Abstract:The objective of this study was to evaluate the effect of low dietary protein sources on the gut microbiota in finishing pigs. Thirty-six Duroc x Landrace x Large White finishing barrows were randomly allocated to four equal groups. Soybean meal (SBM) as a protein source is a control group and had a general protein level of 15% (SBM15). The pigs fed soybean meal (SBM11), soybean-cottonseed corn germ meal (SCCM11), and cottonseed-corn germ meal (CCM11) were offered low protein (11%) diets. The results showed that the ileum and feces microbiota of the SCCM11 group were significantly increased compared to the SBM15 group (P<0.05). At the phylum level, the relative abundance of Proteobacteria phyla was significantly increased in the ileum microbiota of the SCCM11 group compared to the SBM15 group (P<0.05). At the genus level, the abundance of Lactobacillus and Bifidobacterium showed a significant increase in the ileum microbiota of the SCCM11 group (P<0.05). The abundance of Ruminococcaceae, Phascolarctobacterium was significantly increased in the feces microbiota of the SCCM11 group than the other three groups (P<0.05). These results indicated that the SCCM11 group with added crystalline amino acid may modulate intestinal microbiota.
Effect of Dietary Amylose/Amylopectin Ratio on Intestinal Health and Cecal Microbes’ Profiles of Weaned Pigs Undergoing Feed Transition or Challenged With Escherichia coli Lipopolysaccharide

Abstract:Background Dietary amylose/amylopectin ratio (DAR) plays an important role in piglets’ intestinal health. It is controversial whether diarrhea could be relieved by changing DAR in weaning piglets. Methods Sixty (Landrace × Yorkshire) castrated male pigs (initial body weight (BW) 6.51 ± 0.64 kg) were randomly allocated to five groups (one pig per cage and 12 replicates per group) according to their BW. Piglets received diets with different DARs (0.00, 0.20, 0.40, 0.60, and 0.80) for 29 days. Feed transition occurs at day 15. The piglets were challenged with lipopolysaccharides ( Escherichia coli LPS, 100 μg/kg BW) on day 29 by intraperitoneal injection at 12 h before slaughter. Chyme was collected for pH value, short-chain fatty acid (SCFA), and cecal microbe analysis using 16S rRNA gene sequencing; mucosa was sampled for detecting gene expression. Results Rate and degree of diarrhea were higher when DAR was 0.40 than when it was 0.20 and 0.80 during the third week ( P < 0.05). The chyme pH value in the cecum was higher ( P < 0.05) in 0.20 DAR than in 0.00 and 0.80 DARs, but with no significant difference compared with 0.40 and 0.60 DARs ( P > 0.05). Cecal isobutyric acid and isovaleric acid concentrations were higher in 0.20 than in other groups ( P < 0.01). Cecal SCFAs such as acetic acid, propionic acid, and total SCFA, concentrations were higher in 0.40 DAR than in 0.00, 0.60, and 0.80 DARs ( P < 0.05), but with no significant difference when compared with 0.20 ( P > 0.05). Cecal crypt depth was lower ( P < 0.05) in 0.80 than in other groups, but not 0.40. Claudin mRNA expression in the mucosa of the ileum was higher in 0.20 than in other groups ( P < 0.01). The alpha diversity of cecal microbe representative by goods coverage was higher in group 0.40 when compared with group 0.20 ( P < 0.05). At the genus level, the abundances of the Ruminococcaceae_NK4A214_group and Anaerotruncus were higher but that of Cetobacterium was lower in the cecal chyme of group 0.20 than that of group 0.60 ( P < 0.05), with no significant difference compared with other groups ( P > 0.05). The diarrhea rate during the third week was negatively correlated with the abundances of Rikenellaceae_RC9_gut_group and X.Eubacterium_coprostanoligenes_group ( P < 0.05). Conclusion Compared with diet high in amylose or amylopectin, diet with DAR 0.40 showed a worse degree of diarrhea in weaned piglets during feed transition. But the intestinal health will be improved the week after the microbes and metabolites are regulated by DAR.

Integrated metagenomics-metabolomics analysis reveals the cecal microbial composition, function, and metabolites of pigs fed diets with different starch sources

Abstract:This study integrated metagenomics and metabolomics to evaluate the effects of diets with different starch sources on the microbial community, metabolic functions, and resultant metabolites in the cecum of finishing pigs. In this study, 48 crossbred growing barrows were randomly allocated to 2 treatment groups with eight replicate pens of 3 pigs each. Pigs were fed a tapioca starch (TS) diet or a pea starch (PS) diet (the ratio between amylose and amylopectin of the two diets were 0.11 and 0.44, respectively) for 44 days. The results showed that, compared with the TS diet, the PS diet increased (P < 0.05) the relative abundance of amylolytic bacteria, such as Lactobacillus spp., and Streptococcus spp., and decreased (P < 0.05) the relative abundance of some inflammatory bacteria, such as Tyzzerella, Porphyromonas, and Tannerella in the cecum of pigs. In addition, analysis of microbial functions showed that 11 carbohydrate-active enzymes, such as GH73, AA3, and AA6, were enriched in the PS group (P < 0.05), while 26 other enzymes, such as GH2, GH35, and GH53, were enriched in the TS group (P < 0.05). Meanwhile, KEGG pathway analysis showed a decreasing trend (P < 0.1) for energy metabolism and amino acid metabolism pathways, and a significant increase (P < 0.05) in the lipid metabolism pathways in the PS group. Correspondingly, the resultant metabolites related to amino acids and their derivatives (such as decreased histamine and indole, while increased γ-aminobutyric acid and pyroglutamic acid), and fatty acids and lipids (such as increased tetradecanoylcarnitine and monoacylglycerol) were also changed (P < 0.05) in the cecum of pigs fed the PS diet. In summary, these findings indicated that the cecal microbiota and metabolism underwent responsive changes to diets with different starch sources, and a high ratio of amylose to amylopectin in diets may be beneficial to intestinal health of pigs.
Pediatric orbital eosinophilic granuloma with intra- and extracranial extension--case report.

Abstract:A rare case of eosinophilic granuloma of the orbit in a 3-year-old boy presented as right upper eyelid swelling and proptosis. Computed tomographic scanning revealed a soft-tissue dense mass in the lateral wall of the right orbit. Magnetic resonance (MR) imaging demonstrated tumor extension into the orbit, anterior and middle cranial fossae, and extracranial region. The tumor was completely removed. Histological diagnosis was eosinophilic granuloma. No evidence of recurrence was found 14 months later. MR imaging is useful for diagnosis of the lesion, and particularly for surgical management.
Gastrointestinal microbiota and metabolites responses to dietary cereal grains in an adult pig model

Abstract:Corn (C), wheat (W), and paddy rice (PR) are important energy sources and are commonly used in feed production for swine. This study mainly focuses on the variation and regularities of microbiota and metabolites in the gastrointestinal tract (GIT) of pigs in response to C, W, and PR. A total of 18 pigs were allotted into three dietary groups with six replicated pigs and received diets containing C, W, or PR as the sole energy source, respectively. The results showed that digestive parts significantly affected the diversity of microbial communities. Cereal grain sources significantly influenced the β-diversity of microbial communities in the colon and rectum. Campylobacterota and Proteobacteria are mainly distributed in the duodenum, Lactobacillus in the jejunum, and Bacteroidota in the colon and rectum. The W diet increased the Bacteroidota, Spirochaetota, and Prevotellaceae_NK3B31_group abundances and showed the highest concentrations of all short-chain fatty acids (SCFAs) in the hindgut. Fibrobacterota, Bacteroidota, Spirochaetota, Prevotellaceae_NK3B31_group, Prevotella, and Treponema in the colon or rectum were positively correlated with acetate, propionate, butyrate, and total SCFAs. These findings suggested that aerobic bacteria and facultative anaerobes in the foregut will gradually be replaced by anaerobes in the hindgut. The W diet had the best fermentability and was beneficial to the colonization of microbial communities that mainly used carbohydrates. The hindgut flora of the PR diet group may be more balanced with fewer potential pathogenic bacteria. Many microbial communities have been identified to contribute positively to the SCFA production of the hindgut. Collectively, our study revealed the spatial variation regularities of GIT microbial communities in an adult pig model and provided new insights into GIT microbiota and responses of metabolites to cereal grain diets.

Abstract:To investigate the influence of baseline enterotypes and dietary starch type on the concentration of short-chain fatty acids (SCFA), numbers of butyrate producing bacteria and the expression of genes related to intestinal barrier and inflammatory response in the colon of finishing pigs, a 60-d in vivo trial was conducted. A 2-wk pre-trial with 102 crossbred [Duroc × (Landrace × Yorkshire)] finishing barrows (90 d old) was conducted to screen enterotypes. Then, a total of 32 pigs (87.40 ± 2.76 kg) with high (HPBR, ≥ 14) and low (LPBR, ≤ 2) Prevotella-to-Bacteroides ratio (PBR) in equal measure were selected and randomly divided into 4 groups with 8 replicates per group and one pig per replicate. The trial was designed following a 2 (PBR) × 2 (amylose-to-amylopectin ratio, AMR) factorial arrangement. Pigs with different PBR were fed diets based on corn-soybean meal with high AMR (HAMR, 1.24) or low AMR (LAMR, 0.23), respectively. Results showed that neither PBR nor AMR influenced the growth performance of pigs. HPBR pigs fed HAMR diet had a higher number of colonic Clostridium cluster XIVa and higher gene expression of butyrate kinase compared to the LPBR pigs (P < 0.05). The HPBR pigs fed HAMR diets also had increased colonic concentrations of total SCFA and propionate compared to the LPBR pigs (P < 0.05). Comparing with other pigs, HPBR pigs fed HAMR diets showed a lower (P < 0.05) expression of histone deacetylases (HDAC) gene and higher (P < 0.05) expression of G protein-coupled receptor 43 gene (GPR 43) in the colonic mucosa. The interaction (P < 0.05) of HPBR and HAMR was also found to decrease the gene expression of interleukin (IL)-6, IL-12, IL-1β and tumor necrosis factor-α (TNF-α) in colonic mucosa. These findings show that HAMR diet increased the abundance and activity of butyrate-producing bacteria and the concentration and absorption of SCFA, which may be associated with the decreased gene expression of inflammatory cytokines in the colonic mucosa of pigs with Prevotella-rich enterotype. All these alterations are likely to have a positive effect on the intestinal health of finishing pigs.

Methods: A total of 60 (Landrace × Yorkshire) castrated male pigs (initial body weight 6.51 ± 0.64 kg) were allotted to 5 groups (1 pig/cage and 12 replicates/group) in a randomized complete block design according to their BW. Piglets were fed ad libitum with diets different in DAR (0.00, 0.20, 0.40, 0.60, 0.80) for 28 days. Feed transition occurs at day 15. 100 μg/kg body weight lipopolysaccharides or same amount of saline were injected via abdomen on day 29, 12 h before slaughter. Results: Diarrhea rate and diarrhea degree were higher when DAR was 0.40 than in groups 0.20 and 0.80 during third weed ( P <0.05). Iso-valeric acid of ileum decreased after LPS stress ( P <0.05). Cecal iso-butyrate acid and iso-valeric acid concentrations increased after LPS stress ( P <0.05). Iso- and valeric acid concentration of ileal chyme was higher in group 0.20 compared with other groups ( P <0.05). Cecal acetic acid and propionic acid concentration were higher in group 0.40 than in group 0.20 ( P <0.05). Alpha diversity of cecal microbial representative by goods coverage was higher in group 0.40 when compared with group 0.20 ( P <0.05). At the genus level, the abundance of Ruminococcaceae_ NK4A214_ group and Anaerotruncus in cecal chyme of Group 0.20 was significantly higher than that in Group 0.40 ( P <0.05), with no significant difference compared with other groups ( P >0.05). The abundance of Cetobacterium was significant lower in cecal chyme from Group 0.20 compared with pigs from Group 0.40( P <0.01), with no significant difference compared with other groups ( P >0.05). Diarrhea rate during third week negative correlated with abundance of Rikenellaceae_RC9_ gut_ group and X.Eubacterium_coprostanoligenes_group (P <0.05). Conclusions: Diarrhea happened when weaned piglets received diet with DAR 0.40 during feed transition, but they will regulate cecal microorganism and metabolites such as acetic acid and propionic acid to restore their health.
Polysaccharide source altered ecological network, functional profile, and short-chain fatty acid production in a porcine gut microbiota

Abstract:Several validated dynamic in vitro models of the colon have been developed for humans, but there is no dynamic in vitro fermentation model for pigs. This study was conducted to modify the human, dynamic, computer-controlled TNO in vitro model of the colon (TIM-2) for pigs and investigate effects of different starch sources and polysaccharides on swine microbiota structure, ecological network, predictive functional profile, and short-chain fatty acids production. Our study showed that three different types of starch or two polysaccharides greatly impacted microbiota composition. Co-occurrence network analysis indicated that microbiota fed with different sources of starch changed the network topological properties. Functional profiles were predicted to vary significantly among the three starch treatments, and the original pig faecal inoculum was more similar to maize starch treatment. On the other hand, compared with maize starch and arabinoxylans (AX), the microbial composition of the original inoculum was more similar when AX-XG (arabinoxylans and xyloglucan) were added, and the functional profile of the original inoculum also clustered with AX-XG. The cumulative production of acetic, propionic, and butyric acid on maize starch were significantly higher than those on potato starch and wheat starch, while only the amount of acetic acid was significant higher on AX-XG than that on AX. In conclusion, supplementation of maize starch as the starch source together with AX and XG, leads to the bacteria being more stable in the in vitro model and closer to the original inoculum and microbial function compared to potato starch, wheat starch and AX. A maize basal diet may improve energy absorption in the large intestine in growing pigs.
Effects of Dietary Amylose—Amylopectin Ratio on Growth Performance and Intestinal Digestive and Absorptive Function in Weaned Piglet Response to Lipopolysaccharide

Abstract:This study investigated the effects of diet with different amylose–amylopectin ratios (AAR) on the growth performance, intestinal morphology, digestive enzyme activities and mRNA expression of nutrients transporters in piglets with short-term lipopolysaccharide (LPS) intraperitoneal injections. Sixty 21 days-old piglets (Landrace × Yorkshire; 6.504 ± 0.079) were randomly assigned based on their body weight (BW) and litters of origins to five groups with experimental diets with an AAR of 0.00, 0.20, 0.40, 0.60, or 0.80 (namely, the 0.00, 0.20, 0.40, and 0.80 groups), respectively. Each treatment included 12 piglets (one piglet per pen). This experiment lasted for 28 days. On the 28th day, six piglets in each treatment were randomly selected for an LPS intraperitoneal injection (100 μg/kg BW), and other piglets were injected with normal saline. Twelve hours after LPS injection, all piglets were sacrificed to collect small intestinal mucosa for analysis. Although different AAR did not influence the final BW in piglets, the piglets in the 0.40 group represented the poorest feed-to-gain ratio (F/G) in the first, second and fourth week (p < 0.05) and the lowest average daily gain (ADG) in the fourth week (p < 0.05) compared with other groups. In terms of the small intestinal morphology, piglets in the 0.20 and 0.60 groups showed better ileal villous width (p < 0.05). Piglets in the 0.60 group presented greater activities of jejunal maltase, sucrase and alkaline phosphatase (p < 0.05) than those of 0.20 and 0.40. However, a low amylose diet increased the mRNA expression of jejunal glucose and amino acid transporters (p < 0.05). In addition, compared to saline injection, the LPS challenge significantly lessened jejunal digestive enzyme activities (p < 0.01) and, ileal villous width and downregulated the gene expression of glucose and amino acid transporters (p < 0.05) in piglets. Interestingly, certain diet -LPS interactions on duodenal VH/CD, jejunal maltase activity (p < 0.05) and the expression of glucose transporters (p < 0.05) were observed. Taken together, in terms of small intestinal digestion and absorption capacity, these results demonstrated that a diet with an AAR of 0.60 diets could improve the intestinal digestive and absorptive capability by affecting small intestinal morphology, digestive enzymes, and nutrients absorptions in piglets. In addition, the diets containing an AAR of 0.40–0.60 were more likely to resist the damage of LPS stress to intestinal morphology and nutrient absorption.

Effects of dietary crude protein content and resistant starch supplementation on growth performance, intestinal histomorphology and microbial metabolites in weaned pigs

Abstract:A 4-week study was conducted to evaluate the effects of dietary crude protein (CP) content and resistant starch (RS) supplementation on growth performance, intestinal histomorphology and microbial metabolites of weaned pigs. A total of 96 pigs (7.06 ± 0.45 kg body weight) were assigned to 1 of 4 diets in a randomised complete block design involving a 2 (CP levels) × 2 (without or with RS) factorial arrangement to give 8 replicate pens and 3 pigs per pen. Body weight and feed disappearance were recorded weekly, and the faecal consistency score was determined every morning. Blood was sampled on days 1, 14 and 28 from one pig per pen, and the same pig was euthanised on day 28 to collect ileal tissue and ileal and colon digesta. Data were analysed using the MIXED procedure of SAS. The average daily gain and gain:feed ratio were lower (p < 0.05) in pigs fed low crude protein (LCP) diets compared to those fed high CP (HCP) diets during week 3 and overall period. The analysed Lys, Met+Cys and Thr in feed were lower than calculated values, particularly in LCP diets, which may have affected performance. Pigs fed the LCP diets had longer (p < 0.05) ileal villi and higher villus height to crypt depth ratios than those fed the HCP diets, and RS supplementation increased (p < 0.05) ileal villus height. Interactions (p < 0.05) between dietary CP content and RS inclusion were observed for short-chain fatty acid concentration in the ileum and colon in phase 2. There was no difference in propionic acid (ileum) or butyric acid (colon) concentrations among pigs fed HCP diets, however, the butyric acid concentration increased in pigs fed the LCP diet when supplemented with RS. Reducing dietary CP lowered (p < 0.05) faecal score, plasma urea nitrogen and digesta ammonia content. Overall, feeding LCP diets reduced growth performance but improved gut morphology in weaned pigs. Feeding the LCP diet with RS supplementation modulated concentrations of ileal propionic acid and colonic butyric acid in weaned pigs.

In this research, we investigated the fermentation characteristics of amylopectin through an in vitro fermentation method and used an ileal infusion pig model to verify the fermentation trial results and explore the microbiota regulatory effect.

Abstract:This study aimed to determine whether dietary protein content influences pig health as indicated by ileal microbiota structure and coefficients of total tract apparent digestibility (CTTAD) of nutrients. Seventy-two gilts, with an initial body weight of 29.9 +/- 1.5kg, were used in this 42-day feeding study. Pigs were randomly allotted to one of three dietary treatments of corn-soybean meal contained 14, 16 or 18% crude protein (CP). As dietary CP content decreased, the CTTAD of most essential amino acids (AAs), except for arginine and histidine, increased linearly, while those of most nonessential AAs decreased linearly. The concentration of total short-chain fatty acids (SCFA) was higher in pigs fed the diet with 14% CP content than others. Ileal microbiota structure was changed by dietary treatments. In particular, at the phylum level, the relative abundance of Tenericutes in ileal digesta decreased as the dietary protein content reduced, while that of cyanobacteria increased. At the genus level, the relative abundance of Weeksella, Phaseolus acutifolius, Slackia, Sulfurimonas and Aerococcus showed significant differences among the three dietary treatments. In conclusion, ileal microbiota structure was changed by dietary protein content. Moderate reduction of protein intake can benefit gut health by enhancing the gut microbial fermentation and SCFA formation.
Metatranscriptomic analysis of functional response of colonic microbiota to different dietary fibers in growing pigs

Abstract:Abstract Background Dietary fibers are widely considered to be beneficial for health by producing nutrients by gut microbial fermentation while helping with weight management and gut health. So far, the gene expression profiles of CAZymes responsing to different type of fibers (raw potato starch, RPS; inulin, INU; pectin, PEC) in the gut microbes of pigs are not well understood. Therefore, we investigate the functional response of colonic microbiota to different dietary fibers in pigs based on metatranscriptomic analysis. Results Results showed that the microbial composition and carbohydrate active enzymes (CAZymes) structure of three experimental groups changed significantly compared to the control group (CON). As determined by comparative analysis with the control diet, RPS increased the abundances of Parabacteroides, Ruminococcus, Faecalibacterium and Alloprevotella, and decreased Sutterella. INU increased the relative abundance of Fusobacterium and Rhodococcus, while decreased Bacillus. And pectin treatment increased the relative abundance of Streptococcus and Bacteroidetes group, while decreased the relative abundance of Clostridium, Clostridioides, Intestinibacter, Gemmiger, Muribaculum, and Vibrio. The gene expression of CAZymes GH8, GH14, GH24, GH38, GT14, GT31, GT77 and GT91 downregulated while GH77, GH97, GT3, GT10 and GT27 upregulated in the RPS diet group; AA4, AA7, GH14, GH15, GH24, GH26, GH27, GH38, GH101, GT26, GT27 and GT38 downregulated in the INU group; as to the PEC group, the gene expression of PL4, AA1, GT32, GH18, GH37, GH101, and GH112 downregulated while CE14, AA3, AA12, GH5, GH102 and GH103 upregulated. Compared to RPS and INU groups, the composition of colonic microbiota in the PEC group had more diverse changes with the variation of carbohydrate active enzymes and Streptococcus as the main contributor to CBM61, which promoted the digestion of pectin greatly. Conclusions The results of this exploratory study displayed a comprehensive overview on the effects of different fibers on gut microbiota and CAZymes in pig colons, which will provide new insights into the impacts of the use of dietary fibers on animal or human health.

Abstract:The objective of the present study was to evaluate the effects of dietary starch structure (amylose/amylopectin ratio, AR) on intestinal health of weaned pigs by determining the intestinal morphology, barrier functions, epithelium apoptosis and digestive enzymes. A total of one hundred and eight pigs (initial body weight 7.51 ± 0.05 kg) were randomly allotted on the basis of their body weight (BW) and litters to one of the six experimental diet treatments with six replicates per treatment and three pigs per replicate. The six diets included a control (corn-soybean meal basal diet, CON) diet and five experimental diets with AR being 2.90, 1.46, 0.68, 0.31 and 0.14, respectively. The diets and water were provided ad libitum for 21 d. Results showed that pigs fed CON and AR 2.90 diets had higher (P < 0.05) villus height and ratio of villus height : crypt depth in the jejunum and ileum compared with the other four starch groups. Pigs of CON and AR 0.14 groups had greater (P < 0.05) activities of sucrase (SA) and maltase (MA) than the pigs of other groups. Additionally, compared with the high amylopectin (AR 0.31 and 0.14) groups, the total apoptotic percentage was significantly decreased in (P < 0.01) CON and high amylose (AR 2.90) groups. Ingestion of CON and high amylose diets upregulated (P < 0.05) the levels of B-cell lymphoma-2 (Bcl-2) and occludin (OCLN) gene expression in duodenum and jejunum. However, no significant differences in the level of zonula occludens 1 (ZO-1) gene expression were observed in pigs fed with different AR diets. Interestingly, CON and high amylose groups enhanced the localization of ZO-1 protein in apical intercellular region of jejunal epithelium. Together, these results suggest that diets with high amylose can influence the intestinal health of young pigs by improving their intestinal barrier functions and reducing the epithelial cell apoptosis.

Abstract:Background: The absorption and utilization of proteins by animals is affected by the amino acid (AA) release characteristics of their diets. In the present study, we aimed to determine the effects of diets with various amino acid release characteristics on the intestinal barrier function and diversity of gut microbiota of weaned pigs. Results: Forty-eight pigs (7.45 ± 0.58 kg) were fed with diets having different amino acid release characteristics during a period of 28 days. We used a 2 × 3 full-factor (two protein levels and three protein sources with differing amino acid release characteristics) experimental design, with normal (standard terminal ileal digestibility of 17.5%) or low (standard terminal ileal digestibility of 14.9%) protein levels as the first factor. Casein (CAS), corn gluten meal (CGM) and a MIX diet were used as protein sources. Due to the more balanced release of amino acids, the diamine oxidase (DAO) concentrations in the CAS and MIX groups were significantly lower than those in the CGM group (P < 0.05); Reducing the dietary protein content from 17.5% to 14.9% had no significant effects on the levels of serum DAO or D-lactic acid. By contrast, it increased the microbial diversity (chao1 and ACE values) and the number of Lactobacillus in the jejunum (P < 0.05). The CAS-containing diet and the MIX diet resulted in significantly higher microbial diversity (Simpson and Shannon) than the CGM-containing diet in the jejunum. Conclusion: The balanced release of amino acids in CAS and MIX diets maintained intestinal barrier function and increased gut microbiota diversity. These findings could potentially provide a scientific reference for the rational preparation of piglet feed.

Abstract:Increasing the ratio of amylose in the diet can increase the quantity of starch that flows to the large intestine for microbial fermentation. This leads to the alteration of microbiota and metabolite of the hindgut, where the underlying mechanism is not clearly understood. The present study used a combination of 16S amplicon sequencing technology and metabolomics technique to reveal the effects of increasing ratios of amylose/amylopectin on cecal mucosa- and digesta-associated microbiota and their metabolites in young goats. Twenty-seven Xiangdong black female goats with average body weights (9.00 ± 1.12 kg) were used in this study. The goats were randomly allocated to one of the three diets containing starch with 0% amylose corn (T1), 50% high amylose corn (T2), and 100% high amylose corn (T3) for 35 days. Results showed that cecal valerate concentration was higher ( P < 0.05) in the T2 group than those in the T1 and T3 groups. The levels of tumor necrosis factor-α (TNF-α) and interleukin (IL)-6 were decreased ( P < 0.05) in cecal tissue while IL-10 was increased ( P < 0.05) in the T2 group when compared with T1 or T3 groups. At the phylum level, the proportion of mucosa-associated Spirochaetes was increased ( P < 0.05), while Proteobacteria was deceased by feeding high amylose ratios ( P < 0.05). The abundance of Verrucomicrobia was decreased ( P < 0.05) in the T3 group compared with the T1 and T2 groups. The abundance of digesta-associated Firmicutes was increased ( P < 0.05) while Verrucomicrobia and Tenericutes were deceased ( P < 0.05) with the increment of amylose/amylopectin ratios. The LEfSe analysis showed that a diet with 50% high amylose enriched the abundance of beneficial bacteria such as Faecalibacterium and Lactobacillus in the digesta and Akkermansia in the mucosa compared with the T1 diet. The metabolomics results revealed that feeding a diet containing 50% high amylose decreased the concentration of fatty acyls-related metabolites, including dodecanedioic acid, heptadecanoic acid, and stearidonic acid ethyl ester compared with the T1 diet. The results suggested that a diet consisting of 50% high amylose could maintain a better cecal microbiota composition and host immune function.

Abstract:Abstract Background Dietary fibers are widely considered to be beneficial to health as they produce nutrients through gut microbial fermentation while facilitating weight management and boosting gut health. To date, the gene expression profiles of the carbohydrate active enzymes (CAZymes) that respond to different types of fibers (raw potato starch, RPS; inulin, INU; pectin, PEC) in the gut microbes of pigs are not well understood. Therefore, we investigated the functional response of colonic microbiota to different dietary fibers in pigs through metatranscriptomic analysis. Results The results showed that the microbial composition and CAZyme structure of the three experimental groups changed significantly compared with the control group (CON). Based on a comparative analysis with the control diet, RPS increased the abundance of Parabacteroides , Ruminococcus , Faecalibacterium and Alloprevotella but decreased Sutterella ; INU increased the relative abundance of Fusobacterium and Rhodococcus but decreased Bacillus ; and PEC increased the relative abundance of the Streptococcus and Bacteroidetes groups but decreased Clostridium , Clostridioides , Intestinibacter , Gemmiger , Muribaculum and Vibrio . Metabolic and Transcriptomic Responses of Weaned Pigs Induced by Different Dietary Amylose and Amylopectin Ratio

Abstract:Starch is one of the major dietary energy sources for mammals. However, the nutritional value of starch largely depends on its amylose and amylopectin ratio. In this study, the overall metabolic and transcriptomic responses of weaned pigs fed with different dietary starches were assessed. Sixteen weaned pigs were randomly allotted to two experimental diets containing either of pure cassava starch (CS) or maize starch (MS) as the sole energy source (the amylose-amylopectin ratio were 0.25 and 0.43, respectively). Results indicated that the body weight gain was not affected by different dietary starches. However, a moderate fatty liver was observed in CS-fed group. Long-term ingestion of CS not only increased the total liver fat content, but significantly elevated the liver triglyceride and cholesterol content (P<0.05). In addition, the serum insulin and cholesterol concentrations were both elevated in CS-fed group (P<0.05). Microarray analysis led to the identification of 648 genes differentially expressed in liver (P<0.05), and a lot of them were involved in lipid and carbohydrate metabolism. Additionally, pathway analysis indicated that both the insulin and PPAR signaling pathways were acutely affected by dietary amylose-amylopectin ratio. Long-term ingestion of CS activated the transcription of lipogenic genes such as hmgr and fasn, but decreased the expression of lipolytic genes such as aox1, ppara and fbp. The microarray results correlated well with the measurements of several key enzymes involved in hepatic lipid metabolism. Our results suggested that both the metabolic and transcriptomic responses of weaned pigs were tightly regulated by dietary starch composition, and a high amylose ratio starch (i.e MS) may be more healthful for mammals as the long-term energy source by down-regulation of hepatic lipogenesis and steroidogenesis.
