Abstract:This work presents a cost-effective, label-free in point-of-care (POC) biosensor for the sensitive detection of 8-hydroxy-2′-deoxyguanosine (8-OHdG), the most abundant oxidative product of DNA, that may allow a premature assessment of cancer disease, thereby improving diagnosis, prognostics and survival rates. The device targets the direct detection of 8-OHdG by using for the first time a carbon-ink 3-electrode on a paper substrate coupled to Differential Pulse Voltammetry readings. This design was optimized by adding nanostructured carbon materials to the ink and the conducting polymer PEDOT, enhancing the electrocatalytic properties of the sensor towards 8-OHdG detection. Meanwhile, the ability of this oxidative stress biomarker to undertake an oxidation reaction enabled the development of the sensing electrochemical device without the need of chemical probes and long incubation periods. This paper-modified sensor presented high electrochemical performance on the oxidation of 8-OHdG with a wide linear range (50–1000 ng/ml) and a low detection limit (14.4 ng/ml). Thus, our results showed the development of a direct and facile sensor with good reproducibility, stability, sensitivity and more importantly, selectivity. The proposed carbon-based electrochemical sensor is a potential candidate to be miniaturized to small portable size, which make it applicable for in-situ 8-OHdG sensing in real biological samples.

Abstract:8-Hydroxydeoxyguanosine (8-OHdG) is the most widely used oxidative stress biomarker of the free radical-induced oxidative damage product of DNA, which may allow a premature assessment of various diseases. This paper designs a label-free, portable biosensor device to directly detect 8-OHdG by plasma-coupled electrochemistry on a transparent and conductive indium tin oxide (ITO) electrode. We reported a flexible printed ITO electrode made from particle-free silver and carbon inks. After inkjet printing, the working electrode was sequentially assembled by gold nanotriangles (AuNTAs) and platinum nanoparticles (PtNPs). This nanomaterial-modified portable biosensor showed excellent electrochemical performance for 8-OHdG detection from 10 μg/mL to 100 μg/mL by our self-developed constant voltage source integrated circuit system. This work demonstrated a portable biosensor for simultaneously integrating nanostructure, electroconductivity, and biocompatibility to construct advanced biosensors for oxidative damage biomarkers. The proposed nanomaterial-modified ITO-based electrochemical portable device was a potential biosensor to approach 8-OHdG point-of-care testing (POCT) in various biological fluid samples, such as saliva and urine samples.

Abstract:8-Hydroxy-2 '-deoxyguanosine (8-OHdG) is extensively recognized as an oxidative stress biomarker for oxidative DNA damage in the early diagnosis and prevention of various diseases. As a response to the increasing demand for high sensitivity, repeatability, and stability of in-situ sensing platforms, this work designed a flexible, portable and scale-manufacturable microelectrochemical sensor for the sensitive and selective determination of 8-OHdG. The microsensor was fabricated based on polyvinylpyrrolidone (PVP) functionalized laser-induced graphene (LIG), which was prepared by laser direct engraving of commercial polyimide film. Owing to the synergistic effect between the remarkable capabilities for H-bond complexation with the analytes of PVP and the excellent physical and electrochemical properties of LIG, the microsensor demonstrated remarkable electrochemical response to oxidation of 8-OHdG and effectively reduced the interference of uric acid, achieved through the strategic integration of uricase. Under the optimal conditions, the oxidation peak current exhibits a good linear relationship with the concentration of 8-OHdG in the range of 0.005-2.56 mu mol/L (R2 = 0.990) and 2.56 to 20.48 mu mol/L (R2 = 0.972), respectively. The detection limit was calculated to be 0.63 nmol/L. Furthermore, the proposed sensor shows superior stability and repeatability, and was successfully applied to determine the concentrations of 8-OHdG in biological fluid samples, accompanied by good recovery rates. In addition, the field determination capability of the microsensor was evaluated using a microelectrochemical workstation, highlighting its excellent in-situ determination performance. In conclusion, the microelectrochemical sensor is expected to be an alternative technology for early risk assessment of various diseases and has promising application prospects in point-of-care testing.

Abstract:Urolithiasis is a highly prevalent disease with increasing incidence worldwide, which occurs mainly as calcium oxalate. Therefore, measuring urinary oxalate for the early screening of urolithiasis is of growing significance. However, current approaches for oxalate detection are generally based on specific equipment, thus limiting their point-of-care applications. In this study, a colorimetric lateral flow strip was developed based on enzyme catalysis, and a color bar strategy was utilized for equipment-free and semi-quantitative determination of urinary oxalate, which does not require any image acquisition or processing equipment. Rapid semi-quantitative detection of oxalate is achieved by directly counting the number of color bars that is positively correlated with oxalate concentration. Three colorimetric patterns were compared and optimized, among which the one with 3,3',5,5'-tetramethylbenzidine (TMB) scribes presented the best results with minimized drag impact, washing effects and limited inter-reagent reactions in the lateral flow. Under optimized conditions, the assay showed a detection limit less than 0.3 mM and a detection range of 0.3-0.7 mM that covers the urinary oxalate concentrations for urolithiasis screening. Finally, the strips were verified for real urine sample analysis, vali-dating its potential for oxalate detection and urolithiasis screening in point-of-care applications.

Abstract:Ascorbic acid, dopamine, and uric acid are important electroactive biomolecules for health monitoring and they coexist in serum or urine. Their quantitative determination by electrochemistry could provide the accurate reference for diseases diagnosis and treatment. However, the traditional electrochemical workstations are too large for on-field inspection. Hence, the design of handheld electrochemical system for the detection of biomolecules is significant for point-of-care testing (POCT). In this paper, a smartphone-based integrated voltammetry system using modified electrode was developed for simultaneous detection of biomolecules. The system contained a disposable sensor, a coin-size detector, and a smartphone equipped with application program. Screen-printed electrodes were used as sensors for the detection, on which reduced graphene oxide and gold nanoparticles were electrochemically deposited by the system. The detector was used with voltammetric methods, in which excitation voltage was applied on the sensors and subsequent current responses were detected. The smartphone is the core component to communicate with the detector, calculate data, and plot voltammograms in real-time. Then, the system was applied to detect standard solutions of the biomolecules and their mixtures as examples. The results showed that the peak currents of each substance increased with higher concentration and the method allowed the discrimination of the different potentials of the studied species. Finally, the practical applications of the system were tested through detections of the biomolecules in artificial urine. The results exhibited that the system could be used to detect electrochemical activity of biomolecules with linear, high sensitivity, and specific responses in point-of-care testing.
Fast Quantification of Salivary 8-Hydroxy-2′-deoxyguanosine as DNA Damage Biomarker Using CE with Electrochemical Detection

Abstract:The aim of this study was to investigate the correlation between levels of a marker of oxidative DNA damage, 8-hydroxy-2′-deoxyguanosine (8-OHdG), in human saliva and urine, and to explore its potential application in fast diagnosis of many diseases (especially cancer) associated with oxidative damage. A simple and time-efficient method, based on capillary electrophoresis with electrochemical detection, was developed for the determination of salivary and urinary 8-OHdG. Under the optimum conditions, 8-OHdG and its coexisting analytes could be well separated within 16 min at a voltage of 14 kV in 60 mmol L−1 borax running buffer (pH 8.2). A good linear relationship was established between peak current and concentration of analytes over three orders of magnitude with detection limits (S/N = 3) ranging from 0.41 × 10−7 to 2.50 × 10−7 mol L−1 for all analytes.

Abstract:In this work, an electrochemical method for sensitive analysis of 8-hydroxy-2'-deoxyguanosine, a key biomarker that is widely used to study oxidative injury-related diseases, is proposed based on exonuclease-mediated functional nucleic acid. In the design, exonuclease can not only distinguish the existence of target, but also suppress the background noise, thus the sensitivity can be enhanced. Moreover, DNAzyme designed in the functional nucleic acid can further improve the sensitivity of the analysis during signal generation process. Therefore, exonuclease-mediated functional nucleic acid may ensure high sensitivity of the assay. Further studies reveal that the detection of 8-hydroxy-2'-deoxyguanosine can be achieved with a linearity from 0.01 nM to 7.0 M and a detection limit of 6.82 pM. The new method has also been successfully applied to the determination of 8-OHdG in urine with good results, indicating its great potential for practical use.

Abstract:Oxidative stress plays an important role in the pathogenesis of many diseases, while the exact mechanism that hydrogen peroxide (H2O2) as one of the most abundant reactive oxygen species (ROS) exerts its influence on oxidative stress remains unclear. We developed a novel turn-on ratiometric electrochemical sensor for the detection of H2O2 in blood samples. The electrochemical probe 5-(1,2-dithiolan-3-yl)-N-(4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)phenyl)pent-anamide (BA) was designed and synthesized for the selective detection of H2O2 via a one-step amide reaction. Meanwhile, Nile Blue A (NB) was optimized as an internal reference molecule, thus enabling accurate quantification of H2O2 in a complex environment. BA and NB were then co-assembled onto a carbon fiber microelectrode (CFME) coated with Au cones. The oxidation peak current ratio between BA and NB demonstrated good linearity with the logarithm of the H2O2 concentration values ranging from 0.5 μM to 400 μM with a low detection limit of 0.02 μM. The developed sensor showed remarkable selectivity against potential interferences in whole blood samples, especially for ascorbic acid, uric acid, and dopamine. In combination with the unique characteristics of CFME, such as a small size and good biocompatibility, the microsensor was used for rapid analysis of one drop of whole blood sample. This sensor not only creates a new platform for the detection of H2O2 in whole blood samples, but also provides a new design strategy of other ROS analysis for early diagnosis of ROS-related diseases, drug discovery processes, and pathological mechanisms.

Abstract:A sensitive fluorescence resonance energy transfer (FRET) biosensor is proposed to detect 8-hydroxy-2′-deoxyguanosine (8-OHdG), which is a typical DNA oxidation damage product excreted in human urine. The FRET biosensor was based on carbon dots (CDs)-modified nanoporous alumina membrane with CDs as fluorescence donors. Gold nanoparticles were encapsulated in zeolitic imidazolate framework-8 to form Au@ZIF-8 nanoparticles as signal quenchers. CDs and Au@ZIF-8 nanoparticles were biofunctionalized by 8-OHdG antibody. The capture of 8-OHdG on the membrane substrates can bring Au@ZIF-8 nanoparticles closely to CDs. With 350 nm excitation, the fluorescence of CDs was quenched by Au@ZIF-8 nanoparticles and FRET effect occurred. The quenching efficiency was analyzed. The limit of detection (LOD) was 0.31 nM. Interference experiments of the FRET biosensor showed good specificity for 8-OHdG detection. The biosensor could detect urinary 8-OHdG sensitively and selectively with simple sample pretreatment processes. It shows applicability for detecting biomarkers of DNA damage in urine or other biological fluids.

Abstract:The potential reach of point-of-care (POC) diagnostics into daily routines for exposure to reactive oxygen species (ROS) and Cu in aerosolized particulate matter (PM) demands that microfluidic paper-based analytical devices (μPADs) take into consideration the simple detection of these toxic PM components. Here, we propose μPADs with a dual-detection system for simultaneous ROS and Cu(II) detection. For colorimetric ROS detection, the glutathione (GSH) assay with a folding design to delay the reaction yielded complete ROS and GSH oxidation, and improved homogeneity of color development relative to using the lateral flow pattern. For electrochemical Cu(II) determination, 1,10-phenanthroline/Nafion modified graphene screen-printed electrodes showed ability to detect Cu(II) down to pg level being low enough to be applied to PM analysis. No intra- and inter-interference affecting both systems were found. The proposed μPADs obtained LODs for 1,4-naphthoquinone (1,4-NQ), used as the ROS representative, and Cu(II) of 8.3 ng and 3.6 pg, respectively and linear working ranges of 20 to 500 ng for ROS and 1 × 10-2 to 2 × 102 ng for Cu(II). Recovery of the method was between 81.4 and 108.3% for ROS and 80.5-105.3% for Cu(II). Finally, the sensors were utilized for simultaneous ROS and Cu(II) determination in PM samples and the results statistically agreed with those using the conventional methods at 95% confidence.

Abstract:Formamidopyrimidine DNA glycosylase (FPG) is a crucial DNA repair enzyme that specifically recognizes and excises the damaged base 7,8-dihydro-8-oxoguanine (8-oxoG). The current detection technology for FPG is limited due to the need of integrating the relatively independent identification components and signal amplifiers. Herein, we designed an integrated probe (loaded on magnetic beads), which contained 8-oxoG for FPG recognition and a novel chimeric peptide-DNA mimetic enzyme (CPDzyme) for chemiluminescence (CL) signal amplification. Once the FPG recognized the probe, the CPDzyme was excised from the surface of the magnetic beads. Therefore, the change in CL signal caused by CPDzyme on the surface of the magnetic spheres before and after recognition and cleaning could be quantitatively analyzed for FPG. Thanks to the powerful catalytic ability of CPDzyme and the simplicity of the CL system, this method could detect the activity of FPG in a linear range of 0.2-20 U/mL, with the detection limit as low as 0.06 U/mL. Further, we applied the strategy to the detection of FPG activity in human serum and bacterial samples (before and after UV irradiation), demonstrating its potential for the monitoring of oxidative damage. With excellent sensitivity and standardized operation, this strategy demonstrates superior characteristics to commercial assay kits and is expected to provide a new powerful tool for relevant research.
Construction of a damage site-specific fluorescent biosensor for single-molecule detection of DNA damage

Abstract:The 8-oxoguanine (8-oxoG) represents the most common DNA damage type, and it has been regarded as the oxidative stress biomarker, but the reported 8-oxoguanine assays are limited by poor specificity and low sensitivity. Herein, we demonstrate the construction of damage site-specific fluorescent biosensor for 8-oxoG assay by integrating single-molecule detection with hyperbranched signal amplification. In this assay, the 8-oxoG damages in DNA can generate free 3′ OH with the assistance of formamidopyrimidine DNA glycosylase (Fpg) and polynucleotide kinase (PNK), which subsequently triggers the incorporation of abundant Cy5-labeled dUTPs via terminal deoxynucleotidyl transferase (TDT)-mediated site-specific hyperbranched nucleic acid amplification. After digestion of amplification products with nuclease treatment, abundant mononucleotide Cy5-dUTPs are produced, which will be easily monitored via single-molecule imaging and detection. The introduction of hyperbranched nucleic acid amplification and single-molecule detection can greatly improve the sensitivity and achieve a detection limit of 7.62 ×10−18 M. This biosensor is highly specific with the capability of discriminating 0.001% 8-oxoG target from the DNA mixture. Moreover, it can be applied for quantitative detection of 8-oxoG damage in genomic DNAs with a detection limit of 0.0017 ng, and even accurately quantifies the absolute number (7025–8506) of 8-oxoG damage base in single HeLa cell treated with 150 μM H2O2. Importantly, this biosensor can measure the 8-oxoG damage level in different cancer cell lines, facilitating the oxidative damage-associated biomedical researches and clinical diagnosis.

Using a Glucose Meter to Quantitatively Detect Disease Biomarkers Through a Universal Nanozyme Integrated Lateral Fluidic Sensing Platform.

Abstract:Along with the advance in medical research, more biomarkers emerge as useful indicators for both disease and health index. However, majority of them have no practical or economic testing methods available yet, or rely on high-costing methods such as Enzyme-Linked Immuno-Sorbent Assay (ELISA), High-Performance Liquid Chromatography (HPLC), Mass Spectrum, and immunohistochemistry (IHC). In this article, we develop a universal nanozyme integrated testing platform for biological molecules that incorporates the electrochemical measurement of glucose with lateral flow immunostrip (LFS) for target analytes. This design involves the quantitative conversion of analytes into invertase and then glucose, which can be measured by an extremely affordable meter. The feasibility of this design was validated using 8-hydroxy-2'-deoxyguanosine (8-OHdG) and prostate specific antigen (PSA) as representatives for small molecules and moderate to large proteins respectively. Our approach yields results comparable to commercial diagnostic ELISA kits at a substantially reduced cost and reaction time. Specifically, the design has a detection limit of 0.23 ng mL(-1) for 8-OHdG and 1.26 ng mL(-1) for PSA, and a detection range of 0.1-100 ng mL(-1) for 8.0HdG and 1-100 ng mL(-1) for PSA. By combining the accessibility of well-established glucose testing and LFS, our design can serve as a point of care testing method that can be fully integrated into the personal lifestyle without requiring professional assistance.
A stable nanosilver decorated phosphorene nanozyme with phosphorus-doped porous carbon microsphere for intelligent sensing of 8-hydroxy-2′-deoxyguanosine

Abstract:Nanozyme based on the silver nanoparticles (AgNPs) decorated highly water-oxygen stable phosphorene (BP) nanohybrid with phosphorus-doped porous carbon microspheres (P-PCMs) is applied as an intelligent sensing platform for the electrochemical detection of 8-hydroxy-2′-deoxyguanosine (8-OHdG) as a biomarker of oxidative DNA damage in human urine sample. The density functional theory (DFT) is use for investigating the effect of silver on the conductivity level of BP and discussing a possible mechanism for the electrocatalytic oxidation of 8-OHdG. The Ag+ is in-situ reduced as AgNPs that grows onto the BP surface for the controllable preparation of BP-AgNPs with effective surface passivation among different metal ions decorated BP. P-PCMs with 4.9-fold enhancement in specific surface area (1636.73 m2 g−1) are prepared by hydrothermal carbonization of α-cyclodextrin as carbon sources, then calcinate in the presence of phosphoric acid as an activator and dopant. P-PCMs-BP-AgNPs are prepared by the ultrasound-assisted liquid-phase exfoliation with the addition of Ag+, and both P-PCMs and black phosphorus crystals are sufficiently grinded. The P-PCMs-BP-AgNPs nanohybrid displays good long-term water-oxygen stability, extraordinary specific surface area, superior electrocatalytic capacity with 303-fold enhancement, enzyme-like characteristics with <math>Imax</math> of 100 μA and <math>Km</math> of 29 μM. The machine learning (ML) model with artificial neural network (ANN) algorithm is employed for the intelligent output of 8-OHdG in real sample with acceptable recovery in work range from 0.2 to 125 μM.

Abstract:8-Hydroxydeoxyguanosine (8-OHdG) is one of the main products of DNA oxidative stress injury and has been widely used for quantitative analyzing ROS levels. However, the intracellular detection of 8-OHdG is rarely reported and could provide for a novel tool for oxidative damage research. Here, a highly fluorescent silver nanocluster was synthesized by conjugating 12 cytosine bases as Ag nanocluster templates with 8-OHdG aptamer as the specific binding target (named 8-OHdG-12C), resulting in a 10-fold increase in fluorescence intensity of the as synthesized probe. The 8-OHdG-12C@AgNCs showed a good fluorescence property and were stable for weeks. In addition, it can visualize intracellular 8-OHdG in a spatiotemporal manner. We used oxidative or reductive inducers of ROS and showed that the probe was located within cells and proportional to the cell oxidative damage induced. Together, these results indicate that 8-OHdG-12C@AgNCs hold great potential for in vivo 8-OHdG detection and may be a useful method to determine how DNA oxidative damage influences cancer cells and apoptosis.

Abstract:Poultry red mite susceptibility to permethrin, deltamethrin, tetramethrin, trichlorfon, fenitrothion, carbaryl and DDT, respectively, was screened within the territory of Bohemia by a method of diagnostic doses. The survey indicated widely spread resistance to DDT; in a few cases ineffectivity of permethrin, tetramethrin and trichlorfon, respectively, was also documented.

Abstract:A novel microfluidic paper-based chemiluminescence (CL) analytical device (μPAD) with high-throughput, rapid, stable, reusable point-of-care (POC) testing and quantitative response for DNA was designed. Sodium periodate was used to modify μPAD to covalently immobilize capture DNA. The paper device was fabricated by a low-cost, simple, and rapid wax screen-printing method. The developed paper-based CL immunodevice, combined with a typical luminol–H2O2 CL system, showed excellent analytical performance for the detection of DNA. Moreover, gold nanoparticles for signal DNA immobilization were used for signal-on CL assay of target DNA, which greatly improved the sensitivity. This study shows the successful integration of the μPAD and the CL method to afford an easy-to-use, inexpensive, and portable alternative for POC monitoring.

Abstract:Lateral flow immunostrips were newly designed and a sensitive and rapid fluorometric method for the determination of 8-hydroxy-2′-deoxyguanosine (8-OHdG) as a model target of small biomarker molecules was developed. The upconversion nanoparticles (UCNPs, NaYF4:Yb/Er core, and polyacrylic acid (PAA)-modified shell, size ~ 39 nm, excitation wavelength = 980 nm; emission wavelength = 540 nm) were employed as fluorescence signal material. The 8-OHdG antibody (Ab) was taken as the recognition probe while UCNP-labeled Ab was taken as the signal probe. Bovine serum albumin (BSA) was designed as carrier protein for 8-OHdG to form 8-OHdG-BSA conjugate as the capture probe. The lateral flow immunostrips were prepared by laminating a sample pad (glass fiber membrane), a test pad (nitrocellulose membrane), and adsorption pad (filter paper) on PVP backing. The capture probe was immobilized on the test zone while an IgG antibody taken as the control probe was immobilized on the control zone. When the signal probe and the sample were in sequence loaded on the sample pad, 8-OHdG analyte bound with the signal probe, and then the excess of the signal probe move along the strip and is collected by the capture probe on the test zone while the remnant signal probe is collected by the control probe on the control zone. The signal probe and capture probe were synthesized and characterized. The fluorescence intensity on the test zone was inversely proportional to the concentration of 8-OHdG for the quantitative determination while the fluorescence emission on the control zone was observed to validate the assay. The developed method showed a wide linear range from 0.10 to 10 nM, a quite low detection limit of 0.05 nM, small sample volume requirement (100 μL), short assay time (15 min), and good method reproducibility (RSD = 4.4%, nine immunostrips).
