Abstract:Background Participation of compatible living donors and recipients in kidney paired donation (KPD) could double the number of KPD transplants. We determined the willingness of previous directed donors and their recipients to participate in KPD and identified the association of various factors, including financial incentives, with willingness to participate. Methods Survey of previous directed living kidney donors and their recipients in a single Canadian center between 2001 and 2009. Results Among 207 of 222 eligible living donors contacted, 86 (42%) completed the anonymous survey: 93% (78/86) of donors indicated willingness to participate in KPD if this option had been provided at the time of donation. An increased willingness to participate was reported among the majority of respondents if reimbursements for lost wages and travel expenses were provided; however, cash payments between $5 000 and $50 000 had little impact on willingness. Willingness was also increased with an advantage to the recipient (younger donor or better human leukocyte antigen match), whereas delays beyond 3 months and donor travel were associated with reduced willingness to participate. Among 38 recipients approached during routine clinical follow-up visits over a 3-month period, 100% completed the survey, and 36 of 38 (92%) reported they would have been willing to participate in KPD. Conclusions Over 90% of directed donors and recipients were willing to participate in KPD. Reimbursement for the costs of participation and improved efficiency of KPD (i.e., eliminating travel and reducing transplant times), but not cash payments, may increase participation of compatible donors and recipients in KPD.

Abstract:Rotordynamic systems have had many applications for decades. Well known examples include gas turbines and compressors used intensively in many important industry fields. Driven by rotating shafts transmitting large amounts of kinetic energy and utilized with continuous heavy loads, these machines very often suffer from sudden unpredicted failures. Such failure usually results in abrupt release of the transmitted energy, machine damage and a catastrophic accident. Propagating fatigue cracks are dangerous malfunctions, which had been recognized as the most frequent causes of similar accidents in the past. The fatigue shaft crack initiates from a small surface imperfection or a micro-crack due to a combination of corrosive, thermal, loading and other processes. Advancing slowly yet continuously the crack may achieve the critical depth at which an immediate shaft fracture occurs. Therefore the fatigue shaft crack, and especially the methods for its early detection and warning have a great deal of attention. It is widely known that a developing shaft crack manifests itself in the appearance of nonlinear effects resulting in higher harmonics in a vibration spectrum. However, such symptoms are characteristic not only for developing shaft cracks but also for other malfunctions such as a shaft bow, a coupling misalignment, etc. That is why novel shaft crack detection methods introduce a specially designed diagnostic force applied to the shaft in order to amplify the particular symptoms of the crack. Most often a simple harmonic force is used for such purposes, yet the results may not be very reliable. The present paper analyzes the possible application of the multisine excitation technique for an early shaft crack detection problem. This technique has been previously used for many problems where the influence of nonlinear distortions on the linear system behavior is to be evaluated and measured. By using well chosen periodic excitations, called random multisines, it is possible not only to detect but also to quantify these distortions. In the present paper the multisine technique is adopted for the rotating cracked shaft. The approach is illustrated with the numerical results of the flexible rotating shaft modeled with the rigid finite element method. In order to asses the effectiveness of the proposed method the numerical results obtained for the uncracked and cracked rotors are compared. The robustness of the method to additional disturbances is checked by introducing some noise to the system. The results demonstrate a potential of the presented method. Possible problems of its practical implementation are briefly discussed.
Insect fat body cell morphology and response to cold stress is modulated by acclimation

Abstract:Mechanistic understanding about the nature of cellular cryoinjury and mechanisms by which some animals survive freezing while others do not, is currently lacking. Here we exploited the broadly-manipulable freeze tolerance of larval malt flies (Chymomyza costata) to uncover cell and tissue morphological changes associated with freeze mortality. Diapause induction, cold acclimation, and dietary proline supplementation generate malt fly variants ranging from weakly to extremely freeze tolerant. Using confocal microscopy and immunostaining of the fat body, Malpighian tubules, and anterior midgut we described tissue and cytoskeletal (F-actin and α-tubulin) morphologies among these variants after exposure to various cold stresses (from chilling at -5°C to extreme freezing at -196°C), and upon recovery from cold exposure. Fat body tissue appeared to be the most susceptible to cryoinjury; freezing caused coalescence of lipid droplets, loss of α-tubulin structure, and apparent aggregation of F-actin. A combination of diapause and cold acclimation substantially lowered the temperature at which these morphological disruptions occurred. Larvae that recovered from a freezing challenge repaired F-actin aggregation but repaired neither lipid droplet coalescence nor α-tubulin structure. Our observations indicate that lipid coalescence and damage to α-tubulin are non-lethal forms of freeze injury, and suggest that repair or removal (rather than protection) of actin proteins is a potential mechanism of acquired freeze tolerance.

Abstract:Background: Many insect species have evolved the ability to survive extracellular freezing. The search for the underlying principles of their natural freeze tolerance remains hampered by our poor understanding of the mechanistic nature of freezing damage itself. Objectives: Here, in search of potential primary cellular targets of freezing damage, we compared mitochondrial responses (changes in morphology and physical integrity, respiratory chain protein functionality, and mitochondrial inner membrane (IMM) permeability) in freeze-sensitive vs. freeze-tolerant phenotypes of the larvae of the drosophilid fly, Chymomyza costata . Methods: Larvae were exposed to freezing stress at −30°C for 1 h, which is invariably lethal for the freeze-sensitive phenotype but readily survived by the freeze-tolerant phenotype. Immediately after melting, the metabolic activity of muscle cells was assessed by the Alamar Blue assay, the morphology of muscle mitochondria was examined by transmission electron microscopy, and the functionality of the oxidative phosphorylation system was measured by Oxygraph-2K microrespirometry. Results: The muscle mitochondria of freeze-tolerant phenotype larvae remained morphologically and functionally intact after freezing stress. In contrast, most mitochondria of the freeze-sensitive phenotype were swollen, their matrix was diluted and enlarged in volume, and the structure of the IMM cristae was lost. Despite this morphological damage, the electron transfer chain proteins remained partially functional in lethally frozen larvae, still exhibiting strong responses to specific respiratory substrates and transferring electrons to oxygen. However, the coupling of electron transfer to ATP synthesis was severely impaired. Based on these results, we formulated a hypothesis linking the observed mitochondrial swelling to a sudden loss of barrier function of the IMM.

Insect cross-tolerance to freezing and drought stress: role of metabolic rearrangement

Abstract:The accumulation of trehalose has been suggested as a mechanism underlying insect cross-tolerance to cold/freezing and drought. Here we show that exposing diapausing larvae of the drosophilid fly, Chymomyza costata to dry conditions significantly stimulates their freeze tolerance. It does not, however, improve their tolerance to desiccation, nor does it significantly affect trehalose concentrations. Next, we use metabolomics to compare the complex alterations to intermediary metabolism pathways in response to three environmental factors with different ecological meanings: environmental drought (an environmental stressor causing mortality), decreasing ambient temperatures (an acclimation stimulus for improvement of cold hardiness), and short days (an environmental signal inducing diapause). We show that all three factors trigger qualitatively similar metabolic rearrangement and a similar phenotypic outcome—improved larval freeze tolerance. The similarities in metabolic response include (but are not restricted to) the accumulation of typical compatible solutes and the accumulation of energy-rich molecules (phosphagens). Based on these results, we suggest that transition to metabolic suppression (a state in which chemical energy demand is relatively low but need for stabilization of macromolecules is high) represents a common axis of metabolic pathway reorganization towards accumulation of non-toxic cytoprotective compounds, which in turn stimulates larval freeze tolerance.

Abstract:Cryopreservation, one of the most effective preservation methods, is essential for maintaining the safety and quality of food. However, there is no denying the fact that the quality of muscle food deteriorates as a result of the unavoidable production of ice. Advancements in cryoregulatory materials and techniques have effectively mitigated the adverse impacts of ice, thereby enhancing the standard of freezing preservation. The first part of this overview explains how ice forms, including the theoretical foundations of nucleation, growth, and recrystallization as well as the key influencing factors that affect each process. Subsequently, the impact of ice formation on the eating quality and nutritional value of muscle food is delineated. A systematic explanation of cutting-edge strategies based on nucleation intervention, growth control, and recrystallization inhibition is offered. These methods include antifreeze proteins, ice-nucleating proteins, antifreeze peptides, natural deep eutectic solvents, polysaccharides, amino acids, and their derivatives. Furthermore, advanced physical techniques such as electrostatic fields, magnetic fields, acoustic fields, liquid nitrogen, and supercooling preservation techniques are expounded upon, which effectively hinder the formation of ice crystals during cryopreservation. The paper outlines the difficulties and potential directions in ice inhibition for effective cryopreservation.
Dissolved Gases and Ice Fracturing During the Freezing of a Multicellular Organism: Lessons from Tardigrades

Abstract:Three issues are critical for successful cryopreservation of multicellular material: gases dissolved in liquid, thermal conductivity of the tissue, and localization of microstructures. Here we show that heat distribution is controlled by the gas amount dissolved in liquids and that when changing the liquid into solid, the dissolved gases either form bubbles due to the absence of space in the lattice of solids and/or are migrated toward the concentrated salt and sugar solution at the cost of amount of heat required to be removed to complete a solid-state transition. These factors affect the heat distribution in the organs to be cryopreserved. We show that the gas concentration issue controls fracturing of ice when freezing. There are volumetric changes not only when changing the liquid into solid (volume increases) but also reduction of the volume when reaching lower temperatures (volume decreases). We discuss these issues parallel with observations of the cryosurvivability of multicellular organisms, tardigrades, and discuss their analogy for cryopreservation of large organs.
Proline: A Novel Cryoprotectant for the Freeze Preservation of Cultured Cells of Zea mays L.

Abstract:Proline is an effective cryoprotectant for the storage of cultured cells of Zea mays L. in liquid N(2). Increased freeze tolerance can be achieved by pregrowth for 3 to 4 days in medium containing proline. Cells cryoprotected with proline have an increased recovery potential when compared with cells cryoprotected with dimethylsulfoxide and glycerol. They also show a reduced postthaw viability loss and greater tolerance of a range of postthaw culture conditions. It is suggested that the mechanism of action of proline may be similar to that in its putative role of conferring protection against natural stresses. It may be protecting the cell against solution effects caused by dehydration during freezing. These findings are discussed in relation to other freeze tolerance enhancing treatments.

Cold tolerance of the drosophila pupal parasitoid Trichopria drosophilae

Abstract:Trichopria drosophilae (Perkins) (Hymenoptera: Diapriidae) is a pupal parasitoid of drosophila flies recorded from several parts of the world. It is currently considered for augmentative biological programs to control the severe agricultural pest Drosophila suzukii (Matsumura) (Diptera: Drosophilidae). Since D. suzukii has invaded regions that experience zero and sub-zero winter temperatures, cold tolerance of the parasitoid is an important aspect to consider. We characterized low temperature tolerance and overwintering capacity of a colony of T. drosophilae collected in Northern Switzerland. We used copper-constantan thermocouples to determine the supercooling point and pre-freeze mortality. Moreover, we subjected honey-fed and unfed adult T. drosophilae as well as developing stages within their drosophila host to short- and long-term acclimation conditions and assessed the duration of their survival at low temperatures. Finally, we exposed adult and sub-adult stages to winter conditions in a semi-field experiment and evaluated their survival. We found that T. drosophilae is chill susceptible like D. suzukii, but adults froze and survived at colder temperatures than those reported for D. suzukii. Adult parasitoids could tolerate several days of exposure to sub-zero temperatures and could reproduce afterwards, whereas sub-adult stages could survive longer periods under these conditions. The provision of honey and water enhanced the survival of adults and long-term acclimation led to longer survival in all stages. The semi-field experiment supported the results of the laboratory tests. Based on these results we suggest that in Central Europe, T. drosophilae survives winters mainly in developing stages but adults are likely able to tolerate short periods of low spring temperatures.

Abstract:Cryopreservation can keep the bioactivity of biological specimens in long-term storage, but it is hard to retain the structural integrity due to serious thermomechanical stress during cooling and warming procedures, especially for complex living organisms. Few studies have reported on the thermomechanical stress of biological specimens in a pre-freezing supercooled state, which is a widespread phenomenon in slow-rate freezing cryopreservation. Here, we develop a thermomechanical coupling model to study transient thermal and mechanical fields of supercooled biological specimens experiencing freezing procedures. The results show that cryoprotectant accumulation in insects causes pronounced supercooled phenomena with severe deformation and thermomechanical stress in the initial state of phase transition. However, the loss of freezable water induced that final deformation and stress decrease, which is beneficial to organism survival after freezing. This numerical method is proved to be a guideline for optimizing slow-rate freezing cryopreservation protocols efficiently and economically. These results reveal survival mechanisms of insects with supercooled phenomena after freezing and assist researchers in exploring more valuable cryopreservation methods for biological specimens.

To overwinter, insects have adapted a range of strategies to become truly cold hardy. However, while the mechanisms underlying the ability to avoid or tolerate freezing have been well-studied, little attention has been given to the challenge of maintaining ion homeostasis at frigid temperatures in these species, despite this limiting cold tolerance for insects susceptible to mild chilling. Here we investigate how prolonged exposure to temperatures just above the supercooling point affects ion balance in freeze-avoiding mountain pine beetle (Dendroctonus ponderosae) larvae in autumn, mid-winter, and spring, and relate it to organismal recovery times and survival. Hemolymph ion balance was gradually disrupted during the first day of exposure, characterized by hyperkalemia and hyponatremia, after which a plateau was reached and maintained for the rest of the seven-day experiment. The degree of ionoregulatory collapse correlated strongly with recovery times, which followed a similar asymptotical progression. Mortality increased slightly during extensive cold exposures, where hemolymph K+ concentration was highest, and a sigmoidal relationship was found between survival and hyperkalemia. Thus, the cold tolerance of the freeze-avoiding larvae of D. ponderosae appears limited by the ability to prevent ionoregulatory collapse in a manner similar to chill-susceptible insects, albeit at much lower temperatures. Based on these results, we propose that a prerequisite for the evolution of insect freeze-avoidance may be a convergent or ancestral ability to maintain ion homeostasis during extreme cold stress.

Abstract:Background: L-proline is a natural, nontoxic cryoprotectant that helps cells and tissues to tolerate freezing in a variety of plants and animals. The use of L-proline in mammalian oocyte cryopreservation is rare. In this study, we explored the cryobiological characteristics of L-proline and evaluated its protective effect in mouse oocyte cryopreservation.Methods: The freezing property of L-proline was detected by Raman spectroscopy and osmometer. Mature oocytes obtained from 8-week-old B6D2F1 mice were vitrified in a solution consisting various concentration of L-proline with a reduced proportion of dimethyl sulfoxide (DMSO) and ethylene glycol (EG), comparing with the control group (15 % DMSO and 15% EG without L-proline). The survival rate, 5-methylcytosine (5-mC) expression, fertilization rate, two-cell rate, and blastocyst rate in vitro were assessed by immunofluorescence and in vitro fertilization. Data were analyzed by Chi-square test.Results: L-proline can penetrate the oocyte membrane within 1 min. The osmotic pressure of 2.00 mol/L L-proline mixture is similar to that of the control group. The survival rate of the postthawed oocyte in 2.00 mol/L L-proline combining 7.5% DMSO and 10% EG is significantly higher than that of the control group. There is no difference of 5-mC expression between the L-proline combination groups and control. The fertilization rate, two-cell rate, and blastocyst rate in vitro from oocyte vitrified in 2.00 mol/L L-proline combining 7.5% DMSO and 10% EG solution are similar to that of control.Conclusions: It indicated that an appropriate concentration of L-proline can improve the cryopreservation efficiency of mouse oocytes with low concentrations of DMSO and EG, which may be applicable to human oocyte vitrification.

Abstract:Anoplophora glabripennis is a critical global quarantine pest. Recently, its distribution has been extended to colder and higher-latitude regions. The adaptation to low temperatures is vital for the successful colonization of insects in new environments. However, the metabolic pathways of A. glabripennis larvae under cold stress remain undefined. This study analyzed the larval hemolymph under different low-temperature treatments using LC-MS/MS. The results showed that differential metabolites associated with sugar and lipid metabolism are pivotal in the larval chill coma process. Under low-temperature treatments, the glycerol content increased significantly compared with the control group. Cold stress significantly induced the expression of AglaGK2 and AglaGPDHs. After undergoing RNAi treatment for 48 h, larvae exposed to -20 °C for 1 h showed reduced recovery when injected with ds-AglaGK2 and ds-AglaGPDH1 compared to the control group, indicating that glycerol biosynthesis plays a role in the low-temperature adaptation of A. glabripennis larvae. Our results provide a theoretical basis for clarifying the molecular mechanism of A. glabripennis larvae in response to environmental stresses.

Abstract:Cryopreservation of biological samples is an important technology for expanding their applications in the biomedical field. However, the quality and functionality of samples after rewarming are limited by the toxicity of commonly used cryoprotectant agents (CPAs). Here, we developed a novel preservation system by combining the natural amino acid l-proline (L-Pro) with gelatin methacryloyl (GelMA) hydrogels. Compared with dimethyl sulfoxide (DMSO), L-Pro and GelMA demonstrated excellent biocompatibility when co-culturing with cells. Cryopreservation procedures were optimized using 3T3 as model cells. The results showed that rapid cooling was the most suitable cooling procedure for L-Pro and GelMA among the three cooling procedures. Co-culturing with cells for 3 h before cryopreservation, 6 % L-Pro +7 % GelMA had the highest survival rate, reaching up to 80 %. Differential Scanning Calorimetry (DSC) analysis showed that 6 % L-Pro + 7 % GelMA lowered the freezing point of the solution to -4.2 °C and increased the unfrozen water content to 20 %. To the best of our knowledge, this is the first report of cell cryopreservation using a combination of L-Pro and GelMA hydrogels, which provides a new strategy for improving cell cryopreservation.

Abstract:Since biotic interactions within the simple terrestrial communities on the Antarctic Peninsula are limited compared with tropical and temperate regions, survival is largely dictated by the numerous abiotic challenges. Our research focuses on adaptations to environmental stresses experienced by the Antarctic midge (Belgica antarctica Jacobs, 1900), the southernmost free-living insect. Midge larvae can survive freezing and anoxia year-round. Not only can frozen larvae undergo rapid cold-hardening (RCH) at temperatures as low as –12 °C, but RCH develops more rapidly in frozen compared with supercooled larvae. Whether larvae overwinter in a frozen state or cryoprotectively dehydrated may depend on hydration levels within their hibernacula. Larvae constitutively up-regulate genes encoding heat shock proteins, as well as the antioxidant enzymes superoxide dismutase and catalase. Larvae accumulate osmoprotectants in response to freezing, desiccation, and exposure to seawater; exposure to one of these osmotic stressors confers cross-tolerance to the others. Molecular responses to dehydration stress include extensive genome-wide changes that include differential expression of aquaporins among tissues, upregulation of pathways associated with autophagy, inhibition of apoptosis, and downregulation of metabolism and ATP production.

Abstract:Overwintering adults of Pyrrhocoris apterus do not tolerate freezing of their body fluids and rely on a supercooling strategy and seasonal accumulation of polyols to survive at subzero body temperatures. We sampled the adults monthly in the field during the cold season 2008-2009 and found active thermal hysteresis factors (THFs) in hemolymph of winter-sampled adults. The hysteresis between the equilibrium melting and freezing points ranged from 0.18°C to 0.30°C. No signs of THFs activity were found in the autumn- and spring-sampled insects. The total free amino acid pool almost doubled during winter time. The sum concentrations of 27 free amino acids ranged between 35 and 40mM in whole body water and 40-45mM in hemolymph during December-February. Two amino acids, Pro and α-Ala most significantly contributed to the seasonal increase, while Gln showed the most dramatic seasonal decrease. Moderate levels of amino acid accumulation in overwintering P. apterus suggest that they are by-products of protein degradation and pentose pathway activity during the state of metabolic suppression imposed by diapause and low body temperature. Potential colligative effects of accumulated amino acids, extending the supercooling capacity of overwintering P. apterus, are negligible. Non-colligative effects require further study.
Antifreeze protein complements cryoprotective dehydration in the freeze-avoiding springtail Megaphorura arctica

Abstract:Abstract The springtail, Megaphorura arctica , is freeze-avoiding and survives sub-zero temperatures by cryoprotective dehydration. At the onset of dehydration there is some supercooling of body fluids, and the danger of inoculative freezing, which would be lethal. To see if the springtails are protected by antifreeze proteins in this pre-equilibrium phase, we examined extracts from cold-acclimated M. arctica and recorded over 3 °C of freezing point depression. Proteins responsible for this antifreeze activity were isolated by ice affinity. They comprise isoforms ranging from 6.5 to 16.9 kDa, with an amino acid composition dominated by glycine (>35 mol%). Tryptic peptide sequences were used to identify the mRNA sequence coding for the smallest isoform. This antifreeze protein sequence has high similarity to one characterized in Hypogastrura harveyi , from a different springtail order. If these two antifreeze proteins are true homologs, we suggest their origin dates back to the Permian glaciations some 300 million years ago.
