Abstract:Background: Propolis, produced by honey bees, is used around the world, displaying several corroborated biological activities. Brazil is one of the leading producers of propolis, with a great diversity of types, each with a characteristically chemical fingerprint influenced by the flora of the local region. The secondary metabolite's composition of propolis strongly impacts its biological properties, and its chemical characterization is of great importance for its quality control. Several chromatographic techniques have been applied to characterize propolis, highlighting the extraction of its volatiles and its analysis through gas chromatography. Fourteen Brazilian propolis samples collected in four states, including brown, green and red propolis types, were chemically characterized using the automated direct thermal desorption-gas chromatography-mass spectrometry (DTD-GC-MS). Results: Red propolis type was characterized by acyclic saturated hydrocarbons, fatty alcohols, terpenes, and phenylpropanoids as nonacosane, α-copaene, β-amyrin acetate, anethole, and 7-O-methylvestitol. Brown propolis presented hydrocarbons, monoterpenes, and sesquiterpenes, as α-pinene and α-bisabolol. Brazilian green propolis presented polycyclic aromatic hydrocarbons and sesquiterpenes, including 1-methyl-octahydroanthracene, 2,5-dimethyl-γ-oxo-benzenebutanoic acid, nerolidol, and spathulenol. Principal component analysis (PCA) was performed, allowing for clustering brown and red propolis types, indicating a divergence with the chemical composition of the green propolis samples. The hierarchical cluster analysis (HCA) allowed the chemical fingerprint of each propolis type to be differentiated. Conclusion: Red propolis was characterized by sesquiterpenes, pterocarpans, and isoflavans; brown propolis was characterized by hydrocarbons, aldehydes, and monoterpenes, while green propolis samples were characterized by the presence of polycyclic aromatic hydrocarbons, sesquiterpenes, and naphthalene derivatives. © 2022 Society of Chemical Industry.

Abstract:In this paper, we report the characterization of Brazilian red propolis from two origins. The commercial value of this product is due to its composition and bioactivity, which depend on geographical factors. Total and individual phenolic, antioxidant, and medium infrared spectroscopic (MIR) characterization of 72 red propolis samples from two origins (Bahia (BA) and Rio Grande do Norte (RN), Brazil) were performed. Samples from RN showed higher concentrations of biomarkers. The BA samples showed higher antioxidant capacity despite having lower concentrations of the red propolis biomarker molecules when compared to the RN samples. The MIRS showed differences regarding the origin. The principal component analysis confirmed the differences, generating well-defined groups. Linear discriminant analysis or discriminant analysis and and partial least squares discriminant analysis showed excellent model performance. The models with MIRS data from raw propolis have a high correct classification rate and require no sample preparation. These findings can affect red propolis production, boosting sustainable extractivism, and changing the reality of small communities.

Abstract:The problem of practical tracking control is studied for high-order nonlinear uncertain systems, which has been investigated under the assumption that the unmeasurable states in the nonlinear vector field can only grow linearly and the nonlinear systems are low-order. This paper studies the problem that the systems are high-order which have the higher-order growth of unmeasurable states. By means of designing a proper controller and utilizing the homogeneous domination approach, we achieve the objective that the static tracking error arbitrarily small and the systems have nice properties such as robustness to disturbances and universal property to reverence signals. Further, the results are extended to several proper conditions.
A validated ultra-performance liquid chromatography with tandem mass spectrometry method for the quantification of Brazilian green propolis main compounds

Abstract:Brazilian green propolis is used in folk medicine because of its various biological properties. The hydroalcoholic extract of Brazilian green propolis is characteristic for possessing several pharmacological properties. Phytochemical investigations have attributed some of these properties to the presence of compounds, which were chosen as analytical markers. This paper reports the development and analytical validation using UPLC-MS/MS in MRM mode. Veratraldehyde was used as an internal standard in qualitative and quantitative analyses of the extracts. Relative standard deviation values obtained for intra-day and inter-day precision were lower than 4%. Of the five parameters for robustness, wavelength detection and flow rate were the critical ones. Limits of detection and quantification ranged from 0.300 to 39.500 ng.mL −1 and from 1.400 to 85.00 ng.mL −1 , respectively. The recoveries were between 94.00 and 119.00%, with relative standard deviation values around 5.0%. The developed method is precise, sensitive, and reliable for analysing Brazilian green propolis.

Abstract:Specialty green coffee beans have a higher commercial value and some of them have recently been classified in Brazil based on the indication of provenance and denomination of origin. In this context, the classification of the type of coffee bean is still a challenge, using traditional analytical techniques. Thus, alternative analytical techniques, such as ultraviolet–visible spectroscopy (UV–Vis), as non-target analysis can be applied as a quick and reliable method of coffee classification using data science, such as chemometric tools. In the present study, UV–Vis were evaluated as a new strategy for discrimination of green beans of Brazilian specialty canephora coffees with recognized geographical indications (Robusta Amazônico and Conilon from state of Espírito Santo), for the first time. Spectra obtained from the aqueous extract of 222 samples. The Principal Component Analysis (PCA) was performed and subsequently Partial Least Squares with Discriminant Analysis (PLS-DA) model developed. The PCA indicated tendency to group the samples in their respective classes, pointing to the similarities in the spectra of samples of the same origin. The PLS-DA model obtained showed figures of merit values starting at 89.3% in the test set. The VIP scores showed that the variables associated with chlorogenic acids, caffeine and chlorophyll are the most important for differentiating the studied coffees. The results obtained showed that UV–Vis fingerprint − non-targeted analysis associated with PLS-DA is appropriate for the discrimination of green beans of Brazilian specialty coffee from different origins, in a simple way, using common equipment in several laboratories.

Abstract:This study aimed to investigate the content of macro and trace elements in Apis mellifera’s honey in five municipalities in two regions of Pará in the Brazilian Amazon. This state has the largest beekeeping production chain in the Amazon, an essential source of income for family farming. We used the inductively coupled plasma optical emission spectrometry technique (ICP OES) to determine the concentration of metals (Al, Ca, Cu, Fe, Li, K, Mg, Mn, Na and Zn) in the samples. The other studied metals were detected in all five cities, except copper, found only in three locations. Three groups of elements were identified by factor analysis. The first two may indicate anthropic and geographic influences, and the third is related to the higher sodium content in mangrove honey from this ecosystem due to the influence of the maritime environment. The statistical techniques and Machine Learning proved very efficient in discriminating the origin of the honey samples. They can be used to determine geographic origin and authenticity to give subsidies to the quality control of these products, allowing the expansion and inclusion of beekeeping in the region's business portfolio.

Abstract:The variations in the chemical composition, and consequently, on the biological activity of the propolis, are associated with its type and geographic origin. Considering this fact, this study evaluated propolis extracts obtained by supercritical extraction (SCO2) and ethanolic extraction (EtOH), in eight samples of different types of propolis (red, green and brown), collected from different regions in Brazil. The content of phenolic compounds, flavonoids, in vitro antioxidant activity (DPPH and ABTS), Artepillin C, p-coumaric acid and antimicrobial activity against two bacteria were determined for all extracts. For the EtOH extracts, the anti-proliferative activity regarding the cell lines of B16F10, were also evaluated. Amongst the samples evaluated, the red propolis from the Brazilian Northeast (states of Sergipe and Alagoas) showed the higher biological potential, as well as the larger content of antioxidant compounds. The best results were shown for the extracts obtained through the conventional extraction method (EtOH). However, the highest concentrations of Artepillin C and p-coumaric acid were identified in the extracts from SCO2, indicating a higher selectivity for the extraction of these compounds. It was verified that the composition and biological activity of the Brazilian propolis vary significantly, depending on the type of sample and geographical area of collection.

Programmed cell death 4 (Pdcd4) expression in colorectal adenocarcinoma: Association with clinical stage.

Abstract:The aim of this study was to examine the role of Programmed cell death 4 (Pdcd4) in colorectal adenocarcinoma (CRA). Pdcd4 expression was observed in both the nucleus and cytoplasm in colorectal adenocarcinoma, whereas Pdcd4 was expressed in the nucleus in normal colonic epithelial cells. Loss or weak expression of Pdcd4 was identified in 44 cases (40.7%) of cancer cells. Pdcd4 expression was associated with an increase in the nodal and clinical stage (p=0.022 and p=0.016, respectively). Nuclear staining was identified in 66 cases (61.15%), with no correlation with clinicopathological factors. Conversely, cytoplasmic staining for Pdcd4 was observed in 45 cases (41.7%), and increased according to nodal and clinical stage (p=0.011 and p=0.009, respectively), indicating that aberrant Pdcd4 expression leads to tumor progression. However, Pdcd4 expression was not correlated to disease-free survival time. This study demonstrated that during the tumorigenesis of CRA, loss of nuclear Pdcd4 expression occurs, and during tumor progression, aberrant cytoplasmic expression is present, suggesting a higher clinical stage. Although loss of Pdcd4 was not significantly correlated with survival time, as the prognosis of colorectal cancer varies depending on clinical stage including invasion depth, nodal status and metastatic status, cytoplasmic Pdcd4 expression may be a favorable prognostic marker in CRA.

Abstract:Caparaó and the Fluminense northwest regions are nationally recognized by the important contribution on coffee production and exportation. Adulterations involving specialty coffees result in a decrease in the quality of the final product. However, obtaining many different samples from the same region is unfeasible in some cases, needing strategies to work with a limited number of samples for pattern recognition. Thus, this work is the first to use the construction of synthetic samples (SS) for analysis of coffees, and its objective is to identify adulterations in specialty coffees with bark, straw and low-quality beans, using UV-Vis spectroscopy, associated with chemometric methods. The synthetic samples partial least square discriminant analysis (SS-PLS-DA) showed better specificity, sensitivity and reliability rates than the Hard PLS-DA models. One-class methods (soft independent modeling of class analogy (SIMCA) and data driven soft independent modeling of class analogy (DD-SIMCA)) showed low specificity and reliability. The discriminant methods together with the synthetic samples proved to be adequate to identify adulterations in specialty coffees.

Abstract:It is generally accepted that taxa exhibit genetic variation in phenotypic plasticity, but many questions remain unan- swered about how divergent plastic responses evolve under dissimilar ecological conditions. Hormones are signaling molecules that act as proximate mediators of phenotype expression by regulating a variety of cellular, physiological, and behavioral re- sponses. Hormones not only change cellular and physiological states but also influence gene expression directly or indirectly, thereby linking environmental conditions to phenotypic development. Studying how hormonal pathways respond to environ- mental variation and how those responses differ between individuals, populations, and species can expand our understanding of the evolution of phenotypic plasticity. Here, we explore the ways that the study of hormone signaling is providing new insights into the underlying proximate bases for individual, population or species variation in plasticity. Using several studies as exem- plars, we examine how a 'norm of reaction' approach can be used in investigations of hormone-mediated plasticity to inform the following: 1) how environmental cues affect the component hormones, receptors and enzymes that comprise any endocrine sig- naling pathway, 2) how genetic and epigenetic variation in endocrine-associated genes can generate variation in plasticity among these diverse components, and 3) how phenotypes mediated by the same hormone can be coupled and decoupled via independent plastic responses of signaling components across target tissues. Future studies that apply approaches such as reaction norms and network modeling to questions concerning how hormones link environmental stimuli to ecologically-relevant phenotypic re- sponses should help unravel how phenotypic plasticity evolves (Current Zoology 59 (4): 506-525, 2013). Abstract:Propolis is a natural product with many demonstrated biological activities and propolis extract has been used in the food, pharmaceutical and cosmetics industries. Different works have showed the variations in the chemical composition, and consequently, on the biological activity of the propolis that are associated with its type and geographic origin. Due to this study evaluated propolis extracts obtained through supercritical extraction and ethanolic extraction (conventional) in three samples of different types of propolis (red, green and brown), collected from different regions in Brazil (state of Bahia). Analyses were performed to determine the humidity, water activity, the content of total ash, proteins, lipids and fiber in raw propolis samples. The content of phenolic compounds, flavonoids, in vitro antioxidant activity (DPPH), catechin, ferulic acid and luteolin and antimicrobial activity against two bacteria (Staphylococcus aureus and Escherichia coli) were determined for all extracts. For the green and red ethanolic extracts the anti-leishmanicidal potential was also evaluated. The physicochemical profiles showed agreement in relation to the literature. The results identified significant differences among the extracts (p>0.05), which are in conformity with their extraction method, as well as with type and botanical origin of the samples. The extraction with supercritical fluid was not efficient to obtain extracts with the highest contents of antioxidants compounds, when compared with the ethanolic extracts. The best results were shown for the extracts obtained through the conventional extraction method (ethanolic) indicating a higher selectivity for the extraction of antioxidants compounds. The red variety showed the largest biological potential, which included the content of antioxidants compounds. The results found in this study confirm the influence of the type of the raw material on the composition and characteristics of the extracts. The parameters analysis were important to characterize and evaluate the quality of the different Brazilian propolis extracts based on the increased use of propolis by the natural products industry.

Abstract:Propolis is a natural resinous product produced by Apis mellifera bees from the exudates of various plants. The color of propolis (green) is a consequence of its botanical origin, as bees collect young tissues and leaves of Baccaris dracunculifolia. This study evaluated the chemical composition and extraction kinetics of essential oils obtained from Brazilian green propolis by hydrodistillation. Hydrodistillation was performed for 360 min and analyzed at different times (30, 60, 120, 240, and 360 min), allowing the calculation of the accumulated content (% w/w) and the identification of the essential oil chemical profile. The GC/FID and GC/MS analysis led to the annotation of 60 compounds with estragole (13.30%), benzyl propanoate (14.59%), and (E)‐nerolidol (13.57%) as the main compounds. The optimum conditions for extraction of phenylpropanoids (PP), hydrocarbons (HD), monoterpenes (MT), and oxygenated monoterpenes (OMT) are between 30 and 120 min. In comparison, sesquiterpenes (ST) and oxygenated sesquiterpenes (OST) are extracted more efficiently between 240 and 360 min. The optimal extraction speed determination is essential for industrial‐scale processing to obtain components such as sesquiterpenes, which have a high economic value in the cosmetic/perfumery and pharmaceutical industries.

Abstract:This study aimed at the geographical origin discrimination of Jatropha mollissima saps using mid- and near-infrared spectroscopies (MIR and NIR, respectively) and Linear Discriminant Analysis (LDA). For this purpose, a total of 108 sap samples were collected in 3 different geographical regions over 12 months, and their content of total polyphenols, flavonoids, and tannins were quantified. Overall, samples from region C had consistently lower levels of these secondary metabolites throughout most of the months studied. In contrast, samples from regions A and B displayed relatively stable metabolite concentrations over the collection period. Since raw sap samples are subject to deterioration during storage, lyophilization was employed to remove moisture and consequently increase their shelf-life. Thus, MIR spectroscopy was applied to both raw and lyophilized sap samples, while NIR spectroscopy was only applied to lyophilized samples. Then, Principal Component Analysis from the secondary metabolites and spectral data indicated a trend of separation between the region C (located in Sertão ) and the regions A and B (located in Agreste ). Next, the Successive Projection Algorithm (SPA), Genetic Algorithm (GA), and Ant Colony Optimization (ACO) were used for variable selection in LDA. As a result, all constructed models achieved a sensitivity, specificity, and accuracy of 100 %, or very close to it, in both the training and test sets. However, the SPA-LDA models were more parsimonious, selecting fewer variables and presenting reproductive results as it is a deterministic technique. Hence, the proposed analytical methodologies align with the principles of Green Chemistry by requiring a simple sample preparation, avoiding the use of reagents and solvents, and reducing waste generation. Moreover, special attention can be given to NIR spectroscopy, as it offers a cost-effective analytical tool that can be explored in situ using a portable miniaturized device in the future.

Abstract:Studies were performed to examine whether, in addition to T cells, there might be other immune cells also capable of exerting a graft-versus-leukemia (GVL) response following allogeneic marrow transplant. Using an MHC-matched mouse model, consisting of normal B10.S donors and SJL/J Rauscher-retroviral-leukemic recipients, the donor cells were selectively depleted of their Asialo-GM1+ component prior to being infused into the leukemic recipients. The incidence of relapse was then compared against that for matched leukemic control recipients of undepleted cells from the same donors. FCM analysis of the depletion protocol indicated that exposure to anti-Asialo-GM1 antibody eliminated more than half of the donor NK1.1+ cells, but caused no significant losses among the Thy-1+, CD3+, or CD8+ cells. Nevertheless, fatal relapse among the leukemic recipients of the depleted cells was nearly double that found among the leukemic control recipients of undepleted cells, 47.5 vs 25.4% (P = 0.01). In a parallel study, using normal SJL/J recipients, this same depletion protocol was found to have no significant effect on the incidence of graft-versus-host disease (GVHD). These results therefore suggest that Asialo-GM1+ NK cells may be capable of contributing to the suppression of relapse in this type of leukemic recipient of allogeneic marrow, and that this suppression may occur independently of GVHD.

Abstract:Propolis is a resinous bee product with a very complex composition, which is dependent upon the plant sources that bees visit. Due to the promising antimicrobial activities of red Brazilian propolis, it is paramount to identify the compounds responsible for it, which, in most of the cases, are not commercially available. The aim of this study was to develop a quick and clean preparative-scale methodology for preparing fractions of red propolis directly from a complex crude ethanol extract by combining the extractive capacity of counter-current chromatography (CCC) with preparative HPLC. The CCC method development included step gradient elution for the removal of waxes (which can bind to and block HPLC columns), sample injection in a single solvent to improve stationary phase stability, and a change in the mobile phase flow pattern, resulting in the loading of 2.5 g of the Brazilian red propolis crude extract on a 912.5 mL Midi CCC column. Three compounds were subsequently isolated from the concentrated fractions by preparative HPLC and identified by NMR and high-resolution MS: red pigment, retusapurpurin A; the isoflavan 3(R)-7-O-methylvestitol; and the prenylated benzophenone isomers xanthochymol/isoxanthochymol. These compounds are markers of red propolis that contribute to its therapeutic properties, and the amount isolated allows for further biological activities testing and for their use as chromatographic standards.

Abstract:Propolis is a resinous natural product produced by honeybees using beeswax and plant exudates. The chemical composition of propolis is highly complex, and varies with region and season. This inherent chemical variability presents several challenges to its standardisation and quality control. The present review was aimed at highlighting marker compounds for different types of propolis, produced by the species Apis mellifera , from different geographical origins and that display different biological activities, and to discuss strategies for quality control. Over 800 compounds have been reported in the different propolises such as temperate, tropical, birch, Mediterranean, and Pacific propolis; these mainly include alcohols, acids and their esters, benzofuranes, benzopyranes, chalcones, flavonoids and their esters, glycosides (flavonoid and diterpene), glycerol and its esters, lignans, phenylpropanoids, steroids, terpenes and terpenoids. Among these, flavonoids (> 140), terpenes and terpenoids (> 160) were major components. A broad range of biological activities, such as anti-oxidant, antimicrobial, anti-inflammatory, immunomodulatory, and anticancer activities, have been ascribed to propolis constituents, as well as the potential of these compounds to be biomarkers. Several analytical techniques, including non-separation and separation methods have been described in the literature for the quality control assessment of propolis. Mass spectrometry coupled with separation methods, followed by chemometric analysis of the data, was found to be a valuable tool for the profiling and classification of propolis samples, including (bio)marker identification. Due to the rampant chemotypic variability, a multiple-marker assessment strategy considering geographical and biological activity marker(s) with chemometric analysis may be a promising approach for propolis quality assessment.

Abstract:The aim of this study was to evaluate the effect of seasons on the chemical composition and antibacterial activity of Brazilian red propolis (BRP) and its plant source. BRP was collected from Maceio, Alagoas state, north-east of Brazil, during one year. Chemical composition was determined by physicochemical analyses and HPLC while antimicrobial activity was assessed against Streptococcus mutans, Streptococcus sobrinus, Staphylococcus aureus and Actinomyces naeslundii by determining the minimal inhibitory and bactericidal concentrations (MIC and MBC, respectively). The comparative chemical profiles varied quantitatively according to the collection period. Formononetin was the most abundant compound in both propolis and resin, while isoliquiritigenin, (3S)-neovestitol, (3S)-vestitol are suggested to be responsible for antimicrobial activity of Brazilian red propolis. MIC varied from 15.6 to 125 μg/mL, whereas MBC varied from 31.2 to 500 μg/mL. Therefore, season in which propolis and its botanical source are collected indeed influences their chemical compositions, resulting in variations in their antibacterial activity.

Abstract:Brazilian green propolis is known as an appreciable natural antioxidant with abundant polyphenolic compounds. For quality control, a fingerprint-efficacy study of Brazilian green propolis was carried out in this work. Chemical fingerprints of Brazilian green propolis from 22 different sources were determined by HPLC and investigated by similarity analysis. The fingerprint-efficacy relationships between chemical fingerprint and DPPH radical-scavenging activity were established. The results showed that 14 characteristic common peaks were identified, and 9 compounds were discovered with free radical-scavenging activities. Caffeoylquinic acids and artepillin C might be the major effective components for quality control of Brazilian green propolis due to their specificity and strong antioxidant activity. This study provides new markers for the quality assessment of Brazilian green propolis and its derived products.

Abstract:A delayed traumatic intracerebral hematoma was seen in a patient after a shotgun wound to the brain. The cause of the hemorrhage seemed to be a traumatic aneurysm. Both complications are reviewed, and it is suggested that clinical and intracranial pressure monitoring should lead to early diagnosis and may minimize the effects of this problem and that angiography may be appropriate early in the course of patients who have had fragments pass near intracranial vessels.
