Abstract:Individual differences in working memory (WM) predict principled variation in tasks of reasoning, response time, memory, and other abilities. Theoretically, a central function of WM is keeping task-relevant information easily accessible while suppressing irrelevant information. The present experiment was a novel study of mental control, using performance in the game Taboo as a measure. We tested effects of WM capacity on several indices, including perseveration errors (repeating previous guesses or clues) and taboo errors (saying at least part of a taboo or target word). By most measures, high-span participants were superior to low-span participants: High-spans were better at guessing answers, better at encouraging correct guesses from teammates, and less likely to either repeat themselves or produce taboo clues. Differences in taboo errors occurred only in an easy control condition. The results suggest that WM capacity predicts behavior in tasks requiring mental control, extending this finding to an interactive group setting.
Effects of Pb(Ⅱ) exposure on Chlorella protothecoides and Chlorella vulgaris growth, malondialdehyde, and photosynthesis-related gene transcription

Abstract:Greater exposure to Pb(Ⅱ) increases the likelihood of harmful effects in the environment. In this study, the aquatic unicellular alga Chlorella protothecoides (C. protothecoides) and Chlorella vulgaris (C. vulgaris) were chosen to assess the acute and chronic toxicity of Pb(Ⅱ) exposure. Results of the observations show dose-response relationships could be clearly observed between Pb(Ⅱ) concentration and percentage inhibition (PI). Exposure to Pb(Ⅱ) increased malondialdehyde (MDA) content by up to 4.22 times compared with the control, suggesting that there was some oxidative damage. ANOVA analysis shows that Pb(Ⅱ) decreased chlorophyll (chl) content, indicating marked concentration-dependent relationships, and the lowest levels of chl a, chl b, and total-chl were 14.53, 18.80, and 17.95% of the controls, respectively. A real-time PCR assay suggests the changes in transcript abundances of three photosynthetic-related genes. After 120 h exposure Pb(Ⅱ) reduced the transcript abundance of rbcL, psaB, and psbC, and the relative abundances of the three genes of C. protothecoides and C. vulgaris in response to Pb(Ⅱ) were 54.66-98.59, 51.68-95.59, 37.89-95.48, 36.04-94.94, 41.19-91.20, and 58.75-96.80% of those of the controls, respectively. As for 28 d treatments, the three genes displayed similar inhibitory trend. This research provides a basic understanding of Pb(Ⅱ) toxicity to aquatic organisms.

Abstract:Recently, the pollution of microplastics (MPs) in the global freshwater environment has become increasingly problematic, but there are few studies on the freshwater environment risks of MPs. The present study, therefore, has investigated the single and combined effects of MPs and lead (Pb) on the freshwater algal Microcystis aeruginosa. Results showed that Pb-only (>0.05 mg·L−1) promoted the growth of algal cells, while MPs-only (1 mg L−1) resulted in growth inhibition. However, compared with the corresponding concentration of Pb-only groups, the growth of algal cells was promoted in MPs + Pb treatments. MPs-only and Pb-only (0.5 mg L−1) both reduced the content of photosynthetic pigments and affected algal photosynthesis. The MPs-only treatment and MPs + Pb2+ (no pretreatment, 0.5 mg L−1 Pb2+) treatments showed significant cell aggregation. At the same time, MPs-only caused a significant increase in bound extracellular polysaccharides (bEPS), while 0.5 mg L−1 Pb reduced bEPS. Furthermore, under high Pb stress (0.5 mg L−1), the effects of combined MPs and Pb on chlorophyll content, antioxidant enzyme activity (peroxidase (POD), catalase (CAT)), and damage to algal cells were less compared to individual effects, and the combination of MPs and Pb had a synergistic effect on promoting aggregations of M. aeruginosa. These results demonstrate that single and combined effects of MPs and Pb can induce differential responses in the freshwater algal M. aeruginosa, which can have a significant impact on aquatic ecosystems.

Impacts of phosphorus limitation in the toxicity of environmental concentrations of lead and zinc in Raphidocelis subcapitata (Chlorophyceae)

Abstract:Microalgal metabolism is affected by the surrounding environment and nutrients such as phosphorus (P) and nitrogen (N) are essential for optimal metabolism, as well as trace amounts of essential metals such as zinc (Zn); although in higher doses than required, Zn can be toxic. Lead (Pb) is a non-essential metal that can harm organisms from different trophic levels. In the environment, algae are exposed to several stressors simultaneously and adapt their metabolism. In the present study, we evaluated P limitation combined with environmental concentrations of Zn or Pb to the freshwater microalga Raphidocelis subcapitata regarding growth, pigments production, and photosynthetic parameters. Our results indicate that P limitation affected the growth, pigments production, relative maximum electron transport rate (rETR max ), and saturation irradiance; while Pb altered growth, pigments production, and maximum quantum yield; and Zn affected pigment production, photochemical and non-photochemical quenching, and rETR max . However, the combination of metal and P limitation resulted in synergistic responses, i.e., higher damages than the isolated stressors, in growth, maximum and effective quantum yield, and in the rapid light curve parameters. On the other hand, antagonism, i.e., lower damages than isolated stressors, was observed in pigments production and non-photochemical quenching, suggesting that algae activated defense mechanisms to cope with both stressors simultaneously. In addition, our results indicate an algal metabolism adjustment to P limitation and highlight the importance of considering physicochemical water characteristics when defining regulations of acceptable levels of metals in aquatic ecosystems.

Abstract:Copper sulfate (CuSO4) is widely used in the control of algal blooms. Cu can promote or inhibit algal growth, while also affecting trace element uptake, therefore, the response mechanisms of algae cells under Cu2+ interference should be studied. In this study, wild-type Chlamydomonas reinhardtii (C. reinhardtii) and wall-less mutant C. reinhardtii were selected as the research objects. Except for the cell wall, these two algae were physiologically the same. While manipulating the concentration of Cu, the accumulation of Cu, Fe, Zn, and Mn by the two algal cell types was studied. The cell wall hindered the accumulation of Cu by cells and alleviated the toxicity of Cu to C. reinhardtii. The addition of Cu increased the accumulation of Fe by both cell types. In an environment with excess Cu, the total amount of Zn and Mn accumulated by cells also increased. On the one hand, this may be due to the synergistic and antagonistic effects of trace elements in the adsorption and uptake process, and on the other hand, it may be due to the changes in metal speciation in the culture medium. In addition, the difference in the total accumulation of various trace elements between wild-type and wall-less-type C. reinhardtii may be due to the structure and function differences between cell wall and cell membrane. At the same time, by measuring the changes in the levels of glutathione (GSH) in algal cells, the relevant mechanisms underlying the algae's uptake of trace elements by algae were further explored.
Interaction between Haematococcus pluvialis microalgae and lead nitrate: lead adsorption from water

Abstract:Our study aims to investigate the response of the unicellular alga, Haematococcus pluvialis , to the toxicity of lead and propose a low-cost, highly efficient biological adsorbent for the purification of wastewater and lead-contaminated water. The first part examines the effects of lead toxicity on certain physiological indicators of this alga. In the second part, the potential of this alga in lead removal and its adsorption capacity was assessed. The alga was cultivated in a BG11 medium and treated with lead nitrate concentrations of 10, 50, and 200 mg/L during its exponential growth. The results showed that with an increase in lead concentration up to 200 mg/L, the growth rate, chlorophyll a , chlorophyll b , carotenoid and total protein content decreased, while malondialdehyde (MDA) content increased. The astaxanthin content slightly increased at the 10 mg/L but decreased at the 200 mg/L treatment. Maximum lead adsorption was observed at 98.69% under optimal conditions, including a pH of 6, an adsorbent dose of 1 g/L, a lead concentration of 25 mg/L, a temperature of 25 °C, and an exposure time of 120 min. The results of this study demonstrate that Haematococcus pluvialis has the potential for effective lead removal from aquatic environments. While the influence of heavy metals on certain algae species has been explored, research on the impact of lead on Haematococcus pluvialis —a microalga of significant interest for astaxanthin production—remains uncharted territory. Therefore, understanding the impact of this heavy metal and the alga's metal absorption capabilities has profound implications for biotechnology and bioremediation applications. This study promotes H. pluvialis as an economically viable lead absorbent suitable for both industrial and domestic purposes.

Effect of arsenic accumulation on growth and antioxidant defense system of Chlorella thermophila SM01 and Leptolyngbya sp. XZMQ

Abstract:Arsenic pollution is an environmental problem of concern, and algae have shown great potential for its application in bioremediation of arsenic pollution. The growth and antioxidative responses of Chlorella thermophila SM01 (C. thermophila SM01) and Leptolyngbya sp. XZMQ newly isolated from Shimen mine area were investigated in the presence of 0.5–10 mg As(III) L−1. After 3 d of exposure, the growth of C. thermophila SM01 and Leptolyngbya sp. XZMQ was inhibited in 0.5–10 mg As(III) L−1. The malondialdehyde (MDA) contents in the both species algal cell were elevated. The antioxidant enzyme activities (SOD, CAT) and the antioxidant content (GSH) in the cells were enhanced. However, with the extension of exposure time, they showed a downward trend. On the 15th day, they decreased to the lowest but still higher than the control. For C. thermophila SM01, 94.37% conversion of As(III) transformed to As(V) was obtained for initial As(III) concentration of 10 mg L−1 within 2 days. C. thermophila SM01 and Leptolyngbya sp. XZMQ could accumulate 1586.65, 2429.90 μg As g−1 dry weight in 10 mg As(III) L−1 solution, respectively. The shift in FTIR spectra indicated amide, carboxyl, and hydroxyl functional groups were involved in the adsorption of arsenic by algae. Principal component analysis (PCA) and Pearson's correlation indicated that antioxidant system and accumulation were negatively related to OD, biomass, Chl a for C. thermophila SM01. However, for Leptolyngbya sp. XZMQ, it was no correlation between MDA and indicators of OD, biomass, Chl a after 15 days. These results showed that C. thermophila SM01 and Leptolyngbya sp. XZMQ have different As(III) toxicity responses and bioaccumulation ability. The study provided a reference for exploring the role of algae in arsenic geochemical cycle in Shimen mining area. It is significant for further understanding the mechanism of algal bioremediation of arsenic polluted water environments.

Abstract:Elevated levels of pollutants, including heavy metals, resulting from human activities, have raised concerns regarding their adverse impacts on natural ecosystems. Among these, aquatic organisms, particularly non-target microalgae, bear substantial consequences from such contaminants. This study endeavored to evaluate the impact of the heavy metal lead (Pb) at concentrations of 2.5, 25, and 250 μg L−1 on the growth and lipid content of the diatom Cyclotella meneghiniana in a 14-day toxicity test conducted under controlled laboratory conditions. The findings indicate that lower Pb concentrations (2.5 and 25 μg L−1 ) fostered algal proliferation, whereas exposure to 250 μg L−1 resulted in complete inhibition in two distinct endpoints. Additionally, no significant fluctuations in algal lipid content were discernible between exposure to 2.5 and 25 μg Pb L−1 and the control group. This study stands as a pioneering effort in unveiling the toxicity of Pb on C. meneghiniana, shedding light on the species’ susceptibility to this heavy metal and underscoring the necessity of incorporating this species into ongoing ecological risk assessments of Pb.

Lead Induced Changes in the Growth and Antioxidant Metabolism of the Lead Accumulating and Non-accumulating Ecotypes of Sedum Alfredii

Abstract:The phytotoxicity and antioxidative adaptations of lead (Pb) accumulating ecotype (AE) and non-accumulating ecotype (NAE) of Sedum alfredii Hance were investigated under different Pb treatments involving 0, 0.02 mmol/L Pb, 0.1 mmol/L Pb and 0.1 mmol/L Pb/0.1 mmol/L ethylenediaminetetraacetic acid (EDTA) for 6 days. With the increasing Pb level, the Pb concentration in the shoots of AE plants enhanced accordingly, and EDTA supply helped 51% of Pb translocation to shoots of AE compared with those treated with 0.1 mmol/L Pb alone. Moreover, the presence of EDTA alleviated Pb phytotoxicity through changes in plant biomass, root morphology and chlorophyll contents. Lead toxicity induced hydrogen peroxide (H2O2) accumulation and lipid peroxidation in both ecotypes of S. alfredii. The activities of superoxide dismutase (SOD), guaiacol peroxidase (G-POD), ascorbate peroxidase, and dehydroascorbate reductase elevated in both leaves and roots of AE as well as in leaves of NAE with the increasing Pb levels, but SOD and G-POD declined in roots of NAE. Enhancement in glutathione reductase activity was only detected in roots of NAE while a depression in catalase activity was recorded in the leaves of NAE. A significant enhancement in glutathione and ascorbic acid (AsA)levels occurred in both ecotypes exposed to Pb and Pb/EDTA treatment compared with the control, however, the differences between these two treatments were insignificant. The dehydroascorbate (DHA) contents in roots of both ecotypes were 1.41 to 11.22-fold higher than those in leaves, whereas the ratios of AsA to DHA (1.38 to 6.84) in leaves altering more to the reduced AsA form were much higher than those in roots. These results suggested that antioxidative enzymes and antioxidants play an important role in counteracting Pb stress in S. alfredii.

Abstract:We apply a Langevin model by imposing additive and multiplicative noises to study thermally activated diffusion over a fluctuating barrier in underdamped dynamics. The barrier fluctuation is characterized by Gaussian colored noise with exponential correlation. We present the exact solutions for the first and second moments. Furthermore, we use direct simulations to calculate the asymptotic probability for a Brownian particle passing over the fluctuating barrier. The results indicate that the correlation of the fluctuating barrier is crucial for barrier crossing dynamics.

Abstract:Lead (Pb) is a non-essential and toxic metal for plants. Current study was planned to assess Pb-induced toxicity with and without NaCl in a halophyte Arthrocaulon macrostachyum . Three month old seedlings (after germination) were grown in pots containing quartz sand and sub-irrigated with 300 mM NaCl and different Pb(NO 3 ) 2 concentrations (0, 0.75, 3, 6 and 12 mM) in plastic trays where 0 mM NaCl and Pb(NO 3 ) 2 were taken as control for monitoring growth, physiological and biochemical characteristics. Plant growth (height and biomass) was optimized in 300 mM NaCl while progressive increases in fresh weights up to 3 mM Pb(NO 3 ) 2 may be related to better shoot turgor, succulence and more plant pigments. Growth was substantially reduced in 6 and 12 mM Pb(NO 3 ) 2 both with and without NaCl. Plant shoots accumulated higher levels of Pb and Na + of up to 3 mM Pb;while root Pb increased in > 6 mM Pb(NO 3 ) 2 treatments. Decreased pigments, potential quantum yield (F v /F m ) and yield of photosystem II (ɸ PSII) enhanced the accumulation of H 2 O 2 , electrolyte leakage and consequently reactive oxygen species (ROS) in 12 mM Pb(NO 3 ) 2 . The scavenging of ROS was manifested with the increased in vitro activity of superoxide dismutase (SOD) of up to 6 mM Pb(NO 3 ) 2 ; whereas higher Guaiacol peroxidase (GPx) may be related to cell wall hardening and poor growth. The increases in SOD, marginal changes in catalase (CAT) and unchanged APX of up to 6 mM Pb(NO 3 ) 2 demonstrated the activation of stress-responsive metabolism in Arthrocaulon machrostachyum ; and Pb accumulation of up to 200 mg kg -1 making this species a useful candidate for Pb phytoremediation.

Abstract:Cadmium is one of the most common heavy metals in contaminated aquatic environments and one of the most toxic contaminants for phytoplankton. Nevertheless, there are not enough studies focused on the effect of this metal in algae. Through a proteomic approach, this work shows how Cd can alter the growth, cell morphology and metabolism of the microalga Chlorella sorokiniana. Using the sequential window acquisition of all theoretical fragment ion spectra mass spectrometry (SWATH-MS), we concluded that exposure of Chlorella sorokiniana to 250 μM Cd2+ for 40 h caused downregulation of different metabolic pathways, such as photosynthesis, oxidative phosphorylation, glycolysis, TCA cycle and ribosomal proteins biosynthesis. However, photorespiration, antioxidant enzymes, gluconeogenesis, starch catabolism, and biosynthesis of glutamate, cysteine, glycine and serine were upregulated, under the same conditions. Finally, exposure to Cd also led to changes in the metabolism of carotenoids and lipids. In addition, the high tolerance of Chlorella sorokiniana to Cd points to this microalga as a potential microorganism to be used in bioremediation processes.
The role of amino acids in facilitating lead accumulation in microalgae: A quantitative analysis of functional group effects

Abstract:In aquatic ecosystems, dissolved organic matter (DOM) enhances the adsorption of heavy metals onto microalgae by participating in the formation of algae–heavy metal–DOM ternary complexes, thereby increasing environmental risks. The functional groups in the DOM play a crucial role. However, a quantitative description based on these groups is lacking. This study investigates the effects of amino acids (AAs) with varying functional group ratios on the accumulation of lead (Pb) in Chlorella pyrenoidosa ( C. pyrenoidosa ) using batch experiments. Isotherm model fitting, and Fourier transform infrared spectroscopy (FTIR) are employed for characterisation. Results indicate that the addition of AAs facilitated Pb enrichment in microalgae. In particular, incorporating lysine (Lys) led to maximum accumulation, reaching 1.45 mmol g −1 . The adsorption rate decreased after AA addition, particularly with Lys, dropping form 0.0032 g mg −1 min −1 to 0.0009 g mg −1 min −1 . The reaction sequence showed carboxyl groups taking precedence over amino groups. The model developed from the data on Pb adsorption by C. pyrenoidosa following AA addition, demonstrated reliability with an R 2 of 0.91 and an F -value of 157.6. Notably this study employed two-dimensional correlation spectroscopy (2D–COS), offering novel insights into the key roles of functional groups and their distinct contributions. These valuable insights contribute to intensive research on the ternary complex of algae − heavy metal − DOM and elucidating their ecological risks.

Increased Steady-State Mutant Huntingtin mRNA in Huntington's Disease Brain.

Abstract:BACKGROUND Huntington's disease is caused by expansion of CAG trinucleotide repeats in the first exon of the huntingtin gene, which is essential for both development and neurogenesis. Huntington's disease is autosomal dominant. The normal allele contains 6 to 35 CAG triplets (average, 18) and the mutant, disease-causing allele contains >36 CAG triplets (average, 42). OBJECTIVE We examined 279 postmortem brain samples, including 148 HD and 131 non-HD controls. A total of 108 samples from 87 HD patients that are heterozygous at SNP rs362307, with a normal allele (18 to 27 CAG repeats) and a mutant allele (39 to 73 CAG repeats) were used to measure relative abundance of mutant and wild-type huntingtin mRNA. METHODS We used allele-specific, quantitative RT-PCR based on SNP heterozygosity to estimate the relative amount of mutant versus normal huntingtin mRNA in postmortem brain samples from patients with Huntington's disease. RESULTS In the cortex and striatum, the amount of mRNA from the mutant allele exceeds that from the normal allele in 75% of patients. In the cerebellum, no significant difference between the two alleles was evident. Brain tissues from non-HD controls show no significant difference between two alleles of huntingtin mRNAs. Allelic differences were more pronounced at early neuropathological grades (grades 1 and 2) than at late grades (grades 3 and 4). CONCLUSION More mutant HTT than normal could arise from increased transcription of mutant HTT allele, or decreased clearance of mutant HTT mRNA, or both. An implication is that equimolar silencing of both alleles would increase the mutant HTT to normal HTT ratio.

Abstract:Background, aim and scope Lead (Pb) accumulation in soils affects plants primarily through their root systems. The aim of this study was to investigate early symptoms of the loss of membrane integrity and lipid peroxidation in root tissues and physiological adaptation mechanism to Pb in accumulating ecotypes (AE) and non-accumulating ecotypes (NAE) of Sedum alfredii under Pb stress in hydroponics. Methods and results Histochemical in situ analyses, fluorescence imaging, and normal physiological analysis were used in this study. Pb accumulation in roots of both AE and NAE increased linearly with increasing Pb levels (0–200 μM), and a significant difference between both ecotypes was noted. Both loss of plasma membrane integrity and lipid peroxidation in root tissues became serious with increasing Pb levels, maximum tolerable Pb level was 25 and 100 μM for NAE and AE, respectively. Pb supplied at a toxic level caused a burst of reactive oxygen species (ROS) in root cells in both ecotypes. However, the root cells of AE had inherently higher activities of superoxide dismutase (SOD), guaiacol peroxidase (POD), and lipoxygenase (LOX) in control plants, and the induction response of these antioxidant enzymes occurred at lower Pb level in AE than NAE. AE plants maintained higher ascorbic acid and H 2 O 2 concentrations in root cells than NAE when exposed to different Pb levels, and Pb induced more increase in dehydroascorbate (DHA), catalase (CAT), and ascorbate peroxidase (APX) in AE than NAE roots. Discussion and conclusion Results indicate that histochemical in situ analyses of root cell death and lipid peroxidation under Pb short-term stress was sensitive, reliable, and fast. Higher tolerance in roots of accumulating ecotype under Pb stress did depend on effective free oxygen scavenging by making complex function of both constitutively higher activities and sensitive induction of key antioxidant enzymes in root cells of S. alfredii .

Abstract:Hydroponics experiments were conducted to study the effects of Pb on mined ecotype (ME) and non-mined ecotype (NME) of Elsholtzia argyi from Pb/Cu mining and the non-contaminated agricultural areas, respectively. The results showed that at 200μM Pb treatment, although concentrations of Pb in leaves and stem of the ME were 2.6 and 4.5 times higher than those of the NME, these plants exhibited higher tolerance to excessive levels of Pb in the growth medium. In both the ecotypes, Pb caused inhibition of leaf growth and photosynthesis, and induced the membrane damage which was more obvious in the NME. Pb treatment decreased the activities of superoxide dismutase (SOD) and guaiacol peroxidase (G-POD) while activity of catalase (CAT) and levels of total soluble proteins (TSP), ascorbic acid (AsA) and glutathione (GSH) were significantly induced after Pb exposure, however, increase was sharp in the ME plants. Leaf ultrastructural analysis of the spongy mesophyll cells revealed that the excessive Pb concentrations caused adverse effects on chloroplast ultrastructure of both ecotypes whereby internal damage was more severe in NME. The higher tolerance to Pb displayed by ME is mainly attributed to maintenance of its leaf growth and physiology, induction of GSH and integrity of cell organelles especially chloroplast ultrastructure.

Abstract:They not only directly impact aquatic organisms, but also indirectly impact these organisms by interacting with background toxins in the environment. Moreover, under realistic environmental conditions, algae, a natural food for aquatic organisms, may alter the toxicity pattern related to MPs. In this research, we first examined the toxicity of MPs alone, and their effect on the toxicity of lead (Pb) on Ceriodaphnia dubia (C. dubia), a model aquatic organism for toxicity survey. Then, we investigated the effect of algae on the combined toxicity of MPs and Pb. We observed that, MPs significantly increased Pb toxicity, which was related to the increase in soluble Pb concentration and the intake of Pb-loaded MPs, both of which increased the accumulation of Pb in C. dubia. The presence of algae mitigated the combined toxicity of MPs and Pb, although algae alone increased Pb accumulation. Therefore, the toxicity mitigation through algae uptake came from mechanisms other than Pb accumulation, which will need further investigation.
