Abstract:Advances in germplasm enhancement of tea breeding, techniques for identification of new tea cultivars, changes in breeding objectives, improvement of breeding program, achievements of tea breeding, system of propagation and extension of new tea cultivars were reviewed in this paper, which provides useful information for further studies in tea genetics and breeding. Hybridization is still the major method for innovating tea breeding materials, and the physical and chemical mutagenesis methods have been extensively used, while transgenic technique remains to be further improved. Combination of the high resolution inspecting equipments and the gene identification technology with the forecasting models established by computer technology made the early identification of tea cultivars more accurate. The breeding target has been diversified after going through yield breeding, quality breeding and early flushing breeding stages. Improving early identification accuracy and shortening breeding cycle will be the aim for tea breeding technology development.

Abstract:Tea is the most popular beverage, prepared with leaves of Camellia sinensis (L.) O. Kuntze, consumed by almost all communities and has economic, medicinal and cultural importance. The tea plant originated in southwestern China around 5000 years ago and is now grown in over 52 countries. Tea-breeding activities first started in ancient China and the selection of promising plants from natural variability of heterogeneous populations was practiced in the early years. This chapter highlights limitations in the traditional crop improvement programs and the necessity for adopting novel biotechnological tools to overcome them. The role of tea genetic resources in crop improvement, an overview of tea genetic resources in the world and conservation attempts made to date are discussed. Molecular markers have been used to improve the crop in tea-growing countries. The progress and significant achievements of marker-assisted selection for important traits are highlighted. A comprehensive account is given of genetic linkage maps constructed. The recent developments in functional genomic research and the progress of the omics approaches in tea are discussed. Several attempts have been made to identify functional genes associated with important traits. The progress of transgenic research in tea during the last two decades is summarized. The first genome sequence of tea was reported in 2017, which is a milestone in genome research. Mutation breeding has the potential to be used as an alternative tool to develop new cultivars in tea. The progress on controlled hybridization between selected parents, the most preferred strategy for tea crop improvement to date, is discussed. The chapter provides an overview of the current status and future prospective on the application of new technologies to combat global climate change.

Abstract:The genetics and breeding of tea plant are important research areas of tea science disciplines, which involve the study on genetic mechanism and breeding technology of tea plants.In 2016, there were 154 academic papers involving in the research field of genetics and breeding of tea plants, among which 80 were published in domestic journals.The topics of these papers included tea germplasm resources and their classification, innovation of breeding materials, identification and breeding of improved tea cultivars, propagation and cultivation of the obtained thoroughbred tea cultivars.The advances in tea genetics and breeding in 2016 were reviewed in the present paper, which provides reference for related researchers.

Abstract:The achievements in the 20th century and the future development trend of tea breeding and tea cultivation in China were described in the present paper. China, with 1,076,220 ha tea field, is now one of the major tea producing and tea exporting countries in the world and its tea field area ranked the first in the world. Chinese tea industry has been greatly developed in the 20th century because of the development of tea breeding and cultivation techniques.The development of tea breeding in China was benefited from the techniques of vegetable propagation, cross breeding, chemically and physically induced mutation as well as bio-technique and molecular biological technique. There were five stages in the development of tea breeding in China, i.e. the non-organized stage, clonal tea breeding stage, yield breeding stage, quality breeding stage and resistance breeding stage.The tea production in China has been fast developed in the 20th century, especially in the last 50 years. It was mainly benefited from the increase of planting density, the popularization of improved tea cultivars, the manuring technique, the efficient and low-cost cultivation technique.The development trend in Chinese tea breeding will be pest-resistance breeding, high quality breeding and breeding of some special use tea cultivars such as those with high content of catechins, low content of caffeine and low activity of polyphenol oxidase. Intensive management, low-cost and sustainable production will be the important research topics in Chinese tea cultivation in the early decades of the 21st century.

Abstract:The indigenous Assam tea plant was discovered by Robert Bruce in 1823. However, commercial tea cultivation was initiated in India during 1834, using a China tea plant. The better cup quality produced by the Assam tea plant popularized it as an important planting material in the tea industries of the country. Seeds were the only source of propagation until the discovery of the vegetative method. As a result, seed jat populations of unknown parents created a wide range of genetic variation and resulting inconsistency in their performance. Considering the need for improved planting materials for the tea industry, Tocklai Experimental Station (TES) Tea Research Association, initiated a tea breeding programme in 1930, under which germplasms were collected based on trait specific phenotypic characteristics. Promising plants selected from heterogeneous jat populations as well as from wild tea patches were characterized and preserved in the gene bank of TES, along with some of the non-tea Camellia species for utilization in the breeding programmes. The Tea Research Foundation, United Planters’ Association of Southern India (UPASI), initiated a similar program in 1963 and collected germplasms were preserved in their gene bank. The technique of vegetative propagation, standardized in 1955, provided scope for developing improved clonal cultivars as well as biclonal seed cultivars through hybridization. From the selected plants from old seed jats and progenies of biclonal hybrids, 153 locally adapted and 31 universal clones were developed for the tea industry. Under polyploid breeding triploids, tetraploids and aneuploids were produced through hybridization, out of which high yielding quality triploid plants were selected, cloned and made available for plantation. Water logging tolerant genotypes have been selected and used as parents in the breeding programme for the development of tolerant cultivars and progenies. EST’s have been developed from Camellia species and a cDNA library was constructed. Marker development for draught resistance and blister blight disease is under progress using cDNA-AFLP and EST-SSR techniques. Transgenic technology has been developed and vector construction is completed to confer resistance against blister blight. The micropropagation technique has also been standardized for quick multiplication of these biotechnologically modified plantlets. Details of tea breeding in India are discussed in this chapter.

Abstract:During the past 50 years, 10 new tea cultivars of Zhenong series were bred including three national registered tea cultivars (Zhenong 12, Zhenong 113. and Zhenong 21), four provincial registered tea cultivars (Zhenong121, Zhenong25, Zhenong117 and Zhenong139).There are three cultivars to be registered (Zhenong23, Zhenong8-8 and Zhenong6-10) and 20 other new hybrid lines. Besides, early stage evaluation of major economic traits, genetic dynamics of some traits, regeneration system of tea plant and genetic transformation were investigated and some important achievements were obtained. Finally, the following suggestions were summed up: 1. The success of tea breeding mainly depended on the correct selection of the germplasm. 2. The investigation on the early-stage evaluation needs to be enhanced. 3. The spread of registered tea cultivars must be combined with the making of the famous tea. 4. The investigation on the tea breeding theory and new breeding technique must be strengthened.

Abstract:Background: Tea seed (TS), as the mature fruit and growth source of tea plant, has been characterized with appealing bioactive substances, remarkable health benefits and diverse application prospects. As one of the most significant products of TS, tea seed oil (TSO) has gained recognition as the "oriental olive oil". Tea seed meal (TSM) and tea seed shell (TSS) continue to be underutilized and are usually considered as low-value by-products. Nevertheless, current exploration of the underlying mechanisms is inadequate, and investigations concerning deep processing and potential prospects still remain at their infancy. Consequently, there exists a gap in the comprehensive aspects of TS utilization and the related advancements.Scope and approach: In this review, we have summarized the latest research progresses on TS, specifically focused on the bioactive substances, pharmacological activities, manufacturing process and potential applications. Furthermore, we have also elucidated the current challenges in existing studies, and proposed the research orientations and potential prospects of TS.Key findings and conclusions: TS contain abundant bioactive substances, and exhibit outstanding performance in antioxidant, anti-tumor, anti-obesity, anti-microbe, gastric protection, and so on. These properties contribute to its application potentials as crucial materials in industry, agriculture and ecology, as well as new perspective in drug development and clinical translation. Future researches should concentrate on the specific active in-gredients, intricate structure-activity relationships, bioactive mechanisms, undiscovered health benefits and manufacturing process, which would facilitate the transformation of waste resources into valuable treasures, thus fostering sustainable development and socio-economic growth.

Abstract:Research advances in genetics and breeding of tea in 2011 were reviewed. Useful tea germplasms were transformed into productivity through close association of germplasms collection and application. Studies on DNA markers and gene cloning were hot spots in the basic research field. Chemical and morphological identifications were major techniques for appraising quality potential of tea cuhivars. Quality and productivity efficiency were promoted by close combination of good agricultural practices and well-bred tea eultivars. Ecological adoptability of tea cultivars was the focus of attention. Some new tea cultivars were released in this year.

Abstract:Although China and India are the two largest tea-producing countries, the domestication origin and breeding history of the tea plant in these two countries remain unclear. Our previous study suggested that the tea plant includes three distinct lineages (China type tea, Chinese Assam type tea and Indian Assam type tea), which were independently domesticated in China and India, respectively. To determine the origin and historical timeline of tea domestication in these two countries we used a combination of 23 nSSRs (402 samples) and three cpDNA regions (101 samples) to genotype domesticated tea plants and its wild relative. Based on a combination of demographic modeling, NewHybrids and Neighbour joining tree analyses, three independent domestication centers were found. In addition, two origins of Chinese Assam type tea were detected: Southern and Western Yunnan of China. Results from demographic modeling suggested that China type tea and Assam type tea first diverged 22,000 year ago during the last glacial maximum and subsequently split into the Chinese Assam type tea and Indian Assam type tea lineages 2770 year ago, corresponding well with the early record of tea usage in Yunnan, China. Furthermore, we found that the three tea types underwent different breeding histories where hybridization appears to have been the most important approach for tea cultivar breeding and improvements: a high proportion of the hybrid lineages were found to be F2 and BCs. Collectively, our results underscore the necessity for the conservation of Chinese Assam type tea germplasm and landraces as a valuable resource for future tea breeding.

Abstract:Tea plants are an economically important crop and conducting research on tea breeding contributes to enhancing the yield and quality of tea leaves as well as breeding traits that satisfy the requirements of the public. This study reviews the current status of tea plants germplasm resources and their utilization, which has provided genetic material for the application of multi-omics, including genomics and transcriptomics in breeding. Various molecular markers for breeding were designed based on multi-omics, and available approaches in the direction of high yield, quality and resistance in tea plants breeding are proposed. Additionally, future breeding of tea plants based on single-cellomics, pangenomics, plant–microbe interactions and epigenetics are proposed and provided as references. This study aims to provide inspiration and guidance for advancing the development of genetic breeding in tea plants, as well as providing implications for breeding research in other crops.

Abstract:The present paper compares the tea crop in Kenya and China from plant genetic improvement perspective with the associated plant improvement factors. Comparison is made for ecological production conditions noting the vast area lying under different environmental conditions for China that are suitable for growing of the different varieties of tea to the limited choice of conditions in Kenya. China has both subtropical and temperate conditions in both elevated and low altitude areas in which tea is conditioned to grow unlike Kenya that grows its tea specifically in cool high altitude areas that have high and well-disiributed rainfall. Tea is noted to have originated in China and has been under cultivation in that country for thousands of years. Historical development and climatic conditions in the respective countries is noted to have been key in determining the choice in the selection and adaptation of the different varieties within different regions and between the two countries. However, producer preferences, and "tradition" of tea growing and ecological conditions are the main conservative limitation to the range of varieties exploited in Kenya. With historical initial source of planting material and proximity to India, the subtropical China varieties have had remote influence on choice of the types and varieties widely grown in Kenya. With evolution of the crop in Kenya, and challenges to the crop being manifested, lessons and experiences need revisiting. An overview of the crop challenges and management structure of the industry are presented Possible lessons and opportunities for exploitation in genetic improvement of the crop are summarized. The paper is presented with the objective of providing a forum on which the two countries can examine opportunities on which information is shared for technological and biological advancement of the tea crop.

Abstract:Tea is one of the main foreign exchange earners of Sri Lanka. Sri Lanka ranks as one of the largest exporters of black tea in the world. Sri Lanka is well renowned for its high quality orthodox type (95% of the total production) black tea in the international market. The name “Ceylon tea” or “Sri Lankan tea” has been regarded as a sign of high quality throughout the world for a long time and even today, as reflected by the prices. In order to maintain the position of “Ceylon tea” in the world and to serve the local tea industry, it is imperative to focus on efficient tea crop improvement strategies to develop grower acceptable tea cultivars. Commencement of the tea crop improvement program at the Tea Research Institute of Sri Lanka dates back to the 19th century. Since then many significant achievements have been made in developing new tea cultivars, with salient milestones over the years. Growers have benefited immensely by using the improved tea cultivars developed by the Institute, though certain improvements have yet to be accomplished using the modifications made in the current breeding program. This chapter highlights significant achievements in the areas of germplasm collection, characterization and evaluation, and their use in the tea breeding program, with prominence given to cost effective complementary strategies adopted in germplasm conservation and the holistic approach adopted in germplasm characterization. Tea breeding strategies, priorities and the significant contribution made to the industry by developing improved tea cultivars over the years are discussed, emphasizing the diverse needs of the growers in multiregional tea growing areas in the country. The chapter also focuses on the application of biotechnological tools for breeding and crop improvement, highlighting the recent advances made in the development of new technologies and their practical applications in facilitating conventional tea crop improvement programs. Overall, the chapter reviews the achievements, challenges and perspectives of tea breeding in Sri Lanka and overviews the future trends, aims and goals of breeding improved cultivars acceptable to the growers. There is a focus on producing improved tea cultivars that thrive better under changing environments to meet the ever-changing demands of the end-users. Future perspectives on the application of new technologies to address key challenges faced by the industry locally and globally, by focusing on the integration of advanced biotechnological tools and decentralized participatory approaches to the current tea breeding program are also discussed.

Abstract:Endophytic fungi and bacteria are the most ubiquitous and representative commensal members that have been studied so far in various higher plants. Within colonization and interaction with their host plants, endophytic microbiota are reportedly to modulate not only the host’s growth but also holobiont resilience to abiotic and biotic stresses, providing a natural reservoir and a promising solution for sustainable agricultural development challenged by global climate change. Moreover, possessing the talent to produce a wide array of high-value natural products, plant endophytic microbiota also serve as an alternative way for novel drug discovery. In this review, tea, one of the world’s three largest nonalcoholic beverages and a worldwide economic woody crop, was highlighted in the context of endophytic microbiota. We explore the recent studies regarding isolation approaches, distribution characteristics and diversity, and also biological functions of endophytic microbiota in Camellia sinensis (L.) O. Kuntze. Profoundly, the future insight into interaction mechanism between endophytic microbiota and tea plants will shed light on in-depth exploration of tea microbial resources.
Genetic Diversity among Tea Cultivars from China, Japan and Kenya Revealed by Issr Markers and Its Implication for Parental Selection in Tea Breeding Programmes

Abstract:Tea plant [Camellia sinensis (L.) O. Kuntze] is an important beverage crop in the world. In recent years many clonal tea cultivars have been released, and they play major roles in improving the production and quality of tea. It is important to understand the genetic diversity and relatedness of these cultivars to avoid inbreeding and narrow genetic basis in future tea breeding. In the present study, genetic diversity and relationship of 48 tea cultivars from China, Japan and Kenya were evaluated by inter-simple sequence repeat (ISSR) markers. A total of 382 ISSR bands were scored, of which 381 (99.7%) were polymorphic. The ISSR primers showed high ability to distinguish between tea cultivars according to their high Resolving Power (R-P) with an average of 7.4. The mean of Nei's gene diversity (H) and Shannon's information index (I) were 0.22 and 0.35, respectively. More abundant diversity was revealed among cultivars in China than those in Japan and Kenya. Within Chinese populations, the level of diversity in east China was higher than that in other regions. The coefficient of genetic differentiation (G(ST)) was 0.202, which indicates a high degree of genetic variation within populations. This result was further confirmed by analysis of molecular variance, which revealed the variance component within the populations (92.07%) was obviously larger than that among populations (7.93%). The level of gene flow (N-m) was estimated to be 2.0. This could be explained by frequent natural cross-pollination and seed dispersal among tea populations. The pairwise similarity coefficient between the cultivars varied from 0.162 to 0.538. A dendrogram of 48 tea cultivars was constructed where all the tested cultivars were divided into two groups. Our data show that the genetic relationship among tea cultivars can be determined by the ISSR markers. This will provide valuable information to assist parental selection in current and future tea breeding programmes.
Genetic Control Studies to Exploit Flavonoids Synthesis Pathway in Tea Biochemical Improvement

Abstract:The main characteristic components in tea are the tea polyphenols; flavonoids derivatives and caffeine. The basic tea polyphenols are secondary metabolites among the flavonoids group of compounds that accumulate in high concentration in the young tender leaves of tea. The catechins share the major part of the biosynthetic pathway with other derivative flavonoids products, hence similar synthesis mechanisms. The widely studied pathway in several plant species is tightly controlled by a series of structural and regulatory genes determining the main products of the pathway. Understanding the biochemical, molecular and genetic control of tea polyphenols synthesis pathway is necessary in order to derive maximum value from the tea genetic improvement process. The pathway and critical genes in the flavonoids synthesis leading to the synthesis of tea catechins are discussed. Potential value for knowledge of the pathway in tea and its application is enumerated.

Abstract:The quality and high yield tea Cultivar Zhenong 176 has been bred by means of systematic selection and vegetative propagation from the hybrids of Fuding Dabaicha×Zhenong 12.The result of field test showed that the sprouting period of zhenong 176 was similar to Fuding Dabaicha(control),and it was suitble to produce green tea,and its yield and quality were higher than the control. The result of Chemical components analysis showed that content of tea polyphenols and animo acid was higher than the control by 4.1% and 8.7%,respectively. Resistance investgation showed that its resistance to drough and cold conditions was better than the control,and resistance to pests was similar to the control.
