Abstract:This work establishes a hyphenated methodology coupling HPLC with ICP-MS for simultaneous speciation analysis of arsenic, mercury and lead for the first time. Four arsenicals (As(III), DMA, MMA and As(V)), four mercurials (Hg(II), MeHg, EtHg and PhHg) and three lead compounds (Pb(II), TML and TEL) were simultaneously analyzed within only 8 min with acceptable resolution (2.0-8.2 for As, 1.6-6.1 for Hg and 2.7-4.0 for Pb). The detection limits were 0.036-0.20 for As-species, 0.023-0.041 for Hg-species, and 0.0076-0.14 mu g L-1 for Pb-species. The developed method was applied for the measurement of five lotus seed samples, indicating the presence of DMA (19.6-28.2 mu g kg(-1)), TML (1.4-2.9 mu g kg(-1)), MeHg (1.2-4.8 mu g kg(-1)) and EtHg (0.8-2.2 mu g kg(-1)). This method provides a promising tool for studying the toxic, metabolic and bioavailable behaviors of arsenic, mercury and lead.
Simultaneous Measurements of Cr, Cd, Hg and Pb Species in Ng L-1 Levels by Interfacing High Performance Liquid Chromatography and Inductively Coupled Plasma Mass Spectrometry.

Abstract:Toxicity, mobility, bioavailability and biofunctions of chromium, cadmium, mercury and lead are heavily dependent upon their specific chemical forms, leading to a high demand to metal speciation analysis rather than total quantification. Simultaneous speciation analysis of multiple metal(loid)s is attractive to a large sample capacity containing unstable analytes due to its economic and environmental advantages over the conventional single elemental strategies. In this work, an analytical method integrating online solid phase extraction into high performance liquid chromatography interfaced with inductively coupled plasma mass spectrometry (ICP-MS) to simultaneously preconcentrate and quantify Cr, Cd, Hg and Pb forms in pg L-1 levels in water was developed. Cr (III + VI), Cd(II), Hg(II), Pb(II), methylmercury (MeHg), ethylmercury (EtHg), and trimethyl (TML) and triethyl lead (TEL) were captured by the C18 adsorbent (equilibrated with 10 mL of 1.0 mM 2-hydroxyethanethiol at 10 mL min-1) and eluted by mobile phase (5.0 mM Cys at pH 2.0), then completely separated on the C18 column within 8.0 min and eventually determined by ICP-MS. Low limits of detection (0.001-0.007 ng L-1) and quantification (0.003-0.023 ng L-1), good relative standard deviations (<4%) and high enrichment factors (827-2656 folds) were obtained with good linearities. Three reference materials of total cadmium (GBW08602), total mercury (GBW08603) and total lead (GBW08601) in water were analyzed by the developed method to validate the accuracy with good agreement with certified values and satisfactory recoveries (92-100%). This method was proved feasible by the determination of Cr, Cd, Hg and Pb compounds in drinking water, river water, pond water and tap water.
Chemical Speciation of Arsenic and Chromium in Seafood by LC-ICP-MS

Abstract:Two methods were developed for the As and Cr species determination in different kinds of seafood, exploring the LC-ICP-MS potential and chemometric approach to define the extraction conditions. Adequates separation and sensitivity data by LC-ICP-MS were obtained with 0.01 mol L-1 (NH4)2HPO4 in 1% ethanol (pH 8.0) for arsenic, and 0.015 mol L-1 Na2SO4, 0.005 mol L-1 EDTA, and 0.005 mol L-1 NaH2PO4 (pH 7.0) for chromium. The Doehlert matrix and Box-Behnken design defined the ideal extraction conditions for arsenic and chromium species. For As extraction, the optimized conditions employed 0.1 g of sample and 30 mmol L-1 HNO3 at 90°C for 45 min, and Cr, 0.1 g of sample and 0.045 mol L-1 [EDTA] at 90°C for 40 min. Recoveries from 88 to 106% of arsenobetaine (AsB), arsenite [As(III)], dimethylarsinic acid (DMA), monomethylarsonic acid (MMA), arsenate [As(V)], Cr(III) and Cr(VI) were obtained for all evaluated samples. The limits of quantification provided by the proposed methods were 5.3; 52.1; 16.4; 2.8; 83.3; 113.8; 53.9 ng g-1 for AsB, As(III), DMA, MMA, As(V), Cr(III) and Cr(VI) respectively. The trueness was evaluated using certified reference materials and addition and recovery procedures. The sum of the species agreed with the total concentration of arsenic and chromium contents.

Abstract:There are many kinds of micronutrient elements contained in foods that are vital to human health. The strong dependence of the toxicity and bioavailability of microelement upon their chemical forms has made the speciation and bioavailability vital to the food quality control and nutrition evaluation. A brief introduction to the high performance liquid chromatography hyphenated with inductive coupled plasma mass spectrometry ( HPLC-ICP-MS) is given. The hyphenated technique which based on several chromatographic separation mechanisms plays an important role in speciation analysis for its advantage of high sensitivity, high selectivity, wide linearity, low detection and multi-microelement determination simultaneously. The application of HPLC-ICP-MS in microelements speciation analysis and bioavailability research in foods concerning the selenium speciation analysis in selenized yeast, selenium-enriched garlic, selenium nutritional supplement and other foods, the arsenic speciation analysis in sea-products and farm products, the other microelements speciation analysis and the investigation of metabolic and bioactivity of microelement in organism is reviewed in this paper.
Synchronous Extraction and Quantitative Speciation of Arsenic and Chromium in Sediments by High-Performance Liquid Chromatography – Inductively Coupled Plasma – Mass Spectrometry (HPLC-ICP-MS)

Abstract:The optimum chromatographic separation conditions, polyatomic ions interference correction, and extraction parameters were investigated for simultaneous extraction and quantification of As(III), dimethylarsinic acid (DMA), monomethylarsonic acid (MMA), As(V) and Cr(VI) with excellent resolution. The ammonium salts (NH4H2PO4 and NH4NO3) were examined as mobile phases at different pH values by altering the composition during the chromatographic run in gradient elution mode using high-performance liquid chromatography – inductively coupled plasma – mass spectrometry (HPLC-ICP-MS). The 10 mM and 70 mM NH4NO3 composition at pH 8.7 enabled sufficient baseline separation of the target As and Cr species at a total run time of 18 minutes. The identified polyatomic ion interferences were corrected by an inter-element method using mathematical equations. The optimum extraction conditions containing a mixture of 0.375 M (NH4)2HPO4 and 50 mM EDTA at neutral pH as an extraction buffer, extraction temperature of 150 °C and volume of 15 mL were attained by a factorial design technique. The lowest concentrations of the selected elemental species that can be reliably detected by the procedure were in the range from 0.064 to 0.682 ng g−1. The accuracy of the developed procedure was demonstrated by achieving percentage recoveries of 96.8% for As(V) and 97.7% for Cr(VI) using San Joaquin Soil standard reference material 2709a. Proposed method was validated by achieving percentage recoveries from 84.9 to 104% for As species and Cr(VI) using spiking and recovery studies at 3x and 25x the limit of quantification values of the target species. The selected As and Cr species in real sediment were monitored using the optimized analytical method. The differences in concentrations of Cr(VI), As(III), MMA and As(V) between the seasonal state of the river were statistically significant (p ˂0.05) at a 95% confidence level as determined by analysis of variance (ANOVA).

Abstract:The role of calcium ions in the process by which nutrients affect glucagon secretion by pancreatic islet α-cells remains the matter of an apparently endless debate. In the prolongation of recent articles dealing with this matter, the present review draws attention to the dual role of Ca2+ as revealed by prior publications. In such a perspective, emphasis is placed on the increase in glucagon output in response to the omission of extracellular Ca2+ as recorded in the presence of D-glucose or 2-ketoisocaproate, the permissive role of extracellular Ca2+ in the positive secretory response to arginine or a mixture of fumarate, glutamate and pyruvate, and the effects of an organic calcium-antagonist on glucagon output. Considering the role currently ascribed to Ca2+ in the activation of motile events involved in stimulus-secretion coupling, attention is also given to the effects of cytochalasin B, D2O and mitotic-spindle inhibitors upon the secretory response of α-cells exposed to D-glucose in the absence or presence of arginine.

Abstract:Tin and lead are a global concern considering their species-dependent toxicity, bioavailability and transformation. Simultaneous speciation analysis of tin and lead is challenging for a large food capacity containing unstable species. Herein, we developed two sensitive methods for rapid quantification of tin and lead species in Antarctic seafood by high-performance liquid chromatography and inductively coupled plasma mass spectrometry based on strong cation-exchange and Amphion columns. Inorganic tin and lead, four organotin and two organolead compounds can be analysed in 16 min on a 10-cm Amphion II column (mobile phase: 4 mM sodium dodecyl benzene sulfonate at pH 2.0) with 0.02-0.24 mu g L-1 detection limits. The method was applied to Antarctic krill and fish, demonstrating the presence of any tin and lead species down to mu g kg-1 level. Overall, the proposed methods are sensitive, efficient and environment-friendly for routine speciation analysis of tin and lead in food samples.
Simultaneous Arsenic and Selenium Speciation Analysis Using HPLC-ICP-MS

Abstract:Four arsenic species and three selenium species simultaneous analysis method was build with High performance liquid chromatography and Inductively coupled plasma-mass spectrometry(HPLC-ICP-MS).The column of Hamilton PRP-X100 was used for chromatogram seperation,and Collision cell technology(CCT) was used for ICP-MS analysis.The detection limits for arsenite(As(Ⅲ)),dimethylarsinic acid(DMA),monomethylarsonic acid(MMA),arsenate(As(Ⅴ)),selenocystine(SeCys),selenite(Se(Ⅳ)) and selenomethionine(SeMet) were 0.2 μg/L,0.2 μg/L,0.2 μg/L,0.3 μg/L,0.5 μg/L,0.4 μg/L and 2.5 μg/L,respectively.The line range was 10~200 μg/L for all species except the SeMet,all linear correlation coefficients were not less than 0.9995.The species of arsenic and selenium in fresh mushroom and fish was detected with this method,good spiked recoveries from 81.6% to 103.7% were obtained.

Abstract:Nanoliter high-performance liquid chromatography shows low consumption of solvents and samples, offering one of the best choices for arsenic speciation in precious samples in combination with inuctively coupled plasma mass spectrometry. A systematic investigation on coupling nanoliter high-performance liquid chromatography to inductively coupled plasma mass spectrometry from instrument design to injected sample volume and mobile phase was performed in this study. Nanoflow mobile phase was delivered by flow splitting using a conventional high-pressure pump with reuse of mobile phase waste. Dead volume was minimized to 60 nL for the sheathless interface based on the previously developed nanonebulizer. Capillary columns for nanoliter high-performance liquid chromatography were found to be sensitive to sample loading volume. An apparent difference was also found between the mobile phases for nanoliter and conventional high-performance liquid chromatography. Baseline separation of arsenite, arsenate, monomethylarsenic, and dimethylarsenic was achieved within 11 min on a 15 cm C-18 capillary column and within 12 min on a 25 cm strong anion exchange column. Detection limits of 0.9-1.8 mu g/L were obtained with precisions variable in the range of 1.6-4.2%. A good agreement between determined and certified values of a certified reference material of human urine (GBW 09115) validated its accuracy along with good recoveries (87-102%).

Abstract:Multi-elemental speciation analysis in a single run reduces costs, waste and time, and also sample consumption, which is favorable to a large set of instable samples. This work demonstrates mercury and arsenic speciation in a single run using high performance liquid chromatography with inductively coupled plasma mass spectrometric detection (HPLC/ICP-MS). Thiol compounds in mobile phases interfered with the elution of arsenite because of coordination action, which was overcome by gradient elution (elution of As(III) by thiol-free mobile phases at first). Four arsenic forms (As(III), DMA, MMA and As(V)) and three mercury species (Hg(II), MeHg and EtHg) were separated and detected by HPLC/ICP-MS using 4 mM tetrabutylammonium hydroxide (TBAH) (pH 6.0) in the first 1.0 min and 4 mM TBAH + 20 mM L-cysteine (pH 6.0) in the next 9.0 min. The detection limits were obtained at 0.078 for As(III), 0.081 for DMA, 0.078 for MMA and 0.15 mu g L-1 for As(V), and 0.016 for Hg(II), 0.027 for MeHg and 0.032 mu g L-1 for EtHg, and the reproducibilities (50 mu g L-1 for each As-species and 10 mu g L-1 for each Hg-species) were all lower than 3.7%. The proposed method was successfully applied for speciation analyses of arsenic and mercury in five lotus roots, which indicated DMA (3.5-7.9 mu g kg(-1)) as the only arsenic species and MeHg (0.6-1.0 mu g kg(-1)) as the only mercury species. Analyses of two certified reference materials demonstrated good agreement between determined values and certified values. All results prove its advantages including high sensitivity and high analytical efficiency, which are beneficial for routine speciation analyses of arsenic and mercury in a single run in environmental and food fields.

Abstract:This paper presents arsenic speciation studies by high-performance liquid chromatography with inductively coupled plasma-mass spectrometer (HPLC-ICP-MS) using a chromatography column containing a strong anion exchange stationary phase (Agilent Bio SAX G3154A/101) for determination of arsenobetaine (AsB), dimethylarsinic acid (DMA), AsIII, monomethylarsonic acid (MMA), and AsV. The best separation was achieved in 15 min, using a 4 min isocratic step with 1 mM (NH4)2HPO4 pH 10.2, followed by a 2 min salt gradient from 1 to 10 mM, which was kept for 4 min. Compared to the Food and Drug Administration (FDA) method for rice using the PRP-X-100 column, the Bio SAX method was quicker, provided at least equal separation of AsB from the next peak, and led to more symmetrical peaks. The linearity between 0.5 and 60 µg L-1 was shown. The method was tested with 10 µg L-1 fortified rice and fish samples extracted in ultrasound using methanol/water 1:1, and recoveries between 40 and 90% were obtained.

Abstract:A quantitative method was established for the simultaneous speciation of arsenic (As) and selenium (Se) in rice using high performance liquid chromatography (HPLC)- dynamic reaction cell (DRC)-inductively coupled plasma mass spectrometry (ICP-MS). Hamilton PRP- X100 (150 mm × 4.6 mm × 10 μm) and gradient elution using (NH 4) 2CO 3 at pH 9.5 was allowed separation of AsB, As(III), DMA, MMA, As(V), Se(IV) and Se(VI) in less than 14 min. The ion 75As 12CHH + was determined to eliminate the interference of 75ArCl + for 75As + and the potentially interfering 40Ar 40Ar + for 80Se+ was reduced by using CH4 as reactive gas at optimal Rpq (0.35) and DRC cell gas flow rate (0.6 mL min-1). The detection limits for As and Se were in the range of 0.02 - 0.06 ng mL -1. The relative standard deviation of the peak areas for seven injections of 5 ng mL -1 As and Se mixture ranged from 0.32% to 4.67%. The method was applied to the determination of As and Se species from rice extracted by artificial gastric juice which intended to mimic the physical and biochemical process in the human digestive system. In this study, inorganic As containing As(III) and As(V) took up a high content of total As in Chinese rice samples. Se intake from rice samples is lower than the Se Recommended Dietary Allowance (RDA) in humans (70000 ng day -1). Because rice was the predominant Se source in the Chinese diet, lower Se in rice samples might lead to the Se deficiency of inhabitants in China. © 2011 IEEE.
Arsenic speciation based on ion exchange high-performance liquid chromatography hyphenated with hydride generation atomic fluorescence and on-line UV photo oxidation

Abstract:Abstract An on-line method capable of the separation of arsenic species was developed for the speciation of arsenite As(III), arsenate As(V), monomethylarsenic (MMA) and dimethylarsenic acid (DMA) in biological samples. The method is based on the combination of high-performance liquid chromatograph (HPLC) for separation, UV photo oxidation for sample digestion and hydride generation atomic fluorescence spectrometry (HGAFS) for sensitive detection. The best separation results were obtained with an anion-exchange AS11 column protected by an AG11 guard column, and gradient elution with NaH2PO4 and water as mobile phase. The on-line UV photo oxidation with 1.5% K2S2O8 in 0.2 mol L–1 NaOH in an 8 m PTFE coil for 40 s ensures the digestion of organoarsenic compounds. Detection limits for the four species were in the range of 0.11–0.15 ng (20 μL injected). Procedures were validated by analysis of the certified reference materials GBW09103 freeze-dried human urine and the results were in good agreement with the certified values of total arsenic concentration. The method has been successfully applied to speciation studies of blood arsenic species with no need of sample pretreatment. Speciation of arsenic in blood samples collected from two patients after the ingestion of realgar-containing drug reveals slight increase of arsenite and DMA, resulting from the digestion of realgar.
Fast Determination of Toxic Arsenic Species in Food Samples Using Narrow-bore High-Performance Liquid-Chromatography Inductively Coupled Plasma Mass Spectrometry

Abstract:A new method for the speciation analysis of arsenic in food using narrow-bore high-performance liquid-chromatography inductively coupled plasma mass spectrometry (HPLC-ICP-MS) has been developed. Fast separation of arsenite, arsenate, monomethylarsonic acid and dimethylarsinic acid was carried out in 7 min using an anion-exchange narrow-bore Nucleosil 100 SB column and 12 mM ammonium dihydrogen phosphate of pH 5.2 as the mobile phase, at a flow rate of 0.3 mL min(-1). A PFA-ST micronebulizer jointed to a cyclonic spray chamber was used for HPLC-ICP-MS coupling. Compared with standard-bore HPLC-ICP-MS, the new method has provided higher sensitivity, reduced mobile-phase consumption, a lower matrix plasma load and a shorter analysis time. The achieved instrumental limits of detection were in the 0.3 - 0.4 ng As mL(-1) range, and the precision was better than 3%. The arsenic compounds were efficiently (>80%) extracted from various food samples using a 1:5 methanol/water solution, with additional ultrasonic treatment for rice products. The applicability of this method was demonstrated by the analysis of several samples, such as seafood (fish, mussels, shrimps, edible algae) and rice-based products (Jasmine and Arborio rice, spaghetti, flour, crackers), including three certified reference materials.

Abstract:A rapid and novel two-step dispersive liquid-liquid microextraction and dispersive micro-solid phase extraction method was established for the separation and enrichment of trace cadmium, nickel, and copper in food and environmental water prior to determination by inductively coupled plasma-mass spectrometry (ICP-MS). In the first microextraction step, carbon tetrachloride was employed to extract metal-diethyldithiocarbamate chelates from aqueous solution with ultrasound. In the following step, Fe3O4 magnetic nanoparticles were added and used to collect the analytes in the organic solvent. The sample pH, type and volume of extraction solvent, mass of magnetic nanoparticles, concentration of the chelating agent, concentration of sodium chloride, and sonication time were optimized. The linear dynamic range was from 0.01 to 20 micrograms per liter with correlation coefficients between 0.9990 and 0.9992. Enrichment factors were 78, 79, and 81 for cadmium, nickel, and copper, respectively. The limits of detection for cadmium, nickel, and copper were 0.007, 0.009, and 0.017 micrograms per liter, with relative standard deviations from 1.1 to 2.6 percent. The developed method was validated by the determination of cadmium, nickel, and copper in rice and mussel certified reference materials, food, and environmental water with satisfactory results.
Separation and Detection of Arsenic and Selenium Species in Environmental Samples by HPLC-ICP-MS

Abstract:A method is presented for arsenic speciation analysis of an oyster sample using ion chromatography coupled with an inductively coupled plasma mass spectrometry (ICP-MS) instrument. A strong anion exchange resin was employed with a step gradient elution of 0.1 mM/0.1 M K 2 SO 4 at pH 10.2. Arsenobetaine and dimethylarsinic acid were determined following extraction based on trypsin enzymolysis with 95-100% extraction efficiency. Limits of detection in the range 0.1-0.3 mg kg m 1 of arsenic were obtained for organic arsenic species. No inorganic arsenic was detected. Validation was performed using TORT-2 as a certified reference material. Although high performance liquid chromatography (HPLC) coupled to ICP-MS is an effective method for speciation analysis it is not always necessary to obtain such a detailed picture. A simple liquid chromatographic separation technique based upon mini-column technology is presented. It was developed to obtain a fast, efficient and reliable separation of inorganic from organic, i.e. assumed toxic from non-toxic, arsenic and selenium species suitable for use as an initial screening method for environmental analysis. Two types of strong anion exchange resin were tested. Excellent separation was obtained for both min-column resins and analysis times were within 7 min. Limits of detection obtained for inorganic arsenic, organic arsenic, selenomethionine, Se IV and Se VI were 1.6, 1.8, 66, 32 and 22 µg kg m 1 , respectively.

Abstract:Cross-reacting material 197 (CRM197), a specific HB-EGF inhibitor, has been proven to be a promising antitumor agent for ovarian cancer therapy. Our previous studies have shown that CRM197 has potent antitumor activity in human cisplatin-resistant ovarian cancer. However, the relationship between CRM197 and the resistance to cisplatin remains unclear. Here, we report that CRM197 significantly reverses the resistance to cisplatin in cisplatin-resistant ovarian carcinoma cell line (A2780/CDDP). We established xenograft nude mice models with A2780 and A2780/CDDP cells. Notably, we observed that CRM197 suppresses the expression of HB-EGF and epidermal growth factor receptor in A2780/CDDP cells and xenografts harboring the overexpression of HB-EGF and epidermal growth factor receptor. Experiments conducted in vitro and in vivo suggest that CRM197 markedly downregulates the expression of excision repair cross-complementing group 1 (P=0.002) and DNA repair capacity in A2780/CDDP tumor (P<0.001) by inactivation of extracellular signal-regulated kinase signaling, providing novel possible mechanisms for the ability of CRM197 to restore drug sensitivity. These results suggest that CRM197 as an HB-EGF inhibitor might be a cisplatin-chemosensitizing agent for the treatment of ovarian carcinoma with resistance to cisplatin.

Abstract:Clinimetallomics is proposed as a branch of metallomics that focuses on the study of the metallome in clinical samples of urine, blood, and tissues. As the clinical diagnosis of arsenic poisoning is mainly based on the concentration of total arsenic in urine, the toxicity of arsenic varies greatly in different speciation. Analysis of arsenic speciation with excessive total arsenic in urine can provide a basis for precise treatment. It can also be used to understand the fate of arsenic in the body of patients with arsenic poisoning after treatment with sodium dimercaptopropane sulfonate. In this study, a HPLC-ICP-MS method was established for the determination of arsenic species in urine samples from patients diagnosed with arsenism. Use the established method to detect urine samples, which can be directly assayed after simple sample dilution with 20 mmol/L EDTA-2Na. With the concentration of arsenic speciation in the range of 1.0~100.0 ng/mL, the linear correlation coefficient was higher than 0.99996. The recoveries were between 92.4% and 109.0%. The precision of the concentration and retention time (n = 3) were less than 3.0% and 0.3%, respectively, and the detection limit was between 1.42 ng/mL and 1.86 ng/mL. This method can be applied to arsenic speciation in the urine of healthy people, in patients treated for arsenic poisoning, and in patients diagnosed with arsenism.

Abstract:OBJECTIVE: To report the experience of using rasburicase as a single-dose treatment for childhood leukemia presenting with hyperuricemia. CASE SUMMARIES: Three children with acute lymphoblastic leukemia presenting with hyperuricemia received rasburicase as a single intravenous dose just prior to the start of chemotherapy. This was followed by rapid reduction of serum uric acid levels within 24 hours, which remained low throughout induction therapy while allopurinol and hydration therapy without urinary alkalinization ensued. Subclinical tumor lysis was evidenced by the appearance of hyperphosphatemia and hypocalcemia in all cases and hyperkalemia in 1 patient. These abnormalities were transient, and each patient's renal function gradually improved from pretreatment baseline without requiring dialysis. DISCUSSION: Our experience suggests that hyperuricemia in children at risk for tumor lysis can be managed with a briefer regimen of rasburicase than the recommended 5- to 7-day course. CONCLUSIONS: A shorter course of rasburicase treatment, including single-dose injection, is feasible and will improve the cost-effectiveness profile of the otherwise expensive compound.
