Abstract:Immunosensors based on field-effect transistors with nanowire channels (NWFETs) provide fast and real-time detection of a variety of biomarkers without the need for additional labels. The key feature of the developed immunosensor is the coating of silicon NWs with multilayers of polyelectrolytes (polyethylenimine (PEI) and polystyrene sulfonate (PSS)). By causing a macromolecular crowding effect, it ensures the "soft fixation" of the antibodies into the 3-D matrix of the oppositely charged layers. We investigated the interaction of prostate-specific antigen (PSA), a biomarker of prostate cancer, and antibodies adsorbed in the PEI and PSS matrix. In order to visualize the formation of immune complexes between polyelectrolyte layers using SEM and AFM techniques, we employed a second clone of antibodies labeled with gold nanoparticles. PSA was able to penetrate the matrix and concentrate close to the surface layer, which is crucial for its detection on the nanowires. Additionally, this provides the optimal orientation of the antibodies' active centers for interacting with the antigen and improves their mobility. NWFETs were fabricated from SOI material using high-resolution e-beam lithography, thin film vacuum deposition, and reactive-ion etching processes. The immunosensor was characterized by a high sensitivity to pH (71 mV/pH) and an ultra-low limit of detection (LOD) of 0.04 fg/mL for PSA. The response of the immunosensor takes less than a minute, and the measurement is carried out in real time. This approach seems promising for further investigation of its applicability for early screening of prostate cancer and POC systems.

Abstract:This article describes a novel electrochemical on-chip biosensor that utilises the anti-PSA antibody (Ab) and silver nanoparticles (AgNPs) to enhance the sensing and detection capability of the prostate-specific antigen (PSA) in the blood. The AgNPs are prepared, characterised, and applied to a silicon photonic on-chip biosensing receptor platform designed to enhance the accurate detection of PSA. The AgNPs were synthesised by a chemical reduction method using silver nitrate (AgNO3) as the precursor. Transmission electron microscopy (TEM), selected area electron diffraction (SAED), energy dispersion X-ray spectroscopy (EDS), small angle X-ray scattering (SAXS), X-ray diffraction (XRD), and light microscopy were among the methods used in the characterisation and analysis of the AgNPs. Each stage of the immunosensor fabrication was characterised using cyclic voltammetry. The proposed immunosensor was applied in the detection of PSA, a prostate cancer biomarker, with a high sensitivity and a limit of detection of 0.17 ng/mL over a linear concentration range of 2.5 to 11.0 ng/mL. The immunosensor displayed good stability and was selective in the presence of interfering species like immunoglobulin (Ig) in human serum, ascorbic acid (AA), and diclofenac (Dic). The detectivity and sensitivity are significantly higher than previous reports on similar or related technologies.

Abstract:We describe a highly sensitive electrochemical sandwich-type immunosensor for the prostate specific antigen (PSA), a biomarker related to prostate cancer. A glassy carbon electrode was covalently modified with multi-walled carbon nanotubes to which the anti-PSA was immobilized. Following addition of a sample containing PSA, a secondary antibody is added that consists of gold nanoparticles modified with secondary antibody and 6-ferrocenyl hexanethiol acting as the signal molecule. This approach represents a multiple signal amplification strategy in that the functionalized carbon nanotubes improve the electron transfer on the surface of the electrode, while the gold nanoparticles act as carriers for capturing large quantities of Ab2 and ferrocene. The modifying processes were characterized by cyclic voltammetry and electrochemical impedance spectroscopy. Under the optimal conditions, the differential pulse voltammetric signals (acquired at a typical potential of 0.31 V vs. SCE) is linearly related to the concentration of PSA in the 10 pg.mL−1 to 100 ng.mL−1 range. The detection limit is 5.4 pg.mL−1. When applied to the determination of PSA in spiked human serum, the recoveries were between 92.4 and 120.0 %. Abstract:Prostate specific antigen (PSA) is a major biomarker used clinically in the detection of prostate cancer. Current commercial immunoassays for detection of this biomarker rely on spectroscopic absorbance and electro-chemiluminescence which are able to detect as low as 0.03 ng/mL of PSA within 100muL of serum. In this paper, a novel amplified electrochemical technique for detection of prostate specific antigen is described. This electrochemical approach is a highly sensitive method that can have detection limit of 0.004 ng/mL in only 10 mL of serum, a mass detection limit 75 times lower than current commercial techniques. This highly sensitive, low cost and fast detection method not only allows for detection of cancer biomarkers in serum, but also in tissue and cells. Moreover, immunoassay arrays based on this methodology will be highly promising for application in large-scale clinical screening and point-of-care diagnostics. Abstract:Prostate specific antigen (PSA), as a biomarker, plays important roles in early diagnosis of male prostate diseases, especially prostate cancer. In this study, a novel electrochemical probe polymer of poly(ferriporphyrin-co-acrylamide) was synthesized, by further combined with reduced graphene oxide and gold nanoparticles, and oriented immobilization of PSA antibody, a label-free electrochemical immunosensor was obtained, and then successfully applied in the determination of PSA in 10 % non-antigen serum system. The prepared biosensor expressed high selectivity toward PSA, with a LOD of 0.001 ng mL−1, a linear range of 0.01–110 ng mL−1, and a sensitivity of 15.78 µA mL ng−1. What’s more, it showed good stability, well repeatability and high selectivity in the real PSA sample detection. This work provided certain references for the design of the new electrochemical probes and the detection of tumor markers.

Abstract:We describe highly sensitive, label-free, multiplexed electrical detection of cancer markers using silicon-nanowire field-effect devices in which distinct nanowires and surface receptors are incorporated into arrays. Protein markers were routinely detected at femtomolar concentrations with high selectivity, and simultaneous incorporation of control nanowires enabled discrimination against false positives. Nanowire arrays allowed highly selective and sensitive multiplexed detection of prostate specific antigen (PSA), PSA-α1-antichymotrypsin, carcinoembryonic antigen and mucin-1, including detection to at least 0.9 pg/ml in undiluted serum samples. In addition, nucleic acid receptors enabled real-time assays of the binding, activity and small-molecule inhibition of telomerase using unamplified extracts from as few as ten tumor cells. The capability for multiplexed real-time monitoring of protein markers and telomerase activity with high sensitivity and selectivity in clinically relevant samples opens up substantial possibilities for diagnosis and treatment of cancer and other complex diseases.

Abstract:In diagnosing prostate cancer and distinguishing it from other prostate diseases, the ratio of the concentration of free prostate-specific antigen (f-PSA) to total prostate-specific antigen (t-PSA), i.e., (f-PSA%) is more accurate than the concentration of t-PSA alone. Immunoassay based on surface-enhanced Raman scattering (SERS) frequency shift has been proven to be particularly suitable for detecting large biomolecules with high reproducibility. Along similar lines, the present study developed a SERS-based biosensor that simultaneously detects t-PSA and f-PSA. The 4-mercaptobenzoic acid (MBA) on the immunocapture substrate is coupled to the t-PSA antibody through the carboxyl group, and the combination of t-PSA induces the Raman frequency shifts of MBA. The immunocolloidal gold attached with f-PSA antibodies selectively capture the f-PSA that immobilized on the MBA-modified SERS substrates, allowing for f-PSA quantification according to the SERS intensities of the 5, 5'-Dithiobis (succinimidyl-2-nitrobenzoate) (DSNB) probe. The results show that f-PSA and t-PSA have good linear response in the concentration scale of 0.1-20 ng/mL, and 1-200 ng/mL, respectively. The biosensor combines Raman frequency shifts and intensities, which greatly simplifies traditional procedures for f-PSA% detection. All the results demonstrated the great potential of the proposed biosensor in highly reproducible and accurate diagnosis of prostate cancers.

Abstract:Prostate cancer is one of the most common cancers in the world, and its early diagnosis can effectively reduce mortality. A new label-free photoelectrochemical (PEC) immunosensor on the basis of Bi 2 WO 6 /BiOBr nanocomposite materials has been successfully prepared for the test of prostate-specific antigen (PSA) in human serum in this work. The Ag 2 S-sensitized Bi 2 WO 6 /BiOBr heterojunction was used as a photosensitive material, which effectively improved the photocurrent response. On Bi 2 WO 6 /BiOBr surface, dopamine immobilized PSA antibody by self-polymerizing to form polydopamine membrane. Antigen and antibody are specifically combined to achieve quantitative detection of PSA according to the current changes at different concentrations of antigen. Under the optimal experimental conditions, the PEC immunosensor has an ideal linear relationship between 1 pg/mL - 50 ng/mL, and the detection limit is 0.084 pg/mL. In addition, the prepared immunosensor has good stability, reproducibility and selectivity, providing a new method for the detection of PSA in actual sample analysis.

Abstract:Label-free, real-time, parallel and complementary electrical detection of proteins is demonstrated by p-type and n-type silicon nanowire field-effect transistors in the same arrays. Composed of hundreds of individually electrically addressable nanowire devices with highly sensitive and reproducible performances, these nanowire arrays can be controllably modified by monoclonal antibodies, and show discrete conductance changes characteristic of highly selective binding and unbinding of target proteins, such as prostate specific antigens (PSA), thus providing a general and powerful platform for high-throughput real-time parallel detection and rapid screening of libraries of biomolecules. Studies show that the PSA proteins can be routinely detected at femtomolar concentrations with high selectivity, and simultaneously incorporation of both p-type and n-type silicon nanowire devices enable discrimination against false positive/negative signals. The integrated complementary nanowire sensor arrays open up substantial opportunities for diagnosis and treatment of complex diseases such as cancer, detection of biological threats, and fundamental proteomic and biophysical studies.
Electrostatic Nanocage-Confined Probe for Electrochemical Detection of CA19-9 in Human Serum.

Abstract:Prompt and accurate detection of CA19-9 in human serum has great clinical significance for the early diagnosis and disease monitoring of cancer. Herein, we develop a convenient and antifouling electrochemical sensor for CA19-9 determination by immobilization of both an electrochemical redox probe [methylene blue (MB)] and immunorecognition element (CA19-9 antibody) on an electrostatic nanocage consisting of bipolar silica nanochannel array (bp-SNA). bp-SNA is composed of a negatively charged inner layer (n-SNA) and positively charged outer layer (p-SNA), which could be stably prepared on indium tin oxide (ITO) in several seconds using a two-step electrochemically assisted self-assembly approach and display asymmetric surface charges for confinement and enrichment of cationic MB into the inner n-SNA layer through electrostatic interaction. Modification of the CA19-9 antibody on the top surface of bp-SNA confers the sensing interface with specific recognition capacity. An antibody-antigen complex formed at the as-prepared immunosensor causes the decreased electrochemical signals of MB, achieving sensitive determination of CA19-9 with a wider linear dynamic range from 10 μU/mL to 50 U/mL and a low detection limit (3 μU/mL). Furthermore, accurate and feasible analysis of the CA19-9 amount in human serum samples by our proposed probe-integrated electrochemical immunosensor is realized.
Horizontally Aligned Carbon Nanotube Based Biosensors for Protein Detection.

Abstract:A novel horizontally aligned single-walled carbon nanotube (CNT) Field Effect Transistor (FET)-based biosensing platform for real-time and sensitive protein detections is proposed. Aligned nanotubes were synthesized on quartz substrate using catalyst contact stamping, surface-guided morphological growth and chemical vapor deposition gas-guided growth methods. Real-time detection of prostate-specific antigen (PSA) using as-prepared FET biosensors was demonstrated. The kinetic measurements of the biosensor revealed that the drain current (I) decreased exponentially as the concentration of PSA increased, indicating that the proposed FET sensor is capable of quantitative protein detection within a detection window of up to 1 µM. The limit of detection (LOD) achieved by the proposed platform was demonstrated to be 84 pM, which is lower than the clinically relevant level (133 pM) of PSA in blood. Additionally, the reported aligned CNT biosensor is a uniform sensing platform that could be extended to real-time detections of various biomarkers.

Abstract:Prostate-specific antigen (PSA) is the most widely used biomarker for the early diagnosis of prostate cancer. Existing methods for PSA detection are burdened with some limitations and require improvement. Herein, we developed a novel microfluidic-electrochemical (μFEC) detection system for PSA detection. First, we constructed an electrochemical biosensor based on screen-printed electrodes (SPEs) with modification of gold nanoflowers (Au NFs) and DNA tetrahedron structural probes (TSPs), which showed great detection performance. Second, we fabricated microfluidic chips by DNA TSP-Au NF-modified SPEs and a PDMS layer with designed dense meandering microchannels. Finally, the μFEC detection system was achieved based on microfluidic chips integrated with the liquid automatic conveying unit and electrochemical detection platform. The μFEC system we developed acquired great detection performance for PSA detection in PBS solution. For PSA assays in spiked serum samples of the μFEC system, we obtained a linear dynamic range of 1-100 ng/mL with a limit of detection of 0.2 ng/mL and a total reaction time <25 min. Real serum samples of prostate cancer patients presented a strong correlation between the "gold-standard" chemiluminescence assays and the μFEC system. In terms of operation procedure, cost, and reaction time, our method was superior to the current methods for PSA detection and shows great potential for practical clinical application in the future.

Abstract:The development of biosensors capable of averting biofouling and detecting biomarkers in complex biological media remains a challenge. Herein, an ultralow fouling and highly sensitive biosensor based on specifically designed antifouling peptides and a signal amplification strategy was designed for prostate specific antigen (PSA) detection in human serum. A low fouling layer of poly(ethylene glycol) (PEG) doped the conducting polymer poly(3,4-ethylenedioxythiophene) (PEDOT) was electrodeposited on the electrode surface, followed by the immobilization of streptavidin and further attachment of biotin-labelled peptides. The peptide was designed to include PSA specific recognition domain (HSSKLQK) and antifouling domain (PPPPEKEKEKE), and the terminal of the peptide was functionalized with -SH group. DNA functionalized gold nanorods (DNA/AuNRs) were then attached to the electrode, and methylene blue (MB) molecules were adsorbed to the DNA to form the signal amplifier. In the presence of PSA, the peptide was specifically cleaved and resulted in the loss of AuNRs together with DNA and MB, and thus significant decrease of the current signal. The biosensor exhibited a low limit of detection (LOD) of 0.035 pg mL-1 (S/N = 3), with a wide linear range from 0.10 pg mL-1 to 10.0 ng mL-1, and it was able to detect PSA in real human serum owing to the presence of the antifouling peptides, indicating great potential of the constructed biosensor for practical application.

Abstract:Highly sensitive analysis of cancer biomarkers demonstrates an important impact in early diagnosis and therapies of cancer. A novel surface-enhanced Raman scattering (SERS) based immunoassay using microfluidic technique was reported for rapid analysis of prostate-specific antigen (PSA) biomarker. It is a useful screening test to discriminate prostate cancer and other diseases related to prostate. A "sandwich" immunoassay based on SERS nanotags, PSA biomarkers, and magnetic beads was applied on a pump-free microfluidic sensor. Magnetic immunocomplexes are isolated and trapped at the detection chamber by a permanent magnet integrated into the chip. The PBS buffer washed magnetic immunocomplexes and brought the free gold nanoparticles to the downsteam channel for waste. Our results show a good linear response in the range from 0.01 to 100 ng mL-1. The limit of detection of the PSA level is estimated to be below 0.01 ng mL-1 using this chip. This detection level of PSA biomarker in human serum can be accomplished in 5 min without manual incubation and a heavy syringe pump. To the best of our knowledge, this is the first SERS-based immunoassay which applied a pump-free microfluidic chip as a detection platform. We believe that the proposed method reveals a valuable potential tool for the diagnosis of prostate cancer.

Abstract:Prostate cancer is a disease with a high incidence and mortality rate in men worldwide. Serum prostate-specific antigens (PSA) are the main circulating biomarker for this disease in clinical practices. In this work, we present a portable and reusable microfluidic device for PSA quantification. This device comprises a polymethyl methacrylate microfluidic platform coupled with electrochemical detection. The platinum working microelectrode was positioned in the outflow region of the microchannel and was modified with carbon nanofibers (CNF)-decorated gold nanoporous (GNP) structures by the dynamic hydrogen bubble template method, through the simultaneous electrodeposition of metal precursors in the presence of CNF. CNF/GNP structures exhibit attractive properties, such as a large surface to volume ratio, which increases the antibody's immobilization capacity and the electroactive area. CNFs/GNP structures were characterized by scanning electron microscopy, energy dispersive spectrometry, and cyclic voltammetry. Anti-PSA antibodies and HRP were employed for the immune-electrochemical reaction. The detection limit for the device was 5 pg mL-1, with a linear range from 0.01 to 50 ng mL-1. The coefficients of variation within and between assays were lower than 4.40%, and 6.15%, respectively. Additionally, its clinical performance was tested in serum from 30 prostate cancer patients. This novel device was a sensitive, selective, portable, and reusable tool for the serological diagnosis and monitoring of prostate cancer.

Abstract:Microfluidic electrochemical sensors are emerging as favorable platforms for rapid and precise detection of disease markers relying on their superiority of miniaturization, low electrolyte consumption, high precision and integration. Herein, an efficient and facile integrated microfluidic sensor with three-dimensional graphene fiber film/chitosan-gold nanoparticles (3D GF/CS-Au NPs) micro-nano composites as sensing interface was developed for the analysis of prostate specific antigen (PSA). The GF with 3D network structures modified by large amounts of CS-Au nanoparticles could facilitate the enhancement of electrochemical signals. The electrodes and micro-channel were simply assembled and encapsulated by bolts, which made the microfluidic chip had excellent portability and operability. On the basis of this new microfluidic device and the 3D GF/CS-Au NPs composites, a sensitive determination of PSA could be realized with a low detection limit of 0.51 pg/mL and a wide response range from 0.001 -200 ng/mL. Moreover, the proposed biosensor was further used to detect analytes in human blood samples, showing the satisfactory results. This novel microfluidic electrochemical immunosensor demonstrated superior performance for PSA detection, holding potential application in clinical diagnosis and personalized healthcare.

Abstract:How to fabricate scale low-cost microfluidic device for detection of biomarkers owns a great requirement. Herein, it is for the first time reported that a new microfluidic device based on bonding polydimethylsiloxane microfluidic channels onto the substrate of a screen-printed electrode with coating glass solution was fabricated for electrochemical sensing of prostate-specific antigen (PSA). Compared to traditional microfabrication processes, this method is simple, fast, low cost, and also suitable for mass production. The prepared screen-printed electrode-based microfluidic device (CASPE-MFD) was used for the detection of the PSA in human serum. The prepared CASPE-MFD had a detection limit of 0.84 pg/mL (25.8 fM) and a good linearity with PSA concentration ranging from 0.001 to 10 ng/mL, which showed a great promise platform toward the development of miniaturized, low-cost electrochemical microfluidic device for use in human health, environmental monitoring, and other applications.
Nanogold-functionalized G-C3n4 Nanohybrids for Sensitive Impedimetric Immunoassay of Prostate-Specific Antigen Using Enzymatic Biocatalytic Precipitation

Abstract:This work reports on a new impedimetric immunosensing strategy for sensitive detection of prostate-specific antigen (PSA) in biological fluids. The assay was carried out on monoclonal anti-PSA capture antibody-modified glassy carbon electrode with a sandwich-type detection format. Gold nanoparticles-decorated g-C3N4 nanosheets (AuNP/g-C3N4), synthesized by the wet-chemistry method, were utilized for the labeling of polyclonal anti-PSA detection antibody and horseradish peroxidase (HRP). Upon target PSA introduction, the sandwiched immunocomplex could be formed between capture antibody and detection antibody. Followed by the AuNP/g-C3N4, the labeled HRP could catalyze 4-choloro-1-naphthol into benzo-4-chlorohexadienone. The as-generated insoluble product was coated on the electrode surface, thus increasing the Faradaic impedance of Fe(CN)6(4-/3)(-) indicator between the solution and the base electrode. Under the optimal conditions, the impedance increased with the increasing target PSA in the sample, and exhibited a wide linear range from 10pgmL(-1) and 30ngmL(-1) with a detection limit of 5.2pgmL(-1). A repeatability and intermediate precision of <14% was accomplished. The specificity and method accuracy in comparison with commercial PSA ELISA kit for analysis of human serum specimens were relatively satisfactory.

Abstract:This study describes a novel dual-mode immunosensor that combines electrochemical (EC) and surface-enhanced Raman scattering (SERS) techniques for the detection of prostate-specific antigen (PSA), a biomarker associated with prostate cancer. The sensor consists of a nanocomposite of gold nanoparticles (AuNPs) deposited on two-dimensional (2D) molybdenum disulfide (Au@MoS2) modified on a working carbon electrode of a screen-printed electrode (SPE). Subsequently, the primary antibody (Ab1) is immobilized on the modified electrode, creating Ab1/Au@MoS2/SPE for specific recognition of the target PSA. In parallel, AuNPs are conjugated with a secondary antibody (Ab2) and a probe molecule, 3,3',5,5'-tetramethylbenzidine (TMB), leading nanotags (TMB/Ab2/AuNPs) formation exhibiting strong SERS and EC responses. Upon the presence of the target, sandwich immunocomplexes can be formed through antigen-antibody interactions (Ab1-PSA-Ab2). The differential pulse voltammetry (DPV) technique is employed for EC detection mode, while a handheld Raman spectrometer with a 785 nm excitation laser is utilized to collect SERS signals. The developed system demonstrates excellent selectivity and sensitivity, with low limits of detection (LODs) of 3.58 pg mL-1 and 4.83 pg mL-1 for EC and SERS sensing, respectively. Importantly, the dual-mode immunosensor proves effective quantifying PSA protein in human serum samples with good recovery. Given its high sensitivity and proficiency in analyzing biological samples, this proposed immunosensor holds promise as an alternative tool for the early diagnosis of cancers.
