Abstract:Chemical investigation of a co-culture of two endophytic fungi Aspergillus fumigatus D and Fusarium oxysporum R1 from two traditional medicinal plants, Edgeworthia chrysantha Lindl. and Rumex madaio Makino, led to isolation of a new amide 1 and six known compounds, including neovasinin (2), neovasifuranone B (3), N-(2-phenylethyl)acetamide (4), α-linolenic acid (5), α-elaeostearic acid (6), palmitoleic acid (7). On the basis of extensive spectroscopic analysis including 1D and 2D NMR, HR-ESI-MS and optical rotation measurement as well as comparison of literature data, chemical structure of 1 was unambiguous elucidated as (S, E)-methyl-2-(2,4-dimethylhex-2-enamido)acetate. Bioassay results indicated that none of these compounds exhibited strong inhibitory effect on three human pathogens Escherichia coli, Staphyloccocus aureus and Candida albicans with MIC values of ≥25 μM.

Abstract:Fungi are important resource for the discovery of novel bioactive natural products. This study investigated the metabolites produced by Mariana-Trench-associated fungus Aspergillus sp. SY2601 in EY liquid and rice solid media, resulting in the isolation and structure determination of 28 metabolites, including five new compounds, asperindopiperazines A–C (1–3), 5-methoxy-8,9-dihydroxy-8,9-deoxyaspyrone (21), and 12S-aspertetranone D (26). Structures of the new compounds were elucidated based on extensive NMR spectral analyses, HRESIMS data, optical rotation, ECD, and 13C NMR calculations. The new compound 12S-aspertetranone D (26) exhibited antibacterial activity against both methicillin-resistant Staphylococcus aureus and Escherichia coli with MIC values of 3.75 and 5 μg/mL, respectively.

Abstract:Four dimeric diketopiperazine stereoisomers (1-4) including two new ones (1-2) had been isolated from the culture broth of one marine-derived fungus Aspergillus sp. Z3, which was found in the gut of a marine isopod Ligia exotica. The planner structures and absolute configurations of the new compounds were determined by combination of NMR, HRESIMS, electronic circular dichroism calculation, Marfey's method as well as single-crystal X-ray diffraction. Their cytotoxicity against the prostate cancer PC3 cell line was assayed by the MTT method.

Abstract:The endophytic fungus Coniothyrium sp. was isolated from leaves of Quercus robur. Fermentation of this fungus on solid rice medium yielded two new furoic acid derivatives ( 1 and 2 ) and two additional known compounds. The structures of the new compounds were determined by extensive analysis of 1D and 2D nuclear magnetic resonance spectra as well as high‐resolution mass spectrometry data. Compound 1 , containing three aromatic chromophores attached by rotatable sigma bonds and a chirality center in benzylic position, was found to be a scalemic mixture with an excess of the (S) enantiomer, the absolute configuration of which was elucidated as by the solution time‐dependent density functional theory‐electronic circular dichroism approach. The ωB97X/TZVP PCM/MeCN and SOGGA11‐X/TZVP SMD/MeCN methods were used for geometry reoptimization to reproduce the solution conformational ensemble. All isolated compounds were tested for their cytotoxicity but proved to be inactive.

Abstract:Marine actinomycetes are known for their production of remarkable organic molecules, particularly those featuring polyoxygenated long-chain backbones. Determining the absolute configurations of these compounds remains a challenging task even today. In this study, we successfully established the planar structures and absolute configurations of two highly flexible amide alkaloids from Streptomyces sp. WU20: kueishanamides A (1) and B (2). These compounds possess a C-13 linear backbone and each contains five stereogenic carbon centers. Our approach involved a combination of spectroscopic and computational methods, including J-based configurational analysis and VCD calculations, ensuring the unambiguous determination of their configurations. Kueishanamide A (1) and kueishanamide B (2) showed moderate antifungal activity against pathogenic fungus Crytococcus neoformans, with MIC values of 25 mu g/mL each.

Abstract:Six new sesquiterpenes, fusarchlamols A-F ( 1 , 2 , 4 – 7 ); one new natural product of sesquiterpenoid, methyltricinonoate ( 3 ); and ten known compounds were found from Fusarium sp. cultured in two different media by the one strain many compounds strategy. The compounds ( 1 , 2 , and 4 – 11 ) were isolated from Fusarium sp. in PDB medium, and compounds ( 3–5 , 8 , and 10 – 17 ) were discovered from Fusarium sp. in coffee medium. Additionally, the configuration of 8 was first reported in the research by Mosher's method. The structures were established by 1D, 2D NMR, mass spectrometry, calculated ECD spectra and Mosher's method. Compounds 1 , 2 , 6 / 7 , 12 , and 16 indicated significant antifungal activities against the phytopathogen Alternaria alternata isolated from Coffea arabica with MICs of 1 μg/mL. The investigation on the anti-phytopathogen activity of metabolites can provide lead compounds for agrochemicals.

Abstract:In the pursuit of discovering new active metabolites from helicosporous hyphomycetes, rice fermentation products of Neohelicosporium griseum were examined. Eight compounds were isolated from this saprophytic fungus, namely vertixanthone ( 1 ), diaportheone A ( 2 ), 1,3,6,8-tetrahydroxyanthraquinone ( 3 ), lecanoric acid ( 4 ), decarboxycitrinone ( 5 ), 6,8-dihydroxy-4-hydroxymethyl-3,5-dimethyl-isochromen-1-one ( 6 ), decarboxyhydroxycitrinone ( 7 ), and ergosterin ( 8 ). The 1D and 2D NMR characteristics of compound 1 in DMSO- d 6 were detailed for the first time. Antimicrobial testing indicated that compounds 1 – 4 exhibited moderate activity against Pseudomonas aeruginosa , with compound 3 also showing weak activity against Staphylococcus aureus . In-vitro cytotoxicity assays revealed that compounds 1 , 3 , and 4 displayed cytotoxic activity against HELA cell lines, with respective IC 50 values of 30.8, 13.7, and 14.1 μM. Compounds 1 , 3 , and 4 also showed significant cytotoxicity against A549 cell lines, with respective IC 50 values of 24.7, 7.4, and 10.3 μM.

Abstract:Under the guidance of MS/MS-based molecular networking and HPLC-UV, two new alkaloid racemates, (±)-17-hydroxybrevianamide N (1) and (±)-N1-methyl-17-hydroxybrevianamide N (2), featuring a rare o-hydroxyphenylalanine residue and an imide subunit, were isolated from a soft-coral-derived Aspergillus sp. fungus. The true natural products (+)-1 and (+)-2 were further monitored and obtained from the freshly prepared EtOAc extracts, while (−)-1 and (−)-2 are artifacts generated during extraction and purification processes. Simultaneously, the structures including absolute configurations of (+)-13S-1, (−)-13R-1, (+)-13S-2, and (−)-13R-2 were elucidated on the basis of comprehensive spectroscopic analysis, ECD calculations, and X-ray diffraction data. Interestingly, basic solution promotes the racemization of (+)-1 and (−)-1, whereas acidic solution suppresses the transformation. Abstract:Terpenoids are known for their diverse structures and broad bioactivities with significant potential in pharmaceutical applications. However, natural products with low yields are usually ignored in traditional chemical analysis. Feature-based molecular networking (FBMN) was developed recently to cluster compounds with similar skeletons, which can highlight trace amounts of unknown compounds. Fusoxypene A is a sesterterpene synthesized by Fusarium oxysporum fusoxypene synthase (FoFS) with a unique 5/6/7/3/5 ring system. In this study, the FoFS-containing biosynthetic gene cluster was identified from F. oxysporum FO14005, and an efficient FBMN-based strategy was established to characterize four new sesterterpenoids, fusoxyordienoid A–D ( 1 – 4 ), based on a small-scale fermentation strategy. A cytochrome P450 monooxygenase, FusB, was found to be involved in the functionalization of fusoxypene A at C-17 and C-24 and responsible for the hydroxylation of fusoxyordienoid A at C-1 and C-8. This study highlights the potential of FBMN as a powerful tool for the discovery and characterization of natural compounds with low abundance.

Abstract:Four new isocoumarins, alternariethers A‐C (1‐3) and alternariester (4) were separated from the fermentation of the fungus Alternaria malorum FL39, purified from Myoporum bontioides. Their structures were ascertained using NMR and HR‐ESI‐MS spectroscopy. For compound 4, the absolute configuration was solved with the help of ECD calculation and the DP4+ method. Compared with the positive control triadimefon, compound 1 showed more potent antifungal effects on Colletotrichum musae . The antifungal effects of compounds 1, 2, and 3 on Fusarium oxysporum and Fusarium graminearum, of compound 4 on F. oxysporum, were equal to those of triadimefon. Except for compound 4 which was inactive against Escherichia coli with O78 serotype, all compounds showed moderate or weak antibacterial activity against Staphylococcus aureus and E. coli with O6 or O78 serotype.

Abstract:The One Strain Many Compounds (OSMAC) method was applied to explore the chemical diversities of secondary metabolites produced by Neosartorya fischeri NRRL 181. Four pyripyropenes 1–4, eight steroids 5–11, and four prenylated indole alkaloids 12–15, were obtained from the fungus cultured in petri dishes containing potato dextrose agar (PDA). 1,7,11-trideacetylpyripyropene A (1) and 1,11-dideacetyl pyripyropene A (2) were obtained and spectroscopically characterized (1D, 2D NMR, and HR-ESI-MS) from a natural source for the first time. It offered a sustainable source of these two compounds, which were usually used as starting materials in preparing pyripyropene derivatives. In addition, as compared with all the other naturally occurring pyripyropenes, 1 and 2 possessed unique acetylation patterns that did not follow the established late-step biosynthetic rules of pyripyropenes. The natural occurrence of 1 and 2 in the fungus implied that the timing and order of hydroxylation and acetylation in the late-step biosynthetic pathway of pyripyropenes remained to be revealed. The isolation and identification of 1–15 indicated that the OSMAC method could remarkably alter the metabolic profile and enrich the chemical diversities of fungal metabolites. Compounds 1–4 exhibited no obvious cytotoxicity against the triple-negative breast cancer cell line MDA-MB-231 as compared with taxol.

Abstract:During a screening for antifungal secondary metabolites, six new mono-/bis-alkenoic acid derivatives ( 2 – 7 ) and one known alkenoic acid derivative ( 1 ) were isolated from an endophytic fungi Scopulariopsis candelabrum . Their chemical structures were identified by 1 H-NMR, 13 C-NMR, 2D NMR, and high-resolution mass spectrometry, as well as comparisons with previously reported literatures. Among them, fusariumesters C‒F ( 2 – 5 ) are bis-alkenoic acid derivatives dimerized by an ester bond, while acetylfusaridioic acid A ( 6 ) and fusaridioic acid D ( 7 ) are alkenoic acid monomers. All the isolates were submitted to an antifungal assay against Candida albicans and the corn pathogen Exserohilum turcicum using the filter paper agar diffusion method. As a result, only compound 1 decorating with β -lactone ring turned out to be active against these two tested fungi. The broth microdilution assay against Candida albicans showed the minimum inhibitory concentration (MIC) value of 1 to be 20 μ g/ml, while the minimum inhibitory concentration value of the positive control (naystatin) was 10 μ g/ml. And the half maximal inhibitory concentration (IC 50 ) value (21.23 μ g/ml) of 1 against Exserohilum turcicum was determined by analyzing its inhibition effect on the mycelial growth, using cycloheximide (IC 50 = 46.70 μ g/ml) as the positive control.

Abstract:Chemical investigation of the fermentation extract of the mangrove endophytic fungus Aspergillus sp. GXNU-A1, isolated from Acanthus ilicifolius L., discovered an undescribed pair of enantiomers (asperphenyltones A and B (±1)), together with four previously described metabolites: nodulisporol (2), isosclerone (3), 2,3,4-trihydroxy-6-(hydroxymethyl)-5-methylbenzyl alcohol (4), and 4,6-dihydroxy-5-methoxy-7-methyl-1,3-dihydroisobenzofuran (5). Analyses of the 1D and 2D NMR spectroscopic data of the compounds supported their structural assignments. The presence of the asperphenyltones A and B, which are a pair of enantiomers, was established by HR-ESI-MS, 1D and 2D NMR data and confirmed by single-crystal X-ray diffraction analysis. Metabolites 1-5 were evaluated for their anti-inflammatory effects on the production of nitric oxide (NO), and 1, 3, and 4 showed significant potential inhibitory activities against NO production in activated macrophages with IC50 values of 26-40 μM, respectively.

Abstract:Fractures of the shaft of the humerus are common injuries. Historically, the treatment of choice has been functional bracing. Recent technical advances such as angular stable plate and nail constructs lead to a shift in the treatment algorithm for this type of fracture. Surgical treatment is recommended in high grade open fractures, multilevel injury, polytrauma patients, nerve and serious vascular injuries, pathological fractures, and delayed or nonunions. This article aims to describe the published scientific data and current treatment modalities most suitable for each type of fracture.

Abstract:Three new quinazoline derivatives (1-3), one new oxepin-containing natural product (4) and four new cyclopenin derivatives (5-7 and 9) have been isolated from an EtOAc extract of the Taiwan Kueishantao hydrothermal vent crab-associated fungus Aspergillus versicolor XZ-4. Their planar structures were established by HRMS, 1D and 2D NMR spectroscopic data analyses. The absolute configurations for compounds 1 and 4 were determined by chiral phase HPLC analysis of their hydrolysis products. The absolute configurations of 2, 3 and 7 were defined mainly by comparison of the quantum chemical TDDFT calculated and the experimental ECD spectra, and the absolute configuration of 5 was deduced from comparison of the optical rotation values reported in the literature. The presence of two atropisomers of 5 was established by NOE analyses. The Ile & Val units in compounds 1-3 allowed the assignment of a new quinazoline skeleton and it's the first time the configuration of isoleucine in the quinazoline skeleton was defined. A series of 7-methoxy cyclopenin derivatives were reported for the first time in this study. The bioevaluation of compounds 5, 7, 8 and 9 revealed inhibitory activities against E. coli at MIC values around 32 μg mL-1.

Abstract:N -(5-((3E,5E)-10-Acetoxy-5,7,9,11-tetramethyltrideca-3,5-dien-1-yl)-2-oxotetrahydrofuran-3-yl)acetamide ( 1 ), a new amide compound, and five known ones ( 2–6 ) were isolated from the EtOH extract of the solid culture of Fusarium sp. Their structures were determined by means of extensive 1D and 2D NMR techniques, as well as comparison with literature values. Additionally, compounds 1–6 were tested for their cytotoxic activities. The results showed that compounds 2 , 3 , 6 exhibited cytotoxic activities against five human cancer cell lines with IC 50 values ranging from 2.088 to 26.40 μM.

Abstract:Abstract Three new bioactive metabolites, 1,6-dihydroxy- 2-methyl-heptan-4-one ( 1 ), 4-hydroxy-1-(2-methyl-oxiranyl)- pentan-2-one ( 2 ), and 2-(2-hydroxy-propyl)-4-methylfuran- 3-carboxylic acid amide ( 3 ) were isolated from the terrestrial Streptomyces sp. isolate ANK245, along with the new microbial constituent p -vinylanisol ( 4a ) and the known metabolites p -vinyl-phenol ( 4b ) and phenethyl alcohol. Analysis of the nonpolar part of the extract by gas chromatography/mass spectrometry (GC-MS) provided further evidence for tetradecanoic acid, 9-octadecenoic acid, hexadecanoic acid, 2-methoxy-4-vinylphenol ( 4c ), 4-hydroxy-3-methoxy-benzaldehyde, o -hydroxybiphenyl, and 1,5,9-trimethyl-4,8,13-cyclotetradecatrien-1,3-diol ( 5 ). Structures 1–3 of the new compounds were elucidated by nuclear magnetic resonance (NMR) and NMR spectroscopy, but mass spectrometry (MS) techniques and their absolute configuration were determined by density functional theory (DFT) calculations and Mosher derivatisation. Their antimicrobial and cytotoxic activities were evaluated in comparison with the crude bacterial extract.
[Depression among students of the Medical Faculty and doctors in Mostar].

Abstract:UNLABELLED To assess out how many students and doctors have a mild, a moderate and a gravely depressive episode. SUBJECTS The research was carried out among the students and doctors of the Medical Faculty in Mostar, Bosnia and Herzegovina, in March 2003. Out of the total number of 255 medical students and 115 doctors of, 200 (78.4%) medical students and 90 (78.3%) doctors participated in the research. The were analysis in accordance with DSM-IV criteria. RESULTS One hundred and twenty (41.4%) participants experienced mildly depressive episode, and 52 (17.9%) participants experienced moderately depressive episode. Medical students were more depressive (66.5%) than the doctors (43.3%, p=0.001). A majority of the mildly depressive, and in particular the moderately depressive respondents, reported about a mild and moderate, respectively, intensity of disorder of mood loss of interest for the usual activities, fatigue, and the loss of energy. CONCLUSION Medical students and doctors constitute a vulnerable professional group, having a high percentage of depressive disorders.
