Abstract:The study aimed to develop a new reverse-phase high-performance liquid chromatography (RP-HPLC) method with diode array detection (DAD) detection for simultaneous estimation of escitalopram (EST) and clonazepam (CZP) in tablet dosage forms with a quality by design (QbD) approach. The chromatographic conditions were optimized by Box-Behnken design (BBD) and developed method was validated for the linearity, system suitability, accuracy, precision, robustness, sensitivity, and solution stability according to International Council for Harmonization (ICH) guidelines. EST and CZP standard drugs peaks were separated at retention times of 2.668 and 5.046 min by C-18 column with dimension of 4.6 × 100 mm length and particle size packing 2.5 µm. The mobile phase was methanol: 0.1% orthophosphoric acid (OPA) (25:75, v/v), with a flow rate of 0.7 mL/min at temperature of 26 °C. The sample volume injected was 20 µL and peaks were detected at 239 nm. Using the standard calibration curve, the % assay of marketed tablet was founded 98.89 and 98.76 for EST and CZP, respectively. The proposed RP-HPLC method was able to detect EST and CZP in the presence of their degradation products, indicating the stability-indicating property of the developed RP-HPLC method. The validation parameter's results in terms of linearity, system suitability, accuracy, precision, robustness, sensitivity, and solution stability were in an acceptable range as per the ICH guidelines. The newly developed RP-HPLC method with QbD application is simple, accurate, time-saving, and economic.
Spectrophotometric method for simultaneous estimation of escitalopram oxalate and clonazepam in tablet dosage form

Abstract:A simple, accurate and precise spectrophotometric method has been developed for simultaneous estimation of escitalopram oxalate and clonazepam in combined dosage form. Simultaneous equation method is employed for simultaneous determination of escitalopram oxalate and clonazepam from combined dosage forms. In this method, the absorbance was measured at 238 nm for escitalopram oxalate and 273 nm for clonazepam. Linearity was observed in range of 5-100 mug/ml and 5-50 mug/ml for escitalopram and clonazepam respectively. Recovery studies confirmed the accuracy of proposed method and results were validated as per ICH guidelines. The method can be used for routine quality control of pharmaceutical formulation containing escitalopram and clonazepam.

Abstract:Objective: The main objective was to develop stability indicating UPLC technique for simultaneous estimation of Benazepril and Hydrochlorothiazide in bulk and formulation. Methods: 0.1% Triethylamine phosphate: Methanol (25:75v/v) was used as the mobile phase. Benazepril linearity was found to be 4-20 μg/ml and Hydrochlorothiazide linearity was found to be 5-25 g/ml. The detection wavelength was 236 nm, and the retention period of Benazepril was 3.4 min and Hydrochlorothiazide was 5.4 min with a flow rate of 1.0 ml/min. According to the ICH guidlines, the proposed method was validated and stress studies revealed that the drugs are prone to alkali and peroxide stress conditions. Results: The calibration curve was plotted, and the regression equations for Benazepril were y = 2,01,491.67x+60,532.30 with a correlation coefficient (r2) of 0.9997 and Hydrochlorothiazide were y = 64,635.86x-74,607.10 with a correlation coefficient (r2) of 0.9994. According to the accuracy research, the percent recovery of Benazepril is 99.09-100.69 % and that of Hydrochlorothiazide is 98.27-101.88%, both of which are within the ICH recommendations. Benazepril has a limit of detection of 0.08 g/ml-0.24 g/ml and Hydrochlorothiazide has a limit of quantitation of 0.03 g/ml-0.10 g/ml. The procedure was found to be straightforward, linear, rapid, exact, repeatable, and robust. It was determined that the % RSD was within ICH norms. Stress degradation tests showed the drug's vulnerability to oxidative, thermal, photolytic, acid, basic, and neutral hydrolysis stress conditions. Under the circumstances of alkali and peroxide stress, it was discovered that the drug degraded most quickly. Conclusion: The developed chromatographic technique under consideration was suitable for the accurate, precise, and quick simultaneous measurement of hydrochlorothiazide and benazepril in both their bulk and medicinal dose forms.
Mucosal plexus and electrolyte transport across the rat colonic mucosa.

Abstract:Histological and functional studies were performed on a preparation of rat colonic mucosa from which the myenteric and submucosal plexus were removed. This preparation, referred to as the mucosa preparation, was used to investigate the potential influence of the mucosal plexus on electrolyte transport. Two neuropharmacologically active agents were used: sea anemone toxin (ATX II) to stimulate the fibres of the mucosal plexus and tetrodotoxin (TTX) to block the fibres of the mucosal plexus. The morphology of the neuronal network of the mucosal plexus was visualized after the epithelium was removed and whole mount preparations of the lamina propria and circular muscle layer of muscularis mucosae were stained histochemically for acetylcholinesterase activity. Several levels of organization within the mucosal plexus were seen. Each crypt is encircled by a thin bundle of fibres near the top. These thin fibres connect with thicker bundles of fibres that encircle groups of two to five crypts in a broad band. These bundles of fibres are in turn connected to larger bundles of fibres which lie in a flat plane just below the crypts along the circular muscle layer of muscularis mucosae. In addition perikarya and ganglia were revealed within the mucosal plexus. The base‐line net transport of Na+ and Cl‐ across the mucosa preparation was completely inhibited by ATX II (10(‐6) M). This effect of ATX II on net Na+ and Cl‐ transport was accompanied with an increase in the short‐circuit current (Isc), transmural conductance, and open‐circuit potential difference across the mucosa preparation. The effect of ATX II on Isc was dose dependent with a half‐maximal effective concentration at 5 X 10(‐8) M‐ATX II and a maximal effective concentration of 10(‐7) M. ATX II was effective only when added to the serosal solution. Net Na+ and Cl‐ transport was restored by TTX (10(‐6) M) to base‐line values in ATX II‐treated tissue. In addition the value of all three electrical parameters rapidly returned to the values measured before the addition of ATX II. TTX was effective in antagonizing the effects of ATX II only when added to the serosal solution. The results suggest that the regulation of electrolyte transport across the epithelium is at least one function of the mucosal plexus. Stimulation of the neurones within the mucosal plexus leads to the inhibition of electrolyte absorption.(ABSTRACT TRUNCATED AT 400 WORDS)
Development and validation of a stability-indicating, single HPLC method for sacubitril-valsartan and their stereoisomers and identification of forced degradation products using LC-MS/MS

Abstract:The aim of this research work was to develop and validate a stability-indicating, single reversed-phase HPLC method for the separation of five impurities, including enantiomers, diastereomers, and degradation products in sacubitril-valsartan tablets. The method was developed using a Chiralcel OJ-RH column (150 × 4.6 mm, 5 μm) at 45°C with a gradient program of (T/%B) 0.01/25, 10.0/25, 25/38, 37.0/45, 39.0/25, and 45.0/25 at a flow rate of 0.8 ml/min. Mobile phase A consisted of 1 ml of trifluoroacetic acid in 1000 ml of Milli-Q water. Mobile phase B consisted of 1 ml of trifluoroacetic acid in a mixture of acetonitrile and methanol in the ratio of 950:50 (v/v). Sacubitril, valsartan, and their five impurities were monitored at 254 nm. Degradation was not observed when sacubitril-valsartan was subjected to heat, light, hydrolytic, and oxidation conditions. In acid degradation study (1 N HCl/60°C/2 h) impurity 1 (m/z 383.44) was formed, and in base degradation study (0.1 N NaOH/40°C/1 h) impurities 1 and 5 (m/z 265.35) were formed; both impurities were confirmed using LC-MS. The degradation products, enantiomers, and diastereomers were well separated from sacubitril and valsartan, proving the stability-indicating power of the method. The developed method was validated per the International Council for Harmonisation of Technical Requirements for Pharmaceuticals for Human Use guidelines. The inter- and intra-day percentage relative standard deviation for sacubitril, valsartan, and their five impurities was less than 5.2%, recovery of the five impurities was between 93 and 105%, and linearity was ≥0.999. The limit of detection was 0.030-0.048 μg/ml, and the limit of quantification was 0.100-0.160 μg/ml.

Abstract:Objective: The objective of the present work was to develop and validate the stability-indicating method for the simultaneous estimation of itraconazole and terbinafine HCl in bulk and pharmaceutical tablet dosage form by reversed-phase high-performance liquid chromatography (HPLC). This combination of drugs is not reported for simultaneous HPLC analysis as of now. Methods: The analysis of the developed method was carried on Shimadzu LC Prominence-i 2030 model with Lab Solution software and the separation was done on Shim-pack C18 GIST (250 mm×50 mm, 5 μm) column with a flow rate of 1.2 ml/min and run time of 12 min. The injection volume was 10 μl and mobile phase consisted of acetonitrile and 0.1% triethylamine in the ratio of 90:10 and 225 nm was used as a detection wavelength. Results: The retention time was found to be 3.464 min and 8.705 min for itraconazole and terbinafine HCl, respectively. The calibration curve was found to be linear and r2 values were 0.9989 and 0.9995 for itraconazole and terbinafine HCl, respectively. Conclusion: The stability-indicating method was developed by subjecting itraconazole and terbinafine HCl marketed formulation to various stress conditions such as acidic, basic, oxidative, thermal, and water hydrolysis degradation conditions and the degraded product peaks were well resolved from sample peaks.
Stability-indicating RP-LC method for determination of azilsartan medoxomil and chlorthalidone in pharmaceutical dosage forms: application to degradation kinetics

Abstract:A RP-LC method was developed and validated for simultaneous determination of the active components, azilsartan medoxomil (AZL) and chlorthalidone (CLT), in their novel antihypertensive combined recipe. The chromatographic separation was achieved on an Eclipse XDB-C18 (4.6 × 150 mm, 5 μm) column using a mobile phase consisting of methanol/potassium hydrogen phosphate buffer (pH 8, 0.05 M) (40:60, v/v) in isocratic mode. The flow rate was maintained at 0.8 mL min−1 at ambient temperature. Detection was carried out at 210 nm. The method was validated according to the ICH guidelines. Linearity, accuracy, and precision were satisfactory over the concentration range of 5.0–50.0 and 2.5–25.0 μg mL−1 for AZL and CLT, respectively (r2 = 0.9999). LODs for AZL and CLT were 0.90 and 0.32 μg mL−1, whereas LOQs were 2.72 and 0.98 μg mL−1, respectively. Both drugs were subjected to forced degradation studies under hydrolysis (neutral, acidic, and alkaline), oxidative, and photolytic extensive stress conditions. The proposed method is stability indicating by the resolution of the investigated drugs from their degradation products. Moreover, the kinetics of the acidic degradation of AZL as well as the kinetics of the alkaline degradation of CLT were investigated. Arrhenius plots were constructed and the apparent first-order rate constants, half-life times, shelf-life times, and the activation energies of the degradation processes were calculated. The method was successfully applied for the determination of the studied drugs simultaneously in their coformulated tablet. The developed method is specific and stability indicating for the quality control and routine analysis of the cited medications in their pharmaceutical preparations.
Stability‐indicating RP‐HPLC method development and validation for the quantification of amlodipine besylate and valsartan tablets in solid oral dosage form

Abstract:The present study discusses the development of simple, rapid, specific, precision, accuracy, stability indicating the HPLC method for the analysis of amlodipine besylate and valsartan tablet dosage form. The chromatographic separation was achieved using phosphate buffer with 1% triethyl amine (pH 3.0) as mobile phase‐A and mixed Methanol and buffer in the ratio of (65:35)(v/v) as mobile phase‐B. The detection of components was made at 237 nm for amlodipine besylate and valsartan. Analytical techniques should enrich sensitivity and specificity for the estimation of pharmaceutical drug products. Evaluated stress studies under different types of ICH conditions. The optimized HPLC method was validated as per the current ICH guidelines. The validated HPLC method was obtained highly specific with linearity ranging between 25 and 200 μgmL−1 of amlodipine besylate and 40–320 μgmL−1 of valsartan and both components correlation coefficient was > 0.999. The method showed high accuracy more than 97%. In stress studies, amlodipine besylate and valsartan were found to be sensitive to acid stress conditions and oxidation stress conditions. The method was found to be suitable for the quality control of amlodipine besylate and valsartan in the tablet as well as in stability‐indicating studies. The method was applied to the analysis of stability samples.

Abstract:A simple, precise, accurate High-performance thin-layer chromatography (HPTLC) method has been developed and validated for the estimation of escitalopram oxalate (ESC) and flupentixol dihydrochloride (FLU) in pharmaceutical formulation. Separation was achieved on an aluminum HPTLC plate coated with a 0.2 mm layer of silica gel 60 F254 with chloroform: methanol 4:6 (v/v) as mobile phase. The densitometry analysis was carried out in absorbance mode at 254 nm. The Rf values of escitalopram and flupentixol were 0.24 and 0.44 respectively. The linearity of the method was achieved in the range 3-7 μg/spot for ESC with correlation coefficient value 0.998 and 0.3-0.7μg/spot for FLU with correlation coefficient value 0.999. The developed method was validated according to ICH guidelines and the results were statistically validated. The limit of detection and quantification for escitalopram oxalate and flupentixol dihydrochloride was found to be 500 ng/spot and 50 ng/spot, 1000 ng/spot, and 100 ng/spot respectively.

Abstract:Two stability-indicating chromatographic methods namely, reverse-phase high-performance liquid chromatography (RP-HPLC) and high-performance thin-layer chromatography (HPTLC) were developed and validated for the determination of azilsartan medoxomil from pharmaceutical dosage form in presence of its degradation products as per ICH guideline. In RP-HPLC, separation was performed with Hiber® C-18 column (250 mm X 4.6 mm, 5 μm), using mobile phase methanol: acetonitrile: water (88:8:4 v/v/v) at a flow rate of 0.5 ml/min. The analyte was detected at 254 nm over a concentration range of 20 - 70 μg/ml with correlation coefficient of 0.999. In HPTLC, separation was carried out with silica gel G60 F254 aluminum sheet using acetone: toluene: ammonia (8.2:1.7:0.1 v/v/v) as a developing system. Linearity of the proposed HPTLC method was found 500 - 900 ng/band and correlation coefficient 0.9970 at 254 nm. Besides, stress degradation study was also carried out wherein degraded product peaks were well resolved from the analyte peak with difference in their retention time and retardation factor in RP-HPLC and HPTLC methods respectively. Statistical analysis proved that there were no significant differences between the two validated methods. The proposed stability-indicating methods can be applied for the analysis of azilsartan medoxomil in pharmaceutical formulations for quality control.
Development and Validation of Rapid Stability-Indicating RP-HPLC Method for Assay and Related Substances of Solifenacin Succinate

Abstract:A simple, inexpensive, rapid and novel stability-indicating reversed-phase high-performance liquid chromatographic method employing Zorbax-CN column has been developed and validated for the determination of both, assay and related substances of Solifenacin Succinate (SOL) drug substance and in its drug product. Forced degradation study of SOL was performed at acid, base, oxidative, photolytic and thermal stress conditions. Significant degradation was observed only in acidic and basic conditions. Four process and degradation impurities and have been identified by LC–MS and characterized by 1H NMR and FT-IR spectroscopy. The baseline separation of the impurities was achieved by gradient elution using phosphate buffer (pH 3.5) and a mixture of solvents (acetonitrile, methanol, and water), as mobile phase at a flow rate of 1.0 mL min−1. All impurities were quantified using a photodiode array detector (PDA) at 220 nm. The developed RP-HPLC method was validated in agreement with ICH requirements. The forced degradation studies proved the stability-indicating power of the method and hence, the validated method is useful for the intended use in routine analysis of SOL as bulk drug and in respective dosage forms. The salient feature of the presented method could be applied for assay and related substances determination of SOL in pharmaceutical laboratories and industries.

Abstract:Aims: Metformin HCl, Dapagliflozin and Saxagliptin is a new drug combination for the treatment of Diabetes Mellitus which is one of the oldest and lethal diseases of mankind. Aim of the research work was to develop and validate novel, rapid, sensitive, specific, robust stability indicating analytical method for simultaneous estimation of: Metformin HCl, Dapagliflozin and Saxagliptin in pharmaceutical dosage form as fixed dose formulation. Study Design: Method development and validation was performed as recommended in ICH guideline “Validation of analytical procedures: Test and Methodology Q2 (R1)”. Methodology: Method develop with chromatographic parameters as C18 column (250mm×4.6 mm, 5mm particle size), HPLC system with PDA detector and mobile phase contained a mixture of Phosphate Buffer pH 3.5 and Acetonitrile (80:20 v/v) + 1 ml triethylamine per 100 ml mobile phase. The flow rate was set to 1 ml/min with responses measured at 265 nm, injection volume was 20 μl, and run time of 15 mins. Results: The retention time of Metformin HCl, Dapagliflozin and Saxagliptin was 5.8 min, 6.8 mins and 8.4 min respectively with resolution of 3.5 between Metformin HCl and Dapagliflozin and 4.5 between Dapagliflozin and Saxagliptin. Linearity was established in the range of 250-1500 μg/ml for Metformin HCl, 1.25-7.5 μg/ml for Dapagliflozin and Saxagliptin with correlation coefficients more than 0.999. The percentage recoveries were between 98.39-101.66 for Metformin HCl, 99.01-101.77 for Dapagliflozin and 98.88-101.87 for Saxagliptin Validation parameters were evaluated according to the International Conference on Harmonization (ICH) Q2 R1 guidelines. The forced degradation studies were performed by using HCl, NaOH, H2O2, thermal and UV Radiation. The developed method was successfully applied for the quantification and hyphenated instrumental analysis. Conclusion: Significance of developed method is that it can be utilize for routine or unknown sample analysis of assay of Metformin HCl, Dapagliflozin and Saxagliptin in pharmaceutical dosage form developed by various Pharmaceutical Industry.

Abstract:An efficient, cost effective, rapid reverse phase high-performance liquid chromatographic method was developed and validated for the determination of Ezetimibe and Simvastatin in pharmaceutical dosage forms. Chromatography was carried out by using X-terra C18, 150 x 4.6mm, 3.5μ or equivalent internal diameter with a mixture of 0.5 mL glacial acetic acid in 2000 mL water as mobile phase A and Acetonitrile as mobile phase B. Analytical method validation parameters such as specificity, linearity, precision, accuracy, solution stability and robustness, limit of detection (LOD) and limit of quantification (LOQ) was done. The correlation coefficient was found to be linear for each analyte in the desired concentration range. The average recovery was found with range of 96.7 and 107.7 for Ezetimibe and Simvastatin and their respective impurities respectively. The proposed method is highly sensitive, precise and accurate, which was evident from the LOD value range of minimum 0.135 ppm for Ezetimibe and 0.356 ppm for Lovastatin & Epilovastatin for Simvastatin impurities. Hence the present method can be applied successfully for the quantification of finished dosage form in the combined formulations of Ezetimibe and Simvastatin.

Abstract:A stability indicating reversed-phase high-performance liquid-chromatographic method for analysis of empagliflozin was developed and validated as per the ICH guidelines. Statistical design of experiment was applied for optimization, where independent variables used were methanol proportions in mobile phase and flow rate. Experiment was carried out on an analytical reversed phase column Cosmosil C18 (250 × 4.6 mm, 5 μm). Based on the results obtained from these studies, suitable mobile phase with appropriate composition was selected and utilized for method development applying DoE approach. The mobile phase used was methanol: water (85:15 V/V). The flow rate was set at 0.8 mL min-1 and UV detection was carried out at 225 nm. The retention time of empagliflozin was found to be 4.259 min. The lower solvent consumption along with the short analytical run time (≤05 minute) provides a cost effective and environment friendly chromatographic procedure. The measured signal was shown to be precise, accurate and linear over the concentration range tested (10-50 μg mL-1) with a correlation coefficient of 0.9999. Thus, the proposed methodology is rapid, selective and requires simple sample preparation steps and represents a good procedure for analysis of empagliflozin. Central Composite Design (CCD) was used for method development of empagliflozin. Two factors were selected with eight center points and response of empagliflozin was measured in terms of retention time which dependent on two factors namely, methanol content in mobile phase and flow rate. CCD was effective means in optimization of HPLC for analysis of empagliflozin in pharmaceutical formulation. The stability of the drug was examined over different stress conditions as per International Conference on Harmonization (ICH) guidelines. Results obtained from the force degradation studies indicated that the developed method is appropriate for stability studies.

Abstract:Aims: Canagliflozin and Metformin HCl is a new drug combination for the treatment of Diabetes Mellitus which is one of the oldest and lethal diseases of the mankind. Aim of the research work was to develop and validate novel, rapid, sensitive, specific, robust stability indicating analytical method for the simultaneous estimation of Canagliflozin and Metformin HCl in the pharmaceutical dosage form as fixed dose formulation. Study Design: Method development and validation was performed as recommended in ICH guideline “Validation of analytical procedures: Test and Methodology Q2(R1)”. Methodology: Method develop with chromatographic parameters as C18 column (250mm×4.6 mm, 5mm particle size), HPLC system with PDA detector and mobile phase contained a mixture of Phosphate Buffer pH 5.0 and Acetonitrile (60:40 v/v). The flow rate was set to 1ml/min with responses measured at 290 nm, injection volume was 20 μl, and run time of 15 mins. Results: The retention time of Metformin Hydrochloride and Canagliflozin was 5.4 min and 7.6 min respectively with resolution of 7.0. Linearity was established in the range of 10-30 μg/ml for Metformin Hydrochloride and 0.5-1.5 μg/ml for Canagliflozin with correlation coefficients more than 0.999. The percentage recoveries were between (98.62-101.22%) and (98.68-101.27%) for Metformin Hydrochloride and Canagliflozin respectively. Validation parameters were evaluated according to the International Conference on Harmonization (ICH) Q2 R1 guidelines. The forced degradation studies were performed by using HCl, NaOH, H2O2, thermal and UV radiation. The developed method was successfully applied for the quantification and hyphenated instrumental analysis. Conclusion: Significance of developed method is that it can be utilize for routine or unknown sample analysis of assay of Metformin HCl and Canagliflozin in pharmaceutical dosage form developed by various Pharmaceutical Industry.

Abstract:Abstract Background A new, sensitive, suitable, clear, accurate, and robust reversed-phase high-performance liquid chromatography (RP-HPLC) method for the determination of brexpiprazole in bulk drug and tablet formulation was developed and validated in this research. Surface methodology was used to optimize the data, with a three-level Box-Behnken design. Methanol concentration in the mobile phase, flow rate, and pH were chosen as the three variables. The separation was performed using an HPLC method with a UV detector and Openlab EZchrom program, as well as a Water spherisorb C 18 column (100 mm × 4.6; 5m). Acetonitrile was pumped at a flow rate of 1.0 mL/min with a 10 mM phosphate buffer balanced to a pH of 2.50.05 by diluted OPA (65:35% v/v) and detected at 216 nm. Result The developed RP-HPLC method yielded a suitable retention time for brexpiprazole of 4.22 min, which was optimized using the Design Expert-12 software. The linearity of the established method was verified with a correlation coefficient (r 2 ) of 0.999 over the concentration range of 5.05–75.75 g/mL. For API and formulation, the percent assay was 99.46% and 100.91%, respectively. The percentage RSD for the method’s precision was found to be less than 2.0%. The percentage recoveries were discovered to be between 99.38 and 101.07%. 0.64 μg/mL and 1.95 μg/mL were found to be the LOD and LOQ, respectively. Conclusion The developed and validated RP-HPLC system takes less time and can be used in the industry for routine quality control/analysis of bulk drug and marketed brexpiprazole products. Abstract:A sensitive, specific, and accurate stability-indicating high-performance thin-layer chromatographic method for the analysis of ambroxol hydrochloride and doxofylline, for both bulk drug and formulation, was developed and validated according to the International Conference on Harmonization (ICH) guidelines. The densitometric analysis of both drugs was carried out in the absorbance mode at 276 nm using diethyl ether—n-butanol—ammonia (9:0.9:0.1, v/v) as the solvent system. This system was found to give compact spots for ambroxol hydrochloride at RF of 0.74 ± 0.01 and doxofylline at 0.41 ± 0.01. Moreover, both drugs were subjected to acid and alkali hydrolysis, oxidation, and photodegradation. Also, the degraded products were well resolved from the pure drug with different RF values. Linearity was found in the range of 20–100 and 100–500 ng band−1 for ambroxol hydrochloride and doxofylline. The limit of detection (LOD) and limit of quantitation (LOQ) for ambroxol hydrochloride and doxofylline were 1.17 and 3.57 ng band−1 and 30.68 and 92.97 ng band−1, respectively. “Bartlett’s test” and “Lack of fit” applied on peak area for linearity additionally proved validity of the linearity of the developed method. Good accuracy and precision were obtained as revealed from percent relative standard deviation (% RSD) less than 2. Moreover, robustness testing was performed applying fractional factorial design, 24-1. All the four factors: volume of diethyl ether, volume of n-butanol, solvent front, and chamber saturation time, evaluated in the robustness testing, were found to have an insignificant effect on the retention factor. Similarly, no interference was observed from common excipients in tablet formulation as well as degradation product, indicating specificity of the method. As the method could effectively separate both drugs from their degradation products, it can be used as a stability-indicating method.

Abstract:This study validates a stability-indicating LC method for detecting organic impurities in the chlorzoxazone dosage form. Using a Waters X-Select R HSS T3 analytical column, mobile phase of it was made by mixing of water, methanol, and glacial acetic acid in the ratio of 700:300:10 (v/v/v). The drug product and drug substance were subjected to the stress conditions such as acid, base, oxidation, heat, and photolysis as per the recommendations of the International Conference on Harmonization (Q2) methodology. The study revealed the susceptibility of 4-chloro-2-aminophenol to alkaline environments, emphasizing peak homogeneity and stability. The method verification, per ICH guidelines and USP<1225>, established precision, specificity, linearity, accuracy, and robustness for quality control. The mean impurity recovery ranged from 95.5% to 105.2%, the correlation coefficient (r) was greater than 1.000, and the RSD values (n = 6) ranged from 0.6% to 5.1% across the LOQ-150% ranges. Full-factorial design tested final method conditions, evaluating multiple parameters concurrently. Graphical optimization within the design space defined strong method requirements, ensuring consistent and reliable outcomes. The study develops and validates chlorzoxazone stability-indicating methods, employing advanced statistical approaches like design of experiments and factorial design, with resilient conditions established through graphical optimization of the design space.
