Abstract:A rapid and sensitive ultra-high performance liquid chromatography-tandem mass spectrometry method was developed to determine flurbiprofen in rat plasma. A triple quadrupole tandem mass spectrometer equipped with an electrospray ionization (ESI) source was used in negative ion mode. Acetonitrile precipitation was selected to prepare samples. Flurbiprofen and internal standard flurbiprofen-d5 were analyzed on an Acquity UPLC BEH C18 column with the mobile phase consisting of acetonitrile and water, and a gradient procedure was used for separation. The retention time of flurbiprofen was 0.67 min, and the whole running time was only 1.2 min. The detection was performed on a triple quadrupole tandem mass spectrometer using multiple reaction monitoring mode via an ESI source with optimized mass spectrometry parameters. The calibration curve was linear in the range of 25.0-1.00 × 104 ng/mL (r ≥ 0.99). The within-run and between-run relative standard deviations were not more than 13.9%. The within-run and between-run relative errors were from -9.0% to 3.4%. There was no significant matrix effect, and recovery was high. This method was fully validated, including whole blood stability in rat plasma, and successfully applied to the pharmacokinetic study in which 100% incurred sample reanalysis met the criteria.
A Quantitative Determination of Fluorochloridone in Rat Plasma by Uplc-Ms/Ms Method: Application to A Pharmacokinetic Study

Abstract:A precise, high-throughput and sensitive ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method has been developed for the determination of fluorochloridone (FLC) in rat plasma. The extraction of analytes from plasma samples was carried out by protein precipitation procedure using acetonitrile prior to UPLC-MS/MS analysis. Verapamil was proved as a proper internal standard (IS) among many candidates. The chromatographic separation based on UPLC was well optimized. Multiple reaction monitoring in positive electrospray ionization was used with the optimized MS transitions at: m/z 312.0 → 292.0 for FLC and m/z 456.4 → 165.2 for IS. This method was well validated with good linear response (r(2) > 0.998) observed over the investigated range of 3-3000 ng/mL and with satisfactory stability. This method was also characterized with adequate intra- and inter-day precision and accuracy (within 12%) in the quality control samples, and with high selectivity and less matrix effect observed. Total running time was only 1.5 min. This method has been successfully applied to a pilot FLC pharmacokinetic study after oral administration. Copyright © 2016 John Wiley & Sons, Ltd.
Determination of Paeoniflorin in Rat Plasma by Ultra-high Performance Liquid Chromatography-Tandem Mass Spectrometry and its Application to a Pharmacokinetic Study

Abstract:In this study, a rapid and highly sensitive ultra-high performance liquid chromatography-tandem mass spectrometry method was established for the quantification of paeoniflorin in rat plasma. The analyte and the internal standard (IS), hyperoside, were extracted from rat plasma via liquid-liquid extraction with ethyl acetate. LC separation was carried on a Phenomenex Luna C18 column within 2.5 min. Quantitation was performed on a triple quadrupole mass spectrometer employing electrospray ionization and operated in the multiple reaction monitoring and negative ion modes. The precursor to product ion transitions monitored for paeoniflorin and the IS were m/z 524.8→449.0 and 463.1→300.0, respectively. The assay was validated with a linear range of 0.02~25.6 μg/mL for paeoniflorin. The intra- and inter-day precisions (relative standard deviation%) were within 9.7%, and the recoveries were greater than 65.2%. Paeoniflorin was proven to be stable during all sample storage, preparation and analytical procedures. The method was successfully applied to a pharmacokinetic study of the Baixiangdan Capsule in eight female rats.

Abstract:OBJECTIVE To develop a rapid,highly sensitive and selective high performance liquid chromatography tandem mass spectrometry (LC-MS/MS) for the determination of 4-chloro-2-fluoro-3-methoxybenzeneboronic acid(PBA) in rat plasma.Explore and complete the toxicokinetic study of PBA in rats.METHODS To observe the plasma concentration change of PBA in SD rats after treated with different doses of PBA orally and intravenously,respectively.Internal standard method was used in analysis to quantify the PBA plasma concentration.DAS software was used to obtain kinetic parameters such as AUC,Cmax,Tmax,T1/2 and so on.RESULTS The extraction recovery rate of PBA in rat plasma was in the range of 88.8％-94.4％,the intra-day precision was in the range of 6.4％-8.9％,and the inter-day precision was in the range of 3.9％-5.9％,respectively.Accuracy was in the range of 97.8％-105.5％.Linear calibration curves for PBA was observed at 20-2 500 ng·mL-1 with correlation coefficient r＞0.99.The lowest limit of quantification(LLOQ) for PBA was 20 ng·mL-1(RSD=9.67％,n=5).CONCLUSION This method is successfully applied in the rat plasma concentration determination of PBA.The plasma concentrations of PBA in rats has nonlinear dynamics and its systemic exposure did not raise as the administered doses.
Simultaneous Determination of Three Flavonoids in Rat Plasma by RP-LC after Oral Administration of the Total Flavonoids of Scutellaria Barbata

Abstract:A liquid chromatographic method for the simultaneous determination of three flavonoids, scutellarin (SCU), isoscutellarein-8-O-glucuronide (ISO) and luteolin (LUT) in rat plasma was developed and validated. Following a single-step liquid–liquid extraction with ethyl acetate, the analytes and internal standard (IS) (rutin) were successfully separated on a Diamonsil C18 column using a mobile phase composed of acetonitrile (A)–0.2% phosphoric acid aqueous solution (B) (0–5 min, 20% A–29% A; 5–25 min, 29% A, v/v) at a flow rate of 1.0 mL min−1. The linear range was 0.044–2.20 μg mL−1 for SCU, 0.042–2.08 μg mL−1 for ISO, and 0.056–2.80 μg mL−1 for LUT, with the correlation coefficients of 0.9995, 0.9989 and 0.9963, respectively. The limit of quantification of SCU, ISO and LUT were 44, 41.6 and 56 ng mL−1, respectively. The accuracy of assay was between 88.4 and 103.0%. The inter-day and intra-day precisions (RSD) were less than 10.5%. The developed method was simple, rapid and applied successfully to study the pharmacokinetics of SCU, ISO and LUT after oral administration of the total flavonoids of Scutellaria barbata.
Determination of Albiflorin in Rat Plasma by LC-MS/MS and Its Pharmacokinetics

Abstract:OBJECTIVE: To develop a rapid, sensitive and selective liquid chromatography tandem spectrometry method for the determination of albiflorin in rat plasma, and to investigate its pharmacokinetic parameters in rats. METHODS: The plasma samples were pretrealed with solid phase extraction. HPLC analysis was carried out on a Symmetry C18 column (4.6 mm x 50 mm, 3.5 μm) with a mobile phase consisting of acetonitrile and 0.05% formic acid (25:75). Detection was carried out on a triple quadrupole tandem mass spectrometer by multiple-reaction monitoring and an electrospray ionization source was employed as the ionization source. The method was fully validated. The phamacokinetic characteristics of albiflorin was investigated after oral administration to rats in three dosages. RESULTS: The method was sensitive and reproducable. Albiflorin was absorbed and eliminated rapidly. The statistic analysis showed the pharmacokinetics of albiflorin complied with linear kinetic course. CONCLUSION: The method could be applied to the study of pharmacokinetics of albiflorin.
A UPLC-MS/MS Approach for Simultaneous Determination of Eight Flavonoids in Rat Plasma, and Its Application to Pharmacokinetic Studies of Fu-Zhu-Jiang-Tang Tablet in Rats.

Abstract:The purpose of this study is to establish and validate a UPLC-MS/MS approach to determine eight flavonoids in biological samples and apply the method to pharmacokinetic study of Fu-Zhu-Jiang-Tang tablet. A Waters BEH C-18 UPLC column was employed with methanol/0.1% formic acid-water as mobile phases. The mass analysis was carried out in a triple quadrupole mass spectrometer using multiple reaction monitoring with negative scan mode. A one-step protein precipitation by methanol was used to extract the analytes from blood. Eight major flavonoids were selected as markers. Our results showed that calibration curves for 3'-hydroxypuerarin, mirificin, puerarin, 3'-methoxypuerarin, daidzin, rutin, astragalin and daidzein displayed good linear regression (r(2) > 0.9986). The intra-day and inter-day precisions (RSD) of the eight flavonoids at high, medium and low levels were < 8.03% and the bias of the accuracies ranged from -5.20 to 6.75%. The extraction recoveries of the eight flavonoids were from 91.4 to 100.5% and the matrix effects ranged from 89.8 to 103.8%. The validated approach was successfully applied to a pharmacokinetic study in Sprague-Dawley rats after oral administration of FZJT tablet. Double peaks were emerged in curves of mean plasma concentration for 3'-methoxypuerarin, which was reported for the first time.

Abstract:An ultra-high pressure liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was developed for the quantification of three major ingredients in Chinese lantern preparations (CLP) in rat plasma. Following extraction by ethyl acetate, the analytes were separated on an Acquity UPLC BEH Shield RP C(18) column using a gradient mobile phase system of acetonitrile-water. Electrospray ionization (ESI) tandem interface was employed prior to mass spectrometric detection. The calibration curves were linear over the range of 5.0-500.0 ng/ml for physalin D, 2.3-230.0 ng/ml for physalin G and 0.71-71.0 ng/ml for 4,7-didehydroneophysalin B. The average extraction recoveries, examined at four concentration levels, carried from 57.1% to 76.9%, and the accuracies ranged from 94.0% to 113.3% with precision (RSD) <15%. The validated method was successfully applied to the determination of the three physalins in rat plasma after intragastric administration of CLP suspension.

Abstract:Physalin D is an important constituent of some traditional Chinese medicines, and has several known bioactivities. An UPLC-MS/MS method for the determination of physalin D in rat plasma and tissues was developed and the pharmacokinetic and tissue distribution characteristics of physalin D after intravenous administrations were investigated. The bio-samples were prepared by a simple protein precipitation, and the separation of physalin D was achieved on a UPLC HSS T3 column with a mobile phase consisting of methanol/acetonitrile (70:30, v/v) and water (containing 0.1% formic acid and 10 mM ammonium acetate) at a flow rate of 0.3 mL/min. The MS/MS detection was carried out by monitoring the fragmentation of m/z 544.9→508.8 for physalin D and m/z 286.7→152.8 for luteolin (internal standard; IS) on a triple-quadrupole mass spectrometer. The total run time was only 3.6 min. The analyte showed good linearity over a wide concentration range (R(2)>0.995) and its lower limit of quantification was 2 ng/mL. The pharmacokinetic study found that physalin D was distributed and eliminated rapidly in rats (t(1/2)<10 min). Tissue distribution showed the highest level was observed in kidney, then in liver, but no physalin D was detected in brain, which indicated that kidney was the major distribution tissue for physalin D in rats and that physalin D does not cross the blood-brain barrier.
Determination of Oxaceprol in Rat Plasma by Lc-Ms/Ms and Its Application in A Pharmacokinetic Study

Abstract:A sensitive method for the quantification of oxaceprol in rat plasma using high-performance liquid chromatography–tandem mass spectrometry (LC–MS/MS) was developed. Sample pretreatment involved a simple protein precipitation by the addition of 60μL of acetonitrile–methanol (1:2, v/v) to 20μL plasma sample volume. Separation was achieved on a Dikma ODS-C18 (5μm, 150mm×4.6mm) reversed-phase column at 40°C with acetonitrile/0.1% formic acid–4mM ammonium acetate in water (35:65,v/v) at a flow rate of 0.6mL/min. Detection was performed using an electrospray ionization (ESI) operating in negative ion multiple reaction monitoring (MRM) mode by monitoring the ion transitions from m/z 172→130 (oxaceprol) and m/z 153→109 (protocatechuic acid, internal standard). The calibration curve of oxaceprol in plasma showed good linearity over the concentration range of 1.25–800ng/mL. The limit of detection and limit of quantification were 0.400ng/mL and 1.25ng/mL, respectively. Intra- and inter-day precisions in all samples were within 15%. There was no matrix effect. The validated method was successfully applied to a preclinical pharmacokinetic study of oxaceprol in rats. After oral administration of 20mg/kg oxaceprol to rats, the main pharmacokinetic parameters Tmax, Cmax, T1/2, Vz/F and AUC0–t were 1.4h, 1.2μg/mL, 2.3h, 19.7L/kg and 3.4mgh/L, respectively.

Abstract:A precise and sensitive liquid chromatography–tandem mass spectrometry (LC–MS/MS) method for simultaneous determination of vinpocetine (VP) and its primary metabolite, apovincaminic acid (AVA), in rat plasma was developed and validated. The analytes and the internal standard-dimenhydrinate were extracted from 50μL aliquots of rat plasma via solid–liquid extraction. Chromatographic separation was achieved in a run time of 3.5min on a C18 column under isocratic conditions. Detection of analytes and IS was done by tandem mass spectrometry, operating in positive ion and multiple reaction monitoring (MRM) acquisition mode. The protonated precursor to product ion transitions monitored for VP, AVA and IS were m/z 351.4→280.2, 323.2→280.2 and 256.2→167.3 respectively. The method was fully validated for its sensitivity, selectivity, accuracy and precision, matrix effect, stability study and dilution integrity. A linear dynamic range of 0.5–500ng/mL for both VP and AVA was evaluated with mean correlation coefficient (r) of 0.9970 and 0.9984 respectively. The precision of the assay (RSD%) was less than 8.55% at all concentrations levels for both VP and AVA. This method was successfully applied to a pharmacokinetic study of VP in rats after intravenous (1mg/kg) and oral (1mg/kg) administration.

Abstract:Theoretical support, but little empirical evidence confirms recurrent abdominal pain (RAP) as a psychosomatic response of children to stressors in their environment. The differences between children with and without RAP in appraising their own stressors and coping abilities were examined. Of 250 8-12 year olds, 25 (10%) experienced recurrent abdominal pain. An asymptomatic comparison group consisted of 25 children matched for age and gender with the RAP group. Mean total stress scores were significantly higher in the RAP group than the comparison group. Mean coping scores were lower in the RAP group, but not statistically significant. Differences in specific stressors and coping strategies between groups were examined. Prevention and treatment of psychosomatic symptoms requires changing the stressor or changing the methods that children use to cope with stressors that cannot be changed.
A Highly Sensitive and Robust UPLC–MS with Electrospray Ionization Method for Quantitation of Taxifolin in Rat Plasma

Abstract:A sensitive ultra performance liquid chromatography–mass spectrometry method has been developed and validated for the quantification of taxifolin in rat plasma. Following liquid/liquid extraction by ethyl acetate, the analytes were separated on a Sunfire™ (2.1mm×50mm, 3.5μm) column and analyzed in the selected ion recording with a negative electrospray ionization mode. The method was linear over the concentration range of 6–6750ng/mL. Intra- and inter-day precisions were all within 8% and accuracy ranged from 92.9% to 105.1%. The lower limit of quantification was 6ng/mL. The present method was successfully applied to the estimation of the pharmacokinetic parameters of taxifolin following intravenous and oral administration to rats. The absolute bioavailability of taxifolin was 0.17% in rat.
Simultaneous determination of amiodarone, dronedarone, and their principal metabolites in SD rat plasma by UPLC-MS/MS and its application in pharmacokinetics

Abstract:The purpose of the experiment was mainly to establish and verify a precise and straightforward ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) approach for simultaneously analyzing the concentration levels of amiodarone, dronedarone, and their metabolites (desethylamiodarone and desbutyldronedarone) in the plasma of Sprague-Dawley (SD) rats and to investigate the pharmacokinetics of all analytes in SD rats. After rapid protein precipitation by using acetonitrile, we accomplished the chromatographic separation of amiodarone, desethylamiodarone, dronedarone, desbutyldronedarone and ivabradine (internal standard, IS) by using Acquity BEH C18 column and detected through a mass spectrometer with Xevo TQ-S triple quadrupole tandem, choosing the positive ion mode. The approach showed wonderful linearity, and the range of calibration curve for amiodarone was 1-200 ng/mL, desethylamiodarone was 0.1-20 ng/mL, dronedarone was 0.5-100 ng/mL, and desbutyldronedarone was 0.25-50 ng/mL, respectively. For lower limit of quantification (LLOQ), the current method of UPLC-MS/MS can achieve values of 1.0 ng/mL for amiodarone, 0.1 ng/mL for desethylamiodarone, 0.5 ng/mL for dronedarone, and 0.25 ng/mL for desbutyldronedarone, respectively. The accuracy of intra-day and inter-day of all analytes was between -14.8% to 10.9%, while the precision was ≤13.3%. For each substance, the recovery rate was > 82.1%, besides, obvious matrix effect was not found. In all conditions, the stability of all analytes was comfirmed to the plasma sample quantification. In addition, the method of UPLC-MS/MS we developed could also be applied to measure the pharmacokinetic characteristics including amiodarone, desethylamiodarone, dronedarone, and desbutyldronedarone in the plasma of SD rats.

Abstract:A rapid, sensitive and selective liquid chromatography-tandem spectrometry method was developed and validated for determination of paeoniflorin in rat plasma using geniposide as the internal standard. The samples were pretreated with solid-phase extraction using Extract-Clean (TM) cartridges. Separation of paeoniflorin and IS was achieved on a reversed-phase C-18 column (50 x 4.6 mm i.d.) with a mobile phase made up of acetonitrile and 0.05% formic acid (25:75, v/v) at a flow rate of 0.5 mL/min. Detection was carried out on a triple quadrupole tandem mass spectrometer by multiple-reaction monitoring and an electrospray ionization source was employed as the ionization source. The lower limit of quantification obtained was 4 ng/mL (n = 6) using 200 mu L plasma with an accuracy of -3.67% (relative error) and a precision of 4.13% (relative standard deviation). A good linearity was found in the range of 4-1000 ng/mL. The intra- and inter-day relative standard deviations in the measurement of quality control samples 10, 150 and 800 ng/mL ranged from 3.73 to 4.94% and from 4.31 to 6.56%, respectively. The accuracy was from -3.93 to -1.11% in terms of relative error. The analyte and IS were stable in the battery of stability studies. This method was successfully applied to a pharmacokinetic study of paeoniflorin after a single oral administration of 53.36 mg/kg paeoniflorin to rats. Copyright (c) 2005 John Wiley & Sons, Ltd.

Abstract:In this paper, an ultra‐high‐performance liquid chromatography–tandem mass spectrometry (UPLC–MS/MS) method was developed for quantifying the levels of crassicauline A, fuziline, karacoline, and songorine in rat plasma. After processing the rat plasma, the proteins in the plasma were separated by extracting the analytes with acetonitrile–methanol (9:1, v/v). The chromatographic column used was the UPLC HSS T3 column, and the mobile phase (methanol–water with 0.1% formic acid) under a gradient elution profile was used to separate the four compounds, with elution times for each analyte being less than 5 min. Electrospray ionization in positive‐ion mode and operating in multiple reaction monitoring mode was used for quantitative analysis. Crassicauline A, fuziline, karacoline, and songorine were administered to 48 rats (n = 6 per group) orally (5 mg/kg) and intravenously (0.5 mg/kg). The standard curves demonstrated excellent linearity in the range of 1–2500 ng/mL, wherein all r values were greater than 0.99. The UPLC–MS/MS method for the determination of crassicauline A, fuziline, karacoline, and songorine in rat plasma was successfully applied in determining their pharmacokinetics parameters, from which their oral bioavailabilities were calculated to be 18.7%, 4.3%, 6.0%, and 8.4%, respectively.

Determination of Fulvestrant in Rat Plasma by LC–MS–MS: Application to a Pharmacokinetic Study

Abstract:A reversed-phase liquid chromatography coupled to tandem mass spectrometry (LC–MS–MS) method was developed and validated for the determination of fulvestrant in rat plasma. Sample preparation involved a liquid-liquid extraction using 1.0 mL of n -hexane–isopropanol (90:10, v/v ) to extract the analyte from 0.1 mL of rat plasma. The analytes were separated on a phenyl-based column using the mobile phase consisting of methanol/water containing 5 mM ammonium acetate at the flow rate of 0.3 mL min −1 . The analytes were monitored by tandem mass spectrometry under electrospray negative ionization mode. Linear calibration curves were generated over the fulvestrant concentration ranges of 0.05–10.0 ng mL −1 in rat plasma. The accuracy and within- and between-day precisions were within the generally accepted criteria for bioanalytical methods (<15%). This developed and validated assay method was successfully employed to characterize the plasma concentration-time profile of fulvestrant after its intramuscular administration in rats at a dose of 10 mg kg −1 .

Abstract:4-Methyl-piperazine-1-carbodithioc acid 3-cyano-3, 3-diphenylpropyl ester hydrochloride (TM208), a newly synthesized anticancer compound, was quantified using liquid chromatography-tandem mass spectrometry (LC-MS/MS) for the first time. A simple, rapid and sensitive assay method using propranolol as internal standard (IS) after one-step precipitation with acetonitrile was developed and validated to determine TM208 in rat plasma. Separation was achieved on a reverse-phase C(18) column with a mobile phase composed of methanol-water (pH4.0) containing 5 m m ammonium acetate in gradient elution mode. A triple quadrupole tandem mass spectrometer with electrospray ionization source was used as detector and operated by multiple reaction monitoring in the positive ion mode. Calibration curves were linear (r > 0.99) between 0.2 and 500 ng/mL. The quantitative limit was 0.2 ng/mL; reliable precision and accuracy were validated by relative standard deviation values in the range 3.44-13.15% and relative error values between -0.58 and -9.78%. The method was successfully applied to preclinical pharmacokinetic studies of TM208.
LC -MS Determination of Propiverine in Human Plasma

Abstract:Objective:To develop an LC-MS method for the determination of propiverine in human plasma. Method:After addition of internal standard (dipfluzine) and alkalization with potassium phosphate buffer,100 μL of human plasma was extracted by ethyl acetate.The organic layer was evaporated under nitrogen stream.The resid- ual was reconstituted with 100 μL of mobile phase,then a 5μL aliquot was injected into the LC-MS system.The plasma samples were separated on a Shim-pack VP-ODS column 5 μm,150 mm×2.0 mm with a mobile phase of methanol-5 mol·L~(-1) amine acetate buffer (90:10) at a flow rate of 0.20 mL·min~(-1).The analysis was detec- ted by a positive electrospray ionization (ESI) method under selected-ion monitoring (SIM) mode.The SIM ions were m/z 368 ( propiverine,[M+H]~+) and m/z 417 (dipfluzine,[M+H]~+).Results :The retention times for propiverine and dipfluzine were 3.7 min and 3.8 min,respectively,and no endogenous peaks that would interfere with the detection of propiverine was observed.Calibration curves were linear over the range 1-128 ng·mL~(-1) (r 0.99).The lower limit of quantitation was 1 ng·mL~(-1).The extraction recoveries were about 84%.The coefficients of intra-day and inter-day RSD were less than 5% and 9% ,respectively.The relative recoveries were 107% to 110%.In the stability studies,propiverine in human plasma was found to be stable for 4 h at room temperature,for 8 h at room temperature after pretreatment,for 14 days at-20 ℃,and after three freeze-thaw cycles.Conclu- sion:The method is proved to be suitable for the determination of propiverine in human plasma.

Abstract:Moxifloxacin N-sulfate is one of the main metabolites of moxifloxacin in phase II metabolism mediated by sulfotransferases. In this study, a simple, rapid, and sensitive LC/MS/MS method with one-step protein precipitation with methanol was developed and validated to quantify the concentration of moxifloxacin N-sulfate in rat plasma. The chromatographic separation was accomplished by using an Agilent Extend C18 column (4.6×250 mm; 5 μm) with a mobile phase consisting of acetonitrile and distilled water (30:70, v/v) containing 5 mM ammonium formate (pH=8.82 adjusted by ammonia) at a flow rate of 1.0 mL/min. A triple quadrupole tandem mass spectrometer with electrospray ionization source was used as a detector operated by multiple reaction monitoring in the negative-ion mode with m/z 480.2/436.3. The calibration curve was linear ranging from 2 to 200 ng/mL. The intraday and interday precision values (RSD) were less than 7.10%, and the intraday and interday accuracy values (relative error) were within -0.40 to 4.99%. Stabilities of all QC samples were within general assay acceptability criteria according to the U.S. Food and Drug Administration guidelines. No considerable matrix effect was found. A pharmacokinetic study of moxifloxacin N-sulfate after a single oral dose of moxifloxacin in rats was carried out using this new method.
