Abstract:To study the effect of exogenous fecal microbiota intervention on intestinal barrier function in recipient pigs,twelue litters of "Duroc×Landrace×Yorkshire" suckling piglets were randomly allocated into two groups on the basis of similar body weight,birth time and parity.Each group had six litters (10～12 piglets/litter).All piglets were infused by gavage with exogenous fecal suspension or same amount of sterile phosphate buffered solution (PBS) every other day from the first day after birth to 10 days of age.All piglets in each litter had free access to sow and water during the experiment.Piglets were killed and samples were collected on 12 days of age and 27 days of age.The results indicated that:Exogenous fecal microbiota intervention significantly decreased the incidence of diarrhea and increased the average daily gain and survival rate in suckling piglets (P＜0.05).Exogenous fecal microbiota intervention significantly decreased crypt depth in the intestine of suckling piglets at 27 days of age (P＜0.05) and increased villus height/crypt depth in jejunum and ileum at 12 and 27 days of age (P＜0.05).Exogenous fecal microbiota intervention significantly increased the number of goblet cells and SIgA+ cells (12 days of age) and improved the relative expression of Muc2 and tight junction protein ZO-1 and Occludin in the intestine (P＜0.05).While the number of SIgA+ cells had no significant difference between the treatment group and the control group at 27 days of age (P＞0.05).It is suggested that exogenous fecal microbiota intervention can increase the number of goblet cells and SIgA+ cells,improve the expression of Muc2 and tight junction protein and regulate intestinal barrier function in recipient pigs.Exogenous fecal microbiota intervention has the prospect of maintaining intestinal physiology homeostasis and promoting intestinal health in pigs.

Abstract:The objective of this study was to evaluate the effect of low dietary protein sources on the gut microbiota in finishing pigs. Thirty-six Duroc x Landrace x Large White finishing barrows were randomly allocated to four equal groups. Soybean meal (SBM) as a protein source is a control group and had a general protein level of 15% (SBM15). The pigs fed soybean meal (SBM11), soybean-cottonseed corn germ meal (SCCM11), and cottonseed-corn germ meal (CCM11) were offered low protein (11%) diets. The results showed that the ileum and feces microbiota of the SCCM11 group were significantly increased compared to the SBM15 group (P<0.05). At the phylum level, the relative abundance of Proteobacteria phyla was significantly increased in the ileum microbiota of the SCCM11 group compared to the SBM15 group (P<0.05). At the genus level, the abundance of Lactobacillus and Bifidobacterium showed a significant increase in the ileum microbiota of the SCCM11 group (P<0.05). The abundance of Ruminococcaceae, Phascolarctobacterium was significantly increased in the feces microbiota of the SCCM11 group than the other three groups (P<0.05). These results indicated that the SCCM11 group with added crystalline amino acid may modulate intestinal microbiota.
Changes of Gut Microbiota Structure and Morphology in Weaned Piglets Treated with Fresh Fermented Soybean Meal

Abstract:This study investigated the effects of dietary fresh fermented soybean meal (FSM) on the intestinal microbiota and metabolites, bacterial enzyme activity and intestinal morphology of weaning piglets. A total of 64 weaned piglets were randomly allocated into two treatments. A corn-soybean-based diet was used as the control and other treatment was fed the same basal diet containing 15% fresh FSM. The feeding trial lasted for 21 days. Bacterial community structure and diversity in the cecum and colon were assessed using pyrosequencing-based analysis. The results showed that the phylum level, Firmicutes, Bacteroidetes, Proteobacteria and Tenericutes were dominant in the cecum or colon. Gut Firmicutes increased, while Bacteroidetes and Proteobacteria decreased in the fresh FSM-fed piglets. At the genus level, the relative abundances of butyrate-producing bacteria, Lactobacillus and Prevotella were higher in both cecum and colon of fresh FSM fed piglets. Meanwhile, fresh FSM could promote the development of intestinal morphological and reduce the incidence of diarrhea. The results indicated that fresh FSM might change intestinal function by influencing intestinal microenvironment.
Effects of Soybean Antigen Protein on the Growth Performance of Weaning Piglets

Abstract:Forty-eight piglets(Duroc×Large white×Landrace) weaned at 21 days were used to study the effects of soybean antigen protein on the growth performance in a period of 15 days.The results showed that soybean antigen protein significantly decreased AFI by 25.88%(P0.01)and increased diarrhea rate by 58.77%(P0.01) compared with the control group.The serum D-xylose detection indicated that the serum D-xylose concentration was decreased by 62.39%(P0.01),68.54%(P0.01),67.27%(P0.01),67.98%(P0.01) and 69.95%(P0.01) after 1,2,3,4 h and 5 h respectively compared with the control group.The lengths of the villi within the jejunum and ileum tissues of the test group significantly decreased by 27.56% and 22.43%(P0.01) and the crypt depth significantly increased by 30.01% and 53.03%(P0.01) compared with the control group.There was no significant difference between duodenal villi length and crypt length.The activity of NF-κB in jejunum were increased 71.87%(P0.01).The results indicated that soybean antigen protein in diet exerted a negative influence on growth performance of weaning piglets.Soybean antigen protein could regulate the nuclear factor activity of intestinal mucosa tissue,and affect the morphology and structure of intestinal mucosa tissue,furthermore,affect feed intake,diarrhea rate and growth performance of weaning piglets.
Effects of Cello-Oligosaccharide on Intestinal Microbiota and Epithelial Barrier Function of Weanling Pigs

Abstract:A total of 144 piglets (Duroc × Landrace × Yorkshire; average initial weight of 6.13 kg weaned at 21 ± 1 d age) were allotted to 4 treatments for 2 wk, each of which had 6 pens with 6 pigs per pen. After the feeding experiment, 6 pigs per treatment were slaughtered to investigate the effects of cello-oligosaccharide (COS) on intestinal microbiota and epithelial barrier function. The COS was added to the basal diet at 0, 1.5, 3.0, and 4.5 g/kg diet at the expense of corn, respectively. Plasma -lactate, diamine oxidase (DAO), and the Ussing chamber technique were used to determine the intestinal barrier function. 16S rRNA-based methods were used for intestinal microbiota analysis. The results showed that incremental levels of COS had no effect ( > 0.05) on growth performance. Incremental levels of COS increased lactobacilli in jejunal and colonic contents ( < 0.05); decreased in jejunal contents ( < 0.05) and and in colonic contents ( < 0.05); reduced plasma DAO (linear, = 0.013, and quadratic, = 0.037); increased jejunal mucosa DAO (linear, = 0.003, and quadratic, = 0.008); decreased fluorescein isothiocyanate dextran 4 kDa flux of jejunum and colon ( < 0.05); and increased transepithelial electrical resistance (TER) in colon ( < 0.05), claudin-1 protein expression in jejunal mucosa (linear, = 0.001, and quadratic, = 0.003), and protein expressions of claudin-1 and zonula occludens-1 (ZO-1) in colonic mucosa linearly ( = 0.001 and = 0.001, respectively) and quadratically ( = 0.001 and = 0.002, respectively). The results indicated that the improved microbial ecosystem in the presence of COS might contribute to improvement in intestinal barrier function and tight junction proteins. Results also showed that the appropriate dietary COS supplementation level was 3.0 g/kg in weaned pig diets under our trial conditions.
The effect of dietary supplementation of low crude protein on intestinal morphology in pigs

Abstract:To explore the effects of reducing the Cp levels on intestinal barrier function, low Cp (LP) and NRC standard Cp (NP) diets were fed to pigs from 45 to 160 days, and in vitro experiments were performed using monolayers of IPEC-J2 cells. The number of goblet cells, expression of proteins related to cell junction, amino acid transport, glucose transport, transepithelial electrical resistance (TEER), dextran permeability, and IL-6 secretion level were detected in pigs. The results demonstrated that a moderate reduction of Cp levels did not affect intestinal morphology, as demonstrated by a normal villi height, crypt depth and normal numbers of goblet cells. The maintenance of the intestinal structure obtained with LP was also confirmed by stable mRNA expression levels of muc2 and E-cadherin in the jejunum. We also found that LP did not affect the protein expression of cationic amino acid transporter 1 (CAT-1) and alanine serine cysteine transporter 1 (ASCT1) from 45 to 160 days. Moreover, the excitatory amino acid transporter 3 (EAAT3), sodium-glucose cotransporter 1 (SGLT1) and glucose transporter (GLUT2) protein expression levels in the jejunum were significantly increased at a certain age during the rearing period. Furthermore, we also demonstrated that a reduction in protein concentration up to 15% in the cultural medium of IPEC-J2 cells did not impact the mucosal barrier function. This study demonstrated that a moderate reduction of the protein level did not affect intestinal mucosal barrier function and morphology in the jejunum.
Positive Effects of a Clostridium Butyricum-Based Compound Probiotic on Growth Performance, Immune Responses, Intestinal Morphology, Hypothalamic Neurotransmitters, and Colonic Microbiota in Weaned Piglets

Abstract:Weaning stress in piglets can lead to poor health outcomes and reduced production. We investigated the effects of probiotics, one potential antibiotic alternative, on the growth performance, serum biochemical parameters, intestinal morphology, mucosal immunity, hypothalamic neurotransmitters, and colonic microflora in weaned piglets. Thirty-six weaned piglets were fed a basal diet, a diet supplemented with colistin sulphate antibiotic, or a diet supplemented with probiotics including Clostridium butyricum, Bacillus subtilis, and B. licheniformis. Probiotics significantly increased the feed : gain ratio, improved the average day gain from day 1 to day 28, and decreased the diarrhoea index. Probiotics also lowered the serum concentrations of AST, ALT, and ALP on day 14 and lowered the serum concentration of ALT on day 28 compared with the control. Probiotic supplementation caused fewer ileal apoptotic cells. The serum and ileal concentrations of TNF-α and IL-1β on day 28 were significantly lowered, and the serum concentrations of IL-6 were significantly lowered on days 14 and 28. Probiotic-fed piglets exhibited higher contents of hypothalamic serotonin and dopamine as well as serum γ-aminobutyric acid along with higher colonic concentrations of butyrate and valerate on day 28. High-throughput sequencing showed 972 core operational taxonomic units among all groups, of which 48 were unique to the probiotic-treated group. The relative abundance of genus Bacillus and species Bacillus velezensis was enriched in probiotic piglets; the phylogenetic investigation of communities by the reconstruction of unobserved states indicated that amino acid metabolism, DNA repair, replication and recombination proteins, and secretion systems were enriched with probiotics. In conclusion, the Clostridium butyricum-based probiotics improved growth performance, enhanced intestinal morphology, changed hypothalamic neurotransmitters and modulated colonic microflora in weaned piglets.
Dietary protein level affects nutrient digestibility and ileal microbiota structure in growing pigs.

Abstract:This study aimed to determine whether dietary protein content influences pig health as indicated by ileal microbiota structure and coefficients of total tract apparent digestibility (CTTAD) of nutrients. Seventy-two gilts, with an initial body weight of 29.9 +/- 1.5kg, were used in this 42-day feeding study. Pigs were randomly allotted to one of three dietary treatments of corn-soybean meal contained 14, 16 or 18% crude protein (CP). As dietary CP content decreased, the CTTAD of most essential amino acids (AAs), except for arginine and histidine, increased linearly, while those of most nonessential AAs decreased linearly. The concentration of total short-chain fatty acids (SCFA) was higher in pigs fed the diet with 14% CP content than others. Ileal microbiota structure was changed by dietary treatments. In particular, at the phylum level, the relative abundance of Tenericutes in ileal digesta decreased as the dietary protein content reduced, while that of cyanobacteria increased. At the genus level, the relative abundance of Weeksella, Phaseolus acutifolius, Slackia, Sulfurimonas and Aerococcus showed significant differences among the three dietary treatments. In conclusion, ileal microbiota structure was changed by dietary protein content. Moderate reduction of protein intake can benefit gut health by enhancing the gut microbial fermentation and SCFA formation.
Effects of Chitosan Nanoparticle Supplementation on Growth Performance, Humoral Immunity, Gut Microbiota and Immune Responses after Lipopolysaccharide Challenge in Weaned Pigs

Abstract:In this study, we aimed to determine the effects of dietary supplementation with chitosan nanoparticles (CNP) on growth performance, immune status, gut microbiota and immune responses after lipopolysaccharide challenge in weaned pigs. A total of 144 piglets were assigned to four groups receiving different dietary treatments, including basal diets supplemented with 0, 100, 200 and 400 mg/kg CNP fed for 28 days. Each treatment group included six pens (six piglets per pen). The increase in supplemental CNP concentration improved the average daily gain (ADG) and decreased the feed and gain (F/G) and diarrhoea rate (p < .05). However, significant differences in the average daily feed intake (ADFI) among different CNP concentrations were not observed. CNP also increased plasma immunoglobulin (Ig)A and IgG, and C3 and C4 concentrations in piglets in a dose-dependent manner on day 28, whereas IgM concentration was not affected by CNP. A total of 24 piglets in the control diet and control diet with 400 mg/kg CNP supplementation groups were randomly selected for the experiment of immunological stress. Half of the pigs in each group (n = 6) were injected i.p. with Escherichia coli lipopolysaccharide (LPS) at a concentration of 100 μg/kg. The other pigs in each group were injected with sterile saline solution at the same volume. Plasma concentrations of cortisol, prostaglandin E2 (PEG2), interleukin (IL)-6, tumour necrosis factor (TNF)-α and IL-1β dramatically increased after LPS challenge. However, CNP inhibited the increase in cortisol, PEG2, IL-6 and IL-1β levels in plasma, whereas TNF-α level slightly increased. Moreover, the effects of CNP on the gut microbiota were also evaluated. Our results showed that dietary supplementation with CNP modified the composition of colonic microbiota, where it increased the amounts of some presumably beneficial intestinal bacteria and suppressed the growth of potential bacterial pathogens. These findings suggested CNP supplementation improved the growth performance and immune status, alleviated immunological stress and regulated intestinal ecology in weaned piglets. Based on these beneficial effects, CNP could be applied as a functional feed additives supplemented in piglets diet.
Effects of diets with different amino acid release characteristics on the gut microbiota and barrier function of weaned pigs

Abstract:Background: The absorption and utilization of proteins by animals is affected by the amino acid (AA) release characteristics of their diets. In the present study, we aimed to determine the effects of diets with various amino acid release characteristics on the intestinal barrier function and diversity of gut microbiota of weaned pigs. Results: Forty-eight pigs (7.45 ± 0.58 kg) were fed with diets having different amino acid release characteristics during a period of 28 days. We used a 2 × 3 full-factor (two protein levels and three protein sources with differing amino acid release characteristics) experimental design, with normal (standard terminal ileal digestibility of 17.5%) or low (standard terminal ileal digestibility of 14.9%) protein levels as the first factor. Casein (CAS), corn gluten meal (CGM) and a MIX diet were used as protein sources. Due to the more balanced release of amino acids, the diamine oxidase (DAO) concentrations in the CAS and MIX groups were significantly lower than those in the CGM group (P < 0.05); Reducing the dietary protein content from 17.5% to 14.9% had no significant effects on the levels of serum DAO or D-lactic acid. By contrast, it increased the microbial diversity (chao1 and ACE values) and the number of Lactobacillus in the jejunum (P < 0.05). The CAS-containing diet and the MIX diet resulted in significantly higher microbial diversity (Simpson and Shannon) than the CGM-containing diet in the jejunum. Conclusion: The balanced release of amino acids in CAS and MIX diets maintained intestinal barrier function and increased gut microbiota diversity. These findings could potentially provide a scientific reference for the rational preparation of piglet feed.
Xylo-oligosaccharides improve the adverse effects of plant-based proteins on weaned piglet health by maintaining the intestinal barrier and inhibiting harmful bacterial growth

Abstract:Introduction: Piglets are more susceptible to weaning stress syndrome when fed high levels of plant-based proteins that contain abundant food antigens and anti-nutritional factors. Xylo-oligosaccharides (XOS) are a potential prebiotic that may improve the tolerance of weaned piglets to plant-based proteins. The aim of this study was to investigate the effects of XOS supplementation in high and low plant-based protein diets on growth performance, gut morphology, short-chain fatty acid (SCFA) production, and gut microbiota of weaned piglets. Methods: A total of 128 weanling piglets with an average body weight (BW) of 7.63 ± 0.45 kg were randomly allocated to one of the four dietary treatments in a 2 × 2 factorial arrangement, with two levels of plant-based proteins (d 1–14: 68.3 or 81.33%, d 15–28: 81.27 or 100%) and XOS complex (0 or 0.43%) over a 28-day trial. Results: The growth performance of piglets did not differ significantly among groups ( P > 0.05). However, the diarrhea index of weaned piglets fed a high plant-based protein diet (HP) was significantly higher than that of those fed a low plant-based protein diet (LP) at days 1–14 and throughout the experimental period ( P < 0.05). XOS treatment tended to reduce the diarrhea index at days 1–14 ( P = 0.062) and during the whole experiment period ( P = 0.083). However, it significantly increased the digestibility of organic matter at days 15–28 ( P < 0.05). Moreover, dietary XOS supplementation increased ileal mucosa mRNA expression of occludin and ZO-1 ( P < 0.05). Furthermore, the concentration of butyric acid (BA) in the cecal contents and in the concentrations of BA and valeric acid (VA) in colon contents were significantly elevated in the XOS groups ( P < 0.05). Additionally, XOS optimized the gut flora by lowering the number of pathogenic bacteria such as p_Campylobacterota , thereby stabilizing the gut ecosystem. Discussion: In conclusion, the HP diet aggravated diarrhea in weaned piglets while the XOS diet alleviated it by improving nutrient digestibility, protecting intestinal morphology, and optimizing the gut flora.

Abstract:PurposeDietary protein, as important macronutrient, is vital for intestinal function and health status. We aimed to determine the effects of dietary protein source on growth performance and intestinal function of neonates with intrauterine growth retardation (IUGR) in a pig model.MethodsEighteen pairs of IUGR and normal birth weight (NBW) weaned pigs were allotted to be fed starter diet containing soybean protein concentrate (SPC) or spray-dried porcine plasma (SDPP) for 2 weeks. Growth performance, antioxidant variables, intestinal morphology and absorption capability, microbiota composition and short-chain fatty acids (SCFA) were assessed.ResultsIUGR led to poor growth performance, absorption capability and changes on antioxidant variables, while SDPP diet improved the growth performance, diarrhea index, intestinal morphology and antioxidant variables of IUGR or NBW pigs relative to that fed SPC diet. Importantly, SDPP diet improved bacterial diversity and increased the abundance of phylum Firmicutes, but decreased the phylum Proteobacteria in colonic digesta, associating with higher genera Lactobacillus and lower genera Escherichia–Shigella, linking to the increased concentration of SCFA.ConclusionsOur findings indicate that IUGR impairs the growth rate, intestinal function and oxidative status of weaned pigs, which could be partly improved by SDPP diet either for IUGR or NBW pigs, associating with the better antioxidant capability, composition of microbiotas and their metabolites.

Abstract:To the Editor: In March 2008, nicotine patches (15 mg per 16 hours) were authority-listed for Aboriginal and Torres Strait Islander people by the Pharmaceutical Benefits Advisory Committee (PBAC) as part of efforts to improve access to medications.1,2 Although timely and welcome, the decision to list only 15 mg patches should be revisited, as it is not consistent with current smoking cessation clinical guidelines.3-5 Neither is it consistent with evidence from a 2008 tobacco survey that we conducted in remote Aboriginal communities in Arnhem Land, Northern Territory.

Abstract:ABSTRACT This study was to investigate the effects of peptides derived from fermented soybean meal (FSP) on intestinal inflammation and epithelial barrier function of the piglets. Compared to fed soybean meal, piglets fed with fermented soybean meal (FSBM) exhibit lower serum levels of IL-6, IL-1β and D-lactate, and reduced ileum macrophage infiltration. Besides, the FSBM up-regulated (p < 0.05) expression of mucins and tight junction (TJ) proteins in the jejunum and ileum of piglets. The FSP were prepared with alkali extraction and a 3-kDa molecular weight cut-off membrane system. In vitro, FSP reducing the mRNA expression of IL-6, IL-1β, IL-8 and attenuated Escherichia coli K88-induced inflammation in the intestinal porcine epithelial cells-J2 (IPEC-J2). Furthermore, FSP increased (p < 0.05) the expression of TJ proteins and transepithelial electrical resistance in IPEC-J2 cells. These results suggest that FSP could improve small intestinal health by enhancing epithelial barrier function and suppressing inflammation.
Effects of soybean meal fermented by L. plantarum, B. subtilis and S. cerevisieae on growth, immune function and intestinal morphology in weaned piglets

Abstract:BackgroundThe present study compared the effects of soybean meal fermented by three different probiotics organisms with non-fermented soybean meal on growth performance, serum parameters, immune chemistry and intestinal morphology in weaned piglets.MethodsOne hundred and forty-four 35-day old crossbred (Duroc × Landrace × Yorkshire) piglets were randomly allocated into four different dietary treatments (n = 36 per group) containing 0, 5, 10 and 15% fermented soybean meal.ResultsThe piglets fed fermented soybean meal showed an increase (p < 0.05) in average daily weight gain and a reduction in feed consumption (p < 0.05).The piglets fed 10 and 15% fermented soybean meal showed the greatest growth improvement with higher levels of serum alkaline phosphatase and total serum proteins. Serum urea nitrogen in the experimental group was significantly lower than control whereas serum IgG, IgM and IgA levels were all significantly higher. Moreover, villus height in the duodenum, jejunum, and ileum was significantly higher (p < 0.05) and the crypt depth was significantly lower (p < 0.05). The levels of the autophagy factor LC3B in piglets showed a downward trend in the jejunum and ileum compared to control.ConclusionsFermented soybean meal could significantly improve the growth, immune function and intestinal health in weaned piglets, and the best effective benefits showed in 10% FSBM group.

Abstract:Using protein-restriction diets becomes a potential strategy to save the dietary protein resources. However, the mechanism of low-protein diets influencing pigs’ growth performance is still controversial. This study aimed to investigate the effect of protein-restriction diets on gastrointestinal physiology and gut microbiota in pigs. Eighteen weaned piglets were randomly allocated to three groups with different dietary protein levels. After a 16-week trial, the results showed that feeding a low-protein diet to pigs impaired the epithelial morphology of duodenum and jejunum (p < 0.05) and reduced the concentration of many plasma hormones (p < 0.05), such as ghrelin, somatostatin, glucose-dependent insulin-tropic polypeptide, leptin, and gastrin. The relative abundance of Streptococcus and Lactobacillus in colon and microbiota metabolites was also decreased by extreme protein-restriction diets (p < 0.05). These findings suggested that long-term ingestion of a protein-restricted diet could impair intestinal morphology, suppress gut hormone secretion, and change the microbial community and fermentation metabolites in pigs, while the moderately low-protein diet had a minimal effect on gut function and did not impair growth performance.

Abstract:This study aimed to investigate the effects of fermented corn–soybean meal mixed feed (FMF) on growth performance, intestinal barrier function, gut microbiota and short-chain fatty acids in weaned piglets. A total of 128 weaned piglets [Duroc×(Landrace×Yorkshire), male, 21-day-old] were randomly allocated to four groups. Piglets were fed a control diet (CON) or the control diet supplemented with 10%, 50% or 100% FMF (FMF-10, FMF-50 or FMF-100, respectively) for 14 d. The results showed that the FMF-100 group had higher average daily gain and average daily feed intake and lower diarrhea incidence than the CON group (p < 0.05). The FMF-50 and FMF-100 groups had greater villus height in the duodenum and jejunum, and the FMF-10 and FMF-100 groups had higher villus height-to-crypt depth ratio in the duodenum and jejunum than the CON group. Additionally, the FMF-100 group had higher protein expression of duodenal, jejunal and ileal ZO-1 and jejunal claudin-1; higher mRNA expression of duodenal and ileal TJP1 and jejunal CLDN1 and IL10; and lower jejunal IL1B mRNA expression (p < 0.05). The FMF-50 group showed higher jejunal ZO-1 and claudin-1 protein levels, higher mRNA expression levels of IL10 and TJP1 and lower levels of TNF in the jejunum; the FMF-10 group had higher mRNA expression levels of IL10 and lower levels of TNF in the jejunum than the CON group (p < 0.05). Furthermore, the FMF-10 and FMF-50 groups had higher colonic Lactobacillus abundance and butyrate levels; the FMF-100 group had higher abundance of colonic butyrate, Lactobacillus and Faecalibacterium than the CON group (p < 0.05). Collectively, our results suggest that FMF could improve intestinal mucosal barrier function, gut microbiota and their metabolites, thereby enhancing average daily gain and reducing diarrhea incidence in weaned piglets.

Abstract:A 4-week study was conducted to evaluate the effects of dietary crude protein (CP) content and resistant starch (RS) supplementation on growth performance, intestinal histomorphology and microbial metabolites of weaned pigs. A total of 96 pigs (7.06 ± 0.45 kg body weight) were assigned to 1 of 4 diets in a randomised complete block design involving a 2 (CP levels) × 2 (without or with RS) factorial arrangement to give 8 replicate pens and 3 pigs per pen. Body weight and feed disappearance were recorded weekly, and the faecal consistency score was determined every morning. Blood was sampled on days 1, 14 and 28 from one pig per pen, and the same pig was euthanised on day 28 to collect ileal tissue and ileal and colon digesta. Data were analysed using the MIXED procedure of SAS. The average daily gain and gain:feed ratio were lower (p < 0.05) in pigs fed low crude protein (LCP) diets compared to those fed high CP (HCP) diets during week 3 and overall period. The analysed Lys, Met+Cys and Thr in feed were lower than calculated values, particularly in LCP diets, which may have affected performance. Pigs fed the LCP diets had longer (p < 0.05) ileal villi and higher villus height to crypt depth ratios than those fed the HCP diets, and RS supplementation increased (p < 0.05) ileal villus height. Interactions (p < 0.05) between dietary CP content and RS inclusion were observed for short-chain fatty acid concentration in the ileum and colon in phase 2. There was no difference in propionic acid (ileum) or butyric acid (colon) concentrations among pigs fed HCP diets, however, the butyric acid concentration increased in pigs fed the LCP diet when supplemented with RS. Reducing dietary CP lowered (p < 0.05) faecal score, plasma urea nitrogen and digesta ammonia content. Overall, feeding LCP diets reduced growth performance but improved gut morphology in weaned pigs. Feeding the LCP diet with RS supplementation modulated concentrations of ileal propionic acid and colonic butyric acid in weaned pigs.

Abstract:This experiment aimed to investigate the effects of a symbiotic, consisting of microencapsulated Lactobacillus plantarum (MLP) and fructooligosaccharide (FOS) on growth performance, blood immune parameters, and intestinal microbiota in weaned piglets. Ninety weaned piglets were assigned to three dietary groups (CON: basal diet; ANT: basal diet + aureomycin; SYN: basal diet + MLP and FOS) for a four-week trial. Compared to CON, pigs in the SYN group had higher weight gain and feed intake, and lower diarrhea rate (P < .05). Also, pigs in the SYN group had improved plasma IgA and IgG concentrations, and increased (P < .05) numbers of lactic acid bacteria in the colon compared to CON. In conclusion, feeding a synbiotic based on MLP and FOS had beneficial effects on growth performance, plasma immune parameters, and intestinal microbiota, indicating the potential of it to serve as an alternative to feed antibiotics in weaned pig diets.

Abstract:Fermented corn-soybean meal (fermented feed, FF) is commonly used in swine production, but the effects of FF on gut health remain unclear. In this study, serum immunity, mRNA abundances of antimicrobial peptides (AMPs) and Toll-like receptors (TLR1-9), bacterial abundance in the duodenum and colon, and colonic metabolic phenotypes were determined in crossbred barrows (Duroc × Landrace × Large White) fed FF or normal feed (unfermented feed, UF) (n = 6). When compared to the UF group, the results showed that serum levels of IgG and IgM were significantly increased in FF group pigs (P < 0.05). FF significantly decreased the abundances of Bacteroides and Verrucomicrobia in the duodenum and decreased the abundances of Bacteroides, Proteobacteria, and Verrucomicrobia in the colon while it significantly increased the abundances of Firmicutes and Actinobacteria (P < 0.05). Furthermore, a Spearman's correlation analysis showed that serum immunity and the expression of genes related to gut immunity were associated with bacterial strains at the family level. Moreover, differentially abundant colonic microbiota were associated with colonic metabolites. LC-MS data analyses identified a total of 1,351 metabolites that markedly differed between the UF and FF groups. C5-Branched dibasic acid metabolism was significantly upregulated whereas the purine metabolism was significantly downregulated (P < 0.05) in the colonic digesta of pigs in the FF meal group compared to the UF meal group. Collectively, these results indicated that FF meal could influence serum immunity and the expression of genes related to gut immunity, correlating with the gut microbiota and bacterial metabolites in grower-finisher pigs. This study may provide an alternative strategy for improving the intestinal health of grower-finisher pigs.
