Abstract:Studying the impact of land-use changes in wetland systems on the community structure of bacteria in soil aggregates can provide a theoretical basis for elucidating the impact mechanisms of the stability of wetland soil aggregates. The soil samples (0-20 cm) were collected from a natural wetland and paddy field in the Huixian karst wetland. The bacterial community structure in soil aggregates (macroaggregates 0.25-2 mm, microaggregates 0.053-0.25 mm, and silt-clay<0.053 mm) were analyzed using high-throughput sequencing and ecological network analysis. The results showed that ① the proportion of microaggregates in the paddy field was 29.64%, which was significantly higher than that in the natural wetland (22.20%), and ② there were differences in the relative abundance of bacteria between the natural wetland and paddy field. The relative abundance of Chloroflexi in macroaggregates, microaggregates, and silt-clay in the paddy field were 7.97%, 8.56%, and 7.40%, respectively, which were significantly higher than those in the natural wetland (4.93%, 4.81%, and 3.76%). The relative abundance of Anaerolineales in macroaggregates, microaggregates, and silt-clay in the paddy field were 2.35%, 3.03%, and 2.65%, respectively, which were significantly higher than those in the natural wetland (0.92%, 0.91%, and 0.43%).③ Ecological network analysis showed that the number of nodes, edges, and the average numbers of neighbors in microaggregates and the silt-clay network in the paddy field were higher than those in the natural wetland, the average connectivity of the network was greater in the two components, and the characteristic path length was shorter. There was a longer characteristic path length, higher modularity, and lower average connectivity in the macroaggregate network of the paddy field. The above results indicated that the bacterial community structure in soil aggregates was altered after a natural wetland was used as a paddy field. The material circulation and information transmission efficiency of bacterial communities was higher in both microaggregates and silt-clay of the paddy field, whereas the bacterial community structure with low network tightness and high modularity in macroaggregates was more stable.

Abstract:We report a preliminary investigation into soil microbial biomass C, ATP, microbial community composition and gaseous emissions when an upland Chinese and an immediately adjacent paddy soil were incubated with increasing percentage water holding capacities (WHC) from 10% WHC to waterlogging for 10 days. The aim was to see what adaptations, if any, occur when a paddy soil is incubated under conditions of increasing soil moisture, from 10% WHC to waterlogging and an adjacent upland soil is subjected to the opposite moisture changes, from waterlogging to 10% WHC. The main differences were that soil ATP remained quite constant in the paddy soil, irrespective of the different WHCs while in the upland soil, it increased from a low level between 10 and 20% WHC to a maximum at 60% WHC declining to a similar low level as 10 and 20% when waterlogged. The most striking feature was that although there were significant changes in biomass C, ATP and biomass ATP concentrations, of up to 3-fold or more, due to changing soil moisture, the changes in relative abundance of the microbial community composition measured by gene sequencing, particularly for fungi, were small and often insignificant, especially between 40% WHC and waterlogging. There were significant changes in bacterial community composition between 10 and 40%, where 45 to 61% of bacteria responded to the change. However, the changes were very few between 40% WHC and waterlogging. Thus, there was no clear link between the large changes in microbial biomass and microbial community composition.
The Influence of Soil Properties on the Size and Structure of Bacterial and Fungal Communities along A Paddy Soil Chronosequence

Abstract:In agroecosystems, soil bacterial and fungal communities are crucial for soil and plant health because of their diverse metabolic functions. However, little is known about the effect of long term paddy management on microbial communities. This study was conducted to assess the responses of the soil bacterial and fungal communities to cultivation history along a paddy soil chronosequence by using phospholipid fatty acid (PLFA) profiling and Illumina HiSeq sequencing. Soil samples were collected from the paddy fields (50, 100, 300, 1000, and 2000 years of paddy use; P50-P2000-), the adjacent arable chronosequence (50, 100 and 300 years of land use; NP50-NP300) and mudflat (estuarine sediment, representing the parent material for paddy and arable soil reclamation). The results showed that soil microbial biomass PLFAs increased significantly with increasing soil organic C which accumulated more under paddy than arable management. Bacterial taxonomic groups assigned to Proteobacteria and Acidobacteria changed relatively in the transition from tidal wetland to agricultural land. The relative abundances of dominant bacterial phyla in paddy soil orderly changes with cultivation time. The dominant fungal phyla in all samples were Ascomycota, Chytridiomycota, Basidiomycota, Glomeromycota and Zygomycota, representing 18.50%, 18.46%, 10.02%, 7.34%, and 7.19%, respectively. The succession of fungal community structure was mainly associated with changes in Ascomycota. Correlation analysis showed that higher soil total carbon and nitrogen related to long-term cultivation were associated with lower Proteobacteria and Ascomycota, but higher Verrucomicrobia. Furthermore, different land use type differed significantly in their fungal composition, but likely had similar effects on the succession of bacterial composition, mainly the Proteobacteria and Acidobacteria. Our results indicate that orderly succession of soil bacterial and fungal communities occurred along the long-term development of paddy soil, which in turn was associated with changes in soil physicochemical properties over time.

Abstract:Elucidating the biodiversity of CO 2 -assimilating bacterial communities under different land uses is critical for establishing an integrated view of the carbon sequestration in agricultural systems. We therefore determined the abundance and diversity of CO 2 assimilating bacteria using terminal restriction fragment length polymorphism and quantitative PCR of the cbbL gene (which encodes ribulose-1,5-biphosphate carboxylase/oxygenase). These analyses used agricultural soils collected from a long-term experiment (Pantang Agroecosystem) in subtropical China. Soils under three typical land uses, i.e., rice–rice (RR), upland crop (UC), and paddy rice–upland crop rotation (PU), were selected. The abundance of bacterial cbbL (0.04 to 1.25 × 10 8 copies g −1 soil) and 16S rDNA genes (0.05–3.00 × 10 10 copies g −1 soil) were determined in these soils. They generally followed the trend RR > PU > UC. The cbbL -containing bacterial communities were dominated by facultative autotrophic bacteria such as Mycobacterium sp ., Rhodopseudomonas palustris , Bradyrhizobium japonicum , Ralstonia eutropha , and Alcaligenes eutrophus . Additionally, the cbbL -containing bacterial community composition in RR soil differed from that in upland crop and paddy rice–upland crop rotations soils. Soil organic matter was the most highly statistically significant factor which positively influenced the size of the cbbL -containing population. The RR management produced the greatest abundance and diversity of cbbL -containing bacteria. These results offer new insights into the importance of microbial autotrophic CO 2 fixation in soil C cycling.
Effects of paddy field non-grainization consolidation on sustainable eco-functions protection of soil bacterial: Empirical evidence from Zhejiang province, China

Abstract:The increasing "non-grainization" of paddy fields affecting large-scale grain cultivation and impacting the sustainable eco-functions protection of soil bacterial in China. The same problem of "non-grainization" has threatened food security and farmland soil ecological functions in other areas of the world. Although previous research has demonstrated that soil microorganisms are strongly affected by land use change, little is known about the effects of paddy field non-grainization consolidation (NGC) on soil microorganisms. This study examined soil samples before and after paddy field NGC in Zhejiang province, China, measured soil environmental factors and performed 16S rDNA amplicon sequencing to analyze the changes in soilbacterial communities and ecosystem functions before and after NGC. Results show that NGC increased the relative abundances of Proteobacteria (27.89%) and Actinobacteria (25.25%) in the project zones. Total soil bacteria increased in all samples after NGC implementation in terms of absolute soil bacterial community content, but there were large differences. NGC improved the alpha diversity indices, including Ace, Chao1, Coverage, and Shannon indices ( p < 0.01) in terms of soil bacterial community diversity. The data analysis of RDA and the significance test showed that the environmental factors which were close-knitted with the soil bacterial diversity and structure of the sample sites were TN, AP, pH, SOM, FIQ, and AK. Wilcoxon rank sum test results showed that NGC also significantly enhanced the amino acid transport and metabolic functions of soil bacterial. Our findings suggest that NGC is beneficial to improve the paddy field soil bacterial diversity, enhance the versatility of soil ecosystems, and promote sustainable arable land soil ecosystem protection. The quantitative results would be useful for better studying the use of land remediation engineering measures to the remediation and maintenance of bacterial diversity and sustainable eco-functions protection in paddy fields.

[Impact on the Microbial Biomass and Metabolic Function of Carbon Source by Black Soil During Rice Cultivation].

Abstract:The effects of rice cultivation to the black soil microbial communities, which the experimentation area of Shuangyang District Agricultural Technology Extension Station in Changchun city, Jilin Province of northeastern China, were studied by using the method of phospholipid fatty acids and Biolog ECO-microplate culture. Results showed that the content of organic matter in space was the highest, fewer in the field, and the minimum in the rhizosphere, that change trend of total nitrogen and organic matter was similar in soil. The quantity of organic matter in summer sample was the highest. The microbial fun6tional diversity was significantly higher in summer than that in spring and autumn and showed no significant difference between spring and autumn. For summer and the lowest in winter, Shannon-Wiener index and Pielou index of the space were higher than the field and the rhizosphere. The time of microbial growth into the stable period and peak value of the average well color development were different in all samples, that the time was 216 h, 192 h, 216 h, 120 h, which varied from 0.52-0.84, 0.82-1.28, 0.40-0.84, 0.05-0.48, respectively. The result showed that the time of microbial growth into the stable period was similar in spring and autumn, the highest was in summer and the lowest was in winter. Above all, these results would provide more important characteristics of microbial features in the degradation and restoration process of the quality of the black soil habitat scientifically.

Abstract:Understanding the land use pattern relationships regarding the composition, diversity, and abundance of soil microbial communities in a typical karst depression in southwestern China is crucial for assessing the stability of local karst ecosystems. However, these aspects in typical karst depressions within northern tropical karst seasonal rainforests in China remain limited. Therefore, we examined the differences in soil microorganism abundance, diversity, community composition, and co-occurrence networks under five land use types in a tropical karst region in southwestern China: sugarcane fields, orchards, grasslands, plantation forests, and secondary forests. The soil microbial communities of samples from these areas were analyzed using 16S rRNA gene amplification. The abundances of Acidobacteria and Ascomycota were lowest (20.66% and 66.55%, respectively) in secondary forests and highest (35.59% and 89.35%, respectively) in sugarcane fields. Differences in microbial abundance across land use types were related to soil pH and total phosphorus. PCoA and ANOSIM demonstrated significant differences in soil bacterial and fungal community structures among the five land use types. Bacterial alpha-diversity showed no significant variation among the different land uses, whereas fungal alpha-diversity exhibited significant differences. Observed Chao1, ACE, and Shannon indices indicated that secondary forests had the highest fungal alpha-diversity. Land use changes also influenced bacterial and fungal co-occurrence networks, with the networks in secondary forests, plantation forests, and orchards being more complex and stable than those in grasslands and sugarcane fields. Key taxa such as Proteobacteria, Planctomycetes, Chloroflexi, Ascomycota, and Basidiomycota were predominantly connected within the co-occurrence networks, highlighting their high functional potential. This study provides insights that can inform more effective land use planning and management strategies in karst depressions, thereby enhancing ecological sustainability and balance.

[Microbial Metabolism in Typical Flooded Paddy Soils ].

Abstract:[OBJECTIVE] The object of this study is to reveal the composition of active microorganism and their metabolic activities in flooded paddy soils with long-term fertilization ( Mineral nitrogen, phosphorus, and potassium, NPK) and without fertilizer (Control check, CK) by environmental transcriptomics. [METHODS] Flooded soil microcosms were incubated in the laboratory for two weeks, then total RNA were extracted from the soil for transcriptome sequencing. Resulting fastq files were uploaded to the Metagenomics Analysis Server (MG-RAST) for taxonomic analysis, gene annotation and function classification. [RESULTS] Transcripts from diverse active microorganism, including bacteria ( > 95% ) , archaea, eukaryotes and viruses, were detected in both flooded paddy soils of CK and NPK treatments. Most of the transcripts (active genes) of bacteria and archaea were derived from Proteobacteria (more than 50% of total bacterial transcripts) and Thaumarchaaeota (about 70% of total archaeal transcripts ) respectively in both treatments. Transcriptional activity of Acidobacteria in NPK treatment paddy soil was significantly higher than that in CK treatment paddy soil. As for other phyla of bacteria and archaea, there were no significant differences of transcriptional activity of them between CK and NPK treatment paddy soils. The highest expressed gene in both CK and NPK treatment paddy soils is ABC transporter encoding gene which related to the transmembrane transport of substances. Based on gene function category of COG (Clusters of Orthologous Genes), Subsystem and KEGG (Kyoto Encyclopedia of Genes and Genomes) database, we found that the main metabolic activities of microorganisms in both CK and NPK treatment paddy soils were related to energy production and conversion, carbohydrate metabolism, protein metabolism and amino acid metabolism, and the dominant KEGG pathways were oxidative phosphorylation and aminoacyl-tRNA biosynthesis. [ CONCLUSION] Composition of active microorganism in CK and NPK treatment paddy soils was generally similar, except Acidobacteria whose transcriptional activity was significantly different between these two treatment paddy soils. It was also very similar between CK and NPK treatment paddy soils considering the metabolic activities of microorganisms in them, for dominant metabolic processes in these two soils were both related to energy obtaining and protein metabolism. So, dominant metabolic activities of microorganism in flooded paddy soils used in this study were not altered significantly under long - term inorganic fertilization.

Abstract:The current research was aimed to study the effects of different land use types (LUT) and soil depth (SD) on soil enzyme activity, metal content, and soil fungi in the karst area. Soil samples with depths of 0–20 cm and 20–40 cm were collected from different land types, including grassland, forest, Zanthoxylum planispinum land, Hylocereus spp. land and Zea mays land. The metal content and enzyme activity of the samples were determined, and the soil fungi were sequenced. The results showed that LUT had a significant effect on the contents of soil K, Mg, Fe, Cu and Cr; LUT and SD significantly affected the activities of invertase, urease, alkaline phosphatase and catalase. In addition, Shannon and Chao1 index of soil fungal community was affected by different land use types and soil depths. Ascomycota, Basidiomycota and Mortierellomycota were the dominant phyla at 0–20 cm and 20–40 cm soil depths in five different land types. Land use led to significant changes in soil fungal structure, while soil depth had no significant effect on soil fungal structure, probably because the small-scale environmental changes in karst areas were not the dominant factor in changing the structure of fungal communities. Additionally, metal element content and enzyme activity were related to different soil fungal communities. In conclusion, soil mineral elements content, enzyme activity, and soil fungal community in the karst area were strongly affected by land use types and soil depths. This study provides a theoretical basis for rational land use and ecological restoration in karst areas.

Conversion of Land Use from Upland to Paddy Field Changes Soil Bacterial Community Structure in Mollisols of Northeast China

Abstract:Mollisols are extremely important soil resource for crop and forage production. In northeast China, it is a major land use management practice from dry land crops to irrigated rice. However, there is few data regarding soil quality and microbial composition in Mollisols during land use transition. Here, we analyzed the upper 30 cm of soil from land with more than 30 years of paddy use and from adjacent areas with upland crops. Our results showed that land use and soil depth had a significant effect on soil properties and enzyme activities. Soil moisture (SM) and soil organic carbon (SOC) contents were substantially higher in paddy fields than in upland crop lands, while nitrogen-related enzyme activities were lower. Following the land use change, bacterial diversity was increased and bacterial community composition changed. Taxonomic analyses showed that Proteobacteria, Chloroflexi, Firmicutes, and Bacteroidetes were the dominant phyla present. At family level, Gemmatimonadaceae decreased with land use change, while Syntrophorhabdaceae and Syntrophacea that play a part in methane cycling and nitrifying bacteria such as Nitrospiraceae increased, indicating that the structure and composition of the bacterial community might be a promising indicator of Mollisol health. Redundancy analysis indicated that land use type had a stronger effect on the soil bacterial community composition than soil depth. Additionally, bacterial community composition was closely associated with soil parameters such as soil moisture, pH, SOC, NO3−-N, and NH4+-N. Overall, land use change affects the physical and chemical properties of the soil, resulting in changes in the composition of the soil bacterial community and flora. These changes could provide a view of the bacterial community assembly and functional shifts following land use change.

Abstract:Autotrophic carbon-fixing bacteria are the main drivers of carbon sequestration and elemental cycle in wetland ecosystems. Their relationship with environmental factors in karst soils such as soil organic carbon (SOC) fractions, which are affected by natural degradation and human disturbance, is key to understanding the biological mechanisms of karst wetland ecosystem deterioration and restoration. In this study, the abundances of the Calvin cycle functional genes cbbL and cbbM as well as the characteristics of carbon-fixing bacterial communities were compared in soil samples from a native wetland, naturally degraded wetland, and reclaimed farmland wetland in the Huixian karst wetland, Guilin, China. The abundances of the cbbL gene in the degraded wetland and reclaimed wetland soils varied significantly among the seasons (P < 0.05, summer > winter). The influence of the season on the abundances of the cbbL and cbbM genes was more significant than that of the wetland state. The structures of carbon-fixing bacterial communities were similar in the three states of karst wetland soils, mainly consisting of Alphaproteobacteria, Betaproteobacteria and Gammaproteobacteria in the phylum Proteobacteria. However, the relative abundances of some dominant bacterial genera were significantly different. Thiomonas and Bradyrhizobium had a relatively high abundance in the native wetland soil and the degraded wetland soil, respectively, while Ferriphaselus and Sulfuricaulis were more abundant in the reclaimed farmland wetland soil. The main soil factors affecting the structure of the carbon-fixing bacterial communities were the SOC and its fractions dissolved organic carbon (DOC), microbial biomass carbon (MBC) and readily oxidizable organic carbon (ROC), as well as the soil temperature. Anthropogenic activities such as wetland transition to farmland have caused significant changes in karst soils and the characteristics of carbon-fixing bacterial community in this karst wetland soil ecosystem.

Abstract:PurposeSoil microbes play a critical role in the recycling of soil nutrients and soil fertility. Different crop types greatly influence the soil environment, including the carbon and nitrogen biogeochemical cycles, and the effects may be particularly important on the terraces of the Loess Plateau. In this study, the characteristics of soil physicochemical properties and microbial activity were investigated under different crop types to establish relationships among soil microbial biomass, enzyme activities, and physicochemical properties.Materials and methodsSoils under four typical crops (potato, corn, apple, and intercropping (apple and potatoes)) grown on terraces constructed during a similar period were examined at an experimental site on the Loess Plateau, with 5-year abandoned land as the control. The activities of soil invertase, urease, and alkaline phosphatase, soil microbial biomass carbon (MBC), soil microbial biomass nitrogen (MBN), and the ratio MBC/MBN, and their relationships with soil physicochemical properties were investigated using redundancy analysis and partial redundancy analysis.Results and discussion(1) For soil physicochemical properties, including organic carbon, total nitrogen, total phosphorus, nitrate nitrogen, and available phosphorus, the lowest concentrations were in abandoned land and the highest contents were under apple and intercropping; (2) the activities of soil invertase were in the order intercropping > corn > apple > potato > abandoned land, whereas the activities of soil urease in apple and intercropping were more than twofold higher than those in abandoned land; (3) soil microbial biomass carbon and the ratio of MBC/MBN were highest in abandoned land, which indicated that abandoned land had a high proportion of bacterial to fungal biomass; (4) soil microbial activity was affected by the application of organic manure under terrace crop systems.ConclusionsThe contents of soil organic carbon (SOC), total nitrogen (TN), total phosphorus (TP), and available phosphorus (AP) in abandoned land were significantly lower than those under all crop types; thus, directly abandoned farmland was not favorable for soil microbial activity in the typical loess hilly-gully region. Soil TP, moisture (MS), and NO3−–N were identified as the key factors affecting soil microbial activity in the 0–20-cm soil layer, and soil TP, AP, NH4+–N, and NO3−–N were the significant factors in the 20–40-cm layer.

In this study, we investigated the relationships between the composition of the soil bacterial, acidobacterial, and fungal communities and the transformation of wetlands by human activities in the Sanjiang Plain. Soil microbial communities were assessed in wetland soils collected from pristine marsh, neighboring cropland (wetland turned into arable land), and land that had been reforested with Larix gmelinii . The alpha-diversities of bacteria, Acidobacteria, and fungi were affected by land-use change and were highest in the arable land and lowest in the wetland soils. The soil microbial community structures were also altered with changing land-use. Canonical correlation analyses showed that beta-diversity was significantly affected by soil pH, available phosphorus, soil nitrogen, and total organic carbon. Overall, our results showed that the agricultural cultivation of wetlands changes the available soil carbon, nitrogen, and phosphorus pools, thereby influencing the bacterial, acidobacterial, and fungal diversity and community structure. Once the soil microbial community has been altered by human activity, it might be difficult to restore it to its original state. These findings highlight the importance of effectively maintaining the diversity of soil bacterial, Acidobacterial, and fungal communities despite land use change in order to sustain a microbial community diversity and ecosystem function.

Abstract:Irrigation has been applied on a large scale for the improvement of grain yield per hectare and production stability. However, the dryland-to-paddy conversion affects the ecological environment of areas of long-term dry farming, especially soil microorganisms. Little attention has been paid to the changes in microbial communities and the interactions between their populations in this process. Therefore, in this paper, the compositions and diversity of soil bacterial and fungal communities were explored through a combination of high-throughput sequencing technology and molecular ecological network methods using bacterial 16S rRNA and fungal ITS. The results showed that: (1) both the abundance and diversity of soil bacteria and fungi decreased in a short time, and the abundance of Actinobacteria, Firmicutes and Olpidiomycota varied greatly. (2) Compared to dry land, the modular structure of interaction networks and interspecific relationships of bacterial and fungal communities in paddy soil were simpler, and the network became more unstable. A cooperative relationship dominated in the molecular ecological network of bacteria, while a competitive relationship was dominant in the network of fungi. Actinobacteria and Firmicutes were the dominant bacterial species in dry land and paddy field, respectively. Ascomycota was dominant in the fungal communities of both dry land and paddy field. (3) The change in soil environmental factors, such as pH, electrical conductivity (EC), organic matter (OM) and available potassium (AK), directly affected the soil microbial community structure, showing a significant correlation (p < 0.05). These environmental factors also influenced the dominant microbial species. Microorganisms are the most important link in the carbon and nitrogen cycles of soil, and a large-scale dryland-to-paddy conversion may reduce the ecological stability of regional soil.

Abstract:The over-utilizing of nitrogen fertilizers in paddy wetlands potentially threatens to the surrounding waterbody, and a deep understanding of the community and function of microorganisms is crucial for paddy non-point source pollution control. In this study, top soil samples (0–15 cm) of paddy wetlands under groundwater's irrigation at different depths (H1: 6.8 m, H2: 13.7 m, H3: 14.8 m, H4: 15.6 m, H5: 17.0 m, and H6: 17.8 m) were collected to investigate microbial community and function differences and their interrelation with soil properties. Results suggested some soil factor differences for groundwater's irrigation at different depths. Deep-groundwater's irrigation (H2-H6) was beneficial to the accumulation of various electron acceptors. Nitrifying-bacteria Ellin6067 had high abundance under deep groundwater irrigation, which was consistent with its diverse metabolic capacity. Meanwhile, denitrifying bacteria had diverse distribution patterns. Iron-reducing bacteria Geobacter was abundant in H1, and Anaeromyxobacter was abundant under deep groundwater irrigation; both species could participate in Fe-anammox. Furthermore, Geobacter could perform dissimilatory nitrate reduction to ammonia using divalent iron and provide substrate supply for anammox. Intrasporangium and norank_f_Gemmatimonadacea had good chromium- and vanadium-reducting potentials and could promote the occurrence of anammox. Low abundances of methanotrophs Methylocystis and norank_f_Methyloligellaceae were associated with the relatively anoxic environment of paddy wetlands, and the presence of aerobic methane oxidation was favorable for in-situ methane abatement. Moisture, pH, and TP had crucial effects on microbial community under phylum- and genus-levels. Microorganisms under shallow groundwater irrigation were highly sensitive to environmental changes, and Fe-anammox, nitrification, and methane oxidation were favorable under deep groundwater irrigation. This study highlights the importance of comprehensively revealing the microbial community and function of paddy wetlands under groundwater's irrigation and reveals the underlying function of indigenous microorganisms in agricultural non-point pollution control and greenhouse gas abatement.

Abstract:Soil microbial taxa have different functional ecological characteristics that influence the direction and intensity of plant–soil feedback responses to changes in the soil environment. However, the responses of soil microbial survival strategies to wet and dry events are poorly understood. In this study, soil physicochemical properties, enzyme activity, and high–throughput sequencing results were comprehensively anal0079zed in the irrigated cropland ecological zone of the northern plains of the Yellow River floodplain of China, where Oryza sativa was grown for a long period of time, converted to Zea mays after a year, and then Glycine max was planted. The results showed that different plant cultivations in a paddy–dryland rotation system affected soil physicochemical properties and enzyme activity, and G. max field cultivation resulted in higher total carbon, total nitrogen, soil total organic carbon, and available nitrogen content while significantly increasing α–glucosidase, β–glucosidase, and alkaline phosphatase activities in the soil. In addition, crop rotation altered the r/K–strategist bacteria, and the soil environment was the main factor affecting the community structure of r/K–strategist bacteria. The co–occurrence network revealed the inter–relationship between r/K–strategist bacteria and fungi, and with the succession of land rotation, the G. max sample plot exhibited more stable network relationships. Random forest analysis further indicated the importance of soil electrical conductivity, total carbon, total nitrogen, soil total organic carbon, available nitrogen, and α–glucosidase in the composition of soil microbial communities under wet–dry events and revealed significant correlations with r/K–strategist bacteria. Based on the functional predictions of microorganisms, wet–dry conversion altered the functions of bacteria and fungi and led to a more significant correlation between soil nutrient cycling taxa and environmental changes. This study contributes to a deeper understanding of microbial functional groups while helping to further our understanding of the potential functions of soil microbial functional groups in soil ecosystems.

Abstract:The 'Grain for Green' project (GGP) is the largest ecological rehabilitation project in China. A large body of croplands has been abandoned or converted to shrubs or grasslands since 1999. Soil microbes are recognized as sensitive responders of environmental changes, therefore, they are considered as a key component of ecological rehabilitation. However, very limited field experiments have been conducted to investigate the responses of soil microorganisms to restoration projects, especially in karst regions of China. In order to evaluate the response of soil microbial community to ecological restoration, we determined soil microbial community composition by means of qPCR, PLFAs, and high-throughput amplicon sequencing following conversion of cropland to Chinese prickly ash (Zanthoxylum bungeanum Maxim) orchard (CP) along a 20-year chronosequence in a degraded karst ecosystem. Our results showed that soil nutrient contents significantly increased following cropland to CP conversion. qPCR results showed that the highest bacterial abundance was found in the 20-year CP, but bacterial abundance decreased during the first 5-year land-use conversion. Conversion of cropland to CP strongly impacted soil microbial community composition, despite the cropland sites having a long cultivation history (>50 years). However, soil bacterial diversity remained unchanged within a 20-year land-use conversion. Actinobacteria, Proteobacteria, and Acidobacteria were the main bacterial phyla in all land-use sites. In particular, various members of Actinobacteria (e.g., Solirubrobacteraceae) tended to increase their relative abundances in responding to land-use conversion, which may imply that the shifts of soil microbial communities associated with recovering of ecological conditions. Overall, given the rapid yet differential response to ecological restoration, investigation of the belowground microbial community can provide an effective way of assessing ecological recovery of restoration projects in the karst region.

Abstract:Soil microorganisms are greatly affected by their microenvironment. To reveal the influence of different land use patterns on the composition and diversity of soil bacterial and fungal communities, this study analyzed microbial (bacteria and fungi) community composition and diversity under different land use patterns (vegetable land, wasteland, woodland, cultivated land) based on 16S rRNA, 18S rRNA, and high-throughput sequencing method in the Taojia River Basin. Spearman analysis and redundancy analysis (RDA) were used to explore the correlation between soil physicochemical properties and soil fungal and bacterial community composition, and a partial least squares path model (PLS-PM) was constructed to express the causal relationship between soil physicochemical properties and soil bacterial and fungal community diversity. The results showed that the soil bacterial species richness was highest in vegetable land and the lowest in the wasteland. Proteobacteria is the dominant phylum (20.69%–32.70%), and Actinobacteria is the dominant class (7.99%–16.95%). The species richness of fungi in woodland was the highest, while was the lowest in cultivated land. The dominant phylum of fungi in vegetable land, woodland, and cultivated land is Mucoromycota, 29.39%, 41.36%, and 22.67%, respectively. Ascomycota (42.16%) is the dominant phylum in wasteland. Sordariomyetes of Ascomycota is the dominant class in wasteland and cultivated land. Mortierellomycetes and Glomeromycetes of Mucoromycota are the dominant class in vegetable land and woodland. The results of the Spearman analysis revealed that the dominant groups in the bacterial and fungal communities had significant correlations with soil pH, clay, and sand (p < 0.01). The RDA results showed that soil clay, pH, and moisture were the key environmental factors affecting the diversity of soil microbial communities. Fungal diversity is more affected by different land use patterns than bacteria. These results provided a theoretical basis for the changes in soil microbial community composition and diversity in river basins.
