Abstract:The ultimate objective of an aquaculture feed manufacturer and aquaculture food supplier is to ensure that the feed or food produced is both safe and wholesome. Reported food safety risks, which may be associated with the use of commercial animal feeds, including compound aquaculture feeds, usually result from the possible presence of unwanted contaminants, either within the feed ingredients used or from the external contamination of the finished feed on prolonged storage. The major animal feed contaminants that have been reported to date have included Salmonellae, mycotoxins, veterinary drug residues, persistent organic pollutants, agricultural and other chemicals (solvent residues, melamine), heavy metals (mercury, lead, cadmium) and excess mineral salts (hexavalent chromium, arsenic, selenium, flourine), and transmissible spongiform encephalopathies. Apart from the direct negative effect of these possible contaminants on the health of the cultured target species, there is a risk that the feed contaminants may be passed along the food chain, via contaminated aquaculture produce, to consumers. In recent years, public concern regarding food safety has increased as a consequence of the increasing prevalence of antibiotic residues, persistent organic pollutants, and chemicals in farmed seafood. The important role played by the Food and Agriculture Organization of the United Nations (FAO) and the Codex Alimentarius Commission in the development of international standards, guidelines, and recommendations to protect the health of consumers and ensure fair practices in the food trade is discussed.

Abstract:This paper illustrates how supply chain (SC) analytics could provide strategic and operational insights to evaluate the risk-based allocation of regulatory resources in food SCs, for management of food safety and adulteration risks. This paper leverages data on 89,970 tests of aquatic products extracted from a self-constructed data set of 2.6 million food safety tests conducted by the China Food and Drug Administration (CFDA) organizations. The integrated and structured data set is used to conduct innovative analysis that identifies the sources of adulteration risks in China’s food SCs and contrasts them with the current test resource allocations of the CFDA. The analysis highlights multiple strategic insights. Particularly, it suggests potential gaps in the current CFDA testing allocation by SC location, which is heavily focused on retail and supermarkets. Instead, the analysis indicates that high-risk parts of the SC, such as wholesale and wet markets, are undersampled. Additionally, the paper highlights the impact that SC analytics could have on policy-level operational decision making to regulate food SCs and manage food safety. The hope is that the paper will stimulate the interest of academics with expertise in these areas to conduct more work in this important application domain. Abstract:Safety and sustainability of animal feeds is a pillar of the safety of the entire food chain. Feed additive assessment incorporates consumer safety as well as animal health and welfare, which, in turn, can affect productivity and hence food security. The safety of feed users and the environment are other important components of the assessment process which, therefore, builds on a One Health perspective. In several instances the assessment entails a balanced assessment of benefits and risks for humans, animals and/or the environment. Three case studies are selected to discuss issues for a consistent framework on Risk-Benefit Assessment (RBA) of feed additives, based on EFSA opinions and literature: (a) Supplementation of feeds with trace elements with recognized human toxicity (cobalt, iodine) - RBA question: can use levels, hence human exposure, be reduced without increasing the risk of deficiency in animals?; (b) Aflatoxin binders in dairy animals - RBA question: can the use reduce the risk for human health due to aflatoxin M1, without unexpected adverse effects for animals or humans?; (c) Use of formaldehyde as preservative in feedstuffs to prevent microbial contamination - RBA question: is the reduction of microbiological risks outweighed by risks for the consumers, farmed animals or the workers? The case studies indicate that the safety of use of feed additives can involve RBA considerations which fit into a One Health perspective. As in other RBA circumstances, the main issues are defining the question and finding “metrics” that allow a R/B comparison; in the case of feed additives, R and B may concern different species (farm animals and humans). A robust assessment of animal requirements, together with sustainability considerations, might be a significant driving force for a RBA leading to a safe and effective use.

Abstract:Food security has emerged as one of the most pressing socio-economic and health issues of our time. While the formal processes of international and national governance are short-changing the need for action, an increasing number of professional science-based organisations are rallying to reduce the presence and risks of food insecurity. Examples are the Food in Health Security Network (FIHS) for the Asia Pacific region and the 'Healthy Agriculture, Healthy Nutrition, Healthy People’ initiative of the World Council on Genetics, Nutrition and Fitness for Health. The common denominator is the threat to ecosytems which are intrinsic to food and health systems. To increase their prospects for sustainability and health promotion, coordinated partnerships between agriculture and health as well as other sectors are imperative.
Using the rapid alert system for food and feed: potential benefits and problems on data interpretation

Abstract:The Rapid Alert System for Food and Feed (RASFF), where competent authorities in each Member State (MS) submit notifications on the withdrawal of unsafe or illegal products from the market, makes a significant contribution to food safety control in the European Union. The aim of this paper is to frame the potential challenges of interpreting and then acting upon the dataset contained within the RASFF system. As it is largest cause of RASFF notifications, the lens of enquiry used is mycotoxin contamination. The methodological approach is to firstly iteratively review existing literature to frame the problem, and then to interrogate the RASFF system and analyze the data available. Findings are that caution should be exercised in using the RASFF database both as a predictive tool and for trend analysis, because iterative changes in food law impact on the frequency of regulatory sampling associated with border and inland regulatory checks. The study highlights the variability of engagement by MSs with the RASFF database, influencing generalisability of the trends noted. As importing countries raise market standards, there are wider food safety implications for the exporting countries themselves. As this is one of the first studies articulating the complexities and opportunities of using the RASFF database, this research makes a strong contribution to literature.
Application and validation of a statistically derived risk-based sampling plan to improve efficiency of inspection and enforcement

Abstract:The statistically derived risk-based sampling plan for surveillance sample assignments of chemical and biological hazards was designed using binomial probability distribution. The binomial statistics was applied to the past 3-year data to estimate a confidence interval and a sample size aiming to improve efficiency and effectiveness of the agency's sampling and inspectional activities. The accuracy of the statistical models and computed estimates were validated in the following years. The ranges of confidence interval and sample size appeared to be significantly influenced by a level of the violation rate of feed product samples, an acceptable error, a number of the analyzed samples, and a statistical significance level. The violation rates of feed products for target analytes (aflatoxins, fumonisins, Salmonella, and dioxin) in the validation data were lower than those of the average 3-year data in most feed products. Besides, the actual violation rates of the validation samples did not exactly fall within the anticipated range of the confidence interval estimates. Such a discrepancy is considered introduced by several factors such as sample size adequacy, skewed distribution of a target analyte in feed products, and unique analyte/product combination. The overall study results indicate that the risk-based plan of work would provide a more effective and efficient risk management tool to help improve the oversight of the feed industry and the compliance to feed safety standards.

Abstract:Food safety monitoring faces the challenge of tackling multiple chemicals along the various stages of the food supply chain. Our study developed a methodology for optimizing sampling for monitoring multiple chemicals along the dairy supply chain. We used a mixed integer nonlinear programming approach to maximize the performance of the sampling in terms of reducing the risk of the potential disability adjusted life years (DALYs) in the population. Decision variables are the number of samples collected and analyzed at each stage of the food chain (feed mills, dairy farms, milk trucks, and dairy processing plants) for each chemical, given a predefined budget. The model was applied to the case of monitoring for aflatoxin B1/M1(AFB1/M1) and dioxins in a hypothetical Dutch dairy supply chain, and results were calculated for various contamination scenarios defined in terms of contamination fraction and concentrations. Considering various monitoring budgets for both chemicals, monitoring for AFB1/M1 showed to be more effective than for dioxins in most of the considered scenarios, because AFB1/M1 could result into more DALYs than dioxins when both chemicals are in same contamination fraction, and costs for analyzing one AFB1/M1 sample are lower than for one dioxins sample. The results suggest that relatively more resources be spent on monitoring AFB1/M1 when both chemicals' contamination fractions are low; when both contamination fractions are higher, relatively more budget should be addressed to monitoring dioxins.

Abstract:In an increasingly interconnected world, ensuring the safety of the global food supply has emerged as a paramount concern. With foodborne illnesses affecting millions annually and international trade in food products reaching unprecedented levels, the need for robust food safety standards has never been more pressing. This abstract explores the vital role of international collaboration and policy harmonization in enhancing global food safety standards, with a focus on the mechanisms, challenges, and potential solutions involved. At the heart of global food safety efforts lie collaborative endeavors among nations facilitated by international organizations such as the World Health Organization (WHO), the Food and Agriculture Organization (FAO), and the Codex Alimentarius Commission. These entities serve as platforms for harmonizing food safety regulations by developing science-based standards, guidelines, and codes of practice. Through regular consultations, expert committees, and consensus-building processes, these organizations foster agreement on key principles and practices governing food safety across diverse national contexts. However, achieving harmonization in food safety policies poses significant challenges. Divergent regulatory frameworks, varying levels of institutional capacity, and disparities in resources among countries can impede efforts to establish uniform standards. Moreover, geopolitical tensions and trade disputes may hinder cooperation, leading to fragmentation in global food safety governance. Addressing these challenges requires concerted efforts to build trust, enhance communication, and promote transparency among stakeholders. Technological innovations offer promising avenues for overcoming barriers to international collaboration and policy harmonization in food safety. Blockchain technology, for instance, enables transparent and tamper-proof recording of food supply chain data, enhancing traceability and accountability. Internet of Things (IoT) devices provide real-time monitoring of food storage and transportation conditions, helping to prevent contamination and spoilage. Artificial Intelligence (AI) algorithms analyze vast datasets to identify emerging food safety risks and inform regulatory decision-making. By leveraging these technologies, countries can strengthen their capacity to implement harmonized food safety standards and improve compliance with international regulations. Furthermore, public-private partnerships play a crucial role in advancing global food safety initiatives. Collaboration between governments, industry stakeholders, academia, and civil society organizations facilitates knowledge sharing, capacity building, and resource mobilization. By pooling expertise and resources, these partnerships enable more effective surveillance, response, and mitigation of food safety risks on a global scale. Moreover, they promote information exchange and best practice dissemination, fostering a culture of continuous improvement in food safety management. Policy harmonization efforts must also embrace risk-based approaches to prioritize interventions and allocate resources effectively. By identifying and assessing food safety hazards based on their likelihood and severity, countries can tailor their regulatory measures to address the most significant risks. Moreover, regular monitoring, evaluation, and review of food safety policies are essential to ensure their relevance, effectiveness, and adaptability in the face of evolving threats and challenges. Abstract:In view of a continuous trend in replacing synthetic feed additives and especially flavouring compounds by botanical preparations, different aspects of the safety evaluations of plants and plant-derived preparations and components in feed are discussed. This includes risk assessment approaches developed by the European Food Safety Authority (EFSA) for phytotoxins regarding unintentional exposure of target animals and of consumers to animal derived food via carry-over from feed. Relevant regulatory frameworks for feed additives and feed contaminants in the European Union are summarised and the essentials of existing guidelines used in the safety evaluation of botanicals and their preparations and components in feed are outlined. The examples presented illustrate how the safety of the botanicals, their preparations and components present in feed is assessed. An outlook on possible future developments in risk assessment by applying new in vitro and in silico methodologies is given.

Abstract:Background. It is well-established that a range of macronutrients, micronutrients and bioactive compounds found in animal-source foods play unique and important roles in human health as part of a healthy diet. Methods. This narrative review focuses on terrestrial animal source foods (TASFs). It particularly analyzes five groups: poultry eggs, milk, unprocessed meat, foods from hunting and wildlife farming, and insects. The objectives were as follows: (1) examine the nutrient composition of TASFs within and across livestock species, drawing on the country and regional food composition databases; (2) analyze the influence of intrinsic animal characteristics and production practices on TASF nutritional quality. Results. TASFs are rich in high-quality proteins and fats, as well as micronutrients such as vitamin B12, iron or zinc. This study found differences in the nutritional quality of TASFs by livestock species and animal products, as well as by characteristics of livestock production systems. Our findings suggest that there may be public health opportunities by diversifying TASF consumption across species and improving certain aspects of the production systems to provide products that are both more sustainable and of higher quality. Conclusions. Future research should adopt a more holistic approach to examining the food matrix and the dietary patterns that influence TASF digestibility. It is necessary to include meat from hunting and wildlife farming and insects in global food composition databases, as limited literature was found. In addition, scarce research focuses on low- and middle-income countries, highlighting the need for further exploration of TASF food composition analysis and how intrinsic animal characteristics and livestock production system characteristics impact their nutritional value.

Abstract:Our review explores the changing food production, distribution and consumption environment in low and middle-income countries and emerging economies as a basis for framing how to study food systems in order to address public health issues of food safety and nutrition. It presents the state of knowledge on existing food systems science and its use in terms of sustainable actions for food safety and public health. The review identifies a knowledge gap in food system mapping and governance, with value chain mapping of key commodities often missing. Despite a number of initiatives, the application of food systems methods is highly variable in scope and quality. Most analyses concentrate on specific commodities, rarely taking into account the need for a whole diet approach when looking at nutrition or the assessment of a range of infectious agents and their interactions when looking at food safety. Of the studies included in the review there is a growing observation of “informal” food systems, a term used inconsistently and one that requires revision. “Informal” food systems link to the formal sector to provide food security, yet with trade-offs between economic efficiencies and food safety. Efforts to improve food safety are hampered by inadequate food safety capacities and a lack of policy coherence leading to: inadequate investment; fragmented food quality control systems; weak or non-existent traceability mechanisms; weak foodborne disease surveillance; obsolete food regulation; and weak regulatory enforcement. In-depth food systems assessments can complement risk analysis to identify risky behaviors and understand institutional settings in order to improve codes of practice and enforcement. Methods for looking at food safety from a food systems perspective are emerging, yet existing nutrition and food systems science are not advancing sufficiently in response to nutritional public health problems. There is an urgency for improved understanding of the structure and drivers of the food systems, for better planning of changes that leads to nutrients access and healthy levels of eating. It is proposed that countries and international institutions provide an atlas of food system maps for the key commodities based on an agreed common methodology and developed by multidisciplinary teams.
A new dimension on foetal language learning

Abstract:In this issue of Acta Paediatrica, Christine Moon, Hugo Lagercrantz and Patricia K. Kuhl report their findings in phoneme learning before birth (1). They demonstrate that the exposure to the voice of the mother (and possibly other people) in the native language of the mother has altered the processing of phonemes. Specifically, they show that newborn infants seem to perceive variants of phonemes differently depending on whether the phonemes belong to the mother’s native language or not. This result shows a completely new dimension on foetal language learning. Foetal or very early neonatal learning of several key aspects of language have already been demonstrated in several studies. There is plenty of evidence that the early human auditory system is very effective in learning from sounds and language in a variety of ways. For example, high-level abstract rules are automatically extracted by neonatal brains from the auditory environment, allowing them to segregate sounds into streams (2), form coherent sound objects on the basis of sound feature combinations (3) and spot rule-breaking events after forming predictions on the basis of previously heard sound patterns (4). The neonatal capabilities in statistical learning (5) and the amazing speed such learning occurs in a sleeping neonate demonstrate that the neonatal brain is highly capable of differentiating 30 syllables, grouping them according to their statistical co-occurrences, keeping this information in memory, and reacting to syllables with high information content (processing the first syllables of the ‘words’ differentially compared with other syllables). In addition, newborn infants have been shown to react to prosodic aspects of speech and prefer their mother’s native language (6) and even manage to replicate the learned melody and intensity contours of their native language in their cry (7). It thus seems that the neonatal brain is very well tuned towards learning from sounds and specifically from language. The study of Moon, Lagercrantz and Kuhl (1) shows for the first time that already neonates possess information on their native language phonemes and that this information was obtained during the foetal period. This new view of foetal and neonatal language learning challenges the previous views. So far, we have assumed that such language components as slow and clear emotional prosodic cues, or cooccurrences of hundreds of repetitions would form the basis of foetal and neonatal language learning, while these new results suggest that also typically very quickly changing events such as phoneme formant frequencies can be learned by the foetus. This implies that the foetal brain at the time close to birth can actually perceive and learn most of the key aspects of language. Prosody is an interesting part of human language. Prosodic cues can be very strong and one could assume that in many cases in infancy they would override phonemic cues. Infant brains react strongly to changes in voice emotional prosody (8). We share the emotional prosodic features (and gestures) with nonhuman primates: it is very easy for a human to understand emotions in monkey vocalization – a tender call by a female monkey to her offspring compared with an aggressive roar of a violent male. Naturally, also infants are very capable of recognizing such emotional expressions in voice (9). The set of acoustic features that we use to form the emotional prosody are sharednot only in the communication of all primates but also in the expressions of music. Musical expressions are built on the same universal skills of the human auditory system as those of language. Bothmusic and speech use the samemusical features to signal fear with loud, low, noisy sounds and voices, and both express joy using Invited Commentary for Moon et al., Language experienced in utero affects vowel perception after birth:a two-country study, pages 156–160.

Abstract:Food safety and quality and their associated risks pose a major concern worldwide regarding not only the relative economical losses but also the potential danger to consumer's health. Customer's confidence in the integrity of the food supply could be hampered by inappropriate food safety measures. A lack of measures and reliable assays to evaluate and maintain a good control of food characteristics may affect the food industry economy and shatter consumer confidence. It is imperative to create and to establish fast and reliable analytical methods that allow a good and rapid analysis of food products during the whole food chain. Proteomics can represent a powerful tool to address this issue, due to its proven excellent quantitative and qualitative drawbacks in protein analysis. This review illustrates the applications of proteomics in the past few years in food science focusing on food of animal origin with some brief hints on other types. Aim of this review is to highlight the importance of this science as a valuable tool to assess food quality and safety. Emphasis is also posed in food processing, allergies, and possible contaminants like bacteria, fungi, and other pathogens.

Abstract:Food is an elementary requirement for human life, providing nutrients and essential energy needed for optimal health. But at the same time, food can also be a vehicle of hazardous substances or pathogens that could affect human health negatively. Risk-benefit assessment (RBA) of foods, a relatively new methodology for decision support, integrates nutrition, toxicology, microbiology, chemistry and human epidemiology for a comprehensive health impact assessment. By integrating health risks and benefits related to food consumption, RBA facilitates science-based decision-making in food-related areas and the development of policies and consumer advice. The present work programme aimed to allow the fellow to become acquainted with the process of RBA and the associated tools needed to assess quantitatively the risks and the benefits through three main activities (i) to learn the different methodologies used for RBA; (ii) to apply these methodologies to a specific case-study - RBA of raw milk consumption; and (iii) to participate in the main activities of the Risk-Benefit research group at DTU Food regarding risk-benefit issues. For the RBA of raw milk consumption, microbiological pathogens (Listeria monocytogenes, Salmonella spp., Campylobacter jejuni and Shiga toxin-producing Escherichia coli), probiotic bacteria and nutritional components (vitamins B2 and A) were considered, as well as the potential impact of raw milk consumption in the reduction of the allergies' prevalence. Two major approaches were applied: the bottom-up (estimating the disease incidence due to the exposure) and the top-down (using epidemiological and incidence data to the estimate the number of cases attributable to a certain exposure). Through all the training and hands-on activities performed, the present work programme enabled the fellow to extend the knowledge on the quantitative RBA, specifically in the context of raw milk consumption. EU-FORA programme also provided an exceptional opportunity of networking and establishment of future research lines of collaboration.

Abstract:In the course of the last decade, the Appropriate Level of Protection (ALOP), the Food Safety Objective (FSO) and their associated metrics have been proposed by the World Trade Organization and Codex Alimentarius as a means for competent authorities to ultimately translate governmental public health policy regarding food safety into risk-based targets for the food industry. The industry needs to meet these targets through the effective choice of control measures that are part of its operational food safety management system. The aim of this study was to put the practical application of ALOP and FSO to the test in the case of Salmonella in chicken meat in the Netherlands. Two different risk assessment approaches were applied to derive potential ALOP and FSO values, a 'top-down' approach based on epidemiological data and a 'bottom-up' approach based on food supply chain data. To this end, two stochastic models specific to the Dutch situation were built. Comparisons between 23 countries in Europe were also made using the top-down model. The mean estimated current Level Of Protection values were similar for the two approaches applied, with the bottom-up model yielding 87 cases per 100,000 inhabitants per year (95% CI: 0.03, 904) and the top-down model 71 (95% CI: 9.9, 155). The estimated FSO values on the other hand were considerably different with the mean 'top down' FSO being -4.6 log CFU/g (95% CI: -5.4, -4.1) and the mean 'bottom-up' FSO -6.0 log CFU/g (95% CI: -8.1, -2.9) reflecting major differences in the output distributions of this parameter obtained with the two approaches. Significant differences were observed between current LOP values for different EU countries, although it was not clear whether this was due to actual differences in the factors influencing the risk of salmonellosis or due to the quality of the available data.
Foraging animal origin food samples as passive indicators of dioxin-like POPs contamination in industry sites: Method development, characterisation and risk assessment

Abstract:Titanium dioxide (TiO 2 ) is an important industrial chemical, and studies suggest its major production route - the chloride process could lead to the generation of unintentional dl-POPs. However, no relevant studies assessed the occurrence of dl-POPs associated with TiO 2 production in the industrial zones, which is mostly due to the ultra-trace level distribution of these compounds in environmental compartments. The present study explored the novel possibility of utilising foraging animal-origin foods as sensitive indicators for addressing this challenge and generated a globally beneficial dataset by assessing the background levels of dl-POPs in the vicinity of a TiO 2 production house in Southern India. Systematic sampling of foraging cow's milk and free-ranging hen's eggs was carried out from the study site, and the dl-POPs assessments were conducted utilising an in-house developed cost-effective GC-MS/MS-based analytical methodology. The median dl-POPs levels in milk and egg samples were about 3 times higher than the control samples collected from farm-fed animals and retail markets. The contaminant loads in the foraging animal-origin food samples were further traced to their presence in environmental compartments of soil and sediment and admissible degree of correlations were observed in congener fingerprints. Elevated health risks were inferred for the population in the industrial zones with weekly intakes weighing about 0.15–17 times the European Food Safety Authority-assigned levels. The consumption of foraging cow's milk was observed to have a higher contribution towards the hazard indices and cancer risk estimates and were significantly higher (p < 0.05) for children. The study also presents a critical validation of the GC-MS/MS-based method for the purpose of regulatory monitoring of dl-POPs, which could be of practical significance in economies in transition.

Abstract:Background. Animal source foods are under scrutiny for their role in human health, yet some nutritionally vulnerable populations are largely absent from consideration. Methods. Applying a Population Intervention/Exposure Comparator Outcome (PICO/PECO) framework and prioritizing systematic review and meta-analyses, we reviewed the literature on terrestrial animal source foods (TASFs) and human health, by life course phase. Results. There were consistent findings for milk and dairy products on positive health outcomes during pregnancy and lactation, childhood, and among older adults. Eggs were found to promote early childhood growth, depending on context. Unprocessed meat consumption was associated with a reduced risk for anemia during pregnancy, improved cognition among school-age children, and muscle health in older adults. Milk and eggs represent a risk for food sensitivities/allergies, though prevalence is low, and individuals tend to outgrow the allergies. TASFs affect the human microbiome and associated metabolites with both positive and negative health repercussions, varying by type and quantity. Conclusions. There were substantial gaps in the evidence base for studies limiting our review, specifically for studies in populations outside high-income countries and for several TASF types (pig, poultry, less common livestock species, wild animals, and insects). Nonetheless, sufficient evidence supports an important role for TASFs in health during certain periods of the life course.

Abstract:Food products can contain various substances, including essential nutrients, as well as non-nutritive elements and potentially toxic metals. Metal contaminants have the potential to accumulate within the food chain and, when they exceed safe thresholds, can be toxic to humans, leading to health issues. To mitigate health hazards caused by exposure to such harmful substances, accurate monitoring of metal concentrations in various food samples is crucial. Achieving this goal needs understanding the basic principles of various elemental analysis methods. Additionally, selecting the appropriate technique or combination of techniques is critical for obtaining accurate and relevant results. Various advanced analytical techniques, such as atomic absorption spectroscopy, flame emission spectroscopy, inductively coupled plasma-mass spectrometry (ICP-MS), and X-ray fluorescence (XRF) spectrometry, can be used for the quantification of heavy metals and metalloids in food. However, each method has its own limitations, and the accuracy depends on adequate sample preparation. This paper aims to provide a clear overview of commonly used methods and techniques for heavy metal detection in food products, addressing the advantages and limitations of each analytical technique. Additionally, it compares the most important performance parameters of the presented techniques, including the limit of detection (LOD), the limit of quantification (LOQ), recovery, and precision. Moreover, ensuring food safety involves conducting a thorough risk assessment analysis. By integrating risk assessment into the evaluation of heavy metals in food, it becomes possible to determine whether observed concentrations pose significant risks to human health. This step is imperative for establishing regulatory guidelines and implementing control measures to reduce or eliminate potential health risks. Incorporating risk assessment into the broader context of the review enhances its applicability in real-world scenarios, aiding policymakers, regulatory bodies, and researchers in making informed decisions regarding food safety standards and practices.

Abstract:Neuroglobin (Ngb) is a six-coordinate globin that can catalyze the reduction of nitrite to nitric oxide. Although this reaction is common to heme proteins, the molecular interactions in the heme pocket that regulate this reaction are largely unknown. We have shown that the H64L Ngb mutation increases the rate of nitrite reduction by 2000-fold compared to that of wild-type Ngb [Tiso, M., et al. (2011) J. Biol. Chem. 286, 18277–18289]. Here we explore the effect of distal heme pocket mutations on nitrite reduction. For this purpose, we have generated mutations of Ngb residues Phe28(B10), His64(E7), and Val68(E11). Our results indicate a dichotomy in the reactivity of deoxy five- and six-coordinate globins toward nitrite. In hemoglobin and myoglobin, there is a correlation between faster rates and more negative potentials. However, in Ngb, reaction rates are apparently related to the distal pocket volume, and redox potential shows a poor relationship with the rate constants. This suggests a relationship between the nitrite reduction rate and heme accessibility in Ngb, particularly marked for His64(E7) mutants. In five-coordinate globins, His(E7) facilitates nitrite reduction, likely through proton donation. Conversely, in Ngb, the reduction mechanism does not rely on the delivery of a proton from the histidine side chain, as His64 mutants show the fastest reduction rates. In fact, the rate observed for H64A Ngb (1120 M–1 s–1) is to the best of our knowledge the fastest reported for a heme nitrite reductase. These differences may be related to a differential stabilization of the iron–nitrite complexes in five- and six-coordinate globins.
