Abstract:This first serious attempt at an autobiographical accounting has forced me to sit still long enough to compile my thoughts about a long personal and scientific journey. I especially hope that my trajectory will be of interest and perhaps beneficial to much younger women who are just getting started in their careers. To paraphrase from Virginia Woolf's writings in A Room of One's Own at the beginning of the 20th century, "for most of history Anonymous was a Woman." However, Ms. Woolf is also quoted as saying "nothing has really happened until it has been described," a harbinger of the enormous historical changes that were about to be enacted and recorded by women in the sciences and other disciplines. The progress in my chosen field of study-the chemical basis of enzyme action-has also been remarkable, from the first description of an enzyme's 3D structure to a growing and deep understanding of the origins of enzyme catalysis.

Abstract:Since graduating from college as an aspiring structural biologist 30 years ago, I have followed progress in the field with intermittent but genuine interest. To me, structural biology represents 'the road not taken' 1 , as I have instead dedicated my career to other pursuits. Here, I reflect on how keeping in touch with a field I left behind long ago continues to bring me joy and fulfillment. Despite no longer working in structural biology, I felt an unexpected connection to it on 9 October, when I heard the Nobel Prize in Chemistry announcement on the radio as I prepared breakfast. Three researchers — David Baker, Demis Hassabis and John Jumper — had been awarded the prize for pioneering work in protein design and structure prediction. I didn't recognize the names at that point, but I knew of Rosetta and AlphaFold. My mind drifted to memories of staring at stereo protein diagrams, trying — and failing — to see the 3D structures. Then, I went on with my day.

Abstract:I am a bioanalytical chemist using cryo-electron microscopy to investigate biological and nanomaterials. I am interested in the mechanisms by which peptides specifically bind nanoparticles. Additionally, I am interested in the mechanisms by which peptides template the coalescence of atoms into nanoparticles with defined properties. I have always been interested in how drugs work at a molecular level, especially antibiotics. In my opinion, one of the most interesting mechanism-of-action stories in the literature centers around nisin, whose pore forming ability was difficult to recreate in vitro until it was discovered to require lipid II. This paper began as a passion project, for many, to discover new antifungal compounds from nature and diversify our antifungal drug arsenal. I loved working on this project because it was an exciting interdisciplinary story passed on from one researcher to anothereach adding their own diverse perspectives. The aberrant formation of insoluble plaques and fibrils by proteins comprises a pathogenic process in numerous neurodegenerative diseases. Recent proposals link protein aggregation to liquid–liquid phase separation (LLPS). In our current study, we developed a bioluminescent equivalent of a fluorescent intercalating dye, “LUMID”, that switches from blue- to green-light emission upon binding nonspecifically to DNA. LUMID is easily combined with isothermal amplification to enable sensitive, sequence-specific detection of DNA with a smartphone-based readout. My Ph.D. work focused on developing water-soluble azobenzene photoswitches and applying them to photocontrol biological systems. My goal is to work on the interface of chemistry and biology and to use both fields to manipulate and control biological targets to help us unravel their mechanisms or design new therapeutics. Since 1921, insulin has been a life-saving drug for diabetes treatment, but its intricate structure has hindered the development of new types. In my Ph.D. research, we pioneered a phage display method for rapidly selecting novel insulin analogs, using omniligase-1. I have long been fascinated by the tripartite relationship among a host, gut microbes, and small molecules... In this paper, we mined multiple novel gut-derived nisin-like lantibiotics from public gut bacterial genomes and expressed and tested them against human gut pathogens and commensal bacteria. Growing evidence now suggests that RNA plays an active role in genetic regulation via various RNA modifications. My research interest lies in the functional impact of modifications like inosine, m5C, m6A, m1A, and phosphorothioate, both in coding and noncoding RNAs.

Tales from the life and lab of a female social neuroscientist

Abstract:This narrative review charts my unconventional path to becoming a social neuroscientist and describes my research findings - some baffling, some serendipitous, some pivotal - in the field of neuropeptide biology. I trace my childhood as a Bell Labs "brat" to my adolescence as a soccer-playing party girl, to my early days as a graduate student, when I first encountered oxytocin and vasopressin. These two molecules instantly captivated - and held - my attention and imagination. For more than 25 years, a core goal of my research program has been to better understand how these neuropeptides regulate social functioning across a range of species (e.g., meadow voles, mice, squirrel monkeys, rhesus monkeys, and humans), and to translate fundamental insights from this work to guide development of novel pharmacotherapies to treat social impairments in clinical populations. I also discuss my experience of being a woman and a mother in STEM, and identify the important people and events which helped shape my career and the scientist I am today.

Abstract:This personal story recounts the accidental observation, the struggles, the breakthroughs, and the collaborative spirit of a few individuals that led to the discovery that bacterial cells expend energy to effectively fluidize their otherwise "glass-like" cytoplasm and promote the dispersal of large cytoplasmic components. This adventure, which led us into an uncharted world at the intersection of cell biology and condensed matter physics about ten years ago, forever transformed the way I view cells and conduct research.

Abstract:Amphiphilically modified cyclodextrins may form various supramolecular aggregates. Here we report a theoretical study of the aggregation of a few amphiphilic cyclodextrins carrying hydrophobic thioalkyl groups and hydrophilic ethylene glycol moieties at opposite rims, focusing on the initial nucleation stage in an apolar solvent and in water. The study is based on atomistic molecular dynamics methods with a “bottom up” approach that can provide important information about the initial aggregates of few molecules. The focus is on the interaction pattern of amphiphilic cyclodextrin (aCD), which may interact by mutual inclusion of the substituent groups in the hydrophobic cavity of neighbouring molecules or by dispersion interactions at their lateral surface. We suggest that these aggregates can also form the nucleation stage of larger systems as well as the building blocks of micelles, vesicle, membranes, or generally nanoparticles thus opening new perspectives in the design of aggregates correlating their structures with the pharmaceutical properties.

Abstract:This article reflects on the author's career journey, from beginnings in southern Chile to leading a research lab. It emphasizes the importance of relatable role models, resilience and leveraging personal strengths to navigate challenges. The piece offers advice for young scientists and draws parallels between managing a lab and coaching a soccer team. Role models play a crucial role in inspiring and guiding careers in science, offering tangible examples of success and resilience. Reflecting on my journey from a small town in southern Chile to leading a lab at Karolinska Institutet, I've learned that relatable role models are particularly impactful for overcoming imposter syndrome and fostering a sense of belonging in academia. Early in my career, I drew inspiration from peers and mentors, gradually building my confidence and embracing my strengths. Later, exposure to interdisciplinary role models expanded my horizons and shaped my approach to science. Now, as a PI, I see my role as both a coach and mentor, fostering a team dynamic that amplifies individual strengths. Success in science often stems from fearlessness, adaptability and a willingness to seize opportunities, even when the outcome is uncertain. My journey demonstrates that good scientists can come from anywhere, including a small town in southern Chile.

Abstract:Xuemei Chen is a distinguished biologist. She was elected a member of the US National Academy of Sciences in 2013, and is now the Dean of the School of Life Sciences at Peking University. Her main research areas are flower development, plant microRNAs, and RNA modifications. In this interview, Xuemei Chen discussed the prospects of plant research, and emphasized the significance of free exploration to achieve the ground-breaking '0 to 1' innovation-until free exploration leads to the discovery of the '1', one may not know what the '0' is and may not be able to intentionally seek it. She also introduced the upcoming Beijing Advanced Center of RNA Biology (BEACON), and expressed her views on organized scientific research.

Abstract:The DNA nanotechnology outlines a new chapter in biological sciences, opening a new era of scientific and technological advancements. A pioneer of DNA nanotechnology, Professor Liu from the University of Stuttgart in Germany, our latest Light People, has been working at the interface, where nanophotonics meets biology and chemistry. On the path of scientific explorations, she believes in hard working and persistency, as indicated by a line of Chinese poetry she likes, "Don't stop chasing the wind and the moon, the spring mountain is at the end of the plain." She is an outstanding female researcher, radiating wisdom. She is a pioneer in exploring the power of DNA nanotechnology and applying it to other disciplines. Meanwhile, she struggles with work–life balance. Apart from being determined and persistent, Prof. Liu is also introspective. One sees the brilliance of a scientist and the struggles of a mother, as Prof. Laura Na Liu explains to us her very personal "philosophy of life".

Abstract:I was shaped by World War II, years of near starvation as a war refugee, postwar chaos, life in several countries, and relative affluence in later life. The truth is that as I was growing up I wanted to be a writer. My aspirations came to an end when, in order to speed up my graduation from college, I took courses in microbiology. It was my second love at first sight-that of my wife preceded it. I view science as an opportunity to discover the designs in the mosaics of life. What initiates my search of discovery is an observation that makes no sense unless there exists a novel design. Once the design is revealed there is little interest in filling all the gaps. I was fortunate to understand that what lasts are not the scientific reports but rather the generations of scientists whose education I may have influenced.

Abstract:Similar to Dorothy's journey along the yellow brick road in The Wizard of Oz, this perspective carves out the path I took from my early childhood fascinations with science through my independent career at the University of Michigan (maize and blue). The influential research projects and mentors are highlighted, including some fortuitous experimental results that drew me into the field of supramolecular chemistry, specifically, and organic materials, broadly. My research group's efforts toward designing new sensors based on small molecule gelators are described. In particular, I highlight how our design strategy has evolved as we learn more about molecular gelators. This perspective concludes with some predictions about where molecular gels, as well as my personal and professional life, are headed.

Abstract:The Research and Engineering Apprenticeship Program of the Academy of Applied Science has funded several high school student summer internships to work within the Department of Chemistry at the University of Texas at El Paso. Over the last nine years, young Mexican-American scholars have been recruited into STEM-specific (science, technology, engineering, and mathematics) laboratories to cultivate and nurture their interest in research. This commentary describes vignettes from a successful program in which coauthoring students are paired with a graduate student to advance a molecular-level understanding of biomedical intervention by traditional phytoremedials (plant-based remedies) in neurodegenerative disease processes. Considering that the selected phytoremedials originate from Mexico, Latin America, and the Indian subcontinent and are familiar folklore (kitchen theraceuticals) to the participating Hispanic and Indian scholars, the research project provides an enhanced sense of importance, ownership, and enthusiasm. Eventually, it cements the bridge to a future STEM-related college education, engages in the nation’s STEM capacity-building mission, and contributes to the nation’s Hispanic science and technology workforce of tomorrow.

Abstract:This manuscript describes the winding career path of Dr Johnathan Canton – an Assistant Professor at the University of Calgary and recent recipient of the Bhagirath Singh Early Career Award from the Canadian Institutes of Health. My path to becoming a scientist has taken many twists and turns. This is perhaps not unusual to hear. Indeed, in discussions with my colleagues it seems that for many of us the path was never a straight one. Certainly, for me there have been moments when my whole world was encompassed by science and at other times, I have felt strongly that my time in science was up. I like to think that as scientists we ask a lot of questions and, for many of us, those questions extend to our very purpose as a scientist. My intention with this article is not to document my career path in detail or to provide very specific advice. Rather, I hope to describe how questions have defined my journey and to inspire others to occasionally pause and ask themselves what a career in science means to them. Today, I am an Assistant Professor at a major Canadian university, and here are the questions I asked along the way.

Abstract:More than three decades ago, we embarked on a number of bioengineering explorations using the most advanced materials and fabrication methods. In every area we ventured into, it was our intention to ensure fundamental discoveries were deployed into the clinic to benefit patients. When we embarked on this journey, we did so without a road map, not even a compass, and so the path was arduous, sometimes tedious. Now, we can see the doorway to deployment on the near horizon. We now appreciate that overcoming the challenges has made this a rewarding and exciting journey. However, maybe we could have been here a lot sooner, and so maybe the lessons we have learned could benefit others and accelerate progress in clinical translation. Through a number of case studies, including neural regeneration, cartilage regeneration, skin regeneration, the 3D printing of capsules for islet cell transplantation, and the bioengineered cornea, here, we retrace our steps. We will summarise the journey to date, point out the obstacles encountered, and celebrate the translational impact. Then, we will provide a framework for project design with the clinical deployment of bioengineered products as the goal.

Abstract:Emphasis herein is on the early years of my scientific life, primarily in graduate school and at the McCollum-Pratt Institute, Johns Hopkins University, as techniques learned and research performed then became the basis for future scientific endeavors. Studies on the mechanism of conversion of light energy into chemical free energy were a logical consequence of earlier investigations on enzyme-catalyzed hydrogen transfer reactions and pyridine nucleotide coenzyme biochemistry. Identification of several protein factors involved in pyridine nucleotide reduction by illuminated chloroplasts is described and, hopefully, adequately and honestly referenced to complementary research in other laboratories. Coupled with progress were changes in nomenclature of the protein factors and are so noted. In particular, David Wharton proposed the descriptive name, ferredoxin, for the non-heme iron and labile sulfide-containing proteins which serve as redox cofactors in a variety of energy conserving reactions. The inclusion of "Lessons" is adapted from Efraim Racker (1976, A new look at mechanisms in bioenergetics. Academic Press, NY). They are lessons that I learned and are included herein solely for graduate students.
A journey from speech to dance through the field of oxytocin

Abstract:In this article, I am going through my scientific and personal journey using my work on oxytocin as a compass. I recount how my scientific questions were shaped over the years, and how I studied them through the lens of different fields ranging from linguistics and neuroscience to comparative and population genomics in a wide range of vertebrate species. I explain how my evolutionary findings and proposal for a universal gene nomenclature in the oxytocin-vasotocin ligand and receptor families have impacted relevant fields, and how my studies in the oxytocin and vasotocin system in songbirds, humans and non-human primates have led me to now be testing intranasal oxytocin as a candidate treatment for speech deficits. I also discuss my projects on the neurobiology of dance and where oxytocin fits in the picture of studying speech and dance in parallel. Lastly, I briefly communicate the challenges I have been facing as a woman and an international scholar in science and academia, and my personal ways to overcome them.

Abstract:Climate change is driving extreme changes to the environment, posing substantial threats to global food security and bioenergy. Given the direct role of plant roots in mediating plant-environment interactions, engineering the form and function of root systems and their associated microbiota may mitigate these effects. Synthetic genetic circuits have enabled sophisticated control of gene expression in microbial systems for years and a surge of advances has heralded the extension of this approach to multicellular plant species. Targeting these tools to affect root structure, exudation, and microbe activity on root surfaces provide multiple strategies for the advancement of climate-ready crops.
