Abstract:In vitro batch incubations were used to study the rumen biohydrogenation of unsaturated fatty acids. An earlier study using increasing supplementation levels of stearidonic acid (18:4n-3), revealed that the rumen microbial population extensively biohydrogenates 18:4n-3 after 72 h of in vitro incubation, though several intermediates formed were not completely characterized. Therefore, in the present study, samples were reanalyzed in order to identify the 18:2, 18:3 and 18:4 biohydrogenation intermediates of 18:4n-3. Gas-liquid chromatography coupled to mass spectrometry was used to characterize these intermediates. The acetonitrile chemical ionization mass spectrometry of the fatty acid methyl esters derivatives enabled the discrimination of fatty acids as non-conjugated or conjugated biohydrogenation intermediates. In addition, the acetonitrile covalent adduct chemical ionization tandem mass spectrometry yielded prominent ions indicative of the double bond position of the major 18:3 isomers, i.e. Δ5,11,15 18:3. Furthermore, the 4,4-dimethyloxazoline derivatives prepared from the fatty acid methyl esters enabled the structure of novel 18:2, 18:3 and 18:4 biohydrogenation intermediates to be elucidated. The intermediates accumulated in the fermentation media after 72 h of incubation of 18:4n-3 suggest that similar to the biohydrogenation pathways of linoleic (18:2n-6) and α-linolenic (18:3n-3) acids, the pathway of the 18:4n-3 also proceeds with the formation of conjugated fatty acids followed by hydrogenation, although no conjugated dienes were found. The formation of the novel biohydrogenation intermediates of 18:4n-3 seems to follow an uncommon isomerization pattern with distinct double bond migrations.
Impairment of rumen biohydrogenation and bacteria of the Butyrivibrio group in the rumen of goats through a 20:5 n-3 (EPA) rich supplement

Abstract:Background: Marine products can inhibit biohydrogenation in the rumen, but the mechanism is not clear. This study investigated a 20:5 n-3 rich supplement effects on rumen biohydrogenation, microbial change and fermentation characteristics in goats. Results: The supplementation decreased 18:0 proportions in rumen fatty acids (P < 0.001), while it increased cis-9, trans-11 conjugated linoleic acid (CLA) (P < 0.001) and trans-10, cis-12 CLA proportions (P < 0.001). The supplement reduced the number of Butyrivibrio spp. and B. proteoclasticus (P < 0.01). Denaturing gradient gel electrophoresis redundancy analysis indicated that some species, mainly from the rumen of goats receiving the 2.5 and 5.0 g d(-1) supplement, were positively correlated with cis-9, trans-11 CLA proportions; some species, mainly from the rumen of control goats, were positively correlated with 18:0 proportions. The supplement reduced the NH3 -N concentrations and acetate molar proportions in the rumen (P < 0.05), but increased propionate and butyrate molar proportions (P < 0.01), and had no effect on total volatile fatty acid concentration. Conclusion: The supplement rich in 20:5 n-3 reduced the biohydrogenation of 18-carbon unsaturated fatty acids with a significant reduction of the 18:0 proportion and this was coupled with the suppression of the abundance of biohydrogenating bacteria and unknown bacteria.
Fatty acid composition and bacterial community changes in the rumen fluid of lactating sheep fed sunflower oil plus incremental levels of marine algae

Abstract:Supplementation of ruminant diets with plant oils and marine lipids is an effective strategy for lowering saturated fatty acid (FA) content and increasing the concentration of cis-9,trans-11 conjugated linoleic acid and long-chain n-3 FA in ruminant milk. However, changes in populations of ruminal microorganisms associated with altered biohydrogenation of dietary unsaturated FA are not well characterized. Twenty-five lactating Assaf ewes were allocated at random to 1 of 5 treatments composed of dehydrated alfalfa hay and concentrates containing no additional lipid (control), or supplemented with 25 g of sunflower oil and 0 (SO), 8 (SOMA(1)), 16 (SOMA(2)), or 24 (SOMA(3)) g of marine algae/kg of diet dry matter. On d 28 on diet, samples of rumen fluid were collected for lipid analysis and microbial DNA extraction. Appearance and identification of biohydrogenation intermediates was determined based on complementary gas chromatography and Ag+-HPLC analysis of FA methyl esters. Total bacteria and the Butyrivibrio group were studied in microbial DNA by terminal RFLP analysis, and real-time PCR was used to quantify the known Butyrivibrio bacteria that produce trans-11 18:1 or 18:0. Dietary supplements of sunflower oil alone or in combination with marine algae altered the FA profile of rumen fluid, which was associated with changes in populations of specific bacteria. Inclusion of marine algae in diets containing sunflower oil resulted in the accumulation of trans 18:1 and 10-O-18:0 and a marked decrease in 18:0 concentrations in rumen fluid. At the highest levels of supplementation (SOMA(2) and SOMA(3)), marine algae also promoted a shift in ruminal biohydrogenation pathways toward the formation of trans-10 18:1 at the expense of trans-11 18:1. Changes in the concentration of biohydrogenation intermediates were not accompanied by significant variations in the abundance of known cultivated ruminal bacteria capable of hydrogenating unsaturated FA. However, certain bacterial groups detected by terminal RFLP (such as potentially uncultured Lachnospiraceae strains or Quinella-related bacteria) exhibited variations in their relative frequency consistent with a potential role in one or more metabolic pathways of biohydrogenation in the rumen.

Abstract:In this issue of The Journal of Thoracic Disease (JTD) Jonathon Spahr and Shawn West from the Children’s Hospital of Pittsburgh and the University of Pittsburg School of Medicine have presented a perspective on paediatric heart-lung transplantation based on data from their own institution, the Organ Procurement and Transplant Network and supported with data from the International Society for Heart and Lung Transplantation (ISHLT). They have provided a detailed and succinct overview of progress to the current era including indications, contraindications, outcomes and potential future possibilities largely based on the US experience. This editorial aims to extend the review by elaborating on the global perspective of paediatric heart-lung transplantation, particularly in relation to changes in volume and indications over the past 30 years and the influence of organ allocation policies internationally.

Abstract:This study was carried out to investigate effects of type and level of octadeca carbon fatty acids (C18-fatty acids) on rumen fermentation, methane emission and populations of ruminal microbes using an in vitro gas production technique. Four types of C18-fatty acids, stearic, oleic, linoleic and linolenic acids, were added at levels of 0, 35 and 70g/kg of substrate dry matter, respectively. Methane emission was determined by gas chromatography. Total deoxyribonucleic acid (DNA) was extracted from ruminal microbes, and populations of rumen microbes were determined by a real-time quantitative polymerase chain reaction. Populations of total rumen methanogens, protozoa, fungi, Ruminococcus flavefaciens and Fibrobacter succinogenes were expressed as a proportion of total rumen bacterial 16S ribosomal deoxyribonucleic acid (rDNA). The 24h gas production and methane emission were decreased by type, level and their interaction (P<0.001). Compared to the control, C18-fatty acids had little effect on pH, ammonia N and total volatile fatty acids. However, the fermentation patterns were changed, reflected by lower proportion of acetate and higher propionate with increasing levels and unsaturation of C18-fatty acids (P<0.001). Methanogens and protozoa populations relative to total bacterial 16S rDNA were decreased by linoleic and linolenic acids, with the linolenic acid addition being most efficient. However, addition of these unsaturated C18-fatty acids also inhibited growth of fibrolytic microbes including fungi, F. succinogenes and R. flavefaciens. From the present study, it is inferred that there is an effect of unsaturated C18-fatty acids in suppressing methanogenesis, probably mediated by direct action against the rumen microbes involved in methane formation.
Toxicity of unsaturated fatty acids to the biohydrogenating ruminal bacterium, Butyrivibrio fibrisolvens

Abstract:Background: Health-promoting polyunsaturated fatty acids (PUFA) are abundant in forages grazed by ruminants and in vegetable and fish oils used as dietary supplements, but only a small proportion of PUFA finds its way into meat and milk, because of biohydrogenation in the rumen. Butyrivibrio fibrisolvens plays a major role in this activity. The aim of this study was to investigate the mechanisms by which PUFA affect the growth of B. fibrisolvens, how PUFA are metabolized and the metabolic response to growth in the presence of PUFA. Results: Linoleic acid (LA; cis-9, cis-12-18:2) and alpha-linolenic acid (LNA; cis-9, cis-12, cis-15-18:3) increased the lag phase of B. fibrisolvens JW11, LNA having the greater effect. Growth was initiated only when the PUFA had been converted to vaccenic acid (VA; trans-11-18:1). The major fish oil fatty acids, eicosapentaenoic acid (EPA; 20:5(n-3)) and docosahexaenoic acid (DHA; 22:6(n-3)), were not metabolized and prevented growth. Cellular integrity, as determined fluorimetrically by propidium iodide (PI) ingression, was affected as much by 18:1 fatty acids, including VA, as 18:2 fatty acids. The methyl esters of LNA, LA, EPA and DHA had no effect on growth or other measurements. The ATP pool decreased by 2/3 when LA was added to growing bacteria, whereas most acyl CoA pools decreased by >96%. Conclusions: It was concluded that biohydrogenation occurs to enable B. fibrisolvens to survive the bacteriostatic effects of PUFA, and that the toxicity of PUFA is probably mediated via a metabolic effect rather than disruption of membrane integrity.

Abstract:The present study aimed to investigate the effect of hydrogen-consuming compounds on ruminal methane (CH4) production, in vitro fermentation parameters, fatty acids profile, and microbial community in water buffalo. Different sodium nitrate to disodium fumarate ratios [2:1 (F), 1:1 (S), 1:2 (T)] were studied in vitro by batch culture technique in the presence of linoleic acid. Results revealed that the dominant bacterial communities were not affected with sodium nitrate and disodium fumarate, whereas CH4 production and Verrucomicrobia, Succiniclasticum, norank_f__Muribaculaceae, and Prevotellaceae_UCG-003 were reduced (P < 0.05). However, ruminal pH, unsaturated fatty acids/saturated fatty acids (UFA/SFA) and Campilobacterota, Selenomonas, Succinivibrio, Oribacterium, Christensenellaceae_R-7_group, Campylobacter, Shuttleworthia, Schwartzia, and Prevotellaceae_YAB2003_group were increased (P < 0.05). Total volatile fatty acids (TVFA) and Spirochaetae, Fibrobacterota, Verrucomicrobia, Fibrobacter, Treponema, and Prevotellaceae were decreased in F (P < 0.05), but cis-9, trans-11CLA, acetate/propionate and Proteobacteria, Campilobacterota, Selenomonas, Succinivibrio, and Campylobacter were increased in F (P < 0.05). The highly selected bacterial genera in F were Campylobacter and Succinivibrio. The disodium fumarate, enhanced (P < 0.05) the TVFA, propionate, total bacteria, Butyrivibrio proteoclasticus, and Atypical butyrivibrio. The concentrations of C18:3n3, C20:3n6, C21:0, C22:2n6, and C22:1n9, as well as the populations of total fungi, protozoa, methanogens, Butyrivibrio hungatei in T were higher (P < 0.05). The highly selected bacterial genera in T were Fibrobacter and Treponema. Conclusively, the addition of sodium nitrate and disodium fumarate can reduce the CH4 production and optimize ruminal fatty acid composition. Furthermore, disodium fumarate can alleviate the adverse effect of sodium nitrate on the rumen fermentation.

Abstract:The hypothesis that condensed tannins have higher inhibitory effect on ruminal biohydrogenation than hydrolyzable tannins was tested. Condensed tannin extract from mimosa (CT) and hydrolyzable tannin extract from chestnut (HT) or their mixture (MIX) were incorporated (10%) into oil supplemented diets and fed to rumen fistulated sheep. Fatty acid and dimethyl acetal composition of rumen contents and bacterial biomass were determined. Selected rumen bacteria were analyzed by quantitative real time PCR. Lower ( P < 0.05) rumen volatile fatty acids concentrations were observed with CT compared to HT. Moreover, lower concentration ( P < 0.05) of Fibrobacter succinogenes, Ruminococcus flavefaciens, Ruminococcus albus, and Butyrivibrio proteoclasticus were observed with CT compared to HT. The extension of biohydrogenation of 18:2n-6 and 18:3n-3 did not differ among treatments but was much more variable with CT and MIX than with HT. The trans-/ cis-18:1 ratio in bacterial biomass was higher ( P < 0.05) with HT than CT. Thus, mimosa condensed tannins had a higher inhibitory effect on ruminal metabolism and biohydrogenation than chestnut hydrolyzable tannins.

Abstract:Fat supplementation in the ration has a negative impact on rumen microbes and the biohydrogenation of fats. Meanwhile, the effectiveness of post-rumen nutrients may be decreased if high-protein diet is excessively broken down in the rumen. The protection of both fat and protein may help address fat-related issues in the rumen and enhance protein absorption in the intestine. The goal of this study was to examine the fatty acid composition of rumen fluid made with a blend of fat, protein, and Agromix® mineral premix that had been formaldehyde-protected. Two treatments were compared using an independent sample T-test (R1/control = basal diet, and R2 = basal diet with 4.4% DM replacement of protected fat-protein). All treatments were subjected to a 48-hour incubation using Tilley and Terry's in vitro technique. The results indicated that the levels of SFA in R2 treatment showed a significant decrease (p<0.05), while the concentrations of MUFA and PUFA in R2 exhibited a more significant increase (p<0.05) compared to those in R1. It can be concluded that protected fat substitution can reduce biohydrogenation of fat in the rumen, thereby helping elevate the levels of absorbable unsaturated fatty acid levels in the intestine.
Strong-coupling limit of Eliashberg theory.

Abstract:We study the strong-coupling limit of the Eliashberg theory of superconductivity, where the coupling strength \ensuremath{\lambda} goes to infinity and the critical temperature gets large compared to a typical phonon energy. This limit is of interest because it is both universal and simple, and we may hope to obtain from this study a deeper understanding of the conventional strong-coupling regime of superconductivity. Our work on this problem is both analytical and numerical. At T=0, we find that the excitation spectrum is discrete. We interpret physically the excited states as bound states due to a type of polaronic effect. We show that one can solve the Eliashberg equations essentially analytically by working fully on the real frequency axis. At finite temperature we find a thermal smearing of the T=0 structure. Since the critical temperature is small compared to the zero-temperature gap, thermal effects can be treated as a kind of perturbation over almost all the temperature range. In this spirit, we give a simple approximate solution which reproduces almost quantitatively the exact numerical results.

Abstract:Based on controlled clinical trials, vasoactive drugs (pentoxyfylline, naftidrofuryl, buflomedil) and prostanoids (alprostadil, iloprost) are used in symptomatic peripheral arterial occlusive disease: In stage II, vasoactive drugs are prescribed in order to improve leg hyperemia in response to muscular work; in complicated stage II (e.g., non-healing traumatic skin lesions), and in stages III/IV, prostanoids are given in order to increase forefoot skin blood flow. These drugs should only be applied if neither angioplasty nor vascular surgery are recommendable, possible, or successful after complete angiological diagnostic. It should be considered that these substances have a rather limited clinical efficacy in both stage II and stages III/IV and therefore attempts should be made to select patients with a reasonable chance to benefit. These drugs are also not suitable for primary or secondary prophylaxis against the progression of vascular lesions.

Abstract:ABSTRACT This commentary on Gow and Olson questions in three ways their conclusion that speech perception is based on interactive processing. First, it is not clear that the data presented by Gow and Olson reflect normal speech recognition. Second, Gow and Olson's conclusion depends on still-debated assumptions about the functions performed by specific brain regions. Third, the results are compatible with feedforward models of speech perception and appear inconsistent with models in which there are online interactions about phonological content. We suggest that progress in the neuroscience of speech perception requires the generation of testable hypotheses about the function(s) performed by inter-regional connections.
Inhibiting methanogenesis by targeting thermodynamics and enzymatic reactions in mixed cultures of rumen microbes in vitro

Abstract:Mitigation of enteric methane (CH4) emissions from ruminant livestock represents an opportunity to improve the sustainability, productivity, and profitability of beef and dairy production. Ruminal methanogenesis can be mitigated via two primary strategies: (1) alternative electron acceptors and (2) enzymatic inhibition of methanogenic pathways. The former utilizes the thermodynamic favorability of certain reactions such as nitrate/nitrite reduction to ammonia (NH3) while the latter targets specific enzymes using structural analogs of CH4 and methanogenic cofactors such as bromochloromethane (BCM). In this study, we investigated the effects of four additives and their combinations on CH4 production by rumen microbes in batch culture. Sodium nitrate (NaNO3), sodium sulfate (Na2SO4), and 3-nitro-1-propionate (3NPA) were included as thermodynamic inhibitors, whereas BCM was included as a enzymatic inhibitor. Individual additives were evaluated at three levels of inclusion in experiments 1 and 2. Highest level of each additive was used to determine the combined effect of NaNO3 + Na2SO4 (NS), NS + 3NPA (NSP), and NSP + BCM (NSPB) in experiments 3 and 4. Experimental diets were high, medium, and low forage diets (HF, MF, and LF, respectively) and consisted of alfalfa hay and a concentrate mix formulated to obtain the following forage to concentrate ratios: 70:30, 50:50, and 30:70, respectively. Diets with additives were placed in fermentation culture bottles and incubated in a water bath (39°C) for 6, 12, or 24h. Microbial DNA was extracted for 16S rRNA and ITS gene amplicon sequencing. In experiments 1 and 2, CH4 concentrations in control cultures decreased in the order of LF, MF, and HF diets, whereas in experiments 3 and 4, CH4 was highest in MF diet followed by HF and LF diets. Culture pH and NH3 in the control decreased in the order of HF, MF, to LF as expected. NaNO3 decreased (p < 0.001) CH4 and butyrate and increased acetate and propionate (p < 0.03 and 0.003, respectively). Cultures receiving NaNO3 had an enrichment of microorganisms capable of nitrate and nitrite reduction. 3NPA also decreased CH4 at 6h with no further decrease at 24 h (p < 0.001). BCM significantly inhibited methanogenesis regardless of inclusion levels as well as in the presence of the thermodynamic inhibitors (p < 0.001) while enriching succinate producers and assimilators as well as propionate producers (p adj < 0.05). However, individual inclusion of BCM decreased total short chain fatty acid (SCFA) concentrations (p < 0.002). Inhibition of methanogenesis with BCM individually and in combination with the other additives increased gaseous H2 concentrations (p < 0.001 individually and 0.028 in combination) while decreasing acetate to propionate ratio (p < 0.001). Only the cultures treated with BCM in combination with other additives significantly (padj < 0.05) decreased the abundance of Methanobrevibacter expressed as log fold change. Overall, the combination of thermodynamic and enzymatic inhibitors presented a promising effect on ruminal fermentation in-vitro, inhibiting methanogenesis while optimizing the other fermentation parameters such as pH, NH3, and SCFAs. Here, we provide a proof of concept that the combination of an electron acceptor and a methane analog may be exploited to improve microbial efficiency via methanogenesis inhibition.

Abstract:Despite the fact that the ruminant diet is rich in polyunsaturated fatty acids (PUFA), ruminant products - meat, milk and dairy - contain mainly saturated fatty acids (SFA) because of bacterial lipolysis and subsequent biohydrogenation of ingested PUFA in the rumen. The link between SFA consumption by man and coronary heart disease is well established. In contrast, ruminant products also contain fatty acids that are known to be beneficial to human health, namely conjugated linoleic acids (CLAs). The aims of research in this field have been to understand the microbial ecology of lipolysis and biohydrogenation and to find ways of manipulating ruminal microbes to increase the flow of PUFA and CLA from the rumen into meat and milk. This review describes our present understanding of the microbial ecology of ruminal lipid metabolism, including some apparently anomalous and paradoxical observations, and the status of how the metabolism may be manipulated and the possible consequential effects on other aspects of ruminal digestion. Intuitively, it may appear that inhibiting the ruminal lipase would cause more dietary PUFA to reach the mammary gland. However, lipolysis releases the non-esterified fatty acids that form the substrates for biohydrogenation, but which can, if they accumulate, inhibit the whole process. Thus, increasing lipase activity could be beneficial if the increased release of non-esterified PUFA inhibited the metabolism of CLA. Rumen ciliate protozoa do not carry out biohydrogenation, yet protozoal lipids are much more highly enriched in CLA than bacterial lipids. How could this happen if protozoa do not metabolise PUFA? The answer seems to lie in the ingestion of plant organelles, particularly chloroplasts, and the partial metabolism of the fatty acids by contaminating bacteria. Bacteria related to Butyrivibrio fibrisolvens are by far the most active and numerous biohydrogenating bacteria isolated from the rumen. But do we misunderstand the role of different bacterial species in biohydrogenation because there are uncultivated species that we need to understand and include in the analysis? Manipulation methods include dietary vegetable and fish oils and plant-derived chemicals. Their usefulness, efficacy and possible effects on fatty acid metabolism and on ruminal microorganisms and other areas of their metabolism are described, and areas of opportunity identified.
Effects of Lauric Acid and Linolenic Acid on Rumen Fermentation, Methanogenesis and Microbial Flora in Vitro

Abstract:The aim of this study was to investigate the effects of lauric acid(C12) and linolenic acid(C18∶3) on rumen fermentation,methanogenesis and microbial flora of rumen in vitro.Reading pressure treat system and gas chromatography were used to determine gas production and methanogenisis,respectively.Volatile fatty acid(VFA),ammonia nitrogen and pH were also analyzed after incubation of 24 h.Changes of the population of rumen microbial flora were detected and analyzed by real-time PCR assay.Results showed the supplement of C12 and C18∶3 could reduce the production of mathane significantly(P0.01) and inhibit the gas production,acetate and pH compared with the control.Besides,a higher proportion of propionate was observed(P0.01).Ammonia nitrogen in the groups with supplement of lauric acid and linolenic acid decreased(P0.05).With the supplement of fatty acid,the production of the rumen,fungi,protozoa and methanogen were all decreased significantly compared with the control group(P0.01),and the inhibiting effect of C18∶3 were stronger than that of C12(P0.01).In conclusion,C12 had less effect on inhibition of changing degree of methanogenesis,fermentation and growth of microbial flora than C18∶3.
Effects of Culture and 2-Hydroxy-4-(Methylthio)-Butanoic Acid on Rumen Fermentation and Microbial Populations between Different Roughage Sources

Abstract:An in vitro experiment was conducted to evaluate the effects of Aspergillus oryzae culture (AOC) and 2-hydroxy-4-(methylthio)-butanoic acid (HMB) on rumen fermentation and microbial populations between different roughage sources. Two roughage sources (Chinese wild rye [CWR] vs corn silage [CS]) were assigned in a 2×3 factorial arrangement with HMB (0 or 15 mg) and AOC (0, 3, or 6 mg). Gas production (GP), microbial protein (MCP) and total volatile fatty acid (VFA) were increased in response to addition of HMB and AOC (p<0.01) for the two roughages. The HMB and AOC showed inconsistent effects on ammonia-N with different substrates. For CWR, neither HMB nor AOC had significant effect on molar proportion of individual VFA. For CS, acetate was increased (p = 0.02) and butyrate was decreased (p<0.01) by adding HMB and AOC. Increase of propionate was only occurred with AOC (p<0.01). Populations of protozoa (p≤0.03) and fungi (p≤0.02) of CWR were differently influenced by HMB and AOC. Percentages of F. succinogenes, R. albus, and R. flavefaciens (p<0.01) increased when AOC was added to CWR. For CS, HMB decreased the protozoa population (p = 0.01) and increased the populations of F. succinogenes and R. albus (p≤0.03). Populations of fungi, F. succinogenes (p = 0.02) and R. flavefacien (p = 0.03) were increased by adding AOC. The HMB×AOC interactions were noted in MCP, fungi and R. flavefacien for CWR and GP, ammonia-N, MCP, total VFA, propionate, acetate/propionate (A/P) and R. albus for CS. It is inferred that addition of HMB and AOC could influence rumen fermentation of forages by increasing the number of rumen microbes.

Abstract:<h3> Aims</h3><p>This study investigated the effects of ferric citrate, nitrate, and saponin, both individually and in combination, on sulfidogenesis, methanogenesis, rumen fermentation, and abundances of select microbial populations using <i>in vitro</i> rumen cultures. </p><h3> Methods and Results</h3><p>Ferric citrate (50 mg L<sup>−1</sup>), Quillaja saponin (0·6 g L<sup>−1</sup>), and sodium nitrate (5 mmol L<sup>−1</sup>) were used in <i>in vitro</i> ruminal fermentation. Ferric citrate alone, its combination with saponin and/or nitrate lowered the aqueous sulfide concentration and total sulfide production. Methane production was suppressed by nitrate alone (by up to 32·92%), its combination with saponin (25·04%) and with both saponins with nitrate (25·92%). None of the treatments adversely affected feed digestion or rumen fermentation. The population of sulfate‐reducing bacteria was increased by nitrate and saponin individually, while that of total archaea was decreased by nitrate alone and the combination of the three inhibitors. </p><h3> Conclusions</h3><p>Nitrate and its combination with saponin or both ferric citrate and saponin substantially decreased methane production. Most importantly, the decreased methane production was not at the expense of feed digestion or fermentation. Sulfidogenesis from the sulfate present in the high‐sulfur diets can be suppressed competitively by ferric citrate, though it was elevated by saponin and nitrate.</p><h3> Significance and Impact of Study</h3><p>The results of this study demonstrated that combinations of certain methane inhibitors, which have different mechanisms of anti‐methanogenic actions or inhibit different guilds of microbes involved in methane production and sulfate reduction, can be more effective and practical than individual inhibitors, not only in mitigating enteric methane emission but also in lowering the risk of sulfur‐associated polioencephalomalacia in feedlot cattle fed high sulfur diets.</p><p>This article is protected by copyright. Abstract:Nitrate is used as a methane inhibitor while cysteamine is considered as a growth promoter in ruminants. The present study evaluated the effect of sodium nitrate and cysteamine on methane (CH4) production, rumen fermentation, amino acid (AA) metabolism, and rumen microbiota in a low protein diet. Four treatments containing a 0.5 g of substrate were supplemented with 1 mg/mL sodium nitrate (SN), 100 ppm cysteamine hydrochloride (CS), and a combination of SN 1 mg/mL and CS 100 ppm (CS+SN), and a control (no additive) were applied in a completely randomized design. Each treatment group had five replicates. Two experimental runs using in vitro batch culture technique were performed for two consecutive weeks. Total gas and CH4 production were measured in each fermentation bottle at 3, 6, 9, 12, 24, 48, and 72 h of incubation. The results showed that SN and CS+SN reduced the production of total gas and CH4, increased the rumen pH, acetate, acetate to propionate ratio (A/P), and microbial protein (MCP) contents (p < 0.05), but decreased other volatile fatty acids (VFA) and total VFA (p = 0.001). The CS had no effect on CH4 production and rumen fermentation parameters except for increasing A/P. The CSN increased the populations of total bacteria, fungi, and methanogens but decreased the diversity and richness of rumen microorganisms. In conclusion, CS+SN exhibited a positive effect on rumen fermentation by increasing the number of fiber degrading and hydrogen-utilizing bacteria, with a desirable impact on rumen fermentation while reducing total gas and CH4 production.
