Abstract:Ascorbic acid (AsA) and selenium nanoparticles (SeNPs) were versatile plant growth regulators, playing multiple roles in promoting plant growth under heavy metal stresses. This study aimed to evaluate the beneficial role of individual and combined effects of AsA and SeNPs on morpho-physio-biochemical traits of rice with or without chromium (Cr) amendment. The results indicated that Cr negatively affected plant biomass, gas exchange parameters, total soluble sugar, proline, relative water contents, and antioxidant-related gene expression via increasing reactive oxygen species (MDA, H2O2, O2•−) formation, resulting in plant growth reduction. The application of AsA and SeNPs, individually or in combination, decreased the uptake and translocation of Cr in rice seedlings, increased seedlings with tolerance to Cr toxicity, and significantly improved the rice seedling growth. Most notably, AsA + SeNP treatment strengthened the antioxidative defense system through ROS quenching and Cr detoxification. The results collectively suggested that the application of AsA and SeNPs alone or in combination had the potential to alleviate Cr toxicity in rice and possibly other crop species.

Abstract:The multifarious problems created by arsenic (As), for collective environment and human health, serve a cogent case for searching integrative agricultural approaches to attain food security. Rice (Oryza sativa L.) acts as a sponge for heavy metal(loid)s accretion, specifically As, due to anaerobic flooded growth conditions facilitating its uptake. Acclaimed for their positive impact on plant growth, development and phosphorus (P) nutrition, 'mycorrhizas' are able to promote stress tolerance. Albeit, the metabolic alterations underlying Serendipita indica (S. indica; S.i) symbiosis-mediated amelioration of As stress along with nutritional management of P are still understudied. By using biochemical, RT-qPCR and LC-MS/MS based untargeted metabolomics approach, rice roots of ZZY-1 and GD-6 colonized by S. indica, which were later treated with As (10 µM) and P (50 µM), were compared with non-colonized roots under the same treatments with a set of control plants. The responses of secondary metabolism related enzymes, especially polyphenol oxidase (PPO) activities in the foliage of ZZY-1 and GD-6 were enhanced 8.5 and 12-fold, respectively, compared to their respective control counterparts. The current study identified 360 cationic and 287 anionic metabolites in rice roots, and the commonly enriched pathway annotated by Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis was biosynthesis of phenylalanine, tyrosine and tryptophan, which validated the results of biochemical and gene expression analyses associated with secondary metabolic enzymes. Particularly under As+S.i+P comparison, both genotypes exhibited an upregulation of key detoxification and defense related metabolites, including fumaric acid, L-malic acid, choline, 3,4-dihydroxybenzoic acid, to name a few. The results of this study provided the novel insights into the promising role of exogenous P and S. indica in alleviating As stress.

Abstract:Fungal-bacterial combinations have a significant role in increasing and improving plant health under various stress conditions. Metabolites secreted by fungi and bacteria play an important role in this process. Our study emphasizes the significance of secondary metabolites secreted by the fungus Serendipita indica alone and by an actinobacterium Zhihengliuella sp. ISTPL4 under normal growth conditions and arsenic (As) stress condition. Here, we evaluated the arsenic tolerance ability of S. indica alone and in combination with Z. sp. ISTPL4 under in vitro conditions. The growth of S. indica and Z. sp. ISTPL4 was measured in varying concentrations of arsenic and the effect of arsenic on spore size and morphology of S. indica was determined using confocal microscopy and scanning electron microscopy. The metabolomics study indicated that S. indica alone in normal growth conditions and under As stress released pentadecanoic acid, glycerol tricaprylate, L-proline and cyclo(L-prolyl-L-valine). Similarly, d-Ribose, 2-deoxy-bis(thioheptyl)-dithioacetal were secreted by a combination of S. indica and Z. sp. ISTPL4. Confocal studies revealed that spore size of S. indica decreased by 18% at 1.9 mM and by 15% when in combination with Z. sp. ISTPL4 at a 2.4 mM concentration of As. Arsenic above this concentration resulted in spore degeneration and hyphae fragmentation. Scanning electron microscopy (SEM) results indicated an increased spore size of S. indica in the presence of Z. sp. ISTPL4 (18 ± 0.75 μm) compared to S. indica alone (14 ± 0.24 μm) under normal growth conditions. Our study concluded that the suggested combination of microbial consortium can be used to increase sustainable agriculture by combating biotic as well as abiotic stress. This is because the metabolites released by the microbial combination display antifungal and antibacterial properties. The metabolites, besides evading stress, also confer other survival strategies. Therefore, the choice of consortia and combination partners is important and can help in developing strategies for coping with As stress.

Abstract:Arsenic (As) contamination is a major problem affecting soil and groundwater in India, harming agricultural crops and human health. Plant growth-promoting rhizobacteria (PGPR) have emerged as promising agents for reducing As toxicity in plants. This study aimed to isolate and characterize As-tolerant PGPR from rice field soils with varying As levels in five districts of West Bengal, India. A total of 663 bacterial isolates were obtained from the different soil samples, and 10 bacterial strains were selected based on their arsenite (As-III) and arsenate (As-V) tolerant ability and multiple PGP traits, including phosphate solubilization, production of siderophore, indole acetic acid, biofilm formation, alginate, and exopolysaccharide. These isolates were identified by 16S rRNA gene sequencing analysis as Staphylococcus sp. (4), Niallia sp. (2), Priestia sp. (1), Bacillus sp. (1), Pseudomonas sp. (1), and Citricoccus sp. (1). Among the selected bacterial strains, Priestia flexa NBRI4As1 and Pseudomonas chengduensis NBRI12As1 demonstrated significant improvement in rice growth by alleviating arsenic stress under greenhouse conditions. Both strains were also able to modulate photosynthetic pigments, soluble sugar content, proline concentration, and defense enzyme activity. Reduction in As-V accumulation inoculated with NBRI4As1 was recorded highest by 53.02% and 31.48%, while As-III by NBRI12As1 38.84% and 35.98% in the roots and shoots of rice plants, respectively. Overall, this study can lead to developing efficient As-tolerant bacterial strains–based bioinoculant application packages for arsenic stress management in rice.

Abstract:Inadvertent accumulation of arsenic (As) in rice (Oryza sativa L.) is a concern for people depending on it for their subsistence, as it verily causes epigenetic alterations across the genome as well as in specific cells. To ensure food safety, certain attempts have been made to nullify this highest health hazard encompassing physiological, chemical and biological methods. Albeit, the use of mycorrhizal association along with nutrient reinforcement strategy has not been explored yet. Mechanisms of response and resistance of two rice genotypes to As with or without phosphorus (P) nutrition and Serendipita indica (S. indica; S.i) colonization were explored by root transcriptome profiling in the present study. Results revealed that the resistant genotype had higher auxin content and root plasticity, which helped in keeping the As accumulation and P starvation response to a minimum under alone As stress. However, sufficient P supply and symbiotic relationship switched the energy resources towards plant's developmental aspects rather than excessive root proliferation. Higher As accumulating genotype (GD-6) displayed upregulation of ethylene signaling/biosynthesis, root stunting and senescence related genes under As toxicity. Antioxidant defense system and cytokinin biosynthesis/signaling of both genotypes were strengthened under As + S.i + P, while the upregulation of potassium (K) and zinc (Zn) transporters depicted underlying cross-talk with iron (Fe) and P. Differential expression of phosphate transporters, peroxidases and GSTs, metal detoxification/transport proteins, as well as phytohormonal metabolism were responsible for As detoxification. Taken together, S. indica symbiosis fortified with adequate P-fertilizer can prove to be effective in minimizing As acquisition and accumulation in rice plants.
Reduction of arsenic accumulation in rice grain by endophytic fungus Serendipita indica

Abstract:Arsenic contamination in rice is a major health concern that affects millions of people worldwide. Several strategies are recommended to reduce arsenic in rice, with AMF being one of the most important and cost-effective. This work is a pot study under environmental condition on rice plants under arsenic (AsV) treatment to demonstrate the ameliorative property of axenically cultivable AMF Serendipita indica ( S. indica ) in arsenic stress and accumulation in colonised plants. Colonisation of S. indica is 12 times higher under arsenic stress which is correlated to improved growth, biomass, and yield in fungal-colonised plants compared to arsenic-treated plants. Fungal colonisation results in a 1.9 times reduction in H2O2 content with the induction of the antioxidative enzyme system, proline and glutathione levels (up to 1.5 times). The fungal colonisation results in the recovery of pigment content (Chl a, Chl b and carotenoids) with improved photosynthesis, transpiration, and water use efficiency under arsenic stress. S. indica colonisation in plant roots resulted in a massive accumulation of arsenic (31.5 ppm) in the root leading to reduced arsenic mobilisation from the root to shoot, husk and grains. The restriction of arsenic in the root by the fungus was due to the modulation in iron (Fe) partitioning, resulting in the protection of the plant from arsenic stress and reduced level of arsenic in the grain. We show that using axenically cultivable fungus S. indica in highly arsenic-contaminated agricultural soil may reduce arsenic stress and accumulation in rice plants through modulation in Fe homeostasis.

Abstract:Arsenic (As) is a major soil contaminant that restricts plant growth and metabolism. Silicon-Si and arbuscular mycorrhizae-AM have the potential to impart As stress tolerance. The study assessed relative and cumulative effects of Si and mycorrhizal species; Claroideoglomus etunicatum -M1 and Rhizoglomus intraradices -M2 in alleviating AsV and AsIII toxicities in pigeonpea [ Cajanus cajan (L.) Millsp.]. Arsenic reduced mycorrhizal symbiosis, nutrient availability and plant biomass by inducing oxidative stress. Si and AM inoculations decreased metalloid uptake and reactive species generation, with higher benefits under M2, followed by Si, then M1. Higher efficiency of M2 corresponded to its colonization ability and glomalin production as well as its capability of enhancing nutrient uptake by modulating soil enzymes (alkaline phosphate-ALP and arylsulfatase-aryl-S) activities. In addition, S metabolism and antioxidant system got up-regulated, leading to increased synthesis of thiol compounds and antioxidants in the order M2>Si>M1. Accumulated thiols further reduced As translocation by chelating and immobilizing metalloid in plant roots. Moreover, significantly higher As tolerance could be recorded when plants were supplemented with a combination of +Si+M2, indicating functional complementarity between the two amendments. The study, thus, demonstrated introduction of Si and AM in the rhizosphere as a promising strategy for inducing As tolerance to pigeonpea.

Abstract:Bacteria-assisted phytoremediation uses bacteria to promote plant health and improve its ability to remediate toxic heavy metals like Arsenic (As). Here, we isolated rhizobacteria and identified them as Bacillus subtilis strain IU31 using 16S rDNA sequencing. IU31 showed phosphate solubilization potential on Pikovskaya agar medium and produced siderophores, which were detected on Chromium Azurol-S (CAS) agar medium. Indole-3-acetic acid (IAA) and gibberellins (GAs), namely GA 1 , GA 3 , GA 4 , GA 7 , GA 9 , GA 12 , GA 15 , and GA24, were quantified by GC/MS-SIM analysis. The expression levels of genes involved in GA and IAA biosynthesis, such as cyp112, cyp114 , trpA, and trpB , were assessed using semi-quantitative RT-PCR. Plant bioassays showed that As at a 15 mg/kg concentration reduced plant growth, chlorophyll content, and biomass. However, IU31 inoculation significantly improved plant growth dynamics, enhancing As accumulation by up to 50% compared with uninoculated plants. IU31 inoculation induced the bioconcentration factor (BCF) and bioaccumulation factor (BAF) of As in plants compared to uninoculated plants, but the translocation factor (TF) of As was unaffected by IU31 inoculation. IU31 inoculation effectively restored glutathione-S-transferase (GST) and catalase (CAT) enzyme activities, as well as glutathione (GSH) and hydrogen peroxide concentrations to nearly normal levels, which were significantly elevated in plants exposed to As stress. These results show that IU31 improves plant health and growth by producing IAA and GAs, which might contribute to the uptake and detoxification of As.

Abstract:Arsenic contamination of groundwater is a major concern for its usage in crop irrigation in many regions of the world. Arsenic is absorbed by rice plants mainly from arsenic contaminated water during irrigation. It hampers growth and agricultural productivity. The aim of the study was to mitigate the toxic effects of arsenate (As-V) [25 μM, 50 μM, and 75 μM] by silicon (Si) [2 mM] and selenium (Se) [5 μM] amendments on the activity of the TCA cycle, synthesis of γ-aminobutyric acid (GABA) and polyamines (PAs) in rice (Oryza sativa L. cv. MTU-1010) seedlings and to identify which chemical was more potential to combat this threat. As(V) application decreased the activities of tested respiratory enzymes and increased the levels of organic acids (OAs) in the test seedlings. Application of Si with As(V) and Se with As(V) increased the activities of respiratory enzymes and the levels of OAs. The effects were more pronounced during Si amendments. The activities of GABA synthesizing enzymes along with accumulation of GABA were increased under As(V) stress. During joint application of Si with As(V) and Se with As(V) the activity and the level of said parameters were decreased that indicating defensive role of these chemicals to resist As(V) toxicity in rice and Si amendments showed greater potential to reduce As(V) induced damages in the test seedlings. PAs trigger tolerance mechanism against As(V) in plants. PAs such as putrescine, spermidine and spermine were synthesized more during Si and Se amendments in As(V) contaminated rice seedlings to combat the toxic effects of As(V). Si amendments substantially modulated the toxic effects caused by As(V) over Se amendments in the As(V) challenged test seedlings. Thus, in future application of Si enriched fertilizer will be beneficial to grow rice plants with normal vigor in arsenic contaminated soil.

Abstract:This article examines envy as an important cultural link between inequality, institutions, development, and conflict. It argues that envy can be either a source of strife and stagnation or an engine for peaceful competition and growth. The fundamental conditions that activate the constructive side of envy and shut down its destructive side are access to productive investment opportunities, equality, security of property rights, and mild social comparisons. The dominant role of envy in society gives rise to a set of related cultural norms and beliefs that affect economic performance and social relations. While constructive envy is manifested in emulation or even envy-provocation—standard features of a consumer society—destructive envy produces a fear-of-envy culture that hampers economic incentives and creates an environment of suspicion and conflict.
Silicon-Mediated Arsenic Tolerance: Restriction of Arsenic Uptake and Modulation of Antioxidant Defense System in Rice Seedlings

Abstract:Arsenic, a toxic element has been infiltrating the food chain, raising the food safety and security issues. Due to phytotoxicity, mobility and availability of specific transporters for arsenic translocation, it is considered as serious agronomic threat to rice crop. To limit the detrimental effect of this arsenic stress, soil amendment with beneficial substances such as silicon can be helpful in acquiring arsenic tolerance by activating defense mechanism and subsequently improve seedlings vigor. However, its exact mechanism through which it enhances arsenic tolerance is not fully understood. The present study aims to evaluate the potential of exogenous application of silicon to alleviate toxic effects of arsenic stress and the associated morphological, biochemical and physiological mechanism in rice seedlings. In this study, rice seedlings of non-aromatic variety (IR-6) and aromatic variety (Super basmati) were grown in hydroponic system for 28 days. For arsenic stress, 25 μM and 50 μM sodium arsenite were applied along with 1 mM silicic acid (Treatment groups: As0, As25, As50, As25 + Si and As50 + Si). Results indicated that silicon application significantly decreased oxidative stress markers including hydrogen peroxide levels by 34.78%, lipid peroxidation by 20.06% consequently, decrease in relative membrane permeability by 37.79% at 25 μM arsenic stress in Super basmati as compared to IR-6. Furthermore, silicon application under 25 μM arsenic stress also activated enzymatic antioxidant defense system by elevating the activity of superoxide dismutase by 68.89%, guaiacol peroxidase by 53.59% and ascorbate peroxidase by 66.77% in Super basmati except catalase. Additionally, SEM images showed the anatomical adaptation as cuticle thickenings along with deposition of silica in silicon-treated plants. Silicon deposition was also evident by EDX spectra while arsenic was not detected in silicon supplemented rice seedlings under arsenic stress. Hence, these findings suggested that silicon application hinders arsenic translocation and arsenic toxicity. This study would serve as a step towards developing the approach for the rice grains with reduces arsenic content. This mitigation strategy will be helpful in environmental sustainability along with ensuring food security.

Abstract:Arsenic (As) is posing serious health concerns in South East Asia where rice, an efficient accumulator of As, is prominent crop. Salicylic acid (SA) is an important signaling molecule and plays a crucial role in resistance against biotic and abiotic stress in plants. In present study, ameliorative effect of SA against arsenate (As(V)) toxicity has been investigated in rice (Oryza sativa L.). Arsenate stress hampered the plant growth in terms of root, shoots length, and biomass as well as it enhanced the level of H2O2 and MDA in dose dependent manner in shoot. Exogenous application of SA, reverted the growth, and oxidative stress caused by As(V) and significantly decreased As translocation to the shoots. Level of As in shoot was positively correlated with the expression of OsLsi2, efflux transporter responsible for root to shoot translocation of As in the form of arsenite (As(III)). SA also overcame As(V) induced oxidative stress and modulated the activities of antioxidant enzymes in a differential manner in shoots. As treatment hampered the translocation of Fe in the shoot which was compensated by the SA treatment. The level of Fe in root and shoot was positively correlated with the transcript level of transporters responsible for the accumulation of Fe, OsNRAMP5, and OsFRDL1, in the root and shoot, respectively. Co-application of SA was more effective than pre-treatment for reducing As accumulation as well as imposed toxicity.

Abstract:The role of microorganisms in arsenic (As) uptake in association with terrestrial plants is identified, though their rhizosphere specific association and function in rice and barnyard grass in As contaminated soil are not elucidated. We investigated the abundance and diversity of arsenic and other heavy metal tolerant bacteria isolated from barnyard grass and rice rhizospheres, and the bulk soil from an As-contaminated paddy field. Among 83 isolated As resistant bacteria, 22 strains were screened and characterized based on their growth potential in 100 mM arsenate. One strain oxidized and reduced arsenic, whereas five isolates oxidized arsenite to arsenate and seven isolates reduced arsenate to arsenite. All the isolates were tolerant to the highest levels of Zn, Cd, Cr, Cu, Co, and Hg tested and except one, all also survived at the highest level of Pb. Molecular characterization by 16s rRNA gene analysis revealed that the selected bacteria belong to Sphingomonadaceae (13), Moraxellaceae (2), Enterobacteriaceae (2), Planococcaceae (2), Pseudomonadaceae (1), Xanthomonadaceae (1) and Cyanobacteria (1). Bacterial abundances were more in the rhizosphere than in bulk soil and diversity was more in barnyard grass rhizosphere than in rice. Among the isolates, 21 strains solubilized phosphate, produced indole acetic acid and grew in N-free medium. All the isolates that were tested, enhanced the germination and root fresh weight of rice seedlings. These isolates, based on their As and other metals resistance, As detoxification capacity, and plant growth promotion ability, especially the members of Sphingomonadaceae, could be conclusively used for plant growth promotion and microbe assisted remediation of the As and other heavy metal contaminated soil employing barnyard grass and rice.

Abstract:Rapid urbanization and globalization demand increasing agricultural productivity. Soil nutrient supply capacity is continuously decreasing due to soil erosion, degradation, salt deposition, undesired element, metal deposition, water scarcity, and an uneven nutrient delivery system. Rice cultivation requires a large amount of water which is becoming detrimental due to these activities. There is a need to increase its productivity. Microbial inoculants are becoming increasingly important in achieving sustainable agricultural production systems. The current study was conducted to investigate the interaction between the root endophytic fungus Serendipita indica ( S. indica ) and the actinobacterium Zhihengliuella sp. ISTPL4 (Z. sp. ISTPL4) and their synergistic effects on the growth of rice (Oryza sativa L). Both S. indica and Z. sp. ISTPL4 showed positive interactions. Growth of S. indica was observed at different days after Z. sp. ISTPL4 inoculation, and stimulated growth of S. indica was observed when Z. sp. ISTPL4 was inoculated at 5 dafi (days after fungal inoculation). Z. sp. ISTPL4 promoted the growth of S. indica as it increased spore germination. Furthermore, confocal and scanning electron microscopy (SEM) analyses showed a 27% increase in the spore size of S. indica in the presence of Z. sp. ISTPL4. In a liquid chromatography with tandem mass spectrometry (LC-MS/MS) analysis increased production of alanine and glutamic acid was observed in their sequential co-culture as compared with individual cultures. Sequential inoculation of S. indica and Z. sp. ISTPL4 significantly increased the biochemical and physical characteristics of rice as compared with their individual inoculum. Biochemical parameters such as chlorophyll content, total soluble sugar, and flavonoid content in the rice increased by up to 57%, 47%, and 39%, respectively, in the presence of the combined inoculum of S. indica and Z. sp. ISTPL4. This will be the first study, to the best of our knowledge, which shows the fungus and actinobacterium interaction and their synergistic roles in the growth promotion of rice. Furthermore, this novel combination can also be used to boost the growth of other crops to increase the agricultural yield.

Effect of Silicate Supplementation on the Alleviation of Arsenite Toxicity in 93-11 (oryza Sativa L. Indica).

Abstract:Chronic exposure to arsenic (As) in rice has raised many health and environmental problems. As reported, great variation exists among different rice genotypes in As uptake, translocation, and accumulation. Under hydroponic culture, we find that the Chinese wild rice (Oryza rufipogon; acc. 104624) takes up the most arsenic among tested genotypes. Of the cultivated rice, the indica cv. 93-11 has the lowest arsenic translocation factor value but accumulates the maximum concentration of arsenic followed by Nipponbare, Minghui 86, and Zhonghua 11. Higher level of arsenite concentration (50 μM) can induce extensive photosynthesis and root growth inhibition, and cause severe oxidative stress. Interestingly, external silicate (Si) supplementation has significantly increased the net photosynthetic rate, and promoted root elongation, as well as strongly ameliorated the oxidative stress by increasing the activities of antioxidant enzymes superoxide dismutase, ascorbate peroxidase, and peroxidase in roots and/or leaves of 93-11 seedlings. Notably, 1.873 mM concentration of Si considerably decreases the total As uptake and As content in roots, but significantly increases the As translocation from roots to shoots. In contrast, Si supplementation with 1.0 mM concentration significantly increases the total As uptake and As concentrations in roots and shoots of 93-11 seedlings after 50 μM arsenite treatment for 6 days.
Selenium treatment alters the accumulation of osmolytes in arsenic-stressed rice (Oryza sativa L.)

Abstract:Arsenic (As), one of the major pollutants in the soil, is an important environmental concern as its consumption can cause adverse health symptoms in living organisms. Its contamination of rice grown over As-contaminated areas is a serious concern in South Asian countries. Selenium (Se) has been reported to influence various osmolytes under metal stress in plants. The present study reports the role of Se in mitigating As stress in rice by modulating osmolyte metabolism. Rice plants grown in As-amended soil (2.5–10 mg kg −1 ) in pots were treated with sodium selenate (0.5–1.0 mg Se kg −1 soil) in glass house conditions and leaf samples were collected at 60 and 90 days after sowing (DAS). As-treated rice leaves displayed a reduction in relative water content (RWC) and dry weight than control with a maximum reduction of 1.68- and 2.47-fold in RWC and 1.95- and 1.69-fold in dry weight in As10 treatment at 60 and 90 DAS, respectively. Free amino acids (1.38–2.26-fold), proline (3.88–3.93-fold), glycine betaine (GB) (1.27–1.72-fold), choline (1.67–3.1-fold), total soluble sugars (1.29–1.61-fold), and reducing sugars (1.67–2.19-fold) increased in As-treated rice leaves as compared to control at both stages. As stress increased the γ-aminobutyric acid (GABA), putrescine content, and glutamate decarboxylase activity whereas diamine oxidase and polyamine oxidase activities declined by 1.69–1.88-fold and 1.52–1.86-fold, respectively. Se alone or in combination with As improved plant growth, RWC, GB, choline, putrescine, and sugars; lowered proline and GABA; and showed a reverse trend of enzyme activities related to their metabolism than respective As treatments. As stress resulted in a higher accumulation of osmolytes to combat its stress which was further modulated by the Se application. Hence, the current investigation suggested the role of osmoprotectants in Se-induced amelioration of As toxicity in rice plants.

Co-inoculation effects of B. licheniformis and P. aeruginosa on soil Cd and As availability and rice accumulation

Abstract:There have been studies reporting the effects of multiple bacterial strains on the Cd/As immobilization and transformation in culture media. However, there is limited research to validate the effects of microbial strain combination on plant Cd/As accumulation and antioxidant system in the soil-plant system. By planting the rice (Zhefu 7) with the co-inoculation of bacterial strains (i.e. Bacillus licheniformis and Pseudomonas aeruginosa) after two months with the contaminations of Cd (2 mg/kg), As (80 mg/kg) and Cd + As (2 + 80 mg/kg), we found that the bacterial co-inoculation decreased Cd concentrations in the rhizosphere soil porewater, but had limited effects on mitigating plant Cd accumulation. By contrast, the co-inoculation did not affect the As(III) and As(V) concentrations in the rhizosphere soil porewater, but decreased As(III) and As(V) concentrations by 17% and 17% in the root respectively and by 17% and 37% in rice shoot respectively. Using DNA sequencing, we found the increased abundance in both exogenous Bacillus licheniformis and native microorganisms, indicating that the added strains had synergetic interactions with soil native microorganisms. Regarding on plant antioxidant enzyme system, the bacterial co-inoculation decreased the concentrations of superoxide dismutase (SOD), hydrogen peroxide (H2O2) and malondialdehyde (MDA) by 75%, 74% and 22%, mitigating the As damage to rice root and promote plant growth. However, under Cd and As co-stress, the effects of co-inoculation on mitigating plant As accumulation and enhancing plant stress resistance appear to be diminished. Our findings underscore the importance of microbial co-inoculation in reducing plant As accumulation and preserving plant health under heavy metal stress.
Synergistic ameliorative effect of iron oxide nanoparticles and Bacillus subtilis S4 against arsenic toxicity in Cucurbita moschata: polyamines, antioxidants, and physiochemical studies

Abstract:The present study was intended to assess the potential of iron oxide nanoparticles (IONPs) and Bacillus subtilis S4 in mitigation of arsenic (As) stress in Cucurbita moschata. Cucurbita moschata seedlings were subjected to As stress for 60 days. Reduced level of growth parameters including photosynthetic pigments, rate of photosynthesis and gas exchange characteristics was observed in seedlings subjected to As stress. However, IONPs and B. subtilis S4 improved growth attributes and proline contents in supplemented C. moschata seedlings. Bacillus subtilis S4 inoculated seedlings showed higher activity of peroxidase (POD) and superoxide dismutase (SOD) under As toxicity. Similarly, the co-application of IONPs and B. subtilis S4 further increased the activity of these antioxidative enzymes. The As stress alleviation in inoculated C. moschata seedlings is credited to reduced levels of hydrogen peroxide (H2O2), malondialdehyde (MDA) and electrolyte leakage (EL) in IONPs and B. subtilis S4-treated plants. Furthermore, synergism between plant growth promoting bacteria (PGPB) and IONPs enhanced the biosynthesis of stress mitigating polyamines including spermidine and putrescine in As-stressed seedlings. Current research reveals that synergistic application of IONPs and B. subtilis S4 is an effective sustainable and ecofriendly approach for alleviation of As stress in C. moschata seedlings.
Exogenous γ-aminobutyric acid and Bacillus pumilus reduce arsenic uptake and toxicity in rice

Abstract:In this study, the addition of γ-aminobutyric acid (GABA), Bacillus pumilus, or both, was found to enhance rice growth and yield while significantly decreasing arsenic (As) accumulation in Oryza sativa rice tissues. GABA emerged as a regulator of iron (Fe) homeostasis, acting as a signaling modulator that influenced phytosiderophore secretions in the plant. Meanwhile, B. pumilus directly increased Fe levels through siderophore production, promoting the development of Fe-rich rice plants. Subsequently, Fe competed with As uptake at the root surface, leading to decreased As levels and translocation to the grains. Furthermore, the addition of GABA and B. pumilus optimized rice indole-3 acetic acid (IAA) contents, thereby adjusting cell metabolite balance under As stress. This adjustment results in low malondialdehyde (MDA) contents in the leaves and roots during the early and late vegetative phases, effectively reducing oxidative stress. When added to As-contaminated soil, GABA and B. pumilus effectively maintained endogenous GABA levels and exhibited low ROS generation, similar to normal soil. Concurrently, GABA and B. pumilus significantly downregulated the activity of OsLsi1, OsLsi2, and OsABCC1 in roots, reducing As uptake through roots, shoots, and grains, respectively. These findings suggest that GABA and B. pumilus additions impede As translocation through grains, ultimately enhancing rice productivity under As stress.

Abstract:The objective of this research was to determine the effect of zinc oxide nanoparticles (ZnONPs) and/or salicylic acid (SA) under arsenic (As) stress on rice (Oryza sativa). ZnONPs are analyzed for various techniques viz., X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), transmission electron microscopy (TEM), and scanning electron microscopy (SEM). All of these tests established that ZnONPs are pure with no internal defects, and can be potentially used in plant applications. Hence, we further investigated for better understanding of the underlying mechanisms and the extent of ZnONPs and SA induced oxidative stress damages. More restricted plant growth, gas exchange indices, significant reduction in the SPAD index and maximum quantum yield (Fv/Fm) and brutal decline in protein content were noticed in As-applied plants. In contrast, foliar fertigation of ZnONPs and/or SA to As-stressed rice plants lessens the oxidative stress, as exposed by subordinate levels of reactive oxygen species (ROS) synthesis. Improved enzymatic activities of catalase (CAT), peroxidase (POX), and superoxide dismutase (SOD), proline and total soluble protein contents under ZnONPs and SA treatment plays an excellent role in the regulation of various transcriptional pathways participated in oxidative stress tolerance. Higher content of nitrogen (N; 13%), phosphorus (P; 10%), potassium (K; 13%), zinc (Zn; 68%), manganese (Mn; 14%), and iron (Fe; 19) in ZnONPs and SA treated plants under As-stress, thus hampered growth and photosynthetic efficiency of rice plants. Our findings suggest that toxicity of As was conquering by the application of ZnONPs and SA in rice plants.
