Abstract:A new eremophilane sesquiterpene, capnodiumone (1), along with five known eremophilane sesquiterpenes (2-6) and eight other compounds (7-14), have been isolated from a mangrove-associated fungus Capnodium sp. SZ-F22. The chemical structures were elucidated on the basis of extensive spectroscopic analysis. The broth extract of the fungus exhibited a good inhibitory effect on the mycelium growth against Fusarium graminearum at 100g/mL, however, all the 14 compounds showed no expected antifungal activity. Abstract:Azalomycins F5a (1), F4a (2) and F3a (3) are three 36-membered polyhydroxyl macrolides with remarkable antifungal activity. By comparing the NMR spectral data of 1 and its degradation products with those of several universal NMR databases and identical fragments of known compounds, the relative configuration of 1 was assigned for the first time except that the stereochemistry of stereoclusters C6–C11 to C14–C36 was undefined. Moreover, the relative configurations of 2 and 3 were respectively established by comparing their NMR spectral data with those of 1.
[Depression among students of the Medical Faculty and doctors in Mostar].

Abstract:UNLABELLED To assess out how many students and doctors have a mild, a moderate and a gravely depressive episode. SUBJECTS The research was carried out among the students and doctors of the Medical Faculty in Mostar, Bosnia and Herzegovina, in March 2003. Out of the total number of 255 medical students and 115 doctors of, 200 (78.4%) medical students and 90 (78.3%) doctors participated in the research. The were analysis in accordance with DSM-IV criteria. RESULTS One hundred and twenty (41.4%) participants experienced mildly depressive episode, and 52 (17.9%) participants experienced moderately depressive episode. Medical students were more depressive (66.5%) than the doctors (43.3%, p=0.001). A majority of the mildly depressive, and in particular the moderately depressive respondents, reported about a mild and moderate, respectively, intensity of disorder of mood loss of interest for the usual activities, fatigue, and the loss of energy. CONCLUSION Medical students and doctors constitute a vulnerable professional group, having a high percentage of depressive disorders.

Abstract:The endophytic fungus Coniothyrium sp. was isolated from leaves of Quercus robur. Fermentation of this fungus on solid rice medium yielded two new furoic acid derivatives ( 1 and 2 ) and two additional known compounds. The structures of the new compounds were determined by extensive analysis of 1D and 2D nuclear magnetic resonance spectra as well as high‐resolution mass spectrometry data. Compound 1 , containing three aromatic chromophores attached by rotatable sigma bonds and a chirality center in benzylic position, was found to be a scalemic mixture with an excess of the (S) enantiomer, the absolute configuration of which was elucidated as by the solution time‐dependent density functional theory‐electronic circular dichroism approach. The ωB97X/TZVP PCM/MeCN and SOGGA11‐X/TZVP SMD/MeCN methods were used for geometry reoptimization to reproduce the solution conformational ensemble. All isolated compounds were tested for their cytotoxicity but proved to be inactive.

Abstract:Chemical investigation of the endophytic fungus Fusarium oxysporum isolated from fruits of Drepanocarpus lunatus afforded eight new fusaric acid derivatives, fusaricates A-G, 1-7, and 10-hydroxy-11-chlorofusaric acid, 8, along with four known compounds. Their structures were elucidated by one- and two-dimensional NMR as well as MS data and by comparison with the literature. The absolute configurations of fusaricates C-E, 3-5, were determined using chiral GC-MS. Fusaricates A-G, 1-7, represent the first examples of fusaric acid linked to a polyalcohol moiety via an ester bond. All isolated fusaric acid derivatives 1-8 showed significant phytotoxicity to leaves of barley.

Abstract:In order to purify enough material for establishing the absolute stereochemistry of the new antifungal metabolite 3,6,8-trihydroxy-3-[3,5-dimethyl-2-oxo-3(E)-heptenyl]-2,3-diliydronaphthalen-1(4H)-one produced by Keissleriella sp., a marine filamentous fungus (strain number: YS 4108) [1], a repeated growth and fractionation of the fungal culture was performed to give instead a new antimicrobial metabolite, keisslone (1), the structure of which was elucidated on the basis of spectral analyses including homo- and hetero-nuclear correlation NMR experiments (HMQC, COSY, NOESY and HMBC). The absolute configuration of metabolite 1 was determined mainly by its CD data and NOESY spectrum. The compound 1 was shown to be inhibitory against the human pathogenic fungi Candida albicans, Trichophyton rubrum and Aspergillus niger with MICs of 50, 70, 40 mug/mL, respectively.

Abstract:A new sesquiterpenoid derivative, named aspergiketone (1), along with seven known compounds (2-8) were isolated from the coastal saline soil fungus Aspergillus fumigatus. Their structures were elucidated by spectroscopic analysis, and by comparison of experimental and reported data. The absolute configuration of compound 1 was defined by X-ray diffraction analysis. Compound 1 was cytotoxic towards HL-60 and A549 cell lines with IC50 values of 12.4 and 22.1 mu M, respectively.

Abstract:The marine-derived fungus Aspergillus falconensis, isolated from sediment collected from the Canyon at Dahab, Red Sea, yielded two new chlorinated azaphilones, falconensins O and P (1 and 2) in addition to four known azaphilone derivatives (3-6) following fermentation of the fungus on solid rice medium containing 3.5% NaCl. Replacing NaCl with 3.5% NaBr induced accumulation of three additional new azaphilones, falconensins Q-S (7-9) including two brominated derivatives (7 and 8) together with three known analogues (10-12). The structures of the new compounds were elucidated by 1D and 2D NMR spectroscopy and HRESIMS data as well as by comparison with the literature. The absolute configuration of the azaphilone derivatives was established based on single-crystal X-ray diffraction analysis of 5, comparison of NMR data and optical rotations as well as on biogenetic considerations. Compounds 1, 3-9, and 11 showed NF-kappa B inhibitory activity against the triple negative breast cancer cell line MDA-MB-231 with IC50 values ranging from 11.9 to 72.0 mu M.

Abstract:Nine undescribed side chain containing azaphilones, pestaphilones A-I, were isolated from the Anoectochilus roxburghii endophytic fungus Pestalotiopsis oxyanthi. The structures of these isolates were identified by spectroscopic data, electronic circular dichroism (ECD) calculations and comparisons, quantum-chemical C-13 NMR calculations with DP4+ probability analysis, Rh-2(OCOCF3)(4)-induced ECD, acetonide formation, selective oxidation reaction and X-ray crystallographic data. Structurally, pestaphilones A-I were the first azaphilones characteristically formed via a methyl group at C-9 in the C-7 side chain. More importantly, a selective oxidation reaction was firstly set up to resolve the absolute configuration of flexible side chain containing azaphilones, and an acetonide formation based Rh-2(OCOCF3)(4)-induced ECD experiment was performed to identify the configurations of the oxygenated pyranoquinone core in the azaphilones. In bioassay, pestaphilones A-F displayed potential immunosuppressive activity in concanavalin A (Con A)-induced T lymphocyte proliferation, with IC50 values ranging from (9.36 +/- 1.14) mu M to (35.21 +/- 3.25) mu M.

Abstract:The marine-derived fungus Penicillium sp. (strain IO1) isolated from the Mediterranean sponge Ircinia oros yielded a new fusarielin analogue (1) together with the known compounds griseofulvin (4) and dechlorogriseofulvin (5). The structure of 1 was unambiguously elucidated by comprehensive spectroscopic analysis (1D and 2D NMR, and mass spectrometry) as well as by comparison with the literature, while the absolute configuration of 1 was determined on the basis of TDDFT ECD calculations. A further Penicillium sp. (strain IO2) that was isolated from the same sponge I. oros yielded the known compounds dehydrocurvularin (6), curvularin (7), and trichodimerol (8). Co-cultivation of both Penicillium strains (IO1 and IO2) was found to induce the accumulation of the known norlichexanthone (2) and monocerin (3) that were not detected in either of the two axenic fungal controls. Compounds 3 and 6 showed pronounced cytotoxicity against the murine lymphoma (L5178Y) cell line with IC50 values of 8.4 and 4.7μM, respectively.

Abstract:Three new ophiobolin sesterterpenoids, (6R)-16,17,21,21-O-tetrahydroophiobolin G (1), (6R)-16,17-dihydroophiobolin H (2), and (5S,6S)-16,17-dihydroophiobolin H (3), and three new farnesylated phthalide derivatives farnesylemefuranones D–F (9–11), along with five known ophiobolin analogues (4–8), were isolated and identified from the culture extract of Aspergillus insuetus SD-512, a deep-sea-derived fungus obtained from cold seep sediments collected at a depth of 1331 m. Among them, compounds 9–11 are rare examples of phthalide derivatives linked with farnesyl moieties via ether bonds. Their structures were established on the basis of detailed interpretation of the NMR spectroscopic and mass spectrometric data. X-ray crystallographic analysis, ECD calculations, and DP4+ probability analysis were performed to confirm the structures and establish the relative and absolute configurations of compounds 1–4. Compounds 3 and 9–11 showed broad-spectrum antibacterial activities, and differences in potencies could be assigned to structural modifications. Abstract:Two new fusarochromanone derivatives, deacetylfusarochromene (1) and deacetamidofusarochrom-2′,3-diene (2), along with the previously reported metabolites fusarochromanone TDP-2 (3), fusarochromene (4), 2,2-dimethyl-5-amino-6-(2′E-ene-4′-hydroxylbutyryl)-4-chromone (5), fusarochromanone (6), (−)-chrysogine (7), and equisetin (8), were isolated from the marine fungus Fusarium equiseti UBOCC-A-117302. The structures of the compounds were determined by extensive spectrometric (HRMS) and spectroscopic (1D and 2D NMR) analyses, as well as specific rotation. Among them, 2 and 5 showed inhibition of three protein kinases with IC50 values ranging from 1.42 to 25.48 μM. Cytotoxicity and antimicrobial activity of all isolated compounds were also evaluated. Six fusarochromanone derivatives (1–6) exhibited diverse activities against three cell lines, RPE-1, HCT-116, and U2OS (IC50 values ranging from 0.058 to 84.380 μM). Equisetin (8) showed bactericidal activities against Bacillus cereus and Listeria monocytogenes (MBC values of 7.8 and 31.25 μM, respectively), and bacteriostatic activity against Enterococcus faecalis (MIC value of 31.25 μM). Compounds 2 and 4 showed bacteriostatic activities against Listeria monocytogenes (MIC of 125 μM).

Abstract:Simplifusidic acids A-K (1-11), 11 new fusidane-type nortriterpenoids, were isolated from the marine-derived fungus Simplicillium sp. SCSIO 41513. Compound 1 possessed an unprecedented fusidane triterpene skeleton with a 6/6/7/5/5 polycyclic system. Their structures were elucidated by spectroscopic methods, and their absolute configurations were further determined by quantum chemical calculations of ECD spectra, comparison of experimental ECD spectra, and single-crystal X-ray diffraction analysis. Compound 9 showed strong antibacterial activity toward Staphylococcus aureus with an MIC value of 0.078 μg/mL. Their structure-bioactivity relationship was also discussed.

Abstract:Co-cultivation of the endophytic fungus Fusarium tricinctum with Streptomyces lividans on solid rice medium led to the production of four new naphthoquinone dimers, fusatricinones A–D (1–4), and a new lateropyrone derivative, dihydrolateropyrone (5), that were not detected in axenic fungal controls. In addition, four known cryptic compounds, zearalenone (7), (−)-citreoisocoumarin (8), macrocarpon C (9) and 7-hydroxy-2-(2-hydroxypropyl)-5-methylchromone (10), that were likewise undetectable in extracts from fungal controls, were obtained from the co-culture extracts. The known antibiotically active compound lateropyrone (6), the depsipeptides enniatins B (11), B1 (12) and A1 (13), and the lipopeptide fusaristatin A (14), that were present in axenic fungal controls and in co-culture extracts, were upregulated in the latter. The structures of the new compounds were elucidated by 1D and 2D NMR spectra as well as by HRESIMS data. The relative and absolute configuration of dihydrolateropyrone (5) was elucidated by TDDFT-ECD calculations.

Abstract:Two new cyclic depsipeptides, W493 C (1) and D (2), along with two known derivatives W493 A (3) and B (4) were obtained from the endophytic fungus Fusarium sp. isolated from the Mangrove plant Ceriops tagal. The structures of the new compounds were determined on the basis of one- and two dimensional NMR and high-resolution mass spectroscopic data. The absolute configurations of the amino acid residues of 1 and 2 were confirmed by application of Marfey's method. W493 A (3) and B (4) exhibited moderate activity against the fungus Cladosporium cladosporiodes and weak antitumor activity against the human ovarian cancer cell line A2780.

Abstract:A dihydronaphthalene-2,6-dione derivative, isolated from Phoma sp. LN-16, an endophytic fungus associated with Melia azedarach, was identified by spectroscopic data as a previously reported compound, botryosphaeridione (1). The relative configuration was reassigned by quantum chemical analysis of experimental 13C NMR and NOESY data. The first unequivocal assignment of its absolute configuration was made by using circular dichroism spectra and computational model B3LYP/aug-cc-pVDZ as (−)-(5R,6S)-1, and was also established by means of X-ray diffraction. Also, this is the first time that a trinor-eremophilane sesquiterpene is described for Phoma genus.

Abstract:Chemical investigation of the fermentation extract of the mangrove endophytic fungus Aspergillus sp. GXNU-A1, isolated from Acanthus ilicifolius L., discovered an undescribed pair of enantiomers (asperphenyltones A and B (±1)), together with four previously described metabolites: nodulisporol (2), isosclerone (3), 2,3,4-trihydroxy-6-(hydroxymethyl)-5-methylbenzyl alcohol (4), and 4,6-dihydroxy-5-methoxy-7-methyl-1,3-dihydroisobenzofuran (5). Analyses of the 1D and 2D NMR spectroscopic data of the compounds supported their structural assignments. The presence of the asperphenyltones A and B, which are a pair of enantiomers, was established by HR-ESI-MS, 1D and 2D NMR data and confirmed by single-crystal X-ray diffraction analysis. Metabolites 1-5 were evaluated for their anti-inflammatory effects on the production of nitric oxide (NO), and 1, 3, and 4 showed significant potential inhibitory activities against NO production in activated macrophages with IC50 values of 26-40 μM, respectively.
