Abstract:Fat crystals and adsorbed interfacial layer are very important in controlling the stability of partly crystalline fat globules. In this study, the effects of competitive adsorption of emulsifiers, including glyceryl monooleate (GMO) and sucrose ester (SE) with milk proteins and emulsifier-fat (anhydrous milk fat, AMF; coconut oil, CO) interactions on the fat crystallization behavior and stability of whipped-frozen emulsions were investigated. Parameters such as zeta potential, surface protein coverage, rheological property, fat crystallization behavior, overrun, melting rate and microstructure of ice cream were determined. The results indicated both GMO and SE affected the crystallization behavior, changing the crystal growth, morphology and stiffness. Different mechanism was involved in fat partial coalescence in emulsion systems with different fat-emulsifier interactions. The promotion of partial coalescence in whipped-frozen emulsions with increasing concentration of GMO was mainly related to fat crystallization behavior combined with increased fat crystal growth dimension and protrusion length at the fat globule surface, since protein surface coverage was not affected by GMO concentration. However, in the case of emulsions with SE, enhancement of partial coalescence was mainly related to decreased protein surface coverage combined with less resistance of interfacial membranes to penetration by small fat crystals. Ice cream based on CO were distinguished by higher overruns and lower melting rates compared to ice cream based on AMF with the same levels of emulsifiers, especially strong fat crystal network in the presence of GMO accounts for the increased air bubble stability. These findings provides a theoretical basis for aerated food processing.

Effect of Solid Fat Content in Fat Droplets on Creamy Mouthfeel of Acid Milk Gels

Abstract:Previous studies have shown that emulsions with higher solid fat content (SFC) are related to a higher in-mouth coalescence level and fat-related perception. However, the effect of SFC in fat droplets on the fat-related attributes of emulsion-filled gels has not been fully elucidated. In this study, the effect of SFC on the creamy mouthfeel of acid milk gel was investigated. Five kinds of blended milk fats with SFC values ranging from 10.61% to 85.87% were prepared. All crystals in the blended milk fats were needle-like, but the onset melting temperature varied widely. Blended milk fats were then mixed with skim milk to prepare acid milk gels (EG10−EG85, fat content 3.0%). After simulated oral processing, the particle size distribution and confocal images of the gel bolus showed that the degree of droplet coalescence in descending order was EG40 > EG20 > EG60 > EG10 ≥ EG85. There was no significant difference in apparent viscosity measured at a shear rate of 50/s between bolus gels, but the friction coefficients measured at 20 mm/s by a tribological method were negatively correlated with the coalescence result. Furthermore, quantitative descriptive analysis and temporal dominance of sensations analysis showed that SFC significantly affected the ratings of melting, mouth coating, smoothness and overall creaminess, as well as the perceived sequence and the duration of melting, smoothness and mouth coating of acid milk gels. Overall, our study highlights the role of intermediate SFC in fat droplets on the creamy mouthfeel of acid milk gels, which may contribute to the development of low-fat foods with desirable sensory perception.
The impact of glycerol monostearate's similarity to fats and fatty acid composition of fats on fat crystallization, destabilization, and texture properties of ice cream

Abstract:Background: Fat significantly affects the properties of ice cream. Prior studies have investigated the correlation between fat crystallization, fat destabilization, and ice cream quality. However, the role of fatty acid composition, the similarity between fat and emulsifier in these characteristics, and their impact on final product quality remains unclear. Results: To investigate the influence of the fatty acid composition of fats, as well as their similarity to glycerol monostearate (GMS), on fat crystallization and destabilization during the aging and freezing stages, ice creams were formulated using a combination of two types of fats (coconut oil and palm olein) in five different ratios. In oil phases, decreased saturation of fatty acids (from 93.38% to 46.69%) and increased similarity to GMS (from 11.96% to 46.01%) caused a reduction in the maximum solid fat content. Moreover, the rise in unsaturated long-chain fatty acids (from 34.61% to 99.57%) and similarity to GMS enhanced the formation of rare and coarse fat crystals, leading to a sparse crystalline network. This, in turn, reduced the crystallization rate and the stiffness of the fat in emulsions. Assuming consistent overrun across all ice creams, the enhanced interactions between fat globules in ice cream improved its hardness, melting properties, and shrinkage. Conclusion: The crystalline properties of fat in emulsions were influenced by oil phases, impacting fat destabilization and ultimately enhancing the quality of ice cream. The present study offers valuable insights for the optimization of fat and monoglyceride fatty acid ester selection, with the potential to improve ice cream quality. © 2023 Society of Chemical Industry.

Abstract:Commercial shortenings and margarines are mainly produced via rapid cooling with scraped surface heat exchangers (SSHE) and therefore, controlling fat crystallization in the cooling process of these systems plays an important role. In this study, four batches of puff pastry shortening were produced from palm oil blends containing 25% and 50% anhydrous milk fat (AMF) using a bench-top SSHE. The outlet temperature of SSHE was investigated at 20 °C and 12 °C relating to the difference of crystallization temperature between palm oil and milk fat. Physico-chemical properties, crystal morphology and fat stability of these shortenings during storage at 5 °C were determined by various classical technologies including differential scanning calorimetry (DSC), polarized light microscopy (PLM), rheological and texture analyzer. Our results showed that fat blends crystallized at 12 °C had a stronger crystal network after production but the discrepancy of hardness diminished after the first storage week at 5 °C. Besides, the change of crystallization temperature did not affect significantly melting behavior as well as fat polymorphism of products. Upon storage, some big aggregates were detected in shortenings as a combined result of the recrystallization of palm fat crystals and the post-crystallization of milk fat triacylglycerols (TAGs).

Abstract:High-pressure-jet (HPJ) processing of various dairy systems has been shown to disrupt fat droplets and casein micelles and cause a strong association between fat and casein proteins. The present work seeks to better describe this association between fat and casein using a model milk formulated from confectionary coating fat (3.6% wt/wt), micellar casein (3.4% wt/wt), and water (93% wt/wt), which was then pasteurized, homogenized, and then either HPJ-treated (400 MPa) or not (non-HPJ-treated, control). Upon ultracentrifugation, fat in the non-HPJ-treated model milk creamed due to its low density. In the HPJ-treated model milk, fat precipitated with protein into a thick bottom layer upon ultracentrifugation, reflecting a strong association between protein and fat. Differential scanning calorimetry (DSC) and time-domain nuclear magnetic resonance of the non-HPJ-treated model milk revealed fat in 2 physical states: (1) fat that is physically similar to the bulk fat and (2) fat that was in smaller droplets (i.e., homogenized) and crystallized at a lower temperature than the bulk fat. In contrast, DSC of HPJ-treated model milks supported the presence of fat in 3 states: (1) fat that is physically similar to the bulk fat, (2) fat in small droplets that required substantial supercooling beyond the non-HPJ-treated model milk to crystallize, and (3) fat in such small domains that it crystallizes in a less stable polymorphic form than the non-HPJ-treated model milk (or does not crystallize at all). The state of fat within the HPJ-treated model milk changed minimally with acidification, indicating that the association is not dependent on the charge on the casein. Cryogenic transmission electron microscopy (Cryo-TEM) of the non-HPJ-treated model milk revealed uniform casein micelles, which likely adsorbed to the surface of fat globules post-homogenization. In contrast, Cryo-TEM of the HPJ-treated model milk revealed a porous protein aggregate that likely had dispersed fat throughout. Together, these results suggest that HPJ treatment causes fat to be entrapped by casein proteins in very small domains.
Beef tallow/lard blends in O/W emulsions: Characterization of fat crystals, partial coalescence, rheology, and aeration performance

Abstract:This study aimed to find new strategies for enhancing the stability and texture properties of aerated emulsion by combining different animal fats with different ratios. Beef tallow (BT)/ lard (LA) were mixed at different ratios to prepare oil-in-water (O/W) emulsions, with and without aeration. The compatibility, crystallization behavior, stability, and rheology in both O/W and aerated emulsion systems prepared with BT/LA binary blends were further investigated. Larger and inhomogeneous β' and β crystal mixtures appeared as the BT ratio increased. Monotectic or eutectic interaction was displayed according to different BT/LA ratios, solid fat content (SFC) and temperatures. O/W emulsion prepared with BT/LA binary showed higher apparent viscosity with larger fat globules distributed as the BT ratio increased. BT had higher SFC at any given temperature and the fat globule aggregation extent was higher. Partial coalescence occurred as the LA ratio increased when SFC < 35 %. Higher foam firmness of the aerated emulsion was achieved by BT/LA binary with higher BT ratios. As a result, combining BT and LA with different ratios achieved higher emulsion stability and foam properties. This study provides a novel insight into the application of different animal fats and the improvement of high-quality whippable products.

Abstract:In this work, the effect of storage time on the fat crystallization, rheological and whipping characteristics of emulsions was studied and the static destabilization mechanism during storage was explored. As the storage time prolonged, peak melting temperature and onset of melting temperature increased while both the crystallization temperature and crystallization rate increased. Crystal birefringence was more pronounced at the oil/water interface accompanied by the desorption of interfacial proteins from fat droplets. The droplet size (d4,3) began to increase significantly (p < 0.05) from the 5th month. The viscosity and the elastic modulus increased from 505.2 mPa·s to 908.4 mPa·s, and from 23.53 Pa to 51.38 Pa, respectively, as storing from 1st to 7th month. The whipping time decreased while the partial coalescence rate increased from 50.84 % to 65.34 %. The whipped cream at the 3rd month exhibited a smooth surface, whereas a rough surface and lost gloss was observed at the 7th month.

Abstract:Currently, the poor whipping capabilities of anhydrous milk fat (AMF) in aerated emulsion products are a major obstacle for their use in beverages like tea and coffee, as well as in cakes and desserts, presenting fresh hurdles for the food industry. In this study, the mechanism of action of diacylglycerols (DAGs) with different carbon chain lengths and degrees of saturation on the partial coalescence of aerated emulsions was systematically investigated from three fundamental perspectives: fat crystallization, air-liquid interface rheology, and fat globule interface properties. The optimized crystallization of long carbon chain length diacylglycerol (LCD) based on stearate enhances interactions between fat globules at the air-liquid interface (with an elastic modulus E' reaching 246.42 mN/m), leading to a significantly reduced interface membrane strength. This promotes fat crystal-membrane interactions during whipping, resulting in a thermally stable foam structure with excellent shaping capability due to enhanced partial coalescence of fat globules. Although Laurate based medium carbon chain length diacylglycerol (MCD) promoted fat crystallization and optimized interface properties, it showed weaker foam properties because it did not adequately encapsulate air bubbles during whipping. Conversely, oleate long carbon chain length diacylglycerol (OCD) proved to be ineffective in facilitating fat crystal-membrane interaction, causing foam to have a subpar appearance. Hence, drawing from the carefully examined fat crystal-membrane interaction findings, a proposed mechanism sheds light on how DAGs impact the whipping abilities of aerated emulsions. This mechanism serves as a blueprint for creating aerated emulsions with superior whipping capabilities and foam systems that are resistant to heat.

Abstract:<p>Various foods are processed with addition of mono- and diacylglycerol (MAG and DAG) emulsifiers to adjust techno-functional properties. Exposure to and application of high temperatures during production, processing and transport of these emulsifiers can induce compositional changes and thereby affect techno-functional properties. Emulsifiers were stored above their respective melting point and their chemical composition was determined by high-performance thin-layer chromatography–fluorescence detection. Storage for 8 weeks decreased the MAG content of a saturated MAG by about 36% by transesterification into 1,3-DAG, while a rearrangement of 1,2- into 1,3-DAG was observed for a saturated MAG/DAG emulsifier. Emulsifiers were applied to aerosol whipping cream, and viscosity, particle size, overrun, foam firmness and drainage were determined. The increasing 1,3-DAG content of the saturated MAG emulsifier was found to increase the drainage of aerosol whipping cream from 15% to 50%.</p>

Abstract:Abstract The influence of low concentrations (0.1‐5%) of fat crystals on the stability of water‐in‐soybean oil emulsions was examined by light scattering and sedimentation experiments. Both the initial flocculation/coalescence rate and long‐term stability against water separation were determined. The initial flocculation/coalescence rate increased upon addition of small amounts of fat crystals. When the crystal concentration was increased above a critical concentration (specific to a system), a decrease in the flocculation/coalescence rate occurred. The increased flocculation/coalescence rate is likely the effect of bridging of water droplets by fat crystals. Fat crystal wetting by water is an important criterion for this phenomenon to occur. Emulsion stabilization for crystal concentrations above critical is caused by a mechanical screening of water droplets. The presence of considerable amounts of crystals in oil also lowered the density difference between droplet and medium, and enhanced viscosity. The degree of increase in viscosity depended upon the emulsifier. Both a decrease in density difference and an increase in viscosity play a role in hindering flocculation/coalescence of droplets. In long‐term studies of water separation, all concentrations of fat crystals stabilized the water‐in‐oil emulsions. The droplet size of these emulsions increased until the critical droplet size was approached where the screening effect of crystals on the droplets no longer stabilized the emulsions. The stabilizing effect for emulsions with monoolein was continuously improved by increasing the amount of crystals up to 5%. For lecithin‐stabilized emulsions, an optimal effect was achieved for fact crystal concentrations of 1–2%.

Abstract:Milk fat globules or milk fat globule membranes (MFGs/MFGM) have been added in the infant formula to fortify the phospholipids and narrow down the nutritional gap from breast milk. The main aim of this study was to profile the interfacial and thermal properties of MFGs/MFGM prepared from ultrasonicated bovine milk. Bovine milk was sonicated at ultrasonic intensities of 20 kHz and 40 kHz independently or synchronously with the duration time of 0 min (control), 5 min, 10 min and 15 min (work/rest cycles = 5 s: 3 s). Ultrasonic treatments at 20 kHz/ 5 min and 20 + 40 kHz/ 5 min improved the volume density (%) of smaller particles (1–10 μm), while significantly decreased the surface hydrophobicity (H 0 ) (p < 0.05). 40 kHz/5 min samples showed significantly higher ζ- potential than the other samples (p < 0.05), which might be because more negative charges were detected. In comparison with control samples, ultrasonic treatments decreased the interfacial tension (π) between the air and MFGs/MFGM liquid phase. 20 kHz ultra-sonicated treatments decreased the diffusion rate ( k diff ) of MFGs/MFGM interfacial compositions significantly as the duration prolonged from 5 min to 15 min (p < 0.05) but did not affect the adsorption or penetration rate ( k a ) (p > 0.05). X-ray diffraction (XRD) results showed that α-crystal peaks only existed in control and ultrasonicated 5 min samples, but disappeared in all 15 min samples. According to the different scanning calorimetry (DSC), one or two new exothermic events (in the range of 17.29 – 18.81 °C and 22.14 – 25.21 °C) appeared after ultrasonic treatments which, however, were not found in control samples. Ultrasonic treatments resulted in the low-melting fractions (LMF) (T M1 ) peaks undetectable in MFGs/MFGM samples in which only peaks of medium-melting fractions (MMF) (T M2 ) and high-melting fractions (HMF) (T M3 ) were detected. Compared with the control, both enthalpy of crystallisation (ΔH C ) and melting (ΔH M ) decreased in ultrasonicated samples. In conclusion, ultrasonic treatment affects the interfacial and thermal properties of MFGs/MFGM.

Abstract:The effects of whey protein, basil seed gum (BSG), and κ-carrageenan (CGN) on the structure–rheology interactions of low- and high-fat cream were investigated. Pseudoplastic and thixotropic behavior of cream was found for all the samples and the pseudoplasticity was increased with an increased level of stabilizers. The apparent viscosity ( η a ) of the forward curves is greater than that of the backward ones, which may be the result of the breakdown of the fat globule structure under shear stress. The viscosity of cream was reduced, while using a stabilizer (BSG/CGN) can be related to the water binding of hydrocolloid molecules contributing to resistance in flow. For all samples, elastic modulus was greater than viscous modulus, indicating a greater contribution from elastic characteristics. With the increase of BSG/CGN levels, the molecules may be competitively adsorbed onto the surface of fat droplets, thereby changing its surface tension and decreasing its particle size. Increases in whey proteins, fat, and BSG also significantly increased hardness, whereas increases in CGN significantly decreased it. The globular aggregates in the microstructure of high-fat dairy cream were smaller than those in low-fat dairy cream, allowing more water to be retained in the high-fat samples. Therefore, synergistic interactions between polysaccharides and proteins may encourage the formation of a cross-linked network.

Influence of protein and solid fat content on mechanical properties and comminution behavior of structured plant-based lipids

Abstract:Recently an approach has been developed to structure plant-based lipids with the intention to mimic animal fat tissue in processed meat products or analogues. This study investigated the comminution behavior in a bowl chopper of such structured lipids with varying mechanical properties. For products like salami-type sausages these systems need to withstand comminution to yield particles for inclusion in product matrices. Therefore, samples were prepared from protein suspensions with 6%, 8%, 10%, and 12% soy protein isolate (SPI) and 70% (w/w) total fat with varying solid fat contents (0-30%, w/w). The hardness of samples prepared with 6% and 8% SPI varied between 4.5 and 35.9 N. When comminuted in a bowl chopper, these structures had insufficient mechanical strengths to facilitate the formation of small particles and yielded a coarse paste. Higher concentrations of protein increased hardness (15.9-76.2 N and 15.6-96.1 N, for 10% and 12% SPI, respectively). These samples retained their structural integrity upon comminution yielding individual intact particles. The size of these particles increased with sample firmness, i.e. with increasing amount of protein. The shape of the particles was more elongated the higher the solid fat content as indicated by a higher aspect ratio. Taken together, results show that structural characteristics of the gelled emulsions can be tuned to yield desired fat particles after comminution.
In Situ Single-Droplet Analysis of Emulsified Fat by Confocal Raman Microscopy: Insights into Crystal Network Formation within Spatial Resolution

Abstract:Fat crystallization is one of the predominant factors influencing the structure and properties of fat-containing emulsions. In the present study, the role of emulsifiers on fat crystallization dynamics within droplet multiphase systems was evaluated via single-droplet analysis, taking advantage of the non-destructive properties of confocal Raman microscopy. Palm oil droplets dispersed in water were used as a model system, due to palm oil's well-known crystallization properties. Emulsion droplets of the same size were generated using two different emulsifiers (Whey Protein Isolate and Tween 60), at various concentrations. Fast and slow cooling treatments were applied to affect fat crystallisation and network formation as well as droplet morphology, and crystallization dynamics. Raman imaging analysis demonstrated that the chemical structure and concentration of the emulsifier significantly influenced both crystal nucleation within the droplets, as well as the spatial distribution and morphology of the fat crystal network. Additionally, analysis of the spectra of the crystallized phase provided essential information regarding the impact of the emulsifiers on the microstructure, degree of structural order, and structural arrangements of the fat crystal networks. Furthermore, by performing single droplet analysis during cooling it was possible to observe shape distortions in Tween 60 stabilized droplets, as a consequence of the formation of a three-dimensional network of fat crystals that strongly interacted with the interface. On the other hand, the droplets retained their shape when whey proteins were absorbed at the interface. Confocal Raman microscopy, in combination with polarized light microscopy, is, therefore, a well-suited tool for in situ, single-droplet analysis of emulsified oil systems, providing essential information about emulsified fat crystallization dynamics, contributing to better understanding and designing products with enhanced structure and function.

Abstract:In this paper, two emulsion systems with high and low solid fat contents were prepared from 20 % water phase and 80 % oil phase by adjusting the palm oil/palm stearin/soybean oil ratio. Different ultrasonic power and time were used for the pretreatment of emulsion with different solid fat content, and the application characteristics of ultrasonic in W/O emulsions were explored and evaluated. Directly using high-intensity ultrasound to prepare fatty emulsions would weaken the hardness and storage modulus G' of the samples. Although ultrasound reduced the size of fat crystals in emulsions, the interaction between water droplets and fat crystals needs to be considered. After ultrasonic treatment, water droplets were difficult to immobilize on the crystal surface and thus acted as an active filler to stabilize the emulsion together with the fat crystal network. In high solid fat emulsion systems, an increase in ultrasound power (from 100 W to 200 W) could more affect the crystallization behavior of fats than an increase in ultrasound duration (from 30 s to 60 s), and the distribution of crystals and droplets was more uniform. In the low solid fat emulsion system, the texture of the sample after ultrasonic treatment was softer, and the surface was more delicate and smoother. However, the higher ultrasonic intensity (200 W) was not conducive to the preparation of the spread. Although the ultrasound with excessive intensity promoted the formation of small crystals, it would also lead to the aggregation of small crystals. These small crystals cannot form a uniform crystal network, which increases the fluidity of emulsions.

Abstract:The functional and sensory properties of food emulsion are thought to be complicated and influenced by many factors, such as the emulsifier, oil/fat mass fraction, and size of oil/fat droplets. In addition, the perceived texture of food emulsion during oral processing is mainly dominated by its rheological and tribological responses. This study investigated the effect of droplet size distribution as well as the content of oil droplets on the lubrication and sensory properties of o/w emulsion systems. Friction curves for reconstituted milk samples (composition: skimmed milk and milk cream) and Casein sodium salt (hereinafter referred to as CSS) stabilized model emulsions (olive oil as oil phase) were obtained using a soft texture analyzer tribometer with a three ball-on-disc setup combined with a soft surfaces (PDMS) tribology system. Sensory discrimination was conducted by 22 participants using an intensity scoring method. Stribeck curve analyses showed that, for reconstituted milk samples with similar rheological properties, increasing the volume fraction of oil/fat droplets in the size range of 1–10 μm will significantly enhance lubrication, while for CSS-stabilized emulsions, the size effect of oil/fat droplets reduced to around 1 μm. Surprisingly, once the size of oil/fat droplets of both systems reached nano size (d90 = 0.3 μm), increasing the oil/fat content gave no further enhancement, and the friction coefficient showed no significant difference (p > 0.05). Results from sensory analysis show that consumers are capable of discriminating emulsions, which vary in oil/fat droplet size and in oil/fat content (p < 0.01). However, it appeared that the discrimination capability of the panelist was significantly reduced for emulsions containing nano-sized droplets.

Abstract:The immunomodulator AS101 has been found previously by us to stimulate the secretion of high levels of interleukin 1 and colony stimulating factor (CSF) in vitro, as well as the production of CSF in vivo in mice models. These cytokines are known to induce proliferation and differentiation of hematopoietic progenitor cells from the spleen and bone marrow (BM) and to protect mice from DNA-damaging agents. The present studies were designed to evaluate the effects of prolonged treatment with AS101 on myelopoiesis, BM cellularity, and CSF secretion in mice treated with a sublethal dose of cyclophosphamide (CYP) and on the survival of mice undergoing treatment with lethal doses of this compound. In this model, the hematopoietic progenitors were suppressed during the overbound phase of myelopoiesis resulting from the cytotoxic effects of CYP. This allowed the detection of a significant proliferative effect of AS101 in vivo on BM colony-forming units granulocyte-macrophage progenitor cells, BM cellularity, and the secretion of CSF. Moreover, AS101 protected these animals from the lethal effects of high doses of CYP. These protective effects were demonstrable only when AS101 was administered to mice prior to CYP treatment. The only exception was CSF secretion by spleen cells that had been reconstituted when AS101 was administered both prior to and following CYP treatment. AS101 was found to have a synergistic effect with CYP in the treatment of tumor-bearing mice, suggesting that the combination of these two modalities provides a more effective treatment of their tumors. These results strongly suggest an immunoregulatory role for AS101 in counteracting the chemotherapy-induced hematopoietic suppression as well as usefulness as adjunct treatment of cancer when used in combination with CYP.
