Abstract:Although divalent cations and lanthides are well-known inhibitors of voltage-dependent Ca2+ currents (ICa), their ability to selectively inhibit a voltage-gated K+ current is less widely documented. We report that La3+ inhibits the transient K+ current (IA) of crab (Cardisoma carnifex) neurosecretory cells at ED50 approximately 5 microM, similar to that blocking ICa, without effecting the delayed rectifier K+ current (IK). Neurons were dissociated from the major crustacean neuroendocrine system, the X-organ-sinus gland, plated in defined medium, and recorded by whole cell patch clamp after 1-2 days in culture. The bath saline included 0.5 microM TTX and 0.5 mM CdCl2 to eliminate inward currents. Responses to depolarizing steps from a holding potential of -40 mV represented primarily IK. They were unchanged by La3+ up to 500 microM. Currents from -80 mV in the presence of 20 mM TEA were shown to represent primarily IA. La3+ (with TEA) reduced IA and maximum conductance (GA) by approximately 10% for 1 microM and another 10% each in 10 and 100 microM La3+. Normalized GA-V curves were well fit with a single Boltzmann function, with V1/2 +4 mV and slope 15 mV in control; V1/2 was successively approximately 15 mV depolarized and slope increased approximately 2 mV for each of these La3+ concentrations. Cd2+ (1 mM), Zn2+ (200 microM), and Pb2+ (100 microM) or removal of saline Mg2+ (26 mM) had little or no effect on IA. Steady-state inactivation showed similar right shifts (from V1/2 -39 mV) and slope increases (from 2.5 mV) in 10 and 100 microM La3+. Time to peak IA was slowed in 10 and 100 microM La3+, whereas curves of normalized time constants of initial decay from peak IA versus Vc were right-shifted successively approximately 15 mV for the three La3+ concentrations. The observations were fitted by a Woodhull-type model postulating a La3+-selective site that lies 0.26-0.34 of the distance across the membrane electric field, and both block of K+ movement and interaction with voltage-gating mechanisms; block can be relieved by depolarization and/or outward current. The observation of selective inhibition of IA by micromolar La3+ raises concerns about its use in studies of ICa to evaluate contamination by outward current.

Abstract:Calcium and sucrose permeabilities (PCa or Psucrose) were calculated from the fluxes of 45Ca and [3H]sucrose across perfused everted and normal configurations of the perineurial cylinder isolated from the frog sciatic nerve and from fluxes into an intact nerve segment bathed in Ringer. Mean PCa for influx across the isolated perineurium equaled 10.2 +/- 0.6 X 10(-7) cm/s (n = 16) compared with Psucrose = 7.4 +/- 0.4 X 10(-7) cm/s. For efflux, PCa = 27.5 +/- 5.0 X 10(-7) cm/s and Psucrose = 23.2 +/- 4.7 X 10(-7) cm/s. The mean ratio of PCa for efflux to PCa for influx (2.7 +/- 0.5) was not significantly different from the flux ratio for sucrose (3.1 +/- 0.7). No effect on PCa or Psucrose was observed when the calcium concentration in the bath was varied from 0.5 to 20 mM, when Na-free Ringer was perfused, or when ouabain, La3+, or 2,4-dinitrophenol was applied. Asymmetrical fluxes across the perineurial cylinder were due presumably to bulk flow and resultant solvent drag out of the lumen caused by perfusion pressure. Calcium accumulated in the perineurial tissue in a saturable manner with a Km of 80 microM and a Bmax of 0.22 mumol/g wet wt. The half time for calcium exchange from the external medium to the nerve was calculated as 3 h. This long half time and the calcium-sequestering ability of the perineurium suggest that the perineurium can stabilize endoneurial calcium during transient changes in the calcium concentration of plasma.
Potassium currents responsible for inward and outward rectification in rat neostriatal spiny projection neurons

Abstract:Many of the nonlinear membrane properties displayed by neostriatal spiny projection neurons are conferred by their voltage-gated potassium (K+) currents, including an inwardly rectifying current (IKir), fast (IAt), and slowly (IAs)-inactivating A-currents, and a slow, noninactivating current. The relative contribution of these K+ currents to the pronounced inward and outward rectification of the current-voltage (I-V) relationship of spiny neurons was investigated in a neostriatal slice preparation. Manipulation of the equilibrium potential for K+ (EK) showed that the voltage dependence of activation of inward rectification was identical to that of IKir. In addition, application of barium (100 microM), which is known to reduce IKir in a time- and voltage-dependent manner, had equivalent effects on inward rectification. Subsequent application of cesium (3 mM) or tetraethylammonium (TEA, 25 mM) blocked inward rectification in a solely voltage-dependent fashion consistent with the action of these blockers on IKir. Administration of 4-aminopyridine (4-AP, 100 microM) at concentrations that selectively depress IAs, reduced outward rectification of spiny neurons at subthreshold membrane potentials. Higher concentrations of 4-AP (2 mM), which block both IAs and IAt, revealed an early transient overshoot in voltage deflections at potentials near spike threshold, but rectification persisted at the end of the responses. The transient overshoot and the residual rectification were eliminated by TEA (25 mM), a blocker of the slow, noninactivating K+ current. Collectively, these results indicate that all three depolarization-activated K+ currents contribute to outward rectification at different times and membrane potentials defined by their voltage dependence of activation and kinetics of inactivation. The spontaneous activity of neostriatal spiny neurons recorded in intact animals is characterized by sustained and limited shifts in membrane potential from relatively hyperpolarized potentials to depolarized potentials near spike threshold. The present data suggest that the hyperpolarized state is determined principally by IKir and the limits on the depolarized state are defined by IAf, IAs, and the noninactivating current. These outward K+ currents also are hypothesized to govern the spike discharge characteristics once the depolarized state has been reached.
The Pore Helix Dipole Has a Minor Role in Inward Rectifier Channel Function

Abstract:Ion channels lower the energetic barrier for ion passage across cell membranes and enable the generation of bioelectricity. Electrostatic interactions between permeant ions and channel pore helix dipoles have been proposed as a general mechanism for facilitating ion passage. Here, using genetic selections to probe interactions of an exemplar potassium channel blocker, barium, with the inward rectifier Kir2.1, we identify mutants bearing positively charged residues in the potassium channel signature sequence at the pore helix C terminus. We show that these channels are functional, selective, resistant to barium block, and have minimally altered conductance properties. Both the experimental data and model calculations indicate that barium resistance originates from electrostatics. We demonstrate that potassium channel function is remarkably unperturbed when positive charges occur near the permeant ions at a location that should counteract pore helix electrostatic effects. Thus, contrary to accepted models, the pore helix dipole seems to be a minor factor in potassium channel permeation.

Abstract:Although it is a well‐known fact that extracellular calcium has a suppressing effect on neuronal excitability, no consensus exists about the mechanism behind this phenomenon. Using patch‐clamp recordings from CA1 neurons from rat hippocampal acute slices, we investigate the literature‐proposed mechanisms and find that the effect of calcium on excitability does not depend on g‐protein signaling, excluding mechanisms involving the CaSR (calcium‐sensing receptor), neither does it require changes in the concentration of intracellular calcium. Our findings support the model of charge screen, where calcium ions neutralize negative charges on or adjacent to the voltage‐gated sodium channels. It is well recognized that changes in the extracellular concentration of calcium ions influence the excitability of neurons, yet what mechanism(s) mediate these effects is still a matter of debate. Using patch‐clamp recordings from rat hippocampal CA1 pyramidal neurons, we examined the contribution of G‐proteins and intracellular calcium‐dependent signaling mechanisms to changes in intrinsic excitability evoked by altering the extracellular calcium concentration from physiological (1.2 mM) to a commonly used experimental (2 mM) level. We find that the inhibitory effect on intrinsic excitability of calcium ions is mainly expressed as an increased threshold for action potential firing (with no significant effect on resting membrane potential) that is not blocked by either the G‐protein inhibitor GDPβS or the calcium chelator BAPTA. Our results therefore argue that in the concentration range studied, G‐protein coupled calcium‐sensing receptors, non‐selective cation conductances, and intracellular calcium signaling pathways are not involved in mediating the effect of extracellular calcium ions on intrinsic excitability. Analysis of the derivative of the action potential, dV/dt versus membrane potential, indicates a current shift towards more depolarized membrane potentials at the higher calcium concentration. Our results are thus consistent with a mechanism in which extracellular calcium ions act directly on the voltage‐gated sodium channels by neutralizing negative charges on the extracellular surface of these channels to modulate the threshold for action potential activation.

Abstract:During incubation in vivo, exogenously applied ionic lanthanum comes to surround the numerous neurosecretory terminals which are found lying within or immediately beneath the acellular neural lamella ensheathing the nerves from fifth instar and adult specimens of Rhodnius prolixus. The lanthanum does not penetrate beyond the cellular perineurium, which completely surrounds the non-neurosecretory axons in these nerves and constitutes a form of ‘blood-brain barrier’. In some cases, however, lanthanum is found in the vicinity of a neurosecretory axon lying beneath the perineurium, where it can be assumed to have leaked in from the neurosecretory terminal lying free in the neural lamella. When nerves are incubated in calcium-free media, regions with an attenuated perineurium become ‘leaky’, in that lanthanum is found lying in those extracellular spaces between axons and glia which lie immediately below the thin part of the perineurial layer. Bathing solutions made slightly hyperosmotic to the haemolymph with sucrose have no apparent disruptive effects on the barrier. When the tissues are incubated in more hypertonic solutions, the perineurial barrier becomes ‘leaky’ throughout, and tracer pervades beyond its cells into all the intercellular spaced between glia and axons. The possible role of the zonulae occludentes in both the maintenance of the perineurial barrier and in the formation of interglial occlusions to local penetration of exogenous substances is considered.

Rapid adaptation to elevated extracellular potassium in the pyloric circuit of the crab, Cancer borealis

Abstract:Elevated potassium concentration ([K+]) is often used to alter excitability in neurons and networks by shifting the potassium equilibrium potential (EK) and, consequently, the resting membrane potential. We studied the effects of increased extracellular [K+] on the well-described pyloric circuit of the crab Cancer borealis. A 2.5-fold increase in extracellular [K+] (2.5×[K+]) depolarized pyloric dilator (PD) neurons and resulted in short-term loss of their normal bursting activity. This period of silence was followed within 5-10 min by the recovery of spiking and/or bursting activity during continued superfusion of 2.5×[K+] saline. In contrast, when PD neurons were pharmacologically isolated from pyloric presynaptic inputs, they exhibited no transient loss of spiking activity in 2.5×[K+], suggesting the presence of an acute inhibitory effect mediated by circuit interactions. Action potential threshold in PD neurons hyperpolarized during an hour-long exposure to 2.5×[K+] concurrent with the recovery of spiking and/or bursting activity. Thus the initial loss of activity appears to be mediated by synaptic interactions within the network, but the secondary adaptation depends on changes in the intrinsic excitability of the pacemaker neurons. The complex sequence of events in the responses of pyloric neurons to elevated [K+] demonstrates that electrophysiological recordings are necessary to determine both the transient and longer term effects of even modest alterations of K+ concentrations on neuronal activity.NEW & NOTEWORTHY Solutions with elevated extracellular potassium are commonly used as a depolarizing stimulus. We studied the effects of high potassium concentration ([K+]) on the pyloric circuit of the crab stomatogastric ganglion. A 2.5-fold increase in extracellular [K+] caused a transient loss of activity that was not due to depolarization block, followed by a rapid increase in excitability and recovery of spiking within minutes. This suggests that changing extracellular potassium can have complex and nonstationary effects on neuronal circuits.
Alterations in Membrane-Potential after Axotomy at Different Distances from the Soma of an Identified Neuron and the Effect of Depolarization on Neurite Outgrowth and Calcium-Channel Expression

Abstract:1. Intracellular recordings were made from the soma of an identified neuron B5 within the buccal ganglion of the mollusc, Helisoma trivolvis, during axotomy induced by crushing or cutting the esophageal nerve. Axotomy was associated with a rapid depolarization and occasionally a burst of action potentials (injury discharge). The magnitude of the membrane depolarization in the soma in response to axotomy decayed exponentially when the distance between the soma and site of injury was increased. Input resistance measurements taken during axotomy showed that a barrier to current flow formed rapidly and gradually recovered within 2 h. A barrier to the diffusion of intracellularly injected carboxyfluorescein formed at the site of injury within 15 min of axotomy. 2. To examine the effect of chronic depolarization on neurite outgrowth, the extracellular potassium ion concentration [K+]o was manipulated. The membrane potential of neurons B5 exhibited a 51.8 mV/decade potassium dependence between 20 and 150 mM [K+]o. The initiation of neurite outgrowth from axons crushed 800 microns from the soma and bathed in different concentrations of [K+]o was examined by fluorescence microscopy after filling neurons with Lucifer yellow. We compared the percentage of axons with sprouts 9 and 24 h after organ culture in saline containing [K+]o ranging from 0.1 to 50 mM. Sprouting occurred from 33% of neurons B5 in normal saline (1.7 mM [K+]o) after 9 h and from 100% of neurons after 24 h. No sprouting was observed from neurons B5 9 or 24 h after axotomy when bathed in saline containing reduced or elevated concentrations of [K+]o. 3. To examine the effects of chronic depolarizatin on neurite outgrowth over several days, neurons B5 were axotomized close to the soma and maintained in organ culture in Liebovitz medium (defined medium or medium conditioned with central ganglia). Neurite outgrowth was ranked from 0 to 5 after filling neurons with Lucifer yellow, and our analysis indicated that a small increase in neurite outgrowth occurred in medium supplemented with 10 mM potassium. 4. Elevated potassium did not trigger neurite outgrowth from isolated neurons B5 in cell culture within defined medium, but whole-cell patch-clamp analysis revealed that chronic depolarization associated with elevated potassium altered the expression of calcium currents. Kir4.1-mediated spatial buffering of K(+): experimental challenges in determination of its temporal and quantitative contribution to K(+) clearance in the brain

Abstract:Neuronal activity results in release of K(+) into the extracellular space of the central nervous system. If the excess K(+) is allowed to accumulate, neuronal firing will be compromised by the ensuing neuronal membrane depolarization. The surrounding glial cells are involved in clearing K(+) from the extracellular space by molecular mechanism(s), the identity of which have been a matter of controversy for over half a century. Kir4.1-mediated spatial buffering of K(+) has been promoted as a major contributor to K(+) removal although its quantitative and temporal contribution has remained undefined. We discuss the biophysical and experimental challenges regarding determination of the contribution of Kir4.1 to extracellular K(+) management during neuronal activity. It is concluded that 1) the geometry of the experimental preparation is crucial for detection of Kir4.1-mediated spatial buffering and 2) Kir4.1 enacts spatial buffering of K(+) during but not after neuronal activity.
In permeabilised endothelial cells IP3-induced Ca2+ release is dependent on the cytoplasmic concentration of monovalent cations

Abstract:Objective: IP3-induced Ca2+ release from the intracellular stores plays a role in the production of vasoactive substances in the endothelium. In many cells, Ca2+ release is accompanied by an inward movement of K+ whose function may be to dissipate the potential difference created by the loss of positive charge from the internal stores. The existence of such a mechanism in endothelial cells was investigated. Methods: Using saponin-permeabilised bovine aortic endothelial (BAE) cells, the effects of K+ on the IP3-induced 45Ca2+ release were investigated. Results: Replacement of K+ with NMG inhibited IP3 (3 microM)-induced 45Ca2+ release by 55%. The ability of other ions to allow IP3-induced 45Ca2+ release was found to be K+ = Na+ > Cs+ > Rb+ >> Co2+. The K+ channel blockers TEA, 4AP and 3,4-DAP were found to significantly inhibit IP3-induced 45Ca2+ release by 16%, 36% and 27%, respectively. Conclusions: The data suggest that Ca2+ release from intracellular stores is partly dependent on a movement of K+ through K+ channels in the store membranes. In contrast, 9AA (400 microM) and substitution with Co2+ abolished the response. Therefore, K+ is important for IP3-induced 45Ca2+ release, but other ions are also likely to act as counter-ions. 9AA and Co2+ probably act on sites other than those involving ER monovalent cation channels. The possibility that a counter-ion system plays a role in the activation of endothelial cells is discussed.
Effect of Large Conductance Calcium-Activated Potassium Channels on Intracellular Free Calcium Concentration and Excitability of Spinal Cord Neurons

Abstract:Objective To investigate the role of large conductance calcium-activated potassium channels (BK) in the regulation of intracelullar free calcium concentration ([Ca2+]i) and excitability of rat spinal cord neurons. Methods Spinal cord neurons of Wistar rats were cultured. The effect of Iberiotoxin (100 nmol/L) on neuronal action potential and [Ca2+]i level was observed by using patch clamp technique under physiological conditions or during depolarization. The influence of Iberiotoxin on [Ca2+]i was detected by fluorescent microscopy system. [Ca2+]i level in spinal cord neurons in the presence or Absence of Iberiotoxin (100 nmol/L) was compared. Results Under physiological conditions, no significant differences were found in spontaneous action potential frequency and [Ca2+]i level after perfusion of Iberiotoxin (n=7, P>0.05). The frequency of action potential evoked by electric current became higher after perfusion of Iberiotoxin, as well as the [Ca2+]i level during depolarization (n=9, P<0.05). Extracellular high concentration of KCl increased [Ca2+]i level of spinal cord neurons. [Ca2+]i level in spinal cord neurons treated by Iberiotoxin was much higher than that without Iberiotoxin treatment (n=9, P<0.05). ConclusionBK may play an important role in the regulation of intracellular free calcium and excitability of spinal cord neurons.
The Effect of Calcium Ions on Resting Membrane Potential

Abstract:Regulating membrane potential is key to cellular function. For many animal cells, resting membrane potential is predominantly driven by a family of K2P (two-pore domain) potassium channels. These channels are commonly referred to as leak channels, as their presence results in the membrane being permeable to K+ ions. These channels, along with various pumps and exchangers, keep the cell resting membrane potential (Rp) relatively close to potassium's equilibrium potential (EK); however, in many cells, the resting membrane potential is more depolarized than the EK due to a small Na+ ion leak. Raising [Ca2+]O (extracellular Ca2+ concentration) can result in hyperpolarization of the membrane potential from the resting state. The mechanism for this hyperpolarization likely lies in the blockage of a Na+ leak channel (NALCN) and/or voltage-gated Na+ channels. The effects may also be connected to calcium-activated potassium channels. Using Drosophila melanogaster, we here illustrate that changing [Ca2+]O from 0.5 to 3 mM hyperpolarizes the muscle. Replacing NaCl with LiCl or choline chloride still led to hyperpolarization when increasing [Ca2+]O. Replacing CaCl2 with BaCl2 results in depolarization. K2P channel overexpression in the larval muscle greatly reduces the effects of [Ca2+]O on cell membrane potential, likely because potential is heavily driven by the EK in these muscles. These experiments provide an understanding of the mechanisms behind neuronal hypo-excitability during hypercalcemia, as well as the effects of altered expression of K2P channels on membrane potential.
A theoretical examination of ionic interactions between neural and non-neural membranes

Abstract:Evidence from electron microscopy indicates that the separation between adjacent membranes of the central nervous system (CNS) is less than 500 A and perhaps as small as 100-250 A. The rapid K(+) efflux associated with the neural action potential may therefore be sufficient to affect the local extracellular potassium concentration and, via their partial dependence upon the potassium equilibrium potential, alter the electrical states of nearby neural and glial membranes. This new concept of a transient and local depolarizing "ionic interaction" between active and inactive membranes of the CNS is here examined theoretically and its magnitude calculated as a function of (a) the intermembrane separation, (b) the membranes' electrochemical characteristics, and (c) the rate at which K(+) can diffuse away from the vicinity of the active (neural) membrane. My results indicate that the interaction is in the millivolt range and therefore significant in the modulation of postsynaptic and presynaptic information processing; in particular configurations the postulated interaction alone may be suprathreshold. Membrane noise and local synchrony in groups of neurons may reflect these local, K(+)-mediated interactions. The transient ionic interaction between active neural and nearby glial membrane is also in the millivolt range; however, the relevance of neuroglia to neuronal function is obscure. Certain pathological states, such as seizure and spreading depression, have an obvious phenomenological correspondence to the results presented here and are briefly discussed.

Abstract:The α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors (AMPARs) are neurotransmitter-activated cation channels ubiquitously expressed in vertebrate brains. The regulation of calcium flux through the channel pore by RNA-editing is linked to synaptic plasticity while excessive calcium influx poses a risk for neurodegeneration. Unfortunately, the molecular mechanisms underlying this key process are mostly unknown. Here, we investigated calcium conduction in calcium-permeable AMPAR using Molecular Dynamics (MD) simulations with recently introduced multisite force-field parameters for Ca2+. Our calculations are consistent with experiment and explain the distinct calcium permeability in different RNA-edited forms of GluA2. For one of the identified metal binding sites, multiscale Quantum Mechanics/Molecular Mechanics (QM/MM) simulations further validated the results from MD and revealed small but reproducible charge transfer between the metal ion and its first solvation shell. In addition, the ion occupancy derived from MD simulations independently reproduced the Ca2+ binding profile in an X-ray structure of an NaK channel mimicking the AMPAR selectivity filter. This integrated study comprising X-ray crystallography, multisite MD, and multiscale QM/MM simulations provides unprecedented insights into Ca2+ permeation mechanisms in AMPARs, and paves the way for studying other biological processes in which Ca2+ plays a pivotal role.

Apparent intermediate K conductance channel hyposmotic activation in human lens epithelial cells

Abstract:This study explores the nature of K fluxes in human lens epithelial cells (LECs) in hyposmotic solutions. Total ion fluxes, Na-K pump, Cl-dependent Na-K-2Cl (NKCC), K-Cl (KCC) cotransport, and K channels were determined by 85Rb uptake and cell K (Kc) by atomic absorption spectrophotometry, and cell water gravimetrically after exposure to ouabain +/- bumetanide (Na-K pump and NKCC inhibitors), and ion channel inhibitors in varying osmolalities with Na, K, or methyl-d-glucamine and Cl, sulfamate, or nitrate. Reverse transcriptase polymerase chain reaction (RT-PCR), Western blot analyses, and immunochemistry were also performed. In isosmotic (300 mosM) media approximately 90% of the total Rb influx occurred through the Na-K pump and NKCC and approximately 10% through KCC and a residual leak. Hyposmotic media (150 mosM) decreased K(c) by a 16-fold higher K permeability and cell water, but failed to inactivate NKCC and activate KCC. Sucrose replacement or extracellular K to >57 mM, but not Rb or Cs, in hyposmotic media prevented Kc and water loss. Rb influx equaled Kc loss, both blocked by clotrimazole (IC50 approximately 25 microM) and partially by 1-[(2-chlorophenyl) diphenylmethyl]-1H-pyrazole (TRAM-34) inhibitors of the IK channel KCa3.1 but not by other K channel or connexin hemichannel blockers. Of several anion channel blockers (dihydro-indenyl)oxy]alkanoic acid (DIOA), 4-2(butyl-6,7-dichloro-2-cyclopentylindan-1-on-5-yl)oxybutyric acid (DCPIB), and phloretin totally or partially inhibited Kc loss and Rb influx, respectively. RT-PCR and immunochemistry confirmed the presence of KCa3.1 channels, aside of the KCC1, KCC2, KCC3 and KCC4 isoforms. Apparently, IK channels, possibly in parallel with volume-sensitive outwardly rectifying Cl channels, effect regulatory volume decrease in LECs.

Abstract:Cerebral arteries contain two primary and interacting cell types, smooth muscle (SMCs) and endothelial cells (ECs), which are each capable of sensing particular hemodynamic forces to set basal tone and brain perfusion. These biomechanical stimuli help confer tone within arterial networks upon which local neurovascular stimuli function. Tone development is intimately tied to arterial membrane potential (V M ) and changes in intracellular [Ca 2+ ] driven by voltage-gated Ca 2+ channels (VGCCs). Arterial V M is in turn set by the dynamic interplay among ion channel species, the strongly inward rectifying K + (Kir) channel being of special interest. Kir2 channels possess a unique biophysical signature in that they strongly rectify, display negative slope conductance, respond to elevated extracellular K + and are blocked by micromolar Ba 2+ . While functional Kir2 channels are expressed in both smooth muscle and endothelium, they lack classic regulatory control, thus are often viewed as a simple background conductance. Recent literature has provided new insight, with two membrane lipids, phosphatidylinositol 4,5-bisphosphate (PIP 2 ) and cholesterol, noted to (1) stabilize Kir2 channels in a preferred open or closed state, respectively, and (2) confer, in association with the cytoskeleton, caveolin-1 (Cav1) and syntrophin, hemodynamic sensitivity. It is these aspects of vascular Kir2 channels that will be the primary focus of this review.

Ion transport mechanisms responsible for K+ secretion and the transepithelial voltage across marginal cells of stria vascularis in vitro

Abstract:It has long been accepted that marginal cells of stria vascularis are involved in the generation of the endocochlear potential and the secretion of K+. The present study was designed to provide evidence for this hypothesis and for a cell model proposed to explain K+ secretion and the generation of the endocochlear potential. Stria vascularis from the cochlea of the gerbil was isolated and mounted into a micro-Ussing chamber such that the apical and basolateral membrane of marginal cells could be perfused independently. In this preparation, the transepithelial voltage (Vt) and resistance (Rt) were measured across marginal cells and the resulting equivalent short circuit current (Isc) was calculated (Isc = Vt/Rt). Further, K+ secretion (JK+,probe) was measured with a K(+)-selective vibrating probe in the vicinity of the apical membrane. In the absence of extrinsic chemical driving forces, when both sides of the marginal cell epithelium were bathed with a perilymph-like solution, Vt was 8 mV (apical side positive), Rt was 10 ohm-cm2 and Isc was 850 microA/cm2 (N = 27). JK+,probe was outwardly directed from the apical membrane and reversibly inhibited by basolateral bumetanide, a blocker of the Na+/Cl-/K+ cotransporter. On the basolateral but not apical side, oubain and bumetanide each caused a decline of Vt and an increase of Rt suggesting the presence of the Na,K-ATPase and the Na+/Cl-/K+ cotransporter in the basolateral membrane. The responses to [Cl-] steps demonstrated a significant Cl- conductance in the basolateral membrane and a small Cl- conductance in the paracellular pathway or the apical membrane. The responses to [Na+] steps demonstrated no significant Na+ conductance in the basolateral membrane and a small Na+ or nonselective cation conductance in the apical membrane or paracellular pathway. The responses to [K+] steps demonstrated a large K+ conductance in the apical membrane. Apical application of 4,4'-diisothiocyanatostilbene-2,2'-disulfonic acid (DIDS) and basolateral elevation of K+ caused an increase in Vt and a decrease in Rt consistent with stimulation of the apical K+ conductance. Similar observations have been made in vestibular dark cells, which suggest that strial marginal cells and vestibular dark cells are homologous and transport ions by the same pathways. Taken together, these observations are incompatible with a model for the generation of the endocochlear potential which ascribes the entire potential to the strial marginal cells [Offner et al.
