Abstract:Grass-based diets are of increasing social-economic importance in dairy cattle farming, but their low supply of glucogenic nutrients may limit the production of milk. Current evaluation systems that assess the energy supply and requirements are based on metabolisable energy (ME) or net energy (NE). These systems do not consider the characteristics of the energy delivering nutrients. In contrast, mechanistic models take into account the site of digestion, the type of nutrient absorbed and the type of nutrient required for production of milk constituents, and may therefore give a better prediction of supply and requirement of nutrients. The objective of the present study is to compare the ability of three energy evaluation systems, viz. the Dutch NE system, the agricultural and food research council (AFRC) ME system, and the feed into milk (FIM) ME system, and of a mechanistic model based on Dijkstra et al. [Simulation of digestion in cattle fed sugar cane: prediction of nutrient supply for milk production with locally available supplements. J. Agric. Sci., Cambridge 127, 247–60] and Mills et al. [A mechanistic model of whole-tract digestion and methanogenesis in the lactating dairy cow: model development, evaluation and application. J. Anim. Sci. 79, 1584–97] to predict the feed value of grass-based diets for milk production. The dataset for evaluation consists of 41 treatments of grass-based diets (at least 0.75g ryegrass/g diet on DM basis). For each model, the predicted energy or nutrient supply, based on observed intake, was compared with predicted requirement based on observed performance. Assessment of the error of energy or nutrient supply relative to requirement is made by calculation of mean square prediction error (MSPE) and by concordance correlation coefficient (CCC). All energy evaluation systems predicted energy requirement to be lower (6–11%) than energy supply. The root MSPE (expressed as a proportion of the supply) was lowest for the mechanistic model (0.061), followed by the Dutch NE system (0.082), FIM ME system (0.097) and AFRC ME system (0.118). For the energy evaluation systems, the error due to overall bias of prediction dominated the MSPE, whereas for the mechanistic model, proportionally 0.76 of MSPE was due to random variation. CCC analysis confirmed the higher accuracy and precision of the mechanistic model compared with energy evaluation systems. The error of prediction was positively related to grass protein content for the Dutch NE system, and was also positively related to grass DMI level for all models. In conclusion, current energy evaluation systems overestimate energy supply relative to energy requirement on grass-based diets for dairy cattle. The mechanistic model predicted glucogenic nutrients to limit performance of dairy cattle on grass-based diets, and proved to be more accurate and precise than the energy systems. The mechanistic model could be improved by allowing glucose maintenance and utilization requirements parameters to be variable.

Abstract:OBJECTIVE To understand the state of Chlamydia pneumoniae (Cpn) infection in patients with coronary heart disease (CHD), and explore the relationship between Cpn infection and the gonesis and progressin of CHD. METHOD By adopting sandwich method with double antibodies and indirect ELISA method, we detected serum Cpn immune complex and IgG antibody in 160 CHD patients (confirmed by coronary artery angiography) and 40 subjects without CHD. RESULTS The positivity rate of Cpn IgG type immune complex in the CHD group (63.8%) was significantly higher than that in the control group (40.0%, P<0.05), similar to the results of Cpn IgG antibody detection (65.0% vs 32.5%, P<0.05). CONCLUSION Cpn infection is closely connected with the CHD, but their exact relationship needs further exploration and study.
Assessing different cross-validation schemes for predicting novel traits using sensor data: An application to dry matter intake and residual feed intake using milk spectral data

Abstract:Feed efficiency is important for economic profitability of dairy farms; however, recording daily DMI is expensive. Our objective was to investigate the potential use of milk mid-infrared (MIR) spectral data to predict proxy phenotypes for DMI based on different cross-validation schemes. We were specifically interested in comparisons between a model that included only MIR data (model M1); a model that incorporated different energy sink predictors, such as body weight, body weight change, and milk energy (model M2); and an extended model that incorporated both energy sinks and MIR data (model M3). Models M2 and M3 also included various cow-level variables (stage of lactation, age at calving, parity) such that any improvement in model performance from M2 to M3, whether through a smaller root mean squared error (RMSE) or a greater squared predictive correlation (R2), could indicate a potential benefit of MIR to predict residual feed intake. The data used in our study originated from a multi-institutional project on the genetics of feed efficiency in US Holsteins. Analyses were conducted on 2 different trait definitions based on different period lengths: averaged across weeks versus averaged across 28 d. Specifically, there were 19,942 weekly records on 1,812 cows across 46 experiments or cohorts and 3,724 28-d records on 1,700 cows across 43 different cohorts. The cross-validation analyses involved 3 different k-fold schemes. First, a 10-fold cow-independent cross-validation was conducted whereby all records from any one cow were kept together in either training or test sets. Similarly, a 10-fold experiment-independent cross-validation kept entire experiments together, whereas a 4-fold herd-independent cross-validation kept entire herds together in either training or test sets. Based on cow-independent cross-validation for both weekly and 28-d DMI, adding MIR predictors to energy sinks (model M3 vs. M2) significantly (P < 10-10) reduced average RMSE to 1.59 kg and increased average R2 to 0.89. However, adding MIR to energy sinks (M3) to predict DMI either within an experiment-independent or herd-independent cross-validation scheme seemed to demonstrate no merit (P > 0.05) compared with an energy sink model (M2) for either R2 or RMSE (respectively, 0.68 and 2.55 kg for M2 in herd-independent scheme). We further noted that with broader cross-validation schemes (i.e., from cow-independent to experiment-independent to herd-independent schemes), the mean and slope bias increased. Given that proxy DMI phenotypes for cows would need to be almost entirely generated in herds having no DMI or training data of their own, herd-independent cross-validation assessments of predictive performance should be emphasized. Hence, more research on predictive algorithms suitable for broader cross-validation schemes and a more earnest effort on calibration of spectrophotometers against each other should be considered.

Abstract:Corrected fluorescence properties of yeast alcohol dehydrogenase and its coenzyme complexes have been investigated as a function of temperature. Dissociation constants have been obtained for binary and ternary complexes of NAD and NADH by following the enhancement of NADH fluorescence or the quenching of the protein fluorescence. It is found that the presence of pyrazole increases the affinity of NAD to the enzyme approximately 100-fold. The formation of the ternary enzyme - NAD - pyrazole complex is accompanied by a large change in the ultraviolet absorption properties, with a new band in the 290-nm region. Significant optical changes also accompany the formation of the ternary enzyme-NADH-acetamide complex. The possible origin for the quenching of the protein fluorescence upon coenzyme binding is discussed, and it is suggested that a coenzyme-induced conformational change can cause it. Thermodynamic parameters associated with NAD and NADH binding have been evaluated on the basis of the change of the dissociation constants with temperature. Optical and thermodynamic properties of binary and ternary complexes of yeast alcohol dehydrogenase are compared with the analogous properties of horse liver alcohol dehydrogenase.
Predicting Daily Dry Matter Intake Using Feed Intake of First Two Hours after Feeding in Mid and Late Lactation Dairy Cows with Fed Ration Three Times Per Day

Abstract:The objective of this study was to evaluate the feasibility of using the dry matter intake of first 2 h after feeding (DMI-2h), body weight (BW), and milk yield to estimate daily DMI in mid and late lactating dairy cows with fed ration three times per day. Our dataset included 2840 individual observations from 76 cows enrolled in two studies, of which 2259 observations served as development dataset (DDS) from 54 cows and 581 observations acted as the validation dataset (VDS) from 22 cows. The descriptive statistics of these variables were 26.0 ± 2.77 kg/day (mean ± standard deviation) of DMI, 14.9 ± 3.68 kg/day of DMI-2h, 35.0 ± 5.48 kg/day of milk yield, and 636 ± 82.6 kg/day of BW in DDS and 23.2 ± 4.72 kg/day of DMI, 12.6 ± 4.08 kg/day of DMI-2h, 30.4 ± 5.85 kg/day of milk yield, and 597 ± 63.7 kg/day of BW in VDS, respectively. A multiple regression analysis was conducted using the REG procedure of SAS to develop the forecasting models for DMI. The proposed prediction equation was: DMI (kg/day) = 8.499 + 0.2725 × DMI-2h (kg/day) + 0.2132 × Milk yield (kg/day) + 0.0095 × BW (kg/day) (R2 = 0.46, mean bias = 0 kg/day, RMSPE = 1.26 kg/day). Moreover, when compared with the prediction equation for DMI in Nutrient Requirements of Dairy Cattle (2001) using the independent dataset (VDS), our proposed model shows higher R2 (0.22 vs. 0.07) and smaller mean bias (−0.10 vs. 1.52 kg/day) and RMSPE (1.77 vs. 2.34 kg/day). Overall, we constructed a feasible forecasting model with better precision and accuracy in predicting daily DMI of dairy cows in mid and late lactation when fed ration three times per day.
Development and evaluation of predictive models for pregnancy risk in UK dairy cows

Abstract:One suggested approach to improve the reproductive performance of dairy herds is through the targeted management of subgroups of biologically similar animals, such as those with similar probabilities of becoming pregnant, termed pregnancy risk. We aimed to use readily available farm data to develop predictive models of pregnancy risk in dairy cows. Data from a convenience sample of 108 dairy herds in the UK were collated and each herd was randomly allocated, at a ratio of 80:20, to either training or testing data sets. Following data cleaning, there were a total of 78 herds in the training data set and 20 herds in the testing data set. Data were further split by parity into nulliparous, primiparous, and multiparous subsets. An XGBoost model was trained to predict the insemination outcome in each parity subset, with predictors from farm records of breeding, calving and milk recording. Training data comprised 74,511 inseminations in 45,909 nulliparous animals, 86,420 inseminations in 39,439 primiparous animals, and 158,294 inseminations in 32,520 multiparous animals. The final models were evaluated by predicting with the testing data, comprising 31,740 inseminations in 19,647 nulliparous animals, 38,588 inseminations in 16,215 primiparous animals, and 65,049 inseminations in 12,439 multiparous animals. Model discrimination was assessed by calculating the area under receiver operating characteristic curves (AUC); model calibration was assessed by plotting calibration curves and compared across test herds by calculating the expected calibration error (ECE) in each test herd. The models were unable to discriminate between insemination outcomes with high accuracy, with an AUC of 0.63, 0.59 and 0.62 in the nulliparous, primiparous and multiparous subsets, respectively. The models were generally well-calibrated, meaning the model-predicted pregnancy risks were similar to the observed pregnancy risks. The mean (SD) ECE in the test herds was 0.038 (0.023), 0.028 (0.012) and 0.020 (0.008) in the nulliparous, primiparous and multiparous subsets respectively. The predictive models reported here could theoretically be used to identify subgroups of animals with similar pregnancy risk to facilitate targeted reproductive management; or provide information about cows' relative pregnancy risk compared with the herd average, which may support on-farm decision-making. Further research is needed to evaluate the generalizability of these predictive models and understand the source of variation in ECE between herds; however, this study demonstrates that it is possible to accurately predict pregnancy risk in dairy cows using readily available farm data.
Predicting CO2 production of lactating dairy cows from animal, dietary, and production traits using an international dataset

Abstract:Automated measurements of the ratio of concentrations of methane and carbon dioxide, [CH4]:[CO2], in breath from individual animals (the so-called "sniffer technique") and estimated CO2 production can be used to estimate CH4 production, provided that CO2 production can be reliably calculated. This would allow CH4 production from individual cows to be estimated in large cohorts of cows, whereby ranking of cows according to their CH4 production might become possible and their values could be used for breeding of low CH4-emitting animals. Estimates of CO2 production are typically based on predictions of heat production, which can be calculated from body weight (BW), energy-corrected milk yield, and days of pregnancy. The objectives of the present study were to develop predictions of CO2 production directly from milk production, dietary, and animal variables, and furthermore to develop different models to be used for different scenarios, depending on available data. An international dataset with 2,244 records from individual lactating cows including CO2 production and associated traits, as dry matter intake (DMI), diet composition, BW, milk production and composition, days in milk, and days pregnant, was compiled to constitute the training dataset. Research location and experiment nested within research location were included as random intercepts. The method of CO2 production measurement (respiration chamber [RC] or GreenFeed [GF]) was confounded with research location, and therefore excluded from the model. In total, 3 models were developed based on the current training dataset: model 1 ("best model"), where all significant traits were included; model 2 ("on-farm model"), where DMI was excluded; and model 3 ("reduced on-farm model"), where both DMI and BW were excluded. Evaluation on test dat sets with either RC data (n = 103), GF data without additives (n = 478), or GF data only including observations where nitrate, 3-nitrooxypropanol (3-NOP), or a combination of nitrate and 3-NOP were fed to the cows (GF+: n = 295), showed good precision of the 3 models, illustrated by low slope bias both in absolute values (-0.22 to 0.097) and in percentage (0.049 to 4.89) of mean square error (MSE). However, the mean bias (MB) indicated systematic overprediction and underprediction of CO2 production when the models were evaluated on the GF and the RC test datasets, respectively. To address this bias, the 3 models were evaluated on a modified test dataset, where the CO2 production (g/d) was adjusted by subtracting (where measurements were obtained by RC) or adding absolute MB (where measurements were obtained by GF) from evaluation of the specific model on RC, GF, and GF+ test datasets. With this modification, the absolute values of MB and MB as percentage of MSE became negligible. In conclusion, the 3 models were precise in predicting CO2 production from lactating dairy cows.
Predicting nitrogen use efficiency, nitrogen loss and dry matter intake of individual dairy cows in late lactation by including mid-infrared spectra of milk samples

Abstract:Background: Nitrate leaching to groundwater and surface water and ammonia volatilization from dairy farms have negative impacts on the environment. Meanwhile, the increasing demand for dairy products will result in more pollution if N losses are not controlled. Therefore, a more efficient, and environmentally friendly production system is needed, in which nitrogen use efficiency (NUE) of dairy cows plays a key role. To genetically improve NUE, extensively recorded and cost-effective proxies are essential, which can be obtained by including mid-infrared (MIR) spectra of milk in prediction models for NUE. This study aimed to develop and validate the best prediction model of NUE, nitrogen loss (NL) and dry matter intake (DMI) for individual dairy cows in China. Results: A total of 86 lactating Chinese Holstein cows were used in this study. After data editing, 704 records were obtained for calibration and validation. Six prediction models with three different machine learning algorithms and three kinds of pre-processed MIR spectra were developed for each trait. Results showed that the coefficient of determination (R2) of the best model in within-herd validation was 0.66 for NUE, 0.58 for NL and 0.63 for DMI. For external validation, reasonable prediction results were only observed for NUE, with R2 ranging from 0.58 to 0.63, while the R2 of the other two traits was below 0.50. The infrared waves from 973.54 to 988.46 cm-1 and daily milk yield were the most important variables for prediction. Conclusion: The results showed that individual NUE can be predicted with a moderate accuracy in both within-herd and external validations. The model of NUE could be used for the datasets that are similar to the calibration dataset. The prediction models for NL and 3-day moving average of DMI (DMI_a) generated lower accuracies in within-herd validation. Results also indicated that information of MIR spectra variables increased the predictive ability of models. Additionally, pre-processed MIR spectra do not result in higher accuracy than original MIR spectra in the external validation. These models will be applied to large-scale data to further investigate the genetic architecture of N efficiency and further reduce the adverse impacts on the environment after more data is collected.
Development of a dynamic model for prediction of energy in milk protein, lactose, fat, and enteric methane emissions in goats based on energy balance and indirect calorimetry studies

Abstract:Feed costs are overwhelmingly the largest expense for dairy producers. Thus, improving milk production efficiency (milk fat and protein are the main incomes for farmers) is of great economic importance in the dairy industry. The main objective of this study was to develop a dynamic energy partitioning model to describe and quantify how dietary energy from carbohydrate, protein, and fat is transferred to milk (protein, lactose, and fat) in dairy goats. In addition, due to increasing worldwide concerns regarding livestock contribution to global warming, methane (CH4) emission was quantified. For modeling purposes, 158 individual goat observations were used and randomly split into 2/3 for model development and 1/3 for internal evaluation. For external evaluation, 20 different energy balance studies from the literature (77 observations) were evaluated. The Root Mean Square Prediction Error (RMSPE) was 13.2% for loss of energy in CH4, 16.8% for energy in fat, 19.4% for energy in protein, and 22.3 energy in lactose. Mean bias was around zero for all variables and the slope bias was zero for milk energy in lactose, close to 1% for milk fat (1.01%), and around 3% and 10% for protein and CH4, respectively. Random bias was greater than 85% for energy in CH4 and milk energy components indicating non-systematic errors and that the equation in the model fitted the data properly. Analyses of residuals appeared to be randomly distributed around zero. Slopes of regression lines for residuals vs. predicted were positive for milk fat energy, zero for lactose, and negative for milk energy in protein and CH4. This model suggested for use with mixed diets and by-products to obtain balanced macronutrient supply, methane emissions, and milk performance during mid lactation could be an interesting tool to help farmers simulate scenarios that increase milk fat and protein, evaluate CH4 emissions, without the costs of running animal trials.

Abstract:Predicting methane (CH4) emission from milk mid-infrared (MIR) spectra provides large amounts of data which is necessary for genomic selection. Recent prediction equations were developed using the GreenFeed system, which required averaging multiple CH4 measurements to obtain an accurate estimate, resulting in large data loss when animals unfrequently visit the GreenFeed. This study aimed to determine if calibrating equations on CH4 emissions corrected for diurnal variations or modeled throughout lactation would improve the accuracy of the predictions by reducing data loss compared with standard averaging methods used with GreenFeed data. The calibration dataset included 1 822 spectra from 235 cows (Holstein, Montbéliarde, and Abondance), and the validation dataset included 104 spectra from 46 (Holstein and Montbéliarde). The predictive ability of the equations calibrated on MIR spectra only was low to moderate (R2v = 0.22-0.36, RMSE = 57-70 g/d). Equations using CH4 averages that had been pre-corrected for diurnal variations tended to perform better, especially with respect to the error of prediction. Furthermore, pre-correcting CH4 values allowed to use all the data available without requiring a minimum number of spot measures at the GreenFeed device for calculating averages. This study provides advice for developing new prediction equations, in addition to a new set of equations based on a large and diverse population.
Large increase of cytotoxic/suppressor T-lymphoblasts and eosinophils around manifestation of diabetes in BB rats.

Abstract:In order to gain more insight into the pathogenetic mechanism leading to beta cell destruction in BB rats we searched for specific changes in the immune system at the time of diabetes development. We performed a density gradient fractionation of all major leukocyte types in the peripheral blood and the spleen of acutely diabetic or non-diabetic BB rats as well as of normal Wistar rats in order to identify proliferating low density blasts. Acutely diabetic BB rats showed a 4-6 times higher percentage of lymphoblasts among spleen cells than non-diabetic BB or Wistar rats. The majority of blasts was of the T-cytotoxic/suppressor (Ox8+) phenotype (61%) in acutely diabetic BB rats, but of the T-helper (W3/25+) phenotype (51%) in non-diabetic BB or Wistar rats. Plasma cell and monocyte numbers were not increased in acutely diabetic BB rats. At the time of diabetes manifestation BB rats develop a pronounced eosinophilia. These observations indicate a state of enhanced cellular immunity involving cytotoxic/suppressor T cells during diabetes development in BB rats.
A Pipeline for Computer Aided Polyp Detection and Segmentation in Ct Colonography

Abstract:CAD-CTC helps the radiologists for the automatic detection of suspicious polyps in the CT images of the colon. In this paper, a novel method is proposed in order to decrease the computational time taken by the CAD system. In the proposed CAD system, segmentation of the colon is done by Otsu's method of thresholding and clustered by k-means for extraction of the candidates. Classification is performed by SVM (Support Vector Machine) and ANN (Artificial Neural Network) from the training and testing images of candidate extraction. The proposed CAD system is evaluated with 16 slices (each) of normal and abnormal images. The CAD system achieved a minimal computation time (10 minutes) and an increased learning rate of about 1.2608 at an epoch 411.Thus the good results demonstrate that the proposed system may provide relevant additional information

Abstract:Abstract Several studies (1–4) indicated that aspirin-induced gastric or GI bleeding is usually a local effect resulting from contact of aspirin particles, or saturated solutions of aspirin surrounding these particles, with the mucosa. Maudlin (5) also stated that bleeding and ulceration are due to a local focal necrosis. However, Woznicki and Mrtek (6) expressed the opinion that the mechanism of aspirin-induced GI blood loss is due to a combination of both local and systemic effects. They explained the systemic effect by the role of aspirin in reducing platelet aggregation through elimination of the second wave of platelet aggregation induced by adenosine diphosphate. These authors defended their opinion by the fact that aspirin is being used experimentally to prevent formation of thromboemboli. However, no work has been published to support this theory.

Abstract:Feed efficiency is an important trait of dairy production. However, assessing feed efficiency is constrained by the associated cost and difficulty in measuring individual feed intake, especially on pastures. The objective of this study was to investigate short-term feed efficiency traits of herbage-fed dairy cows and screening of potential biomarkers (n = 238). Derived feed efficiency traits were ratio-based (i.e., feed conversion ratio (FCR) and N use efficiency (NUE)) or residual-based (i.e., residual feed intake (RFI), residual energy intake (REI), and residual N intake (RNI)). Thirty-eight Holstein and 16 Swiss Fleckvieh dairy cows underwent a 7-d measurement period during mid- and/or late-lactation. The experimental data (n = 100 measurement points) covered different lactational and herbage-fed system situations: mid-lactation grazing (n = 56), late-lactation grazing (n = 28), and late-lactation barn feeding (n = 16). During each measuring period, the individual herbage intake of each cow was estimated using the n-alkane marker technique. For each cow, biomarkers representing milk constituents (n = 109), animal characteristics (n = 13), behaviour, and activity (n = 46), breath emissions (n = 3), blood constituents (n = 35), surface, and rectal temperature (n = 29), hair cortisol (n = 1), and near-infrared (NIR) spectra of faeces and milk (n = 2) were obtained. The relationships between biomarkers and efficiency traits were statistically analysed with univariate linear regression and for NIR spectra using partial least squares regression with feed efficiency traits. The feed efficiency traits were interrelated with each other (r: -0.57 to -0.86 and 0.49-0.81). The biomarkers showed varying R2 values in explaining the variability of feed efficiency traits (FCR: 0.00-0.66, NUE: 0.00-0.74, RFI: 0.00-0.56, REI: 0.00-0.69, RNI: 0.00-0.89). Overall, the feed efficiency traits were best explained by NIR spectral characteristics of milk and faeces (R2: 0.25-0.89). Biomarkers show potential for predicting feed efficiency in herbage-fed dairy cows. NIR spectra data analysis of milk and faeces presents a promising method for estimating individual feed efficiency upon further validation of prediction models. Future applications will depend on the ability to improve the robustness of biomarkers to predict feed efficiency in a greater variety of environments (locations), managing conditions, feeding systems, production intensities, and other aspects.

Abstract:Fifty Holstein cows (2.6 ± 1.0 lactations) in early lactation (52.6 ± 16.4 days in milk) calved in autumn and producing 34.3 (±4.4) kg milk per cow ·day -1 were randomly assigned for 27 weeks to one of two treatments (feeding systems) in 10 groups (pens) of 5 animals each (5 groups/treatment). The confined treatment was a total mixed ration (TMR_100) whereas the supplemented grazing system was defined by a partially mixed ration (PMR_75) with 75% TMR and 25% oat pasture ( Avena sativa L.) during the autum-winter (Period I) and alfalfa pasture ( Medicago sativa L.) plus concentrate (7.0 kg per cow ·day -1) (P + C) during the next spring (Period II). Milk production was daily and individually recorded throughout the trial while the chemical composition of milk was measured on individual samples every 2 weeks. Live weight (LW) and body condition score (BCS) using a scale of 1 to 5 were recorded every 3 weeks after the morning milking. Simultaneously, blood samples were taken for determinations of glucose, urea, non-esterified fatty acids (NEFA), insulin, somatotrophin (GH) and somatomedin C (IGF-I). Total DM intake was measured using the difference method four times per repetition in the months of July, August, September and October. The experimental herd was inseminated at fixed time at the start of the service (May-July) and on the return with estrus detection. The diagnosis of pregnancy was made by transrectal ultrasonography at 30 and 60 days after the start of the service. The productive data were analyzed according to a model with repeated observations in time adjusted by covariate with 2 periods and using the animal as the experimental unit. Data of DM intake were analyzed using ANOVA for 2 treatments with 5 repetitions. Cows from the TMR_100 group produced more milk (kg ·cow -1 ·day -1) during Period I (33.7 vs. 32.3) and Period II (28.8 vs. 27.8) (P < 0.01). Yields were also higher (P < 0.05) in TMR_100 for milk fat (kg ·cow -1 ·day -1) in Periods I (1.33 vs. 1.24) and II (1.10 vs. 0.92) and for milk protein (1.19 vs. 1.13 and 0.99 vs. 0.93). The lower energy intake of grazing cows (45.0 vs. 43.1 and 40.9 vs. 38.9 Mcal per cow · day -1 for TMR_100 vs. PMR_75 and TMR_100 vs. P + C) coupled to the extra energy cost associated with grazing activity would explain the difference in milk production. Milk protein content (%) resulted higher (P < 0.05) in TMR_100 over the two experimental periods (3.53 vs. 3.47 and 3.42 vs. 3.31) while milk fat content (%) was similar between TMR_100 vs. PMR_75 (3.90) but lower in P + C (3.30) compared to TMR_100 (3.87). The greatest LW gain ( kg · cow -1 · day -1) in the cows of the TMR_100 group in both periods (0.69 vs. 0.34 and 0.49 vs. -0.22) was consistent with the increase in the plasmatic levels of glucose and IGF-I and the reduction in the circulating levels of GH, but not with the lack of increases in plasma insulin concentration or decreases in circulating levels of NEFA, parameters that were not affected by treatments. Despite the positive effects of TMR_100 on parameters linked to energy balance, no significant differences were detected in any of the reproductive parameters evaluated and the final rate of pregnancy was 80.0 and 91.3% for the confined and grazing system with supplementation respectively. Free milk liters over feeding costs were higher in the grazing system with supplementation compared to the confined system (18.5 vs. 15.5 l, respectively). The results indicate that the grazing system with supplementation can be economically competitive compared to the confined system of milk production.

Abstract:The residual feed intake (RFI) model has recently gained popularity for ranking dairy cows for feed efficiency. The RFI model ranks the cows based on their expected feed intake compared to the observed feed intake, where a negative phenotype (eating less than expected) is favourable. Yet interpreting the biological implications of the regression coefficients derived from RFI models has proven challenging. In addition, multitrait modelling of RFI has been proposed as an alternative to the least square RFI in nutrition and genetic studies. To solve the challenge with the biological interpretation of RFI regression coefficients and suggest ways to improve the modelling of RFI, an interdisciplinary effort was required between nutritionists and geneticists. Therefore, this paper aimed to explore the challenges with the traditional least square RFI model and propose solutions to improve the modelling of RFI. In the traditional least square RFI model, one set of fixed effects is used to solve systematic effects (e.g., seasonal effects and age at calving) for traits with different means and variances. Thereby, measurement and model fitting errors can accumulate in the phenotype, resulting in undesirable effects. A multivariate RFI model will likely reduce this problem, as trait-specific fixed effects are used. In addition, regression coefficients for DM intake on milk energy tend to have more biologically meaningful estimates in multitrait RFI models, which indicates a confounding effect between the fixed effects and regression coefficients in the least square RFI model. However, defining precise expectations for regression coefficients from RFI models or sourcing for accurate feed norm coefficients seems difficult, especially if the coefficients are applied to a wide cattle population with varying diets or management systems, for example. To improve multitrait modelling of RFI, we suggest improving the modelling of changes in energy status. Furthermore, a novel method to derive the energy density of the diet and individual digestive efficiency is proposed. Digestive efficiency is defined as the part of the efficiency associated with digestive processes, which primarily reflects the conversion from gross energy to metabolisable energy. We show the model was insensitive to prior values of energy density in feed and that there was individual variation in digestive efficiency. The proposed method needs further development and validation. In summary, using multitrait RFI can improve the accuracy of the ranking of dairy cows' feed efficiency, consequently improving economic and environmental sustainability on dairy farms.

Abstract:Abstract The pathophysiological changes in the pericontusional region after traumatic brain injury (TBI) have classically been considered to be ischemic. Using [F-18]fluorodeoxyglucose (FDG) and triple-oxygen PET studies, we examined the pericontusional "penumbra" to assess for increased oxygen extraction fraction (OEF), anaerobic metabolism, and tissue viability. Acute (≤4 days) CT, MRI, and PET studies were performed in eight patients with TBI who had contusions. Four regions-of-interest (ROI) containing the contusion core, pericontusional hypodense gray matter (GM), pericontusional normal-appearing GM, and remote normal-appearing GM, were defined using a semi-automatic method. The correlations of cerebral blood flow (CBF) with OEF, cerebral metabolic rate of oxygen (CMRO2), and cerebral metabolic rate of glucose (CMRglc) were examined. The oxygen-glucose ratio (OGR) in each brain region was evaluated for anaerobic metabolism. The results show that pericontusional tissue had progressively diminishing OEF, CBF, CMRO2, or CMRglc approaching the contusion core. In general, there was a preserved ratio of CBF to CMRO2 in pericontusional hypodense GM. The OGR of the pericontusional hypodense GM was low (<4.0) and was inversely correlated (r=-0.68) with time after injury. A large proportion (%area: 22-76%) of pericontusional hypodense GM tissue had CMRO2 values less than 35 μmol/100 g/min, with this percentage increased with time after injury.

Abstract:The transverse cerebellar diameter was measured in 62 nondiabetic and 30 diabetic women with large for gestational age fetuses. Using reference curves from 675 women with normal gestations between 14-42 weeks, head circumference and abdominal circumference overestimated gestational age significantly in both study groups, whereas the transverse cerebellar diameter did not.

Abstract:With multiparous Jersey cows, colostrum production seems to be variable. Due to this, we aimed to identify specific variables involved in colostrum production and quality. From 2021 to 2023, data from 28 US farms (415 multiparous Jersey cows) were used to investigate if colostrum yield, IgG concentration (g/L), and IgG yield (g) could be predicted by farm variables and transmitting abilities. With the data collected, multiple regression equations were developed to aid in predicting colostrum yield, IgG concentration, and IgG yield. Colostrum was weighed and sampled for IgG analysis. Dairy Herd Information (DHI), calving, diet, and management information data were compiled. Days below 5°C (D<), days above 23°C (D>), and days between 5 and 23°C (D) were recorded. We evaluated transmitting abilities for milk, fat, protein, and dollars; previous lactation milk yield, fat percent, fat yield, protein percent, protein yield, previous lactation somatic cell score, previous lactation days open, previous lactation days dry, previous lactation days in milk, and previous parity; and current lactation parity, days dry, and calving information, birth ordinal day, and latitude. Colostrum yield, IgG yield, and concentration had 1 added to correct for values = 0. After addition, values >0 were transformed to ln or log10. Nontransformed variables were also used to develop the model. Variance inflation factor analysis was conducted, followed by backward elimination. The log10 colostrum yield model (R2 = 0.55; β in parentheses) included herd size (-0.0001), ordinal days (-0.001), ln ordinal days (0.07), latitude (-0.02), dry period length (0.004), D< (-0.005), D (-0.003), time to harvest (0.05), ln time to harvest (-0.35), IgG (-0.004), log10 IgG (0.46), feedings per day (0.06), ln pasture access (-0.13), and ln previous lactation days open (0.14). The model showed that previous lactation days open contributed the most toward increasing and latitude contributed the most toward decreasing colostrum yield. The IgG model (R2 = 0.21) included herd size (0.02), D> (0.38), ln time to harvest (-19.42), colostrum yield (-4.29), ln diet type (18.00), ln previous lactation fat percent (74.43), and previous parity (5.72). The model showed that previous lactation milkfat percent contributed the most toward increasing and time from parturition to colostrum harvest contributed the most toward decreasing colostrum IgG concentration. The log10 IgG yield model (R2 = 0.79) included ln ordinal days (0.03), time to harvest (-0.01), colostrum yield (-0.11), ln colostrum yield (1.20), ln pasture access (-0.09), ln previous lactation fat percent (0.53), and previous parity (0.02). The model showed that colostrum yield contributed the most toward increasing IgG yield, followed by previous lactation milkfat percentage. Pasture access contributed the most toward decreasing IgG yield, although the contribution was very small. These models were validated using 39 samples from 22 farms. Actual minus predicted colostrum yield and IgG concentration and yield were 0.89 kg, -21.10 g/L, and -65.15 g, respectively. These models indicate that dry period management and cow information can predict colostrum yield and IgG concentration and yield.
