Abstract:Objectives We hypothesized that autonomic atrial remodeling can be reversed by low-level (LL) vagosympathetic nerve stimulation (VNS).Background Previously, we showed that VNS can be antiarrhythmogenic.Methods Thirty-three dogs were subjected to electrical stimulation (20 Hz) applied to both vagosympathetic trunks at voltages 10% to 50% below the threshold that slowed sinus rate or AV conduction. Group 1 (n = 7): Programmed stimulation (PS) was performed at baseline and during 6-h rapid atrial pacing (RAP). PS allowed determination of effective refractory period (ERP) and AF inducibility measured by window of vulnerability (WOV). LL-VNS was continuously applied from the 4th to 6th hours. Group 2 (n = 4): After baseline ERP and WOV determinations, 6-h concomitant RAP+LL-VNS was applied. Sustained AF was induced by injecting acetylcholine (ACh) 10 mM into the anterior right ganglionated plexus (Group 3, n = 10) or applying ACh 10 mM to right atrial appendage (Group 4, n = 9).Results Group 1: The ERP progressively shortened and the Sigma WOV (sum of WOV from all tested sites) progressively increased (p < 0.05) during 3-h RAP then returned toward baseline during 3-h RAP+LL-VNS (p < 0.05). Group 2: 6-h concomitant RAP+LL-VNS did not induce any significant change in ERP and Sigma WOV. Group 3 and Group 4: AF duration (AF-D) and cycle length (AF-CL) were markedly altered by 3-h LL-VNS (Group 3: baseline: AF-D = 389 +/- 90 s, AF-CL = 45.1 +/- 7.8 ms; LL-VNS: AF-D = 50 +/- 15 s, AF-CL = 82.0 +/- 13.7 ms [ both p < 0.001]; Group 4: baseline: AF-D = 505 +/- 162 s, AF-CL = 48.8 +/- 6.6 ms; LL-VNS: AF-D = 71 +/- 21 s, AF-CL = 101.3 +/- 20.9 ms [ both p < 0.001]).Conclusions LL-VNS can prevent and reverse atrial remodeling induced by RAP as well as suppress AF induced by strong cholinergic stimulation. Inhibition of the intrinsic cardiac autonomic nervous system by LL-VNS may be responsible for these salutary results. Abstract:We report a patient with atrioventricular reentrant tachycardia (AVRT) with bidirectional conduction over an anteroseptal accessory pathway (AP) who underwent successful ablation in the non-coronary aortic sinus (AS). In three previous attempts, the intracardiac recordings showed an anteroseptal, AP with antegrade and retrograde conduction that failed to be ablated in spite of radiofrequency (RF) applications from the right and left anteroseptal regions. During the study, the earliest atrial. activation during tachycardia was recorded in the non-coronary AS preceding the atrial activation at the His bundle (HB) region by 24 ms, and the anteroseptal AP was successfully blocked by one single ablation in the non-coronary AS. These data strongly suggest that careful mapping of an anteroseptal. AP in the non-coronary AS may provide an alternative ablation approach in patients with previously failed ablation.

Abstract:Objective To report the effect of management of arrhythmia induced by coronary artery spasm.Method Ten patients with syncope or pre-syncope were enrolled in the study,eight patients with complete atrial-ventricular block and inferior ST segment elevation with no-evidence to make the diagnosis clear in the other two patients.All patients accepted the arrhythmia monitoring and coronary angiography(CAG).Results All patients had normal electrolyte and heart structure.Eight patients had normal CAG,the combination of pacemakers implantation and Ca2+ channel antagonist agents were used in them.The other two patients without diagnostic evidence had V1-V6 ST segment elevation followed by ventricular tachycardia.CAG showed plaque less than 50% in left anterior descending artery,and stents were implanted in the spasm-relative artery.Result of follow-up for period of 6mo-11yr showed all were alive without syncope or pre-syncope.Conclusion Methods to focus on unstable plaque and coronary spasm under pacemaker backup are effective.

Abstract:1. Ketamine is widely used for the induction of anaesthesia in high-risk patients with cardiovascular instability or severe hypovolaemia. However, the ionic mechanisms involved in the effects of ketamine at therapeutically relevant concentrations in human cardiac myocytes are unclear. The present study was designed to investigate the effects of ketamine on L-type Ca2+ (I-Ca), transient outward K+ (I-to), ultra-rapid delayed rectifier K+ (I-Kur) and inward rectifier potassium (I-K1) currents, as well as on action potentials, in human isolated atrial myocytes.2. Atrial myocytes were isolated enzymatically from specimens of human atrial appendage obtained from patients undergoing coronary artery bypass grafting. The action potential and membrane currents were recorded in both current- and voltage-clamp modes using the patch-clamp technique.3. Ketamine inhibited I-Ca with an IC50 of 1.8 mu mol/L. In addition, 10 mu mol/L ketamine decreased the I-Ca peak current at +10 mV from 5.1 +/- 0.3 to 2.1 +/- 0.4 pA/pF (P < 0.01), but did not change the threshold potential, peak current potential and reverse potential.4. Ketamine had no effect on I-to, I-Kur or I-K1, but it reversibly shortened the duration of the action potential in human atrial myocytes.5. In conclusion, ketamine, at a clinically relevant concentration, shortens the action potential duration of the human atrial myocytes, probably by inhibiting I-Ca.
Value of the Adenosine Test for Diagnosis of Dual AV Nodal Physiology in Patients with AV Nodal Reentrant Tachycardia

Abstract:Objectives: This study was aimed at assessing the value of the adenosine test for noninvasive diagnosis of dual AV nodal physiology (DAVNP) in patients with AV nodal reentrant tachycardia (AVNRT). Methods: 53 patients with paroxysmal supraventricular tachycardia (PSVT) were given incremental doses of adenosine intravenously during sinus rhythm before electrophysiological study. The adenosine test was repeated on a subset of 18 patients with AVNRT after radiofrequency catheter ablation. Results: Sudden increments of PR interval of more than 60 msec between two consecutive beats were observed in 26 (83.9%) of 31 patients with typical AVNRT and 2 (9.1%) of 22 patients with AVRT and AT (P<0.01). The maximal PR increment between 2 consecutive beats in the AVNRT group (105±45ms) was significantly greater than that in the AVRT and AT group (20±13ms) (P<0.01). In postablation adenosine test, DAVNP was eliminated in all 8 patients who underwent slow pathway abolition that EPS showed the slow pathway disappeared and 4 of 10 patients who underwent slow pathway modification that EPS showed the slow pathway persisted. Six of 10 patients who exhibited persistent duality showed a marked reduction in the number of beats conducted in the slow pathway after adenosine injection (P<0.01). Conclusions: Administration of adenosine during sinus rhythm may be a useful bedside test for diagnosis of DAVNP in high percentage of patients with typical AVNRT and additionally for evaluating the effects of radiofrequency ablation.

Abstract:Ethnopharmacological relevance: Arnebiae Radix, a common herbal medicine in China, is often utilized to treat blood-heat syndrome and has been reported to exert an effect on the heart. A im of the study: The combination of acetylcholine (Ach) and CaCl2 has been widely used to induce atrial fibrillation (AF) in animals. However, whether Amebiae Radix displays any preventive action on Ach-CaCl2 induced AF in rats remains uncertain. In our study, we attempted to investigate the protective effects of Arnebiae Radix on Ach-CaCl2 induced AF compared to amiodarone, which was employed as the positive control. Materials and methods: To establish the AF model, SD rats were treated with a mixture of 0.1 mL/100 g Ach-CaCl2 (60 mu g/mL Ach and 10 mg/mL CaCl2) by tail vein injection for 7 days. Rats were also given a gavage of Arnebiae Radix (0.18 g/mL) one week before or concurrently with the establishment of the AF model. At the end of the experimental period, the induction, duration and timing of AF were monitored using electrocardiogram recordings. Left atrial tissues were stained to observe the level of fibrosis. Electrophysiological measurements were used to examine atrial size and function. Results: In Ach-CaCl2-induced AF rats, Amebiae Radix decreased AF induction, duration and susceptibility to AF. In addition, Arnebiae Radix significantly reduced atrial fibrosis and inhibited atrial enlargement induced by Ach-CaCl2. Moreover, there was an apparent improvement in cardiac function in the Arnebiae Radix-treated group. Conclusions: Our findings indicate that Arnebiae Radix treatment can attenuate Ach-CaCl2-induced atrial injury and serve as an effective therapeutic strategy for the treatment of AF in the future.

Abstract:Objective:To study the cause and appropriate treatment of ventricular fibrillation during selective coronary angiography.Methods:Retrospectively analyzed the data of 5050 patients who were treated with selective coronary angiograph.Results:The seven of 5050 patients complicated with ventricular fibrillation when right coronary angiography was in progress.Five of seven patients were trans-radial artery coronary angiography,four patients using 5F Terumo TIG cather.In these seven patients,two was with three branches lesion,one was with double branch lesion,three with small right coronary artery(one with catheter into Sinus artery,two with deep insert),five of seven patients with contast media retention,five of seven patients with the pressure of coronary artery descending.All patents were successful cardioversion only with one electrical conversion.Conclusion:Small right coronary artery,contrast medium retention,and unfit instrument selection are contributed ventricular fibrillation during selective coronary angiography,but the complication can be avoid if be stressed.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Background</h3><p>Post-operative atrial fibrillation (POAF) is associated with worse long-term cardiovascular outcomes.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Objectives</h3><p>This study hypothesized that injecting calcium chloride (CaCl<sub>2</sub>) into the major atrial ganglionated plexi (GPs) during isolated coronary artery bypass grafting (CABG) can reduce the incidence of POAF by calcium-induced autonomic neurotoxicity.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Methods</h3><p>This proof-of-concept study randomized 200 patients undergoing isolated, off-pump CABG to CaCl<sub>2</sub> (n = 100) or sodium chloride (sham, n = 100) injection. Two milliliters of CaCl<sub>2</sub> (5%) or sodium chloride (0.9%) was injected into the 4 major atrial GPs during CABG. All patients received 7-day continuous telemetry and Holter monitoring. The primary outcome was incidence of POAF (≥30 s) in 7 days. Secondary outcomes included length of hospitalization, POAF burden, average ventricular rate during AF, plasma level of inflammatory markers, and actionable antiarrhythmic therapy to treat POAF.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>The POAF incidence was reduced from 36% to 15% (hazard ratio: 0.366; 95% confidence interval: 0.211 to 0.635; p = 0.001). Length of hospitalization did not differ between the 2 groups. POAF burden (first 7 post-operative days), the use of amiodarone or esmolol, and the incidence of atrial couplets and nonsustained atrial tachyarrhythmias were significantly reduced in the CaCl<sub>2</sub> group. Heart rate variability data showed a decrease in both high-frequency and low-frequency power in the CaCl<sub>2</sub> group with a preserved low-frequency/high-frequency ratio, suggesting that the sympathetic/parasympathetic balance was not perturbed by CaCl<sub>2</sub> injection.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusions</h3><p>Injection of CaCl<sub>2</sub> into the 4 major atrial GPs reduced the POAF hazard by 63%. Inhibition of GP function by Ca-mediated neurotoxicity may underlie the therapeutic effect. Safety and efficacy of aminophylline in the prevention of bradyarrhythmia during coronary atherectomy

Abstract:Coronary calcified lesions are commonly encountered and coronary atherectomy is commonly used for lesion modification during percutaneous coronary interventions (PCI). The release of adenosine during atherectomy can result in bradyarrhythmias and aminophylline is commonly used to prevent this reaction. We identified 138 patients to evaluate the safety and efficacy of intravenous (IV) aminophylline administration prior to coronary atherectomy. A total of 159 calcified lesions were treated, and the atherectomy device was orbital atherectomy, rotational atherectomy, and both in 52 %, 42 %, and 6 %; respectively. After administration of aminophylline, 4.3 % of patients required intraprocedural insertion of a transvenous pacer (TVP), and 18.1 % of patients required administration of IV atropine. Technical success was achieved in 98.6 % of patients, and no adverse reactions to aminophylline were reported. All patients survived to discharge. In conclusion, aminophylline administration prior to coronary atherectomy was safe and effective. No adverse effects of aminophylline were seen, and the rate of bailout TVP placement was low.

Abstract:Supraventricular tachycardia (SVT), a heterogeneous group of rapid arrhythmias, is primarily attributed to ectopic foci or reentrant circuits positioned superior to the bifurcation of the His bundle. [ 1 ] SVT can be stratified into multiple subtypes, such as atrioventricular nodal reentrant tachycardia (AVNRT), sinus tachycardia, atrial fibrillation, focal atrial tachycardia, and atrioventricular reentrant tachycardia, based on the site of abnormality within the conduction system and the pathophysiological principles involved. Notably, AVNRT represents approximately 50% to 60% of all SVT instances. [ 2 , 3 ] AVNRT predominantly afflicts young to middle-aged adults, often manifesting with symptoms such as restlessness, palpitations, angina, and agitation, and it may precipitate shock in severe cases. [ 4 ] At present, the underlying pathogenesis of AVNRT remains incompletely understood. However, it is generally posited that anomalies in the development of the congenital cardiac conduction pathway and functional dissociation within the atrioventricular node play significant roles. [ 5 , 6 ] Traditional management strategies for AVNRT primarily encompass pharmacological interventions (with a focus on beta-blockers and calcium channel blockers), physical therapies, and electrical cardioversion. While these approaches can yield moderate efficacy, their significant limitation lies in the high recurrence rates observed posttreatment.

[The exercise test with atropine]

Abstract:Background: Exercise testing remains the most accessible and widely used technique for the detection of coronary artery disease (CAD) and for the assessment of its severity. Clinical usefulness of the simple exercise test is limited by poor sensitivity and imperfect specificity. Many patients referred for diagnostic stress testing cannot achieve an adequate increase in heart rate due to lack of motivation, poor physical condition, or medications. Atropine increases the heart rate and therefore myocardial oxygen consumption, and might improve the diagnostic accuracy of exercise testing, either reducing the number of non-diagnostic examinations, or increasing the positivity of the test in patients with non severe coronary disease. Methods: One-hundred-seventy-two consecutive patients (144 males and 28 females, age 58 +/- 8 years) with suspected coronary artery disease underwent exercise testing and coronary angiography. Exercise testing was considered positive in the presence of ST segment depression > or = 0.10 mV horizontal or downsloping, > or = 0.15 mV upsloping, or in the presence of ST elevation without Q waves. The test was considered negative in the absence of significant ST depression or elevation if the exercise heart rate was > 85% of age-predicted maximum, non-diagnostic if the heart rate was < 85%. In 148 patients (86%) coronary angiography showed CAD (> 70% luminal reduction in at least one major vessel), in 24 patients (14%) non significant CAD or angiographically normal coronary arteries (NoCAD). Exercise testing was positive in 134 patients (78%: 85% in CAD, 33% in NoCAD), negative in 13 patients (8%: 3% in CAD, 38% in NoCAD), non-diagnostic in 25 patients (14%; 12% in CAD, 29% in NoCAD). The sensitivity was 85%, specificity was 38%, and diagnostic accuracy 78%. In the 25 patients with non-diagnostic test (18 CAD, 7 NoCAD) the exercise was repeated 30-90 minutes later, immediately after i.v. injection of 1-2 mg of atropine, and was stopped at the same workload of the index test. Results: The atropine-exercise test was well tolerated and accomplished in all cases. The maximal heart rate (139 +/- 11 vs 121 +/- 11) and the double product (25,308 +/- 4082 vs 22,166 +/- 3569) were significantly greater after atropine. The increase of the maximal heart rate improved the detection of the electrocardiographic signs of exercise-induced myocardial ischemia only in CAD patients. In the 18 CAD patients the atropine-exercise test was positive in 8, negative in 5, non-diagnostic in 5. In the 7 NoCAD patients the atropine-exercise test was positive in 1, negative in 6. The test remained non-diagnostic only in 3% of patients. The sensitivity of the test with atropine was 91%, specificity was 63%, and diagnostic accuracy 87%. Conclusions: The addition of atropine to exercise testing, which causes further chronotropic stress to the myocardium, is well tolerated and safe, and improves the diagnostic accuracy of the test for the detection of coronary artery disease in patients who cannot achieve an adequate exercise heart rate. The combination with atropine increases the utility and the cost-effectiveness of exercise testing.
[Heart rate reduction using edrophonium during coronary artery bypass grafting without cardiopulmonary bypass]

Abstract:Coronary artery bypass grafting without the use of cardiopulmonary bypass (CPB) is performed with increasing frequency. Performing revascularization on a beating heart is technically more demanding than performing revascularization on the arrested heart, especially in high-risk patients. beta-Blockers and calcium-channel antagonists have been used for the reduction of heart rate (HR) for the local immobilization of the anastomotic site. However, their negative inotropic actions often lead to serious hypotension. Therefore, we investigated the effect of edrophonium on HR reduction in high-risk patients undergoing CABG without CPB. Ten high-risk patients undergoing CABG without CPB were selected. To reduce HR during anastomosis, edrophonium was administered during the procedure. Systemic blood pressure (sBP), HR, and cardiac index (CI) were measured from the induction of anesthesia to the end of surgery. All surgeries were successfully performed without serious complications. To keep the rate under 60 bpm, edrophonium was administered at the time of anastomosis and this decreased the cardiac index from 2.19 to 1.95, while the sPB was maintained easily over 90 mmHg with the infusion of methoxamine. Edrophonium may be useful for the reduction of HR during coronary anastomosis in high-risk patients undergoing CABG without CPB.
Arctigenin Attenuates Ischemia/Reperfusion Induced Ventricular Arrhythmias by Decreasing Oxidative Stress in Rats.

Abstract:BACKGROUND/AIMS:Arctigenin (ATG) has been shown to possess anti-inflammatory, immunemodulatory, anti-viral, anti-microbial, anti-carcinogenic, vasodilatory and anti-platelet aggregation properties. However, the protective role of ATG in prevention of arrhythmias induced by myocardial ischemia/reperfusion is unknown. The aim of this study was to investigate the anti-arrhythmia effect of ATG in an ischemia/reperfusion injured rat heart model and explore the related mechanisms.METHODS:Rats were randomly exposed to sham operation, myocardial ischemia/ reperfusion (MI/R) alone, ATG+ MI/R, pretreated with ATG in low (12.5 mg/kg/day), medium (50 mg/kg/day) and high dose (200 mg/kg/day), respectively. Ventricular arrhythmias were assessed. The activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and the level of malondialdehyde (MDA) in myocardial tissue were determined by chemical analysis.RESULTS:Compared to MI/R, rats pretreated with ATG in doses of 50 mg/kg/day and 200 mg/kg/day showed significantly reduced incidence and duration of ventricular fibrillation, ventricular tachycardia and ventricular ectopic beat (VEB), and decreased the arrhythmia score during the 30-min ischemia. Incidence and duration of ventricular tachycardia, infarction size and arrhythmia scores in these groups were significantly decreased during the 120-min reperfusion. No ventricular fibrillation occurred during the period of reperfusion. Rats pretreated with ATG in doses of 50 mg/kg/day and 200 mg/kg/ day markedly enhanced the activities of antioxidant enzymes SOD and GSH-Px, reduced the level of MDA. No differences were observed between the group pretreated with a low dose of ATG and the sham group. Administration of ATG significantly increased the expression of antioxidant stress protein Nrf2, Trx1 and Nox1.CONCLUSION:Our data suggested that ATG plays anti-arrhythmia role in ischemia/reperfusion injury, which is probably associated with attenuating oxidative stress by Nrf2 signaling pathway.
Arctigenin, a potential anti-arrhythmic agent, inhibits aconitine-induced arrhythmia by regulating multi-ion channels

Abstract:Background/aims: Arctigenin possesses biological activities, but its underlying mechanisms at the cellular and ion channel levels are not completely understood. Therefore, the present study was designed to identify the anti-arrhythmia effect of arctigenin in vivo, as well as its cellular targets and mechanisms. Methods: A rat arrhythmia model was established via continuous aconitine infusion, and the onset times of ventricular premature contraction, ventricular tachycardia and death were recorded. The Action Potential Duration (APD), sodium current (I(Na)), L-type calcium current (I(Ca, L)) and transient outward potassium current (I(to)) were measured and analysed using a patch-clamp recording technique in normal rat cardiomyocytes and myocytes of arrhythmia aconitine-induced by. Results: Arctigenin significantly delayed the arrhythmia onset in the aconitine-induced rat model. The 50% and 90% repolarisations (APD50 and APD90) were shortened by 100 µM arctigenin; the arctigenin dose also inhibited the prolongation of APD50 and APD90 caused by 1 µM aconitine. Arctigenin inhibited I(Na) and I(Ca,L) and attenuated the aconitine-increased I(Na) and I(Ca,L) by accelerating the activation process and delaying the inactivation process. Arctigenin enhanced Ito by facilitating the activation process and delaying the inactivation process, and recoverd the decreased Ito induced by aconitine. Conclusions: Arctigenin has displayed anti-arrhythmia effects, both in vivo and in vitro. In the context of electrophysiology, I(Na), I(Ca, L), and I(to) may be multiple targets of arctigenin, leading to its antiarrhythmic effect.

Abstract:Objective To summarize different kinds of arrhythm which occurred during percutaneous radiofrequency themocoagulation for idiopathic trigeminal neuralgia and to provide grounds for preventing and reducing adverse reactions during operation.Methods Four hundred and twelve patients were included and analysed who were given an local anaesthesia operation of percutaneous radiofrequency thermorhizotomy for idiopathic trigeminal neuralgia during 2005.1～2008.1.Among the 412 patients,297 did not use atropine and 115 used atropine before operation. We evaluated and analysed statistically all kinds of arrhythmia and adverse reactions according to the electrocardiographic monitoring results.Results Eighty-one (19.7%) patients had sinus bradycardia,13 (3.2%) had sinus arrest,2 (0.4%) had Adame-Strokes syndrom and 5 (1.2%) had acute cardiogenic shock. Fifty-five patients had sinus bradycardia and 8 had sinus arrest in atropine-injection group. Twenty-six patients had sinus bradycardia and 5 had sinus arrest in non-atropine-injection group.there was no significant difference between these two groups.Among the 412 patients,93 (22.6%) had sinus tachycardia,66 (16.0%) had atrial premature beats,59 (14.3%) had ventricular premature beats,3 (0.7%) had supraventricular tachycardia and 1 (0.2%) had atrial fibrillation.Conclusion Bradycardia arrhythmia or tachycardia arrhythmia occurred during operation of percutaneous radiofrequency thermorhizotomy. Atropine injection before the operation can not avoid occurrence of bradycardia arrhythm. It is critical to strengthen the cardiac monitoring in order to assure the safety of the operation.
Oxygen-dependent lactate utilization by Actinomyces viscosus and Actinomyces naeslundii.

Abstract:Actinomyces viscosus and Actinomyces naeslundii were grown in chemostat cultures with glucose or lactate as the sole sources of energy. Growth on lactate was dependent on oxygen. Lactate was oxidised partly to acetic acid. As judged from the low carbon recoveries in acidic degradation products, it is concluded that a substantial part of the lactate was oxidised to carbon dioxide. Assuming that 1 ATP is generated in the transfer of an electron pair to oxygen, YATP values of 10-11 were calculated from the molar growth yield on lactate and product formation. Although all cell 3 test strains oxidised lactate, one strain could not grow on lactate as the sole source of energy. The rate of oxygen uptake with lactate as substrate was higher for cells grown aerobically than of cells grown anaerobically, indicating that lactate oxidation was induced by oxygen. It is reasonable to assume that lactate oxidation is mediated either by lactate dehydrogenase or lactate oxidase, but the pathway in Actinomyces sp. is not yet known. Lactate consumption may be an important trait of Actinomyces sp. that live in an environment limited in energy sources for most of the time.

Abstract:Atropine is commonly used to counter the effects of the parasympathetic neurotransmitter acetylcholine on heart rate in clinical practice, such as in the perioperative period; however, individual differences in the response to atropine are huge. The association between <i>SCN10A</i>/voltage-gated sodium channel 1.8 (Na<sub>V</sub>1.8) and cardiac conduction has been demonstrated; however, the exact role of <i>SCN10A</i>/Na<sub>V</sub>1.8 in the heart rate response to atropine remains unclear. To identify the role of <i>SCN10A</i> variants that influence the heart rate responses to atropine, we carried out a retrospective study in 1,005 Han Chinese subjects. Our results showed that rs6795970 was associated with the heart rate response to atropine. The heart rate responses to atropine and methoctramine in Na<sub>V</sub>1.8 knockout mice were lower, whereas the heart rate response to isoproterenol was like those in wild type mice. Furthermore, we observed that the Na<sub>V</sub>1.8 blocker A-803467 alleviated the heart rate response to atropine in wild type mice. The retrospective study revealed a previously unknown role of Na<sub>V</sub>1.8 in controlling the heart rate response to atropine, as shown by the animal study, a speculative mechanism that may involve the cardiac muscarinic acetylcholine receptor M2.
