Abstract:The ANAMMOX process with sequential biocatalyst addition (SBA-ANAMMOX process) was developed in sequencing batch reactor (SBR) to enhance nitrogen removal from ammonium-rich wastewater containing glucose content up to 800 mgCOD/L. The nitrogen removal performance of the SBR operated with 800 mgCOD/L was significantly enhanced after sequential adding 0.5 gVSS/d of high-rate ANAMMOX granules (HASGs). Kinetics analyses showed that the HASGs possessed high nitrite affinity and strong resistance to organic matter, both of which contributed to the enhancement of autotrophic nitrogen removal. Consequently. ANAMMOX became the dominant reaction to metabolize nitrite under organic conditions; denitrification was limited due to the lack of electron acceptors (nitrite and nitrate); while sulfate reduction emerged as an important process for consumption of organic matter in the SBA-ANAMMOX system. It is suggested that employing the SBA-ANAMMOX process for treatment of organic ammonium-rich wastewater, the simultaneous removal of nitrogenous compounds as well as organic matter was quite possible. Abstract:In this work, two identical sequencing batch reactors (SBRs) with mature aerobic granules were utilized to treat synthetic high-strength ammonium wastewaters with chemical oxygen demand (COD)/total nitrogen (TN) ratios of 3.9-6.9. The contributions of various mechanisms to the removal of ammonium were determined. Ammonium levels of 600-2000mg-Nl(-1) had little adverse effect on the COD removal rate (91.6%-95.3%) with an influent COD of 4490-9860mgl(-1). The TN removal rate was slightly reduced from 71.3% to 59.6% as the influent ammonium concentration was increased from 600 to 2000mg-Nl(-1). Experimental results indicated that aerobic granules removed 94.5% of COD and 59.6% of TN in the treatment of synthetic high-strength wastewater (9860mg-CODl(-1) and 2000mg -Nl(-1)) during a 12h cycle. Granular adsorption, air stripping and conversion by nitrification/denitrification were responsible for removing 9%, 15% and 76%, respectively, of the total removed -N. Dissolved oxygen (DO) was a useful process indicator of the biological reactions in the treatment of high-level ammonium wastewaters.

Abstract:Since nitrification is the rate-limiting step in the biological nitrogen removal from wastewater, many studies have been conducted on the immobilization of nitrifying bacteria. A laboratory-scale investigation was conducted to examine the effectiveness of a continuous-flow airlift reactor (ALR) on the granulation of nitrifying sludge and the nitrification efficiency of the reactor after granulation. The results showed that the granular sludge began to appear on day 30 and matured in 75 days. The mature granules had an average diameter of 1.54mm, settling velocity higher than 82.4mh−1 and specific gravity of 1.07. The granules cultured in the present study had aerobic ammonia oxidation activity of 13.3mgNH4+-N(gVSS)−1day−1 and anaerobic ammonium oxidation (ANAMMOX) activity of 3.22mgNH4+-N(gVSS)−1day−1, which demonstrated that the nitrifying granules possessed the potential to be used as seed sludge for ANAMMOX and CANON (completely autotrophic nitrogen-removal over nitrite) reactors. After granulation, the ALR exhibited an excellent nitrification performance. It had strong tolerance to influent NH4+-N of 1100mgL−1. When operated at influent NH4+-N concentration of 546mgL−1, the reactor could remove over 94.4% of ammonium even at a nitrogen loading rate (NLR) of 2.37fm−3day−1 with a short hydraulic retention time (HRT) of 5.4h. With the influent NH4+-N concentration of 547mgL−1, HRT 6.9h and NLR of 1.90kgm−3day−1, superior effluent quality could be achieved robustly, with an effluent NH4+-N of less than 5mgL−1, satisfying the national primary discharging standard of China (GB 8978-1996).

Abstract:Partial nitrification on ammonia laden wastewater using aerobic granule continuous-flow reactors was studied. Effects of temperature and carbon sources on performance of partial nitrification and characteristics of aerobic granules were reported. High nitrite accumulation ratio (>90%) was observed in all reactors after an acclimation phase of 6 days. Decrease in temperature or inorganic carbon concentration limited nitrite accumulation ratio; while nitrite accumulation was hard to recover once complete nitrification to nitrate was established in granules. The aerobic granules collected at continuous-flow reactor were stronger in strength than the seed granules from sequential batch reactor. Clone libraries revealed that Nitrosomonas europaea and Nitrosomonas sp. were the ammonia-oxidizing bacteria (AOB), whereas Nitrobacter sp. and Nitrospira cf moscoviensis were nitrite-oxidizing bacteria (NOB). Polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) analysis revealed that temperature and carbon sources shifted the microbial community in granules. Partial Nitrification Adjusted by Hydroxylamine in Aerobic Granules under High DO and Ambient Temperature and Subsequent Anammox for Low C/N Wastewater Treatment

Abstract:Partial nitritation–Anammox process is regarded as a promising nitrogen removal method for nitrogen removal from low C/N wastewaters. In the present work, stable partial nitrification was successfully achieved by 10mg/l hydroxylamine dosing in aerobic granules under experimental conditions of pH 7.8–8.2, dissolved oxygen (DO) above 5mg/l and temperature around 25°C. When the NH4–N and COD were fixed at 100mg/l and 400mg/l, the TN removal efficiency was 57% with a NO2–N/NOx–N (NO2+NO3)–N ratio of 99.8% in the effluent. Meanwhile, 79% COD was removed by aerobic granules, which was regarded as beneficial to the following Anammox. Sequentially, a mixture of the SBR effluent and raw influent with an average NO2–N/NH4–N ratio of 1.67 was fed to an Anammox reactor. The COD and TN removal efficiencies reached 77% and 81%, respectively, through coupling of Anammox and denitrification. SEM detection showed that the main composition of aerobic granules was coccus and bacilli bacteria. FISH results demonstrated that ammonia-oxidizing bacteria (AOB) became the dominant bacteria of aerobic granules, which further proved the important effects of hydroxylamine on the achievement of partial nitrification by aerobic granules under conditions of high DO and ambient temperature.
Successful operation of nitrifying granules at low pH in a continuous-flow reactor: Nitrification performance, granule stability, and microbial community

Abstract:Acidic nitrification, as a novel process for treating wastewater without sufficient alkalinity, has received increasing attention over the years. In this study, a continuous-flow reactor with aerobic granular sludge was successful operated at low pH (<6.5) performing high-rate acidic nitrification. Volumetric ammonium oxidation rate of 0.4-1.2 kg/(m3·d) were achieved with the specific biomass activities of 5.8-13.9 mg N/(gVSS·h). Stable partial nitritation with nitrite accumulation efficiency over 85% could be maintained at pH above 6 with the aid of residual ammonium, whereas the nitrite accumulation disappeared when pH was below 6. Interestingly, the granule morphology significantly improved during the acidic operation. The increased secretion of extracellular polymeric substances (especially polysaccharides) suggested a self-protective behavior of microbes in the aerobic granules against acidic stress. 16S rRNA gene sequencing analyses indicated that Candidatus Nitrospira defluvii was always the dominant nitrite-oxidizing bacteria, while the dominant ammonia-oxidizing bacteria shifted from Nitrosomonas europaea to Nitrosomonas mobilis. This study, for the first time, demonstrated the improved stability of aerobic granules under acidic conditions, and also highlighted aerobic granules as a useful solution to achieve high-rate acidic nitrification.
Granulation and Characteristics of Extracellular Polymers of Anammox Sludge in High-Load EGSB Reactor

Abstract:High effectiveness is one of the most attractive advantages of the anaerobic ammonium oxidation (anammox) process for nitrogen removal from ammonium-rich wastewater.Granulation is considered to play a vital role in development of high-load bioreactors.The hydraulic retention time (HRT) of the 1.1 L anammox EGSB (expanded-granular sludge bed) reactor was progressively shortened from 6.9 h to 0.30 h;while the influent nitrite concentration was fixed at 360 mg·L-1 and the effluent recycling was maintained at 0.5 constantly.A super high nitrogen removal rate,50.75 kg·m-3·d-1 was recorded,which was 2 times of the highest value reported before.Granulation of the anammox microorganisms occurred in the high-load anammox EGSB reactor.The average diameter of the high-load anammox granules was (2.51±0.91)mm and the specific anammox activity reached 1.899 kg·(g VSS)-1·d-1.The extracellular polymers (ECP) content was 143.00 mg·(g VSS)-1,which was amazingly high when compared to heterotrophic microbial granules.The extracellular proteins increased at a higher rate along with the increase of hydrodynamic shear stress,resulting in over-production of extracellular proteins.Thus the stability of the anammox granules decreased and sludge washout became inevitable.

Abstract:Nitrogen pollution of waters has sometimes caused severe eutrophication, leading to the death of fishes and most aquatic life. There is therefore a need for efficient and cost-effective methods to remove nitrogen from ammonium-rich wastewaters. Anaerobic ammonium oxidation (ANAMMOX) is a promising process to remove nitrogen because this process directly oxidizes ammonium (NH4+) to dinitrogen gas (N2) under anoxic condition. Nonetheless, a challenge of this process is that chemolithoautotrophic Anammox bacteria grow slowly at the beginning, thus resulting in low Anammox biomass and instability of reactors. Such issues can be overcome by granulation of the Anammox sludge. Here, we review the characteristics of the Anammox bacteria, and the formation, structure and flotation of Anammox granules under high hydraulic loadings. We also evaluate the performances of full-scale granular Anammox processes. The major points are: 1) Anammox bacteria secrete a large amount of extracellular polymeric substances (EPS), up to 415 mg g−1 of volatile suspended solids (VSS), containing many hydrophobic functional groups that facilitate biomass granulation. 2) Granulation enhances the sludge settling property and retention time, which contributes to the extremely high nitrogen removal rate of 77 kg m−3 d−1 of Anammox upflow reactors. 3) Flotation of Anammox granules frequently occurs under nitrogen removal rate higher than 10 kg m−3 d−1, which is mainly due to the overproduction of EPS under high hydraulic conditions.

Treatment of low-C/N nitrate wastewater using a partial denitrification-anammox granule system: Granule reconstruction, stability, and microbial structure analyses

Abstract:Industrial wastewater discharged into sewer systems is often characterized by high nitrate contents and low C/N ratios, resulting in high treatment costs when using conventional activated sludge methods. This study introduces a partial denitrification-anammox (PD/A) granular process to address this challenge. The PD/A granular process achieved an effluent TN level of 3.7 mg/L at a low C/N ratio of 2.3. Analysis of a typical cycle showed that the partial denitrification peaked within 15 min and achieved a nitrate-to-nitrite transformation ratio of 86.9%. Anammox, which was activated from 15 to 120 min, contributed 86.2% of the TN removal. The system exhibited rapid recovery from post-organic shock, which was attributed to significant increases in protein content within TB-EPS. Microbial dispersion and reassembly were observed after coexistence of the granules, with Thauera (39.12%) and Candidatus Brocadia (1.25%) identified as key functional microorganisms. This study underscores the efficacy of PD/A granular sludge technology for treating low-C/N nitrate wastewater.

Abstract:Partial nitrification for the biological nitrogen removal (BNR) via nitrite has recently gained interest for treating high-strength ammonium wastewater with low organic matter content. However, stability of partial nitrification is still a challenge in activated sludge system. In the present study, the technique of aerobic granulation was developed to produce granules for stable nitritation, treating ammonia-rich inorganic influent with 400 mg NH4+-N/L. The morphology, physical properties, bacterial community structure and partial nitrification performance of the sludge were characterized throughout the experiments. The results indicated that aerobic granules could be produced for partial nitrification through selective discharge of small and slow-settling sludge flocs. Sludge granulation help to achieve ammonium oxidation to the level of nitritation, or partial nitrification, other than to complete nitrification. Based on DNA-base molecular analysis, aerobic granulation resulted in an enrichment of AOB and a reduction of nitrite-oxidizing bacteria (NOB) in the granular sludge, which is highly favorable to a stable operation of partial nitrification.
Performance and granular characteristics of salt-tolerant aerobic granular reactors response to multiple hypersaline wastewater

Abstract:Aerobic granular sludge (AGS) technology has been recognized as a promising alternative to alleviate the osmotic stress of hypersaline wastewater. However, the response of AGS process to composite hypersaline wastewater on removal performance and populations was yet to be understood. In this work, two sequenced batch reactors were operated in parallel in absence (R0) and presence (R1) of high concentration sulfate as proxy for single and mixed salts (30 g salt·L−1) respectively. Results demonstrated that the presence of sulfate in hypersaline wastewater enhanced chemical oxygen demand (COD) and total nitrogen (TN) removals of 95.3% and 65.5% respectively with lower accumulations of nitrite. High-throughput 16 S rRNA gene sequencing technique elucidated that Denitromonas (31.6%) and Xanthomarina (17.0%) were the more dominant genera in AGS response to mixed salts with high sulfate and laid the biological basis for strengthening removal performance. The enrichment of halophilic Luteococcus (23.5%) in the AGS surface indicated the potential role of mixed salts in shaping the physical properties and surface population structure of AGS. Our work could facilitate the potential applications of AGS technology for industrial hypersaline wastewater treatment with complicated compositions.

Abstract:The partial-denitrification/anammox (PDA) process is a promising method to achieve mainstream anammox in wastewater treatment plants (WWTPs). To investigate the feasibility of developing a granule-based process for effective nitrogen removal via PDA, an upflow anaerobic sludge bed reactor was used as a PDA reactor treating low C/N sewage for over 200 days. Granules were formed with an average particle size of 1.92 mm. Metagenomic analysis revealed that the two most abundant genera in granules were Thauera (17.46%) and Candidatus Brocadia (6.24%) which played important roles in achieving partial-denitrification and anammox, respectively. Effective nitrogen removal was achieved with an average effluent TN concentration of 8.74 mg/L when influent TN concentration and COD/TN ratio were 42.56 mg/L and 1.52, respectively. Nitrogen removal via anammox accounted for 90% of dinitrogen production in the PDA reactor. Finally, a granule-based combined process of PDA with nitrification was proposed for achieving anammox in mainstream WWTPs.

Facilitating sludge granulation and favoring glycogen accumulating organisms by increased salinity in an anaerobic/micro-aerobic simultaneous partial nitrification, denitrification and phosphorus removal (SPNDPR) process

Abstract:This study used salinity (0.5 wt%, 0.75 wt%) to accelerate the formation of ammonia oxidizing bacteria (AOB)-enriched aerobic granular sludge in a lab-scale anaerobic/micro-aerobic simultaneous partial nitrification, denitrification and phosphorus removal (SPNDPR) reactor. Results confirmed that the average granule diameter increased from 298.7 to 425.4 µm after 45 days of salinity stress even with low dissolved oxygen. Extracellular polymeric substances increased from 149.5 to 387.7 mg/g VSS after salinity (0.75 wt%) treatment, in turn accelerating granulation. Partial nitrification was maintained under the salinity condition due to the relative high activity and abundance of AOB, and the observed nitrite accumulation ratio averaged 98.9%. Salinity favored glycogen-accumulating organisms over polyphosphate-accumulating organisms (PAOs)/denitrifying-PAOs, with the abundance of Candidatus_Competibacter increasing from 4.86% to 15.34% and the simultaneous partial nitrification-denitrification efficiency increasing from 74.4% to 91.1%, promoting N-removal potential. The P-removal performance was good under 0.5 wt% salinity but was inhibited under 0.75 wt% salinity.
Aerobic granulation in a continuous-flow simultaneous nitrification, endogenous denitrification, and phosphorus removal system fed with low-strength wastewater: Granulation mechanism and microbial succession

Abstract:The culture of aerobic granular sludge (AGS) in continuous-flow system for treating low-strength municipal wastewater is still a challenge for its widespread application. This study developed a continuous-flow reactor based on metabolic and hydraulic selection pressure to explore the feasibility of culturing AGS in conventional continuous-flow mode. The results showed that aerobic granulation was successfully achieved with a mean particle size of 373 μm and almost all granules (95.6 %) were larger than 200 μm. An obvious three-stages granulation process comprised of initial adhesion, the formation of early aggregates that acted as nuclei of granules, and the final AGS maturation, was observed in this work. The selective enrichment of slow-growing bacteria in alternating feast/famine condition played a critical role in the aerobic granulation. The analysis results of extracellular polymeric substances revealed that β-polysaccharides and proteins distributed uniformly within the mature granules, while α-polysaccharides were found mainly concentrated around the granules, which was believed to enhance the mechanical strength of granules. The continuous-flow AGS system exhibited an excellent simultaneous carbon, nitrogen, and phosphorus removal performance with high removal efficiencies of NH4 +-N (97.6 %), TIN (91.0 %), COD (93.7 %), and TP (94.8 %). Moreover, a low sludge yield of 0.1 gMLSS/gCOD was obtained for dramatic in situ sludge reduction. Illumina MiSeq sequencing results confirmed the outstanding accumulation of GAOs (Candidatus_Competibacter) and PAOs (Rhodocyclaceae) in the system, which were responsible for simultaneous C, N, P removal and sludge reduction. Overall, this study demonstrated the feasibility of aerobic granulation in conventional continuous-flow mode and offered insights into the granulation mechanism, providing implications and references for the application of AGS technology in existing wastewater treatment plants.

Achieving high-rate partial nitritation with aerobic granular sludge at low temperatures

Abstract:Partial nitritation is necessary for the implementation of the mainstream anammox (anaerobic ammonium oxidation) process in wastewater treatment plants. However, the difficulty in outcompeting nitrite-oxidizing bacteria (NOB) at mainstream conditions hinders the performance of partial nitritation. The present work aimed to develop a high-rate partial nitritation process for low-ammonium wastewater treatment at low temperatures by seeding aerobic granules. Experimental results suggested that both stratified structure of nitrifiers developed in the granules and sufficient residual ammonium concentration (18–35 mg N L−1) in the bulk liquid contributed to efficient NOB repression. With the hydraulic retention time progressively shortened from 1.0 to 0.17 h, the influent nitrogen loading rate of the partial nitritation process reached 6.8 ± 0.4 kg N m−3 d−1 even at 10–15 °C. The high concentration (7.5 gVSS L−1) and activity (0.48 g N g−1 VSS d−1 at 11 °C) of granular sludge made the reactor possess an overcapacity evaluated by the ratio between the actual ammonium oxidation rate of the granules and their maximum potential. The overcapacity helped the reactor to face the adverse effect of decreasing temperatures. Overall, this work indicated the great potential of applying aerobic granules to achieve high-rate partial nitritation at mainstream conditions. Moreover, anammox bacteria with a relative abundance of 2.8% was also identified in the partial nitritation granules at the end of this study, suggesting that the granules provided a habitable niche for anammox bacteria growth. Note that these results cannot fully relate to the treatment of real domestic/municipal wastewater, they are a source of important information increasing the knowledge about low temperature partial nitrification.Graphical abstract

Abstract:The denitrification of high-strength nitrate wastewater is a major problem for many industries, especially for those involved in the production of petrochemicals, explosives, fertilizers, pectin, nuclear, and many metal-finishing industries, whose wastewater usually contain large amount of salts. What is more, the denitrification of synthetic high-nitrate wastewater containing 14,000mgNL(-1) in an expanded granular sludge bed (EGSB) reactor had been achieved in our previous study. The activated sludge culture was acclimatized by a stepwise increase in the nitrate concentration of synthetic waste. In the present work, the denitrification of synthetic high-salinity and high-nitrate wastewater containing 6,000mgL(-1)-N and 11% (w/v) salinity was achieved in the similar reactor. Meanwhile, The phylogenetic analysis of isolated 16S rRNA gene sequences indicated that halophilic species, Halomonas sp. and Marinobacter sp., were predominant at 11% salinity, suggesting that these bacteria show a high denitrifying activity in the EGSB reactor. The Shannon-Wiener index (H), representing the bacterium community diversity of the sludge sample, was calculated as 3.79. The EGSB reactor offers bright prospects for the treatment of high-salinity nitrate wastewater.
Successful operation performance and syntrophic micro-granule in partial nitritation and anammox reactor treating low-strength ammonia wastewater

Abstract:The stable operation of the partial nitritation and anammox (PN/A) process is a challenge in the treatment of low-strength ammonia wastewater like sewage mainstream. This study demonstrated the feasibility of achieving stable operation in the treatment of 50 mg/L ammonia wastewater with a micro granule-based PN/A reactor. The long-term operation results showed nitrogen removal efficiencies of 71.8 ± 9.9% were stably obtained under a relatively short hydraulic retention time (HRT) of 2 h. The analysis on the physicochemical properties of the granules indicated most of the granules were in a size in a range of 265-536 μm, and the elementary composition of the granules was determined to be CH1.61O0.61N0.17S0.01P0.03. The microbial analysis revealed Candidatus Kuenenia stuttgartiensis anammox bacteria and Nitrosomonas-like AOB were the two most dominant bacteria with 27.6% and 10.5% abundance, respectively, both of which formed spatially syntrophic co-immobilization within the micro-granules. The ex-situ activity tests showed the activity of NOB was well limited through DO regulation in the reactor. These results provide an alternative PN/A process configuration for low-strength wastewater treatment by sustaining microstate granules. Optimization of the nitrogen sludge loading rate and DO regulation are important for the successful performance.

Abstract:Aerobic granular sludge (AGS) technology is increasingly considered for wastewater treatment. AGS stability particularly under lower COD/N ratios is an impediment for AGS technology. This study evaluated AGS stability and nitrogen removal at different loading rates of 0.03 to 4 kg NH4+-N m-3 d-1 and COD/N ratios of 18.3 to 0.13. Ammoniacal and total nitrogen removals were high at 99.9% and 99.3%, respectively, during 440 days. MiSeq sequencing revealed a reduction in bacterial diversity and enrichment of ammonia oxidizing bacteria (AOB), anammox and denitrifying bacteria. Quantitative PCR showed enrichment of AOB, anammox bacteria, Nitrospira and denitrifiers. Chemical data and bacterial community supported occurrence of nitritation and anammox pathways. AGS had stable granular structure with excellent settling properties at lower COD/N≤ 1. Removal of high-strength ammonium could be partly explained by the existing nitrogen pathways suggesting novel mechanisms. Nevertheless, results presented here support implementation of AGS process for ammonium wastewaters.

Abstract:This study investigated the effects of hydraulic retention time (HRT) at 12 h, 7.2 h and 2.4 h on partial nitrification efficiency of continuous-flow aerobic granular reactors (CFAGRs) with mature aerobic granules (500 +/- 20mg l-1). At HRT 12 h and 7.2h, the removal efficiency of both ammonia-nitrogen (NH4+ - N) and nitrite accumulation rate were exceeding 90%. At HRT 2.4 h, NH4+ - N removal efficiency was reduced but most of the conversion efficiency to nitrite was only slightly reduced. At HRT < 2.4 h, washout of aerobic granules occurred. In all tests conducted herein, the chemical oxygen demand removal efficiencies exceeded 90%. The clone library results noted the presence of ammonia-oxidizing bacteria belonged to beta-Proteobacteria subclass, including 94% of Nitrosomonas europaea and 6% of Nitrosomonas sp. The polymerase chain reaction and denaturing gradient gel electrophoresis results suggested that Alpha proteobacterium, Pseudoxanthomonas mexicana strain, Sphaerotilus natans and Uncultured gamma proteobacterium were responsible for the aerobic granular stability and processing performance. The present CFAGR successfully implemented continuous partial nitrification using aerobic granules at low HRT.
