Abstract:The purpose of this study was to explore whether essential amino acids (EAAs) in the particulate and sedimentary organic matter (OM) can potentially limit the nutritional needs for benthic macrofauna of different phyla, and which factors regulate the nutritional value with respect to EAAs in a large river-dominated shelf sea, the East China Sea. The EAA excess percentage of threonine, leucine, phenylalanine, histidine and arginine in suspended particles was significantly higher than those in surface sediments (p < 0.01), suggesting that these EAAs for the macrofauna were from fresher suspended particles' source, while methionine was from more degraded organic matter in surface sediments. The EAA excess percentage of valine, isoleucine and lysine showed no significant difference between suspended particles and sediments, indicating that macrofauna may uptake these EAAs from both suspended particle and sediment sources. Deeper offshore region had a lower nutritional potential for valine, leucine, isoleucine and phenylalanine. Bottom dissolved oxygen was not a regulator of the nutritional potential, due to dynamic oxygen conditions and much lower oxygen thresholds for the survival of most macrofauna.

Abstract:Essential biomolecules can critically influence the performance of consumers. A deficiency in dietary sterols has been shown to constrain the food quality of prokaryotic food sources for aquatic consumers. Here, we assessed the importance of dietary cholesterol for life history traits (survival, growth and egg production) of the freshwater herbivore Daphnia magna in supplementation experiments with various sterol‐containing eukaryotic phytoplankton diets (dinoflagellates, diatoms, and golden algae). We combined cholesterol supplementation via liposomes with 13C‐labelling of the phytoplankton diets and traced the origin of cholesterol in Daphnia using compound‐specific stable isotope analysis. All phytoplankton strains used here were rich in polyunsaturated fatty acids (PUFA) but differed in their phytosterol composition. We show that growth and reproduction of D. magna can be limited by sterols even when feeding on sterol‐containing eukaryotic phytoplankton diets. The impact of cholesterol supplementation on growth and reproduction of D. magna differed among phytoplankton diets (strains). The positive effect of cholesterol supplementation was most pronounced on diatom diets. Estimation of source proportions using stable isotopes revealed that D. magna preferentially assimilated the supplemented cholesterol rather than synthesizing it from dietary phytosterols. Our experiments suggest that the different responses to cholesterol supplementation on the various phytoplankton diets were unrelated to the suitability of dietary phytosterols to serve as cholesterol precursors but were caused by other biochemical or morphological food quality constraints. The combination of methods applied here could be very useful for uncovering nutritional constraints and thus for assessing the importance of essential biomolecules for the performance of herbivorous consumers.

Abstract:The objective of this study was to investigate the effects of black sea bream, Acanthopagrus schlegelii, juveniles fed with arginine‐deficient and replete diets. One hundred and eighty fish were randomly allocated into three arginine level diets with triplicate groups: T1, 1.88% (arginine deficient diet without supplementation); T2, 2.49% (supplemented with 0.6% arginine); and T3, 3.13% (supplemented with 1.2% arginine) of dietary arginine, respectively. The results showed that the daily growth increments of fish in T1 group was significantly lower than those of fish fed T2 and T3 diets, while no significant differences were observed between two replete arginine treatments. Fish fed with replete arginine diets had higher pepsin/trypsin activities in stomach, foregut, and midgut. Amylase activities in stomach and foregut were also enhanced in the arginine groups than the T1 group. Arginine and EAA content of dorsal muscle increased significantly with dietary increasing arginine levels. The relative mRNA expression levels of growth hormone receptor gene in liver and insulin‐like growth factor‐I (IGF‐I) gene in dorsal muscle were stimulated significantly by dietary arginine supplementation, but no significant differences were observed between T2 and T3 diets. IGF‐I expression in liver was markedly increased with dietary increasing arginine level. The present findings indicated that black sea bream fed with replete arginine diets could improve growth performance by enhancing pepsin/trypsin and amylase enzymes activities, as well as stimulating growth‐related genes expression level.
Fear and food: Effects of predator‐derived chemical cues and stoichiometric food quality on Daphnia

Abstract:While resource quality and predator‐derived chemical cues can each have profound effects on zooplankton populations and their function in ecosystems, the strength and direction of their interactive effects remain unclear. We conducted laboratory experiments to evaluate how stoichiometric food quality (i.e., algal carbon [C] : phosphorus [P] ratios) affects responses of the zooplankter, Daphnia pulicaria, to predator‐derived chemical cues. We compared growth rates, body P content, metabolic rates, life‐history shifts, and survival of differentially P‐nourished Daphnia in the presence and absence of chemical cues derived from fish predators. We found effects of predator cues and/or stoichiometric food quality on all measured traits of Daphnia. Exposure to fish cues led to reduced growth and increased metabolic rates but had little effect on the body %P content of Daphnia. Elevated algal C : P ratios reduced growth and body %P and increased mass‐specific respiration rates. While most of the effects of predator cues and algal C : P ratios of Daphnia were non‐interactive, reduced survival and relatedly reduced population growth rates that resulted from P‐poor food were amplified in the presence of predator‐derived cues. Our results demonstrate that stoichiometric food quality interacts with antipredator responses of Daphnia, but these effects are largely trait dependent and appear connected to animal life‐history evolution. Given the ubiquity of predators and P‐poor food in lake ecosystems, our results highlight the importance of the interactive responses of animals to predator cues and poor nutrition.

Abstract:Abstract The nutritional quality of phytoplankton is essential for the fitness of herbivorous zooplankton and for efficient carbon fluxes in pelagic ecosystems. In freshwater lakes, cladocerans and calanoid copepods are the main pelagic herbivores in terms of both numbers and grazing impact. However, most studies focused on the easily cultivable cladocerans, while only few studies addressed the impact of the diet on freshwater calanoid copepods due to their more complex life cycle. We here supplied five different phytoplankton diets to the freshwater calanoid copepod Eudiaptomus sp. to investigate their dietary quality for the copepods’ fitness traits over the copepod’s entire life cycle. While all tested diets supported comparable reproductive success in adults, egg production, hatching success and survival rate differed markedly between diets. In the offspring generation, diet affected developmental and reproductive periods, size at first reproduction and clutch size. Eudiaptomus body fatty acid composition only partially reflected their diet, indicating that the copepods are able to selectively accumulate and interconvert certain essential fatty acids. This capability may allow them to cope with nutritional deficiencies and may thus be interpreted as an ecological adaptation strategy to the fluctuating environmental conditions and resource availabilities in freshwater plankton.

Abstract:Abstract Diet composition, especially the relative abundance of key macronutrients, is well known to affect animal wellbeing by changing reproductive output, metabolism and length of life. However, less attention has been paid to the ways the quality of these nutrients modify these macronutrient interactions. Nutritional Geometry can be used to model the effects of multiple dietary components on life-history traits and to compare these responses when diet quality is varied. Previous studies have shown that dietary protein quality can be increased for egg production in Drosophila melanogaster by matching the dietary amino acid proportions to the balance of amino acids used by the sum of proteins in the fly’s in silico translated exome. Here, we show that dietary protein quality dramatically alters the effect of protein quantity on female reproduction across a broad range of diets varying in both protein and carbohydrate concentrations. These data show that when sources of ingredients vary, their relative value to the consumer can vastly differ and yield very different physiological outcomes. Such variations could be particularly important for meta analyses that look to draw generalisable conclusions from diverse studies.

Abstract:The first few months of life is the most vulnerable period for fish and their optimal hatching time with zooplankton prey is favored by natural selection. Traditionally, however, prey abundance (i.e., zooplankton density) has been considered important, whereas prey nutritional composition has been largely neglected in natural settings. High-quality zooplankton, rich in both essential amino acids (EAAs) and fatty acids (FAs), are required as starting prey to initiate development and fast juvenile growth. Prey quality is dependent on environmental conditions, and, for example, eutrophication and browning are two major factors defining primary producer community structures that will directly determine the nutritional quality of the basal food sources (algae, bacteria, terrestrial matter) for zooplankton. We experimentally tested how eutrophication and browning affect the growth and survival of juvenile rainbow trout (Oncorhynchus mykiss) by changing the quality of basal resources. We fed the fish on herbivorous zooplankton (Daphnia) grown with foods of different nutritional quality (algae, bacteria, terrestrial matter), and used GC-MS, stable isotope labeling as well as bulk and compound-specific stable isotope analyses for detecting the effects of different diets on the nutritional status of fish. The content of EAAs and omega-3 (ω-3) polyunsaturated FAs (PUFAs) in basal foods and zooplankton decreased in both eutrophication and browning treatments. The decrease in ω-3 PUFA and especially docosahexaenoic acid (DHA) was reflected to fish juveniles, but they were able to compensate for low availability of EAAs in their food. Therefore, the reduced growth and survival of the juvenile fish was linked to the low availability of DHA. Fish showed very low ability to convert alpha-linolenic acid (ALA) to DHA. We conclude that eutrophication and browning decrease the availability of the originally phytoplankton-derived DHA for zooplankton and juvenile fish, suggesting bottom-up regulation of food web quality.

Evaluating food quality of Arthrospira platensis for culturing Daphnia magna

Abstract:Arthrospira is a filamentous cyanobacterium, rich in proteins and bioactive compounds, which is a dietary supplement for humans and animals. However, the contribution of this "superfood" on the life history traits of zooplankton is poorly addressed. We conducted Daphnia magna growth experiment using Arthrospira platensis comparing it with Chlamydomonas reinhardtii as quality food, to evaluate the nutritional adequacy of A. platensis for rearing daphnids. The trichomes of A. platensis was fragmented using a bead crusher for easier ingestion by the daphnids. The growth experiments revealed that A. platensis alone did not support both survival and growth of D. magna , but the daphnid survival increased after the addition of 20% C. reinhardtii. When A. platensis was supplemented with 50% and more of C. reinhardtii , however, growth was better than to those daphnids exclusively fed on C. reinhardtii . This suggests that A. platensis have some nutritional limitations essential for daphnids survival and growth instead of less digestibility or toxicity. Carbon (C) to Phosphorus (P) ratio (C:P = 46) of A. platensis was far below the threshold for daphnid growth (C:P = 116). This implies that P limitation is unlikely in A. platensis as feed for daphnids. Although the lack of some essential biochemicals such as sterols and polyunsaturated fatty acids may be a more plausible explanation, a small addition of A. platensis to standard microalgal foods would result in better growth of zooplankton, including D. magna .

Abstract:Zooplankton displays different inducible defenses against invertebrate and vertebrate predators. The response pattern to gape‐limited invertebrate predators involves increased somatic growth and offspring body size but delayed maturity and reduced offspring numbers. In contrast to this general pattern, the freshwater model organism Daphnia magna has been reported to exhibit a different response when encountering the gape‐limited tadpole shrimp Triops cancriformis. Under laboratory conditions, D. magna showed increased somatic growth, earlier maturation, and an increase in both offspring number and size. We propose here that the discrepancy between the previously observed and the theory‐based response patterns against invertebrate predators is due to differences in food availability in the applied laboratory settings and assessed whether the defensive response of D. magna against T. cancriformis is modulated differently by food quantity and quality. We found a strong impact of food quantity and quality on the defense response of D. magna to T. cancriformis kairomones. The prey seem to be able to overcome trade‐offs between morphological defense traits and reproductive traits, but distinctly between high food quantity and high food quality. Thereby, reproductive traits were preferred over morphological defenses. Furthermore, the removal of particles from the T. cancriformis‐conditioned water caused a defense pattern in D. magna that was consistent with the general response pattern known from other invertebrate predators, thus explaining the described discrepancy to previous studies with T. cancriformis. Our study highlights the importance of assessing food‐related effects on predator–prey interactions to understand trophic relationships and food web processes.

Abstract:Ecological stoichiometry predicts that the extent to which herbivores are limited by nutrients depends on their metabolic demands. Nonconsumptive effects (NCEs) caused by predators have been demonstrated to trigger not only inducible defenses but also physiological stresses in prey. Stressed prey always exhibit increased metabolism and decreased nutrient deposition (e.g., nitrogen and phosphorus) as physiological stress responses to their predators. Consequently, prey exposed to predation risk should reduce demands for nutrients intake and become adaptive to nutrient limitations. In this study, results obtained from classic Brachionus – Asplanchna model revealed that Brachionus calyciflorus fed with full nutrient algal food showed energetic costs, e.g., reduced investment in asexual (lifetime fecundity) and sexual (mixis ratio) reproductions, in response to predation risk. However, reduction in sexual and asexual reproductions diminished when herbivorous Brachionus were supplied with nitrogen- or phosphorus-limited algal food. Moreover, long posterolateral spines and high posterolateral spine-body length ratio developed in either nitrogen- or phosphorus-deficient conditions containing Asplanchna kairomones, indicating an additive effect of nutrient limitation and predation risk on posterolateral spine development in Brachionus . The findings of this study highlighted predator-facilitated adaptation of prey to nutrient limitations of primary producers, which may have implications in understanding roles of NCEs in regulating trophic interactions.
Grazing resistance and poor food quality of a widespread mixotroph impair zooplankton secondary production

Abstract:Abstract Growing evidence suggests that global climate change promotes the dominance of mixotrophic algae especially in oligotrophic aquatic ecosystems. While theory predicts that mixotrophy increases trophic transfer efficiency in aquatic food webs, deleterious effects of some mixotrophs on consumers have also been reported. Here, using a widespread mixotrophic algal genus Dinobryon , we aimed to quantify how colonial taxa contribute to secondary production in lakes. We, therefore, studied the dietary effects of Dinobryon divergens on Cladocera ( Daphnia longispina ) and Copepoda ( Eudiaptomus gracilis ), representing two main taxonomic and functional groups of zooplankton. In feeding experiments, we showed that Dinobryon was largely grazing resistant and even inhibited the uptake of the high-quality reference food in Daphnia. Eudiaptomus could to some extent compensate with selective feeding, but a negative long-term food quality effect was also evident. Besides, Eudiaptomus was more sensitive to the pure diet of Dinobryon than Daphnia . Low lipid content and high C:P elemental ratio further supported the low nutritional value of the mixotroph. In a stable isotope approach analysing a natural plankton community, we found further evidence that carbon of Dinobryon was not conveyed efficiently to zooplankton. Our results show that the increasing dominance of colonial mixotrophs can result in reduced dietary energy transfer to consumers at higher trophic levels. In a wider perspective, global climate change favours the dominance of some detrimental mixotrophic algae which may constrain pelagic trophic transfer efficiency in oligotrophic systems, similarly to cyanobacteria in eutrophic lakes.

Abstract:<ol class=""><li>According to the River Continuum Concept, headwater streams are richer in allochthonous (e.g. terrestrial leaves) than autochthonous (e.g. algae) sources of organic matter for consumers. However, compared to algae, leaf litter is of lower food quality, particularly ω‐3 polyunsaturated fatty acids (n‐3 PUFA), and would constrain the somatic growth, maintenance, and reproduction of stream invertebrates. It may be thus assumed that shredders, such as Gammarus, receive lower quality diets than grazers, e.g. Ecdyonurus, that typically feed on algae. </li><li>The objective of this study was to assess the provision of dietary PUFA from leaf litter and algae to the shredder Gammarus and the grazer Ecdyonurus. Three different diets (algae, terrestrial leaves, and an algae–leaf litter mix) were supplied to these macroinvertebrates in a flume experiment for 2 weeks. To differentiate how diet sources were retained in these consumers, algae were isotopically labelled with 13C. </li><li>Both consumers became enriched with 13C in all treatments, demonstrating that both assimilated algae. For Gammarus, n‐3 PUFA increased, whereas n‐6 PUFA stayed constant. By contrast, the n‐3 PUFA content of Ecdyonurus decreased as a consequence of declining algal supply. </li><li>Results from compound‐specific stable isotope analysis provided evidence that the long‐chain n‐3 PUFA eicosapentaenoic acid (EPA) in both consumers was more enriched in 13C than the short‐chain n‐3 PUFA α‐linolenic acid, suggesting that EPA was taken up directly from algae and not from heterotrophic biofilms on leaf litter. Both consumers depended on algae as their carbon and EPA source and retained their EPA from high‐quality algae. </li></ol>

Abstract:Chytrid fungal parasites convert dietary energy and essential dietary molecules, such as long‐chain (LC) polyunsaturated fatty acids (PUFA), from inedible algal/cyanobacteria hosts into edible zoospores. How the improved biochemical PUFA composition of chytrid‐infected diet may extend to zooplankton, linking diet quality to consumer fitness, remains unexplored. Here, we assessed the trophic role of chytrids in supporting dietary energy and PUFA requirements of the crustacean zooplankton Daphnia, when feeding on the filamentous cyanobacterium Planktothrix. Only Daphnia feeding on chytrid‐infected Planktothrix reproduced successfully and had significantly higher survival and growth rates compared with Daphnia feeding on the sole Planktothrix diet. While the presence of chytrids resulted in a two‐fold increase of carbon ingested by Daphnia, carbon assimilation increased by a factor of four, clearly indicating enhanced carbon transfer efficiency with chytrid presence. Bulk carbon (δ13C) and nitrogen (δ15N) stable isotopes did not indicate any treatment‐specific dietary effects on Daphnia, nor differences in trophic position among diet sources and the consumer. Compound‐specific carbon isotopes of fatty acids (δ13CFA), however, revealed that chytrids bioconverted short‐chain to LC‐PUFA, making it available for Daphnia. Chytrids synthesised the ω‐3 PUFA stearidonic acid de novo, which was selectively retained by Daphnia. Values of δ13CFA demonstrated that Daphnia also bioconverted short‐chain to LC‐PUFA. We provide isotopic evidence that chytrids improved the dietary provision of LC‐PUFA for Daphnia and enhanced their fitness. We argue for the existence of a positive feedback loop between enhanced Daphnia growth and herbivory in response to chytrid‐mediated improved diet quality. Chytrids upgrade carbon from the primary producer and facilitate energy and PUFA transfer to primary consumers, potentially also benefitting upper trophic levels of pelagic food webs.

Abstract:The diets of animals are essential to support development, and protein is key. Accumulation of stored nutrients can support developmental events such as molting and initiation of reproduction. Agricultural studies have addressed how dietary protein quality affects growth, but few studies have addressed the effects of dietary protein quality on developmental transitions. Studies on how dietary quality may affect protein storage and development are possible in arthropods, which store proteins in the hemolymph. We hypothesized that diets with a composition of amino acids that matches the precursor of egg yolk protein (vitellogenin, Vg) will be high quality and support both egg production and accumulation of storage proteins. Grasshoppers were fed one of two isonitrogenous solutions of amino acids daily: Vg-balanced (matched to Vg) or Unbalanced (same total moles of amino acids, but not matched to egg yolk). We measured reproduction and storage protein levels in serial hemolymph samples from individuals. The Vg-balanced group had greater reproduction and greater cumulative levels of storage proteins than did the Unbalanced group. This occurred even though amino acids fed to the Vg-balanced group were not a better match to storage protein than were the amino acids fed to the Unbalanced group. Further, oviposition timing was best explained by a combination of diet, age at the maximum level of storage protein hexamerin-270 and accumulation of hexamerin-90. Our study tightens the link between storage proteins and commitment to reproduction, and shows that dietary protein quality is vital for protein storage and reproduction.

Abstract:Changes in food quality can dramatically impair zooplankton fitness, especially in eutrophic water bodies where cyanobacteria are usually predominant. Cyanobacteria are considered a food with low nutritional value, and some species can produce bioactive secondary metabolites reported as toxic to zooplankton. Considering that cyanobacteria can limit the survival, growth and reproduction of zooplankton, we hypothesized that the dietary exposure of neotropical Daphnia species (D. laevis and D. gessneri) to saxitoxin-producing cyanobacteria impairs Daphnia feeding rates and fitness regardless of a high availability of nutritious algae. Life table and grazing assays were conducted with different diets: (1) without nutritional restriction, where neonates were fed with diets at a constant green algae biomass (as a nutritious food source), and an increasing cyanobacterial concentration (toxic and poor food source), and (2) with diets consisting of different proportions of green algae (nutritious) and cyanobacteria (poor food) at a total biomass 1.0 mg C L-1. In general, the presence of high proportions of cyanobacteria promoted a decrease in Daphnia somatic growth, reproduction and the intrinsic rate of population increase (r) in both diets with more pronounced effects in the nutritionally restricted diet (90% R. raciborskii). A two-way ANOVA revealed the significant effects of species/clone and treatments in both assays, with significant interaction between those factors only in the second assay. Regarding the grazing assay, only D. laevis was negatively affected by increased cyanobacterial proportions in the diet. In the life table assay with constant nutritious food, a reduction in the reproduction and the intrinsic rate of the population increase (r) of all species were observed. In conclusion, we found adverse effects of the toxic cyanobacterial strain R. raciborskii on Daphnia fitness, regardless of the constant amount of nutritious food available, proving the toxic effect of R. raciborskii and that the nutritional quality of the food has a greater influence on the fitness of these animals.
Food Quantity and Quality Interactions at Phytoplankton–Zooplankton Interface: Chemical and Reproductive Responses in a Calanoid Copepod

Abstract:Marine food webs form the major component of the biological pump and play a central role in the global carbon (C) cycle. Understanding the response of particular processes in marine food webs to changing environments is a prerequisite to predict changes in ecological functioning in the future ocean. Here, we experimentally assessed the effects of nitrogen:phosphorus (N:P) supply ratios (the molar ratios 10:1, 24:1, and 63:1) on elemental and biochemical quality of marine phytoplankton Rhodomonas sp., and the interactions between food quantity and quality on stoichiometric C:N:P, fatty acids (FAs) and reproductions in copepods Acartia tonsa. Overall, the stoichiometry of A. tonsa was to some extent homeostatic in response to the changing algal C:N and C:P ratios, with significant changes in C:N ratios of A. tonsa observed, especially under higher food quantities. The relative gross growth efficiencies (GGEs) for C and N (and P) were analyzed, revealing that copepods may achieve homeostasis by lowering the GGE for C while increasing it for the limiting nutrient. Egg production rates in A. tonsa were lowest on nutrient deficient diets under low food quantities. Reduced egg production rates may be attributed to the lowered GGEs for C and reduced transfer efficiency of essential FAs between phytoplankton and copepods, indicating interactive-essential effects of elements and FAs on copepod production. Our results highlight that nutrient deficiency in the environments may reduce energy transfer efficiency at the base of food webs by altering phytoplankton chemical composition, which can interact with food quantity and have implications on food web dynamics in the changing ocean.

Abstract:Anti-grazer responses of phytoplankton have been widely documented to vary with external abiotic factors (e. g. nutrient status), suggesting an environment-dependence of phytoplankton-zooplankton dynamics. Here, we examined the trophic interaction between the cyanobacteria Coleofasciculus chthonoplastes and the cladoceran Moina micrura under three nutrient levels from oligotrophic to hypereutrophic conditions (0.3, 3.0 and 100.0 mu M PO43-; molar ratio NO3- : PO43- - 16 : 1) to determine the effect of nutrients on the stability of a phytoplankton-zooplankton system. In the absence of M. micrura, cell density and filament (single-filament) length of C. chthonoplastes were lower at the low nutrient level compared with those at the medium and high nutrient levels. When M. micrura was present, cell density, filament length, colony proportion and size (the mean number of cells per colony) of C. chthonoplastes were highest under the medium nutrient level, corresponding to the lowest amplitude of density fluctuations in M. micrura. Regression analyses revealed that low cell densities of C. chthonoplastes (due to its limited ability to form colonies and long filaments and hence a high consumption rate of M. micrura) at low and high nutrient levels limited the growth of M. micrura adult females, leading to a sharp decline in embryo length, clutch size and population density. In general, our results suggest that the stability of phytoplankton-zooplankton systems varies with nutrients because of the nutrient-dependent morphology in phytoplankton, and imply that extremely low or high nutrient load may destabilize phytoplankton-zooplankton systems.
