Abstract:Aims: To investigate an in-line Raman method capable of detecting accidental microbial contamination in pharmaceutical vessels, such as bioreactors producing monoclonal antibodies via cell culture. Methods and results: The Raman method consists of a multivariate model built from Raman spectra collected in-line during reduced-scale bioreactor batches producing a monoclonal antibody, as well as a reduced-scale process with intentional spiking of representative compendial method microorganisms (n = 4). The orthogonal partial least squares regression discriminant analysis model (OPLS-DA) area under the curve (AUC), specificity and sensitivity were 0.96, 0.99, and 0.95, respectively. Furthermore, the model successfully detected contamination in an accidentally contaminated manufacturing-scale batch. In all cases, the time to detection (TTD) for Raman was superior compared to offline, traditional microbiological culturing. Conclusions: The Raman OPLS-DA method met acceptance criteria for equivalent decision making to be considered a viable alternative to the compendial method for in-process bioburden testing. The in-line method is automated, non-destructive, and provides a continuous assessment of bioburden compared to an offline compendial method, which is manual, results in loss of product, and in practice is only collected once daily and requires 3-5 days for enumeration.
Infrared thermography can detect previsual bacterial growth in a laboratory setting via metabolic heat detection

Abstract:Aims: Detection of bacterial contamination in healthcare and industry takes many hours if not days. Thermal imaging, the measurement of heat by an infrared camera, was investigated as a potential noninvasive method of detecting bacterial growth. Methods and results: Infrared thermography can detect the presence of Escherichia coli and Staphylococcus aureus on solid growth media by an increase in temperature before they are visually observable. A heat decrease is observed after treatment with ultraviolet light and heat increased after incubation with dinitrophenol. Conclusions: Infrared thermography can detect early growth of bacteria before they are detectable by other microbiology-based method. The heat observed is due to the cells being viable and metabolically active, as cells killed with ultraviolet light exhibit reduced increase in temperature and treatment with dinitrophenol increases heat detected. Significance and impact of the study: Infrared thermography detects bacterial growth without the need for specialized temperature control facilities. The method is statistically robust and can be undertaken in situ, thus is highly versatile. These data support the application of infrared thermography in a laboratory, clinical and industrial setting for vegetative bacteria, thus may become into an important methodology for the timely and straightforward detection of early-stage bacterial growth.
Non-Contact, Real-Time Laser-Induced Fluorescence Detection and Monitoring of Microbial Contaminants on Solid Surfaces Before, During and After Decontamination

Abstract:A real-time detection and monitoring (RTDM) of microbial contamination on solid surfaces is mandatory in a range of security, safety and bio-medical applications where surfaces are exposed to accidental, natural or intentional microbial contamination. This work presents a new device, the BC-Sense, which allows a rapid and user-friendly RTDM of microbial contamination on various surfaces while assessing the decontamination kinetics and degree of cleanliness. The BC-Sense LIDAR (Light Detection and Ranging) device uses the Laser-Induced Fluorescence (LIF) method based on dual wavelength sensing with multispectral pattern recognition system to rapidly detect microbial contamination on a solid surface. Microbial simulants (bacteria, bacterial spores, fungal conidia and virus) were spread at varying concentrations on a panel of solid surfaces which were assessed by BC-Sense. The spectra of dead and living E. coli showed differences at various sensing wavelengths. The limit of detection (LoD) of E. coli and MS2 virus was 2.9 × 104 and 9.5 × 104 PFU and CFU/cm2, respectively. Random samples (n=200) tested against a training dataset (n=800) were optimally discriminated for contamination versus background with a threshold of predicted response (PR) >0.55 and <0.4, respectively. Decontamination kinetics on copper surface showed a complete disappearance of fluorescence in 1 min with MS2 versus >10 min with spores and E. coli.
Application of non-radioactive BACTEC system to sterility testing and hygiene

Abstract:The applicability of the non-radioactive BACTEC NR 660 system for the detection of low viable counts of aerobic and anaerobic bacteria (including non-fermenters and aerobic spore-forming bacteria) and fungi was investigated. The BACTEC system together with the usual BACTEC NR 6A and NR 7A media was able to detect most of the organisms without supplementation of the media; for growth of lactobacilli and other acidophilic organisms, sterile orange juice was used as a supplement. Microbial growth was detected from pure cultures and also from artificially contaminated beverages and ointment bases. Detection times varied according to the types of metabolism of the respective strains and their initial viable counts, but usually they were detected after 20 to 42 h of incubation. The BACTEC NR 660 system seems to be applicable not only for blood cultures in medical microbiology but also in pharmaceutical and food industry.
Comparison of time-to-detection of Mindray TDR and BacT/ALERT®3D blood culture systems using simulated blood cultures

Abstract:Purpose: Blood culture (BC) is the standard for diagnosing bloodstream infections. Available blood culture (BC) systems have been developed to shorten the time to detection (TTD) of positive BCs. This study aimed to evaluate the performance of the Mindray TDR automatic BC system by comparing it with the BacT/ALERT®3D system. Methods: Sixteen reference strains and 14 clinical isolates were used. Serial dilutions were prepared from all bacterial and yeast colonies with a final concentration of 100 CFU/ml and 10 CFU/ml. The prepared solutions were simultaneously inoculated into the bottles of both systems and placed in blood culture devices. Results: Three hundred and fifty-two (176 BacT/ALERT®3D and 176 Mindray TDR-X060) blood culture bottles were evaluated, 336 aerobic and 16 anaerobic. At both 10 CFU/ml and 100 CFU/ml dilution, there was no significant difference between the two systems in terms of mean detection times for all isolates (p = 0.965, p = 0.245). When evaluated according to the type of organism, the detection time of gram-positive bacteria at 10 CFU/ml dilution was significantly shorter in the BacT/ALERT system (p = 0.019), whereas detection time for yeasts was significantly shorter with the Mindray system (p = 0.047). The number of anaerobic bacteria was too small to draw statistical conclusions, but we observed a trend of shorter detection times in the Mindray TDR-X060 system. Conclusion: Two systems with similar operating principles showed different concentrations-dependent performances in terms of positivity detection times depending on the type of microorganism. Mindray TDR-X060 system has been found to be safe to use at high concentrations with this at lower concentrations further comparative studies are needed on the newly introduced Mindray system.

Abstract:The emergence of antibacterial resistance impacts healthcare networks globally, with mortality rates and linked burdens of infection disproportionately affecting the developing world. Rapid alternatives to antibiotic susceptibility testing (AST) allow for swifter, more effective treatment, though they are limited in use in low-resource settings due to significant cost barriers. Herein we demonstrate a simple, cost-effective diffraction sensing-based approach for rapidly detecting bacterial growth (a precursor to AST). Diffraction gratings (1D, lined) directly comprised of our test bacteria ( Escherichia coli DH5α) were produced using soft agar-based gel templates designed to direct bacterial attachment and produce a near-zero background signal. The diffraction spot intensities from the live bacterial gratings were monitored in growth and no growth (ampicillin) conditions at room temperature, using a simple fixed laser and photodetector setup. Growth-induced differences in signal were observed within 10–20 minutes, highlighting the sensitivity of this approach and its potential to be adapted as a rapid and accessible AST alternative.

Abstract:Prophylactic antibiotics should be given as close to the time of incision as possible to ensure that tissue antimicrobial levels are adequate and maintained for the duration of the procedure. The choice of antibiotic should be based on the organisms most likely to be encountered--usually staphylococcal skin florae. The choice of vancomycin over a cephalosporin may be justified in patients who are known carriers of MRSA. A full therapeutic dose of antibiotic should be used for prophylaxis. Morbidly obese patients should be given twice the standard dose. Redosing during an operation is recommended if the duration of the procedure exceeds two half-lives of the antibiotic administered. Prophylactic antibiotics should not continue to be administered more than 48 hours postoperatively.

The goal of the research was to determine experimentally and compare the LODs of microorganisms by direct inoculation and using a BacT/ALERT 3D Microbial Detection System automated bacteriological analyzer. Statistically significant differences between the results obtained by the different methods were not found.

Abstract:The current status of microbiological testing methods for the determination of viable bacteria in complex sample matrices, such as food samples, is the focus of this review. Established methods for the enumeration of microorganisms, particularly, the 'gold standard' agar plating method for the determination of total aerobic viable counts (TVC), bioluminescent detection of total ATP, selective molecular methods (immunoassays, DNA/RNA amplification, sequencing) and instrumental methods (flow cytometry, Raman spectroscopy, mass spectrometry, calorimetry), are analyzed and compared with emerging oxygen sensor-based respirometry techniques. The basic principles of optical O2 sensing and respirometry and the primary materials, detection modes and assay formats employed are described. The existing platforms for bacterial cell respirometry are then described, and examples of particular assays are provided, including the use of rapid TVC tests of food samples and swabs, the toxicological screening and profiling of cells and antimicrobial sterility testing. Overall, O2 sensor-based respirometry and TVC assays have high application potential in the food industry and related areas. They detect viable bacteria via their growth and respiration; the assay is fast (time to result is 2–8 h and dependent on TVC load), operates with complex samples (crude homogenates of food samples) in a simple mix-and-measure format, has low set-up and instrumentation costs and is inexpensive and portable.

Abstract:Adequate food packaging is important to ensure food quality and prolong the shelf life of food. Frequently, the method of modified atmosphere packaging (MAP) is used. We used a tunable diode laser spectroscopy technique to detect the oxygen content in food packages, e.g., in packaged milk and bread. Different detection geometries were adopted to be able to collect scattering light that had passed through the gas volume in the packages, and to avoid the disturbance of ambient air. Furthermore, studies of the bread-baking process have been performed by measuring the water vapor signals in a fermenting dough. The results demonstrate that the technique has a great potential for nonintrusive assessment of food quality and the bread-baking process.

Abstract:BackgroundAcute treatment and long-term secondary prevention of noncardioembolic ischemic stroke and transient ischemic attack (TIA) include initiation of antiplatelet therapy. Antiplatelet agents currently used in the treatment of ischemic stroke and TIA are aspirin, clopidogrel, and dipyridamole. Review SummaryThe safety and efficacy of antiplatelet therapy in patients with ischemic stroke, including a discussion of recent trial data and its influence on treatment guidelines, are presented. A brief discussion of the use of antiplatelet therapy in preventing stroke and embolism in patients with atrial fibrillation is also presented. For secondary prevention of ischemic events in patients with a history of stroke, clinical trials have shown the addition of dipyridamole to aspirin to be more effective than aspirin alone. The therapies are also similar from a standpoint of bleeding. The combination of aspirin and clopidogrel was not shown to be more efficacious and caused more bleeding than aspirin alone when evaluated for secondary prevention. However, dual antiplatelet therapy with aspirin and clopidogrel may have some benefit in the acute stroke setting or in the prevention of thrombotic events in patients with atrial fibrillation who cannot or will not take warfarin. ConclusionsAntiplatelet therapy is an important component of acute and long-term treatment of ischemic stroke and TIA. Ongoing clinical trials may help to refine what treatment regimens are best suited for acute and long-term therapy.

Abstract:Timely pathogen identification in bloodstream infections is crucial for patient care. A comparison is made between positive blood culture (BC) pellets from serum separator tubes using a direct identification (DI) method and colonies on agar plates from a short-term incubation (STI) method with a matrix-assisted laser desorption/ionization Biotyper for the evaluation of 354 monomicrobial BCs. Both the DI and STI methods exhibited similar identification rates for different types of bacteria, except for Gram-positive and anaerobic bacteria. The DI method's results aligned closely with the STI method's results for Enterobacterales, glucose-non-fermenting Gram-negative bacilli (GNB), and carbapenem-resistant Enterobacterales. The DI method exhibited high concordance with the conventional method for GNB identification, achieving 88.2 and 87.5% accuracy at the genus and species levels, respectively. Compared with the STI method, the DI method showed a less successful performance for Gram-positive bacterial identification (50.5 vs. 71.3%; p < 0.01). The DI method was useful for anaerobic bacterial identification of slow-growing microorganisms without any need for colony growth, unlike in the STI method (46.7 vs. 13.3%; p = 0.04). However, both methods could not identify yeast in positive BCs. Overall, the DI method provided reliable results for GNB identification, offering many advantages over the STI method by significantly reducing the turnaround time and enabling quicker pathogen identification in positive BCs.

Abstract:The interactions of host cells and fungi during infection represent a complex interplay. Although T helper 1 (Th1)-mediated immunity is primarily responsible for acquired resistance to Paracoccidioides brasiliensis, studies have demonstrated that polymorphonuclear neutrophils play a critical role in providing an early resistance to this organism. One study has shown that the invasiveness of Candida albicans requires adherence, particularly to endothelial cells, which in turn are stimulated to express various cell-markers and pro-inflammatory cytokines as part of a proactive resistance to invasion. Somewhat in contrast to infection with C. albicans, it has been shown that the capsular glucuronoxylomannan of Cryptococcus neoformans causes the shedding of host-cell adherence molecules (L-selectins) needed for the migration of host-inflammatory cells to sites of infection and likely explains, in part, the reduced host inflammatory response to this organism. Resistance to aspergillosis is often associated with the immune status of the host. In one set of studies, it has been demonstrated that lymphocytes have little direct effect on the organism, but that antigen-presenting dendritic cells stimulate the production of Th1 cytokines, suggesting a positive role for the dendritic cell in host-response. Similarly, another study has shown that among the regulatory cytokine networks that Th2-associated cytokines (e.g., interleukin-10) likely play a detrimental role in the resistance of the host to Aspergillus fumigatus.

Abstract:Microbial density is an important biochemical parameters of the training process. This article adopts the method of online measuring optical density of the reactor to obtain information on the growth of microorganisms, and flow photoelectric colorimetric instrument was developed based on this principles. Through MCU STM32F107VC output PWM to control peristaltic pump, to achieve the culture of continuous flow through the optical flow cuvette, using photocell to detect culture absorbed light intensity. The instrument can display detected parameters independently and adjust the control parameters, also can communicate with the PC by RS485. The instrument can achieve real-time monitoring of the growth of microorganisms in the reactor.

Abstract:BACKGROUND: Automated growth-based methods for sterility testing of cell-therapy products should be qualified to demonstrate that they are equivalent to, or better than, the conventional reference method. The aim of the present study was to assess the ability of the BACTEC FX40 system to detect low microbial contamination and to confirm the suitability of the method in the presence of seven different human mesenchymal cell-based products, according to Ph. Eur. 2.6.27. Additionally, a study to select the best vial to detect fungus contamination was performed.METHODS: Microorganisms representing Gram-negative, Gram-positive, aerobic, anaerobic, spore-forming, slow-growing bacteria, yeast and mold were prepared in either Dulbecco's PBS or seven biological matrices containing approximately 5, 10, and 15 colony-forming units (CFU) per sample. These preparations were inoculated to the specific media required for each test method: (i) BACTEC aerobic and anaerobic vials; (ii) aerobic and anaerobic media for direct inoculation; and (iii) Trypcase soy 3P or Brucella blood agar plates. Colonies from cultures were identified using MALDI-TOF mass spectrometry.RESULTS: The BACTEC FX40 system, in both Dulbecco's PBS and the biological matrices with a 5-CFU inoculum, detected most of the microorganisms significantly faster than the conventional method, despite the presence of a matrix containing gentamicin and several matrices containing 10% DMSO. Conversely, it showed an extremely delayed detection of Candida albicans compared with the conventional method. The addition of a Mycosis IC/F (MYC) vial decreased radically the time to detection (TTD) of C. albicans (28.2 ± 1.8 h) compared with the conventional method (36 h).CONCLUSIONS: When a MYC vial was added to the standard aerobic and anaerobic vials to test each sample, BACTEC FX40 was shown to be a superior alternative sterility method for cell-therapy products contaminated with low inocula, with a faster TTD for microbial growth compared with the conventional method (5 versus 14 days). The studies were carried out in different cell-based matrices with sensitivities and specificities of 100% for all the tested strains at 15-, 10- and 5-CFU inoculum, with the exception of Kocuria rhizophila at 5 CFU (90.48% sensitivity and 100% specificity).

Abstract:(1) Background: Due to the aging population in industrialized countries and due to the increase in the number of traffic or sports accidents, the number of artificial joints and implants for osteosynthesis will increase in the coming years. Therefore, the risk of postoperative infections will be higher as well. (2) Methods: For this study, we combined classical bacterial identification with the description of bacterial growth curves using microcalorimetry. (3) Results: We evaluated the growth of S. aureus and S. epedermidis, but we believe that this can be applied to any anaerobic or aerobic bacterial colony. We discovered that the time interval after which we can identify a growth curve does not exceed 15–20 h. (4) Conclusions: The diagnosis made by combining the methods of sonication and microcalorimetry manages to provide a great deal of information about the bacteria we studied. Microcalorimetry has real potential as a method for obtaining quick diagnosis in various cases of infection, but many more experiments need to be done to ensure the correct use of this technique. A detailed investigation (including kinetic analysis) of the reproducible thermal signal of bacterial growth can lead to the development of alternative means of rapid bacterial identification.

Abstract:Liquid micro-respirometry, Liq-μRM, is a popular, more rapid alternative method for measuring Total Viable Count of aerobes, i.e., TVC, units: colony forming units (CFU)/mL, to the traditional, time-consuming, plate counting method. Liq-μRM is based on monitoring the consumption of dissolved O 2 in a liquid growth medium as a function of incubation time. However, in Liq-μRM, the O 2 indicator is difficult to make, non-reusable and not well suited for use in well-plates, due to agitation issues. In this work a new, Solid-State micro-respirometry method, i.e., SS-μRM, is described based on a solid , rather than liquid , growth medium, in which the consumption of O 2 in the headspace is monitored, using a 3D printed O 2 indicator set in the lid of a small Petri dish or well plate containing the solid growth medium. The performance of the SS-μRM in a small Petri dish is compared with that of a commercial Liq-μRM system, using Escherichia coli , i.e., E. coli , as the test bacterium over the range 10 1 – 10 8 CFU/mL and demonstrated to be equivalent. The same system is used to measure the TVC of Pseudomonas aeruginosa , P. aeruginosa, and Enterobacter cloacae , E. cloacae, over the range 10 1 – 10 8 CFU/mL. When used to measure the TVC of E. coli , in a well plate, SS-μRM is shown to be insensitive to agitation, in contrast to Liq-μRM.
