Abstract:Cu2+ and Ni2+ complexes of azo‐thiosemicarbazone named, 2‐(‐2‐hydroxy‐5‐(phenyldiazenyl)benzylidene) hydrazine‐1‐carbothioamide (H4Azo‐TSC, 1) are prepared and characterized theoretically based on DFT and experimentally via 1D/2D‐NMR, UV–Vis, FT‐IR, thermal, and E‐mass. The in vitro anticancer and antimicrobic activities of synthetics were investigated. This study focuses on the synthesis of azo‐thiosemicarbazone ligand, which is designed by reacting 2‐hydroxy‐5‐(phenyldiazenyl)benzaldehyde with hydrazine carbothioamide. Hence, this ligand was exploited for chelation with nickel and cupric salts in (1 M:1 L) molar ratio. These synthetics were structurally investigated using a variety of physical, analytical, and spectroscopic (1D/2D‐NMR, UV–Vis, FT‐IR, and E‐mass) tools as well as theoretically. The finding demonstrated that the resulting azo‐thiosemicarbazone (H4Azo‐TSC) bonded with the nickel and cupric ions as mono/di‐basic tridentate chelators, binding them via deprotonated phenolic oxygen, imino nitrogen, and thionic/thiol sulfur atoms adopting square planar geometry. The theoretical investigation was examined by DFT/B3LYP/LanL2dZ, including energetic parameters, HOMO‐LUMO energy gap, dipole moment, and geometrical optimization, which were applied to support the complexes' geometrical structure. The in vitro microbicidal activities of the azo‐thiosemicarbazone, compared with its Ni2+ and Cu2+ complexes, display superior activity over the metal complexes against different bacterial and fungal species. The anticancer efficacy of the synthetics was tested against human lung fibroblast (WI38) and mammary gland breast cancer (MCF‐7) compared with Doxorubicin as a standard drug. The cytotoxic efficiency of metallic complexes exceeded that of un‐chelated azo‐thiosemicarbazone. The most effective complex is Ni2+ complex with IC50 equal to 11.75 and 14.05 μg/ml, respectively. It has been demonstrated that Ni2+ and Cu2+ complexes are more biocompatible than free azo‐thiosemicarbazone; this finding may be related to the chelation process.

Abstract:Four new complexes (Ni2+, Cu2+, Ag+, and Hg2+) were prepared from the ligand N-(4-chlorophenyl)-2-(phenylglycyl)hydrazine-1-carbothioamide (H2L). Analytical and spectroscopic techniques were used to clarify the structural composition of the new chelates. In addition, all chelates were tested against bacterial strains and the HepG2 cell line to determine their antiseptic and carcinogenic properties. The Ni(II) complex was preferable to the other chelates. Molecular optimization revealed that H2L had the highest reactivity, followed by Hg-chelate, Ag-chelate, Ni-chelate, and Cu-chelate. Moreover, molecular docking was investigated against two different proteins: the ribosyltransferase enzyme (code: 3GEY) and the EGFR tyrosine kinase receptor (code: 1m17).

Abstract:Copper (II) complex of the ligand, thiophene-2-aldehyde semicarbazone was synthesized and characterized. Their structures were explored on the basis of CHN analysis, conductance and magnetic susceptibility measurements, spectral studies (FT-infrared and ESI-MS) and thermogravimetric analysis (TGA). It has been observed from spectral and analytical studies, the copper complex have the composition of [CuL(CH3COO)2(H2O)] [where L is thiophene-2-aldehyde semicarbazone]. The electrospray mass spectrum of the ligand and complex exhibited ion at mass-to charge ratio (m/z) of 169.13 and 368.86 matching the calculated values for the molecular ion of the ligand and complex, respectively. One mole of the ligand behaves as bidentate chelating agents around the Cu(II) ion. From conductivity studies, we obtained that the copper complex of the ligand, thiophene-2-aldehyde semicarbazone had no ionic properties. All data confirmed a trigonal bipyramidal geometry of the complex. Antibacterial activity of the compounds against the microbe Enterobacter aerogenes and Bacillus cereus have been screened to reflect that the complex has higher antimicrobial activity than the free ligand.

Abstract:A series of thiosemicarbazone (TSCN) compounds including ((E)-2-((E)-1-(2-(p-tolyl)hydrazono)propan-2-ylidene)hydrazine-1-carbothioamide (TSC1), (E)-N-ethyl-2-((E)-1-(2-(p-tolyl)hydrazono)propan-2-ylidene)hydrazine-1-carbothioamide (TSC2), and (E)-N-phenyl-2-((E)-1-(2-(p-tolyl)hydrazono)propan-2-ylidene)hydrazine-1-carbothioamide) (TSC3) were synthesized and fully characterized by diverse spectroscopies, such as X-ray single-crystal, infrared, mass, proton nuclear magnetic resonance, and ultraviolet-visible. Potentiometric measurements, molecular modeling, and biological and antitumor activity screening were studied. The thermodynamics and protonation constants of TSC1 as a representative of the synthesized TSCs were calculated and discussed. The solution speciation of different species was studied with pH. The molecular parameters of the optimized structures were calculated and discussed. The X-ray single crystals of TSC2 and TSC3 were established. Considering the antimicrobial activities and correlating structure-activity relationship of the synthesized compounds, the TSC1 molecule was considered a promising candidate as an antifungal agent against Candida albicans. Thus, it would be extremely helpful in the field of medicinal chemistry, particularly as an antimicrobial agent. The results are of significance to the chemistry of antimicrobial agents.

Abstract:New Ni (II) and Cu (II) complexes with pyridoxal-semicarbazone were synthesized and their structures were solved by X-ray crystallography. This analysis showed the bis-ligand octahedral structure of [Ni(PLSC-H)2]·H2O and the dimer octahedral structure of [Cu(PLSC)(SO4)(H2O)]2·2H2O. Hirshfeld surface analysis was employed to determine the most important intermolecular interactions in the crystallographic structures. The structures of both complexes were further examined using density functional theory and natural bond orbital analysis. The photocatalytic decomposition of methylene blue in the presence of both compounds was investigated. Both compounds were active toward E. coli and S. aureus, with a minimum inhibition concentration similar to that of chloramphenicol. The obtained complexes led to the formation of free radical species, as was demonstrated in an experiment with dichlorofluorescein-diacetate. It is postulated that this is the mechanistic pathway of the antibacterial and photocatalytic activities. Cyclic voltammograms of the compounds showed the peaks of the reduction of metal ions. A molecular docking study showed that the Ni(II) complex exhibited promising activity towards Janus kinase (JAK), as a potential therapy for inflammatory diseases, cancers, and immunologic disorders.

Abstract:New binary and ternary nickel (II) hyrdazone complexes were synthesized and characterized by analytical and spectral methods. The molecular structures of the complexes were investigated by density functional theory. Complexes showed anticancer action against Ehrlich Ascites Carcinoma, and the results are supported by molecular docking studies. New binary and mixed‐ligand nickel (II) chelates of a hyrdazone ligand (HYQX) carrying alloxan and quinoline moieties were synthesized. Spectroscopic and analytical techniques were effectively used for identifying the composition and structure of the prepared nickel (II) HYQX chelates. The neutral nature of Ni‐HYQX complexes was demonstrated by molar conductivity studies, and magnetic moment values revealed octahedral geometry. The results demonstrated that HYQX behaves as a monoanionic di‐/tridentate chelating agent in all complexes, except nitrato C2 and acetato C4 complexes, which are bi‐ and tri‐nuclear complexes, respectively, with a unique mode of coordination. The thermal decomposition patterns of Ni‐HYQX complexes were investigated in relation to structure, and thermodynamic parameters were successfully calculated. The molecular structural features of HYQX and its Ni (II) chelates were investigated based on density functional theory (DFT) level at B3LYP/6‐311G(d,p) and LanL2dz level. Ni‐HYQX complexes were found to have anticancer action against Ehrlich Ascites Carcinoma. A molecular docking investigation was conducted to determine in what way the title compounds connect to the CDK‐5 inhibitor‐crystal structure of the inhibitor EFP with CDK‐2 (PDB ID: 3IG7) in order to verify their biological activity.

Abstract:Synthesis for new Cu(II) complexes from acetamidopyridine derivitives and their characterization. Applying analytical, spectral and conformational techniques, the characterization for all new synthesizes, was achieved. Equimolar ratio (1:1) was proposed for all complexes in which the ligands behaved as bidentate toward copper center. The molar conductivity measurements defined state of acetate presence according to coordination sphere. Two structural forms were proposed for five complexes, square-planer and octahedral based on UV–Vis and ESR data. Conformational study was executed for all compounds by DFT method in Gaussian 09 under 6-31G basis set. The optimal structural forms were obtained besides other significant files (log, chk & fchk). Also, functional parameters were calculated, to put a well view about physical features. QSAR parameters were obtained from HyperChem program, also to assert on some characteristics. MOE-docking module, which considered the most powerful program simulates interaction of tested compound with biological systems, was also used to strengthen the study. The data extracted orient to great expectation for biological efficiency of complexes against breast cancer cells. The most shining point was the anticancer screening results, which reflect vigorous toxic behavior for most tested complexes against breast cancer cell line that actually exceeds than doxorubicin itself.

Abstract:Preparation, characterization, and application of thiophene Schiff base complexes. Novel thiophene Schiff base ligand (L) was synthesized through the condensation of 2‐thiophene‐methylamine and 3‐formyl‐4‐methoxyphenylboronic acid. Transition metal complexes with the formulae [M(L)(H2O)2Cl]Cl2.xH2O (M = Er(III), x = 4 and La(III), x = 5) and [M(L)(H2O)Cl2]Cl.xH2O (M = Yb(III), x = 4 and Ru(III), x = 0) were synthesized. Different spectral and physicochemical methods were used to confirm and identify the stoichiometry, structure, and bonding style of transition metal chelates. The creation of 1:1 molar ratio (M:L) complexes was supported by elemental data. According to IR spectra, the thiophene Schiff base ligand (L) acted as a tridentate ligand that is neutrally coordinated by S‐thiophene, N‐azomethine, and o‐methoxy groups. All chelates formed in octahedral structures according to the findings data. Thermogravimetric analysis (TG and DTG) was employed to examine the heat decomposition of these compounds. DFT/B3LYP method, a molecular modeling methodology, is used to support the expected shape of the produced ligand. Chemical hardness and softness, HOMO and LUMO energy levels, electrophilic index, bond lengths, angles, dipole moment, electronegativity, and other factors were calculated. Against HepG‐2 cell line, the in vitro anticancer activity of all chelates and free ligands was evaluated. These synthesized compounds showed good effectiveness. Additionally, DPPH scavenging was used to assess antioxidant activity, with extremely positive results. Molecular docking with the hepatic protein receptor 3ELJ was used to confirm the anticancer results. Finally, the effectiveness of the current thiophene ligand's ability to block Al‐Si alloys in 1 M HCl solution was examined utilizing electrochemical methods. The high inhibition efficiency was obtained revealing the effectiveness of using this thiophene Schiff base ligand on an industrial scale.

Abstract:Structural features and catalytic activities of five ternary copper (II)-Schiff base complexes are investigated. The ligands are derived from 3-methoxysalicylaldehyde (MS) or 4-diethylaminosalicylaldehyde (DS) and amino acids {L-phenylalanine (Phe), L histidine (His) or DL-tryptophan (Trp)}, as primary ligands, and 2,4′-bipyridyl (DP) as a secondary ligand. Cu(II)-complexes are characterized by various physicochemical tools. The catalytic efficiency of Cu(II)-complexes is studied in the oxidation of benzyl alcohol by an aqueous H2O2 in different reaction conditions. Temperature and catalyst features are involved in order to obtain the optimized catalytic oxidation conditions of benzaldehyde production. The Schiff base ligands and their ternary complexes are screened for their antimicrobial activities in various types of fungi and bacteria. The interaction between Cu(II)-complexes and (CT-DNA) was examined by employing various techniques, including viscosity, spectral and gel electrophoreses studies.

Abstract:The objective of our work is the preparation of the new catalysts for catecholase, whose principle is based on the catechol oxidation reaction which presents a major challenge in both biology and medicine. First, we synthesized nine pyrazole and triazole ligands, then we evaluated the catalytic properties of certain of those ligands in situ complexes to catalyze the oxidation reaction of catechol to o-quinone. The aim of this study is to find the right models to reproduce the catalytic activity of the enzyme (catecholase), so we used complexes formed in situ by pyrazole and triazole derivatives with copper (II) salts. The reason behind the interest of these complexes is their resemblance to biological systems, capability of activating the catalyst for many chemical reactions. Among these complexes, some of them showed good catalytic activity for this reaction. We have demonstrated that the nature of ligand, the concentration of ligand, the nature of the solvent and the nature of the copper (II) salt, influences the efficiency of the catecholase activity. Pyrocatechol is benzene-1,2-diol (nomenclature IUPAC), also known as catechol with formula C 6 H 6 O 2 crude used in many organic syntheses. Some catecholamines have important physiological functions. The Government of Canada has determined that catechol is "toxic" as defined by the Canadian Environmental Protection Act, 1999. So, to reduce the toxic effect of catechol we try to oxidize o-quinone which is less dangerous than catechol. The DFT study has big interest in many researches to know the reactivity of the ligands by calculating different quantum descriptors as: EHOMO, ELUMO, The gap energy, Ionisation potential, The electron affinity and The hardness.

Abstract:The divalent transition metal ions Co, Ni, and Cu coordination compounds with unusual bidentate mixed Schiff base-based 1,2,4-triazole-3-thione ligand in a protonated specific coordination stoichiometry and structures forms were synthesized. The structural composition of the ligand 5-(1-naphthylmethyl)-4-{( Z )-[(2 E )-3-phenyl-2-propen-1-ylidene]amino}-2,4-dihydro-3 H -1,2,4-triazole-3-thione (LH) was fully identified by 1 H and 13 C NMR, FTIR, UV-Vis, mass spectrometry as well elemental analysis. The UV-Vis, FTIR spectroscopy, magnetic susceptibility measurements, molar conductance, flame atomic absorption, and elemental analysis (CHNS) techniques were used to characterize the synthesized complexes. Three complexes of Ni(II), Co(II), and Cu(II), in addition to the ligand LH, were the subject of computational investigations. Benchmark analyses were carried out to determine the best degree of calculation for the complexes under the study. The B3LYP approach was chosen as the computation method. The B3LYP/6-31G (d,p) (LANL2DZ) was the best calculation rank. IR spectra and structural studies were carried out. Furthermore, a thorough investigation and examination of biological characteristics were conducted. Complex (Cu) was determined to be the most excellent choice for biological applications based on calculation findings.

Abstract:By combining hydrazide with 2-Acetylpyridine, a hydrazone ligand (HL) was successfully created. Several copper (II) salts have been used to create three copper (II) hydrazone complexes (acetate, sulphate, and chloride). The hydrazide ligand and its copper (II) complexes (1-3) were studied via variety of analytical techniques, including elemental analysis, electronic, infrared, UV-vis Spectrum, XRD study, thermal analysis, also molar conductivity amounts. The spectrum results indicate that in all complexes, the ligand exhibits monobasic tridentate behavior. Octahedral geometries were present in all metal complexes. The Coats-Redfern equations were used to compute and describe the dynamics properties of several steps of TGA (Ea, A, ΔH*, ΔS*, and ΔG*). Calculations using the density functional theory (DFT) were done at the molecular studio software toward examine ligands agent's and its complexes' best structures. The MCF-7 in addition to HepG-2 cell lines was resistant to tumor-inducing effects of the copper (II) chelates. The in vitro antioxidant capacities of all complexes have been estimated via DPPH free radical scavenger assays. Furthermore, zones of inhibition length accustomed to test antimicrobial effect of particular complexes in vitro towards Staphylococcus aureus (Gram positive bacteria) E. coli (Gram negative bacteria). Both absorption spectra and viscosity measurements in calf thymus DNA binding have been used to study the complexes. In order to explore docking research of copper (II) chelates, the crystallographic construction of the SARS-active CoV-2's site protein (PDB ID:6XBH) was used (COVID-19) and breast cancer distorted (PDB ID: 3hb5).

Abstract:Three tridentate imine ligands containing N 2 S donors were synthesized via Schiff condensation between derivatives of both amino triazine and 2-carbonyl pyridine. The reaction of these ligands with CuCI 2 in a molar ratio of 1:1 provides three Cu(II) complexes with the general formula [CuL n ·Cl 2 ]. Analytical, electrical, magnetic, and spectroscopic studies were used to assign the molecular formulae of these metallic chelates. Density function theory (DFT) calculations confirmed the structural analysis results obtained from spectroscopic studies. The various characterization techniques used demonstrated the pentacoordinated slightly distorted square pyramidal structure for the present Cu(II) complexes 1, 2 , and 3 . Measurements of cyclic voltammetry were done in methanol to define the electrochemical behavior of the current Cu(II) complexes. The biomimetics of catechol oxidase (C.O.) and phenoxazinone synthase (PHS) have been studied in the aerobic oxidation of some phenolic substrates, such as 3,5-di- tert -butylcatechol (3,5-DTBCH 2 ) and ortho -aminophenol ( o -APH 3 ). The three candidate oxidase mimetics showed promising activity in the order 3>1>2 . The catalytic activity related to the structural properties of existing oxidase mimetics was discussed. The driving force (−Δ G °) controlling the redox reactions of the present biomolecules was calculated from the redox data of Cu(II) complexes 1, 2 , and 3 . The potential catalytic reaction pathway for the oxidation of the studied phenolic substrates was discussed.

Abstract:The long-standing interest in thiosemicarbazones (TSCs) has been largely driven by their potential toward theranostic applications including cellular imaging assays and multimodality imaging. We focus herein on the results of our new investigations into: (a) the structural chemistry of a family of rigid mono(thiosemicarbazone) ligands characterized by extended and aromatic backbones and (b) the formation of their corresponding thiosemicarbazonato Zn(II) and Cu(II) metal complexes. The synthesis of new ligands and their Zn(II) complexes was performed using a rapid, efficient and straightforward microwave-assisted method which superseded their preparation by conventional heating. We describe hereby new microwave irradiation protocols that are suitable for both imine bond formation reactions in the thiosemicabazone ligand synthesis and for Zn(II) metalation reactions. The new thiosemicarbazone ligands, denoted HL, mono(4-R-3-thiosemicarbazone)quinone, and their corresponding Zn(II) complexes, denoted ZnL2, mono(4-R-3-thiosemicarbazone)quinone, where R = H, Me, Ethyl, Allyl, and Phenyl, quinone = acenapthnenequinone (AN), aceanthrenequinone (AA), phenanthrenequinone (PH), and pyrene-4,5-dione (PY) were isolated and fully characterized spectroscopically and by mass spectrometry. A plethora of single crystal X-ray diffraction structures were obtained and analyzed and the geometries were also validated by DFT calculations. The Zn(II) complexes presented either distorted octahedral geometry or tetrahedral arrangements of the O/N/S donors around the metal center. The modification of the thiosemicarbazide moiety at the exocyclic N atoms with a range of organic linkers was also explored, opening the way to bioconjugation protocols for these compounds. The radiolabeling of these thiosemicarbazones with 64Cu was achieved under mild conditions for the first time: this cyclotron-available radioisotope of copper (t1/2 = 12.7 h; β+ 17.8%; β- 38.4%) is well-known for its proficiency in positron emission tomography (PET) imaging and for its theranostic potential, on the basis of the preclinical and clinical cancer research of established bis(thiosemicarbazones), such as the hypoxia tracer 64Cu-labeled copper(diacetyl-bis(N4-methylthiosemicarbazone)], [64Cu]Cu(ATSM). Our labeling reactions proceeded in high radiochemical incorporation (>80% for the most sterically unencumbered ligands) showing promise of these species as building blocks for theranostics and synthetic scaffolds for multimodality imaging probes. The corresponding "cold" Cu(II) metalations were also performed under the mild conditions mimicking the radiolabeling protocols. Interestingly, room temperature or mild heating led to Cu(II) incorporation in the 1:1, as well as 1:2 metal: ligand ratios in the new complexes, as evident from extensive mass spectrometry investigations backed by EPR measurements, and the formation of Cu(L)2-type species prevails, especially for the AN-Ph thiosemicarbazone ligand (L-). The cytotoxicity levels of a selection of ligands and Zn(II) complexes in this class were further tested in commonly used human cancer cell lines (HeLa, human cervical cancer cells, and PC-3, human prostate cancer cells). Tests showed that their IC50 levels are comparable to that of the clinical drug cis-platin, evaluated under similar conditions. The cellular internalizations of the selected ZnL2-type compounds Zn(AN-Allyl)2, Zn(AA-Allyl)2, Zn(PH-Allyl)2, and Zn(PY-Allyl)2 were evaluated in living PC-3 cells using laser confocal fluorescent spectroscopy and these experiments showed exclusively cytoplasmic distributions.

Abstract:Novel Co(II), Ni(II), Cd(II) and Hg(II)-Schiff base complexes were prepared and characterized by all possible techniques. 1:2 molar ratio (M:L) was the only one isolated for all solid complexes. The conductometric titrations for one metal ion (for instance, copper) with the ligand, were carried out. Such simplifies the calculation for complex molar ratio in solution beside association and formation constants. Conformational study was elaborately handled by using Material Studio package (DMOL3 program) and Gaussian 09 under double numerical basis sets plus polarization functional (DNP). The best structural forms as well as other important data were obtained and discussed for differentiation. The molecular docking study was done by using MOE-approach, to discriminate and evaluate the inhibition grade of all new synthesizes, against two elected functional proteins. 3mng and 5j71 were chosen for this study, which are responsible for blocking antioxidant pathway and kinase dehydrogenase that regulates cancer, respectively. Ni(II)-complex plays well inhibitory role towards the two functional proteins. Furthermore, extensive biological screening was done concerning all compounds as, antimicrobial, antioxidant and antitumor. The moderate activity was commonly dominated according to most complexes-results. This screening matches good with the data expected from MOE-docking methodology.

Abstract:In this study, firstly, bis(thiosemicarbazone) ligand [L: 2,2′-(2-(2-(4-methoxyphenyl)hydrazineylidene)cyclohexane-1,3-diylidene)bis(hydrazine-1-carbothioamide)] was synthesized by the condensation reaction of thiosemicarbazide and ketone compound (2-(2-(4-methoxyphenyl)hydrazone)cyclohexane-1,3-dione). The metal complexes were synthesized by the reaction of obtained ligand (L) with CuCl 2 ·2H 2 O, NiCl 2 ·6H 2 O, CoCl 2 ·6H 2 O, and MnCl 2 ·4H 2 O salts. The structures of synthesized ligand and their complexes were characterized using elemental analysis, IR, UV–Vis, 1 H-NMR spectra, 13 C-NMR spectra, magnetic susceptibility, mass spectra (LC–MS), thermogravimetry analysis-differential thermal analysis (TGA-DTA), and differential scanning calorimetry techniques. According to the results of the analysis, square plane geometry was suggested for Cu and Co complexes. However, the structures of Ni and Mn complexes were in agreement with octahedral geometry. Molecular docking analysis and pharmacological potential of the compound were evaluated to determine the inhibitory potential against acetylcholinesterase (AChE) and Glutathione-S-transferases (GST) enzymes. The compound exhibited strong binding/docking indices of − 5.708 and − 5.928 kcal/mol for the respective receptors. In addition, L–Ni(II) complex was found to be the most effective inhibitor for AChE enzyme with a K i value of 0.519. However, with a K i value of 1.119, L–Cu(II) complex was also found to be an effective inhibitor for the GST enzyme.

Abstract:Schiff base derivative of 2‐aminothiazole and 2,4‐dihydroxybenzaldehyde in addition to its Co (II), Cu (II), Zn (II) and Th (IV) chelates were prepared. Structure characterization of the prepared compounds by applying all available analytical and spectroscopic tools. The acquired data showed the chelation of the ligand with metal through the nitrogen of the azo group and deprotonated ortho phenolic oxygen. Investigation of antimicrobial and anticancer activity for the ligand and in addition to docking studied through interaction with 3S7S and 2JW2. For further pharmaceutical applications, a novel Schiff base derivative of aminothiazole and its Co (II), Cu (II), Zn (II), and Th (IV) chelates were designed and prepared. For this purpose, 2‐aminothiazole and 2,4‐dihydroxybenzaldehyde have been coupled before the complexation reaction with the metal salts. Elemental analysis, FT‐IR, 1HNMR, UV–Vis spectra, transmission electron microscopy (TEM), X‐ray powder diffraction (XRD), molar conductance, and magnetic moment measurements were used to determine the structural formula of the ligand and its produced chelates. The acquired data pointed out that the ligand was chelated with metal through the nitrogen of azo group and deprotonated ortho phenolic oxygen atoms. In Cu (II) complex the copper ion chelated with two ligand molecules, whereas in other complexes the metal binds with one molecule only of ligand. It was shown by XRD patterns and high‐resolution TEM images that the complex particles had a uniform distribution across the chelates' surfaces and were nanometric in size. Extremely, antimicrobial and anticancer activity for the prepared ligand and complexes were investigated, which showed high activity, especially after complex formation. Docking studies have been performed to predict the compounds' efficacy as antitumor agents through their interaction with the co‐crystal structure of proteins for breast cancer cells (PDB ID: 3S7S) and protein of liver cancer 2 (PDB ID: 2JW2). The experimental results of the anticancer activity showed promising results, especially for Cu (II) compound which exhibited the greatest antitumor efficacy (IC50 = 12.81 μg/ml) contra HepG‐2 carcinoma and Mcf‐7 cells (IC50 = 18.92 ± 0.91 μg/ml). Antimicrobial activity against the strains Klebsiella pneumoniae (K. pneumoniae), Escherichia coli (E. coli), Staphylococcus aureus (S. aureus), Bacillus subtilis (B. subtilis), C. albicans (C. albicans), and Aspergillus niger (A. niger) also gave very promising results when it is compared with the standard drugs.

Abstract:New mononuclear copper(II), nickel(II) and cobalt(II) hydrazone complexes of 2-cyano-N′-((4-oxo-4H-chromen-3-yl)methylene)acetohydrazide have been synthesized. Structure identification of the prepared complexes was carried out by physicochemical and spectroscopic techniques including elemental analyses, infrared, electronic, mass and electron spin resonance spectra, thermal analysis, conductivity and magnetic susceptibility measurements, powder x-ray diffraction and transmission electron microscope. The isolated binary complexes are neutral mononuclear complexes (with 1:1 and 1:2; M:L stoichiometries) and ternary complexes (with 1:1:1; M:L:L′ stoichiometry). The analytical and spectroscopic techniques illustrated the monoanionic tridentate behavior of the hydrazone ligand with γ-pyrone oxygen, enolic oxygen and azomethine nitrogen coordinating sites. The metal complexes displayed square planar, tetrahedral and octahedral geometries. Density Functional Theory calculations were carried out for the chelating agent and its copper(II) chelates. The phenoxazinone synthase activity of the binary copper(II) complexes was studied and discussed. The antimicrobial activity of the hydrazone ligand and its complexes was evaluated toward some sorts of Gram–positive bacteria, Gram–negative bacteria, yeast and fungus.

Abstract:The 2-benzyl-4‐hydroxy-1,1-dioxo-1,2-benzothiazine-3-carboxylic acid methyl ester i.e., ligand 1 was utilized to synthesize Mn(II) 2 , Co(II) 3 , Ni(II) 4 , Cu(II) 5 and Zn(II) 6 derived metal complexes in mild basic conditions. The structural investigation was carried out by UV–vis, FT-IR, NMR spectroscopy, CHN analysis, conductivity measurement, ESI-MS analysis and magnetic susceptibility techniques. The X-ray diffraction analysis confirmed the octahedral and distorted square pyramidal geometries of the cobalt and copper complexes. The ligand 1 coordinated with metal centres through enolic oxygen atoms of the thiazine ring and ester group while methanol coordinated with metal center via oxygen atom. The Hirshfeld surface analysis (HSA) was utilized to determine the intermolecular interactions. In support of the experimental data, DFT studies were also performed for the cobalt and copper complexes. The NBO analysis resulted in the large stabilization values 11.91 and 12.35 kcal/mol for the cobalt and copper complexes. The global reactivity indicated the higher values of hardness and the large values of FMO analysis which support the higher stability of the metal complexes. The synthesized compounds were also evaluated against six bacterial strains to check their antibacterial potential. It was observed from results that highest activity was depicted by copper complex 5 against B. subtilis and the zone of inhibition is 23.4 ± 0.6 mm which is comparable to ampicillin (standard drug).
