Abstract:In this work, the suitability of gas chromatography-mass spectrometry (GC-MS) and liquid chromatography-tandem mass spectrometry (LC-MS/MS) for the multi-class determination of different families of neutral per- and polyfluoroalkyl substances (PFASs), such as fluorotelomer olefins (FTOs), alcohols (FTOHs) and fluorooctanesulfonamides (FOSAs) and sulfonamido-ethanols (FOSEs), was investigated and compared. Regarding GC-MS, the use of a semi-polar GC column (DB-624, 6%-cyanopropilphenyl 94%-dimethyl polysiloxane) allowed the adequate separation of all the compounds while chemical ionisation (CI) of positive ions as ionisation technique provided the best responses. Concerning UHPLC-MS/MS, atmospheric pressure chemical ionisation (APCI) and photoionisation (APPI) sources allowed the ionisation of all studied neutral PFASs, including FTOs for the first time. High vaporizer temperatures (450 °C) and acetonitrile/water mobile phase mixtures were required to favour the ionisation of FTOs, with adequate ionisation for FTOHs, FOSAs and FOSEs. The chromatographic separation, performed on a totally porous column (Luna C18), allowed the successful separation of the four families of neutral PFASs. After comparing the performance of the studied methods, the highest detectability was achieved using UHPLC-APCI-MS/MS and it was chosen in combination with a solid-phase extraction (SPE) procedure for the analysis of neutral PFASs in water samples. The whole method provided low limits of detection (0.003-6 µg L-1), good precision (RSD < 9%) and trueness (relative error < 10%). The methodology was applied to the analysis of river water samples and the presence of some neutral PFASs were detected (8:2 FTO) and quantified (4:2 FTOH and N-EtFOSA) at low concentration levels (ng L-1).

Abstract:Fluoro-solid phase extraction (fluoro-SPE) has been well established to selectively extract targets after they have been fluoro-tagged. Here we report that this technique can also be used to selectively extract fluorosurfactants of PFAS (per- and poly-fluoroakyl substances), which have fluoro-carbon skeletons, from other anionic surfactants that have no fluoro-carbon skeletons. We advance the fluoro-SPE protocol to enable the extraction of fluorosurfactants using 4-5 times (in volume) more solvent for fluorophilic eluting in effort to overcome the strong interaction between the fluorosurfactants and the fluoro-Gel. Based on a smartphone-based app we developed recently to assess the colouration of fluorosurfactants with a dye (ethyl violet) that is similar to MBAS (methylene blue active substances), plus an SPE for pre-concentration and fluoro-SPE for pre-selection, we successfully achieve a detection limit of 1 ppb for the PFAS including perfluorooctanoic acid (PFOA), perfluorooctanesulfonic acid (PFOS) and 6:2 fluorotelomer sulfonate (6:2FTS), which is encouraging and can be conducted in a general laboratory.
Simultaneous characterization of perfluoroalkyl carboxylate, sulfonate, and sulfonamide isomers by liquid chromatography-tandem mass spectrometry

Abstract:A comprehensive method was developed to simultaneously separate and detect perfluorinated acid (PFA) and PFA-precursor isomers using liquid chromatography-tandem mass spectrometry (LC-MS/MS). A linear perfluorooctyl stationary phase and acidified mobile phase increased separation efficiency, relative to alkyl stationary phases, for the many perfluoroalkyl carboxylate (PFCA), perfluoroalkyl sulfonate (PFSA), and perfluorooctyl sulfonamide (PFOSA) isomers and in combination with their distinct MS/MS transitions allowed full resolution of most isomers in standards. Utilizing the absence of the "9-series" and "0-series" product ions, several perfluorooctane sulfonate (C8F17SO3-, PFOS) isomers were structurally elucidated. In human serum, only perfluorooctane sulfonamide (C8F17SO2NH2, FOSA) and PFOS consisted of significant quantities of branched isomers, whereas PFCAs were predominantly linear. Interferences that coelute with the m/z 499 --> 80 transition of PFOS on alkyl stationary phases were simultaneously separated and identified as taurodeoxycholate isomers, removal of which permitted the use of the more sensitive m/z 80 product ion and a resulting 20-fold decrease in PFOS detection limits compared to the m/z 499 --> 99 transition (0.8 pg versus 20 pg using m/z 80 and 99, respectively). Interferences in human serum which caused a 10-20-fold over-reporting of perfluorohexane sulfonate (C6F13SO3-, PFHxS) concentrations on alkyl stationary phases were also simultaneously separated from linear PFHxS and identified as endogenous steroid sulfates. PFOSA isomers, generated with human microsomes, had different rates of metabolism, suggesting that the perfluoroalkyl branching pattern may affect the biological properties of individual isomers. This fact, and for reasons of improved accuracy and sensitivity, investigators are urged to utilize more efficient separation methods capable of isomer characterization in perfluoroalkyl research.

Abstract:The scientific foundation for per- and polyfluoroalkyl substances (PFAS) measurements in water, soils, sediments, biosolids, biota, and outdoor air has rapidly expanded; however, there are limited efforts devoted to developing analytical methods to measure vapor-phase PFAS in indoor air. A gas chromatography-tandem mass spectrometry (GC-MS/MS) method coupled with thermal desorption (TD) sorbent tube analysis was developed to quantify trace levels of fluorotelomer alcohols (FTOHs) emitted from consumer products in the indoor environment. Method evaluation included determination of instrument detection limits (IDLs), quality assurance checks of target standards purchased from different vendors, sample loss during storage, and TD sorbent breakthrough with tubes coupled in-series. The IDLs for TD-GC-MS/MS analyses ranged from 0.07 – 0.09 ng/tube. No significant loss of FTOHs was observed during stability tests over 28 days with relative standard deviations (RSDs) of spiked TD tubes ranging from 3.1 – 7.7% and the RSDs of polypropylene copolymer vial storage of standard solutions ranging from 4.3 – 8.4%. TD tube breakthrough was minimal with recovered FTOHs in the second tubes <1% of the spiked concentrations in the first tubes with carrier gas volume up to 20 L. The method has been applied to determine FTOH emissions from three consumer products in micro-scale chambers. A liquid stone cleaner/sealer product contained the highest levels of 6:2, 8:2, and 10:2 FTOHs, while the mattress pad products contained lower levels of 8:2 and 10:2 FTOHs. The emission parameters, including the initial emission factors and first order decay rate constants, were obtained based on the experimental data. The developed methods are sensitive and specific for analysis of all four target FTOHs (4:2, 6:2, 8:2, 10:2 FTOHs) with chamber testing. The methods can be extended to indoor air sampling and could be applicable to ambient air sampling.

Abstract:Fluorotelomer alcohols (FTOHs) are the main precursors of environmentally ubiquitous perfluorinated acids, and determination of FTOHs at low concentrations presents significant challenges. In this study, a new liquid chromatography–electrospray mass spectrometry (LC–ESI-MS) method in conjunction with low-energy collision dissociation tandem mass spectrometry (CID-MS/MS) was developed by employing an optimized derivatization reaction with dansyl chloride (DNS) in acetonitrile under catalysis of 4-(dimethylamino)-pyridine (DMAP). The instrument detection limits (IDLs) of the newly developed method were 0.014, 0.015, 0.014, 0.0075 and 0.0093μg/L for 4:2 FTOH, 6:2 FTOH, 8:2 FTOH, 10:1 FTOH and 10:2 FTOH respectively, which were 7.5–241 times lower than those without derivatizaiton and 57–357 times lower than previous GC/MS method. The method was successfully applied to analyze FTOHs in sediments combined with WAX and silica cartridges cleanup. The overall method recoveries were from 67±6.0% to 83±9.4% with matrix effects of <15%. The limits of quantification for all FTOHs were 0.017–0.060ng/gdry weight (dw). The method was applied to analyze six marine sediment samples from Liaodong Bay, China. All FTOHs except for 10:1 FTOH were detected, and the total concentrations of FTOHs were 0.19–0.52ng/gdw. The developed method provides a new method to sensitively determine FTOHs in environmental matrices.

Abstract:The mass spectral characteristics of perfluorooctane sulfonate (PFOS, C(8)F(17)SO(3)-) isomers present in technical PFOS were obtained using high-resolution gas chromatography (HRGC) combined with mass spectrometry (MS). To make PFOS amenable to HRGC separation, a simple derivatization procedure was developed. The method involved the conversion of PFOS into the iso-propyl ester using iso-propanol as the derivatization reagent under acidic conditions. Mass spectra were generated employing electron ionization (EI) and negative chemical ionization (NCI). Interpretation of fragment ions was possible due to the use of deuterium-labeled iso-propanol as derivatization reagent, which induced mass shifts in the electron ionization (EI) and negative chemical ionization (NCI) mass spectra. HRMS allowed the accurate mass measurement of important EI fragments and confirmed the derivatization reaction as well as the proposed fragmentation pathway involving rearrangement. Moreover, the high resolution provided by HRGC enabled the separation of eleven PFOS isomers present in the technical product. This is an improvement over the previously reported high-performance liquid chromatography (HPLC) separation. A complete identification of all isomers was not possible due to lack of pure reference materials. Finally, the developed derivatization procedure was successfully applied to perfluoroalkyl carboxylates (PFCA) and corresponding fragmentation involving rearrangement of the derivatized PFCA was observed. The described qualitative derivatization offers a promising alternative technique for the separation and identification of isomers of perfluoroalkyl sulfonates and carboxylates by HRGC/MS.

Application of High-Resolution Mass Spectrometry to Evaluate UV-Sulfite-Induced Transformations of Per- and Polyfluoroalkyl Substances (PFASs) in Aqueous Film-Forming Foam (AFFF)

Abstract:UV-sulfite has been shown to effectively degrade per- and polyfluoroalkyl substances (PFASs) in single-solute experiments. We recently reported treatment of 15 PFASs, including perfluoroalkyl sulfonic acids (PFSAs), perfluoroalkyl carboxylic acids (PFCAs), and fluorotelomer sulfonic acids (FTSs), detected in aqueous film-forming foam (AFFF) using high-resolution liquid chromatography quadrupole time-of-flight mass spectrometry (LC-QTOF-MS) targeted analysis. Here, we extend the analysis within those original reaction solutions to include the wider set of PFASs in AFFF for which reactivity is largely unknown by applying recently established LC-QTOF-MS suspect screening and semiquantitative analysis protocols. Sixty-eight additional PFASs were detected (15 targeted + 68 suspect screening = 83 PFASs) with semiquantitative analysis, and their behavior was binned on the basis of (1) detection in untreated AFFF, (2) PFAS photogeneration, and (3) reactivity. These 68 structures account for an additional 20% of the total fluorine content in the AFFF (targeted + suspect screening = 57% of total fluorine content). Structure-reactivity trends were also revealed. During treatment, transformations of highly reactive structures containing sulfonamide (-SO2N-) and reduced sulfur groups (e.g., -S- and -SO-) adjacent to the perfluoroalkyl [F(CF2)n-] or fluorotelomer [F(CF2)n(CH2)2-] chain are likely sources of PFCA, PFSA, and FTS generation previously reported during the early stages of reactions. The results also show the character of headgroup moieties adjacent to the F(CF2)n-/F(CF2)n(CH2)2- chain (e.g., sulfur oxidation state, sulfonamide type, and carboxylic acids) and substitution along the F(CF2)n- chain (e.g., H-, ketone, and ether) together may determine chain length-dependent reactivity trends. The results highlight the importance of monitoring PFASs outside conventional targeted analytical methodologies.

Abstract:Volatile per- and polyfluoroalkyl substances (PFASs) are often used as precursors in the synthesis of nonvolatile PFASs. The volatile PFASs, which include the perfluoroalkyl iodides (PFAIs), fluorotelomer iodides (FTIs), fluorotelomer alcohols (FTOHs), fluorotelomer olefins (FTOs), fluorotelomer acrylates (FTACs), and fluorotelomer methacrylates (FTMACs), are often produced starting from the telomerization process. These volatile compounds can be present in the air and water environment and can be transformed into highly persistent perfluoroalkyl carboxylic acids. With the exception of FTOHs, which are well studied, the determination of other volatile PFASs is also of prime importance in studying the sources and fate of PFASs. In this study, a method was developed to determine representative precursor compounds that included PFAIs, FTIs, FTOs, FTACs, and FTMACs in wastewater treatment plant (WWTP) air and water samples. The sampling and sample preparation step involved the use of solid-phase extraction (SPE) cartridges with HLB™ material to enrich the analyte. Gas chromatography with mass spectrometry was employed for the detection and quantification of the analytes. Method validation results showed high linearity and sensitivity in the positive electron ionization-selected ion monitoring mode (+EI-SIM). The absolute instrumental limits of detection were in the range of 0.5 to 2 pg. The method detection limit (MDL) in air was 1 ng/m3 with the exception of the FTACs which could be only be detected at concentrations higher than 40 ng/m3. The MDL in water was 10 ng/L. Direct spiking of the cartridges and analyte introduction by volatilization from the glass surface onto the SPE material had recoveries between 86 and 100%. The volatile PFASs were shown to readily partition into the air rather than into water. Consequently, large losses in the amount of PFASs were observed when these were spiked into the water.Graphical abstractWastewater treatment plant air and water samples were passed through HLB™ solid-phase materials. Abstract:A rapid (<23 min) new HPLC-MS/MS method was developed for simultaneous characterization of 24 per- and polyfluoroalkyl compounds in landfill leachate. In addition to isomer-specific analysis of perfluorooctane sulfonate and perfluorooctanoate, branched from linear isomer separation was accomplished for C6 and C10 perfluoroalkyl sulfonates, C6, C7 and C9-C11 perfluoroalkyl carboxylates, perfluorooctane sulfonamide and, for the first time, 3 perfluorooctane sulfonamidoacetates. The method utilizes a fused-core pentafluorophenylpropyl (PFP) stationary phase and is approximately 4 times faster than previous comprehensive isomer-specific HPLC-MS/MS methods. This is the first isomer-specific methodology which can be adopted for routine analysis without sacrificing throughput from lengthy run times or limited target lists.

Abstract:Per- and polyfluoroalkyl substances (PFAS) have caused widespread environmental concern in recent years. Among them, the levels of perfluoroalkane sulfonyl fluorides (PFASFs) in the environment have rarely been reported due to the lack of sensitive analytical methods. Herein, a novel liquid chromatography-microwave plasma torch ionization-mass spectrometry (LC-MPTI-MS) technique was designed for the direct analysis of PFASFs in the environment. The collaborative action of reactive oxygen species (such as hydroxyl radicals) and the elevated temperature within the ambient MPTI environment results in the replacement of the fluorine atom in sulfonyl fluoride by oxygen, leading to the detection of perfluoroalkanesulfonic acid (PFSA) ions by MS. Concurrently, LC was employed to separate other PFSAs that are present in the environment. Three PFASFs exhibited good linearity within the range of 1-500 mu g/L with R-2 > 0.994. The limit of detections (LODs) and the limit of quantifications (LOQs) were measured at 39.32-87.87 and 131.07-292.90 ng/L, respectively. The method was utilized for the direct detection of spiked perfluorooctane sulfonyl fluoride (PFOSF) in wastewater with recoveries of 77.16 to 124.81%. Our approach circumvents the laborious process of chemical derivatization and is anticipated to serve as a robust tool for determining the levels and behaviors of PFASFs in the environment.
Molecular detection of per- and polyfluoroalkyl substances in water using time-of-flight secondary ion mass spectrometry

Abstract:Detection of per- and polyfluoroalkyl substances (PFASs) is crucial in environmental mitigation and remediation of these persistent pollutants. We demonstrate that time-of-flight secondary ion mass spectrometry (ToF-SIMS) is a viable technique to analyze and identify these substances at parts per trillion (ppt) level in real field samples without complicated sample preparation due to its superior surface sensitivity. Several representative PFAS compounds, such as perfluorooctanesulfonic acid (PFOS), perfluorobutanoic acid (PFBA), perfluoropentanoic acid (PFPeA), perfluoheptanoic acid (PFHpA), and perfluorononanoic acid (PFNA), and real-world groundwater samples collected from monitoring wells installed around at a municipal wastewater treatment plant located in Southern California were analyzed in this work. ToF-SIMS spectral comparison depicts sensitive identification of pseudo-molecular ions, characteristic of reference PFASs. Additionally, principal component analysis (PCA) shows clear discrimination among real samples and reference compounds. Our results show that characteristic molecular ion and fragments peaks can be used to identify PFASs. Furthermore, SIMS two-dimensional (2D) images directly exhibit the distribution of perfluorocarboxylic acid (PFCA) and PFOS in simulated mixtures and real wastewater samples. Such findings indicate that ToF-SIMS is useable to determine PFAS compounds in complex environmental water samples. In conclusion, ToF-SIMS provides simple sample preparation and high sensitivity in mass spectral imaging, offering an alternative solution for environmental forensic analysis of PFASs in wastewater in the future.

Abstract:A procedure for the determination of perfluorocarboxylic acids (i.e. PFC7-10A) in sediment by pressurized fluid extraction (PFE), derivatization, headspace solid-phase microextraction and GC-MS determination in the negative ion chemical ionisation mode was developed. The PFE extraction variables such as solvent composition, number and time per extraction cycle, and extraction temperature were optimised. In the optimum extraction conditions, recoveries exceeding 95% with a limit of detection and RSDs of 0.5-0.8 ng g(-1) and 15.5-16.8%, respectively, were obtained. The developed analytical procedure was applied to harbour sediments where PFC8A and PFC10A were detected for the first time at low ppb concentrations (i.e. 8-11 ng g(-1).

Abstract:To date, poly- and perfluoroalkyl substances (PFAS) represent a real threat for their environmental persistence, wide physicochemical variability, and their potential toxicity. Thus far a large portion of these chemicals remain structurally unknown. These chemicals, therefore, require the implementation of complex non-targeted analysis workflows using liquid chromatography coupled with high-resolution mass spectrometry (LC-HRMS) for their comprehensive detection and monitoring. This approach, even though comprehensive, does not always provide the much-needed analytical resolution for the analysis of complex PFAS mixtures such as fire-fighting aqueous film-forming foams (AFFFs). This study consolidates the advantages of the LC×LC technique hyphenated with high-resolution tandem mass spectrometry (HRMS/MS) for the identification of PFAS in AFFF mixtures. A total of 57 PFAS homolog series (HS) were identified in 3M and Orchidee AFFF mixtures thanks to the (i) high chromatographic peak capacity ( n' 2D,c ~ 300) and the (i) increased mass domain resolution provided by the "remainder of Kendrick Mass" (RKM) analysis on the HRMS data. Then, we attempted to annotate the PFAS of each HS by exploiting the available reference standards and the FluoroMatch workflow in combination with the RKM defect by different fluorine repeating units, such as CF 2 , CF 2 O, and C 2 F 4 O. This approach resulted in 12 identified PFAS HS, including compounds belonging to the HS of perfluoroalkyl carboxylic acids (PFACAs), perfluoroalkyl sulfonic acids (PFASAs), (N-pentafluoro(5)sulfide)-perfluoroalkane sulfonates (SF5-PFASAs), N -sulfopropyldimethylammoniopropyl perfluoroalkane sulfonamides (N-SPAmP-FASA), and N -carboxymethyldimethylammoniopropyl perfluoroalkane sulfonamide (N-CMAmP-FASA). The annotated categories of perfluoroalkyl aldehydes and chlorinated PFASAs represent the first record of PFAS HS in the investigated AFFF samples.

Abstract:To date, poly- and perfluoroalkyl substances (PFASs) represent a real threat for both their environmental persistence, wide physicochemical variability, and their potential toxicity. Thus far a large portion of these chemicals remain structurally unknown. These chemicals, therefore, require the implementation of complex non-targeted analysis workflows using high resolution mass spectrometry coupled with liquid chromatography (LC-HRMS) for their comprehensive detection and monitoring. This approach, even though comprehensive, does not always provide the much needed analytical resolution for the analysis of complex PFAS mixtures such as fire-fighting aqueous-film forming foams (AFFFs). This study consolidates the advantages of LC×LC technique hyphenated with high-resolution tandem mass spectrometry (HRMS/MS) for the identification of PFASs in AFFF mixtures. A total of 57 PFAS homologue series (HSs) were identified in 3M and Orchidee AFFF mixtures thanks to the (i) high chromatographic peak capacity (n’2D,c~300) and the (i) increased mass domain resolution provided by the “remainder of Kendrick Mass” (RKM) analysis on the HRMS data. Then, we attempted to annotate PFASs of each HSs exploiting the available reference standards and the FluoroMatch workflow in combination with the RKM defect by different fluorine repeating units, such as CF2, CF2O, and C2F4O. This approach resulted in 12 identified PFAS HSs, including compounds belonging to the HSs of PFACAs, PFASAs, SF5-PFASAs, N-SPAmP-FASA, and N-CMAmP-FASA. The annotated categories of perfluoroalkyl aldehydes, and chlorinated PFASAs represent a first record of PFAS HSs in the investigated AFFF samples.

Tandem Mass Spectrometry of Perfluorocarboxylate Anions: Fragmentation Induced by Reactive Species Formed From Microwave Excited Hydrogen and Water Plasmas

Abstract:ABSTRACT Rationale Polyfluoroalkyl substances (PFAS) like perfluorooctanoic acid have persistent environmental and physiological effects. This study investigates the degradation of C n F 2n+1 CO 2 − ( n = 1–7) with neutral radical fragmentation under oxygen attachment dissociation (OAD). Unique fragments absent from collision‐induced dissociation (CID) are observed. Further, potential mechanisms are uncovered by density functional theory (DFT) calculations. Methods From a standard mixture of PFAS, straight‐chain perfluorinated carboxylic acids with carbon chain lengths of one to eight were separated via liquid chromatography and transferred to the gas phase via negative‐mode electrospray ionisation. Each C n F 2n+1 CO 2 − of interest was mass selected and fragmented via both CID and OAD in a quadrupole time‐of‐flight mass spectrometer. DFT optimisations of structures were performed at M06/6‐31+g(d), and single point energy calculations were performed at M06‐2X/aug‐cc‐pVTZ for C 3 F 7 CO 2 − . Results Decarboxylation was observed from both CID and OAD, but fluorine abstraction and hydroxyl addition only occurred with OAD. The DFT calculations suggest that C 3 F 6 –• ( m / z 150) is most likely formed from by H• attack onto a β‐ C‐F bond, then loss of HF, finally decarboxylation. Further, C 3 F 5 O − ( m / z 147) likely arises from C 3 F 6 –• recombining with OH• to produce energised C 3 F 6 OH − ions, followed by α‐ or β‐ elimination of HF to give enolate and/or epoxide‐type products. Conclusions OAD of C 3 F 7 CO 2 − yields unique product ions C 3 F 6 –• ( m / z 150) and C 3 F 5 O − ( m / z 147) absent from collision‐induced dissociation. DFT calculations suggest an intricate pathway of H• attack onto a β C‐F bond, then loss of HF, decarboxylation, recombination with OH•, and finally α‐ or β‐ elimination of HF to give the products.

Abstract:A method is reported for the sensitive, simultaneous determination of mono- (MFA), di- (DFA), and trifluoroacetates (TFA) by ion chromatography (IC). These species were separated using a Dionex AS17 anion-exchange column employed with a potassium hydroxide gradient (via a Dionex EG40 eluent generator) and suppressed conductivity detection. The fluoroacetates were successfully separated from a range of inorganic and organic species likely to be present in environmental samples, in a total analysis time of 35min (including re-equilibration of the column). Detection limits for mono-, di- and trifluoroacetate were 21, 38 and 36μg/l, respectively, determined using a signal-to-noise ratio of 3, and were obtained using a sample injection volume of 50μl. Precision was less than 0.83% relative standard deviation (RSD) for replicate injections performed over a period of 30 days. The method was applied to the determination of monofluoroacetate in river water samples and also in carrot baits.

Abstract:Perfluorooctanesulfonate (PFOS) is a common environmental pollutant and has been detected in various environmental media. Several major isomers have been identified, and their profiles provide important signatures from the source of contamination. Gas chromatography has a significant resolving power for the isomers. However, methods for derivatizing sulfonates for a sensitive analysis are limited. In this study, in-port derivatization with diaryl iodonium salts into aryl sulfonates was used. A technical mixture of PFOS was separated on gas chromatography to afford 11 isomers. In electron-capture negative ionization, the PFOS derivatives form specific fragmented anions (sulfinate anions). The derivatization was also applied to other per- or polyfluoroalkyl acids (PFASs; 13 perfluoroalkyl carboxylic acids and 10 sulfonates). Analysis of human serum samples was achieved with as little as 50 μL of sample and indicated the presence of PFOS isomers and PFASs. The detection limits of PFOS isomers were 0.02–0.09 ng mL<sup>–1</sup> with sufficient recoveries (85%–98%), and those for other PFASs ranged from 0.01–1 ng mL<sup>–1</sup> (recoveries, 80%–100%), which are suitable for the analysis of human serum samples.

Abstract:Organofluorine mass balance approaches are increasingly applied to investigate the occurrence of per- and polyfluoroalkyl substances (PFAS) and other organofluorines in environmental samples more comprehensively. Usually, complex samples prevent the identification and quantification of every fluorine-containing molecule. Consequently, large unidentified fractions between fluorine sum parameters such as extractable organic fluorine (EOF) and the sum of quantified analytes are frequently reported. We propose using oxidative conversion to separate (unidentified) weakly fluorinated compounds (e.g., pesticides, pharmaceuticals) from PFAA-precursors (perfluoroalkyl chain lengths ≥ C 6 ). We show with three organofluorine model substances (flufenamic acid, diflufenican, pantoprazole) that CF 3 -groups or aromatic fluorine can be quantitatively converted to inorganic fluoride and trifluoroacetic acid (TFA) by applying PhotoTOP oxidation (UV/TiO 2 ). The principle of fluorine separation in mixtures is demonstrated by the oxidation of the three weakly fluorinated compounds together with the PFAA-precursor 6:2/6:2 fluorotelomer mercaptoalkyl phosphate diester (FTMAP). After oxidation, the products F − and TFA were separated from PFCAs (> C 4 ) by SPE, and the fractions were analyzed individually. Closed mass balances both with and without the addition of organic matrix were achieved. Eventually, the fluorine balance was verified by total fluorine measurements with combustion ion chromatography (CIC). The proposed methods should be considered a proof of concept to potentially explain unidentified fractions of the EOF, especially if compounds with low fluorine content such as pesticides, pharmaceuticals, and their transformation products contribute largely to the EOF. Future studies are needed to show the applicability to the complexity of environmental samples.

External Liquid Calibration Method for Iodide Chemical Ionization Mass Spectrometry Enables Quantification of Gas-Phase Per- and Polyfluoroalkyl Substances (PFAS) Dynamics in Indoor Air

Abstract:Per- and polyfluoroalkyl substances (PFAS) are manufactured chemicals that have been detected across the globe. Fluorotelomer alcohols (FTOHs) are one PFAS class commonly found in indoor air due to emissions from consumer products (e.g., textiles and food packaging) and are human metabolic, atmospheric oxidative, and industrial precursors of perfluoroalkyl carboxylic acids (PFCAs). We developed a quantitative method for real-time analysis of gas-phase FTOHs, perfluoroalkyl acids (PFCAs and GenX), one perfluorooctane sulfonamide (EtFOSA), one fluorotelomer diol (FTdiOH), and one fluorinated ether (E2) using high-resolution time-of-flight chemical ionization mass spectrometry equipped with iodide reagent ion chemistry (I-HR-ToF-CIMS). Herein, we present a direct liquid injection method for external calibration, providing detection limits of 0.19-3.1 pptv for 3-s averaging and 0.02-0.44 pptv for 120-s averaging, with the exception of E2, which had detection limits of 1700 and 220 pptv for 3- and 120-s averaging, respectively. These calibrations enabled real-time gas-phase quantification of 6:2 FTOH in room air while microwaving popcorn, with an average peak air concentration of 31.6 ± 4.5 pptv measured 2 meters from a closed microwave. Additionally, 8:2 and 10:2 FTOH concentrations in indoor air were measured in the presence and absence of a rain jacket, with observed peak concentrations of 110 and 25 pptv, respectively. Our work demonstrates the ability of I-HR-ToF-CIMS to provide real-time air measurements of PFAS relevant to indoor human exposure settings and allow for PFAS source identification. We expect that real-time quantification of other gas-phase PFAS classes is possible, enabling advances in understanding PFAS sources, chemistry, and partitioning.

Abstract:Analysis of neutral and ionic per- and polyfluoroalkyl substances (PFAS) required two discrete techniques- with methods based on gas chromatography–mass spectrometry (GC-MS) for the former and ultrahigh performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) for the latter. In this study, we developed a method for simultaneous analysis of 58 neutral and ionic PFAS in environmental samples, obviating the need for distinct extraction and instrumental methods. The UPLC-MS/MS method was optimized and validated for sensitivity, selectivity, linearity, precision, and accuracy. The instrumental detection limits for fluorotelomer alcohols (FTOHs) ranged from 0.08 to 0.50 pg on the column. Select environmental samples, including food packaging, indoor dust, roadside soil, polyurethane foam, carpet, and consumer products, were analyzed. The highest concentrations of polyfluoroalkyl phosphates (Σ6PAPs, 650 ± 910 ng/g) and fluorotelomer sulfonates (Σ4FTSs, 36 ± 51 ng/g) were found in indoor dust, whereas Σ5FTOHs (81 ± 69 ng/g) were found in carpet. Emerging PFAS, including F-53B, ADONA, and Gen-X, were found at sub ng/g concentrations. Hydrogen-substituted polyfluorocarboxylates (H-PFCAs), such as 7H-PFHpA, were found at concentrations of up to 4 ng/g in indoor dust. This method can expedite the analysis of neutral PFAS precursors, including FTOHs, PAPs, and FTSs along with legacy iPFAS in environmental samples.
