Abstract:Phenolic compounds impact human health and the environment in both beneficial and undesirable manners. While some phenols are known to be antioxidants, others function as hormones or neurotransmitters, some are significant environmental contaminants, and others have the potential to cause cancer or disturb the endocrine system. To track the amount of toxicity, it is essential to identify and measure these phenols in food, the environment, and human samples. A novel phenol sensing amperometric enzymatic biosensor with a gold electrode was fabricated based on covalent immobilisation of synthesised laccase nanoparticles (LacNPs). The process of synthesising laccase nanoparticles was examined using UV–visible spectrophotometry, FTIR, transmission electron microscopy, zeta potential, and dynamic light scattering techniques. For analysis of electrode fabrication (LacNPs-AuE), scanning electron microscopy, cyclic voltammetry, and electrochemical impedance spectra including Nyquist plots and Bode's plot were examined. Optimisation and evaluation of fabricated biosensor were investigated using cyclic voltammetry studies. The LacNPs-AuE biosensor's overall characteristics were enhanced by the direct immobilisation of laccase nanoparticles, enabling the analysis at a lower detection limit (0.3 μM), wider linear range (0.1–100 μM and 100 to 600 μM), faster response time (3 s), and high recovery (92–98%). Tea, alcohol, and pharmaceutical samples were tested for total phenolic content using the biosensor; the results were compared with spectrophotometric data.

Construction of Bacterial Laccase Displayed on the Microbial Surface for Ultrasensitive Biosensing of Phenolic Pollutants with Nanohybrids-Enhanced Performance.

Abstract:Although bacterial laccase (BLac) has many advantages including short fermentation period and adaptable activity to wide temperature and pH ranges, it is of challenge and significance to apply BLac to the biosensors, due to the intracellular secretion and poor electron transfer efficiency of BLac. Here, cell surface-displayed BLac (CSDBLac) was successfully constructed as whole-cell biocatalyst through microbial surface display technology, eliminating the mass transfer restriction and laborious purification steps. Meanwhile, MXenes/polyetherimide-multiwalled carbon nanotubes (MXenes/PEI-MWCNTs) nanohybrids were designed to immobilize CSDBLac and improve their electrochemical activity. Then, an electrochemical biosensor was successfully constructed to detect common phenolic pollutants (catechol and hydroquinone) by the co-immobilization of CSDBLac and MXenes/PEI-MWCNTs nanohybrids onto a glassy carbon electrode. Subsequently, it was successfully applied to the water samples assay with good reliability and repeatability. This work innovatively used BLac and nanohybrid as the core elements of biosensor, which not only effectively solved the application bottleneck of BLac on biosensors, but also dramatically promote the electro transfer efficiency between whole-cell biocatalyst and electrode. This method is of profound meanings for significantly improving the performance of phenolic biosensors and other biosensors from the origin.

Abstract:A biosensor was fabricated by immobilizing glucose oxidase (GOD) into nanostructured graphene (GRA)-conducting polyaniline (PANI) nanocomposite, which was based on electrochemical polymerization of aniline in GRA synthesized by using electrochemical expansion of graphite in propylene carbonate electrolyte. Scanning electron spectroscopy (SEM), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) were used to characterize the morphology and performance of the as-prepared biosensor, respectively. Amperometric measurements were carried out to optimize test conditions (pH and applied potential) of the biosensor. Under the optimal conditions, the biosensor showed a linear range from 10.0μM to 1.48mM (R2=0.9988) with a sensitivity of 22.1μAmM−1cm−2, and a detection limit of 2.769μM (S/N=3). The apparent Michaelis–Menten constant (KMa) was estimated to be 3.26mM. The interference from glycine (Gly), D-galactose (D-Gal), urea (Urea), L-phenylalanine (L-Phe), ascorbic acid (AA), and L-tyrosine (L-Tyr) was also investigated. The results indicated that the biosensor exhibit high sensitivity and superior selectivity, providing a hopeful candidate for glucose biosensing.

Abstract:Polyaniline nanotube was prepared with aid of lactic acid assembly. A chemical prototype sensor was constructed based on nanotube-structured polyaniline and two typical structured chemical sensors, one is an interdigital electrodes on flexible polymer substrate, the other is a highly sensitive quartz resonators. The gas-sensing behavior of the two typical sensors to some organic vapors operating at room temperature was investigated. Results showed rapid response, and good reversibility of these two typical sensors. The response time is 10-20 s, the reversible time is about 100 s. This study provides a simple approach for preparation of materials needed for a chemical sensor to selected organic volatiles with rapid response or organic flexible nanoelectronic devices.

Abstract:The use of nanomaterials allows the design of ultrasensitive biosensors with advantages in the detection of organic molecules. Catechol and catechin are molecules that occur naturally in fruits, and their presence in products like dyes and wines affects quality standards. In this study, catechol and catechin were measured at the nanoscale by means of cyclic voltammetry. The oxidation of Coriolopsis gallica laccase immobilized on nitrogen-doped multiwalled carbon nanotubes (Lac/CN x -MWCNT) and on graphene oxide (Lac/GO) was used to measure the concentrations of catechol and catechin. Nitrogen-doped multiwalled carbon nanotubes (CN x -MWCNT) were synthesized by spray pyrolysis and characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), and x-ray photoelectron spectroscopy (XPS). Covalently bonded hybrids with laccase (Lac/CN x -MWCNT and Lac/GO) were generated. Catalytic activity of free enzymes determined with syringaldazine yielded 14 584 UmL-1. With Lac/CN x -MWCNT at concentrations of 6.4 mmol L-1 activity was 9326 U mL-1, while enzyme activity measured with Lac/GO at concentration of 6.4 mmol L-1 was 9 234 U mL-1. The Lac/CN x -MWCNT hybrid showed higher stability than Lac/GO at different ethyl alcohol concentrations. The Lac/CN x -MWCNT hybrid can measure concentrations, not previously reported, as low as 1 × 10-8 mol L-1 by measuring the electric current responses.

Abstract:The successful fabrication of biosensors is greatly limited by the immobilization of their bioreceptor, thus we propose a facile and reproducible two-step method to modify graphite electrodes with a bacterial laccase, relying on a fast and controllable potentiostatic process to coat graphite surfaces with biomolecule-compatible thin films of polynorepinephrine (ePNE) and polydopamine (ePDA). Both polymers, synthesized with a similar thickness, were functionalized with bacterial laccase, displaying distinct electrochemical transducing behaviours at pH 5.0 and 7.0. ePNE layer enables adequate electron transfer of anionic and cationic species in acidic and neutral media, whereas transduction across ePDA strongly depends on pH and redox probe charge. ePNE stands out by improving the amperometric responses of the biosensing interface towards a phenolic acid (gallic acid) and a flavonoid (catechin), in respect to ePDA. The optimal graphite/ePNE/laccase interface outperforms biosensing interfaces based on fungal laccases at neutral pH, displaying detection sensitivities of 104 and 14.4 µA cm-2 mM-1for gallic acid and catechin, respectively. The fine synthetic control of the ePNE bio-inspired transduction layer and the use of an alkaliphilic bacterial laccase enabled the construction of an amperometric biosensing interface with extended pH range of polyphenols detection present in food products and agro-industrial waste.

Abstract:This study describes the development of a highly sensitive amperometric biosensor for the analysis of phenolic compounds such as catechol. The biosensor architecture is based on the immobilization of tyrosinase (Tyr) on a screen-printed carbon electrode (SPE) modified with nanodiamond particles (ND), 1-butyl-3-methylimidazolium hexafluorophosphate (IL) and poly-l-lysine (PLL). Surface morphologies of the electrodes during the modification process were evaluated by scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDX). Cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) were used to investigate the electrochemical characteristics of the modified electrodes. Owing to the synergistic effect of the modification materials, the Tyr/PLL/ND-IL/SPE exhibited high sensitivity (328.2 μA mM-1) towards catechol with a wide linear range (5.0 × 10-8 - 1.2 × 10-5 M) and low detection limit (1.1 × 10-8 M). Furthermore, the method demonstrated good reproducibility and stability. The amperometric response of the biosensor towards other phenolic compounds such as bisphenol A, phenol, p-nitrophenol, m-cresol, p-cresol and o-cresol was also investigated. The analytical applicability of the biosensor was tested by the analysis of catechol in tap water. The results of the tap water analysis showed that the Tyr/PLL/ND-IL/SPE can be used as a practical and effective method for catechol determination.

Abstract:A novel polyaniline–ionic liquid–carbon nanofiber (PANI–IL–CNF) composite was greenly prepared by in situ one-step electropolymerization of aniline in the presence of IL and CNF for fabrication of amperometric biosensors. The scanning electron micrographs confirmed that the PANI uniformly grew along with the structure of CNF and the PANI–IL–CNF composite film showed a fibrillar morphology with the diameter of around 95nm. A phenol biosensor was constructed by immobilizing tyrosinase on the surface of the composite modified glassy carbon electrode via the cross-linking step with glutaraldehyde. The biosensor exhibited a wide linear response to catechol ranging from 4.0×10−10 to 2.1×10−6M with a high sensitivity of 296±4AM−1cm−2, a limit of detection down to 0.1nM at the signal to noise ratio of 3 and applied potential of −0.05V. According to the Arrhenius equation, the activation energy for enzymatic reaction was calculated to be 38.8kJmol−1 using catechol as the substrate. The apparent Michaelis–Menten constants of the enzyme electrode were estimated to be 1.44, 1.33, 1.16, 0.65μM for catechol, p-cresol, phenol, m-cresol, respectively. The functionalization of CNF with PANI in IL provided good biocompatible platform for biosensing and biocatalysis.

Abstract:A novel biosensor for phenols was constructed by immobilizing polyphenol oxidase (PPO) into polyaniline–polyacrylonitrile composite matrix. The sensing film was prepared by electropolymerization of aniline into polyacrylonitrile (PAN)-coated platinum electrode in the presence of PPO. The scanning electron micrographs (SEM) showed that PAN had microporous structure and polyaniline and the enzyme could co-intercalated into PAN matrix. The obtained biosensor exhibited high sensitivity and excellent stability, which had no apparent loss of activity after 100 consecutive measurements and intermittent usage for 8 months with storage in a phosphate buffer at 4°C. The construction and operational conditions of the enzyme electrode were optimized. The sensitivities of the enzyme electrode for phenol, p-cresol, m-cresol and catechol were 0.96, 1.38, 1.5 and 2.03 AM−1cm−2, respectively.

Abstract:Bienzyme electrode with horseradish peroxidase (HRP) and glucose oxidase (GOD) multilayers was constructed based on sugar-lectin biospecific interactions for amperometric determination of phenolic compounds and aromatic amines. Atomic force microscopy (AFM) was applied to monitor the uniform layer-by-layer assembly of concanavalin A (Con A) and HRP or GOD on polyelectrolyte precursor film-modified Au electrode. Substituted phenolic compounds and aromatic amines could be determined with in situ generation of H2O2 by GOD-catalyzed oxidation of glucose. The parameters of the biosensor including the number of assembled HRP and GOD layer, and the concentrations of glucose were optimized. The linear range for the determination of catechol and p-phenyldiamine was 6.0-60.0 mu mol L-1 and 7.6-68.4 mu mol L-1 with detection limit of 0.9 mu mol L-1 and 0.4 mu mol L-1, respectively. The biosensor possessed high sensitivity and fast response for phenolic compounds and 95% of the maximum response could be reached in about 3 s. Glucose, ascorbic acid, tartaric acid, citric acid and starch exhibited no interference for the detection. Construction of Platinum Nanozyme by Using Carboxymethylcellulose with Improved Laccase-Like Activity for Phenolic Compounds Detection

Abstract:Natural laccases are multicopper oxidases widely applied in enzymatic biotransformation. However, using natural laccases in real-world settings is expensive and usually hampered by their stability. Most reported laccase-like nanozymes are limited to copper-containing, while other materials have yet to receive sufficient attention. Herein, platinum (Pt) nanoparticles (NPs) have been constructed as a laccase-like nanozyme using sodium carboxymethylcellulose (CMC-PtNPs). The as-prepared CMC-PtNPs have small particle size (3.8 ± 0.03 nm), good dispersion, and clear interaction mode between polysaccharides and nanozyme. CMC-PtNPs were found to own applicable laccase-like activity by producing O 2 •− as oxidants to oxidize laccase-related substrates. According to the steady-state dynamic assay, the apparent K m value of CMC-PtNPs was calculated as 0.218 mM. Compared with natural laccase or laccase-like materials, the laccase-like CMC-PtNPs have the advantage of higher substrate affinity and catalytic effect. More significantly, CMC-PtNPs were found to be highly stable at wide-ranged pH and temperature. On a practical level, the adequacy of the developed CMC-PtNPs was also confirmed by the efficient detection of the laccase-related phenolic compounds (e.g., 2,4-dichlorophenol, dopamine, noradrenaline, and adrenaline). A highly linear relationship (R 2 = 0.994) was obtained by plotting the absorbance at 510 nm and 2,4-DCP concentration in a range of 6.25 to 225 μM with a low limit of detection (LOD) of 900 nM. Also, the colorimetric sensor exhibited outstanding sensing performance toward other phenolic compounds. Looking forward, this work provides a new design of laccase-like Pt nanozyme and demonstrates its utilization potentiality in nanozyme research, environmental remediation, and other fields.

Conduction velocities of corticospinal axons in the rat studied by recording cortical antidromic responses.

Abstract:The rat corticospinal tract was stimulated at the medullary pyramid and at different levels in the spinal cord (segments C2/3, T2, T12) and responses were recorded from the surface of the cerebral cortex and extracellularly from individual cortical neurones. Irrespective of the site stimulated, the earliest surface and single unit responses had frequency‐following and other characteristics which indicated they resulted from antidromic invasion of corticospinal neurones. Synaptically mediated discharges with longer latency were also evoked in cortical neurones other than corticospinal neurones. At least in part these discharges probably resulted from stimulus spread to the dorsal column‐medial lemniscus pathway. Corticospinal neurones were almost all between 1.0 and 1.5 mm beneath the cortical surface while synaptically excited units were at all depths greater than 0.4 mm. By stimulating at two sites, estimates of conduction velocity were obtained for single corticospinal axons. For those reaching at least as far as T12, velocities caudal to the pyramid ranged from 5 to 19 m/s (mean 11.4 +/‐ 2.9 m/s; S.D.). Slow axons in the pyramid (antidromic latency greater than 2.5 ms) could rarely be excited from T12. By stimulating at three sites (pyramid, T2, T12) most axons reaching T12 were found to have similar conduction velocities in the ‘cervical’ (pyramid‐T2) and ‘thoracic’ (T2‐T12) cord. However, in 15% of the axons the ‘thoracic’ velocity was at least 25% less than the cervical. The results are discussed and related to those from previous investigations.

Abstract:Phenolic compounds (PhCs) are ubiquitously distributed phytochemicals found in many plants, body fluids, food items, medicines, pesticides, dyes, etc. Many PhCs are priority pollutants that are highly toxic, teratogenic, and carcinogenic. Some of these are present in body fluids and affect metabolism, while others possess numerous bioactive properties such as retaining antioxidant and antimicrobial activity in plants and food products. Therefore, there is an urgency for developing an effective, rapid, sensitive, and reliable tool for the analysis of these PhCs to address their environmental and health concern. In this context, carbonaceous nanomaterials have emerged as a promising material for the fabrication of electrochemical biosensors as they provide remarkable characteristics such as lightweight, high surface: volume, excellent conductivity, extraordinary tensile strength, and biocompatibility. This review outlines the current status of the applications of carbonaceous nanomaterials (CNTs, graphene, etc.) based enzymatic electrochemical biosensors for the detection of PhCs. Efforts have also been made to discuss the mechanism of action of the laccase enzyme for the detection of PhCs. The limitations, advanced emerging carbon-based material, current state of artificial intelligence in PhCs detection, and future scopes have also been summarized.

Reagentless Biosensor for Phenolic Compounds Based on Tyrosinase Entrapped Within Gelatine Film

Abstract:A simple and new reagentless phenolic compound biosensor was constructed with tyrosinase immobilized in the gelatine matrix cross-linked with formaldehyde. The morphologies of gelatine and gelatine/tryosinase were characterized by SEM. The tyrosinase retains its bioactivity when being immobilized by the gelatine film. Phenolic compounds were determined by the direct reduction of biocatalytically liberated quinone at -0.1 V vs SCE. The process parameters for the fabrication of the enzyme electrode were studied. Optimization of the experimental parameters has been performed with regard to pH, operating potential, temperature and storage stability. This biosensor exhibits a fast amperometric response to phenolic compounds. The linear range for catechol, phenol, and p-Cresol determination was from 5x10(-8) to 1.4x10(-4) M, 5x10(-8) to 7.1x10(-5) M, and 1x10(-7) to 3.6x10(-5) M, with a detection limit of 2.1x10(-8) M, 1.5x10(-8) M, and 7.1x10(-8) M, respectively. The enzyme electrode retained ca.77% of its activity after 7 days of storage at 4 degrees C in a dry state. The proposed sensor presented good repeatability, evaluated in terms of relative standard deviation (R.S.D.=8.6%) for eight different biosensors and was applied for determination in water sample. The recovery for the sample was from 99.0% to 99.8%.
Glottic and cervical tracheal narrowing in patients with obstructive sleep apnea.

Abstract:There are several studies showing that patients with idiopathic obstructive sleep apnea (OSA) have a narrow and collapsible pharynx that may predispose them to repeated upper airway occlusions during sleep. We hypothesized that this structural abnormality may also extend to the glottic and tracheal region. Consequently, we measured pharyngeal (Aph), glottic (Agl), cervical tracheal (Atr1), midtracheal (Atr2), and distal (Atr3) tracheal areas during tidal breathing in 66 patients with OSA (16 nonobese and 50 obese) and 8 nonapneic controls. We found that Aph, Agl, and Atr1, but not Atr2 or Atr3, were significantly smaller in the OSA group than in the control group. Obese patients with OSA had the smallest upper airway area, although the nonapneic controls had the largest areas. Multiple linear regression analysis revealed that the pharyngeal area, cervical tracheal area, and body mass index were all independent determinants of the apnea-hypopnea index, accounting for 31% of the variability in apnea-hypopnea index. Aph, Agl, and Atr showed significant correlation with the body mass index. We conclude that sleep-disordered breathing is associated with diffuse upper airway narrowing and that obesity contributes to this narrowing. Furthermore, we speculate that a common pathophysiological mechanism may be responsible for this reduction in upper airway area extending from the pharynx to the proximal trachea.

Abstract:Magnesium-containing mesoporous silica sieve (Mg-MCM-41) provided a suitable immobilization of biomolecule matrix due to its uniform pore structure, high surface areas, fast electron-transfer rate, and good biocompatibility. Based on this, an amperometric biosensor was developed by entrapping laccase into the Mg-MCM-41/PVA composite matrix. Laccase from Trametes versicolor was assembled on a composite film of Mg-MCM-41/PVA modified Au electrode and the electrode was investigated by cyclic voltammetry, impedance spectroscopy, and chronoamperometry. The results indicated that the Mg-MCM-41/PVA/Lac modified electrode exhibited excellent catalytic activity towards catechol at room temperature in pH 4.8 acetate buffer solution. The optimum experimental conditions of biosensor for the detection of catechol were studied in detail. Under the optimal conditions, the linear range was from 0.94 to 10.23 μ M with the sensitivity of 16.9227 A/M, the detection limit of 0.00531 μ M, and the response time of less than 14 s. The Michaelis-Menten constant ( K M app ) was estimated by Lineweaver-Burk equation and the K M app value was about 1.01 μ M. In addition, the biosensor exhibited high reproducibility and long-time stability. This work demonstrated that Mg-MCM-41/PVA composite provides a suitable support for laccase immobilization and construction of biosensor.

Abstract:Polyaniline–polyacrylonitrile composite films prepared by in situ electropolymerization of aniline on polyacrylonitrile-coated platinum electrode were used to immobilize polyphenol oxidase forming an enzyme biosensor. The novel polyphenol oxidase-based biosensor exhibited high sensitivity, low background and excellent stability, which showed no loss of activity after 100 consecutive measurements and intermittent usage for six months with storage in a phosphate buffer at 4 °C. The construction parameters of the enzyme electrode were optimized. The influences of pH and temperature on the biosensor response were explored. The sensitivities of the enzyme electrode for catechol, phenol, p -cresol and m -cresol were 2.03, 0.96, 1.38 and 1.5 A M −1 cm −2 respectively.

Abstract:A novel glucose biosensor was prepared by in situ electropolymerization of aniline in GRA synthesized by using electrochemical expansion of graphite in propylene carbonate electrolyte onto platinum (Pt) electrode, followed by electrodeposition of platinum nanoparticles (PtNPs) on Pt/PANI-GRA electrode. Glucose oxidase (GOD) was immobilized on this modified Pt electrode through chitosan. The fabricated process and the electrochemical behaviors of resulting biosensor were investigated by scanning electron microscopy (SEM) and cyclic voltammetry (CV). The developed glucose biosensor exhibited superior sensitivity of 52.67 mu A mM(-1) cm(-2) and selectivity, which showed no response to the interference from glycine (Gly), Urea (Urea), L-phenylalanine (L-Phe), ascorbic acid (AA), tyrosine (L-Tyr) and D-galactose (D-Gal). The biosensor also showed a fast response time within 3 s. The biosensor was used for the detection of glucose in human blood samples with satisfactory results compared with standard hospital laboratory analysis.
Fabrication of an Amperometric Flow-Injection Microfluidic Biosensor Based on Laccase for In Situ Determination of Phenolic Compounds

Abstract:We aim to develop an in situ microfluidic biosensor based on laccase from Trametes pubescens with flow-injection and amperometry as the transducer method. The enzyme was directly immobilized by potential step chronoamperometry, and the immobilization was studied using cyclic voltammetry and electrochemical impedance spectroscopy. The electrode response by amperometry was probed using ABTS and syringaldazine. A shift of interfacial electron transfer resistance and the electron transfer rate constant from 18.1 kΩ to 3.9 MΩ and 4.6 × 10 −2 cm s −1 to 2.1 × 10 −4 cm s −1 , respectively, evidenced that laccase was immobilized on the electrode by the proposed method. We established the optimum operating conditions of temperature (55°C), pH (4.5), injection flow rate (200 μ L min −1 ), and applied potential (0.4 V). Finally, the microfluidic biosensor showed better lower limit of detection (0.149 μ M) and sensitivity (0.2341 nA μ M −1 ) for ABTS than previous laccase-based biosensors and the in situ operation capacity.
