Abstract:OBJECTIVE To present results from HIV testing, knowledge of HIV status and socioeconomic factors associated with the probability of having a HIV positive result among transwomen (TW) in Mexico. MATERIALS AND METHODS In 2012, we conducted an HIV seroprevalence survey to 585 TW in Mexico City in three strata: gathering places, the Condesa HIV Clinic and in four detention centers. We estimated the prevalence of HIV in each strata and applied a probit model to the overall sample to analyze factors associated with the probability of a HIV positive result. RESULTS The prevalence of HIV was 19.8% in meeting places; 31.9% in detention centers and 64% among the participants of the clinic. Age, low education and number of sexual partners was positively associated with HIV. CONCLUSIONS Results from the study provide relevant information to design HIV prevention interventions tailored to the needs of the TW population.
Overexpression of GmDREB1 Improves Salt Tolerance in Transgenic Wheat and Leaf Protein Response to High Salinity

Abstract:The transcription factor dehydration-responsive element binding protein（DREB）is able to improve tolerance to abiotic stress in plants by regulating the expression of downstream genes involved in environmental stress resistance.The objectives of this study were to evaluate the salt tolerance of GmDREB1 transgenic wheat（Triticum aestivum L.）and to evaluate its physiological and protein responses to salt stress.Compared with the wild type,the transgenic lines overexpressing GmDREB1 showed longer coleoptiles and radicles and a greater radicle number at the germination stage,as well as greater root length,fresh weight,and tiller number per plant at the seedling stage.The yield-related traits of transgenic lines were also improved compared with the wild type,indicating enhanced salt tolerance in transgenic lines overexpressing GmDREB1.Proteomics analysis revealed that osmotic-and oxidative-stressrelated proteins were up-regulated in transgenic wheat leaves under salt stress conditions.Transgenic wheat had higher levels of proline and betaine and lower levels of malondialdehyde and relative electrolyte leakage than the wild type.These results suggest that GmDREB1 regulates the expression of osmotic-and oxidative-stress-related proteins that reduce the occurrence of cell injury caused by high salinity,thus improving the salt tolerance of transgenic wheat.

Abstract:Under stress conditions such as drought, high-salinity and low-temperature, the transcription factor of DREB (dehydration responsive element binding proteins) improved efficiently stress resistance by regulating the expression of its downstream genes with various environmental stress resistance in plants.GmDREB gene (GenBank Accession No. AF514908) encoding a stress-inducible transcription factor was cloned by screening a cDNA library ofGlycine max cv. Jinong 27 with yeast one-hybrid method.GmDREB gene was 910 bp in length and encoded 174 amino acids containing a conserved AP2/EREBP DNA-binding domain of 58 amino acids. Two conserved functional amino acids, valine and glutamic acid, were located on the 14th and the 19th amino acid residues in the conserved structural domain. An alkaline amino acid region (KKR) related to a nuclear localization signal was at the N-terminal, while an acidic amino acid region (DDD) related totrans-activation was at the C-terminal. Plant expression vectors were constructed and transformed into wheat by bombardment. In total, 13 transgenic plants withUbi::GmDREB and 11 transgenic plants withrd29A::GmDREB were identified from 103 regeneration plants by molecular analysis. The drought and salt tolerances of T1 transgenic lines withUbi::GmDREB orrd29A::GmDREB were demonstrated to be improved as compared to wild type. The result also suggested that bothUbiquitin andrd29A promoters could effectively drive the expression of theGmDREB gene and enhance drought and salt tolerance of T1 plants.

Abstract:Plants have evolved complex signaling networks that enable them to adapt to adverse environmental conditions. The dehydration-responsive element-binding (DREB) transcription factors are important for plant responses to abiotic stresses. In this study, a new member of the AP2/ERF transcription factor gene family, PcDREB2A, was cloned and characterized from Polygonum cuspidatum, a traditional Chinese medicinal herb. PcDREB2A, which includes a typical AP2 domain, was clustered in the A-2 subgroup of the DREB subfamily. At the seedling stage, PcDREB2A expression was induced by cold, salt, and drought stresses. A yeast one-hybrid assay and an analysis of transiently transformed tobacco revealed that PcDREB2A can specifically bind to the DRE motif and transactivate reporter gene expression. Following 200 and 250 mM mannitol treatments, the PcDREB2A-overexpressing Arabidopsis thaliana lines had longer roots and a significantly higher fresh weight than the wild-type plants. Furthermore, under drought stress conditions, the PcDREB2A-overexpressing A. thaliana plants accumulated less malondialdehyde than the control plants. These results indicate that PcDREB2A encodes a novel DREB transcription factor in P. cuspidatum. Furthermore, the data generated in this study may be useful for researchers and breeders interested in genetically engineering plants to increase drought tolerance without inhibiting growth.

Abstract:Peroxisomal ascorbate peroxidase gene (SbpAPX) of an extreme halophyte Salicornia brachiata imparts abiotic stress endurance and plays a key role in the protection against oxidative stress. The cloned SbpAPX gene was transformed to local variety of peanut and about 100 transgenic plants were developed using optimized in vitro regeneration and Agrobacterium mediated genetic transformation method. The T0 transgenic plants were confirmed for the gene integration; grown under controlled condition in containment green house facility; seeds were harvested and T1 plants were raised. Transgenic plants (T1) were further confirmed by PCR using gene specific primers and histochemical GUS assay. About 40 transgenic plants (T1) were selected randomly and subjected for salt stress tolerance study. Transgenic plants remained green however non-transgenic plants showed bleaching and yellowish leaves under salt stress conditions. Under stress condition, transgenic plants continued normal growth and completed their life cycle. Transgenic peanut plants exhibited adequate tolerance under salt stress condition and thus could be explored for the cultivation in salt affected areas for the sustainable agriculture.

Abstract:Abiotic stresses comprise all nonliving factors, such as soil salinity, drought, extreme temperatures, and metal toxicity, posing a serious threat to agriculture and affecting the plant production around the world. Peanut ( Arachis hypogaea L.) is one of the most important crops for vegetable oil, proteins, minerals, and vitamins in the world. Therefore, it is of importance to understand the molecular mechanism of peanut against salt stress. Six transcriptome sequencing libraries including 24-hour salt treatments and control samples were constructed from the young leaves of peanut. A comprehensive analysis between two groups detected 3,425 differentially expressed genes (DEGs) including 2,013 upregulated genes and 1,412 downregulated genes. Of these DEGs, 141 transcription factors (TFs) mainly consisting of MYB, AP2/ERF, WRKY, bHLH, and HSF were identified in response to salinity stress. Further, GO categories of the DEGs highly related to regulation of cell growth, cell periphery, sustained external encapsulating structure, cell wall organization or biogenesis, antioxidant activity, and peroxidase activity were significantly enriched for upregulated DEGs. The function of downregulated DEGs was mainly enriched in regulation of metabolic processes, oxidoreductase activity, and catalytic activity. Fourteen DEGs with response to salt tolerance were validated by real-time PCR. Taken together, the identification of DEGs’ response to salt tolerance of cultivated peanut will provide a solid foundation for improving salt-tolerant peanut genetic manipulation in the future.

Abstract:The DREB transcription factors regulate multiple stress response genes, and are therefore useful for molecular plant breeding. AhDREB, a stress-inducible gene, was isolated from Atriplex hortensis L. and introduced into Populus tomentosa Carrière under the control of the CaMV35S promoter. Under salt stress, the chlorophyll content and net photosynthetic rate were higher in transgenic lines than in the wild type (WT). Moreover, the rate of electrolyte penetration (REC) was lower in the transgenic lines. Additional analyses revealed that the AhDREB transgenic plants generally displayed lower malondialdehyde (MDA) activity but higher superoxide dismutase (SOD) and peroxidase (POD) activities and proline content than the WT under salt stress. RNA sequencing indicated that AhDREB could enhance tolerance to salt by activating various downstream genes in the transgenic plants. Furthermore, no growth inhibition was detected in transgenic plants expressing AhDREB driven by the constitutive CaMV35S promoter. The transcriptome showed 165 and 52 differentially expressed genes in transgenic plants under stress and non-stress conditions, respectively, among which no significant metabolic pathway was enriched and no unintended effects have yet been identified. Together, these results suggest that AhDREB may be a good candidate gene for increasing salt tolerance in transgenic poplar breeding.

Abstract:The peanut (Arachis hypogaea) is an important crop species that is threatened by drought stress. The genome sequences of peanut, which was officially released in 2016, may help explain the molecular mechanisms that underlie drought tolerance in this species. We report here a gene expression profiling of A. hypogaea to gain a global view of its drought resistance. Using whole-transcriptome sequencing, we analysed differential gene expression in response to drought stress in the drought-resistant peanut cultivar J11. Pooled samples obtained at 6, 12, 18, 24, and 48 h were compared with control samples at 0 h. In total, 51,554 genes were found, including 49,289 known genes and 2265 unknown genes. We identified 224 differentially expressed transcription factors, 296,335 SNPs and 28,391 InDELs. In addition, we detected significant differences in the gene expression profiles of the treatment and control groups. After comparing the two groups, 4648 genes were identified. An in-depth analysis of the data revealed that a large number of genes were associated with drought stress, including transcription factors and genes involved in photosynthesis–antenna proteins, carbon metabolism and the citrate cycle. The results of this study provide insights into the diverse mechanisms that underlie the successful establishment of drought resistance in the peanut, thereby facilitating the identification of important genes in the peanut related to drought management. Transcriptome analysis based on RNA-Seq is a powerful approach for gene discovery and molecular marker development for this species.

Abstract:AbstractThe cultivation of rice (Oryza sativa L.), a major food crop, requires ample water (30 % of the fresh water available worldwide), and its productivity is greatly affected by drought, the most significant environmental factor. Much research has focussed on identifying quantitative trait loci, stress-regulated genes and transcription factors that will contribute towards the development of climate-resilient/tolerant crop plants in general and rice in particular. The transcription factor DREB1A, identified from the model plant Arabidopsis thaliana, has been reported to enhance stress tolerance against drought stress. We developed transgenic rice plants with AtDREB1A in the background of indica rice cultivar Samba Mahsuri through Agrobacterium-mediated transformation. The AtDREB1A gene was stably inherited and expressed in T1 and T2 plants and in subsequent generations, as indicated by the results of PCR, Southern blot and RT-PCR analyses. Expression of AtDREB1A was induced by drought stress in transgenic rice lines, which were highly tolerant to severe water deficit stress in both the vegetative and reproductive stages without affecting their morphological or agronomic traits. The physiological studies revealed that the expression of AtDREB1A was associated with an increased accumulation of the osmotic substance proline, maintenance of chlorophyll, increased relative water content and decreased ion leakage under drought stress. Most of the homozygous lines were highly tolerant to drought stress and showed significantly a higher grain yield and spikelet fertility relative to the nontransgenic control plants under both stressed and unstressed conditions. The improvement in drought stress tolerance in combination with agronomic traits is very essential in high premium indica rice cultivars, such as Samba Mahsuri, so that farmers can benefit in times of seasonal droughts and water scarcity.

Comparative Transcriptome Analyses Reveal Potential Mechanisms of Enhanced Drought Tolerance in Transgenic Salvia Miltiorrhiza Plants Expressing AtDREB1A from Arabidopsis

Abstract:In our previous study, drought-resistant transgenic plants of Salvia miltiorrhiza were produced via overexpression of the transcription factor AtDREB1A. To unravel the molecular mechanisms underpinning elevated drought tolerance in transgenic plants, in the present study we compared the global transcriptional profiles of wild-type (WT) and AtDREB1A-expressing transgenic plants using RNA-sequencing (RNA-seq). Using cluster analysis, we identified 3904 differentially expressed genes (DEGs). Compared with WT plants, 423 unigenes were up-regulated in pRD29A::AtDREB1A-31 before drought treatment, while 936 were down-regulated and 1580 and 1313 unigenes were up- and down-regulated after six days of drought. COG analysis revealed that the 'signal transduction mechanisms' category was highly enriched among these DEGs both before and after drought stress. Based on the Kyoto Encyclopedia of Genes and Genomes (KEGG) annotation, DEGs associated with "ribosome", "plant hormone signal transduction", photosynthesis", "plant-pathogen interaction", "glycolysis/gluconeogenesis" and "carbon fixation" are hypothesized to perform major functions in drought resistance in AtDREB1A-expressing transgenic plants. Furthermore, the number of DEGs associated with different transcription factors increased significantly after drought stress, especially the AP2/ERF, bZIP and MYB protein families. Taken together, this study substantially expands the transcriptomic information for S. miltiorrhiza and provides valuable clues for elucidating the mechanism of AtDREB1A-mediated drought tolerance in transgenic plants.

Abstract:Peanut(Arachis hypogaea) is an important crop,but it is badly affected by drought stress.The draft peanut genome sequence released in 2016 can be used to help understand the molecular mechanisms of drought tolerance in this species.Transcription factors (TFs) play central roles in defense responses in peanut.In this study,the differential expression of TFs in response to drought stress in the drought-resistant peanut cultivar Jll by RNA-seq was analyzed.Pooled samples obtained at 6,12,18,24,and 48 h were compared with control samples at 0 h.In total,this study identified 236 differentially expressed TFs based on the transcriptome data;142 were up-regulated and 94 were down-regulated.Among the differentially expressed TFs,members of the MYB and bHLH families were the most abundant.In up-regulated TFs group,the fold change range was from 2.034 1 to 43.968 3,and 69 TFs was＞10.In down-regulated TFs group,the fold change range was from-2.1031 to-11.6103,and 58 TFs was ＜-4.In this study,25 MYB genes were induced and 17 were down-regulated;17 bHLH genes were up-regulated and 14 were down-regulated;all NAC TFs was used to were up-regulated.In order to validate the gene expression data obtained by RNA-seq,qRT-PCR was used determine the expression levels of 14TF genes.Although the data obtained from qRT-PCR and RNA-seq were different,the trends in the expression levels determined by qRT-PCR were consistent with those obtained by RNA-Seq,thereby confirming the accuracy of the RNA-seq results.The results confirm the idea that drought resistance in peanut was largely dependent upon multiple TF families.The results of this study will provide a useful resource for further molecular research in this species.

Abstract:APETALA2/ethylene response element-binding factor (AP2/ERF) transcription factors (TFs) have been found to regulate plant growth and development and response to various abiotic stresses. However, detailed information of AP2/ERF genes in peanut against drought has not yet been performed. Herein, 185 AP2/ERF TF members were identified from the cultivated peanut (A. hypogaea cv. Tifrunner) genome, clustered into five subfamilies: AP2 (APETALA2), ERF (ethylene-responsive-element-binding), DREB (dehydration-responsive-element-binding), RAV (related to ABI3/VP), and Soloist (few unclassified factors)). Subsequently, the phylogenetic relationship, intron-exon structure, and chromosomal location of AhAP2/ERF were further characterized. All of these AhAP2/ERF genes were distributed unevenly across the 20 chromosomes, and 14 tandem and 85 segmental duplicated gene pairs were identified which originated from ancient duplication events. Gene evolution analysis showed that A. hypogaea cv. Tifrunner were separated 64.07 and 66.44 Mya from Medicago truncatula L. and Glycine max L., respectively. Promoter analysis discovered many cis-acting elements related to light, hormones, tissues, and stress responsiveness process. The protein interaction network predicted the exitance of functional interaction among families or subgroups. Expression profiles showed that genes from AP2, ERF, and dehydration-responsive-element-binding subfamilies were significantly upregulated under drought stress conditions. Our study laid a foundation and provided a panel of candidate AP2/ERF TFs for further functional validation to uplift breeding programs of drought-resistant peanut cultivars.

Abstract:OBJECTIVES We wished to determine the independent contribution of craniofacial dimensions of the upper airway to sleep-disordered breathing (SDB) in subjects who spanned the entire continuum of SDB. We also determined the interactive effects of body mass index (BMI) and age on the relationship between airway dimensions and SDB. DESIGN AND SUBJECTS We studied 142 nonclinical male subjects in a working community population (average age, 47 years; average BMI, 29; average +/- SD apnea/hypopnea index [AHI], 20 +/- 20/h), and 62 patients with obstructive sleep apnea (average age, 47 years; average BMI, 32; average +/- SD AHI, 48 +/- 35/h. We determined the AHI from overnight polysomnography and the number of oxygen desaturations (> or = 2%) per hour of sleep. We used lateral facial cephalometric radiographs to measure 41 anatomic landmarks and 55 dimensions in the upper airway. SETTING A university hospital and a sleep-disorders clinic. DATA ANALYSIS We used stepwise regression analysis to determine the independent contributions of measured variables to SDB. MEASUREMENTS AND RESULTS In the entire study population (n = 204), variations in BMI and six measures of craniofacial morphology accounted equally for one half of the total variance in AHI, and their interactive effects accounted for an additional 15%. Membership in the clinical or nonclinical group per se had no significant influence on these relationships. The single most important cephalometric variable in predicting AHI severity was the horizontal dimension of the maxilla (ie, porion vertical to supradentale [PV-A] distance). When the PV-A distance was relatively narrow (< 97 mm) the probability of having mild (AHI, 15 to 30/h) to severe (AHI > 30/h) SDB increased fivefold to sevenfold in nonobese subjects and threefold in obese subjects. Thus, in nonobese subjects (average BMI, 25 +/- 2) and in subjects with narrow upper airway dimensions, four cephalometric dimensions were the dominant predictors of AHI, accounting for 50% of the variance. However, in subjects with a large anteroposterior facial dimension, BMI was the major predictor of AHI and a BMI > 28 increased the probability of moderate-to-severe sleep apnea by approximately fivefold. Finally, the combination of cephalometric dimensions and BMI accounted for an increasing amount of the variance in AHI as the severity of AHI increased. CONCLUSIONS Across the population spectrum of SDB, four cephalometric dimensions of the upper airway in combination with BMI accounted independently for up to two thirds of the variation in AHI; and the relative contribution of these two sets of determinants of AHI varied depending on airway size, obesity, and the amount of SDB.

Abstract:Dehydration-responsive element-binding (DREB) transcription factors are important abiotic stress tolerance related genes, and some reports on the roles of DREB have primarily addressed herbal plants. To explore the abiotic stress tolerance role of DREB (ThDREB) from Tamarix hispida, a ThDREB gene with a complete ORF of 783 bp that encodes a 28.74 kDa protein with 260 amino acids, was isolated and functionally annotated. ThDREB expression was highly induced by NaCl, PEG, NaHCO3 and CdCl2 treatments, and the highest expression level (369.2-fold of control) was found for the roots that were under NaCl stress for 6 h. The tobacco plants that were transformed by ThDREB were conferred with higher germination rates, fresh weights and root lengths than the wild type (WT) tobacco plants under NaCl and mannitol treatments. The total chlorophyll content (tcc), superoxide dismutase (SOD) and peroxidase (POD) activities were also higher in the transgenic lines in comparison with the WT, and the malondialdehyde (MDA) and H2O2 content, electrolyte leakage (EL) rate and ROS as tracked by staining were generated to a lesser degree in ThDREB transgenic plants than in the WT under NaCl and mannitol stress. Furthermore, the transient overexpression analysis of ThDREB in T. hispida also improved plant salt and drought tolerance in comparison with the empty vector-transformed lines. Our results indicated that ThDREB expression could effectively improve tolerance to salt and drought stress by enhancing the antioxidase activity that keeps the ROS at a low accumulation level and makes them easy to scavenge.
