Abstract:We investigated further the mechanism whereby albumin interacts with the walls of capillary blood vessels to maintain normal permeability properties. In individually perfused capillaries of frog mesentery, hydraulic conductivity was measured as the albumin perfusate concentration was first reduced in three steps from 0.1 to 0 g/dl (intermediate concns 0.01 and 0.001 g/dl), then increased in up to four steps to 1 g/dl (intermediate concns 0.001, 0.01, and 0.1 g/dl). The albumin concentration required to return hydraulic conductivity toward control values, following Ringer perfusion, was at least an order of magnitude larger than that required to maintain permeability close to control values prior to Ringer perfusion. The experiments indicate that the affinity of albumin for binding sites on the capillary wall following Ringer perfusion is less than the albumin affinity for binding sites after the capillary has been perfused with solutions containing 0.1 g/dl albumin. Before the hysteresis of hydraulic conductivity on albumin concentration can be understood, the factors determining the transport of albumin into the membrane must be investigated further.

The Effect of Calcium Ions on Resting Membrane Potential

Abstract:Regulating membrane potential is key to cellular function. For many animal cells, resting membrane potential is predominantly driven by a family of K2P (two-pore domain) potassium channels. These channels are commonly referred to as leak channels, as their presence results in the membrane being permeable to K+ ions. These channels, along with various pumps and exchangers, keep the cell resting membrane potential (Rp) relatively close to potassium's equilibrium potential (EK); however, in many cells, the resting membrane potential is more depolarized than the EK due to a small Na+ ion leak. Raising [Ca2+]O (extracellular Ca2+ concentration) can result in hyperpolarization of the membrane potential from the resting state. The mechanism for this hyperpolarization likely lies in the blockage of a Na+ leak channel (NALCN) and/or voltage-gated Na+ channels. The effects may also be connected to calcium-activated potassium channels. Using Drosophila melanogaster, we here illustrate that changing [Ca2+]O from 0.5 to 3 mM hyperpolarizes the muscle. Replacing NaCl with LiCl or choline chloride still led to hyperpolarization when increasing [Ca2+]O. Replacing CaCl2 with BaCl2 results in depolarization. K2P channel overexpression in the larval muscle greatly reduces the effects of [Ca2+]O on cell membrane potential, likely because potential is heavily driven by the EK in these muscles. These experiments provide an understanding of the mechanisms behind neuronal hypo-excitability during hypercalcemia, as well as the effects of altered expression of K2P channels on membrane potential.

Abstract:Involvement of extracellular Ca2+ in stomatal movement through the regulation of water channels was investigated in broad bean (Vicia faba L.). Leaf peels were first incubated to open stomata, and then transferred to buffers in the presence of different CaCl2 concentrations. Stomatal status was observed under magnification and stomatal aperture (pore width/length) was measured. Stomatal closure was significantly induced and aperture oscillation occurred at lower extracellular concentrations of calcium ([Ca2+](ext)), while at higher concentrations, no significant change in stomatal aperture was observed, which was similar to the response recorded with HgCl2. Lower [Ca2+](ext)-induced stomatal closure could be reversed using depolarizing buffer. It is suggested that lower [Ca2+](ext) regulates water channels through an indirect way and at higher concentrations, extracellular Ca2+ is involved in regulating stomatal aperture by directly influencing water channels to retard aperture change.

Abstract:Connexin hemichannels (HCs) expressed at the plasma membrane of mammalian cells are of paramount importance for intercellular communication. In physiological conditions, HCs can form gap junction (GJ) channels, providing a direct diffusive path between neighbouring cells. In addition, unpaired HCs provide conduits for the exchange of solutes between the cytoplasm and the extracellular milieu, including messenger molecules involved in paracrine signalling. The synergistic action of membrane potential and Ca2+ ions controls the gating of the large and relatively unselective pore of connexin HCs. The four orders of magnitude difference in gating sensitivity to the extracellular ([Ca2+]e) and the cytosolic ([Ca2+]c) Ca2+ concentrations suggests that at least two different Ca2+ sensors may exist. While [Ca2+]e acts as a spatial modulator of the HC opening, which is most likely dependent on the cell layer, compartment, and organ, [Ca2+]c triggers HC opening and the release of extracellular bursts of messenger molecules. Such molecules include ATP, cAMP, glutamate, NAD+, glutathione, D-serine, and prostaglandins. Lost or abnormal HC regulation by Ca2+ has been associated with several diseases, including deafness, keratitis ichthyosis, palmoplantar keratoderma, Charcot–Marie–Tooth neuropathy, oculodentodigital dysplasia, and congenital cataracts. The fact that both an increased and a decreased Ca2+ sensitivity has been linked to pathological conditions suggests that Ca2+ in healthy cells finely tunes the normal HC function. Overall, further investigation is needed to clarify the structural and chemical modifications of connexin HCs during [Ca2+]e and [Ca2+]c variations. A molecular model that accounts for changes in both Ca2+ and the transmembrane voltage will undoubtedly enhance our interpretation of the experimental results and pave the way for developing therapeutic compounds targeting specific HC dysfunctions.

In permeabilised endothelial cells IP3-induced Ca2+ release is dependent on the cytoplasmic concentration of monovalent cations

Abstract:Objective: IP3-induced Ca2+ release from the intracellular stores plays a role in the production of vasoactive substances in the endothelium. In many cells, Ca2+ release is accompanied by an inward movement of K+ whose function may be to dissipate the potential difference created by the loss of positive charge from the internal stores. The existence of such a mechanism in endothelial cells was investigated. Methods: Using saponin-permeabilised bovine aortic endothelial (BAE) cells, the effects of K+ on the IP3-induced 45Ca2+ release were investigated. Results: Replacement of K+ with NMG inhibited IP3 (3 microM)-induced 45Ca2+ release by 55%. The ability of other ions to allow IP3-induced 45Ca2+ release was found to be K+ = Na+ > Cs+ > Rb+ >> Co2+. The K+ channel blockers TEA, 4AP and 3,4-DAP were found to significantly inhibit IP3-induced 45Ca2+ release by 16%, 36% and 27%, respectively. Conclusions: The data suggest that Ca2+ release from intracellular stores is partly dependent on a movement of K+ through K+ channels in the store membranes. In contrast, 9AA (400 microM) and substitution with Co2+ abolished the response. Therefore, K+ is important for IP3-induced 45Ca2+ release, but other ions are also likely to act as counter-ions. 9AA and Co2+ probably act on sites other than those involving ER monovalent cation channels. The possibility that a counter-ion system plays a role in the activation of endothelial cells is discussed.

Abstract:change in volume, and proton permeability of the inner membrane of mitochondria isolated from cotyledons of etiolated seedlings of narrow-leaved lupine (Lupinus angustifolius L.) was studied. The mitochondria used in the work were characterized by a strong coupling of oxidation and phosphorylation processes. The high functional activity of mitochondria was confirmed by their ability to generate a transmembrane gradient of protons on the inner membrane (membrane potential or ∆Ψ) during the oxidation of succinate and also sustainably maintain it for a long time , both due to the operation of the electron transport chain and due to ATP hydrolysis under conditions of anaerobiosis. It was shown that the presence of 60–120 μM CaCl2 in the mitochondrial incubation medium had no significant effect on the rate of succinate oxidation and the parameters of oxidative phosphorylation; however, it induced ∆Ψ dissipation under conditions of oxygen depletion in the incubation medium. The most complete removal Ca2+ from the medium in the presence of chelators (EGTA, EDTA) prevented the membrane potential dissipation. Ca2+-dependent depolarization of the inner membrane was inhibited by dithiothreitol, suggesting involvement in this process of reactive oxygen species. The reset of the membrane potential was not accompanied by swelling of mitochondria and was not sensitive to cyclosporine A. Using metallochromic Ca2+-indicator arsenazo III, it was shown that the mitochondria of lupine cotyledons are able to actively absorb exogenous Ca2+ and store it in the matrix. Ca2+-induced dissipation of ∆Ψ under conditions of anaerobiosis was accompanied by the release of Ca2+ from mitochondria, the rate of which sharply increased in the presence of calcium ionophore A23 (A23187). It is assumed that the accumulation of Ca2+ and an increase in the level of reactive oxygen species in the matrix induces reversible permeabilization of the inner mitochondrial membrane of lupine cotyledons under conditions of anaerobiosis, which is due to the opening of a pore of nonspecific permeability in a state of low conductivity permeable to protons and, possibly, to other small cations (Na+, K+, Ca2+).
Permeation and gating properties of the L-type calcium channel in mouse pancreatic beta cells

Abstract:Ba2+ currents through L-type Ca2+ channels were recorded from cell-attached patches on mouse pancreatic beta cells. In 10 mM Ba2+, single-channel currents were recorded at -70 mV, the beta cell resting membrane potential. This suggests that Ca2+ influx at negative membrane potentials may contribute to the resting intracellular Ca2+ concentration and thus to basal insulin release. Increasing external Ba2+ increased the single-channel current amplitude and shifted the current-voltage relation to more positive potentials. This voltage shift could be modeled by assuming that divalent cations both screen and bind to surface charges located at the channel mouth. The single-channel conductance was related to the bulk Ba2+ concentration by a Langmuir isotherm with a dissociation constant (Kd(gamma)) of 5.5 mM and a maximum single-channel conductance (gamma max) of 22 pS. A closer fit to the data was obtained when the barium concentration at the membrane surface was used (Kd(gamma) = 200 mM and gamma max = 47 pS), which suggests that saturation of the concentration-conductance curve may be due to saturation of the surface Ba2+ concentration. Increasing external Ba2+ also shifted the voltage dependence of ensemble currents to positive potentials, consistent with Ba2+ screening and binding to membrane surface charge associated with gating. Ensemble currents recorded with 10 mM Ca2+ activated at more positive potentials than in 10 mM Ba2+, suggesting that external Ca2+ binds more tightly to membrane surface charge associated with gating. The perforated-patch technique was used to record whole-cell currents flowing through L-type Ca2+ channels. Inward currents in 10 mM Ba2+ had a similar voltage dependence to those recorded at a physiological Ca2+ concentration (2.6 mM). BAY-K 8644 (1 microM) increased the amplitude of the ensemble and whole-cell currents but did not alter their voltage dependence. Our results suggest that the high divalent cation solutions usually used to record single L-type Ca2+ channel activity produce a positive shift in the voltage dependence of activation (approximately 32 mV in 100 mM Ba2+).
The Pump and Leak Steady-State Concept with a Variety of Regulated Leak Pathways

Abstract:The paper will reflect on how Ussing has affected my own scientific work and how he created much of the framework within which I have been working. I have used five examples: (i) The first description of a 1:1 exchange diffusion was introduced by Ussing in 1947 and has been found to be of great physiological significance in most cells. We found that Cl- transport in Ehrlich ascites tumor cells (EATC) was completely dominated by an exchange diffusion process, as defined by Ussing, and, thus, the Cl- conductance was much lower than previously estimated from measurements of unidirectional tracer fluxes. This had a major influence on my later description of a swelling-activated Cl- conductance. (ii) The pump-leak steady-state concept for cell volume control was introduced by Krogh in 1946, but it was developed in detail by Leaf and Ussing in 1959. This concept was the basis for me and others, when we later found that the passive ion leaks play an active role in cell volume control. (iii) The use of isotopes and Ussing's famous flux ratio equation provided an ingenious instrument for distinguishing the various transport routes. We used this to identify the Na,K,2Cl cotransport system as accounting for maintaining a [Cl-]i in the EATC far above thermodynamic equilibrium, as well as accounting for the ion uptake during a regulatory volume increase (RVI) in EATC, similar to what Ussing had found in frog skin. (iv) Short-circuit current setup in the Ussing chamber is still used in laboratories around the world to study ion transport across epithelia. A few results on Cl- transport across the operculum epithelium of the small eurohaline fish Fundulus heteroclitus mounted in small Ussing chambers are presented. (v) Shrinkage-activated Na+ conductance and its possible role in isotonic secretion in frog skin glands is finally discussed.

Identification of a stretch-activated monovalent cation channel from teleost urinary bladder cells

Abstract:The urinary bladder of euryhaline teleost is an important osmoregulatory organ which absorbs Na+, Cl-, and water from urine. Using patch clamp technique, single stretch-activated channels, which were permeable to K+ and Na+ (PNa/PK approximately 0.75) and had conductances of 55 and 116 pS, were studied. In excised, inside-out patches which were voltage-clamped in the physiological range of membrane potential, the single-channel open probability (Po) was low (approximately 0.02), and increased to a maximum of 0.9 with applied pipette suction. Single-channel conductance also increased with suction. The channels showed adaptation to applied suction and relaxed to a steady-state activity about 20 seconds after application of suction. The Po increased up to 0.9 with strong membrane depolarization (Vm = 0 to +80 mV); however, there was little dependence of Po on membrane potential in the physiological range. The kinetic data suggest that there is one conducting state and at least two non-conducting states of the channel. The open-time constant increased with suction but remained unchanged with membrane potential (Vm = -70 to +60 mV). The mean closed-time of the channel decreased with suction and membrane depolarization. These results demonstrate the presence of a non-selective monovalent cation channel which may be involved in cell volume regulation in the goby urinary bladder. Additionally, this channel may function as an enhancer of Na+ influx and K+ efflux across the bladder cell as part of transepithelial ion transport if it is located in apical membrane.
Calcium Ion Binding at the Lipid-Water Interface Alters the Ion Permeability of Phospholipid Bilayers.

Abstract:Calcium ions (Ca2+) play a fundamental role in membrane-associated physiological processes. Ca2+ can also significantly modulate the physicochemical properties of phospholipid bilayers, but whether this occurs at physiologically relevant concentrations is difficult to determine because of the uncertainty in the reported affinity of Ca2+ for phospholipid bilayers. In this article, we determine the apparent affinity of Ca2+ for zwitterionic phospholipid bilayers using tethered bilayer lipid membranes (tBLMs) used in conjunction with swept-frequency electrical impedance spectroscopy (EIS). We report that Ca2+ binds to phospholipid bilayers at physiologically relevant concentrations and modulates membrane permeability. We present direct experimental evidence that this effect is governed by specific interactions with select lipid headgroup moieties, which is supported by data from molecular dynamics (MD) simulations. This is the first reported use of tBLM/EIS to estimate cation-membrane affinity. Combined with MD simulations, this technique provides a novel methodology to elucidate the molecular details of cation-membrane interactions at the water-phospholipid interface.

Na-caprate-induced increase in MDCK II epithelial cell leak pathway permeability and opening number is associated with disruption of basal F-actin organization

Abstract:Confluent populations of the epithelial cell line, MDCK II, develop circumferential tight junctions joining adjacent cells to create a barrier to the paracellular movement of solutes and water. Treatment of MDCK II cell populations from the apical surface with 1 mM Na-caprate increased permeability to macromolecules (Leak Pathway) without increasing monolayer disruption or cell death. Graphical analysis of the apparent permeability versus solute Stokes radius for a size range of fluorescein-dextran species indicates apical 1 mM Na-caprate enhances Leak Pathway permeability by increasing the number of Leak Pathway openings without significantly affecting opening size. Na-caprate treatment did not alter the content of any tight junction protein examined. Treatment of MDCK II cell populations with apical 1 mM Na-caprate disrupted basal F-actin stress fibers and decreased the tortuosity of the tight junctions. Treatment of MDCK II cell populations with blebbistatin, a myosin ATPase inhibitor, alone had little effect on Leak Pathway permeability but synergistically increased Leak Pathway permeability when added with 1 mM Na-caprate. Na-caprate exhibited a similar ability to increase Leak Pathway permeability in wild-type MDCK II cell monolayers and ZO-1 knockdown MDCK II cell monolayers but an enhanced ability to increase Leak Pathway permeability in monolayers of TOCA-1 knockout MDCK II cells. These results demonstrate that Na-caprate increases MDCK II cell population Leak Pathway permeability by increasing the number of Leak Pathway openings. This action is likely mediated by alterations in F-actin organization, primarily involving disruption of basal F-actin stress fibers.NEW & NOTEWORTHY This study determines the underlying change in the openings in the epithelial tight junction permeability barrier structure that leads to a change in the paracellular permeability to macromolecules (the Leak Pathway) and connects this to disruption of specific F-actin structures within the cells. It provides important and novel insights into how tight junction permeability to macromolecules is modulated by specific changes to cellular and tight junction composition/organization.
Nernst–Planck–Gaussian finite element modelling of Ca2+ electrodiffusion in amphibian striated muscle transverse tubule–sarcoplasmic reticular triadic junctional domains

Abstract:Introduction: Intracellular Ca 2+ signalling regulates membrane permeabilities, enzyme activity, and gene transcription amongst other functions. Large transmembrane Ca 2+ electrochemical gradients and low diffusibility between cell compartments potentially generate short-lived, localised, high-[Ca 2+ ] microdomains. The highest concentration domains likely form between closely apposed membranes, as at amphibian skeletal muscle transverse tubule–sarcoplasmic reticular (T-SR, triad) junctions. Materials and methods: Finite element computational analysis characterised the formation and steady state and kinetic properties of the Ca 2+ microdomains using established empirical physiological and anatomical values. It progressively incorporated Fick diffusion and Nernst–Planck electrodiffusion gradients, K + , Cl − , and Donnan protein, and calmodulin (CaM)-mediated Ca 2+ buffering. It solved for temporal–spatial patterns of free and buffered Ca 2+ , Gaussian charge differences, and membrane potential changes, following Ca 2+ release into the T-SR junction. Results: Computational runs using established low and high Ca 2+ diffusibility ( D Ca2+ ) limits both showed that voltages arising from intracytosolic total [Ca 2+ ] gradients and the counterions little affected microdomain formation, although elevated D Ca2+ reduced attained [Ca 2+ ] and facilitated its kinetics. Contrastingly, adopting known cytosolic CaM concentrations and CaM-Ca 2+ affinities markedly increased steady-state free ([Ca 2+ ] free ) and total ([Ca 2+ ]), albeit slowing microdomain formation, all to extents reduced by high D Ca2+ . However, both low and high D Ca2+ yielded predictions of similar, physiologically effective, [Ca 2+ -CaM]. This Ca 2+ trapping by the relatively immobile CaM particularly increased [Ca 2+ ] at the junction centre. [Ca 2+ ] free , [Ca 2+ -CaM], [Ca 2+ ], and microdomain kinetics all depended on both CaM-Ca 2+ affinity and D Ca2+. These changes accompanied only small Gaussian (∼6 mV) and surface charge (∼1 mV) effects on tubular transmembrane potential at either D Ca2+ . Conclusion: These physical predictions of T-SR Ca 2+ microdomain formation and properties are compatible with the microdomain roles in Ca 2+ and Ca 2+ -CaM-mediated signalling but limited the effects on tubular transmembrane potentials. CaM emerges as a potential major regulator of both the kinetics and the extent of microdomain formation. These possible cellular Ca 2+ signalling roles are discussed in relation to possible feedback modulation processes sensitive to the μM domain but not nM bulk cytosolic, [Ca 2+ ] free , and [Ca 2+ -CaM], including ryanodine receptor-mediated SR Ca 2+ release; Na + , K + , and Cl − channel-mediated membrane excitation and stabilisation; and Na + /Ca 2+ exchange transport.

Abstract:Objective— The present study is aimed at investigating the interaction of TRPV4 with TRPC1 and the functional role of such an interaction in flow-induced Ca 2+ influx. Hemodynamic blood flow is an important physiological factor that modulates vascular tone. One critical early event in this process is a cytosolic Ca 2+ ([Ca 2+ ] i ) rise in endothelial cells in response to flow. Methods and Results— With the use of fluorescence resonance energy transfer, coimmunoprecipitation, and subcellular colocalization methods, it was found that TRPC1 interacts physically with TRPV4 to form a complex. In functional studies, flow elicited a transient [Ca 2+ ] i increase in TRPV4-expressing human embryonic kidney (HEK) 293 cells. Coexpression of TRPC1 with TRPV4 markedly prolonged this [Ca 2+ ] i transient; it also enabled this [Ca 2+ ] i transient to be negatively modulated by protein kinase G. Furthermore, this flow-induced [Ca 2+ ] i increase was markedly inhibited by anti–TRPC1-blocking antibody T1E3 and a dominant-negative construct TRPC1Δ567-793 in TRPV4-C1–coexpressing HEK cells and human umbilical vein endothelial cells. T1E3 also inhibited flow-induced vascular dilation in isolated rat small mesenteric artery segments. Conclusion— This study shows that TRPC1 interacts physically with TRPV4 to form a complex, and this TRPV4-C1 complex may mediate flow-induced Ca 2+ influx in vascular endothelial cells. The association of TRPC1 with TRPV4 prolongs the flow-induced [Ca 2+ ] i transient, and it also enables this [Ca 2+ ] i transient to be negatively modulated by protein kinase G. This TRPV4-C1 complex plays a key role in flow-induced endothelial Ca 2+ influx.

Abstract:Previous studies have reported that the KcsA potassium channel has an osmotic permeability coefficient of 4.8 x 10(-12) cm3/s, giving it a significantly higher osmotic permeability coefficient than that of some membrane channels specialized in water transport. This high osmotic permeability is proposed to occur when the channel is depleted of potassium ions, the presence of which slow down the water permeation process. The atomic structure of the potassium-depleted KcsA channel and the mechanisms of water permeation have not been well characterized so far. Here, all-atom molecular dynamics simulations, in conjunction with an umbrella sampling strategy and a nonequilibrium approach to simulate pressure gradients are employed to illustrate the permeation of water in the absence of ions through the KcsA K+ channel. Equilibrium molecular dynamics simulations (95 ns combined total length) identified a possible structure of the potassium-depleted KcsA channel, and umbrella sampling calculations (160 ns combined total length) revealed that this structure is not permeable by water molecules moving along the channel axis. The simulation of a pressure gradient across the channel (30 ns combined total length) identified an alternative permeation pathway with a computed osmotic permeability of approximately (2.7 +/- 0.9) x 10(-13) cm3/s. Water fluxes along this pathway did not proceed through collective water motions or transitions to vapor state. All of the major results of this study were robust against variations in a wide set of simulation parameters (force field, water model, membrane model, and channel conformation).
Numerical Study of Pressure Response to Action Potential by Water Permeation With Ion Transports

Abstract:Abstract While the permeation mechanism of solute (e.g., ions and glucose) through biological membrane has been studied extensively, the mechanical role of water transport in intracellular phenomena has not received much attention. In the present study, to investigate the effect of water permeation on the intracellular pressure response, a novel permeation flux model through a biological membrane is developed by incorporating the coupling permeabilities (between water and ion fluxes) as the water–ion interaction in the ion channels. The proposed model is applied to a two–dimensional permeation problem of water and ions in a closed cell separated by a thin membrane. The permeation flux model reproduces the typical time response of intracellular pressure to action potentials with reasonable agreement with experimental results in the literature, indicating that the pressure response can be characterized by the following three parameters: water permeability, the mass ratio of water and ion, and the ratio of the permeation fluxes of water and ion. In particular, the permeation flux ratio plays an essential role in intracellular phenomena; depending on the value of the permeation flux ratio, the time lag between the action potential and the pressure response is 0.1 times smaller than that expected by the previous researchers, indicating that water transport associated with ions may trigger a pressure response. This study demonstrates the importance of water permeation in intracellular mechanical response through coupling of the fluid motion and electric fields.

Abstract:I am delighted to welcome you to this short, special edition on compassion-focused therapy (CFT). One of the core principles of CFT is that it is rooted in clinical science rather than any particular approach to therapy. If we use psychological science to better understand the way the brain evolved, functions, and can create mental health problems, we can begin to target and develop therapeutic interventions based on that science. Indeed, clinical psychology is at a point where it can begin to be less focused on allegiance to particular schools of therapy and root itself in basic science. We can distinguish the science of process from the science of intervention because they have not developed in tandem. There is considerable evidence now that a whole range of interventions that involve: developing an appropriate and supportive therapeutic relationship, creating opportunities for guided discovery and behavioural experiments, gradual exposure for avoided situations and emotions, practising imagery, learning behavioural and emotional regulation skills – to name a few – play a central role in the personal change and recovery journey. However, CFT suggests that how individuals engage in these processes can play a big role in their willingness to engage with them and how effective they are. Compassion-focused therapy highlights the fact that if we look at the evolution of our minds then one thing stands out above all else – the human brain evolved for social processing (Cozolino, 2007, 2008; Siegel, 2012). Over 100 millions years ago, the advent of the mammalian ‘caring for offspring’ and group living introduced a whole range of emotion regulation processes that work through relationships – one of which was obviously the attachment system and capacity for affiliation and caring (Carter, 1998; Porges, 2007). Much of our evolution of intelligence was driven by social challenges and our cognitive architecture is adapted for social processing (Dunbar, 2010). Social relationships matter immensely to our well-being in many ways. For example, the extent anddegree of affiliation thatweexperience from the daywe are born to the day thatwedie has a huge impact on the quality of our lives (Cozolino, 2007, 2008). Affiliative processes influence the architecture of our motivational and emotional systems and even influence genetic expression (Belsky & Pluess, 2009; Siegel, 2012). So, if we put the capacity for affiliation central to the regulation of mind this directs attention to how people can experience kindness and affiliation from others, show kindness, empathy and affiliation toothers and relate tooneself in thoseways. Lipid-mediated hydrophobic gating in the BK potassium channel

Abstract:The large-conductance, calcium-activated potassium (BK) channel lacks the typical intracellular bundle-crossing gate present in most ion channels of the 6TM family. This observation, initially inferred from Ca$^{2+}$-free-pore accessibility experiments and recently corroborated by a CryoEM structure of the non-conductive state, raises a puzzling question: how can gating occur in absence of steric hindrance? To answer this question, we carried out molecular simulations and accurate free energy calculations to obtain a microscopic picture of the sequence of events that, starting from a Ca$^{2+}$-free state leads to ion conduction upon Ca$^{2+}$ binding. Our results highlight an unexpected role for annular lipids, which turn out to be an integral part of the gating machinery. Due to the presence of fenestrations, the "closed" Ca$^{2+}$-free pore can be occupied by the methyl groups from the lipid alkyl chains. This dynamic occupancy triggers and stabilizes the nucleation of a vapor bubble into the inner pore cavity, thus hindering ion conduction. By contrast, Ca$^{2+}$ binding results into a displacement of these lipids outside the inner cavity, lowering the hydrophobicity of this region and thus allowing for pore hydration and conduction. This lipid-mediated hydrophobic gating rationalizes several seemingly problematic experimental observations, including the state-dependent pore accessibility of blockers.

Abstract:Significance Calcium channel dysfunction is implicated in cardiac arrhythmias and immunodeficiency disorders. The recent publication of the crystal structure of a calcium ion channel denoted CRAC has allowed us to use atomic-level computer simulation to begin to understand the mechanism of ion permeation through this ubiquitous and vital cell entry pathway. Our computations identify pore water molecules as a crucial contributor in governing the channel conductance characteristics not only of the wild-type protein but also a disease-related mutant. The present study should be helpful for research designed to improve treatment of immune diseases caused by disrupted calcium signaling.
