Abstract:Nanoparticles exhibiting diverse shapes, high porosity and chemical stability reveal, upon appropriate chemical engineering, enzyme-like catalytic activities, "nanozymes", providing a plethora of nanomaterials for diverse applications. The present review article addresses the sensing applications of the catalytic functions of nanozymes consisting of metal nanoparticles, metal oxides, metal sulfides and cyanometallate nanoparticles, carbon-based nanomaterials and metal-organic-framework nanoparticles. The nanozymes emulate catalytic functions of oxidases or peroxidases and are employed as amplifying agents for sensing diverse analytes such as glucose, dopamine, NADH, thiols, phosphates and more. Moreover, the immobilization of nanozymes on electrodes provides versatile means to develop electrochemical sensing platforms. Different principles of the electrochemical sensing platforms, synthetic methodologies to deposit nanozymes on electrodes, and methods to establish electrical communication between the bulk conductive support and nanozyme particles are introduced. Electrochemical sensing platforms applying nanozyme-modified electrodes for the detection of analytes such as organophosphates, glucose and more are discussed. In particular, the application of nanozymes as amplifying labels for biosensor devices detecting proteins, DNA and microRNAs are addressed. Finally, the uses of nanozymes as functional constituents to design sense-and-treat systems are discussed. This is exemplified with the assembly of a bioreactor system for the sensing of glucose, the nanozyme-promoted generation of reactive oxygen species as cytotoxic agents towards cancer cells, and the autonomous nanozyme-based glucose-controlled release of insulin from nanocarrier devices. The future challenges in developing nanozyme-based sensors and sense-and-treat systems are presented.

Abstract:Nanozymes are a class of artificial enzymes that have dimensions in the nanometer range and can be composed of simple metal and metal oxide nanoparticles, metal nanoclusters, dots (both quantum and carbon), nanotubes, nanowires, or multiple metal-organic frameworks (MOFs). They exhibit excellent catalytic activities with low cost, high operational robustness, and a stable shelf-life. More importantly, they are amenable to modifications that can change their surface structures and increase the range of their applications. There are three main classes of nanozymes including the peroxidase-like, the oxidase-like, and the antioxidant nanozymes. Each of these classes catalyzes a specific group of reactions. With the development of nanoscience and nanotechnology, the variety of applications for nanozymes in diverse fields has expanded dramatically, with the most popular applications in biosensing. Nanozyme-based novel biosensors have been designed to detect ions, small molecules, nucleic acids, proteins, and cancer cells. The current review focuses on the catalytic mechanism of nanozymes, their application in biosensing, and the identification of future directions for the field.

Abstract:The introduction of functional nanomaterials into biosensing fields advanced analytical chemistry and opened a series of concepts for design of new analytical and tracing tools. It not only improved the stability and biocompatibility of biosensing interfaces, also has greatly enhanced the biosensing response to recognition event, thus met the need for detection of trace amounts of a wide variety of analytes and acquisition of ultraweak biological signals, and promoted the development of life science, biomedicine, food and environmental science. Different nanomaterials have been functionalized to modify the biosensing interfaces for accelerating the electron transfer, lowing the activation energy, and/or concentrating the target analytes, or to act as nanoprobes for labeling the recognition molecules, in which the nanomaterials can act as catalysts, enzyme mimics, signal emitters or carriers for loading of signal molecules to produce amplified signals. By coupling with the specificity of biological recognition events, the functional nanomaterials and nanoprobes have been quickly developed for highly sensitive and specific biosensing both in vitro and in vivo. This review focuses on the significant advances in use of functional nanomaterials for design of amplified biosensing strategies. Abstract:In comparison with bioenzymes, nanozymes exhibit excellent robustness against extreme conditions, a low production cost, and easy-to-adjust properties, as well as potential versatility. These superiorities have attracted abundant interest in the last 15 years, to develop various nanozymes for applications including analytical sensing, environmental engineering, and biomedicine. In particular, for analytical sensing, a lot of nanozyme-involved principles and methods have been explored and applied to clinical diagnosis, environmental monitoring, food safety detection, and forensic analysis. Moreover, rational exploitation and use of nanozyme materials promote the performance of analytical methods. To highlight the latest progress in this attractive field, recent design concepts of nanozymes for advanced biochemical sensing are summarized. The development of single-atom nanozymes, self-cascade nanozymes, structurally biomimetic nanozymes, molecularly imprinted nanozymes, nanozymes breaking the pH limit, and multifunctional nanozymes is discussed in detail, to enhance detection sensitivity and selectivity, as well as expand application scenarios. Finally, some challenges and trends related to nanozyme-based sensors are reported, to satisfy the increasing needs of biochemical analysis with nanozymes.

Abstract:Nanozymes are nanomaterials that exhibit enzyme-like catalytic activity, which have drawn increasing attention on account of their unique superiorities including very high robustness, low cost, and ease of modification. Metal–organic frameworks (MOFs) and covalent–organic frameworks (COFs) have emerged as promising candidates for nanozymes due to their abundant catalytic activity centers, inherent porosity, and tunable chemical functionalities. In this review, we first compare the enzyme-mimicking activity centers and catalytic mechanisms between MOF and COF nanozymes, and then summarize the recent research on designing and modifying MOF and COF nanozymes with inherent catalytic activity. Moreover, typical examples of sensing applications based on these nanozymes are presented, as well as the translation of enzyme catalytic activity into a visible signal response. At last, a discussion of current challenges is presented, followed by some future prospects to provide guidance for designing nanozyme sensors based on MOFs and COFs for practical applications.

Abstract:This review summarizes recent progress in the development of biosensors by integrating functional DNA molecules with different types of nanomaterials, including metallic nanoparticles, semiconductor nanoparticles, magnetic nanoparticles, and carbon nanotubes. On one hand, advances in nanoscale science and technology have generated nanomaterials with unique optical, electrical, magnetic and catalytic properties. On the other hand, recent progress in biology has resulted in functional DNAs, a new class of DNAs that can either bind to a target molecule (known as aptamers) or perform catalytic reactions (known as DNAzymes) with the ability to recognize a broad range of targets from metal ions to organic molecules, proteins and cells specifically. By taking advantage of the strengths in both fields, the physical and chemical properties of nanomaterials have been modulated by the target recognition and catalytic activity of functional DNAs in the presence of a target analyte, resulting in a large number of colorimetric, fluorescent, electrochemical, surface-enhanced Raman scattering and magnetic resonance imaging sensors for the detection of a broad range of analytes with high sensitivity and selectivity.

Abstract:DNAzymes, screened through in vitro selection, were artificial nucleic acids with catalytic function. They could cleave specific substrates in the presence of cofactors with unique characteristics, such as high catalytic activity, high specificity for cofactors, excellent stability, and easy to synthesize and modify. The combination of DNAzymes with nanomaterials could retain the DNAzyme activity and realize the functional integration of recognition and signal transduction, promoting rapid development of biosensors. In the current paper, we have mainly reviewed the recent progress of DNAzyme-nanomaterial based biosensors, and the nanomaterials included gold nanoparticles, graphene, quantum dots, magnetic nanomaterials, and so on.

Abstract:Given the superiorities in catalytic stability, production cost and performance tunability over natural bio-enzymes, artificial nanomaterials featuring enzyme-like characteristics (nanozymes) have drawn extensive attention from the academic community in the past decade. With these merits, they are intensively tested for sensing, biomedicine and environmental engineering. Especially in the analytical sensing field, enzyme mimics have found wide use for biochemical detection, environmental monitoring and food analysis. More fascinatingly, rational design enables one fabrication of enzyme-like materials with versatile activities, which show great promise for further advancement of the nanozyme-involved biochemical sensing field. To understand the progress in such an exciting field, here we offer a review of nanozymes with multiple catalytic activities and their analytical application prospects. The main types of enzyme-mimetic activities are first introduced, followed by a summary of current strategies that can be employed to design multi-activity nanozymes. In particular, typical materials with at least two enzyme-like activities are reviewed. Finally, opportunities for multi-activity nanozymes applied in the sensing field are discussed, and potential challenges are also presented, to better guide the development of analytical methods and sensors using nanozymes with different catalytic features.

Abstract:In the past few decades, researchers have developed lots of artificial enzymes with various materials to mimic the structures and functions of natural enzymes. Recently, nanozymes, nanomaterials with enzyme-like characteristics, are emerging as novel artificial enzymes, and attracting researchers' enormous interest. Remarkable advances have been made in the area of nanozymes due to their unique properties compared with natural enzymes and classic artificial enzymes. Until now, lots of nanomaterials have been studied to mimic various natural enzymes for wide applications. To highlight the recent progress of nanozymes (especially in bionanotechnology), here we discuss the diverse applications of nanozymes, which range from sensing, imaging, and therapeutics, to logic gates, pollutant removal, water treatment, etc. Finally, we address the current challenges facing nanozyme research as well as possible directions to fulfill their great potential in future.

Abstract:Nanozymes are nanomaterials with enzyme-mimicking catalytic activity. Compared to natural enzymes, nanozymes show various properties such as easy to manufacture, stable, adjustable, and inexpensive. Nanozymes play key roles in biosensing, biocatalysis, and disease treatment. As an important kind of nanozymes, metal-organic framework (MOF)-based nanozymes are receiving a lot of attention due to their structural properties and composition. Rationally developing MOF with enzymes-like catalytic properties has opened new perspectives in biosensing. This review summarizes the up-to-date developments in synthesizing two-dimensional and three-dimensional MOF-based nanozymes and their applications in biosensing. Firstly, classification of nanozymes obtained by MOFs is categorized, and different properties of MOF-based nanozymes are described. Then, the distinctive applications of MOF-based nanozymes in identifying various analytes are thoroughly summarized. Finally, the recent challenges and progressive directions in this area are highlighted.

Abstract:The first decade of the 21st century has been labeled as “the sensing decade”. The functional nanomaterials offer excellent platforms for fabrication of sensitive biosensing devices, including optical and electronic biosensors. A lot of works have focused on the biofunctionalization of different nanomaterials, such as metal nanoparticles, semiconductor nanoparticles and carbon nanostructures, by physical adsorption, electrostatic binding, specific recognition or covalent coupling. These biofunctionalized nanomaterials can be used as catalysts, electronic conductors, optical emitters, carriers or tracers to obtain the amplified detection signal and the stabilized recognition probes or biosensing interface. The designed signal amplification strategies have greatly promoted the development of stable, specific, selective and sensitive biosensors in different fields. This review introduces some novel principles and detection strategies in the area of biosensing, based on functional nanomaterials. The general methods for biofunctionalization of nanomaterials with biomolecules and their biosensing application in immunoassay of protein, DNA detection, carbohydrate analysis and cytosensing are also described.

Abstract:<p>The past decade has seen a period for the rapid development of nanozymes. As a class of nanoscale materials with some enzyme-like properties, nanozymes not only possess catalytic activity comparable to that of natural enzymes with the help of advanced technologies, but also have unique superiorities including excellent robustness in harsh environments, large-scale production, low cost, and ease of performance modulation. These advantages make them a promising generation of artificial enzymes for various applications. Especially in the analytical field, the nature of nanozyme catalysis offers amplified signals for high-sensitivity detection. However, most of currently developed nanozymes lack essential catalytic specificity, which becomes one of their biggest challenges in sensing. How to realize selective detection turns to be a vital problem in the nanozyme analytical community. In this review, we first summarize strategies available to make nanozymes work for the analytical application with good specificity. Then, typical examples utilizing these strategies to achieve selective detection are introduced, followed by the discussion of their merits as well as challenges. In the end, trends of nanozymes in this promising area are presented, to provide some guides for designing nanozyme sensing methods and devices for practical applications.</p>

Abstract:In recent years, the development of highly sensitive sensors has become a popular research topic. Some functional nanomaterials occupy an important position in the sensing field by virtue of their unique structures and catalytic properties, but there are still problems such as low sensitivity and poor specificity. Single-atom nanomaterials (SANs) show significant advantages in amplifying sensing signals and improving sensor interference resistance due to their high atomic utilization, structural simplicity, and homogeneity. They are expected to achieve high sensitivity and high specificity monitoring by modulating the active sites. In this review, the recent progress on SANs for electrochemical sensing applications was summarized. We first briefly summarize the features and advantages of single-atom catalysts. Then recent advances in the regulation of reaction sites in noble and non-noble metal-based SANs, including the introduction of defects in the carrier, other metal atoms, and ligand atoms, were highlighted. After that, the SANs for the construction of electrochemical, electrochemiluminescent (ECL), and photoelectrochemical (PEC) sensors and their applications in biochemical and environmental analysis were demonstrated. Finally, the future research aspect of SANs-based electrochemical sensing and the challenges of the SANs design and structure-properties revelation were illustrated, giving guidance on sensitive and accurate biosensing toward clinic diagnostic and environmental analysis.

Abstract:With the rapidly increasing demands for ultrasensitive biodetection, the design and applications of functional nanoprobes have attracted substantial interest for biosensing with optical, electrochemical, and various other means. In particular, given the comparable sizes of nanomaterials and biomolecules, there exists plenty of opportunities to develop functional nanoprobes with biomolecules for highly sensitive and selective biosensing. Over the past decade, numerous nanoprobes have been developed for ultrasensitive bioaffinity sensing of proteins and nucleic acids in both laboratory and clinical applications. In this review, we provide an update on the recent advances in this direction, particularly in the past two years, which reflects new progress since the publication of our last review on the same topic in Chem. Soc. Rev. The types of probes under discussion include: (i) nanoamplifier probes: one nanomaterial loaded with multiple biomolecules; (ii) quantum dots probes: fluorescent nanomaterials with high brightness; (iii) superquenching nanoprobes: fluorescent background suppression; (iv) nanoscale Raman probes: nanoscale surface-enhanced Raman resonance scattering; (v) nanoFETs: nanomaterial-based electrical detection; and (vi) nanoscale enhancers: nanomaterial-induced metal deposition.

Abstract:Nanozymes have been widely used to construct colorimetric sensors due to their advantages of cost-effectiveness, high stability, good biocompatibility, and ease of modification. The emergence of nanozymes greatly enhanced the detection sensitivity and stability of the colorimetric sensing platform. Recent significant research has focused on designing various sensors based on nanozymes with peroxidase-like activity for colorimetric analysis. However, with the deepening of research, nanozymes with peroxidase-like activity has also exposed some problems, such as weak affinity and low catalytic activity. In view of the above issues, existing investigations have shown that the catalytic properties of nanozymes can be improved by adding surface modification and changing the structure of nanomaterials. In this review, we summarize the recent trends and advances of colorimetric sensors based on several typical nanozymes with peroxidase-like activities, including noble metals, metal oxides, metal sulfides/metal selenides, and carbon and metal-organic frameworks (MOF). Finally, the current challenges and prospects of colorimetric sensors based on nanozymes with peroxidase-like activity are summarized and discussed to provide a reference for researchers in related fields.

Abstract:Over decades, as alternatives to natural enzymes, highly-stable and low-cost artificial enzymes have been widely explored for various applications. In the field of artificial enzymes, functional nanomaterials with enzyme-like characteristics, termed as nanozymes, are currently attracting immense attention. Significant progress has been made in nanozyme research due to the exquisite control and impressive development of nanomaterials. Since nanozymes are endowed with unique properties from nanomaterials, an interesting investigation is multifunctionality, which opens up new potential applications for biomedical sensing and sustainable chemistry due to the combination of two or more distinct functions of high-performance nanozymes. To highlight the progress, in this review, we discuss two representative types of multifunctional nanozymes, including iron oxide nanomaterials with magnetic properties and metal nanomaterials with surface plasmon resonance. The applications are also covered to show the great promise of such multifunctional nanozymes. Future challenges and prospects are discussed at the end of this review.

The unique features of nanozymes (such as high stability, low cost, large surface area for bioconjugation, ease of storage, and multi-functionalities) offer unprecedented opportunities for designing electrochemical biosensors. Recent years have witnessed the rapid development of nanozyme-based electrochemical biosensors. Abstract:Nanozyme, a term defined for nanomaterial with enzyme-like properties, has attracted significant research attention owing to its striking merits. Recently, a surge of nanozymes have been demonstrated to catalyze some typical enzymatic reactions mimicking oxidase, peroxidase and catalase. Especially, nanozymes with peroxidase-like activity have grown into a big family due to their broad range of applications in the field of biosensing and immunoassay. Since inorganic nanoparticles possess the advantages of high stability and easy surface modification, nanozymes have been emerging alternatives to natural enzymes to some extent. In this Review, we briefly summarize several typical nanozymes and then focus our attention on their enormous applications with respect to analytical chemistry. Representative examples would be discussed in detail from the literatures of last 10 years.
