Abstract:Arbuscular mycorrhizal fungi (AMF) are beneficial for the plant growth under heavy metal stress. Such beneficial effect is improved by elevated CO2 (eCO2). However, the mechanisms by which eCO2 improves AMF symbiotic associations under arsenite (AsIII) toxicity are hardly studied. Herein, we compared these regulatory mechanisms in species from two agronomical important plant families - grasses (wheat) and legumes (soybean). AsIII decreased plant growth (i.e., 53.75 and 60.29% of wheat and soybean, respectively) and photosynthesis. It also increased photorespiration and oxidative injury in both species, but soybean was more sensitive to oxidative stress as indicated by higher H2O2 accumulation and oxidation of protein and lipid. eCO2 significantly improved AMF colonization by increasing auxin levels, which induced high carotenoid cleavage dioxygenase (CCDs) activity, particularly in soybean roots. The improved sugar metabolism in plant shoots by co-application of eCO2 and AsIII allocated more sugars to roots sequentially. Sugar accumulation in plant roots is further induced by AMF, resulting in more C skeletons to produce organic acids, which are effectively exudated into the soil to reduce AsIII uptake. Exposure to eCO2 reduced oxidative damage and this mitigation was stronger in soybean. This could be attributed to a greater reduction in photorespiration as well as a stronger antioxidant and detoxification defence systems. The grass/legume-specificity was supported by principal component analysis, which revealed that soybean was more affected by AsIII stress and more responsive to AMF and eCO2. This study provided a mechanistic understanding of the impact of AMF, eCO2 and their interaction on As-stressed grass and legume plants, allowing better practical strategies to mitigate AsIII phytotoxicity.
Arbuscular Mycorrhiza Improves Photosynthesis and Restores Alteration in Sugar Metabolism in Triticum aestivum L. Grown in Arsenic Contaminated Soil

Abstract:Contamination of agricultural soil by arsenic (As) is a serious menace to environmental safety and global food security. Symbiotic plant–microbe interaction, such as arbuscular mycorrhiza (AM), is a promising approach to minimize hazards of As contamination in agricultural soil. Even though the potential of AM fungi (AMF) in redeeming As tolerance and improving growth is well recognized, the detailed metabolic and physiological mechanisms behind such beneficial effects are far from being completely unraveled. The present study investigated the ability of an AM fungus, Rhizophagus intraradices , in mitigating As-mediated negative effects on photosynthesis and sugar metabolism in wheat ( Triticum aestivum ) subjected to three levels of As, viz., 0, 25, and 50 mg As kg –1 of soil, supplied as sodium arsenate. As exposure caused significant decrease in photosynthetic pigments, Hill reaction activity, and gas exchange parameters such as net photosynthetic rate, stomatal conductance, transpiration rate, and intercellular CO 2 concentration. In addition, As exposure also altered the activities of starch-hydrolyzing, sucrose-synthesizing, and sucrose-degrading enzymes in leaves. Colonization by R. intraradices not only promoted plant growth but also restored As-mediated impairments in plant physiology. The symbiosis augmented the concentration of photosynthetic pigments, enhanced Hill reaction activity, and improved leaf gas exchange parameters and water use efficiency of T. aestivum even at high dose of 50 mg As kg –1 of soil. Furthermore, inoculation with R. intraradices also restored As-mediated alteration in sugar metabolism by modulating the activities of starch phosphorylase, α-amylase, β-amylase, acid invertase, sucrose synthase, and sucrose-phosphate synthase in leaves. This ensured improved sugar and starch levels in mycorrhizal plants. Overall, the study advocates the potential of R. intraradices in bio-amelioration of As-induced physiological disturbances in wheat plant.

Abstract:Arbuscular mycorrhiza (AM) can help plants to tolerate arsenic (As) toxicity. However, plant responses are found to vary with the host plant and the AM fungal species. The present study compares the efficacy of two AM fungi Rhizoglomus intraradices (M1) and Glomus etunicatum (M2) in amelioration of As stress in wheat (Triticum aestivum L. var. HD-2967). Mycorrhizal (M) and non-mycorrhizal (NM) wheat plants were subjected to four levels of As (0, 25, 50, and 100 mg As kg-1 soil). Although As additions had variable effects on the percentage of root colonized by the two fungal inoculants, each mycobiont conferred benefits to the host plant. Mycorrhizal plants continued to display better growth than NM plants. Formation of AM helped the host plant to overcome As-induced P deficiency and maintained favorable P:As ratio. Inoculation of AMF had variable effects on the distribution of As in plant tissues. While As translocation factor decreased in low As (25 mg kg-1 soil), it increased under high As (50 and 100 mg As kg-1 soil). Further As translocation to grain was reduced (As grain:shoot ratio) in M plants compared with NM plants. Arsenic-induced oxidative stress (generation of H2O2 and lipid peroxidation) in plants reduced significantly by AMF inoculation. The alleviation potential of AM was more evident with increase in severity of As stress. Colonization of AMF resulted in higher activities of the antioxidant enzymes (superoxide dismutase, catalase, and guaiacol peroxidase). It increased the concentrations of the antioxidant molecules (carotenoids, proline, and α-tocopherol) than their NM counterparts at high As addition level. Comparatively higher activities of enzymes of glutathione-ascorbate cycle in M plants led to higher ascorbate:dehydroascorbate (AsA:DHA) and glutathione:glutathione disulphide (GSH:GSSG) ratios. Inoculation by AMF also augmented the glyoxalase system by increasing the activities of both glyoxalase I and glyoxalase II enzymes. Mycorrhizal colonization increased concentrations of cysteine, glutathione, non-protein thiols, and activity of glutathione-S-transferase that facilitated sequestration of As into non-toxic complexes. The study reveals multifarious role of AMF in alleviation of As toxicity.

Abstract:The inoculation with arbuscular mycorrhizal (AM) fungi shows great potential to increase the tolerance of host plants toward toxic arsenic. In many Asian countries and elsewhere, crops are increasingly being irrigated with arsenic‐contaminated groundwater, which causes loss of yield and the risk that arsenic enters the food chain. Greenhouse cultivation experiments demonstrated that AM fungi are able to increase the arsenic tolerance in crop plants, but the results are highly variable. This meta‐analysis demonstrates that AM fungi significantly reduce the arsenic concentration, and at the same time, positively modify the nutrient supply and the biomass in host crops that are grown under arsenic stress. Thus, the inoculation with AM fungi represents a promising approach to alleviate the negative effects of arsenic on crop growth, which deserves to be developed in future studies. SummaryRecent greenhouse experiments showed that the inoculation of crop plants with arbuscular mycorrhizal (AM) fungi does not only improve plant nutrition but also shows great potential to increase the host plants' tolerance toward the toxic metalloid arsenic (As). This approach could alleviate negative effects of crop plants grown under As stress, which causes substantial loss of yield and the risk of As entering the food chain. This is especially relevant in Asian countries and elsewhere where As subsequently accumulates in soil due to irrigation with As‐polluted groundwater. Overall effects of AM fungi symbiosis on seven crop plants grown under As stress were assessed by a meta‐analysis that included 299 studies obtained from 27 independent publications. Biomass, concentrations of As and phosphorus (P), and P/As ratios in plant tissue were used to evaluate the effects of AM fungi in host crops. Inoculation with AM fungi reduced As concentrations in tissue of host plants by 19% on average, while it increased the P/As ratio and biomass at the same time by 64% and 53%, respectively. With the exception of Helianthus annuus, AM effects were highly significant in all crop plants. The meta‐analysis results illustrate that AM fungi are able to significantly reduce As concentration, and at the same time, positively modify the nutrient supply and the biomass in host crops. These promising results strongly encourage the implementation of field trials as a next step toward the development of AM fungi‐based approaches to alleviate the negative effects of As stress on crop growth.

Abstract:Abstract Rice polluted by metal(loid)s, especially arsenic (As) and cadmium (Cd), imposes serious health risks. Numerous studies have demonstrated that the obligate plant symbionts arbuscular mycorrhizal fungi (AMF) can reduce As and Cd concentrations in rice. The behaviours of metal(loid)s in the soil–rice–AMF system are of significant interest for scientists in the fields of plant biology, microbiology, agriculture, and environmental science. We review the mechanisms of As and Cd accumulation in rice with and without the involvement of AMF. In the context of the soil–rice–AMF system, we assess and discuss the role of AMF in affecting soil ion mobility, chemical forms, transport pathways (including the symplast and apoplast), and genotype variation. A potential strategy for AMF application in rice fields is considered, followed by future research directions to improve theoretical understanding and encourage field application.

Abstract:Arsenic (As) pollution in wetlands has emerged as a serious global concern, posing potential threat to the growth of wetland plants. Arbuscular mycorrhizal fungi (AMF) can alleviate As phytotoxicity to host plants, but their ecological functions in wetland plants under flooding conditions remain largely unknown. Thus, a pot experiment was conducted using Rhizophagus irregularis and Iris tectorum Maxim. exposed to light (15 and 30 mg/kg As) and high (75 and 100 mg/kg As) levels of As, to investigate the intrinsic mechanisms underlying the effects of mycorrhizal inoculation on plant As tolerance under flooding conditions. The mycorrhizal colonization rates ranged from 31.47 ± 3.92 % to 60.69 ± 5.58 %, which were higher than the colonization rate (29.55 ± 13.60%) before flooding. AMF significantly increased biomass of I. tectorum under light As levels, together with increased phosphorus (P) and As uptake. Moreover, expression of arsenate reductase gene RiarsC and a trace of dimethylarsenic (1.87 mg/kg in shoots) were detected in mycorrhizal plants, suggesting As transformation and detoxification by AMF exposed to light levels of As. However, under high As levels, AMF inhibited As translocation from roots to shoots, and facilitated the formation of iron plaque. The immobilized As concentrations in iron plaque of mycorrhizal plants were respectively 1133.68 ± 179.17 mg/kg and 869.11 ± 248.90 mg/kg at 75 and 100 mg/kg As addition level, both significantly higher than that in non-inoculated plants. Irrespective of As exposure levels, mycorrhizal symbiosis decreased soil As bioavailability. Overall, the study provides insights into the alleviation of As phytotoxicity in natural wetland plants through mycorrhizal symbiosis, and potentially indicates function diversity of AMF under flooding conditions and As stress, supporting the subsequent phytoremediation and restoration of As-contaminated wetlands.

Abstract:Arsenic (As) accumulation in plants is a global concern. Although the application of arbuscular mycorrhizal fungi (AMF) has been suggested as a potential solution to decrease As concentration in plants, there is currently a gap in a comprehensive, quantitative assessment of the abiotic and biotic factors influencing As accumulation. A meta-analysis was performed to quantitatively investigate the findings of 76 publications on the impacts of AMF, plant properties, and soil on As accumulation in plants. Results showed a significant dose-dependent As reduction with higher mycorrhizal infection rates, leading to a 19.3% decrease in As concentration. AMF reduced As(V) by 19.4% but increased dimethylarsenic acid (DMA) by 50.8%. AMF significantly decreased grain As concentration by 34.1%. AMF also improved plant P concentration and dry biomass by 33.0% and 62.0%, respectively. The most significant reducing effects of As on AMF properties were seen in single inoculation and experiments with intermediate durations. Additionally, the benefits of AMF were significantly enhanced when soil texture, soil organic carbon (SOC), pH level, Olsen-P, and DTPA-As were sandy soil, 0.8%–1.5%, ≥7.5, ≥9.1 mg/kg, and 30–60 mg/kg, respectively. AMF increased easily extractable glomalin-related soil protein (EE-GRSP) and total glomalin-related soil protein (T-GRSP) by 23.0% and 28.0%, respectively. Overall, the investigated factors had significant implications in developing AMF-based methods for alleviating the negative effects of As stress on plants.

Abstract:Arbuscular mycorrhizal fungi (AMF) appear to be highly associated with arsenic (As) uptake in host plants because arsenate (As(V)) and phosphorus (P) share the same transporter, whereby AMF can enhance P uptake. A short-term experiment was conducted for low- (0 to 0.05 mM As) and high-affinity (0 to 2.5 mM As) uptake systems, to investigate the AMF role on As uptake mechanism in plants, which may explain As uptake kinetics in upland rice cultivar: Zhonghan 221. When concentration of As ranged from 0 to 0.05 mM, Funneliformis geosporum (Fg) significantly decreased arsenite (As(III)) and monomethylarsonicacid (MMA) uptake when (p < 0.05) compared to non-mycorrhizal (NM) treatment, since the major route for (As(III)) in rice roots-rice silicon transporter Lsi1 would be influenced by Fg inoculation at high As concentrations. Fg can also reduce As(V) uptake significantly (p < 0.05) under both uptake systems relative to NM treatment, whereas, Funneliformis mosseae (Fm) increased As(V) and MMA uptake in rice roots, with MMA uptake rate generally lower than As(III) and As(V). Using suitable AMF species inoculation with rice, As uptake and accumulation in rice grains can be reduced and the risk to human health, once consumed, can be minimized.

Abstract:It is well known that arbuscular mycorrhizal (AM) fungi can enhance plant arsenic (As) resistance by influencing As uptake, translocation, and speciation; however, As transformation and volatilization by an entire plant inoculated with AM fungus remains uninvestigated. In the present study, AM symbiosis of Rhizophagus irregularis with unbroken Medicago sativa was successfully established in vitro. Afterwards, five concentrations of arsenate were applied to the culture media. The results showed that AM inoculation could methylate inorganic As into dimethylarsinic acid (DMA), dimethylarsine (DMAsH), and trimethylarsine (TMAs), which were detected in the plants, media, or air. Volatile As, accounting for a small proportion of total organic As, appeared under high arsenate exposure, accompanied by remarkable upregulation of root RiMT-11, an arsenite methyltransferase gene in R. irregularis. In addition, AM colonization significantly increased arsenite percentages in plant tissues and external media. Regardless of As species, AM inoculation tended to release the transformed As into the environment rather than transfer them to plant tissues. Our present study, for the first time, comprehensively verified As methylation, volatilization, and reduction by AM fungus associated with the entire plant under absolute axenic conditions and gained a deeper insight into As metabolism in AM symbionts.

Abstract:Arsenic (As) is a major soil contaminant that restricts plant growth and metabolism. Silicon-Si and arbuscular mycorrhizae-AM have the potential to impart As stress tolerance. The study assessed relative and cumulative effects of Si and mycorrhizal species; Claroideoglomus etunicatum -M1 and Rhizoglomus intraradices -M2 in alleviating AsV and AsIII toxicities in pigeonpea [ Cajanus cajan (L.) Millsp.]. Arsenic reduced mycorrhizal symbiosis, nutrient availability and plant biomass by inducing oxidative stress. Si and AM inoculations decreased metalloid uptake and reactive species generation, with higher benefits under M2, followed by Si, then M1. Higher efficiency of M2 corresponded to its colonization ability and glomalin production as well as its capability of enhancing nutrient uptake by modulating soil enzymes (alkaline phosphate-ALP and arylsulfatase-aryl-S) activities. In addition, S metabolism and antioxidant system got up-regulated, leading to increased synthesis of thiol compounds and antioxidants in the order M2>Si>M1. Accumulated thiols further reduced As translocation by chelating and immobilizing metalloid in plant roots. Moreover, significantly higher As tolerance could be recorded when plants were supplemented with a combination of +Si+M2, indicating functional complementarity between the two amendments. The study, thus, demonstrated introduction of Si and AM in the rhizosphere as a promising strategy for inducing As tolerance to pigeonpea.

Abstract:Accumulation of Arsenic (As) generates oxidative stress by reducing nutrients availability in plants. Arbuscular mycorrhizal (AM) symbiosis can impart metalloid tolerance in plants by enhancing the synthesis of sulfur (S)-rich peptides (glutathione- GSH) and low-molecular-weight nitrogenous (N) osmolytes (proline- Pro). The present study, therefore investigated the efficiency of 3 AM fungal species ( Rhizoglomus intraradices -Ri, Funneliformis mosseae -Fm and Claroideoglomus claroideum - Cc) in imparting As (arsenate-AsV −40 at 60 mg kg −1 and arsenite- AsIII at 5 and 10 mg kg −1 ) tolerance in two Cicer arietinum (chickpea) genotypes (HC 3 and C 235). As induced significantly higher negative impacts in roots than shoots, which was in accordance with proportionately higher reactive oxygen species (ROS) in the former, with AsIII more toxic than AsV. Mycorrhizal symbiosis overcame oxidative stress by providing the host plants with necessary nutrients (P, N, and S) through enhanced microbial enzyme activities (MEAs) in soil, which increased the synthesis of Pro and GSH and established a redox balance in the two genotypes. This coordination between nutrient status, Pro-GSH levels, and antioxidant defense was stronger in HC 3 than C 235 due to its higher responsiveness to the three AM species. However, Ri was most beneficial in inducing redox homeostasis, followed by Fm and Cc, since the Cicer arietinum-Ri combination displayed the maximum ability to boost antioxidant defense mechanisms and establish a coordination with Pro synthesis. Thus, the results highlighted the importance of selecting specific chickpea genotypes having an ability to establish effective mycorrhizal symbiosis for imparting As stress tolerance.

Abstract:A pot experiment examined the biomass and As uptake of Medicago truncatula colonized by the arbuscular mycorrhizal (AM) fungus Glomus mosseae in low-P soil experimentally contaminated with different levels of arsenate. The biomass of G. mosseae external mycelium was unaffected by the highest addition level of As studied (200 mg kg(-1)) but shoot and root biomass declined in both mycorrhizal and non-mycorrhizal plants, indicating that the AM fungus was more tolerant than M. truncatula to arsenate. Mycorrhizal inoculation increased shoot and root dry weights by enhancing host plant P nutrition and lowering shoot and root As concentrations compared with uninoculated plants. The AM fungus may have been highly tolerant to As and conferred enhanced tolerance to arsenate on the host plant by enhancing P nutrition and restricting root As uptake.

Abstract:Arbuscular mycorrhizal fungi (AMF) play key roles in enhancing plant tolerance to heavy metals, and iron (Fe) compounds can reduce the bioavailability of arsenic (As) in soil, thereby alleviating As toxicity. However, there have been limited studies of the synergistic antioxidant mechanisms of AMF (Funneliformis mosseae) and Fe compounds in the alleviation of As toxicity on leaves of maize (Zea mays L.) with low and moderate As contamination. In this study, a pot experiment was conducted with different concentrations of As (0, 25, 50 mgꞏkg−1) and Fe (0, 50 mgꞏkg−1) and AMF treatments. Results showed that under low and moderate As concentrations (As25 and As50), the co-inoculation of AMF and Fe compound significantly increased the biomass of maize stems and roots, phosphorus (P) concentration, and P-to-As uptake ratio. Moreover, the co-inoculation of AMF and Fe compound addition significantly reduced the As concentration in stem and root, malondialdehyde (MDA) content in leaf, and soluble protein and non-protein thiol (NPT) contents in leaf of maize under As25 and As50 treatments. In addition, co-inoculation with AMF and Fe compound addition significantly increased the activities of catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD) in the leaves of maize under As25 treatment. Correlation analysis showed that stem biomass and leaf MDA content were very significantly negatively correlated with stem As content, respectively. In conclusion, the results indicated that the co-inoculation of AMF and Fe compound addition can inhibit As uptake and promote P uptake by maize under low and moderate As contamination, thereby mitigating the lipid peroxidation on maize leaves and reducing As toxicity by enhancing the activities of antioxidant enzymes under low As contamination. Effects of Antimony on Rice Growth and Its Existing Forms in Rice Under Arbuscular Mycorrhizal Fungi Environment

Abstract:Arbuscular mycorrhizal fungi (AMF) can form symbiotic relationships with most terrestrial plants and regulate the uptake and distribution of antimony (Sb) in rice. The effect of AMF on the uptake and transport of Sb in rice was observed using pot experiments in the greenhouse. The results showed that AMF inoculation increased the contact area between roots and metals by forming mycelium, and changed the pH and Eh of the root soil, leading to more Sb entering various parts of the rice, especially at an Sb concentration of 1,200 mg/kg. The increase in metal toxicity further led to a decrease in the rice chlorophyll content, which directly resulted in a 22.7% decrease in aboveground biomass, 21.7% in underground biomass, and 11.3% in grain biomass. In addition, the antioxidant enzyme results showed that inoculation of AMF decreased 22.3% in superoxide dismutase, 9.9% in catalase, and 20.7% in peroxidase compared to the non-inoculation groups, further verifying the negative synergistic effect of AMF inoculation on the uptake of Sb in rice. The present study demonstrated the effect of AMF on the uptake and transport of Sb in the soil–rice system, facilitating future research on the related mechanism in the soil–rice system under Sb stress.

Abstract:The multifarious problems created by arsenic (As), for collective environment and human health, serve a cogent case for searching integrative agricultural approaches to attain food security. Rice (Oryza sativa L.) acts as a sponge for heavy metal(loid)s accretion, specifically As, due to anaerobic flooded growth conditions facilitating its uptake. Acclaimed for their positive impact on plant growth, development and phosphorus (P) nutrition, 'mycorrhizas' are able to promote stress tolerance. Albeit, the metabolic alterations underlying Serendipita indica (S. indica; S.i) symbiosis-mediated amelioration of As stress along with nutritional management of P are still understudied. By using biochemical, RT-qPCR and LC-MS/MS based untargeted metabolomics approach, rice roots of ZZY-1 and GD-6 colonized by S. indica, which were later treated with As (10 µM) and P (50 µM), were compared with non-colonized roots under the same treatments with a set of control plants. The responses of secondary metabolism related enzymes, especially polyphenol oxidase (PPO) activities in the foliage of ZZY-1 and GD-6 were enhanced 8.5 and 12-fold, respectively, compared to their respective control counterparts. The current study identified 360 cationic and 287 anionic metabolites in rice roots, and the commonly enriched pathway annotated by Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis was biosynthesis of phenylalanine, tyrosine and tryptophan, which validated the results of biochemical and gene expression analyses associated with secondary metabolic enzymes. Particularly under As+S.i+P comparison, both genotypes exhibited an upregulation of key detoxification and defense related metabolites, including fumaric acid, L-malic acid, choline, 3,4-dihydroxybenzoic acid, to name a few. The results of this study provided the novel insights into the promising role of exogenous P and S. indica in alleviating As stress.

Abstract:Arsenic (As) is naturally present in soils and groundwater in agricultural areas, mainly in the form of arsenate (AsV) and arsenite (AsIII). It can enter soybean plants and decrease their yield and growth, therefore posing a serious problem. Our aim was to evaluate its effects on physiological events closely related to plant growth and the possible signaling pathways involved. Photosynthesis related parameters, gas exchange, mineral concentration, phytohormone content, and the expression of genes related to abscisic acid (ABA) and jasmonic acid (JA), were analyzed in soybean leaves and roots exposed to AsV and AsIII. A reduction in the CO2 assimilation rate was associated with decreased stomatal conductance, transpiration rate and intercellular CO2 concentration in treated plants. The alterations observed in the photochemical phase of the photosynthesis (OJIP -test) suggest a reduction in electron transport, mainly under AsV treatment. Arsenic also induced changes in NH4+, NO3−, PO43−, Cl−, SO42− and Na+ content in roots, while the mineral status seemed to remain unchanged in leaves. Surprisingly, ABA content was significantly lower in As-treated plants and so was the expression of GmPYL1 and GmbZIP1, genes potentially involved in ABA signaling. However, treatment caused an increase in JA content and a decrease in its precursor, 12-oxo-phytodienoic acid (OPDA), which suggests that JA could play a key role in regulating the response to the metalloid. These results build on those of previous studies and contribute to elucidating the complexity of As-triggered responses in soybean.

Abstract:Arsenic (As) bioavailability in the rice rhizosphere is influenced by many microbial interactions, particularly by metal-transforming functional groups at the root-soil interface. This study was conducted to examine As-transforming microbes and As-speciation in the rice rhizosphere compartments, in response to two different water management practices (continuous and intermittently flooded), established on fields with high to low soil-As concentration. Microbial functional gene composition in the rhizosphere and root-plaque compartments were characterized using the GeoChip 4.0 microarray. Arsenic speciation and concentrations were analyzed in the rhizosphere soil, root-plaque, pore water, and grain samples. Results confirmed several As-biotransformation processes in the rice rhizosphere compartments, and distinct assemblage of As-reducing and methylating bacteria was observed between the root-plaque and rhizosphere. Results confirmed higher potential for microbial As-reduction and As-methylation in continuously flooded, long term As-contaminated fields, which accumulated highest concentrations of As-III and methyl-As concentrations in pore water and rice grains. Water management treatment significantly altered As-speciation in the rhizosphere, and intermittent flooding reduced methyl-As and As-III concentrations in the pore water, root-plaque and rice grain. Ordination and taxonomic analysis of detected gene-probes indicated that root-plaque and rhizosphere assembled significantly different microbial functional groups demonstrating niche separation. Taxonomic non-redundancy was evident, suggesting that As-reduction, -oxidation and -methylation processes were performed by different microbial functional groups. It was also evident that As transformation was coupled to different biogeochemical cycling processes (nutrient assimilation, carbon metabolism etc.) in the compartments and between treatments, revealing functional non-redundancy of rice-rhizosphere microbiome in response to local biogeochemical conditions and As contamination. This study provided novel insights on As-biotransformation processes and their implications on As-chemistry at the root-soil interface and their responses to water management, which could be applied for mitigating As-bioavailability and accumulation in rice grains.

Abstract:Arsenic (As) is a highly toxic metalloid for all forms of life including plants. Rice is the main food source for different countries worldwide, although it can take up high amounts of As in comparison with other crops, showing toxic profiles such as decreases in plant growth and yield. The induction of oxidative stress is the main process underlying arsenic toxicity in plants, including rice, due to an alteration of the reactive oxygen species (ROS) metabolism. The aim of this work was to gain better knowledge on how the ROS metabolism and its interaction with nitric oxide (NO) operate under As stress conditions in rice plants. Thus, physiological and ROS-related biochemical parameters in roots and shoots from rice (Oryza sativa L.) were studied under 50 μM arsenate (AsV) stress, and the involvement of the main antioxidative systems and NO in the response of plants to those conditions was investigated. A decrease of 51% in root length and 27% in plant biomass was observed with 50 μM AsV treatment, as compared to control plants. The results of the activity of superoxide dismutase (SOD) isozymes, catalase, peroxidase (POD: total and isoenzymatic), and the enzymes of the ascorbate–glutathione cycle, besides the ascorbate and glutathione contents, showed that As accumulation provoked an overall significant increase of most of them, but with different profiles depending on the plant organ, either root or shoot. Among the seven identified POD isozymes, the induction of the POD-3 in shoots under As stress could help to maintain the hydrogen peroxide (H2O2) redox homeostasis and compensate the loss of the ascorbate peroxidase (APX) activity in both roots and shoots. Lipid peroxidation was slightly increased in roots and shoots from As-treated plants. The H2O2 and NO contents were enhanced in roots and shoots against arsenic stress. In spite of the increase of most antioxidative systems, a mild oxidative stress situation appears to be consolidated overall, since the growth parameters and those from the oxidative damage could not be totally counteracted. In these conditions, the higher levels of H2O2 and NO suggest that signaling events are simultaneously occurring in the whole plant.

Abstract:Soil salinization is one of the most serious abiotic stress factors affecting plant productivity through reduction of soil water potential, decreasing the absorptive capacity of the roots for water and nutrients. A weighted meta-analysis was conducted to study the effects of arbuscular mycorrhizal fungi (AMF) inoculation in alleviating salt stress in C3 and C4 plants. We analyzed the salt stress influence on seven independent variables such as chlorophyll, leaf area, photosynthetic rate (Amax), stomatal conductance (Gs), transpiration rate (E), relative water content (RWC), and water use efficiency (WUE) on AMF inoculated plants. Responses were compared between C3 and C4 plants, AMF species, plant functional groups, level of salinity, and environmental conditions. Our results showed that AMF inoculated plants had a positive impact on gas exchange and water status under salt stress. The total chlorophyll contents of C3 plants were higher than C4 plants. However, C3 plants responses regarding Gs, Amax, and E were more positive compared to C4 plants. The increase in Gs mainly maintained E and it explains the increase in Amax and increase in E. When the two major AMF species (Rhizophagus intraradices and Funnelliformis mosseae) were considered, the effect sizes of RWC and WUE in R. intraradices were lower than those in F. mosseae indicating that F. mosseae inoculated plants performed better under salt stress. In terms of C3 and C4 plant photosynthetic pathways, the effect size of C4 was lower than C3 plants indicating that AMF inoculation more effectively alleviated salt stress in C3 compared to C4 plants.
