Abstract:The present study aimed to conduct a meta-analysis of previously published studies in order to clarify the association of long noncoding RNA (lncRNAs) LINC00673 rs11655237 C> T polymorphism with cancer risk.Systematic review and meta-analysis.Electronic databases of PubMed, EMBASE, Web of Science, Cochrane Library, Chinese National Knowledge Infrastructure, and Wanfang Database were used to search relevant studies. Studies published up to October 20, 2019 were included. The included studies were assessed in the following genetic model: allelic model, homozygote model, Heterozygote model, dominant model, recessive model. Data syntheses were conducted using STATA 12.0.Participants with various types cancers were included.Odds ratio (ORs) and 95% confidence interval (CIs) were calculated to assess the risk of tumor.Seven articles including 7 case-control studies, 7423 cases and 11,049 controls were adopted for meta-analysis. Our result demonstrated that LINC00673 rs11655237 C> T was related to the cancer among all model including allelic model (T vs C: pooled OR = 1.24, 95% CI = 1.16-1.41, P < .001), homozygous model (TT vs CC: pooled OR=1.54, 95% CI = 1.36-1.76, P < .001), heterozygous model (CT vs CC: pooled OR=1.24, 95% CI = 1.16-1.32, P < .001), dominant model (CT + TT vs CC: pooled OR=1.28, 95% CI = 1.20-1.36, P < .001) and recessive model (TT vs CT+ CC: pooled OR=1.42, 95% CI = 1.25-1.61, P < .001). Subgroup analysis also demonstrated that polymorphisms at this site also increased the risk of neuroblastoma.Our results find that rs11655237 contributed to occurrence of cancer in all models in Chinese population.
Association of 3 Common Polymorphisms of IL-27 Gene with Susceptibility to Cancer in Chinese: Evidence from an Updated Meta-Analysis of 27 Studies.

Abstract:Background: Many epidemiology studies have indicated that several functional polymorphisms of the IL-27 gene may contribute to individual susceptibility to cancer. Nevertheless, the data arising from these studies were inconclusive. Therefore, we conducted the current meta-analysis aiming to elucidate the effects of IL-27 polymorphisms (rs153109, rs17855750, and rs181206) on cancer susceptibility. Material/Methods: We searched the CNKI (Chinese National Knowledge Infrastructure), Wanfang database, PubMed, Web of Science, and Google Scholar for all eligible publications. We used odds ratios (ORs) corresponding with 95% confidence intervals (CIs) by using the random/fixed-effects model to evaluate the association. Finally, a total of 12 publications, including 27 case-control studies comprising of 7570 patients and 9839 controls, were enrolled in our meta-analysis. Results: Our work demonstrates that IL-27 rs17855750 polymorphism is significantly associated with cancer susceptibility, particularly for bladder cancer. However, no association between IL-27 rs153109 and rs181206 polymorphisms and cancer susceptibility was identified. When a stratification analysis was performed by cancer type, we identified an increased susceptibility of bladder cancer in rs153109 polymorphism. Moreover, in the stratification analysis by genotyping method, we identified an increased susceptibility for PCR-RFLP group in rs17855750 polymorphism, whereas a decreased susceptibility was identified in rs153109 polymorphism. Conclusions: Our study shows that IL-27 rs17855750 polymorphism is significantly associated with increased susceptibility to cancer in Chinese.
Effect of Polymorphism on IL1A to Cancer Susceptibility: Evidence Based on 34,016 Subjects

Abstract:IL-1α is closely related to the development and metastasis of cancer, and its polymorphisms have been reported affecting the susceptibility of malignancy tumors, yet the conclusions are controversial. Present an overall meta-analysis was performed to reach more general findings. Relevant literature was searched from Google Scholar, Web of Science, PubMed, EMBASE, CNKI database and Wanfang database, and the association among polymorphism and cancer risk was appraised by counting ORs and 95% CIs of overall and stratification analysis. The date from 15,586 cases and 18,430 controls in 40 publications were enrolled. There is significantly upregulated risk leads by rs3783553 in all genetic models, while the same tendency was found in all cancer types. The results also suggest a high risk of cancer susceptibility in patients carried rs1800587 polymorphism, of which draw out form allelic and homozygote models in overall studies, especially for cervical cancer. However, there are no significant results in analysis of rs17561. In a word, IL1A SNPs play an essential role in upregulating cancer susceptibility, and current analysis provides detail date for further study in the future.
Correlation of Interleukin-10-1082G/A Single Nucleotide Polymorphism to the Susceptibility of Bladder Cancer

Abstract:Objective To investigate the relationship between polymorphism at-1082G/A in the promoter region of the IL-10 gene and the risk of bladder cancer.Methods In a hospital based case-control study for the Chinese population in Zhejiang,a total of 400 patients with bladder cancer and 400 age-and sex-matched healthy controls subjects were collected.We analyzed the genotypes of IL-10-1082G/A by allele specific PCR amplification(AS-PCR) and DNA sequencing,and determined their association with bladder cancer risk and clinical stage.Results The genotype distribution and allele frequency of IL-10-1082G/A gene had significant differences between patient group and control group(χ2=8.375,P<0.05;χ2=8.322,P<0.01).Compared to the individual carrying AA genotype,the risk of bladder cancer with GG genotype and GA genotype increased 2.055 times(OR=2.055,95% CI:1.252-3.374).Compared to the individual carrying allele A,the risk of bladder cancer with allele G increased 1.990 times(OR=1.990,95% CI: 1.237-3.203).We did not observe any association between the genotypes of IL-10-1082G/A with the clinical stage of bladder cancer.Conclusion IL-10-1082G/A gene polymorphism may be a risk factor for bladder cancer and it may serve as a molecular marker for detecting bladder cancer.
Distinct effects of rs895819 on risk of different cancers: an update meta-analysis.

Abstract:Previous studies have indicated an association between the genetic variant in pre-miR-27a rs895819 with A->G transition and cancer risk; however, the results remain inconsistent and somehow conflicting in different cancers. Therefore, to obtain a more reliable conclusion, we performed an update meta-analysis by searching PubMed database or other databases. Odds ratio (ORs) and 95% confidence interval (CIs) were calculated to evaluate cancer risk. A total of 34 case-control studies involving 15,388 cases and 18,704 controls were included. The results showed that rs895819 was associated with an increased cancer risk (GG vs. AA/AG: OR = 1.15, 95% CI = 1.02-1.29). Furthermore, stratification analyses revealed an association of rs895819 with increased cancer risk among Asians (GG vs. AA: OR = 1.17, 95% CI = 1.01-1.36; GG vs. AA/AG: OR = 1.18, 95% CI = 1.03-1.35), but not Caucasians. Interestingly, the [G] allele of rs895819 was significantly associated with decreased risk of breast cancer (G vs. A: OR = 0.91, 95% CI = 0.86-0.97). However, rs895819 was associated with increased risk of colorectal cancer (GG vs. AA: OR = 1.56, 95% CI = 1.31-1.85; GG vs. AA/AG: OR = 1.53, 95% CI = 1.30-1.79; G vs. A: OR = 1.19, 95% CI = 1.09-1.30) and lung cancer (GG vs. AA/AG: OR = 1.43, 95% CI = 1.00-2.04). In addition, no association was found between rs895819 and risk of gastric cancer or esophageal cancer. In conclusion, our findings suggest distinct effects of rs895819 on risk of different cancers, and future well-designed studies with large samples are required to further validate our results.
Polymorphisms of Inflammation-Related Genes and Colorectal Cancer Risk: a Population-Based Case-Control Study in China.

Abstract:The previous studies found that chronic inflammation related to an increased risk of colorectal cancer (CRC). This study aims to explore the associations of polymorphisms in inflammation-related genes (IL10, IL10RA, IL6R, TNFRSF1A, TNFRSF1B, LTA and IL4) and their interactions with the risk of colorectal cancer among Chinese population. A population-based case-control study including 299 cases and 296 controls was conducted from January 2001 to December 2009. Multivariate unconditional logistic regression was used to analyse the association of nine SNPs in inflammation-related genes with the risk of CRC, colon cancer and rectal cancer, respectively. Generalized multifactor dimensionality reduction (GMDR) was implemented to explore the gene-gene interactions among all SNPs on CRC. A decreased risk of colorectal cancer in subjects with rs1800872 AC genotype of IL10 (OR = 0.643, 95%CI = 0.453, 0.912) or AC/CC genotype (OR = 0.636, 95%CI = 0.457, 0.885) was observed, compared with those with AA genotype. Meanwhile, similar associations were observed between rs1800872 and rectal cancer. Additionally, in rs1061624 of TNFRSF1B gene, AG genotype (OR=0.566; 95% CI= 0.362, 0.885) and AG/GG genotype (OR=0.638; 95% CI=0.420, 0.971) were significantly associated with a decreased risk of rectal cancer, respectively. Our findings indicated that mutants in IL10 and TNFRSF1B genes may change the CRC risk. However, there is no interaction between inflammation-related genes on CRC risk.
IL-8 –251A/T Polymorphism is Associated with Decreased Cancer Risk among Population-Based Studies: Evidence from a Meta-Analysis

Abstract:Growing evidence suggests that interleukin-8 (IL-8) play pivotal roles in the pathogenesis of cancer through the modulation of tumour immune response or enhanced angiogenesis. A single nucleotide polymorphism, –251A/T, has been identified in the promoter region of the IL-8 gene and has been shown to influence its production. Results from previous studies on the association of –251A/T polymorphism with different cancer types remained contradictory. To assess the effect of –251A/T of IL-8 on cancer susceptibility, we conducted a meta-analysis, up to May 2009, of 14,876 cases with different cancer types and 18,465 controls from 45 published case–control studies. Summary odds ratios and corresponding 95% confidence intervals (CIs) for IL-8 polymorphism and cancer were estimated using fixed- and random-effects models when appropriate. The AA/AT genotypes were associated with a significantly increased risk of nasopharyngeal carcinoma when compared with TT genotype (OR = 1.48; 95% CI, 1.16–1.89). Moreover, significantly elevated risks were observed in 'other cancers', and also in African population when population is concerned. Interestingly, when stratified separately by population-based studies and hospital-based studies, significantly elevated risk was found among hospital-based studies (OR = 1.21, 95% CI, 1.07–1.37), whereas significantly decreased risk was found among population-based studies (OR = 0.90, 95% CI, 0.83–0.97). This meta-analysis shows that IL-8 –251A/T polymorphism may play a complex role in cancer development.
Association between IL13 gene polymorphisms and susceptibility to cancer: a meta-analysis.

Abstract:Interleukin-13 (IL13) is an immunoregulatory cytokine which plays an important role in carcinogenesis through affecting tumor immunosurveillance. Many studies had reported the influence of IL13 rs1800925 and rs20541 polymorphisms on cancer risk, however, with inconclusive results. The aim of the present study was to conduct a meta-analysis to clarify the relationship.Twenty studies including a total of 6713 cancer cases and 8693 controls for IL13 rs20541 polymorphism and 4081 cancer cases and 6202 controls for IL13 rs1800925 polymorphism were included in the meta-analysis. Data were extracted from these studies and odds ratios with corresponding 95% confidence intervals were computed to estimate the strength of the association.Overall, the IL13 rs20541 polymorphism were associated with significantly decreased cancer risk in all genetic models (AA vs. GG: OR=0.82, 95%CI=0.71-0.95; GA vs. GG: OR=0.92, 95%CI=0.85-0.99; GA/AA vs. GG: OR=0.90, 95%CI=0.85-0.97; AA vs. GG/GA: OR=0.85, 95CI%=0.74-0.98). In the stratified analyses, significant effects were found among European populations, studies with population-based controls and studies of glioma. No influence of the IL13 rs1800925 polymorphism on the overall cancer risk was observed. However, in the stratified analyses, we found the IL13 rs1800925 polymorphism was significantly associated with decreased risk for glioma (CT vs. TT: OR=0.72, 95%CI=0.55-0.93; CT/TT vs. TT: OR=0.76, 95%CI=0.62-0.89).Our meta-analysis suggests that the IL13 rs20541 polymorphism contributes to susceptibility to cancer, especially for glioma; and the IL13 rs1800925 polymorphism may be associated with glioma risk.
Associations of Interleukin-6 Gene Polymorphisms with Cancer Risk: Evidence Based on 49,408 Cancer Cases and 61,790 Controls.

Abstract:Many molecular epidemiologic studies have shown that interleukin-6 (IL-6) polymorphisms are significantly associated with susceptibility for various cancers. However, the conclusions of these studies are inconsistent. The purpose of the present study was to explore the association between three common IL-6 loci (rs1800795, rs1800796, and rs1800797) and the risk for various cancers. We systematically searched the PubMed, Web of Science, Wanfang and China national knowledge infrastructure (CNKI) databases for relevant publications and obtained 108 eligible studies, involving 49,408 cancer patients and 61,790 cancer-free controls. Odds ratio (OR), 95% confidence interval (CI), and false positive reporting probability (FPRP) were used to evaluate cancer risk. All statistical analyses were performed using the R software meta package. We observed a non-significant association between rs1800795 and overall cancer risk, while rs1800797 was found to have a false positive association with overall risk of cancer. Subgroup analyses of rs1800797 also suggested non-significant association and rs1800795 played a protective role in liver cancer. Rs1800796 was found to be associated with overall cancer risk, particularly in Asian patients and those with prostate cancer. These findings provide evidence that IL-6 polymorphisms may affect cancer risk.
Associations of IL-17A -197G/A and IL-17F 7488T/C polymorphisms with cancer risk in asians: An updated meta-analysis from 43 studies

Abstract:OBJECTIVE: Numerous epidemiological studies have been published to elucidate the potential associations of IL-17A -197G/A (rs2275913) and IL-17F 7488T/C (rs763780) with cancer risk in Asians. Nevertheless, the results from different studies remain controversial. To identify the roles of the two polymorphisms in cancer risk, we performed this current meta-analysis.METHODS: The available literature was derived from five databases, covering relevant articles updated through February 17, 2021. Five different analysis models with corresponding odds ratios (ORs) and 95% confidence intervals (CIs) were applied to appraise the gene-disease correlation.RESULTS: In total, 43 case-control studies with 31,237 subjects were enrolled. Overall analyses indicated that there was significantly increased cancer risk led by IL-17A -197G/A under the five analysis models. A similar tendency was also identified in the subgroup analysis of cancer type, especially for gastric cancer, cervical cancer, colorectal cancer, and oral carcinoma. As for IL-17F 7488T/C, we revealed that patients who carried this variant had a higher cancer risk in the recessive model among the overall analyses, as well as subgroup analyses of cervical cancer or oral carcinoma.CONCLUSIONS: In summary, our work confirmed that IL-17A -197G/A acted as a risk factor for diverse cancer types and that IL-17F 7488T/C might be involved in cervical cancer and oral carcinoma.

Association of LINC00673 rs11655237 polymorphism with cancer susceptibility: A meta-analysis based on 23,478 subjects

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Background</h3><p>Some studies on the relationship between LINC00673 polymorphism and cancer susceptibility have been inconsistent. To perform a more comprehensively quantitative assessment of LINC00673 rs11655237 and risk of overall cancer, we operated this meta-analysis for the first time.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Methods</h3><p>A comprehensive search was conducted to obtain relevant literature up to November 20, 2019. Pooled odds ratios and 95% confidence intervals were utilized to assess rs11655237 and cancer susceptibility under five different genetic models.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>Eventually, 11 case-control studies from 9 articles were included. We found that LINC00673 rs11655237 polymorphism increased the susceptibility to overall cancer under all genetic models in the overall population. By dividing ethnicity and cancer type into subgroups, we also obtained similar positive results in subgroups of Chinese population, pancreatic cancer, cervical cancer, neuroblastoma, hepatoblastoma and gastric cancer.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusion</h3><p>Overall, this meta-analysis has demonstrated for the first time that LINC00673 rs11655237 could increase susceptibility to cancer.</p>

Association between the interleukin-17A -197G>A (rs2275913) polymorphism and risk of digestive cancer.

Abstract:Interleukin-17A (IL-17A) is a multifunctional cytokine which plays a crucial role in the initiation and progression of cancer. To date, several studies have investigated associations between IL-17A-197G>A (rs2275913) polymorphism and digestive cancer risk, but the results remain conflicting. We here aimed to confirm the role of this single nucleotide polymorphism (SNP) in susceptibility to digestive cancer through a systemic review and meta-analysis. Ten eligible case-control studies were identified by searching electronic databases, involving 3,087 cases and 3,815 controls. Odds ratios (ORs) and corresponding 95% confidence intervals (CIs) were used to estimate the strength of the association. The results of overall analyses indicated that the variant A allele was associated with an increased risk of digestive cancer (AA vs GG: OR=1.51, 95% CI=1.18-1.93; AA vs GG+GA: OR=1.45, 95% CI=1.12-1.87; A vs G: OR=1.21, 95% CI=1.05-1.39). In subgroup analysis stratified by specific cancer type, elevated risk among studies of gastric cancer was found (AA vs GG: OR=1.68, 95% CI=1.24-2.28; AA vs GG+GA: OR=1.62, 95% CI=1.16-2.26; A vs G: OR=1.23, 95% CI=1.04-1.46). According to ethnicity, there was evidence in the Asian populations for an association between this polymorphism and cancer risk (GA vs GG: OR=1.19, 95% CI=1.05-1.36; AA vs GG: OR=1.56, 95% CI=1.15-2.12; AA+GA vs GG: OR=1.28, 95% CI=1.131.44; AA vs GG+GA: OR=1.42, 95% CI=1.01-2.00; A vs G: OR=1.24, 95% CI=1.08-1.44), while in the Caucasian populations an association was found in the recessive model (AA vs GG+GA: OR=1.62, 95% CI=1.17-2.24). In conclusion, the results of this meta-analysis suggest that the IL-17A-197G>A polymorphism contributes to an increased risk of human digestive cancer, both in the Asian and Caucasian populations and especially for gastric cancer.
Role of interleukin-12 gene polymorphisms in the onset risk of cancer: a meta-analysis

Abstract:Many molecular epidemiologic studies have explored the possible links between interleukin-12 (IL-12) polymorphisms and various cancers. However, results from these studies remain inconsistent. This meta-analysis is aimed to shed light on the associations between three common loci (rs568408, rs2243115, rs3212227) of IL-12 gene and overall cancer risk. Our meta-analysis finally included 33 studies comprising 10,587 cancer cases and 12,040 cancer-free controls. Odds ratios (ORs) and 95% confidence intervals (CIs) were used to assess the cancer risk. We observed a significant association between IL-12B rs3212227 and overall cancer risk, especially in hepatocellular carcinoma, nasopharyngeal cancer, and among Asians. IL-12A polymorphisms (rs2243115 and rs568408) were found no influence on overall cancer risk. Nevertheless, stratification analyses demonstrated that rs568408 polymorphism contributed to increasing cancer risk of Caucasians and cervical cancer. And, rs2243115 may enhance the risk of brain tumor. These findings provided evidence that IL-12 polymorphisms may play a potential role in cancer risk.
Association Of A Pre-Mir-27a Polymorphism With Cancer Risk: An Updated Meta-Analysis

Abstract:MicroRNA-27a is highly expressed in cancers and has been identified as an oncogenic microRNA. A genetic variant in pre-miR-27a (rs895819) with a transition of A to G has been demonstrated to be associated with cancer risk; however, the results of these studies remain conflicting rather than conclusive. Therefore, we performed a meta-analysis to derive a more precise estimation. Through searching PubMed or other databases up to March 2014 using the following MeSH terms and keywords, "miR-27a", "polymorphism" and "cancer", seventeen case-control studies were identified in this meta-analysis, including 7,813 cases and 9,602. Crude odds ratios (ORs) and corresponding 95% confidence intervals (CIs) were calculated to investigate the association strength between rs895819 and the susceptibility of cancer. The results of the overall meta-analysis did not suggest any association between rs895819 polymorphism and cancer susceptibility, and this remained in Asians as a subgroup. In Caucasians, however, the rs895819 was associated with a reduced cancer risk in heterozygous (OR, 0.83; 95% CI, 0.75-0.93) and dominant models (OR, 0.84; 95% CI, 0.76-0.93), and the [G] allele of rs895819 showed a protective effect (OR, 0.90, 95% CI, 0.84-0.97). Further studies showed a significant association between the [G] allele of rs895819 and decreased risk of breast cancer (0.91; 95% CI, 0.85-0.98), and stratified analyses indicated a protective effect of the [G] allele in Caucasians (OR, 0.89; 95% CI, 0.82-0.98), younger breast cancer cases (OR, 0.87; 95% CI, 0.79-0.96), and in the group of unilateral breast cancer patients (OR, 0.90; 95% CI, 0.83-0.97). These findings suggest an association between pre-miR-27a polymorphism rs895819 and cancer risk in Caucasians. The protective effect of rs895819 [G] allele in younger breast cancer and in the group of unilateral breast cancer patients await further confirmation since the included studies in this meta-analysis were limited.
Association Ofil-17Polymorphisms with Gastric Cancer Risk in Asian Populations

Abstract:AIM:To investigate associations between the IL-17 rs2275913 G>A and rs763780 T>C polymorphisms and susceptibility to gastric cancer in Asian populations. METHODS:We reviewed studies published up to 2014 on IL-17 polymorphisms with gastric cancer susceptibility systematically. Relevant articles were identified in the MEDLINE, Science Citation Index, Cochrane Library, PubMed, EMBASE, CINAHL and Current Contents Index databases. We used version 12.0 STATA statistical software to evaluate the statistical data. Two reviewers abstracted the data independently. Odds ratios (ORs) and 95% confidence intervals (95%CIs) were calculated. RESULTS:Seven independent, case-control studies were chosen for the meta-analysis, which included 3210 gastric cancer patients and 3889 healthy controls. The overall estimation showed a positive association between the IL-17 rs2275913 G>A polymorphism and the occurrence of gastric cancer for five genetic models (all P < 0.05) and similar results were observed for the IL-17 rs763780 T>C variation with four genetic models (all P < 0.05), but not for the dominant model (P > 0.05). Subgroup analysis by country revealed that the rs2275913 G>A and rs763780 T>C polymorphisms may be the main risk factor for gastric cancer in Chinese and Japanese populations. CONCLUSION:The IL-17 gene may be significantly correlated with gastric cancer risk in Asian populations, especially those carrying the rs2275913 G>A and rs763780 T>C polymorphisms.
A Meta-Analysis of Association Between Interleukin Polymorphisms (rs4073, rs1800925, rs1179251, rs1179246, rs2227485, rs17855750, and rs153109) and Colorectal Cancer Risk

Abstract:Interleukins (ILs) play a significant role in triggering the inflammatory response in blood vessels and immune cells. A systematic review and meta-analysis were conducted to investigate the relationship between IL-8 (rs4073), IL-13 (rs1800925), IL-22 (rs1179251, rs1179246, and rs2227485), and IL-27 (rs17855750 and rs153109) polymorphisms and the risk of developing colorectal cancer (CRC). Four databases were searched up until October 13, 2023, without any restrictions, to find relevant studies. The association was evaluated using crude odds ratios (ORs) and 95% confidence intervals in five genetic models. A total of twenty-three articles were entered into the meta-analysis. The pooled ORs ( p -values) for the IL-8 (rs4073) polymorphism were 0.98 (0.63), 0.93 (0.44), 0.89 (0.13), 0.94 (0.38), and 0.99 (0.90) for studies following HWE without heterogeneity, and for all studies with high heterogeneity were 1.03 (0.69), 1.30 (0.07), 1.04 (0.71), 1.12 (0.20), and 1.23 (0.06). For the IL-13 (rs1800925 ) polymorphism, the pooled ORs were 1.44 (0.06), 2.58 (0.0004), 1.72 (0.16), 1.82 (0.09), and 2.37 (0.001) in AHHDR models, respectively. The pooled ORs of IL-22 (rs1179251) polymorphism for AHHDR models were 0.97 (0.92), 0.92 (0.90), 0.98 ( p = 0.95), 1.08 (0.87), and 0.96 (0.82), respectively. The pooled ORs of IL-22 (rs1179246) polymorphism for AHHDR models were 0.98 (0.67), 0.97 (0.80), 0.92 (0.36), 0.93 (0.42), and 1.02 (0.84), respectively. The pooled ORs of IL-22 (rs2227485) polymorphism for AHHDR models were 1.47 (0.02), 2.03 (0.02), 1.28 (0.29), 1.52 (0.06), and 1.70 (0.04), respectively. The pooled ORs of IL-27 (rs17855750) polymorphism for AHHDR models were 0.53 (0.46), 0.19 (0.28), 1.10 (0.60), 0.55 (0.58), and 0.27 ( p = 0.05), respectively. The pooled ORs of IL-27 (rs153109) polymorphism for AHHDR models were 1.28 (0.007), 1.45 (0.002), 1.40 (0.0002), 1.41 (< 0.0001), and 1.20 (0.09), respectively. The results reported that just the TT genotype of IL-13 (rs1800925), the T allele and TT genotype of IL-22 (rs2227485), and the G allele and GG, AG and GG + AG genotypes of IL-27 (rs153109) polymorphisms had an elevated risk in CRC patients.

Abstract:Background: Interleukin-1 beta (IL-1 beta), a pro-inflammatory cytokine, is emerging as a key mediator of carcinogenesis that characterizes host-environment interactions. Epidemiological studies investigating the association between two polymorphisms of IL-1B (-511C/T and +3954C/T) and cancer susceptibility have shown conflicting results. The aim of this study is to derive a more precise estimation of the relationship.Methods: Related studies were identified through a systematic literature search of PubMed and Web of Science from their inception to September 15, 2012. Summary odds ratios (ORs) and 95% confidence intervals (CIs) for the IL-1B -511C/T and +3954C/T polymorphisms and cancer risk were calculated. Heterogeneity among studies and publication bias were also tested.Results: The meta-analysis included 91 case-control studies in 85 publications, 81 studies for the -511C/T (19547 cases and 23935 controls) and 26 studies for the +3954C/T polymorphisms (8083 cases and 9183). The pooled results indicated that IL-1B +3954C/T (dominant model: OR = 1.15, 95% CI: 1.01-1.30) was significantly associated with increased overall cancer risk, especially among hospital-based case-control studies (dominant model: OR = 1.30, 95% CI: 1.02-1.66). As for -511C/T, we observed an inverse relationship in cervical cancer (dominant model: OR = 1.74, 95% CI: 1.35-2.23) and hepatocellular carcinoma (dominant model: OR = 0.68, 95% CI: 0.47-0.99). Moreover, -511C/T was associated with risk of specific subtypes of gastric carcinoma.Conclusion: This meta-analysis suggested that both the IL-1B -511C/T and +3954C/T polymorphisms might modulate cancer susceptibility. Further well-designed studies based on larger sample sizes should be performed to confirm the findings.
Role of IL-17F T7488C Polymorphism in Carcinogenesis: a Meta-Analysis

Abstract:Previous case-control studies on the association of interleukin-17F (IL-17F) T7488C polymorphism and cancer risk have yielded conflicting and inconclusive findings. We performed a meta-analysis by pooling all currently available data to acquire a more precise estimation of the association. A comprehensive literature screening from the PubMed, Embase, Web of Science, and Wanfang databases was performed for eligible publications without language restrictions. The pooled odds ratios (ORs) with corresponding 95 % confidence intervals (95 % CIs) were calculated. According to the inclusion criteria, a total of nine case-control studies with 3,034 cases and 3,694 controls were included. Overall, the pooled ORs showed that IL-17F T7488C polymorphism was associated with neither increased nor decreased risk of cancer. However, the IL-17F T7488C polymorphism exerted risk effect on cancer in population-based case-control studies when stratifying analysis by source of controls (C vs T OR = 1.24, 95 % CI, 1.10–1.40, pooled OR (POR) < 0.001; TC vs TT OR = 1.28, 95 % CI, 1.11–1.48, POR = 0.001; CC + TC vs TT OR = 1.29, 95 % CI, 1.12–1.48, POR < 0.001). Additionally, the variant genotypes of IL-17F T7488C could alter the risk of gastric cancer under the following comparisons (C vs T OR = 1.29, 95 % CI, 1.13–1.47, POR < 0.001; TC vs TT OR = 1.35, 95 % CI, 1.14–1.60, POR < 0.001; CC + TC vs TT OR = 1.35, 95 % CI, 1.15–1.58, POR < 0.001). Sensitivity analysis by sequential omission of single study did not materially alter the pooled findings. The present meta-analysis suggests that the IL-17F T7488C polymorphism may modify the risk of cancer, particularly gastric cancer. However, the precise association needs to be elucidated by more individual studies with sufficient statistical power.
Review and pooled analysis of studies on -607(C/A) and -137(G/C) polymorphisms in IL-18 and cancer risk

Abstract:Interleukin-18 (IL-18) is a pleiotropic, pro-inflammatory cytokine with dual effects on tumor development and progression. The -607(C/A) and -137(G/C) polymorphisms in IL-18 gene region have been implicated in cancer risk; however, data from published studies with individually low statistical power are conflicting. To clarify the role of IL-18 -607(C/A) and -137(G/C) genotype in global cancer, we examined all the available published studies through a pooled analysis approach. Overall, IL-18 -607A allele was associated with increased total cancer risk when compared with -607C allele (OR = 1.14, 95% CI = 1.01–1.28, P = 0.010), as well as in the heterozygote comparison (OR = 1.10, 95% CI = 1.04–1.15, P = 0.256) and the dominant model (OR = 1.07, 95% CI = 1.03–1.11, P = 0.124). Furthermore, IL-18 -137(G/C) polymorphism was associated with increased nasopharyngeal carcinoma risk. In the stratified analysis for -607(C/A) polymorphism, a significantly increased cancer risk in Asian population was found, as well as subgroup in source of control. Similar results were found in the stratified analysis for -137(G/C) polymorphism. Our pooled analysis supported that IL-18 is a good candidate for large-scale epidemiological case–control studies that may be a low-penetrance susceptibility biomarker for cancer.

Abstract:Introduction: It has been suggested that allergies are inversely associated with glioma risk. Single nucleotide polymorphisms in two allergy-related genes [interleukin (IL)-4Rα, IL-13] have been implicated in susceptibility to glioma; however, results from the published studies remained inconclusive. Methods: To derive a more precise relationship, we conducted a meta-analysis including seven case–control studies that investigated the influence of IL-4Rα rs1801275 and IL13 rs20541 polymorphisms on glioma risk. Data were extracted from these studies and pooled odds ratios (OR) with 95% confidence intervals (CI) were used to investigate the strength of the association. Results: Overall, the pooled analysis showed that there was no significant association between the IL-4Rα rs1801275 polymorphism and glioma risk (OR=0.99, 95%CI: 0.79–1.25, AG/GG vs. AA). However, we found that the IL13 rs20541 variant genotypes (GA/AA) were significantly associated with reduced risk for glioma (OR=0.85, 95%CI: 0.75–0.97, GA/AA vs. GG). In the stratified analyses by ethnicity, marginally significant association between the IL13 rs20541 polymorphism and decreased glioma risk was found among Asian populations in dominant models (OR=0.84, 95%CI: 0.70–1.00, GA/AA vs. GG). Conclusions: This meta-analysis suggests that the IL13 rs20541 but not the IL-4Rα rs1801275 polymorphism may be a genetic predictor for glioma. More studies with larger sample size are warranted to further elucidate the impact of the IL13 rs20541 polymorphism on glioma risk.
