Abstract:In this study, a calibration model based on Near-infrared spectroscopy (NIR) technique and chemometrics method was developed for rapid and non-destructive detecting the polysaccharide contents of lentinula edodes samples collected from different regions. The polysaccharide contents of these samples were firstly determined by standard phenol-sulphruic acid method. Then, NIR spectra of these samples were collected by using Fourier transform infrared spectrometry. Based on these experimental data, a random forest method was further used to distinguish the regions of these samples, with a classification accuracy of 96.6%. After that, a rapid, accurate, and quantitative model was established for predicting the polysaccharide contents of these samples. In the model establishing process, some signal pre-treatment methods were optimized, and the validation results with highest determination coefficient (R2) and low root mean square errors of prediction (RMSEP) were, 0.925 and 0.720, respectively. These results showed that combined NIR technique with chemometrics was an effective and green method for lentinula edodes quality control.
Rapid Analysis of the In-Process Extract Solutions of Compound E Jiao Oral Liquid Using Near Infrared Spectroscopy and Partial Least-Squares Regression

Abstract:A rapid and simultaneous quantification of the main active compounds of the in-process extract solutions is beneficial to the process monitoring as well as ensuring quality consistency of the end products during the production of traditional Chinese medicine. Here we present a near infrared (NIR) spectroscopy-based method for rapid analysis of extract solutions in the production of compound E Jiao oral liquid. The partial least-squares regression (PLSR) models for four quality indicators (viz. total flavonoids, total saponins, total saccharides and soluble solid contents) were established and validated. The results showed that all of the four models exhibited satisfactory fitting and predictive capacity. The root mean squares error of prediction (RMSEP) was 0.0384 mg mL−1, 0.0154 mg mL−1, 3.80 mg mL−1 and 0.199% for total flavonoids, total saponins, total saccharides and soluble solid contents, respectively. This work here demonstrated that NIR spectroscopy coupled with PLSR calibration can offer a reliable and non-destructive alternative in the routine monitoring of the extraction process in the production of compound E Jiao oral liquid. The presented approach is expected to be equally applicable to the mixed decoction of other herbal medicines.

Abstract:There is a lack of methods to analyze large numbers of traditional Chinese medicine (TCM) samples quickly and efficiently. Here we present a near infrared (NIR) spectroscopy-based method for the rapid analysis of Honeysuckle extracts. NIR spectra of 124 Honeysuckle extract samples produced within a four-year period were collected in the diffuse reflection mode and pretreated with multiplicative scatter correction (MSC), smoothing and derivative. Using partial least squares regression (PLSR), including PLS1 and PLS2, we established and validated the calibration models of six organic acids. Our results showed that both accuracy and precision of the NIR spectroscopy-based method meet the requirements of a traditional Chinese medicine (TCM) factory. This new method has the potential to save time, efforts and cost associated with the analysis of large quantities of unknown samples and will be particularly applicable to TCM factories for cost reduction.

Abstract:The paper presents a novel strategy to identify analytical markers of traditional Chinese medicine preparation (TCMP) rapidly via direct analysis in real time mass spectrometry (DART-MS). A commonly used TCMP, Danshen injection, was employed as a model. The optimal analysis conditions were achieved by measuring the contribution of various experimental parameters to the mass spectra. Salvianolic acids and saccharides were simultaneously determined within a single 1-min DART-MS run. Furthermore, spectra of Danshen injections supplied by five manufacturers were processed with principal component analysis (PCA). Obvious clustering was observed in the PCA score plot, and candidate markers were recognized from the contribution plots of PCA. The suitability of potential markers was then confirmed by contrasting with the results of traditional analysis methods. Using this strategy, fructose, glucose, sucrose, protocatechuic aldehyde and salvianolic acid A were rapidly identified as the markers of Danshen injections. The combination of DART-MS with PCA provides a reliable approach to the identification of analytical markers for quality control of TCMP.

Abstract:The extract of Chinese propolis (ECP) has recently been adulterated with that of poplar buds (EPB), because most of ECP is derived from the poplar plant, and ECP and EPB have almost identical chemical compositions. It is very difficult to differentiate them by using the chromatographic methods such as high performance liquid chromatography (HPLC) and gas chromatography (GC). Therefore, how to effectively discriminate these two mixtures is a problem to be solved urgently. In this paper, a rapid method for discriminating ECP and EPB was established by the Fourier transform infrared (FT-IR) spectra combined with the two-dimensional infrared correlation (2D IR) analysis. Forty-three ECP and five EPB samples collected from different areas of China were analyzed by the FT-IR spectroscopy. All the ECP and EPB samples tested show similar IR spectral profiles. The significant differences between ECP and EPB appear in the region of 3000–2800cm−1 of the spectra. Based on such differences, the two species were successfully classified with the soft independent modeling of class analogy (SIMCA) pattern recognition technique. Furthermore, these differences were well validated by a series of temperature-dependent dynamic FT-IR spectra and the corresponding 2D IR plots. The results indicate that the differences in these two natural products are caused by the amounts of long-chain alkyl compounds (including long-chain alkanes, long-chain alkyl esters and long chain alkyl alcohols) in them, rather than the flavonoid compounds, generally recognized as the bioactive substances of propolis. There are much more long-chain alkyl compounds in ECP than those in EPB, and the carbon atoms of the compounds in ECP remain in an order Z-shaped array, but those in EPB are disorder. It suggests that FT-IR and 2D IR spectroscopy can provide a valuable method for the rapid differentiation of similar natural products, ECP and EPB. The IR spectra could directly reflect the integrated chemical compositions of complicated mixtures, and it may be available for use in further chromatographic analysis.

Abstract:Background: American ginseng (Panax quinquefolius L.) and Asian ginseng (Panax ginseng Meyer) products, such as slices, have a similar appearance, but they have significantly different prices, leading to widespread adulteration in the commercial market. Their aroma characteristics are attracting increasing attention and are supposed to be effective and nondestructive markers to determine adulteration. Methods: The aroma characteristics of American and Asian ginseng were investigated using gas chromatography-mass spectrometry(GC-MS) and an electronic nose (E-nose). Their volatile organic compounds were separated, classified, compared, and analyzed with different pattern recognition. Results: The E-nose showed a good performance in grouping with a principle component analysis explaining 94.45% of variance. A total of 69 aroma components were identified by GC-MS, with 35.6% common components and 64.6% special ingredients between the two ginsengs. It was observed that the components and the number of terpenes and alcohols were markedly different, indicating possible reasons for their difference. The results of pattern recognition confirmed that the E-nose processing result is similar to that of GC-MS. The interrelation between aroma constituents and sensors indicated that special sensors were highly related to some terpenes and alcohols. Accordingly, the contents of selected constituents were accurately predicted by corresponding sensors with most R-2 reaching 90%. Conclusion: Combined with advanced chemometrics, the E-nose is capable of discriminating between American and Asian ginseng in both qualitative and quantitative angles, presenting an accurate, rapid, and nondestructive reference approach. Rapid monitoring approaches for concentration process of lanqin oral solution by near-infrared spectroscopy and chemometric models

Abstract:Qualitative and quantitative detection methods based on near-infrared spectroscopy (NIRs) have been proposed in the process analysis of traditional Chinese medicine in recent years. In this study, rapid monitoring methods were developed for quality control of concentration process of lanqin oral solution (LOS). Partial least squares regression (PLSR) method was adopted to construct quantitative models for epigoitrin, geniposide, baicalin, berberine hydrochloride and density. Simultaneously, the genetic algorithm joint extreme learning machine (GA-ELM) was first applied in qualitative analysis of NIRs to distinguish end point of concentration process. Results of PLSR models were satisfactory with the relative standard error of calibration valued at 3.80%, 3.75%, 3.79%, 11.5% and 1.22% for epigoitrin, geniposide, baicalin, berberine hydrochloride and density respectively, and the residual predictive deviation values were higher than 3. For qualitative analysis, the GA-ELM model obtained 100% prediction accuracy. The PLSR quantitative models and the end point discrimination model constructed by GA-ELM correspond with the requirements of practical applications. Abstract:A novel method for the rapid determination of chrysin and galangin in Chinese propolis of poplar origin by means of visible and near infrared spectroscopy (Vis-NIR) was developed. Spectral data of 114 Chinese propolis samples were acquired in the 325 to 1,075 nm wavelength range using a Vis-NIR spectroradiometer. The reference values of chrysin and galangin of the samples were determined by high performance liquid chromatography (HPLC). Partial least squares (PLS) models were established using the spectra analyzed by different preprocessing methods. The effective wavelengths were selected by successive projections algorithm (SPA) and employed as the inputs of PLS, back propagation-artificial neural networks (BP-ANN), multiple linear regression (MLR) and least square-support vector machine (LS-SVM) models. The best results were achieved by SPA-BP-ANN models established with the Savitzky-Golay smoothing (SG) preprocessed spectra, where the r and RMSEP were 0.9823 and 1.5239 for galangin determination and 0.9668 and 2.4841 for chrysin determination, respectively. The results show that Vis-NIR demosntrates powerful capability for the rapid determination of chrysin and galangin contents in Chinese propolis.

Abstract:Process monitoring is essential for the pharmaceutical industry to control the quality uniformity of final products. In this study, the prompt quantitative methods of geniposide in Lanqin oral solution (LOS) during the process of extraction and concentration were developed by near-infrared spectroscopy (NIRs). Simultaneously, the feasibility of the method for universally monitoring the two processes was investigated. Methods including interval partial least square (iPLS), synergy interval partial least square (siPLS) and competitive adaptive reweighted sampling (CARS) were performed for characteristic variables selection. The single-process models of extraction, concentration and the multi-process model based on the combination of the two processes were constructed and discussed. Results showed that the performance of siPLS models were superior to others. For the extraction, concentration and the combination of these two processes, the Rc values were 0.9486, 0.9746 and 0.9980; while the RSEP values were 10.42 %, 5.41 % and 6.17 %, respectively. Compared with the specific models, the strong accuracy and stability of the multi-process model were also demonstrated. In addition, Pearson correlation analysis and paired t-test were also applied to verify the predictive performance of models. The overall results indicate that NIRs combined with siPLS algorithm is a robust method for predicting the geniposide in the extraction and concentration of LOS. Furthermore, the multi-process model of combination is feasible for the universal supervision of geniposide, which is convenient for industrial production applications.

Abstract:A simple, sensitive and specific high-performance liquid chromatography-UV (HPLC-UV) method has been developed to simultaneously quantify the eight major bioactive phenolic compounds in Chinese propolis, namely caffeic acid, isoferulic acid, 3,4-dimethoxycinnamic acid, pinobanksin 5-methyl ether, pinocembrin, benzyl caffeate, chrysin and galangin. This HPLC assay was performed on an Agilent Zorbax Extend-C18 (250 x 4.6 mm, 5 microm) column with a gradient of methanol and 0.2% aqueous acetic acid (v/v) in 50 min, at a flow rate of 1.0 mL/min, and detected at 290 nm. All calibration curves showed good linearity (r2 > 0.999) within the test ranges. The intra- and inter-day assay precision (RSD) of eight phenolic compounds were in the range of 0.07-4.92%. The recoveries were between 98.3% and 104.8%. This assay was applied to the evaluation of nineteen samples from different origins in China. The results indicated that the developed assay could be readily utilized for the quality control of propolis.
Improvement in ß-cell function in patients with normal and hyperglycemia following Roux-en-Y gastric bypass surgery.

Abstract:Glycemic disorders resolve following Roux-en-Y gastric bypass (RYGB) surgery, but early and longer-term mechanisms regarding effects on β-cell dysfunction as well as relationships with decreasing adiposity are not well understood. We evaluated longitudinal changes in peripheral insulin sensitivity (Si), the acute insulin response to glucose (AIRg), and the composite estimate of β-cell function, the disposition index (DI), over 24 mo via frequently sampled intravenous glucose tolerance testing in severely obese women who had fasting normoglycemia (n = 16) and hyperglycemia (n = 11) before RYGB surgery; homeostatic model assessment (HOMA-IR) estimated insulin resistance; air displacement plethysmography determined adipose tissue mass. At baseline, subjects with normoglycemia had adequate DI associated with elevated AIRg, but DI was markedly reduced in subjects with hyperglycemia. Within 1-6 mo post-RYGB, glycemic control was normalized in subjects with hyperglycemia related to reduced HOMA-IR (-54% at 1 mo, P < 0.005) and increased DI (23-fold at 6 mo vs. baseline, P < 0.05). Over 24 mo, DI improved in subjects with hyperglycemia (15-fold vs. baseline, P < 0.005) and also modestly in subjects with normoglycemia (58%, P < 0.05), due largely to increased Si. Decreasing adiposity correlated with longer-term HOMA-IR and Si values at 6 and 24 mo, respectively. In patients exhibiting fasting hyperglycemia before surgery, β-cell function improved early following RYGB, due largely to increases in insulin secretion. For both normoglycemic and hyperglycemic subjects, further improvement or stabilization of β-cell function over the 2 yr is due largely to improved Si associated with reduced adiposity.
Chemometric Determination of the Botanical Origin for Chinese Honeys on the Basis of Mineral Elements Determined by ICP-MS

Abstract:In this work, the potential of mineral elements and chemometric methods as a tool to classify Chinese honeys according to their botanical origin was examined. Twelve mineral elements (Na(23), Mg(24), P(31), K(39), Ca(43), Mn(55), Fe(56), Cu(63), Zn(66), Rb(85), Sr(88), and Ba(137)) of 163 Chinese honey samples, including linden, vitex, rape, and acacia, collected from Heilongjiang, Beijing, Hebei, and Shaanxi, China, in 2013 were determined by the ICP-MS method. Principal component analysis (PCA) reduced 10 variables to four principal components and could explain 93.06% of the total variance. Partial least-squares discriminant analysis (PLS-DA) and back-propagation artificial neural network (BP-ANN) were explored to construct a classification model. By PLS-DA, the total correct classification rates for model training and cross-validation were 90.9 and 88.4%, respectively. By BP-ANN, the total correct classification rates for model training and cross-validation were 100 and 92.6%, respectively. The performance of BP-ANN was better than that of PLS-DA. The validation of the developed BP-ANN model was tested by the independent test set of 42 honey samples. Linden, vitex, and rape honey samples were predicted with an accuracy of 100%, whereas one acacia honey was predicted as rape honey with an accuracy of 92.3%. It is concluded that the profiles of mineral elements by ICP-MS with chemometric methods could be a potential and powerful tool for the classification of Chinese honey samples from different botanical origins.

Abstract:The rapid and accurate authentication of traditional Chinese medicines (TCMs) has always been a key scientific and technical problem in the field of pharmaceutical analysis. Herein, a novel heating online extraction electrospray ionization mass spectrometry (H-oEESI-MS) was developed for the rapid and direct analysis of extremely complex substances without the requirement for any sample pretreatment or pre-separation steps. The overall molecular profile and fragment structure features of various herbal medicines could be completely captured within 10-15 s, with minimal sample (<0.5 mg) and solvent consumption (<20 μL for one sample). Furthermore, a rapid differentiation and authentication strategy for TCMs based on H-oEESI-MS was proposed, including metabolic profile characterization, characteristic marker screening and identification, and multivariate statistical analysis model validation. In an analysis of 52 batches of seven types of Aconitum medicinal materials, 20 and 21 key compounds were screened out as the characteristic markers of raw and processed Aconitum herbal medicines, respectively, and the possible structures of all the characteristic markers were comprehensively identified based on Compound Discoverer databases. Finally, multivariate statistical analysis showed that all the different types of herbal medicines were well differentiated and identified (R2X > 0.87, R2Y > 0.91, and Q2 > 0.72), which further verified the feasibility and reliability of this comprehensive strategy for the rapid authentication of different TCMs based on H-oEESI-MS. In summary, this rapid authentication strategy realized the ultra-high-throughput, low-cost, and standardized detection of various complex TCMs for the first time, thereby demonstrating wide applicability and value for the development of quality standards for TCMs.

Abstract:PURPOSE To compare the pathologic complete response (pCR) rate and relapse-free survival (RFS) and overall survival (OS) after neoadjuvant systemic chemotherapy (NST) in patients with breast cancer with and without deleterious BRCA1 and BRCA2 mutations. PATIENTS AND METHODS A total of 317 women who underwent BRCA genetic testing and were treated with NST for breast cancer between 1997 and 2009 were included in the study. The Kaplan-Meier product-limit method was used to estimate RFS and OS rates. Logistic regression models were fit to determine the associations between BRCA status, pCR, and survival. RESULTS Fifty-seven (18%) and 23 (7%) patients had BRCA1 and BRCA2 mutations, respectively. Twenty-six (46%) of 57 BRCA1 carriers achieved a pCR, compared with three (13%) of 23 BRCA2 carriers and 53 (22%) of 237 BRCA noncarriers (P < .001). In the multivariate logistic model, BRCA1 status (odds ratio [OR] = 3.16; 95% CI, 1.55 to 6.42; P = .002), estrogen receptor (ER) negativity (OR = 1.96; 95% CI:1.05 to 3.65; P = .03) and concurrent trastuzumab use (OR = 4.18; 95% CI, 2.04 to 8.57; P < .001) remained as independent significant predictors for a pCR. At a median follow-up of 3.2 years, 69 patients (22%) experienced a disease recurrence or death. No significant differences were noted in survival outcomes with respect to BRCA status and type of NST received. However, among BRCA1 carriers, patients who achieved a pCR had better 5-year RFS (P = .001) and OS (P = .01) rates than patients who did not. CONCLUSION BRCA1 status and ER negativity are independently associated with higher pCR rates in patients with breast cancer. Overall prognosis of breast cancer in BRCA carriers is similar to sporadic breast cancers.

Abstract:Characterization of the botanical origin and quality of honeys is of great importance and interest in agriculture. In this study, an electronic nose (e-nose) was applied for identifying the botanical origin of honey as well as determining their main quality components such as glucose, fructose, hydroxymethylfurfural (HMF), amylase activity (AA), and acidity. Principal component analysis (PCA) and discriminant factor analysis (DFA) were employed to generate scatter plots of honey samples from 14 botanical origins. Origin discrimination models with 100 % overall accuracy were established by least squares support vector machines (LS-SVM). LS-SVM outperformed the linear regression method of partial least squares regression (PLSR) for quality prediction, showing that the non-linear correlations between e-nose responses were important for the analysis of honey. Moreover, three sensor selection algorithms, namely, uninformation variable elimination (UVE), successive projections algorithm (SPA), and competitive adaptive reweighted sampling (CARS) were applied for the first time to analyze e-nose fingerprints of honey. After the calculation of the above three algorithms and the comparison of their results, from a total of 18 sensors, the important ones were selected for glucose (three), fructose (five), HMF (three), AA (five), and acidity (four) prediction, respectively. The results of sensor selection show the advantages of reducing redundancy of e-nose data, optimizing the sensor array of an e-nose, and improving the performance of models in terms of robustness. The overall results show that the laborious, time-consuming, and destructive analytical methods like high-performance liquid chromatography (HPLC), acid-base titration, and spectrophotometry could be replaced by e-nose to provide a rapid and non-invasive determination of the botanical origin and quality of honey.

Abstract:Background Natural medicines present a considerable analytical challenge due to their diverse botanical origins and complex multi‐species composition. This inherent complexity complicates their rapid identification and analysis. Tangerine peel, a product of the Citrus species from the Rutaceae family, is widely used both as a culinary ingredient and in traditional Chinese medicine. It is classified into two primary types in China: Citri Reticulatae Pericarpium (CP) and Citri Reticulatae Pericarpium Viride (QP), differentiated by harvest time. A notable price disparity exists between CP and another variety, Citri reticulatae “Chachi” (GCP), with differences being based on the original variety. Methods This study introduces an innovative method using portable miniature mass spectrometry for swift on‐site analysis of QP, CP, and GCP, requiring less than a minute per sample. And combined with machine learning to differentiate the three types on site, the method was used to try to distinguish GCP from different storage years. Results This novel method using portable miniature mass spectrometry for swift on‐site analysis of tangerine peels enabled the characterization of 22 compounds in less than one minute per sample. The method simplifies sample processing and integrates machine learning to distinguish between the CP, QP, and GCP varieties. Moreover, a multiple‐perceptron neural network model is further employed to specifically differentiate between CP and GCP, addressing the significant price gap between them. Conclusions The entire analytical time of the method is about 1 minute, and samples can be analyzed on site, greatly reducing the cost of testing. Besides, this approach is versatile, operates independently of location and environmental conditions, and offers a valuable tool for assessing the quality of natural medicines.

Abstract:A novel headspace integrated E-nose based on metal oxide gas sensor arrays is reported. And its performance to distinguish six groups of Chinese medical herbs which might be mixed easily by appearance is presented. The headspace integrated E-nose is mainly consisted of gas sensing module, signal procession module and human-machine interaction module. The E-nose, integrated with an innovative headspace diffusion sampling way, has a compact structure with an approximate size of Φ 48×100mm. For the characteristics of the small size and the headspace diffusion sampling way, it is suitable for on-site volatiles analysis in food and raw material detection. Temperature modulation technology was combined with pattern recognition analysis to enhance the selectivity of the E-nose. Principal components analysis and Sammon mapping were used in the feature extraction, while the sample was discriminated by Fisher discriminant analysis. The correct classification rate of the headspace integrated E-nose for discriminating thirteen species of herbs is 100%, which shows the potential application of the E-nose for on-site volatiles analysis.

Abstract:Abstract Zhejiang Suichang native honey, which is included in the list of China’s National Geographical Indication Agricultural Products Protection Project, is very popular. This study proposes a method of Raman spectroscopy combined with machine learning algorithms to accurately detect low-concentration adulterated Suichang native honey. In this study, the native honey collected by local beekeepers in Suichang was selected for adulteration detection. The spectral data was compressed by Savitzky–Golay smoothing and partial least squares (PLS) in sequence. The PLS features taken for further analysis were selected according to the contribution rate. In this study, three classification modeling methods including support vector machine, probabilistic neural network and convolutional neural network were adopted to correctly classify pure and adulterated honey samples. The total accuracy was 100%, 100% and 99.75% respectively. The research result shows that Raman spectroscopy combined with machine learning algorithms has great potential in accurately detecting adulteration of low-concentration honey.

Abstract:Honey manufacturers and traders have suffered economic damage due to the spurious labelling of honey botanical sources. The traditional detection methods for distinguishing honey origins have inherent performance disadvantages, such as costly equipment, complicated operations and need more time. This study proposes a novel method for identifying the origin of honey by applying electronic tongue (ET) and deep learning algorithm. First, a self-developed ET device is utilized to collect unique fingerprints of honey samples from different botanical origin. Due to the high dimensionality and complex nonlinearity of the collected signal, an unsupervised convolutional autoencoder (CAE) is developed to reduce the dimension of the ET signals and achieve effective feature extraction. Moreover, in order to avoid the gradient disappearance and suppress redundant information, the self-adaptive attention mechanism and residual learning is introduced in the CAE model to improve the feature extraction ability of the model. The proposed model is further used to measure actual honey samples and contrast with multiple machine learning and deep learning models. The experiment results show that the proposed model demonstrates better classification capabilities for honey from different botanical sources, and the accuracy, precision, F1 and AUC score reach 99.25%, 99.14%, 0.9919, and 0.9957, respectively. Compared with the other detection methods, the proposed method exhibited advantages in terms of convenience and accuracy of detection, which has great application prospects in the field of on-site measurements.
