenticity of beef can help to ensure the security and quality of beef.

Abstract:Because of the increased demand for processed meat, there is an urgent need to introduce specific identification methods. Strategies such as molecular genetics and the physical condition of meat are used to quickly explore multi-component products. However, a single methodology does not always unambiguously classify a product as counterfeit. In laboratory practice, as a rule, screening techniques are rarely used in the first stage, followed by arbitration. This work aimed to study individual methodologies using artificially falsified meat samples as examples and to identify their composition based on muscle tissue. For the experiments, the three most common types of raw meat were selected: pork, beef, and chicken. The calculation of the content of muscle tissue was carried out according to the BEFFE method. The study of muscle protein was carried out by ICA, ELISA, PCR, microstructural analysis, and mass spectrometric identification. In this connection, we proposed a multilevel control system for multicomponent meat products. Both classical methodologies, such as calculation by prescription bookmarks (BEFFE) and microstructural analysis, and approaches of highly sensitive methodologies, such as identification of muscle tissue by marker peptides (LC/MS-MRM) and semi-quantitative PCR analysis, were evaluated.

Abstract:Epimedium brevicornum Maxim is a famous medicinal herb which has been widely used for the treatment of impotence, infertility, osteoporosis, cardiovascular diseases, amnesia, and senile functional diseases. A dual-mode high-speed counter-current chromatographic method is developed for the separation and purification of four bioactive flavonoids from the medicinal herb Epimedium brevicornum Maxim. The crude flavonoids are obtained by extraction with ethyl acetate and ethanol from the dried aerial parts of Epimedium brevicornum Maxim under sonication. High-speed counter-current chromatography with a two-phase solvent system composed of n-butanol-ethyl acetate-water (3:7:10, v/v) is performed using a dual-mode method, which yields 3.2 mg of epimedin A at a purity of 98.2%, 5.5 mg of epimedin B at a purity of 92.6%, 12.7 mg of epimedin C at a purity of 90.4%, and 42.9 mg of icariin at a purity of 96.8% based on the HPLC analysis. The recoveries are 95.2%, 89.4%, 91.1%, and 94.8%, respectively. The isolation of these flavonoids in adequate amounts makes them readily available for structure-activity relationship studies and for quality control of the herbal medicine.

Abstract:Meat species identification and animal authentication in meat products is a significant subject, attention to which would contribute to fair-trade, and would enable consumers to make informed choices. Analytical methods are often based on protein or DNA measurements. Methods based on protein fractions include electrophoretic, chromatographic and immunological techniques and are often not suitable for compound food products, nor are they sensitive in processed products to differentiate closely related meat species. Advances in DNA technology have led to the rapid development of alternative approaches to species identification. Recently, application of polymerase chain reaction in food analysis has increased in the light of their simplicity, specificity and sensitivity. This review discusses a wide range of analytical methods with a focus on their ability to quantify meat and authentication of meat products.

Simultaneous identification of animal-derived components in meats using high-throughput sequencing in combination with a custom-built mitochondrial genome database

Abstract:Abstract Currently, the inspection and supervision of animal ingredients relies primarily upon specific amplification-dependent methods, whose efficiency and accuracy are being seriously challenged by the increasing diversity and complexity of meat products. High-throughput sequencing (HTS) technology was employed to develop an alternative method to detect animal-derived ingredients in meat products. A custom-built database containing 2,354 complete mitochondrial genomic sequences from animals, an identification analysis pipeline based on short-sequence alignment, and a web-based server were built to facilitate this detection. The entire process, including DNA extraction, gene amplification, and sequencing, was established and optimized for both marker gene (part of the CYTB gene)-based detection and total DNA-based detection. Using simulated samples containing various levels of pig, cattle, sheep, chicken, rabbit, and mice ingredients, the detection capability and accuracy of this method were investigated. The results of this study indicated that the method is capable of detecting animal components in meats that are present at levels as low as 1%. Our method was then tested using 28 batches of real meat products such as raw meat slices, raw meat mince, cooked dried meat, cooked meat sausage, and other supermarket samples, with a traditional qPCR method as the control. The results demonstrated an accuracy of 97.65% for the qualitative detection method, which indicate that the developed method is reliable for the detection of animal components. The method is also effective for the identification of unknown food samples containing mixed animal components, which suggests a good future in application.
