Abstract:Emerald ash borer (EAB, Agrilus planipennis), an exotic invasive pest, has killed millions of ash trees (Fraxinus spp.) in North America and continues to threaten the very survival of the entire Fraxinus genus. Despite its high-impact status, to date very little knowledge exists for this devastating insect pest at the molecular level. Mariner-like elements (MLEs) are transposable elements, which are ubiquitous in occurrence in insects and other invertebrates. Because of their low specificity and broad host range, they can be used for epitope-tagging, gene mapping, and in vitro mutagenesis. The majority of the known MLEs are inactive due to in-frame shifts and stop codons within the open reading frame (ORF). We report on the cloning and characterization of two MLEs in A. planipennis genome (Apmar1 and Apmar2). Southern analysis indicated a very high copy number for Apmar1 and a moderate copy number for Apmar2. Phylogenetic analysis revealed that both elements belong to the irritans subfamily. Based on the high copy number for Apmar1, the full-length sequence was obtained using degenerate primers designed to the inverted terminal repeat (ITR) sequences of irritans MLEs. The recovered nucleotide sequence for Apmar1 consisted of 1,292 bases with perfect ITRs, and an ORF of 1,050 bases encoding a putative transposase of 349 amino acids. The deduced amino acid sequence of Apmar1 contained the conserved regions of mariner transposases including WVPHEL and YSPDLAP, and the D,D(34)D motif. Both Apmar1 and Apmar2 could represent useful genetic tools and provide insights on EAB adaptation.
Sucralose, a Persistent Artificial Sweetener in the Urban Water Cycle: Insights into Occurrence, Chlorinated Byproducts Formation, and Human Exposure

Abstract:As a globally important artificial sweetener, sucralose (SUC) is a persistent emerging contaminant in global aquatic environments. This research comprehensively track the fate and transport of SUC throughout an urban water cycle; to perform laboratory experiments to determine chlorinated disinfection by-products (DBPs) of SUC sebsequent to advanced oxidation with ozone; and to perform a risk evaluation of SUC related DBPs.; The results demonstrated that SUC was consistently present throughout the urban water cycle, including consumer's tap water. The inluent to the municipal wastewater treatment plant contained 1033.4-2626.3 ng/L SUC, and as expected for this non-biodegradable artificial sweetener, the concentration in the effluent was slightly reduced to 917.6-2031.2 ng/L. The concentration of SUC varied in surface waters with a peak value of 2070.0 ng/L. In finished drinking water 288.1-505.3 ng/L SUC was found, and values of 177.7-409.7 ng/L were present in the distribution system. Up to 16.3% of SUC concentration discharged as municipal wastewater was delivered to residents in their drinking water. While conventional treatment little removed SUC, the mean removal efficiency of SUC with ozonation followed by activated carbon filtration was 39.6% at a full-scale drinking water treatment plant. The fate and transformation of SUC in drinking water treated by ozonation and chlorination were investigated in laboratory experiments. Ozonation partially degraded SUC; however, intermediates reacted with chlorine to generate DBPs, including trichloromethane (TCM), dichloroacetic acid (DCAA) and trichloroacetic acid (TCAA). Peak 7-days potential formation of TCM, DCAA and TCAA were 0.016, 0.51 and 23.34 mu g/mg SUC, respectively. DBP yields increased with increasing chlorine dosage, chlorination time, temperature, and solution pH. The presence of ammonia nitrogen in water facilitated dichloroacetonitrile (DCAN) production up to 0.78 mu g/mg SUC, while simultaneously reducing the yield of TCM, DCAA and TCAA. Human exposure analysis revealed that carcinogenic risks of DBPs caused only by SUC ozonation were in the range of 1.86 x 10(-12)-6.12 x 10(-9), while non-carcinogenic risks were in the range of 6.0 x 10(-9)-4.37 x 10(-6). The pervasive occurrence of SUC in an urban water cycle, couple to its resistance to biological and chemical treatment, confirms that SUC is a persistent contaminant in the aquatic system, including tap water.
Variable persistence of artificial sweeteners during wastewater treatment: Implications for future use as tracers

Abstract:For more than a decade the artificial sweeteners acesulfame (ACE) and sucralose (SUC) have been applied as tracers of the input of wastewater to environmental waters. Recently concerns have been raised that degradation of ACE during treatment may hinder or restrict its use as a wastewater tracer. In this study the value of ACE and SUC as tracers was reassessed based on samples of wastewater at 12 municipal wastewater treatment (MWWT) plants and from 7 septic systems and associated septic plumes in groundwater. The results indicated stability of SUC during MWWT at most plants, and variable removal of both sweeteners during some MWWT and in the septic wastewater systems. However, the residual concentrations of ACE and SUC in municipal effluent and in septic plumes indicate that both sweeteners remain valuable wastewater tracers. The mass ratio SUC/ACE was found to be a useful parameter for examining the relative persistence of these sweeteners.
Occurrence, distribution, and ecological risk assessment of artificial sweeteners in surface and ground waters of the middle and lower reaches of the Yellow River (Henan section, China)

Abstract:As a new class of water contaminants, artificial sweeteners (ASs) have attracted much attention due to their environmental persistence and potential adverse effects to human and the environment. This study systematically investigated the occurrence and distribution of four commonly used ASs in the effluent of wastewater treatment plants (WWTPs), surface water and groundwater in the middle and lower reaches of the Yellow River (Henan section). Sucralose (SUC) was dominant in WWTP effluents and had the highest mass loading. Acesulfame (ACE), cyclamate (CYC), saccharin (SAC), and SUC were consistently detected in surface water at concentrations ranging from 1.364 ng/L (CYC) to 7786 ng/L (ACE). Spatial analysis showed that the pollution level of ASs in the trunk stream was lower than that in most tributaries. The total concentrations of ASs detected in surface water ranged between 308.7 and 10,498 ng/L, while in groundwater, the total concentration of ASs detected was between ND-4863 ng/L. ACE and SUC are the main pollutants in surface water and groundwater within this survey area. The risk assessment showed that the risks of the four target ASs to aquatic organisms were negligible (risk quotient (RQ) values < 0.1), and the maximum risk quotient of the mixtures (MRQ) values of all rivers were all much less than 0.1.
Levels, consumption, and variations of eight artificial sweeteners in the wastewater treatment plants of Dalian city, China

Abstract:Artificial sweeteners (ASs) are emerging contaminants in the environment, primarily derived from wastewater treatment plant (WWTP) effluents. In this study, the influents and effluents of three WWTPs in the Dalian urban area, China, were analyzed for the distribution of 8 typical ASs to investigate their seasonal fluctuations in the WWTPs. The results showed that acesulfame (ACE), sucralose (SUC), cyclamate (CYC), and saccharin (SAC) were both detected in the influent and effluent water samples of WWTPs, with concentrations ranging from not detected (ND) to 14.02 μg·L-1. In addition, SUC was the most abundant ASs type, accounting for 40 %-49 % and 78 %-96 % of the total ASs in the influent and effluent water, respectively. The WWTPs revealed high removal efficiencies of CYC, SAC, and ACE, while the SUC removal efficiency was poor (26 % ± 36 %). The ACE and SUC concentrations were higher in spring and summer, and all ASs showed lower levels in winter, which may be caused by the high consumption of ice-cream in warmer months. The per capita ASs loads in the WWTPs were determined in this study based on the wastewater analysis results. The calculated per capita daily mas loads for individual ASs ranged from 0.45 g·d-1·1000p-1 (ACE) to 2.04 g·d-1·1000p-1 (SUC). In addition, the relationship between per capita ASs consumption and socioeconomic status showed no significant correlation.
Calibration and field validation of POCIS passive samplers for tracking artificial sweeteners as indicators of municipal wastewater contamination in surface waters

Abstract:Polar organic chemical integrative samplers (POCIS) are widely used to track contaminants in surface waters. However, POCIS have not been used previously to monitor for artificial sweeteners as an indicator of wastewater pollution. In this study, we report for the first time the POCIS sampling rates (Rscal) for four artificial sweetener compounds, acesulfame (0.001 L/day), sucralose (0.114 L/day), cyclamate (0.001 L/day), and saccharin (0.002 L/day). We also prepared a modified POCIS with Strata X-AW anion exchange resin as a sorbent (i.e., ax-POCIS) and determined the sampling rates for sucralose (0.060 L/day) and acesulfame (0.128 L/day). Rscal values were adjusted according to the rate of loss of the performance reference compound, metoprolol-d6 from deployed POCIS to yield field sampling rates (i.e., Rsfield). Field validation of the monitoring method was conducted in Presqu'ile Bay on the north-central coast of Lake Ontario that is impacted by discharges from a sewage lagoon. POCIS were deployed at four sites within the bay and in the lagoon discharge. The four artificial sweeteners, as well as caffeine, ibuprofen, and other microcontaminants of sewage origin, were present throughout the bay at estimated concentrations in the ng/L range, and in the lagoon discharge at estimated concentrations higher by approximately one order of magnitude. Because acesulfame is present in ionic form over the pH range of natural waters, there are uncertainties related to the sampling rates using the standard POCIS. Sucralose is recommended as the best choice for source tracking using POCIS. There was good agreement between the concentrations of sucralose estimated from POCIS and the measured concentrations in grab samples of surface water in the bay. The present study provides key data for monitoring artificial sweeteners using POCIS.
Tracing pesticide dynamics: High resolution offers new insights to karst groundwater quality

Abstract:Generally, karst aquifers and springs are highly susceptible to contamination due to the high permeability and, therefore, groundwater flow velocities. The often thin soil cover, accompanied by dolines, can lead to fast infiltration of precipitation water loaded with mobilized contaminants such as pesticides and their transformation products. To date, continuous, temporally highly resolved in-situ monitoring to decipher concentration dynamics for a broad range of pesticides is missing. Therefore, a transportable HPLC-HRMS/MS system (MS2field) was positioned at two karst study sites in the Swiss Jura. Water samples were collected and analyzed for pesticides and their transformation products in-situ every 20 min for 6 weeks in 2021 and 8 weeks in 2022. During the spraying season in 2021, six rain events at site 1 and three at site 2 in 2022 were captured. Concurrently, the water quality parameters electrical conductivity, pH, nitrate, turbidity, and water level, were monitored continuously at high temporal resolution. Further, bacterial cell counts were monitored via online flow cytometry. In 2021, several pesticides and pesticide transformation products were detected in peak concentrations after rain events, of which metamitron showed the highest concentration of up to 1000 ng/L. In one rain event, the Swiss federal and EU drinking water limit of 100 ng/L was exceeded for up to 38 h. Compared with highly frequent MS2field samples collected every 20 min, 42-hours composite samples severely underestimated peak concentrations for all compounds, especially for labile ones. Therefore, it was demonstrated that exceedences of the regulatory limit would have been missed if just composite sampling would have been conducted. Peak concentrations of pesticides coincided with peaks in nitrate concentration and bacterial cell counts following rain events. The correlation analysis showed strong correlations between the three analyzed contaminants (pesticides, nitrate and bacteria), and the proxy parameters electrical conductivity, and pH. The investigation of a second spring revealed similar dynamics indicating that these can be expected in other karst aquifers as well.
Ecotoxicity of artificial sweeteners and stevioside

Abstract:Produced, consumed and globally released into the environment in considerable quantities, artificial sweeteners have been identified as emerging pollutants. Studies of environmental concentrations have confirmed the widespread distribution of acesulfame (ACE), cyclamate (CYC), saccharin (SAC) and sucralose (SUC) in the water cycle at levels that are among the highest known for anthropogenic trace pollutants. Their ecotoxicity, however, has yet to be investigated at a larger scale. The present study aimed to fill this knowledge gap by systematically assessing the influence of ACE, CYC and SAC and complementing the data on SUC. Therefore we examined their toxicity towards an activated sewage sludge community (30min) and applying tests with green algae Scenedesmus vacuolatus (24h), water fleas Daphnia magna (48h) and duckweed Lemna minor (7d). We also examined the effects caused by the natural sweetener stevioside. The high No Observed Effect Concentrations (NOECs) yielded by this initial evaluation indicated a low hazard and risk potential towards these aquatic organisms. For a complete risk assessment, however, several kinds of data are still lacking. In this context, obligatory ecotoxicity testing and stricter environmental regulations regarding food additives appear to be necessary.
Seasonal occurrence, removal and mass loads of artificial sweeteners in the largest water reclamation plant in China

Abstract:Artificial sweeteners (ASs) are of growing concern as an emerging contaminant. In the study, the seasonal occurrence, removal and mass load of six ASs in sewage, suspended particulate matter (SPM) and sludge were investigated throughout the treatment process of the largest water reclamation plant in China. The highest ASs concentrations in the influent (13.0 μg/L), effluent (2.22 μg/L), SPM (4.48 μg/g) and sludge (0.15 μg/g) were observed in the dry season, which were 1.24– to 5.0-fold higher than in the normal season and 1.06– to 37.5-fold higher than the flood season. Following treatment, ASs concentrations decreased by 24.3 %, 51.7 % and 5.1 % (on average) in primary, secondary and reclaimed processes, respectively. Among the investigated ASs, acesulfame (93.1 %) and cyclamate (98.4 %) were removed most efficiently, with removal occurring mainly in secondary processes, while sucralose exhibited the lowest removal efficiency (38.7 %). Seasonal characteristics affect the consumption of ASs, which subsequently changes the input and discharge ASs loads of STPs. The maximum mass load of ASs occurred in the dry season, ranging from 0.002 (neotame) to 1.33 mg/d/person (cyclamate), while the maximum emission load occurred in the flood season, ranging from 0.003 (neotame) to 0.83 mg/d/person (sucralose). The mass and emission load of ASs in Beijing is significantly lower than in European or the United States, due to Beijing having low per capita consumption of ASs (5.50 mg/d/person). The highest ASs risk in the receiving water occurred in the flood season due to the input of other pollution sources by rainfall runoff. Meanwhile, attention should be paid to the risk of receiving water close to the STP outlet in the dry seasons for the highest ASs concentration in the STP effluent in the season. The present study provides important guidance on controlling the input and reducing the emission of ASs in different seasons.

Abstract:The accurate determination of widespread artificial sweeteners (ASs) and the information of their distributions in environments are of significance to investigate the environmental behaviors. This paper firstly reviews the typical analytic methodologies for ASs and the main influencing factors during the analytic processes. Solid-phase extraction (SPE) with LC-ESI-MS is currently the leading-edge method. However, the efficiency and accuracy for ASs analysis in environmental samples are also dependent on the SPE cartridges, buffers and pH, matrix effects, and sample stability. A basic procedure for ASs determination in different environmental samples is proposed. The current occurrences of ASs in environments are then evaluated. The ASs, especially the acesulfame and sucralose, are widely detected in various environmental medium. The concentrations of investigated ASs are generally in the order of wastewater treatment plants (WWTPs) influent > WWTPs effluent > surface water > groundwater > drinking water; and atmosphere > soil. The ASs levels in the environment exhibit significant differences among different regions. Further analysis indicates that the phenomenon is highly correlated with the consumption patterns and the removal efficiency of WWTPs in a specific country.

Abstract:Karst aquifers are important resources for drinking water supply and are very vulnerable to contamination. Microbial concentrations at karst springs, in particular, often vary quickly over a short period of time. In this study, the response of microbial water quality and particle-size distribution of two alpine karst springs to rainfall events was investigated to test and validate parameters that can be used as early-warning systems for fecal contamination. At both investigated karst springs, total organic carbon, particle-size distribution (especially small particle fractions), and particle load show a good correlation to the fecal indicator bacteria E. coli and can therefore be used as a real-time indicator of fecal contamination at the investigated springs. In addition to conventional bacterial determination methods, the β-D-glucuronidase activity, which can be measured in near real-time, was used as a novel indicator parameter for fecal contamination. At the event scale, the β-D-glucuronidase (GLUC) activity shows a good correlation to E. coli and can be used as an additional real-time indicator of fecal contamination. For the studied springs, when they show two peaks in turbidity and small particles, these two parameters are suitable for an early warning system because the bacterial contamination occurs during the secondary peak of these parameters. These results highlight the vulnerability of karst aquifers and demonstrate the applicability of advanced measurement techniques in detecting fecal contamination in real-time, which is especially important given the time-consuming nature of conventional bacterial detection methods.

Abstract:Artificial sweeteners (ASs), as a new emerging pollutant, the fate from wastewater treatment plants (WWTPs) to sea is growing concerned. In this study, the distribution and polluted loading of six typical ASs were analyzed based on the measurement in influent, primary effluent, secondary effluent, tertiary effluent, suspended particulate matter (SPM), and sludge from four WWTPs and receiving waters near the coast of Shenzhen city, China. Elevated levels of ASs were detected in WWTPs located near a highly populated area (about 14,000 people km-2). Sucralose was predominant in all water samples (0.1–22.2 μg L-1), and had a 1.6–704.0 times higher concentration than the means of other ASs detected. Aspartame were mainly distributed in SPM and sludge, and the fractions in offshore water exceeded 45%. Acesulfame, sucralose, cyclamate and saccharin could be effectively removed by secondary biochemical treatment. The average daily loading (4.2 g d-1 1000people-1) and discharge loading (0.4 g d-1 1000people-1) of sucralose in WWTPs was higher than those of the other ASs. Dissolved organic carbon and UV254 can affect the fate of ASs, and SPM mainly affects the distributions of aspartame and neotame. As a potential sewage indicator, neotame deserves further attention.

Abstract:Artificial sweeteners are a class of low-level emerging organic contaminants that occur in the environment around the world. Once ingested by humans, major amounts of artificial sweeteners are excreted unchanged from the body and are added to the water environment via sewage systems. Consequently, artificial sweeteners pose a new threat to the aquatic environment, which is the primary recipient of these substances. Recently, they have been detected in the area of the Káraný waterworks, Czech Republic, where there is a riverbank filtration system. Considerable attention has been given to Acesulfame-K, which has proved to be the most frequently occurring contaminant in the Jizera river, with a concentration range from 72.0 to 591.0 ng/L. Although the riverbank filtration systems remove many anthropogenic contaminants in water, Acesulfame-K has continued to be detected in the groundwater, with a concentration range from <49.9 to 71.7 ng/L. Therefore, the main purpose of this study is to describe and quantify the attenuation behaviour within a riverbank filtration site by providing model-based estimates of the first-order degradation rate constant. For Acesulfame-K, the first-order degradation rate was identified: λ = 0.0358 ± 0.0022 1/d. This result was further confirmed by a small-scale laboratory experiment.

Effects of common artificial sweeteners at environmentally relevant concentrations on soil springtails and their gut microbiota

Abstract:Artificial sweeteners (AS) are extensively utilized as sugar substitutes and have been recognized as emerging environmental contaminants. While the effect of AS on aquatic organisms has garnered recent attention, their effects on soil invertebrates and gut microbial communities remain unclear. To address this knowledge gap, we exposed springtails (Folsomia candida) to both single and combined treatments of four typical AS (sucralose [SUC], saccharin [SAC], cyclamate [CYC], and acesulfame [ACE]) at environmentally relevant concentrations of 0.01, 0.1 and 1 mg kg-1 in soil. Following the first-generational exposure, the reproduction of juveniles showed a significant increase under all the AS treatments of 0.1 mg kg-1. The transcriptomic analysis revealed significant enrichment of several Kyoto Encyclopedia of Gene and Genome pathways (e.g., glycolysis/gluconeogenesis, pentose and glucuronate interconversions, amino sugar, and nucleotide sugar metabolism, ribosome, and lysosome) in springtails under all AS treatments. Analysis of gut bacterial microbiota indicated that three AS (SUC, CYC, and ACE) significantly decreased alpha diversity, and all AS treatments increased the abundance of the genus Achromobacter. After the sixth-generational exposure to CYC, weight increased, but reproduction was inhibited. The pathways that changed significantly (e.g., extracellular matrix-receptor interaction, amino sugar and nucleotide sugar metabolism, lysosome) were generally similar to those altered in first-generational exposure, but with opposite regulation directions. Furthermore, the effect on the alpha diversity of gut microbiota was contrary to that after first-generational exposure, and more noticeable disturbances in microbiota composition were observed. These findings underscore the ecological risk of AS in soils and improve our understanding of the toxicity effects of AS on living organisms.

All four ASs (acesulfame (ACE), sucralose (SUC), cyclamate (CYC) and saccharin (SAC)) were detected, ranging from 0.43 to 27.34μg/L in the influent. Higher concentrations of ASs were measured in winter. ACE could be partly removed by 7.11-50.76% through biodegradation and especially through the denitrifying process. The A2/O process was the most efficient at biodegrading ASs. Adsorption (by granular activated carbon (GAC) and magnetic resin) and ultraviolet radiation-based advanced oxidation processes (UV/AOPs) were evaluated to remove ASs in laboratory-scale tests. The amounts of resin adsorbed were 3.33-18.51 times more than those of GAC except for SUC. The adsorption ability of resin decreased in the order of SAC > ACE > CYC > SUC in accordance with the pKa. Degradation of ASs followed pseudo-first-order kinetics in UV/H2O2 and UV/PDS. When applied to the secondary effluent, ASs could be degraded from 30.87 to 99.93% using UV/PDS in 30 minutes and UV/PDS was more efficient and economic.
Outcomes of community re-entry programmes for brain injury survivors. Part 1: Independent living and productive activities.

Abstract:This is a study of the multiple outcomes of residential community re-entry programmes for disabled survivors of brain injury. The sample consisted of 82 clients from nine facilities across the USA. The level of functioning of the client at and before admission was determined from clinical evaluations reported in programme records. Telephone interviews assessed outcomes 1 year after discharge. The participation in the re-entry programmes led to enduring improvements in independent living and productive activities. Total supervision requirements decreased substantially. Paid employment increased, but benefits went much beyond this to include improvements in educational activities, unpaid vocational activities, and especially household management. For the most part, these improvements cannot be attributed to chronicity or natural healing. The benefits of the re-entry programmes were clearly multiple. The results generally affirm the effectiveness and utility of community re-entry programmes.
Untreated wastewater impact and environmental risk assessment of artificial sweeteners in river water and sediments of the Danube River Basin in Serbia

Abstract:AbstractArtificial sweeteners are receiving increasing attention as newly recognized emerging contaminants that mainly reach the aquatic environment through the discharge of municipal wastewater containing large amount of these compounds. In this study, the impact of raw untreated wastewater discharges on the levels and the water/sediment distribution of artificial sweeteners in the Danube River and its largest tributaries in Serbia was evaluated, and a comprehensive assessment of environmental risks for freshwater and benthic organisms was performed. Acesulfame and sucralose were detected in all river water samples (100%), while saccharin (59%) and cyclamate (12%) were less frequently found, indicating long-term continuous sewage-derived pollution. Aspartame (100%) and neotame (60%) were the only artificial sweeteners recorded in the sediment samples due to their preference to sorb to particulate matter in the water/sediment system. In terms of ecotoxicological risk, a low risk for aquatic organisms was determined at the detected levels of saccharin in river water, while a high to medium risk was found for benthic biota at the concentrations of neotame and aspartame detected in sediments. The largest contribution to the pollution of the Danube River Basin with artificial sweeteners, and consequently the highest environmental risk, was determined in the two largest cities, the capital Belgrade and Novi Sad, which raises the issue of transboundary pollution.

Abstract:Tracking contaminants in karst aquifers is challenging because of the high heterogeneity encountered in carbonate rocks. Multi-tracer tests, combined with chemical and isotopic analyses, were conducted to solve a groundwater contamination incident within a complex karst aquifer in Southwest China. Results showed that: (1) the wastewater from a paper mill, public sewers, and septic tanks were the three main potential contaminant sources identified by chemical and isotopic methods; (2) a direct effect of the paper mill wastewater with high Na+ (up to 2230.5 mg/L) and chemical oxygen demand (COD) concentrations on spring water quality was confirmed by multi-tracer tests, which changed the water type from Ca-HCO3 in the 1970s to Ca-Na-HCO3 in the present study and resulted in a depleted carbon isotope value (-16.5‰); and (3) the studied aquifer is a highly complex karst system, due to two conduits crossed each other without mixing, contaminants traveled a long distance (up to 14 km) within the lower conduit, paper mill-contaminated groundwater flowed across a river bottom and discharged to the opposite bank, and an active subsurface divide occurred. After several months of operation, the groundwater restoration measure based on karst hydrogeologic conditions proved that cutting off contaminant sources for karst aquifer self-restore was effective in practice, which contributed to the decline in NH4+ (from 7.81 mg/L to 0.04 mg/L), Na+ (from 50.12 mg/L to 4.78 mg/L), and COD (from 16.42 mg/L to 0.9 mg/L) concentrations coupled with an increase in δ13C-DIC value (from -16.5‰ to -8.4‰) in the earlier contaminated karst spring. This study's integrated method is expected to screen and confirm contaminant sources within complex karst systems rapidly and effectively, thereby contributing to karst groundwater environmental management.

Abstract:Septic systems are a common contributor of contaminants to groundwater that have implications for source water protection, particularly in fractured sedimentary bedrock environments. Two 24-h tracer experiments were performed that applied (1) the dye Lissamine Flavine FF and (2) three artificial sweeteners (acesulfame, sucralose, and cyclamate) in the leaching bed to examine solute transport from a single-family septic bed to a multilevel monitoring well installed in fractured sedimentary bedrock on a First Nation reserve in Southern Ontario, Canada. Tracer was first observed 3 h and 20 min after deployment, and breakthrough curves showed that multiple pathways likely exist between the septic bed and the monitoring well. Cyclamate concentrations were more elevated than expected compared to other studies that examined cyclamate’s attenuation in the laboratory and in porous media aquifers. Solute transport through the septic bed was analyzed with the numerical modeling software Hydrus 1D, which indicated that the septic bed may be too thin, located directly on bedrock, underlain by fractured soils, or bypassed through a short-circuit. The rapid transport of septic leachate to fractured sedimentary aquifers is problematic for First Nation and rural communities. More stringent regulations are needed for the design and use of septic systems in these environments.
