Abstract:Amphiphilically modified cyclodextrins may form various supramolecular aggregates. Here we report a theoretical study of the aggregation of a few amphiphilic cyclodextrins carrying hydrophobic thioalkyl groups and hydrophilic ethylene glycol moieties at opposite rims, focusing on the initial nucleation stage in an apolar solvent and in water. The study is based on atomistic molecular dynamics methods with a “bottom up” approach that can provide important information about the initial aggregates of few molecules. The focus is on the interaction pattern of amphiphilic cyclodextrin (aCD), which may interact by mutual inclusion of the substituent groups in the hydrophobic cavity of neighbouring molecules or by dispersion interactions at their lateral surface. We suggest that these aggregates can also form the nucleation stage of larger systems as well as the building blocks of micelles, vesicle, membranes, or generally nanoparticles thus opening new perspectives in the design of aggregates correlating their structures with the pharmaceutical properties.

Abstract:This article reflects on the author's career journey, from beginnings in southern Chile to leading a research lab. It emphasizes the importance of relatable role models, resilience and leveraging personal strengths to navigate challenges. The piece offers advice for young scientists and draws parallels between managing a lab and coaching a soccer team. Role models play a crucial role in inspiring and guiding careers in science, offering tangible examples of success and resilience. Reflecting on my journey from a small town in southern Chile to leading a lab at Karolinska Institutet, I've learned that relatable role models are particularly impactful for overcoming imposter syndrome and fostering a sense of belonging in academia. Early in my career, I drew inspiration from peers and mentors, gradually building my confidence and embracing my strengths. Later, exposure to interdisciplinary role models expanded my horizons and shaped my approach to science. Now, as a PI, I see my role as both a coach and mentor, fostering a team dynamic that amplifies individual strengths. Success in science often stems from fearlessness, adaptability and a willingness to seize opportunities, even when the outcome is uncertain. My journey demonstrates that good scientists can come from anywhere, including a small town in southern Chile.

Abstract:This manuscript describes the winding career path of Dr Johnathan Canton – an Assistant Professor at the University of Calgary and recent recipient of the Bhagirath Singh Early Career Award from the Canadian Institutes of Health. My path to becoming a scientist has taken many twists and turns. This is perhaps not unusual to hear. Indeed, in discussions with my colleagues it seems that for many of us the path was never a straight one. Certainly, for me there have been moments when my whole world was encompassed by science and at other times, I have felt strongly that my time in science was up. I like to think that as scientists we ask a lot of questions and, for many of us, those questions extend to our very purpose as a scientist. My intention with this article is not to document my career path in detail or to provide very specific advice. Rather, I hope to describe how questions have defined my journey and to inspire others to occasionally pause and ask themselves what a career in science means to them. Today, I am an Assistant Professor at a major Canadian university, and here are the questions I asked along the way.

Abstract:I was shaped by World War II, years of near starvation as a war refugee, postwar chaos, life in several countries, and relative affluence in later life. The truth is that as I was growing up I wanted to be a writer. My aspirations came to an end when, in order to speed up my graduation from college, I took courses in microbiology. It was my second love at first sight-that of my wife preceded it. I view science as an opportunity to discover the designs in the mosaics of life. What initiates my search of discovery is an observation that makes no sense unless there exists a novel design. Once the design is revealed there is little interest in filling all the gaps. I was fortunate to understand that what lasts are not the scientific reports but rather the generations of scientists whose education I may have influenced.

Abstract:Abstract &#160; &#160;With an aim to inspire young scientists, the idea of motivational journeys came into the picture. There are plenty of online materials available in the form of books, podcasts, animations and videos. However, these materials can lack the field specific knowledge and thus young scientists find it more difficult to relate to. In the &#8216;Motivational Journeys&#8217; series we have share the struggles and advice of experienced scientists who are currently working in planetary science with the aim of inspiring early career researchers in our field. &#160;&#160; Project and philosophy The idea for the &#8216;Motivational Journeys&#8217; series came from a personal experience. Being an international student, working towards a PhD project in a different country away from my comfort zone has affected my self-confidence and ability to focus. Under such challenging conditions, I either chose to read a book on my favourite subject or try to watch short movies or series based on autobiographies to look for inspiration. After following this approach for some time, I realised rather than looking for the motivation outside, why not get it within my own professional field?&#160; With this thought, I started to plan the project and further discussed with my colleagues and peers too who positively supported the idea. We planned a short series of interviews in which experienced scientist could share their stories of how they overcame the more difficult hurdles during their early career span. They told us how, with their passion for science and dedication and &#8216;never give up&#8217; attitude, they conquered the difficulties and pushed forward their professional career. In this series, we aimed to have a dialogue and discussion on various aspects of struggles. All the scientist we interviewed came from a diverse range of backgrounds and cultures. However, the passion and struggles described by the interviewees to achieve their dreams connects each Motivational Journey to the others in the series. Today, when so many early career professionals face issues related to mental health and self-esteemed, we hope that this series of interviews may help to them to find out how others have approached related struggles and to motivate them in to overcome difficulties. We successfully recorded 8 interviews during the EPSC-DPS Joint Meeting 2019 in Geneva. Our first interview was launched in October 2019 and since then we released one interview per month up to May 2020. Abstract:When I reflect on how I became a cell biologist and why I love being one today, one thing that comes to mind is the many terrific collaborations I have had. The science I am most proud of from my graduate and postdoctoral training would not have been possible without working in teams with other scientists. Now, in my own group, much of our best work is being done collaboratively, both within the lab and with other labs. In this essay, I will highlight my experiences working in teams as a trainee, the role teamwork has played in my own research group, and how important I think collaborative science is for the future of biological research.

Abstract:This first serious attempt at an autobiographical accounting has forced me to sit still long enough to compile my thoughts about a long personal and scientific journey. I especially hope that my trajectory will be of interest and perhaps beneficial to much younger women who are just getting started in their careers. To paraphrase from Virginia Woolf's writings in A Room of One's Own at the beginning of the 20th century, "for most of history Anonymous was a Woman." However, Ms. Woolf is also quoted as saying "nothing has really happened until it has been described," a harbinger of the enormous historical changes that were about to be enacted and recorded by women in the sciences and other disciplines. The progress in my chosen field of study-the chemical basis of enzyme action-has also been remarkable, from the first description of an enzyme's 3D structure to a growing and deep understanding of the origins of enzyme catalysis.

Abstract:Since graduating from college as an aspiring structural biologist 30 years ago, I have followed progress in the field with intermittent but genuine interest. To me, structural biology represents 'the road not taken' 1 , as I have instead dedicated my career to other pursuits. Here, I reflect on how keeping in touch with a field I left behind long ago continues to bring me joy and fulfillment. Despite no longer working in structural biology, I felt an unexpected connection to it on 9 October, when I heard the Nobel Prize in Chemistry announcement on the radio as I prepared breakfast. Three researchers — David Baker, Demis Hassabis and John Jumper — had been awarded the prize for pioneering work in protein design and structure prediction. I didn't recognize the names at that point, but I knew of Rosetta and AlphaFold. My mind drifted to memories of staring at stereo protein diagrams, trying — and failing — to see the 3D structures. Then, I went on with my day.

Abstract:What sets someone on a life trajectory? This question is at the heart of studies of 21st-century neurosciences that build on scientific models developed over the last 150 years that attempt to link psychopathology risk and human development. Historically, this research has documented persistent effects of singular, negative life experiences on people's subsequent development. More recently, studies have documented neuromolecular effects of early life adversity on life trajectories, resulting in models that frame lives as disproportionately affected by early negative experiences. This view is dominant, despite little evidence of the stability of the presumably early-developed molecular traits and their potential effects on phenotypes. We argue that in the context of gaps in knowledge and the need for scientists to reason across molecular and phenotypic scales, as well as time spans that can extend beyond an individual's life, specific interpretative frameworks shape the ways in which individual scientific findings are assessed. In the process, scientific reasoning oscillates between understandings of cellular homeostasis and organisms' homeorhesis, or life trajectory. Biologist and historian François Jacob described this framework as the "attitude" that researchers bring to bear on their "objects" of study. Through an analysis of, first, historical and contemporary scientific literature and then ethnographic research with neuroscientists, we consider how early life trauma came to be associated with specific psychological and neurobiological effects grounded in understandings of life trajectories. We conclude with a consideration of the conceptual, ontological, and ethical implications of interpreting life trajectories as the result of the persistence of long-embodied biological traits, persistent life environments, or both.
Focus on People and the Science Will Follow: Motivating Forces for Professional Movement in Stem Cell Research

Abstract:The migration of researchers across geographic borders, or "brain drain" as it is commonly called, remains an important issue for governments around the world as loss or gain of highly qualified personnel in research can have substantial social, economic and political consequences. In the present study we seek to examine the forces that drive international professional migration of stem cell (SC) researchers, for which variation of SC policy in different jurisdictions has previously been implicated as a driving force. Structured interviews were carried out with a purposive sample of SC researchers in the professoriate who had made international moves after postdoctoral work between the years 2001-2014, or were actively anticipating a future move. Participants were asked to rank motivators of international movement on a 5-point Likert scale and prompted to elaborate on their answers. The results suggest that career considerations, availability of research funding, and personal considerations are of high importance to the participants when considering an international move, while the permissiveness or restrictiveness SC research policy is of comparably lower importance. Participants also expressed that international movements are beneficial to scientific careers overall. The findings have important implications for policy and strategies to attract and retain members of the SC research community.

Abstract:More than three decades ago, we embarked on a number of bioengineering explorations using the most advanced materials and fabrication methods. In every area we ventured into, it was our intention to ensure fundamental discoveries were deployed into the clinic to benefit patients. When we embarked on this journey, we did so without a road map, not even a compass, and so the path was arduous, sometimes tedious. Now, we can see the doorway to deployment on the near horizon. We now appreciate that overcoming the challenges has made this a rewarding and exciting journey. However, maybe we could have been here a lot sooner, and so maybe the lessons we have learned could benefit others and accelerate progress in clinical translation. Through a number of case studies, including neural regeneration, cartilage regeneration, skin regeneration, the 3D printing of capsules for islet cell transplantation, and the bioengineered cornea, here, we retrace our steps. We will summarise the journey to date, point out the obstacles encountered, and celebrate the translational impact. Then, we will provide a framework for project design with the clinical deployment of bioengineered products as the goal.

Abstract:A hybrid molecular mechanics-statistical mechanics-solvent accessibility-based computational protocol is developed to calculate DNA-ligand binding affinity without any database training and is validated on 50 DNA-ligand complexes. The calculated binding energies yield high correlation coefficients of 0.95 (R2 = 0.90) and 0.96 (R2 = 0.93) in linear plots against experimental binding free energies (DeltaGo) and DeltaTm, respectively. The protocol is translated into a swift, web-enabled, freely accessible computational tool, http://www.scfbio-iitd.res.in/preddicta, for DeltaGo and DeltaTm prediction for DNA-ligand complexes to aid and expedite rational drug design attempts.
