Abstract:The self-assembly adsorption of function protein on crystal surfaces, as a common phenomenon, broadly takes place in many applications of biosensors, biocapsules and bioMEMS/bioNEMS. To systematically investigate the different adsorption characteristic of the same function protein on two different crystal surfaces under the identical environment, a hybrid surface composing silica and discontinuous Gold Nano Film (GNF) was fabricated by Physical Vapor Deposition (PVD) and ultrasonic cleaning method, where the dynamic process of the self-assembly adsorption of Bovine Serum Albumin (BSA) was in situ observed by Atomic Force Microscope (AFM). The variations on the junction area of the two different surfaces were studied in the aqueous solution before and after injecting BSA with 0.05 mg/ml concentration. It was found that silica, compared with same hydrophilic GNF, took on a fairly weak adsorption force. The results indicated that the adsorption strength of BSA on the hydrophilic crystal surface was determined not only by hydrophilic property, but also other interaction forces, like Van der Waals and so on. Moreover, observed under the contact mode of AFM, BSA adsorbed on GNF had great tendency to forming a ridge-like topography. These results may be helpful in the application of immunosensors and other areas.

Abstract:We have investigated the sensing performance of protein-based microcantilever biosensors prepared from multiple surface conjugation chemistries. The 11-mercaptoundecanoic acid monolayers were prepared according to both traditional and modified processes. In three protein-based biosensors, the modified process improved microcantilever sensing performance by increasing the bending amplitude, a critical step toward developing a cost-effective microcantilever-based sensor platform for medical diagnostics and environmental and drug screening applications. Scanning electron microscopy (SEM) images demonstrated that proteins immobilized on the microcantilever surface using the modified chemistry approach formed a compact layer.

Abstract:In this study, a versatile fabrication method for coating enzyme-based biosensors with ultrathin antifouling zwitterionic polymer films to meet the challenge of the long-time stability of sensors in vivo was developed. Electrochemically mediated atom transfer radical polymerization (eATRP) was applied to polymerize zwitterionic sulfobetaine methacrylate monomers on the rough enzyme-absorbed electrode surfaces; meanwhile, a refined overall bromination was developed to improve the coverage of polymers on the biosensor surfaces and to maintain the enzyme activity simultaneously for the first time. X-ray photoelectron spectroscopy and atomic force microscopy were used to characterize the properties of the polymer layers. The antifouling performance and long-time stability in 37 °C undiluted bovine serum in vitro were evaluated. The results showed that the polymer brush coatings diminished over 99% nonspecific protein adsorption and that the sensitivity of the evaluated sensor was maintained at 94% after 15 days. The overall sensitivity deviation of 7% was nearly 50% lower than that of the polyurethane-coated ones and also much smaller than the current commercially available glucose biosensors. The results suggested that this highly controllable electrodeposition procedure could be a promising method to develop implantable biosensors with long-time stability.

Abstract:Many gerontologists propose definitions of wisdom. Usually these are “empirical,” as opposed to a-priori or “real” definitions. In this article we defend an a-priori definition of wisdom. We briefly explain a-priori and empirical definitions, and how they relate to each other in research. After rejecting two classical a-priori definitions of wisdom, we present and defend our own, and examine its ability to predict key findings of recent empirical studies. Finally, we describe some implications of our approach for future empirical studies of wisdom.

Abstract:This study introduces the use of an IgA isotype aflatoxin (AF) specific monoclonal antibody for the development of a highly sensitive Quartz Crystal Microbalance (QCM) immunobiosensor for the detection of AF in inhibitory immunoassay format. The higher molecular weight of IgA antibodies proved an advantage over commonly used IgG antibodies in label free immunobiosensor measurements. IgA and IgG antibodies with similar affinity for AF were used in the comparative studies. Sensor surface was prepared by covalent immobilization of AFB1, using self assembled monolayer (SAM) formed on gold coated Quartz Crystal, with 1-Ethyl-3-(3-dimethylaminopropyl) carbodiimide/N-hydroxy succinimide (EDC/NHS) method using a diamine linker. Nonspecific binding to the surface was decreased by minimizing the duration of EDC/NHS activation. Sensor surface was chemically blocked after AF immobilization without any need for protein blocking. This protein free sensor chip endured harsh solutions with strong ionic detergent at high pH, which is required for the regeneration of the high affinity antibody-antigen interaction. According to the obtained results, the detection range with IgA antibodies was higher than IgG antibodies in QCM immunosensor developed for AFB1.

Abstract:A quartz crystal microbalance (QCM) biosensor integrated into a flow injection analysis (FIA) system was used for the real-time investigation of molecular recognition between a protein and small molecular medicinal agents. Two sulfa-drugs, sulfamethazine (SMZ) and sulfamethoxazole (SMO), were, respectively, immobilized on the gold electrodes of the piezoelectric crystals using appropriate procedures based on self-assembly of the dithiothreitol (DTT). The binding interactions of the two immobilized drug ligands, with various proteins in solution, were followed as changes in the resonant frequency of the modified crystals. Results obtained from this rapid screen analysis clearly indicated that the two drug ligands appeared quite different in this molecular recognition procedure although their structures were similar. SMZ-immobilized sensor showed specific interaction only with IgG, while SMO-immobilized sensor showed negligible specific binding with IgG, but binding with trypsin and chymotrypsin. Further studies on the specific interaction between immobilized SMZ and three different species of IgG—human IgG, goat IgG and mouse IgG were carried out and the marked species-dependent difference was observed. The resultant sensorgrams were rapidly analyzed by using an in-house kinetic analysis software based on genetic algorithm (GA) to derive both the kinetic rate constants (kass and kdiss) and equilibrium association constants (KA) for IgG–SMZ interactions. For the interactions, KA were 5.48×l05, 2.75×l05 and 1.86×l05 M−1 for human IgG, goat IgG and mouse IgG, respectively. The kinetic data provided further insight into the structural/functional relationships of different IgG on a molecular level.

Abstract:Multi-hydroxyl amines including tris(hydroxymethyl)aminomethane (Tris), serinol and ethanolamine were selected as weak affinity ligands using a rapid screening by quartz crystal microbalance (QCM) biosensor. Based on the specific recognition between the ligands and two proteins, lysozyme (LZM) and cytochrome c (Cyt c), a weak affinity chromatography method was developed for specific separation of the two proteins. The frontal analysis results showed that the apparent dissociation constants (K(D)) of ligand-protein complexes were all in the order of weak affinity (10(-4) M). By weak affinity columns modified with the three multi-hydroxyl amines individually, LZM and Cyt c were baseline separated as retarded peaks from non-specific protein and each other in a single cycle of loading and eluting. Moreover, the Tris-modified column typically showed the satisfactory repeatability and stability as a new type of weak affinity columns. The present strategy composed of QCM selecting and affinity chromatography separating was promising to extend the variety of weak affinity ligands and develop inexpensive specific affinity methods for separation and purification of multiple proteins on one single column.
Biomolecular Surface Functionalization and Stabilization Method to Fabricate Quantum Dots Nanobeads for Accurate Biosensing Detection

Abstract:The surface functionalization of quantum dots (QDs) is essential for their application as a label material in a biological field. Here, a protein surface functionalization approach was introduced to combine with silica encapsulation for the sustainable and stable synthesis of QDs nanobeads for biomarker detection. The formation of QDs nanobeads was achieved by multiple mercapto groups in bovine serum albumin (BSA) macromolecules as multidentate ligands to replace hydrophobic ligands on the surface of QDs and decompression. The resulting QDs nanobeads exhibited 20 times more photoluminescence than the corresponding hydrophobic QDs and presented excellent stability under physiological conditions due to the protection of BSA and silica. The nanobeads served as a robust signal-generating reagent to construct the lateral flow immunoassay (LFIA) biosensor for the detection of glycosylated hemoglobin (HbA1c). The concentration of HbA1c was determined within 10 min with high specificity using only 60 μL of whole blood samples collected clinically. The nanobeads-based LFIA biosensor exhibited linear detection of HbA1c from 4.2% to 13.6%. The accuracy and stability of this approach in clinical utility was demonstrated by the detection of HbA1c after a long-term storage of test strips. Denatured bovine serum albumin hydrogel–based electrochemical biosensors for detection of IgG

Abstract:An antifouling sensing surface was constructed by crosslinking two-dimensional nanomaterial MXene with bovine serum albumin (BSA) denatured by urea previously. The immunoglobulin G (IgG) capture peptide was then modified to the surface to construct a highly selective antifouling electrochemical biosensor. Due to the large specific surface area and good electrical conductivity of MXene, the sensitivity of the biosensor is significantly enhanced. The biosensor at a working potential of around − 0.18 V (vs. Ag/AgCl) provides a wide linear detection range (0.1 ng/mL to 10 μg/mL) for IgG with a limit of detection of 23 pg/mL (3 σ / k ). The result is consistent with that obtained from the commercial enzyme-linked immunosorbent kit. Compared with BSA, which is usually used as a passivator or blocker for biosensing platforms, the hydrogel formed through the peptide chain obtained from BSA with good hydrophilicity can provide a better antifouling sensing surface to resist nonspecific adsorption. The prepared biosensor can quantitatively detect the concentration of IgG in complex human serum with high sensitivity. Thus, the antifouling sensing surface constructed without expensive antifouling materials and complex process is expected to develop as a variety of electrochemical biosensors and used for the clinical testing of biomarkers.
Development of an Aptamer-Based QCM-D Biosensor for the Detection of Thrombin Using Supported Lipid Bilayers as Surface Functionalization

Abstract:Biosensors play an important role in numerous research fields. Quartz crystal microbalances with dissipation monitoring (QCM-Ds) are sensitive devices, and binding events can be observed in real-time. In combination with aptamers, they have great potential for selective and label-free detection of various targets. In this study, an alternative surface functionalization for a QCM-D-based aptasensor was developed, which mimics an artificial cell membrane and thus creates a physiologically close environment for the binding of the target to the sensor. Vesicle spreading was used to form a supported lipid bilayer (SLB) of 1-palmitoyl-2-oleoyl-glycero-3-phosphocholine (POPC) and 1,2-dipalmitoyl-sn-glycero-3-phosphethanolamine-N-(cap biotinyl) (biotin-PE). The SLB was then coated with streptavidin followed by applying a biotinylated aptamer against thrombin. SLB formation was investigated in terms of temperature and composition. Temperatures of 25 °C and below led to incomplete SLB formation, whereas a full bilayer was built at higher temperatures. We observed only a small influence of the content of biotinylated lipids in the mixture on the further binding of streptavidin. The functionalization of the sensor surface with the thrombin aptamer and the subsequent thrombin binding were investigated at different concentrations. The sensor could be reconstituted by incubation with a 5 M urea solution, which resulted in the release of the thrombin from the sensor surface. Thereafter, it was possible to rebind thrombin. Thrombin in spiked samples of human serum was successfully detected. The developed system can be easily applied to other target analytes using the desired aptamers.

Abstract:Application-oriented hydrogel properties can be obtained by modifying the synthesis conditions of the materials. The purpose of this study is to achieve customized properties for sensing applications of hydrogel membranes based on poly(2-hydroxyethyl methacrylate), HEMA and N,N-dimethylacrylamide, DMAa. Copolymer p(HEMA-co-DMAa) hydrogels were prepared by varying the DMAa monomer ratio from 0–100% in 20% increments. Hydrogel membranes were characterized by attenuated infrared spectroscopy. Swelling and sorption were evaluated using cation solutions. Copolymers were also synthesized on the gold surface of quartz crystal microbalances (QCM) as coating membranes. A proof of concept was conducted for approaching the design and development of QCM sensors based on P(DMAa-co-HEMA)-membranes. Results showed that the water and ion adsorption capacity of hydrogel membranes increased with higher DMAa content. Membranes are not selective to a specific location but did show different transport features with each cation. The QCM coated with the selected membrane presented linear relationships between resonance frequency and ions concentration in solution (10–120 ppm). As a consequence, hydrogel membranes obtained are promising for the development of future biosensing devices.

Selection of Ligands for Affinity Chromatography Using Quartz Crystal Biosensor

Abstract:This paper described a new strategy for rapid selecting ligands for application in affinity chromatography using a quartz crystal microbalance (QCM) biosensor. An aminoglycoside antibiotic drug, kanamycin (KM), was immobilized on the gold electrodes of the QCM sensor chip. The binding interactions of the immobilized KM with various proteins in solution were monitored as the variations of the resonant frequency of the modified sensor. Such a rapid screen analysis of interactions indicated clearly that KM-immobilized sensor showed strong specific interaction only with lysozyme (LZM). The resultant sensorgrams were rapidly analyzed by using a kinetic analysis software based on a genetic algorithm to derive both the kinetic rate constants (k(ass) and k(diss)) and equilibrium dissociation constants (K(D)) for LZM-KM interactions. The immobilized KM showed higher affinity to LZM with a dissociation constant on the order of 10(-5) M, which is within the range of 10(-4)-10(-8) M and suitable for an affinity ligand. Therefore, KM was demonstrated for the first time as a novel affinity ligand for purification of LZM and immobilized onto the epoxy-activated silica in the presence of a high potassium phosphate concentration. The KM immobilized affinity column has proved useful for a very convenient purification of LZM from chicken egg white. The purity of LZM obtained was higher than 90%, as determined by densitometric scanning of sodium dodecyl sulfate-polyacrylamide gel electrophoresis of purified fraction. These results confirmed that the selected KM ligand is indeed a valuable affinity ligand for purification of LZM. The new screening strategy based on a QCM biosensor is expected to be a promising way for rapid selecting specific ligands for purifying other valuable proteins.

Abstract:A robust, efficient and sensitive quartz crystal microbalance (QCM) for glucose detection has been constructed using Au@bovine serum albumin (Au@BSA) nanoparticles as an active layer. The nanoparticles serve as tandem nanozymes and their stability over natural enzymes enable the sensor to show a wider linear dynamic range between 0.05 and 15 mM, a higher acid-resistance (pH 2.0–8.0) and heat-resistance (35–60 °C) than conventional glucose oxidase (GOx)-based sensors. The sensor has been further applied to measure glucose content in artificial urine directly without dilution, where the recovery of 99.6–105.2% and the relative standard deviations (RSDs) below 0.88% confirm a good reproducibility for the measurement results. In addition, the developed Au@BSA QCM sensor can retain 95% of its initial activity after 40 days of storage. Overall, the Au@BSA sensor shows better comprehensive performance than the commercial sensor strips for urine glucose analysis and provides a promising approach in a more precise and robust manner.

Abstract:An electrochemical platform was designed using biocompatible quaternary ammonium salts containing alkyl groups with different chain lengths as electrode materials for visible protein immobilization on a glassy carbon (GC) electrode. The electrode was constructed using a simple self-assembly method relying on the electrostatic interaction between negatively charged hemoglobin (Hb) and positively charged quaternary ammonium materials. The Hb/quaternary ammonium salts/GC assembly exhibited excellent catalytic and electrochemical activities. Additionally, the structure-function properties of the quaternary ammonium salts on the electrochemical behavior of Hb was systematically investigated for various alkyl chain lengths between monomer and polymeric structures. Meanwhile, the corresponding bactericidal activities of the monomers and related polymers were evaluated by determining the minimum bactericidal concentration (MBC), minimum inhibitory concentration (MIC), and inhibitory zone diameters against bacteria. The results of these studies demonstrated that the quaternary ammonium monomers not only immobilized more proteins, but also displayed better antibacterial activity as alkyl chain length increased. Moreover, polymers possessed higher antimicrobial activities than their monomeric counterparts. However, the efficiency of the direct electron transfer process and the antibacterial properties of long-chain polymers were limited because they were prone to aggregation and blistering. In summary, the present results provide convenient access to direct electrochemistry using an immobilized redox protein. Furthermore, the potential to use the obtained materials in the construction of third-generation electrochemical biosensors was evaluated.

Abstract:A biocompatible surface has been constructed on the electrode surface via layer-by-layer assembly of beta-nanozeolites and polydiallyldimethylammonium (PDDA) for the adsorption of enzymes towards sensitive biosensing. The film assembly process and enzyme adsorption were monitored by Quartz Crystal Microbalance measurements. The nanozeolite film exhibited an amazing adsorption capacity (about 350mgg−1) for tyrosinase as a model enzyme. The tyrosinase biosensor showed a high sensitivity (400μAmM−1), short response time (reaching 95% within 5s), broad linear response range from 10nM to 18μM, very low detection limit (0.5nM) and high operational and storage stability (more than 2 months). The apparent Michaelis–Menten constant KMapp was calculated to be 24μM using phenol as the substrate. The assembly-controlled nanozeolite film could provide a biocompatible surface for the interaction study between enzymes and target molecules.

Abstract:Quartz crystal microbalance (QCM) biosensor integrated in a flow injection analysis (FIA) system was used for the real-time investigation of the interaction between human TNF-α (hTNF-α) and its monoclonal antibodies (mAbs). Biologically active hTNF-α trimer was immobilized on the gold electrodes of the piezoelectric crystals and the binding interactions of the two immobilized antigen with two anti-hTNF-α monoclonal antibodies (Z8 and Z12) were followed as changes in the resonant frequency of the modified crystals. The resultant sensorgrams were rapidly analyzed by using an in-house kinetic analysis software based on genetic algorithm (GA) to derive both the kinetic rate constants (kass and kdiss) and equilibrium dissociation constant (KD) for antigen–antibody interactions. For the interactions, KD were 2.31×10−7 and 5.42×10−7 M for Z8 and Z12, respectively. The generally good agreement found between biosensor data and the affinities known from conventional equilibrium enzyme-linked immunosorbent assay (ELISA) validates this analytical method. The study shows the potential usefulness of QCM biosensor technology for biomolecular interaction analysis.

Abstract:Biomolecule immobilization on nanomaterials is attractive for biosensors since it enables the capture of a higher concentration of bioreceptor units while also serving as a transduction element. The technique could enhance the accuracy, specificity, and sensitivity of the analytical measurements of biomolecules. However, it was found that the limitation in chemically binding biomolecules on nanoparticle surfaces could only cross-link between the C-terminal and N-terminal. Here, we report the facile one-step synthesis of amine-functionalized silica nanoparticles (AFSNPs). (3-Aminopropyl)triethoxysilane was used as a precursor to modify the functional surface of nanoparticles via the Stöber process. The biomolecules were immobilized to the AFSNPs through itaconic acid, a novel cross-linker that binds between the N-terminal and N-terminal and potentially improves proteins and nucleic acid immobilization onto the nanoparticle surface. The newly developed immobilization approach on AFSNPs for biomolecular detection enhanced the efficiency of ELISA, resulting in increased sensitivity. It might also be easily used to identify different pathogens for clinical diagnostics.

Abstract:In the present study, the feasibility of electrospun polyethersolfone (PES) nanofibrous membrane as the solid substrate for microfluidic based immunoassays to enhance the density of immobilized antibody on the surface of membrane was assessed. Conversely, the efficacy of antibody immobilization was compared by two different strategies as 1-Ethyl-3-(3-dimethylaminopropyl)-carbodiimide (EDC)/N-Hydroxysuccinimide (NHS) coupling chemistry and hydrophobic interaction. Compared to conventional immunoassays carried out in plates or gels, microfluidic based immunoassays grant a lot of advantages such as a consumption of little samples and reagents, shorter analysis time, and higher efficiency. Therefore, microfluidic immunoassays can be efficiently used as a point-of-care device in medical diagnosis. Surprisingly, we found the increase of specific surface areas of the microfluidic channels improve density of immobilized proteins and leads to higher signal strength. Anti-staphylococcus enterotoxin B (anti-SEB) was used as an analyte model to demonstrate the utility of our proposed platform. Fluorescent microscopy, Fourier transform infrared spectroscopic (FTIR), gas adsorption, contact angle, X-ray photoelectron spectroscopy (XPS), scanning electron microscopy (SEM), Uv-Vis spectrophotometer and atomic force microscopy (AFM) techniques were used to assess the efficacy of antibody immobilization on the surface. To understand dominant mechanism of protein immobilization, zeta potential measurement was also carried out and it was found electrostatic attraction play significant role in antibody immobilization running into micro- channels containing through EDC/NHS. Moreover, incorporation of nanofibrous membrane causes significant improvement in the signal detection of microfluidic based immunoassay. © 2017 Wiley Periodicals, Inc.
