Abstract:It has been a challenge to achieve rapid, simple, and effective discrimination of organophosphorus nerve agents (typical chemical warfare agents) due to the similar chemical properties of the targets such as sarin, soman, cyclosarin and VX. In this study, we propose a chemiluminescence sensor array that can effectively discriminate organophosphorus nerve agents by organophosphorus-H2O2 reaction, which produces peroxyphosphonate intermediate and regulates the chemiluminescence intensity. A simple chemiluminescence sensor array based on different chemiluminescence characteristics of the four organophosphorus nerve agents in the luminol-H2O2 system and layered double hydroxide-luminol-H2O2 system has been constructed. Four agents can be well distinguished at a concentration of 1.0 mg L-1 when linear discriminant analyses and hierarchical cluster analyses are smartly combined. The high accuracy (100%) evaluation of 20 blind samples demonstrates the practicability of this proposed chemiluminescence sensor array.

Abstract:Sensitive, selective and fast detection of chemical warfare agents is necessary for anti-terrorism purposes. In our search for functional materials sensitive to dimethyl methylphosphonate (DMMP), a simulant of sarin and other toxic organophosphorus compounds, we found that zinc oxide (ZnO) modification potentially enhances the absorption of DMMP on a manganese dioxide (MnO2) surface. The adsorption behavior of DMMP was evaluated through the detection of tiny organophosphonate compounds with quartz crystal microbalance (QCM) sensors coated with ZnO-modified MnO2 nanofibers and pure MnO2 nanofibers. Experimental results indicated that the QCM sensor coated with ZnO-modified nanostructured MnO2 film exhibited much higher sensitivity and better selectivity in comparison with the one coated with pure MnO2 nanofiber film. Therefore, the DMMP sensor developed with this composite nanostructured material should possess excellent selectivity and reasonable sensitivity towards the tiny gaseous DMMP species.

Abstract:We report a biosensor based on organophosphorus hydrolase (OPH) multilayer modified microcantilever (MCL) for detection of organophosphorus compounds (OPs). The assay is based on substrate-dependent bending of the OPH functionalized MCLs. The cantilever bending amplitude at equilibrium was a function of the concentration of paraoxon with the dynamic range extending from 10−7 to 10−3M. The lower detection limit of approximately 10−7M for paraoxon was an order of magnitude better than the OPH-based potentiometric and optical biosensors based on pH modulation. There was a good intra-sensor and an acceptable inter-sensor reproducibility as evidenced by the standard errors of 5% and 15%, respectively. OPs measured using this technique included parathion and diisopropyl fluorophosphate (DFP) in the order of paraoxon>DFP>parathion. The conformational change of the OPH was most likely the main origin of MCL bending.

Abstract:Chemical warfare agents (CWAs) are extremely toxic chemicals, necessitating rapid, easy-to-use, sensitive and selective sensors for detection. Dimethyl methylphosphonate (DMMP) serves as a simulant of nerve agents and is commonly used to evaluate the efficacy of sensors in detecting these hazardous compounds. In this context, this paper proposes a one-dimensional (1D) photonic crystal (PC) gas sensor functionalised with UIO-66-COOH, leveraging a hydrogen-bonding-based acidic strategy, for rapid and sensitive DMMP detection. To optimise DMMP uptake, the organic ligand, UIO-66, is functionalised by grafting additional carboxyl groups to facilitate hydrogen-bonding interactions with the PO groups of organophosphorus gases such as DMMP. Subsequently, attenuated total reflection surface-enhanced infrared absorption spectroscopy is performed to quantitatively monitor the impact of strong interactions between the PC bonds and PO bonds of DMMP and the adsorption sites of metal–organic frameworks (MOFs) during adsorption. The results reveal an increase in the number of hydrogen-bonding sites available for DMMP adsorption, thus promoting hydrogen-bonding interactions. Furthermore, density functional theory calculations are employed to analyse the presence and strength of hydrogen bonds between the MOFs and DMMP. The results reveal that the chemisorption energy of UIO-66-COOH exceeds those of its variants, corroborating the findings of sensing experiments. Overall, the developed 1D PC gas sensor comprising UIO-66-COOH/TiO 2 demonstrates excellent stability and reproducibility, with a limit of detection of 0.3 ppm. This sensor also demonstrates enhanced selectivity towards organophosphorus gases compared to volatile organic compounds.

Abstract:Nerve agents, one of the most toxic chemical warfare agents, seriously threaten human life and public security. The high toxicity of nerve agents makes the development of fluorescence sensors with suitable limit of detection challenging. Here, we propose a sensor design based on a conjugated microporous polymer film for the detection of diethyl chlorophosphate, a substitute of Sarin, with low detection limit of 2.5 ppt. This is due to the synergy of the susceptible on-off effect of hybridization and de-hybridization of hybrid local and charge transfer (HLCT) materials and the microporous structure of CMP films facilitating the inward diffusion of DCP vapors, and the extended π-conjugated structure. This strategy provides a new idea for the future development of gas sensors. In addition, a portable sensor is successfully integrated based on TCzP-CMP films that enables wireless, remote, ultrasensitive, and real-time detection of DCP vapors.

Abstract:A facile and highly sensitive signaling strategy for organophosphorus compounds (OPs) based on acetylcholinesterase (AChE)-acetylthiocholine (ATCh) catalytic reaction and II-VI group photoluminescent CdTe quantum dots (QDs) prepared from low molar ratio of thiols to Cd2+ has been developed. Photoluminescence (PL) of the ensemble system can be modulated by the enzyme catalytic reaction and OPs, as the resulting catalytic product, namely thiolcholine (TCh) and H+, can significantly decrease PL of the prepared QDs, while OPs can inhibit this AChE-ATCh catalytic reaction. This signaling platform shows much more enhanced sensitivity to highly toxic OPs such as nerve agents and organophosphate pesticides, compared with previous reports.

Abstract:The ability to analyze highly toxic chemical warfare agents (CWAs) and related chemicals in a rapid and precise manner is essential in order to alleviate serious threats to humankind and public security caused by unexpected terrorist attacks and industrial accidents. In this investigation, we designed a o-phenylenediamine-pyronin linked dye that is capable of both fluorogenic and colorimetric discrimination between phosgene and the prototypical nerve-agent mimic, diethyl chlorophosphate (DCP) in the solution or gas phase. Moreover, this dye has been used to construct a portable kit that can be employed for real-time monitoring of DCP and phosgene in the field, both in a discriminatory manner, and in a simple and safe way.

Abstract:Organophosphorus compounds are widely distributed and highly toxic to the environment and living organisms. The current detection of organophosphorus compounds is based on a single-mode method, which makes it challenging to achieve good portability, accuracy, and sensitivity simultaneously. This study designed a multifunctional microfluidic chip to develop a dual-mode biosensor employing a DNA hydrogel as a carrier and aptamers as recognition probes for the colorimetric/electrochemical detection of malathion, an organophosphorus compound. The biosensor balanced portability and stability by combining a microfluidic chip and target-triggered DNA hydrogel-sensing technologies. Moreover, the biosensor based on target-triggered DNA hydrogel modified microfluidic developed in this study exhibited a dual-mode response to malathion, providing both colorimetric and electrochemical signals. The colorimetric mode enables rapid visualization and qualitative detection and, when combined with a smartphone, allows on-site quantitative analysis with a detection limit of 56 nM. The electrochemical mode offers a broad linear range (0.01-3000 μM) and high sensitivity (a limit of detection of 5 nM). The two modes could validate each other and improve the accuracy of detection. The colorimetric/electrochemical dual-mode biosensor based on target-triggered DNA hydrogel modified microfluidic chip offers a portable, simple, accurate, and sensitive strategy for detecting harmful environmental and food substances.

Abstract:Organophosphorus (OP)-based nerve agents are extremely toxic and potent acetylcholinesterase inhibitors and recent attacks involving nerve agents highlight the need for fast detection and intervention. Fluorescence-based detection, where the sensing material undergoes a chemical reaction with the agent causing a measurable change in the luminescence, is one method for sensing and identifying nerve agents. Most studies use the simulants diethylchlorophosphate and di-iso-propylfluorophosphate to evaluate the performance of sensors due to their reduced toxicity relative to OP nerve agents. While detection of nerve agent simulants in solution is relatively widely reported, there are fewer reports on vapor detection using solid-state sensors. Herein, progress in organic semiconductor sensing materials developed for solid-state detection of OP-based nerve agent vapors is reviewed. The effect of acid impurities arising from the hydrolysis of simulants and nerve agents on the efficacy and selectivity of the reported sensing materials is also discussed. Indeed, in some cases it is unclear whether it is the simulant that is detected or the acid hydrolysis products. Finally, it is highlighted that while analyte diffusion into the sensing film is critical in the design of fast, responsive sensing systems, it is an area that is currently not well studied.

Abstract:Developing reliable sensors that accurately detect deadly chemical gases is critical to global security. Nerve agents are one of the most dangerous chemicals in the world and are often found in gaseous forms in the environment, which remain a challenge to detect because of their low levels. In this paper, a fluorescent probe based on a Zr-based metal-organic framework UiO-66-NH2 was proposed. The specific binding between the Zr-O site of UiO-66-NH2 and diethyl chlorophosphate (DCP) blocked the ligand-to-metal charge transfer (LMCT) process in UiO-66-NH2, thereby enabling the fluorescence turn-on detection of DCP. More importantly, a simple and portable hydrogel soft-solid platform (UiO-66-NH2@Aga) was constructed by incorporating UiO-66-NH2 into the formation process of agarose (Aga) hydrogel for fast and sensitive detection of gaseous DCP. When the hydrogel was exposed to a low concentration of DCP vapor, its fluorescence changed from colorless to bright blue, allowing visualization of the DCP gas for analysis. The UiO-66-NH2@Aga integrated solid-state platform showed an excellent response to DCP vapor in the detection range of 1.98 to 9.90 ppm and with a detection limit of 1.16 ppm. This work opened up a unique way to design a convenient, low cost and practical gas physical examination platform.

Abstract:Among all kinds of chemical warfare agents, only cyanide and nerve agents can cause massive mortality at low concentrations. In this work, a dual-channel fluorescent probe CWAs-Thia capable of detecting cyanide and nerve agents is presented. The two reactive recognition units, pyridine and the thiazole-2-carbonyl group, of the probe for cyanide and nerve agents, respectively, produced red and blue fluorescent responses, respectively, which were attributed to excited-state intramolecular proton transfer and intramolecular charge transfer. CWAs-Thia is the first probe that can selectively recognize cyanide and nerve agent. And it has proven to be effective in visualizing cyanide and nerve agents in living cells.

Abstract:Developing a fluorescence sensor capable of precisely identifying highly toxic nerve agents has become a focus of research, given that they are easily available and can be used by terrorists to threaten the security of the world. In this work, we designed a ratiometric fluorescence sensing platform with Cu 2+ -modified UiO-66-NH 2 (Cu 2+ @UiO-66-NH 2 ) and o-phenenediamine (OPD) as signal indicators and successfully used for the liquid and gas detection of the nerve agent simulant diethyl chlorophosphate (DCP). UiO-66-NH 2 obtained oxidase activity by simple Cu 2+ modification, which catalyzed the oxidation of OPD to give a yellow fluorescent product 2, 3-diaminophenazine (DAP). In this detection system, the fluorescence of UiO-66-NH 2 was greatly enhanced by blocking the ligand metal charge transfer (LMCT) process inside the framework by DCP. At the same time, the amino group of DAP was phosphorylated by DCP, and the attenuation of DAP fluorescence was observed by the static quenching effect. In addition, the Cu 2+ @UiO-66-NH 2 /OPD loaded agarose hydrogel device can be used to detect DCP vapor with naked eyes and showed satisfactory detection result (LOD: 6.65 ppb).

Abstract:In this paper, two donor (D)-acceptor (A) type of small organic fluorescent molecules (T1 and T2) based on terpyridine group are synthesized, characterized and used as colorimetric/fluorometric dual-channel probes towards diethylchlorophosphate (DCP, the mimic of chemical warfare agent sarin) not only in solution but also in gas phase featuring instantaneous responses, excellent recyclability, high selectivity and sensitivity. Interestingly though the discriminated units of both chemosensors are terpyridine, their fluorescent responded signals are different, which is due to the different electron-donating substituents of T1 and T2 caused the different responded mechanism to DCP. And the possible sensing mechanism was proved by using nuclear magnetic resonance (1H NMR, 31P NMR) spectra and natural transition orbitals calculations. Furthermore, facile testing filter paper-constructed strips with the visualization of colorimetric/fluorometric dual-channel responses based on T1 and T2 have been fabricated for real-time, on-site high selective and sensitive, recyclable monitor of DCP vapor.

Abstract:Enzyme-inhibited electrochemical sensor is a promising strategy for detecting organophosphorus pesticides (OPs). However, the poor stability of enzymes and the high oxidation potential of thiocholine signal probe limit their potential applications. To address this issue, an indirect strategy was proposed for highly sensitive and reliable detection of chlorpyrifos by integrating homogeneous reaction and heterogeneous catalysis. In the homogeneous reaction, Hg2+ with low oxidation potential was employed as signal probe for chlorpyrifos detection since its electroactivity can be inhibited by thiocholine, which was the hydrolysate of acetylthiocholine catalyzed by acetylcholinesterase. Additionally, Co,N-doped hollow porous carbon nanocage@carbon nanotubes (Co,N-HPNC@CNT) derived from ZIF-8@ZIF-67 was utilized as high-performance electrode material to amplify the stripping voltammetry signal of Hg2+. Thanks to their synergistic effect, the sensor exhibited outstanding sensing performance, excellent stability and good anti-interference ability. This strategy paves the way for the development of high-performance OP sensors and their application in food safety.

Abstract:Organophosphorus compounds are toxic organic phosphorus molecules including nerve agents and pesticides. Organophosphorus pesticides play a significant role in improving large-scale modern agriculture. Nerve agents are neurotoxic chemicals. Therefore, there is a demand for assays to measure organophosphorus analogs in a rapid, sensitive, and convenient way. Here, colorimetric and fluorescence sensors can be an effective approach for sensing these substances. They have been extensively explored due to their portability and high sensitivity. This review summarizes recent development in colorimetric and fluorescent materials including small organic fluorophores, nanomaterials, complexes, and polymers since 2015. We describe advances in sensing a variety of organophosphorus analogs. We also discuss recent challenges and design strategies in developing colorimetric and fluorescent assays with high performance and specific sensing.

Abstract:Organophosphorus pesticides (OPs) are widely used in pest control. The residual OPs can cause pollution to the environment and harm to the human body. Although OPs can inhibit the activity of acetylcholinesterase (AChE) and lead to neurological diseases, it also provides enlightenment for the development of a simple and sensitive method for the detection of OPs. Herein, we report a colorimetric sensor array using nanozymes (Fe-N-C nanozymes and Cu-N-C nanozymes) combined with natural enzymes to discriminate eight OPs (Iprobenfos (Ipr), Diazinon (Dia), Crotoxyphos (Cro), Bromophos ethyl (Bro), Dipterex (Dip), Chlorpyrifos methyl (Chl), Ethion (Eth), and Omethoate (Ome)) through a cascade reaction system. Surprisingly, eight OPs were successfully identified in tap water at 1 ng mL−1.

Ultrabright chemiluminophores are highly desired in CL-based analytical approaches, imaging, for example, although the design of CL scaffold with ultra-brightness remains a challenge. We report a feasible strategy for constructing an ultrabright chemiluminophore, BTCL-Cl, by grafting benzothiazole moiety to the dioxetane center of Schaap's adamantylidene-dioxetane. Compared with the classical undecorated chemiluminophores, the CL quantum yield of BTCL-Cl has improved by 2.4-folds to 17.6% with a short half-life due to deprotonation under alkaline conditions. Probe CL-HMPC, the thiol-recognizer-decorated BTCL-Cl, has demonstrated ultrahigh sensitivity (detection limit is 6.0 ng/mL) in monitoring organophosphorus compounds (OPs) by immediate releasing of thiols after the alkaline hydrolysis of OPs. The OPs-triggered remarkable CL enhancement has ensured quantitively visualization of OPs in the naked eye by imaging with a smartphone in situ . Using the versatile and ultrasensitive CL platform, we succeeded in feasibly visualizing the concentration of OPs on cabbage leaves on farmland by portable setups. The simple and promising approach for analyzing pesticides and potentially other targets brings tremendous imagination in related areas.
