Abstract:Beef is one of the favourite meats consumed by people worldwide due to its high nutrition value needed by human development. It is highly susceptible for the adulteration practice by substituting beef with lower price meats by unethical meat traders due to the economic reasons. Therefore, the authenticity of beef meat (BM) is important because it is also related to halal status of meat which is required for certain religions. This research aimed to differentiate metabolites of BM from different origins using liquid chromatography-high resolution mass spectrometry (LC-HRMS) combined with chemometrics for the authentication purposes. Various metabolites mostly amino acids and lipids could be detected using methanol extraction. Principal component analysis (PCA), partial least square-discriminant analysis (PLS-DA) and sparse PLS-DA were successfully used to discriminate BMs from different origins. Fifty potential metabolite markers which are important for discrimination have been identified using variable importance projection (VIP) value extracted from PLS-DA analysis. Metabolites of (4S)-4-{[(9Z)-3-Hydroxy-9-hexadecenoyl]oxy}-4-(trimethylammonio)butanoate, N,N-Diisopropylethylamine (DIPEA), D-sphingosine, (2E,4Z)-N-Isobutyl-2,4-octadecadienamide, 1-(14-methylhexadecanoyl)pyrrolidine, linoleic acid, 12-HAS, dodecylamine1, myristamide, and tributyl phosphate had high responsibility in discriminating BMs from different origins (VIP value > 2.0). It can be concluded that LC-HRMS based untargeted metabolomics combined with chemometrics could be used for authentication of BMs from different regions.

Abstract:With the rapid developments of food industry and the improvement of living condi-tions,the phenomenon of economically motivated adulteration( EMA)is an extremely issue of concern due to people’s increasing requirements for food safety. Nowadays,the major EMA incidents include substitution of varieties,mislabeling geographical origin,interfusion of poor quality,incorporation of inferiority, etc. Therefore, an efficient authentication technology needs to be established. Metabolomics is defined as the quantitative and qualitative analyses of large quantity of metabolites as a whole in a given sample,which has huge advantages in non-targeted analysis. Metabolomics has been adopted extensively by both domestic and abroad researchers to study the food safety problems,which provided an alternative research tool to identify the EMA incidents. In this paper,the progress and applications of metabolomics in identifying food varieties,geographical origin,quality and counterfeits have been reviewed, which provide technical support to ensure food quality and safety and protect the interests of consumers.

Abstract:The immune system of any organism must maintain a fine balance between activation and inhibition. It must possess adequate reactivity to raise an effective immune response to target non-self molecules while not harming the organism itself. Essential to this process is the ability to control the timing and place of activation and to limit the extent of, and eventually terminate, activation. Failure to maintain this balance will result in either immunodeficiency or autoimmunity. Inhibitory receptors are involved in this regulation, a number having been shown to be critical in controlling the B cell immune response. There are two broad classes of inhibitory receptor most are of the immunoglobulin (Ig) superfamily while the remainder are lectin-like molecules. They share a number of structural and functional similarities. Each inhibitory receptor contains one or more immunoreceptor tyrosine-based inhibitory motifs (ITIMs) within its cytoplasmic domain essential for generation and transduction of inhibitory signals. The ITIM consists of a six amino acid consensus sequence (Ile/Val/Leu/Ser)-X-Tyr-X-X(Leu/Val). Ligation of the inhibitory receptor to an immunoreceptor tyrosine-based activatory motif (ITAM)-containing activatory molecule results in tyrosine kinase phosphorylation of the tyrosine residue within the ITIM by lyn. Tyrosine phosphorylation of the ITIM allows it to bind and activate phosphatases containing an src homology 2 (SH2) domain. Two classes of SH2-containing inhibitory phosphatases have been identified: the protein tyrosine phosphatases SHP-1 and SHP-2, and the phosphoinositol phosphatases SHIP and SHIP2. These classes have separate downstream signalling pathways through which they modulate cellular inhibition. In general, each class of phosphatase interacts with the ITIMs of different inhibitory receptors but each inhibitory receptor appears to act predominantly through only one class of phosphatase. A number of inhibitory receptors have been described on B cells, the details of which are summarized in Fig. 1 and Table 1. We will concentrate on three of these, FcgRII, CD22 and PD 1, and in addition will discuss lyn, SHP-1 and SHIP, which are crucial elements in the signalling pathways of the inhibitory receptors. We will describe their probable physiological roles in immune regulation, and then review the evidence from knockout mice, spontaneous mouse models of autoimmunity and human disease that defective regulation by B cell inhibitory receptors can lead to autoimmunity.
Non-invasive metabolomics biomarkers of production efficiency and beef carcass quality traits

Abstract:Abstract The inter-cattle growth variations stem from the interaction of many metabolic processes making animal selection difficult. We hypothesized that growth could be predicted using metabolomics. Urinary biomarkers of cattle feed efficiency were explored using mass spectrometry-based untargeted and targeted metabolomics. Feed intake and weight-gain was measured in steers (n = 75) on forage-based growing rations (stage-1, 84 days) followed by high-concentrate finishing rations (stage-2, 84 days). Urine from days 0, 21, 42, 63, and 83 in each stage were analyzed from steers with the greater (n = 14) and least (n = 14) average-daily-gain (ADG) and comparable dry-matter-intake (DMI; within 0.32 SD of the mean). Steers were slaughtered after stage-2. Adjusted fat-thickness and carcass-yield-grade increased in greater-ADG-cattle selected in stage-1, but carcass traits did not differ between ADG-selected in stage-2. Overall 85 untargeted metabolites segregated greater- and least-ADG animals, with overlap across diets (both stages) and breed type, despite sampling time effects. Total 18-bile acids (BAs) and 5-steroids were quantified and associated with performance and carcass quality across ADG-classification depending on the stage. Stepwise logistic regression of urinary BA and steroids had > 90% accuracy identifying efficient-ADG-steers. Urine metabolomics provides new insight into the physiological mechanisms and potential biomarkers for feed efficiency.

Abstract:Abstract This study presents the application of metabolomics to evaluate changes in the rumen metabolites of beef cattle fed with three different diet types: forage-rich, mixed and concentrate-rich. Rumen fluid samples were analysed by 1 H-NMR spectroscopy and the resulting spectra were used to characterise and compare metabolomic profiles between diet types and assess the potential for NMR metabolite signals to be used as proxies of methane emissions (CH 4 in g/kg DMI). The dataset available consisted of 128 measurements taken from 4 experiments with CH 4 measurements taken in respiration chambers. Predictive modelling of CH 4 was conducted by partial least squares (PLS) regression, fitting calibration models either using metabolite signals only as predictors or using metabolite signals as well as other diet and animal covariates (DMI, ME, weight, BW 0.75 , DMI/BW 0.75 ). Cross-validated R 2 were 0.57 and 0.70 for the two models respectively. The cattle offered the concentrate-rich diet showed increases in alanine, valerate, propionate, glucose, tyrosine, proline and isoleucine. Lower methane yield was associated with the concentrate-rich diet (p < 0.001). The results provided new insight into the relationship between rumen metabolites, CH 4 production and diets, as well as showing that metabolites alone have an acceptable association with the variation in CH 4 production from beef cattle.

Abstract:Nowadays, stable isotope technology, DNA technology, spectroscopic technology, volatilomics technology, metabolomic analysis, mineral elements analysis and fatty acid analysis have been used to trace the origin of beef. Stable isotope technique is often combined with mineral elements analysis. Both of them have the advantages of high accuracy and low detection limit. Stable isotopes may be affected by geography and dietary history. The mineral elements analysis has high requirements for experimental operation. DNA technology is not affected by geography and dietary history, so it has unique advantages. It faces the problem of selecting molecular markers. Spectroscopic technology and volatilomics technology based on electronic nose do not require sample pretreatment. The limitation is that infrared and Raman are insensitive, and the data processing of hyperspectral image is difficult. Raman is much less sensitive than Infrared. The use of electronic nose has higher requirements for environment. Volatilomics technology based on mass spectrometry (MS), metabolomic analysis and fatty acid analysis have been used to analyze organic components. They can provide comprehensive information, but they have higher requirements for sample preservation. Metabolomic analysis equipment is expensive. The research on traceability and authenticity of beef can help to ensure the security and quality of beef.

Abstract:The liquid chromatography mass spectrometry (LC-MS)-based metabolomic and lipidomic methodology has great sensitivity and can describe the fingerprint of metabolites and lipids in pork and beef. This approach is commonly used to identify and characterize small molecules such as metabolites and lipids, in meat products with high accuracy. Since the metabolites and lipids can be used as markers for many properties of a food, they can provide further evidence of the foods authenticity claim. Chromatography coupled to mass spectrometry is used to separate lipids and metabolites from meat samples. The research data usually is compared to lipid and metabolite databases and evaluated using multivariate statistics. LC-MS instruments directly connected to the metabolite and lipid databases software can be used to assess the authenticity of meat products. LC-MS has good selectivity and sensitivity for metabolomic and lipidomic analysis. This review highlighted the combination of metabolomics and lipidomics can be used as a reference for analyzing authentication meat products.

Identification of Predictive Biomarkers of Lameness in Transition Dairy Cows

Abstract:The aim of this study was to identify with a high level of confidence metabolites previously identified as predictors of lameness and understand their biological relevance by carrying out pathway analyses. For the dairy cattle sector, lameness is a major challenge with a large impact on animal welfare and farm economics. Understanding metabolic alterations during the transition period associated with lameness before the appearance of clinical signs may allow its early detection and risk prevention. The annotation with high confidence of metabolite predictors of lameness and the understanding of interactions between metabolism and immunity are crucial for a better understanding of this condition. Using liquid chromatography-tandem mass spectrometry (LC-MS/MS) with authentic standards to increase confidence in the putative annotations of metabolites previously determined as predictive for lameness in transition dairy cows, it was possible to identify cresol, valproic acid, and gluconolactone as L1, L2, and L1, respectively which are the highest levels of confidence in identification. The metabolite set enrichment analysis of biological pathways in which predictors of lameness are involved identified six significant pathways (p < 0.05). In comparison, over-representation analysis and topology analysis identified two significant pathways (p < 0.05). Overall, our LC-MS/MS analysis proved to be adequate to confidently identify metabolites in urine samples previously found to be predictive of lameness, and understand their potential biological relevance, despite the challenges of metabolite identification and pathway analysis when performing untargeted metabolomics. This approach shows potential as a reliable method to identify biomarkers that can be used in the future to predict the risk of lameness before calving. Validation with a larger cohort is required to assess the generalization of these findings.
Visible-NIR spectroscopic authentication assay for the classification of lamb meat according to pasture-finishing duration

Abstract:Spectral analysis of meat combined with chemometric analysis has been identified as a promising tool for authenticating livestock-animal diets. The objectives of this study were (i) to determine whether the visible-NIR spectrum of perirenal adipose tissue (PAT) and caudal adipose tissue (CAT) can reliably discriminate lambs pasture-finished for different durations before slaughter, and (ii) to analyze the kinetics of appearance and stabilization of the visible-NIR spectrum-based pasture signature in PAT and CAT. Four groups of 50-55 lambs were used over three years: lambs finished on lucerne pasture for 0 (L0, concentrate-fed in stall), 21 (L21), 42 (L42) and 63 (L63) days before slaughter. Partial least squares discriminant analysis was applied on PAT or CAT visible-NIR spectra to discriminate the groups. No one adipose tissue reliably discriminated the four groups, with less than 62 % lambs correctly classified. However, visible-NIR spectroscopy was able to discriminate stall-fed (L0) from pasture-finished (L21 + L42 + L63) lambs, with an accuracy of 93.8 % and 87.5 % lambs correctly classified based on PAT and CAT spectra, respectively. The lucerne pasture fingerprint (or signature) on visible-NIR spectrum appeared between 0 and 42 days in more than 95 % of lambs. It stabilized between 42 and 63 days in CAT, but had not stabilized within the range of grazing durations pre-slaughter explored in PAT. Further research into shorter and longer pasture-finishing durations could help determine more precisely the time required for the pasture signature to appear and stabilize in animal tissues.
Metabolomics-based biomarkers of fermented dairy and red meat intake: a randomized controlled trial in healthy adults

Abstract:Background: Dietary assessment is usually performed through imprecise tools, leading to error-prone associations between diet and health-related outcomes. Metabolomics has been applied in recent years to develop biomarkers of food intake (BFIs) and to study metabolites in the diet-microbiome crosstalk. Candidate BFIs exist to detect intake of meat and to a lesser extent dairy, but validation and further development of BFIs are needed. Here, we aim to identify biomarkers that differentiate between intakes of red meat and dairy, to validate previously reported BFIs for these foods, and to explore the effect of protein-matched meals on selected microbial metabolites. Methods: We conducted a randomized, controlled, cross-over single-meal study comparing a meal with highly fermented yogurt and cheese, and a meal with beef and pork meatballs. Postprandial urine samples from 17 subjects were collected sequentially after each meal up to 24 h and analyzed by untargeted metabolomics through ultra-high-performance-liquid chromatography (UHPLC) coupled via electrospray (ESI) source to a qTOF mass spectrometer. Univariate (repeated measures ANOVA) and multivariate (PLSDA, ML-PLSDA) data analyses were used to select BFIs differentiating the two meals. 3-Indoxyl sulfate, p-cresol sulfate, and several other microbial amino acid catabolites were additionally explored within the urine profiles. Results: Thirty-eight markers of meat and dairy intake were selected and are presented along with their excretion kinetics. Carnosine, taurine, and creatine, as well as hydroxyproline-based dipeptides are confirmed as meat BFIs. For dairy, previously reported metabolites such as acyl-glycines are confirmed, while proline-based dipeptides are reported as novel putative BFIs. Microbial metabolites showed only marginal evidence of differential formation after the two meals. Conclusion: This study allowed us to validate the postprandial kinetics of previously suggested biomarkers of meat and dairy intake and to identify new potential biomarkers. The excretion kinetics are useful to ensure that the collection of urine covers the correct time window in future dietary studies. The BFIs add to the existing body of biomarkers and may further be used in combination to provide a more reliable assessment of meat and dairy intake. Proteolytic microbial metabolites should be further investigated to assess the effect of different protein sources on health.

The potentiality of NMR-based metabolomics in food science and food authentication assessment

Abstract:In the last years, there was an increasing interest on nuclear magnetic resonance (NMR) spectroscopy, whose applications experienced an exponential growth in several research fields, particularly in food science. NMR was initially developed as the elective technique for structure elucidation of single molecules and nowadays is playing a dominant role in complex mixtures investigations. In the era of the "omics" techniques, NMR was rapidly enrolled as one of the most powerful methods to approach metabolomics studies. Its use in analytical routines, characterized by rapid and reproducible measurements, would provide the identification of a wide range of chemical compounds simultaneously, disclosing sophisticated frauds or addressing the geographical origin, as well as revealing potential markers for other authentication purposes. The great economic value of high-quality or guaranteed foods demands highly detailed characterization to protect both consumers and producers from frauds. The present scenario suggests metabolomics as the privileged approach of modern analytical studies for the next decades. The large potentiality of high-resolution NMR techniques is here presented through specific applications and using different approaches focused on the authentication process of some foods, like tomato paste, saffron, honey, roasted coffee, and balsamic and traditional balsamic vinegar of Modena, with a particular focus on geographical origin characterization, ageing determination, and fraud detection.
Detection of Pork in Beef Meatballs Using LC-HRMS Based Untargeted Metabolomics and Chemometrics for Halal Authentication

Abstract:Adulteration of high-quality meat products using lower-priced meats, such as pork, is a crucial issue that could harm consumers. The consumption of pork is strictly forbidden in certain religions, such as Islam and Judaism. Therefore, the objective of this research was to develop untargeted metabolomics using liquid chromatography-high resolution mass spectrometry (LC-HRMS) combined with chemometrics for analysis of pork in beef meatballs for halal authentication. We investigated the use of non-targeted LC-HRMS as a method to detect such food adulteration. As a proof of concept using six technical replicates of pooled samples from beef and pork meat, we could show that metabolomics using LC-HRMS could be used for high-throughput screening of metabolites in meatballs made from beef and pork. Chemometrics of principal component analysis (PCA) was successfully used to differentiate beef meatballs and pork meatball samples. Partial least square-discriminant analysis (PLS-DA) clearly discriminated between halal and non-halal beef meatball samples with 100% accuracy. Orthogonal projection to latent structures-discriminant analysis (OPLS-DA) perfectly discriminated and classified meatballs made from beef, pork, and a mixture of beef-pork with a good level of fitness (R2X = 0.88, R2Y = 0.71) and good predictivity (Q2 = 0.55). Partial least square (PLS) and orthogonal PLS (OPLS) were successfully applied to predict the concentration of pork present in beef meatballs with high accuracy (R2 = 0.99) and high precision. Thirty-five potential metabolite markers were identified through VIP (variable important for projections) analysis. Metabolites of 1-(1Z-hexadecenyl)-sn-glycero-3-phosphocholine, acetyl-l-carnitine, dl-carnitine, anserine, hypoxanthine, linoleic acid, and prolylleucine had important roles for predicting pork in beef meatballs through S-line plot analysis. It can be concluded that a combination of untargeted metabolomics using LC-HRMS and chemometrics is promising to be developed as a standard analytical method for halal authentication of highly processed meat products.

Validation of Raman spectroscopic models to verify the origin of Australian beef grown under different production systems

Abstract:Verification of beef production systems and authentication of origin is becoming increasingly important as consumers base purchase decisions on a greater number of perceived values including the healthiness and environmental impact of products. Previously Raman spectroscopy has been explored as a tool to classify carcases from grass and grain fed cattle. Thus, the aim of the current study was to validate Partial Least Squares Discriminant Analysis (PLS-DA) models created using independent samples from carcases sampled from northern and southern Australian production systems in 2019, 2020 and 2021. Validation of the robustness of discrimination models was undertaken using spectral measures of fat from 585 carcases which were measured in 2022 using a Raman handheld device with a sample excised for fatty acid analysis. PLS-DA models were constructed and then employed to classify samples as either grass or grain fed in a two-class model. Overall, predictions were high with accuracies of up to 95.7% however, variation in the predictive ability was noted with models created for southern cattle yielding an accuracy of 73.2%. While some variation in fatty acids and therefore models can be attributed to differences in genetics, management and diet, the impact of duration of feeding is currently unknown and thus further work is warranted.
A promising 31P NMR-multivariate analysis approach for the identification of milk phosphorylated metabolites and for rapid authentication of milk samples

Abstract:A fast and reliable method for the identification of milk from different mammalians was developed by using 31P NMR metabolite profile of milk serum coupled to multivariate analysis (PCA and classification models UNEQ, SIMCA and K-NN). Ten milk samples from six different mammalians, relevant to human nutrition (human, cow, donkey, mare, goat, sheep), were analyzed and eight monophosphorylated components were identified and quantified: phosphocreatine (PCr), glycerophosphorylcholine (GPC), glycerophosphorylethanolamine (GPE), N-acetylglucosamine-1-phosphate (NAcGlu-1P), lactose-1-phosphate (Lac-1P), galactose-1-phosphate (Gal-1P), phosphorylcholine (PC), glucose-6-phosphate (Glu-6P). PCA showed interesting clustering based on the animal genus. K-NN can be successfully used to discriminate between donkey and cow samples while UNEQ class-modeling resulted more suitable for compliance verification. Results confirm the natural variability of milk samples among different species. These data highlight the great potentials of NMR/multivariate analysis combined method in the rapid analysis of phosphorylated milk serum metabolites for milk origin assessment and milk adulteration detection.

Abstract:Fraudulent blending of food products with meat from undeclared species is a problem on a global scale, as exemplified by the European horse meat scandal in 2013. Routinely used methods such as ELISA and PCR can suffer from limited sensitivity or specificity when processed food samples are analyzed. In this study, we have developed an optimized method for the detection of horse and pork in different processed food matrices using MRM and MRM(3) detection of species-specific tryptic marker peptides. Identified marker peptides were sufficiently stable to resist thermal processing of different meat products and thus allow the sensitive and specific detection of pork or horse in processed food down to 0.24% in a beef matrix system. In addition, we were able to establish a rapid 2-min extraction protocol for the efficient protein extraction from processed food using high molar urea and thiourea buffers. Together, we present here the specific and sensitive detection of horse and pork meat in different processed food matrices using MRM-based detection of marker peptides. Notably, prefractionation of proteins using 2D-PAGE or off-gel fractionation is not necessary. The presented method is therefore easily applicable in analytical routine laboratories without dedicated proteomics background.

Abstract:One‐way mixed leukocyte cultures, supplemented by the daily addition of small numbers of fresh stimulating cells between days 5 and 15 of incubation, failed to give a secondary proliferative or cell‐mediated lyrnpholytic response upon challenge with activating concentrations of the same stimulating cells. However, these cultures could be fully activated by blastogenic factor (BF). The unresponsiveness induced was immunologically specific: when reponding cells were primed simultaneously with stimulating cells from two donors and one stimulator was added daily, the cultures were unresponsive to challenge with cells of the stimulator which were added daily, but good secondary responses were elicited both by cells of the other stimulator used in priming and by BF. The induced unresponsiveness may be a consequence of specific inactivation of helper cells.

Abstract:Because of the increased demand for processed meat, there is an urgent need to introduce specific identification methods. Strategies such as molecular genetics and the physical condition of meat are used to quickly explore multi-component products. However, a single methodology does not always unambiguously classify a product as counterfeit. In laboratory practice, as a rule, screening techniques are rarely used in the first stage, followed by arbitration. This work aimed to study individual methodologies using artificially falsified meat samples as examples and to identify their composition based on muscle tissue. For the experiments, the three most common types of raw meat were selected: pork, beef, and chicken. The calculation of the content of muscle tissue was carried out according to the BEFFE method. The study of muscle protein was carried out by ICA, ELISA, PCR, microstructural analysis, and mass spectrometric identification. In this connection, we proposed a multilevel control system for multicomponent meat products. Both classical methodologies, such as calculation by prescription bookmarks (BEFFE) and microstructural analysis, and approaches of highly sensitive methodologies, such as identification of muscle tissue by marker peptides (LC/MS-MRM) and semi-quantitative PCR analysis, were evaluated.

Abstract:Meat species identification and animal authentication in meat products is a significant subject, attention to which would contribute to fair-trade, and would enable consumers to make informed choices. Analytical methods are often based on protein or DNA measurements. Methods based on protein fractions include electrophoretic, chromatographic and immunological techniques and are often not suitable for compound food products, nor are they sensitive in processed products to differentiate closely related meat species. Advances in DNA technology have led to the rapid development of alternative approaches to species identification. Recently, application of polymerase chain reaction in food analysis has increased in the light of their simplicity, specificity and sensitivity. This review discusses a wide range of analytical methods with a focus on their ability to quantify meat and authentication of meat products.
