Abstract:In this paper, we report a non-PCR-based electrochemical assay that can detect telomerase activity. Telomerase from HeLa cells may induce telomerization of thiolated primers immobilized on a gold electrode surface. With the telomerization reaction, more and more guanine-rich telomeric repeats are formed, so the electrochemical oxidation signal of guanine at about 1.00V, which is utilized to indicate the elongated guanine-rich telomeric repeats tethered to the primers, will be increased. This assay method can detect the telomerase activity originated from 3000 HeLa cells and thus holds promise as a simple and sensitive approach in clinical diagnosis of cancer.
Graphene-mesoporous silica-dispersed palladium nanoparticles-based probe carrier platform for electrocatalytic sensing of telomerase activity at less than single-cell level

Abstract:Human telomerase is a specialized ribonucleoprotein polymerase and is used as a clinical cancer marker and special target for chemotherapy. Traditional telomerase detection uses "telomerase repeat amplification protocol" (TRAP) assay. However, TRAP is questioned because it requires use of DNA polymerases, which is susceptible to polymerase chain reaction (PCR)-derived artifacts and time-consuming. Here, a novel PCR-free, electrocatalytic assay for signal-amplified detection of telomerase activity using graphene-mesoporous silica-dispersed palladium nanoparticles-based probe carrier platform to improve probe-target recognition is reported. The low background noise is achieved by using DNA site-specific cleavage endonuclease and the detection signal is amplified by using hemoglobin. As a highly efficient electron sink, hemoglobin does not carry out direct reduction at the surface, minimizing false positives. These merits make this assay highly sensitive, achieving the sensitivity comparable to TRAP. The developed electrocatalysis assay is PCR-free, simple in design, and fast in operation, therefore avoiding PCR amplification-related errors, which make it more reliable to evaluate telomerase activity for clinical use.

Abstract:A simple yet powerful polymerase chain reaction (PCR)-free strategy for visualized assay of human telomerase activity was reported in this work. Gold nanoparticles (AuNPs) based colorimetric strategy was applied with well-designed enzyme-aided cyclic amplification. Briefly, the detection relies on the elongated primers of telomerase substrate (TS) induced by telomerase, which open the hairpin DNA and hybridize with linker DNA, the trigger of AuNPs aggregation. Nicking endonuclease was added in the sensing system, which cleaved linker DNA after hybridization and released complimentary strand for cyclic hybridization with linker DNA, resulted in high sensitivity for the detection of telomerase. Down to 25 HeLa cells with high expression of telomerase could be recognized. The proposed strategy provides a good platform for the determination of telomerase activity, differentiation of cancer cell lines from normal cell line and screening of telomerase-targeted anticancer drugs.

Abstract:Telomerase is regarded as a crucial biomarker for the early diagnosis of malignant tumors and a valuable therapeutic target. In this work, a telomerase-triggered amplification strategy was designed on the basis of a catalyzed hairpin assembly (CHA) for bridging a signal probe of platinum nanoparticles (Pt NPs) anchored on three-dimensional (3D) epoxy-functionalized macroporous carbon (Pt/MPC-COOH) in an ultrasensitive electrochemical biosensor. Pt/MPC-COOH nanomaterials with interconnected macroporous structure not only immobilized hairpin DNA probe 2 (H2) via an amide reaction (Pt/MPC-COOH-H2), but they also generated an obvious electrochemical signal in response to acetaminophen (AP) oxidation. After the introduction of telomerase, telomerase primer (TP) was extended to a telomerase extension product (TEP) with several hexamer repeats (TTAGGG)n to initiate the CHA cycle, leading to signal amplification. Subsequently, with the TEP-triggered CHA cycle amplification strategy, a large amount of Pt/MPC-COOH-H2 was introduced on the electrode surface for the construction of the electrochemical platform, which realized the sensitive detection of telomerase activity from 102 to107 cells mL-1 with a limit of detection (LOD) of 9.02 cells mL-1. This strategy provides a sensitive method for the detection of biomolecules that could be useful for bioanalysis and early clinical diagnoses of diseases.

Abstract:We report an electrochemical sensor for telomerase activity detection based on spherical nucleic acids gold nanoparticles (SNAs AuNPs) triggered mimic-hybridization chain reaction (mimic-HCR) enzyme-free dual signal amplification. In the detection strategy, SNAs AuNPs and two hairpin probes were employed. SNAs AuNPs as the primary amplification element, not only hybridized with the telomeric repeats on the electrode to amplify signal but also initiated the subsequent secondary amplification, mimic-hybridization chain reaction of two hairpin probes. If the cells' extracts were positive for telomerase activity, SNAs AuNPs could be captured on the electrode. The carried initiators could trigger an alternative hybridization reaction of two hairpin probes that yielded nicked double helices. The signal was further amplified enzyme-free by numerous hexaammineruthenium(III) chloride ([Ru(NH3)6](3+), RuHex) inserting into double-helix DNA long chain by electrostatic interaction, each of which could generate an electrochemical signal at appropriate potential. With this method, a detection limit of down to 2 HeLa cells and a dynamic range of 10-10,000 cells were achieved. Telomerase activities of different cell lines were also successfully evaluated.

Abstract:A simple and rapid electrochemical sensor is constructed for the detection of telomerase activity based on the electrocatalysis of platinum nanoparticle (Pt NP) encapsulated metal-organic frameworks (MOFs), which are synthesized by one-pot encapsulation of Pt NPs into prototypal MOFs, UiO-66-NH2. Integrating with the efficient electrocatalysis of Pt@MOFs towards NaBH4 oxidation, this biosensor shows the wide dynamic correlation of telomerase activity from 5 × 10(2) to 10(7) HeLa cells mL(-1) and the telomerase activity in a single HeLa cell was calculated to be 2.0 × 10(-11) IU, providing a powerful platform for detecting telomerase activity.

Abstract:In the present study, an electrochemical method for highly sensitive detection of human telomerase activity was developed based on bio-barcode amplification assay. Telomerase was extracted from HeLa cells, then the extract was mixed with telomerase substrate (TS) primer to perform extension reaction. The extension product was hybridized with the capture DNA immobilized on the Au electrode and then reacted with the signal DNA on Au nanoparticles to form a sandwich hybridization mode. Electrochemical signals were generated by chronocoulometric interrogation of [Ru(NH(3))(6)](3+) that quantitatively binds to the DNA on Au nanoparticles via electrostatic interaction. This method can detect the telomerase activity from as little as 10 cultured cancer cells without the polymerase chain reaction (PCR) amplification of telomerase extension product.
Quantum dot-based electrochemical biosensor for stripping voltammetric detection of telomerase at the single-cell level

Abstract:Human telomerase is responsible for the maintenance of chromosome end structures and is a valuable biomarker for malignant growth. However, the accurate measurement of telomerase activity at the single-cell level has remained a great challenge. Here we develop a simple quantum dot (QD)-based electrochemical biosensor for stripping voltammetric detection of telomerase activity at the single-cell level. We designed a thiol-modified capture DNA which may be immobilized on the gold electrode by the gold-sulfur bond. The presence of telomerase enables the addition of the telomere repeats of (TTAGGG)n to the 3' end of the primer, accompanied by the incorporation of abundant biotins in the extension product with the assistance of the biotin-tagged dATP. The subsequent hybridization of extension product with the capture DNA and the addition of streptavidin-coated QDs induce the assembly of large amounts of QDs onto the electrode via specific biotin-streptavidin binding. After the acidic dissolution of QDs, the released Cd (II) can be simply quantified by anodic stripping voltammetry (ASV). Due to the introduction of large amounts of QDs by telomerase-induced primer extension reaction and the synergistic signal amplification induced by the release of Cd (II) from the QDs, this biosensor can detect telomerase activity at the single-cell level without the involvement of any thermal cycling and extra enzymes for signal amplification. Moreover, this assay exhibits a large dynamic range over four orders of magnitude and it is very simple without the involvement of specific hairpin probe design and complicated labelling, holding great potential in point-of-need testing.
DNAzyme-Mediated Biodeposition Coupling Adjustable Cascade Electric Fields for Photoelectrochemical Telomerase Activity Monitoring

Abstract:Telomerase, as a specialized reverse transcriptase, plays a vital role in early cancer diagnostics and prognosis; thus, developing efficient sensing technologies is of vital importance. Herein, an innovative "signal-on-off" photoelectrochemical (PEC) sensing platform was developed for ultrasensitive evaluation of telomerase activity based on an electron-transfer tunneling distance regulation strategy and DNAzyme-triggerable biocatalytic precipitation. Concretely, cascade internal electric fields between CuInS2 quantum dots (QDs), graphitic carbon nitride nanosheets (g-C3N4 NSs), and TiO2 nanorod arrays (NRAs) were developed to realize cascade electron extraction and hole transfer. Enabled by such a design, an effective "signal-on" state to gain a progressively enhanced PEC output was designed by suppressing the photogenerated electron-hole pair recombination. With the introduction of hairpin probe H2 and the subsequent extension of the primer sequence driven by the target telomerase, the CuInS2 QDs labeled with hairpin probe H1 were programmatically unfolded, resulting in CuInS2 QDs' close proximity to the working electrode away from the cascade interface, accompanied by the formation of G-quadruplex/hemin complexes. The gradual undermining of tunneling distance and implantation of DNAzyme-initiating biocatalytic precipitation tremendously induced the sluggish migration kinetics of the photoinduced charge, accompanied by the photocurrent intensity decrement, leading to the "signal-off" state. Under optimized conditions, the as-prepared PEC biosensor realizes ultrasensitive detection of telomerase activity from 10 to 105 cell·mL-1 with a detection limitation of 3 cells·mL-1. As a proof of concept, this well-designed method provides new insights into signal amplification for telomerase activity evaluation and also presents promising potential for further development in drug screening, healthcare diagnostics, and biological assays.

Abstract:We report a new assay for human telomerase activity that relies on polyvalent oligonucleotide nanoparticle conjugates as diagnostic probes and amplification units. Gold nanoparticles functionalized with specific oligonucleotide sequences can efficiently capture telomerase enzymes and subsequently be elongated. Both the elongated and unmodified oligonucleotide sequences are simultaneously measured. The two strands not only serve as internal positive controls for each other but also provide a way of amplifying signal. At high concentrations, both elongated and unmodified strands exhibit measurable responses. At low telomerase concentrations (e.g., from 10 HeLa cells), elongated strands cannot be detected, but the unmodified sequences, which come from the same probe particles, can be detected because their concentration is higher, providing a novel form of amplification. This new assay rivals the sensitivity of the conventional PCR-based method of telomerase detection.

Abstract:A new electrochemical method for telomerase activity assay was developed on the basis of hybridization chain reaction (HCR). assisted multiple signal amplification, aiming at improving the sensitivity and specificity of telomerase assay. The experiments utilized HeLa cells as original source of the telomerase in the electrochemical studies. The telomerase primer was firstly self-assembled on the surface of gold electrode. The telomerase catalyzed the elongation of the primer, producing the complementary sequences of hairpin probe H1. In this case, HCR was then initiated by interacting with two hairpin probes H1 and H2. Because both H1 and H2 were modified by biotin, horseradish peroxidase could be captured on the electrode surface through the high. affinity interaction between biotin and streptavidin, catalyzing the oxidation of o. phenylenediamine to produce 2,3-diaminophenazine. Therefore, the telomerase assay was realized by tracing the electrochemical signals with differential pulse voltammetry. This electrochemical method was of high efficiency and feasibility for detecting telomerase activity, and could trace the telomerase activity down to 10 cells/mL HeLa cells with a wide linear range. Besides, it could also easily distinguish the target enzyme from the control proteins with high specificity.

Abstract:The general electrochemical biosensors for telomerase detection require the immobilization of primers on the electrode surface for telomeric extension and hybridization reactions. However, immobilization of primers may suffer from the challenges of hindrance effect and configuration freedom, thus reducing the extension and hybridization efficiency. Herein, we developed a sensitive electrochemical biosensor for telomerase detection by integration of homogeneous extension and hybridization reactions and surface-tethered detection. In the presence of telomerase, the biotinylated primer (bio-primer) was efficiently elongated with telomeric repeats of (TTAGGG) n at the 3' end in solution. Then, the extension product (bio-DNA) was hybridized with the signal probe DNA modified on the surface of ferrocene (Fc)-capped gold nanoparticle (AuNP). The bio-DNA/DNA/Fc-AuNP hybrids were then tethered by streptavidin-modified electrodes through the specific avidin-biotin interactions, thus producing strong electrochemical signals from the oxidation of Fc tags. The biosensor was successfully used to determine telomerase in HeLa cells and monitor the inhibition efficiency of inhibitor. A wide linear range for the detection of telomerase extracted from HeLa cells was attained. This method has great potential in clinical diagnosis and anti-cancer drug development, and should be beneficial for the fabrication of novel biosensors by integration of homogeneous catalysis and hybridization reactions.

Abstract:We developed a novel telomere complementary (TC) oligonucleotide modified AuNP probe (TC-AuNPs) for colorimetric analysis of telomerase activity. The mechanism of this method is that the telomerase reaction products (TRP), which can hybridize with the TC-AuNPs, are able to protect the AuNPs from the aggregation induced by salt. It is demonstrated that the colorimetric method enabled the analysis of the telomerase activity in 1000 HeLa cells with the naked eye, and down to 100 HeLa cells with the aid of UV-Vis spectroscopy. This strategy is not only convenient and sensitive, but also has a tunable dynamic range. The platform is also applicable for the initial screening of a telomerase inhibitor to discover new anticancer drugs.
A sensitive, label-free electrochemical detection of telomerase activity without modification or immobilization

Abstract:Telomerase has become one of the most typical tumor marker because it is closely related to cancers. In this paper, a simple label-free electrochemical detection of telomerase activity by using methylene blue (MB) as a G-quadruplex binding probe was proposed, avoiding commonly used complex label procedures, nano-probe synthesis, complicated electrode modification, probe immobilization or signal amplification. In the presence of telomerase substrate (TS) primer, the binding of MB on primer was weak. When repeats of (TTAGGG) were extended on the TS primer under the action of telomerase, they formed multiple G-quadruplexes with the help of K+. As a result, a large amount of MB bounded to multiple G-quadruplexes because they have more strong interaction with G-quadruplexes than TS primer. As a result, the diffusion current of MB decreased sharply, which was strongly dependent on the telomerase activity. The DPV current change has a linear correlation with the logarithm of HeLa cell number in the range of 10-10,000 cells, with the detection limit of 3 cells. The high sensitivity was due to the formed multiple G-quadruplexes. Using indium tin oxide (ITO) as working electrode without modification ensured the good reproducibility of the method. The method was also simple, rapid, and has been successfully applied in the telomerase activity detection in urine with good selectivity and reproducibility, which is significant for cancer diagnosis, anticancer drugs screening, and cancer therapy evaluation.

Abstract:Enzyme-mediated signal amplification strategies have gained substantial attention in photoelectrochemical (PEC) biosensing, while natural enzyme on the photoelectrode inevitably obstructs the interfacial electron transfer, in turn deteriorating the photocurrent responses. Herein, Au nanoparticles and Cu2+-modified boron nitride nanosheets (AuNPs/Cu2+-BNNS) behaved as nanozyme to achieve remarkable magnification in the PEC signals from a novel signal-off aptasensor for ultra-sensitive assay of telomerase (TE) activity based on Ag2S/Ag nanoparticles decorated ZnIn2S4/C3N4 Z-scheme heterostructures (termed as Ag2S/Ag/ZnIn2S4/C3N4, synthesized by hydrothermal treatment). Specifically, telomerase primer sequences (TS) were extended by TE in the presence of deoxyribonucleoside triphosphates (dNTPs), which was directly bond with the thiol modified complementary DNA (cDNA), achieving efficient linkage with the nanozyme via Au-S bond. The immobilized nanoenzyme catalyzed the oxidation between 4-chloro-1-naphthol (4-CN) and H2O2 to generate insoluble precipitation on the photo-electrode. By virtue of the inhibited PEC signals with the TE-enabled TS extension, an aptasensor for assay of TE activity was developed, showing the wide linear range of 50-5×105 cell mL-1 and a low detection limit of 19 cell mL-1. This work provides some valuable guidelines for developing advanced nanozyme-based PEC bioanalysis of diverse cancer cells.

Abstract:The level and activity of telomerase are often considered as an important tumor biomarker index in clinical diagnosis. In this work, we developed a rapid and sensitive telomerase activity assay based on the 3,3′,5,5′-tetramethylbenzidine dilation (TMB2+) etching of gold nanorods (AuNRs) at a single-particle level. In principle, telomerase primers are triggered by telomerase to extend and form a hemin/G-quadruplex DNAzyme, which then catalyzes the hydroxyl radical oxidation of TMB to TMB2+, which in turn is able to efficiently etch gold nanorods (AuNRs), making the localized plasmonic resonance (LSPR) scattering features change under dark-field microscopy (DFM). Compared to the common strategy of direct etching of AuNRs based on a hydroxyl radical that has a short lifetime (3 μs), the TMB2+ is much more stable and greatly overcomes the drawbacks of the hydroxyl radical that is not able to effectively etch AuNRs owing to its short lifetime. The LSPR DFM scattering intensity reduction was found to have a good linear relationship with the telomerase level in the range of 100–24 000 cells and the limit of detection (LOD) reaching 43 cells. Telomerase extracted from different cancer cells such as MCF-7, HeLa, and HepG2 demonstrated good accuracy of this assay, showing that this TMB2+ etching strategy of AuNRs for the telomerase activity assay is successful and reliable.

Abstract:Telomerase, which is regarded as a common biomarker for early cancer diagnostics and a potential target for clinical therapies, has attracted considerable interests concerning its detection and monitoring. Herein, we propose a sensitive method by designing a gold nanoparticle (AuNP) probe for visually intracellular detection of telomerase activity. The AuNPs were functionalized with a telomerase substrate primer (SH‐prime). A 6‐carboxy‐fluorescein (FAM) modified strand (FAM‐probe) was attached to the surface of AuNP through its complementary stand (SH‐attach).In the absence of telomerase, the fluorescence resonance energy transfer (FRET) from FAM to AuNPs results in efficient fluorescence quenching. In the presence of telomerase, SH‐primers on AuNPs were extended with the repeat units (TTAGGG)n. The extension sequence triggered the strand displacement of FAM‐probe to restore the fluorescence signals. It is worth mentioning that the proposed strategy does not need to design complex hairpin structure and allows the measurement of telomerase in crude cell extracts down to 0.5 HeLa cells/μL in 2 h. In addition, the present sensing platform can be applied to the visually intracellular detection of telomerase activity in living cells.

An Amplified Fluorescence Biosensor for Intracellular Telomerase Determination and in Situ Imaging Based on Thioflavin T and Conjugated Polymer Nanoparticles

Abstract:Telomerase, as a significant tumor biomarker, is highly associated with cancers. Highly sensitive determination of telomerase activity is beneficial to accurate diagnosis and treatment of malignant tumors. A new simple and elegant fluorescence sensing strategy based on thioflavin T (ThT) and conjugated polymer nanoparticles was established to determine telomerase activity in living cells. In the presence of telomerase, the primer is prolonged with repeated sequence of (TTAGGG)(n). This long G-riched sequence can form G-quadruplex in the mediation of ThT. The combination of ThT and G-quadruplex allows ThT to emit robust fluorescence. The fluorescence can be further amplified by positively charged conjugated polymer nanoparticles (PPE-PLL). The remarkable amplification is attributed to the disaggregation of PPE-PLL nanoparticles and a good superposition of fluorescence emission from ThT and PPE-PLL. The results demonstrated a new amplification assay for the determination of telomerase activity in cell extracts and living cells with high specificity and sensitivity. The limit of detection is estimated to be 3 HeLa cells in 400 mu L. It is not only more sensitive than most reported biosensors for telomerase detection. Still, it is extremely simple, rapid, and low-cost since it is PCR-free, label-free, and enzyme-free.
CuS quantum dots activated DNAzyme for ratiometric electrochemical detection of telomerase activity

Abstract:Telomerase activity detection has attracted much attention concerning its importance for early cancer diagnosis. Here, we established a ratiometric electrochemical biosensor for telomerase detection based on CuS quantum dots (CuS QDs) dependent DNAzyme-regulated dual signals. The telomerase substrate probe was used as the linker to combine the DNA fabricated magnetic beads and CuS QDs. In this way, telomerase extended the substrate probe with repeated sequence to from hairpin structure, releasing CuS QDs as an input to DNAzyme modified electrode. DNAzyme was cleaved with high current of ferrocene (Fc) and low current of methylene blue (MB). On the basis of the obtained ratiometric signals, telomerase activity detection was achieved in the range of 1.0 × 10-12-1.0 × 10-6 IU/L, with the limit of detection down to 2.75 × 10-14 IU/L. Moreover, telomerase activity from HeLa extracts was also tested to verify the clinical application.

Abstract:In this work, we designed a nanoscaled porphyrinic metal-organic framework (PorMOF) with iron porphyrin as linker and zirconium ion as node for electrochemical detection of telomerase activity. The as-prepared PorMOF was characterized with scanning electron microscopy, powder X-ray diffraction, and spectroscopic techniques and demonstrated excellent electrocatalytic activity toward O2 reduction. Sequentially, the functionalization of PorMOF with streptavidin results in a water-stable electrochemical tracer for detection of telomerase. Upon the telomerase-triggered extension, the assistant DNA 1 (aDNA1)-assistant DNA 2 (aDNA2) duplex could switch into a hairpin structure, and thus, the aDNA2 was released and then hybridized with the capture DNA. Therefore, the PorMOF@SA tracer could be introduced on the electrode surface via biotin-streptavidin recognition, leading to the strong electrochemical signal for readout. The developed approach displayed desirable dynamic range and limitation of detection down to 30 HeLa cells mL-1. The telomerase activity was 2.2 X 10(-11) IU in a single HeLa cell with good reproducibility and stability. The nanoscaled porphyrinic MOF provided a powerful platform for electrochemical signal transduction and had a promising application in the determination of various biomolecules.
